Abstract

A survey of chilli fields in the state of Karnataka, India, showed the presence of bacterial wilt disease in important chilli growing regions. The disease incidence ranged from 26?–?32%. The pathogen was isolated from infected plant material and seeds. Infected plant material showed the release of milky white bacterial ooze. Burkholderia solanacearum was detected from chilli seeds by liquid assay and its identity was confirmed by biochemical tests, hypersensitive reaction and pathogenicity tests. Seed transmission of the pathogen up to 45% was observed in seeds artificially infested with the pathogen. Among different tissues of the seed, endosperm showed the presence of the pathogen. Biological seed treatment with antagonistic Pseudomonas fluorescens significantly (p?=?0.05) improved the seed quality parameters under laboratory conditions and drastically reduced the bacterial wilt incidence under field conditions. Seed-borne nature, transmission and effect of Pseudomonas fluorescens in both the forms of pure culture and formulation on seed quality parameters and bacterial wilt incidence are discussed in the present work.  相似文献

An efficient DNA extraction protocol and polymerase chain reaction (PCR) assay for detecting Leptosphaeria maculans from infected seed lots of oilseed rape were developed. L. maculans, the causal agent of blackleg, a damaging disease in oilseeds rape/canola worldwide, was listed as a quarantine disease by China in 2009. China imports several millions of tons of oilseeds every year. So there is a high risk that this pathogen will be introduced to China via contaminated seeds. Seed contamination is one of the most significant factors in the global spread of phytopathogens. Detection of L. maculans in infected seed lots by PCR assay is difficult due to the low level of pathogen mycelium/spores on seeds and PCR inhibitors associated with the seeds of oilseed rape. In our study, these two major obstacles were overcome by the development of a two‐step extraction protocol combined with a nested PCR. This extraction protocol (kit extraction after CTAB method) can efficiently extract high‐quality DNA for PCR. Amplification results showed that the detection threshold for conventional PCR and nested PCR was, respectively, 1 ng and 10 fg of DNA per μl in mycelia samples. On contaminated seed lots of oilseed rape, the detection threshold of conventional and nested PCR was 709 fg/μl and 709 ag/μl of DNA, respectively. The DNA extraction protocol and PCR assay developed in this study can be used for rapid and reliable detection of L. maculans from infected seeds of oilseed rape .  相似文献

Downy mildew on sweet basil (Ocimum basilicum L.) occurs worldwide. Contaminated seeds are considered as the primary inoculum source. So far no strategy to control the disease is available. Hence, the use of pathogen-free seeds is the only alternative to prevent disease outbreaks. Therefore, a rapid diagnostic method for seed testing is urgently needed. The sensitivity of a specific PCR method for direct detection of the downy mildew pathogen Peronospora belbahrii on basil samples, particularly on seeds, was evaluated. The applied PCR method proved to be very sensitive for direct detection of the pathogen on seeds and plant samples. The PCR detection limit of P. belbahrii in artificially infested seeds corresponded to the DNA amount of a single spore per seed. Additionally, the systemic spread of the pathogen from naturally infected seeds was investigated. The experiments showed that outgrowing basil plants were latently infected with the downy mildew pathogen, and the infection continued within the plant. Contaminated seeds were harvested from symptomless latently infected plants. These results support the implementation of PCR-based detection in a seed certification scheme and the necessity to control the pathogen on seeds. The PCR method can also be used for evaluation of pathogen control on seeds based on detection of the pathogen in outgrowing plants.  相似文献

Aims: To evaluate the effectiveness of the optimized immunomagnetic separation (IMS)‐plating protocol in relation to other culture, serological and molecular techniques currently used for Clavibacter michiganensis subsp. michiganensis in seed‐testing laboratories. Methods and results: Bacterial suspensions, tomato seed extracts spiked with the pathogen and naturally infected seeds were IMS‐plated for the detection of C. m. subsp. michiganensis. These results were compared with plating on general (YPGA) and semiselective (mSCM) media, double‐antibody sandwich enzyme‐linked immunosorbent assay (DAS‐ELISA), immunofluorescent assay (IF) or polymerase chain reaction (PCR). Different seed lots and pathogen strains were also tested. IMS‐plating allowed the detection of less than 10 CFU ml?1 of pathogen in all assayed samples. The mSCM medium provided positive results for 10 CFU ml?1 in naturally infected seeds, but up to 14 days was necessary for the typical colonies of the target to be come visible. By serological techniques, 103 and up to 104 CFU ml?1 were detected by IF and ELISA, respectively. DNA extraction was required to obtain positive results by PCR in seed extracts containing 103 CFU ml?1 or more. Conclusions: Among the evaluated methods, IMS‐plating provided the best results regarding sensitivity and specificity for C. m. subsp. michiganensis detection, allowing the recovery of viable bacteria from seed extracts. Significance and impact of the study: IMS‐plating increases isolation rates of C. m. subsp. michiganensis and could improve standard protocols currently used for routine analysis.  相似文献

