Seven enrichment media (two proposed by the authors) for detecting salmonellas from polluted freshwater were compared. The Most Probable Number technique for enumeration of salmonellas in water samples was used, directly adding filtered water to buffered peptone water as the pre-enrichment medium. The results indicate that Rappaport-Vassiliadis/43 and Rappaport-Vassiliadis/43 supplemented with 10 micrograms of sodium novobiocin per ml are the best media for the recovery and enumeration of salmonellas from water samples.  相似文献

The efficiency of Rappaport's broth (RB10) and Rappaport's broth containing novobiocin (NRB10) were compared for the isolation of salmonellas from polluted water, both as direct enrichment media and after pre-enrichment in buffered peptone water. Ninety samples were examined and 41 were found to contain salmonellas by at least one of the procedures used. Direct inoculation of the sample into RB10 resulted in the recovery of salmonellas from only 29.3% of the samples found to