AIM: To develop a diagnostic assay based on polymerase chain reaction for the detection of Magnaporthe grisea from infested rice seeds. METHODS AND RESULTS: Primers were designed based on the nucleotide sequence of the mif 23, an infection-specific gene of M. grisea. The primers amplified target DNA from genetically and geographically diverse isolates of the pathogen. The lowest concentration of template DNA that led to amplification was 20 rhog. No PCR product was detected when DNA from other fungi was used, indicating the specificity of the primers. With this PCR based seed assay, M. grisea was detected in rice seedlots with infestation rates as low as 0.2%. CONCLUSION: The PCR detection of M. grisea is simple, rapid, specific, sensitive and suitable for the routine detection of the pathogen in infested seeds. SIGNIFICANCE AND IMPACT OF THE STUDY: Introduction of the blast fungus into new areas where it has not been previously recorded could be avoided by the detection of infested seedlots. A PCR-based seed assay could facilitate risk assessment of naturally infested rice seeds; help design management programs and optimize fungicide use.  相似文献

Seed‐borne pathogenic fungi can cause serious damage to soybean crops by reducing the germination, vigour and emergence of the seeds. Special attention should be paid to pathogen detection in seeds to prevent its introduction in disease‐free areas. Considering the importance of rapid and successful diagnosis of seed‐borne pathogenic fungi in soybeans, this study evaluated a method to detect Sclerotinia sclerotiorum and Phomopsis spp. in seeds using quantitative polymerase chain reaction (qPCR). Naturally infested samples were subjected to detection using qPCR and blotter test, and the findings were compared. Using soybean seeds soaked in water, both pathogens were detected at an infestation level up a 0.0625% (one infected seed out of 1,599 healthy seeds) by qPCR. This technique allowed the detection of 300 fg of S. sclerotiorum and 30 fg of Phomopsis spp. DNA in the seed samples. Phomopsis spp. was detected in 40.7% of the evaluated seed batches (81 batches) and S. sclerotiorum was detected in 32.1% of the evaluated batches, although most of the seeds had low infestation levels. It was up to 28.5 times more efficient to use qPCR rather than blotter test to detect pathogens with a low incidence of occurrence in soybean seeds. If routinely used to test healthy seeds, qPCR would contribute to reducing soybean losses due to diseases as well as decreasing the costs required to control those diseases.  相似文献

The results of experiments on transmission of Corynebacterium michiganense pv. michiganense showed that the spread of the bacterium by artificially infested seeds varies, but it was confirmed that from artificially infested seeds few diseased plants can be grown. The results depend on the environmental conditions during experimentation and the method of assaying the seedlings. High relative humidity favours the spread of the pathogen. The use of selective media helped the detection ofthe pathogen in seemingly healthy plants. It was proved that host can have population of C. michiganense pv. michiganense in a resident phase. It is assumed that when conditions are favorable the pathogen multiplies and cause disease.  相似文献

Aflatoxin, a potent carcinogen, is produced by the fungus Aspergillus flavus Link: Fr. Drought, high temperatures, and insect damage contribute to increased levels of aflatoxin contamination in corn, Zea mays L. Plant resistance is widely considered a desirable method of reducing aflatoxin contamination. Germplasm lines with aflatoxin resistance have been developed. This investigation was undertaken to determine whether crosses among these lines exhibited resistance to southwestern corn borer, Diatraea grandiosella Dyar, and to assess the effects of southwestern corn borer feeding on aflatoxin accumulation. Differences in ear damage among southwestern corn borer infested hybrids were significant. Estimates of general combining ability effects indicated that the lines Mp80:04, Mp420, and Mp488 contributed to reduced ear damage, and SC213 and T165 contributed to greater damage when used in hybrids. Mean aflatoxin levels were 254 ng/g for hybrids infested with southwestern corn borer larvae and 164 ng/g for noninfested hybrids in 2000 when environmental conditions were conducive to aflatoxin production. In contrast, the overall mean aflatoxin level for southwestern corn borer infested hybrids was only 5 ng/g in 1999 when environmental conditions did not favor aflatoxin accumulation. Crosses that included lines selected for aflatoxin resistance as parents (Mp80:04 and Mp313E) exhibited lower levels of aflatoxin contamination both with and without southwestern corn borer infestation in 2000. Only the experimental line Mp80:04 contributed significantly to both reduced southwestern corn borer damage and reduced aflatoxin contamination.  相似文献

Clusterbean seed health testing is warranted since the pathogen (Xanthomonas campestris pv. cyamopsidis (Xccy)) is seed-borne and seed-transmitted. A polyclonal antibody was developed in rabbit via subcutaneous and intramuscular injections and characterized for sensitivity, specificity and its applicability to ELISA which: (i) was sensitive in detecting as few as 102 cells ml - 1 at a titre of 1: 4000; (ii) was specific, since it reacted only with Xccy and not with other xanthomonads; (iii) reacted both with Xccy cells and culture filtrate, indicating that the antigenic determinant is a secretory component; (iv) was applicable and reliable in seed health testing since it reacted only with infected seeds and plant materials and not with healthy seeds and (v) a purified fraction of antibody was virulent-specific since heat-denatured and avirulent isolates were not detected. The ELISA thus developed is highly reproducible and therefore suitable for the evaluation of the potential disease status of seeds and plant health, which is appropriate for routine seed health testing.  相似文献

Nitrogen (N) and phosphorus (P) fertilizers have differing effects on southwestern corn borer (SWCB). Diatraea grandiosella Dyar, densities and damage to field corn, Zea mays L. Nitrogen fertilization of corn is associated with an increase in number of stalks infested and girdled by corn borers, while addition of P has the opposite effect. When combinations of N and P were applied, there were no significant differences in SWCB infestation or damage from plots receiving no fertilizer. The effect of combinations of N and P on SWCB were intermediate to either element alone.
Résumé Nous avons étudié les corrélations entre l'utilisation d'engrais azotés et phosphatés d'une part, l'infestation et l'alimentation de D. grandiosella sur maïs en champs d'autre part. Les formules d'engrais comparées étaient: pas d'engrais, 90 et 179 kg N/ha sans P, 45 et 90 kg P/ha sans N, et les combinaisons de ces deux teneurs en N et P.La contamination était plus élevée dans les lots ayant reçu de l'azote que dans ceux traités sans N, mais la présence d'azote n'a pas réduit le décapage des tiges et la verse dus à l'alimentatin larvaire. A l'inverse, les lots n'ayant reçu que du phosphore avaient des pourcentages plus faibles de tiges contaminées par D. grandiosella et ainsi décapées et versées, que ceux qui n'avaient pas reçu P. Il n'a pas eu de différences significatives dans les contaminations ou les dégats entre les lots ayant reçu différentes combinaisons de N et P, et les effets de ces combinaisons sur l'insecte étaient intermédiaires à ceux de N et P seuls.

Fusarium oxysporum f.sp. asparagi (Foa) incites crown and root rot of asparagus which causes early decline of asparagus plantings. The aim of the present study was to identify the main inoculum sources of the pathogen in the Netherlands. As has been reported for foreign seed lots, Dutch seed lots can be infested with Foa at low levels. We found that seed infestation occurs mainly during the seed harvesting process through infested soil adhering to fallen berries. Soil samples from 59 fields without a history of asparagus growing and differing in their distance from asparagus plantings were tested for infestation with Foa, using a bioassay with asparagus as a bait plant. A high correlation was found between the incidence of infestation and proximity to asparagus fields; Foa was found in 69% of the samples from fresh fields in an asparagus production centre, and in only 6% of the samples from fields at a distance of 1 km and more from asparagus fields and outside a production centre. To evaluate planting material as an inoculum source of Foa, 49 lots of one-year-old crowns from 23 nurseries were collected and rated for disease symptoms. Infestation was found to be common with only two lots free of symptomatic plants. Most of the lots had more than 75% of symptomatic plants. Although most of the plants were infested, they showed only slight root rot symptoms. The procedure for production of Foa-free planting material is discussed. Persistence and infestation of asparagus root residues in former asparagus fields was assessed by retrieving the residues from eight former asparagus fields with an asparagus-free period of one to 25 years, and three fields with a standing asparagus crop. Even after an asparagus-free period of 25 yr asparagus root residues were retrieved from soil, although at low levels. Mean population densities of Fusarium spp. declined from 2 times 106 to 1 times 105 colony forming units g_1 air-dry root tissue during the first 10 years and were still > 104 c.f.u. g“1 air-dry root tissue 20 to 25 yr after asparagus produced was stopped. The population was dominated by F. oxysporum. Eighty-three of the 112 isolates (74%) of F. oxysporum belonged to the forma specialis asparagi. The proportion of Foa in the population did not decrease in time. It was concluded that persistence of Foa in asparagus root residues is a major reason for its long-term survival.  相似文献

Testing of seed and grain lots is essential in the enforcement of GM labelling legislation and needs reliable procedures for which associated errors have been identified and minimised. In this paper we consider the testing of oilseed rape seed lots obtained from the harvest of a non-GM crop known to be contaminated by volunteer plants from a GM herbicide tolerant variety. The objective was to identify and quantify the error associated with the testing of these lots from the initial sampling to completion of the real-time PCR assay with which the level of GM contamination was quantified. The results showed that, under the controlled conditions of a single laboratory, the error associated with the real-time PCR assay to be negligible in comparison with sampling error, which was exacerbated by heterogeneity in the distribution of GM seeds, most notably at a small scale, i.e. 25 cm3. Sampling error was reduced by one to two thirds on the application of appropriate homogenisation procedures.  相似文献

Tests in Britain on samples of basic and commercial Brassica oleracea seed between 1976 and 1978 showed that many lots were infected with Alternaria brassicicola. A. brassicae was uncommon in basic seed in these years and in commercial seed harvested in 1976 and 1977 but was frequent in seed harvested in 1978. Most affected seeds were contaminated by surface-borne spores and mycelium of A. brassicicola but many were internally infected by the fungus situated within the seed-coat and in some seeds in the embryo tissues. Superficial contamination by the fungus declined rapidly after 2 yr in cabbage seeds stored at 10 °C, 50% r.h. but internal infection persisted for up to 12 yr. In some samples, internal infection was commonly associated with small shrivelled seeds. Surface contaminated and internally infected seeds transmitted the disease but seedling infection was more closely correlated with the latter.  相似文献

Black scurf and stem canker on potato is an economically important disease complex, causing both quantitative and qualitative damage to potato crops which occurs in potato production areas throughout the world. The ribosomal DNA internal transcribed spacer sequence analysis is currently accepted and a commonly used method for classifying Rhizoctonia species and anastomosis groups (AGs). To date, 13 AGs have been recognized. The updated AG distribution in potato worldwide production areas confirm the status of AG‐3 as the most prevalent AG in potato and reflects the population dynamics of the pathogen probably due to global trading of tubers. As R. solani is a tuber‐ and soilborne pathogen, the ability to detect its levels in the seed tubers and in the soil and predict the potential damage is an important factor in controlling the disease. Effective disease management of Rhizoctonia disease requires implementation of an integrated disease management approach and knowledge of each of its stages. Although the most important control measures are cultural, chemical control (either by seed tuber‐ or in‐furrow treatments) is still an important tool in reducing the damages caused by R. solani.  相似文献

In the present study we attempt to determine the principal factors which influence the contamination by Penicillium of 116 samples of pre-harvest corn from five crop areas in Valencia (Spain).In the corn tested, Penicillium is the most abundant genus after Fusarium, with an incidence of 71.5% in infected samples, counts of about 642 cfu/g and kernel infection of 8.7%. The most frequently isolated species are Penicillium chrysogenum, P. steckii and P. purpurogenum.There is a high correlation between the contamination from this genus and the amout of kernels broken and infested by insects, as well as a correlation with crop areas.We have found no difference in the contamination of the two varieties of corn investigated.  相似文献

A semi‐solid fermentation product of the potential biocontrol fungus Stilbella aciculosa was formulated on wheat bran: water (1:1, w/w) and incubated 5, 10 and 15 days before addition to soil infested with the pathogen Rhizoctonia solani. Generally, preparations did not reduce survival of the pathogen in infested beet seed but they did prevent saprophytic growth of the pathogen from beet seed into soil. The magnitude of reduction by the 15‐day‐old inoculum was greater than that by the 5‐day‐old inoculum. Ten‐day‐old bran preparations of S. aciculosa at rates of 0.5 and 1.0% (w/w) in soil prevented post‐emergence damping‐off of cotton, radish and sugar beet in the glasshouse and a rate of 1.0% gave stands similar to those in the non‐infested control soil. The antagonist, grown on perlite formulated with molasses, cornmeal, alfalfa tissue or corn stover, prevented damping‐off of cotton in a naturally infested soil. However, the stands were not as great as that in soil planted with pentachloronitrobenzene (PCNB)‐treated seed. Toxic metabolites, produced by S. aciculosa developing on various substrates, slightly inhibited the growth of R. solani in culture and induced cytoplasmic leakage of the pathogen mycelium.  相似文献

Dollar spot is the most economically important disease of amenity turfgrasses in the United States, yet little is known about the source of primary inoculum for this disease. With the exception of a few isolates from the United Kingdom, Sclerotinia homoeocarpa, the causal agent of dollar spot, does not produce spores. Consequently, it was assumed that overwintering of this organism in soil, thatch, and plant debris provides primary inoculum for dollar spot epidemics. Overwintering of S. homoeocarpa in roots and shoots of symptomatic and asymptomatic creeping bentgrass turfgrass was quantified over the course of a three-year field experiment. Roots did not consistently harbor S. homoeocarpa, whereas S. homoeocarpa was isolated from 30% of symptomatic shoots and 10% of asymptomatic shoots in the spring of two out of three years. The presence of stroma-like pathogen material on leaf blades was associated with an increase in S. homoeocarpa isolation and colony diameter at 48 hpi. Commercial seed has also been hypothesized to be a potential source of initial inoculum for S. homoeocarpa. Two or more commercial seed lots of six creeping bentgrass cultivars were tested for contamination with S. homoeocarpa using culture-based and molecular detection methods. A viable, pathogenic isolate of S. homoeocarpa was isolated from one commercial seed lot and contamination of this lot was confirmed with nested PCR using S. homoeocarpa specific primers. A sensitive nested PCR assay detected S. homoeocarpa contamination in eight of twelve (75%) commercial seed lots. Seed source, but not cultivar or resistance to dollar spot, influenced contamination by S. homoeocarpa. Overall, this research suggests that seeds are a potential source of initial inoculum for dollar spot epidemics and presents the need for further research in this area.  相似文献

Douglas fir seeds infested by diapausing larvae of a chalcid, Megastigmus spermotrophus, were transferred under different temperature and photoperiod regimes at various dates during autumn and winter. Both winter diapause and prolonged diapause were compared to the diapause patterns observed in control lots kept under natural conditions. The results showed that exposure to low temperatures in autumn and winter is required for the completion of diapause development. Median adult emergence dates and the date of transfer of larvae to 20°C were negatively correlated. The frequency of prolonged diapause of larvae transferred to 20°C decreased significantly after November. When larvae were submitted to different constant temperatures, the longer the daylength, the higher and faster the reactivation. These results suggest that chalcid larvae can perceive daylength through the seed coat. This phenomenon is probably related to the fact that most of the infested seeds falling in autumn are usually covered by a layer of leaf litter during the winter. BACKGROUND AND AIMS: Pathogen-seed interactions may involve a race for seed resources, so that seeds that germinate more quickly, mobilizing reserves, will be more likely to escape seed death than slow-germinating seeds. This race-for-survival hypothesis was tested for the North American seed pathogen Pyrenophora semeniperda on seeds of the annual grass Bromus tectorum, an invasive plant in North America. In this species, the seed germination rate varies as a function of dormancy status; dormant seeds germinate slowly if at all, whereas non-dormant seeds germinate quickly. METHODS: Three experimental approaches were utilized: (a) artificial inoculations of mature seeds that varied in primary dormancy status and wounding treatment; (b) naturally inoculated undispersed seeds that varied in primary dormancy status; and (c) naturally inoculated seeds from the carry-over seed bank that varied in degree of secondary dormancy, habitat of origin and seed age. KEY RESULTS: In all three approaches, seeds that germinated slowly were usually killed by the pathogen, whereas seeds that germinated quickly frequently escaped. Pyrenophora semeniperda reduced B. tectorum seed banks. Populations in drier habitats sustained 50 times more seed mortality than a population in a mesic habitat. Older carry-over seeds experienced 30 % more mortality than younger seeds. CONCLUSIONS: Given the dramatic levels of seed death and the ability of this pathogen to reduce seed carry-over, it is intriguing to consider whether P. semeniperda could be used to control B. tectorum through direct reduction of its seed bank.  相似文献
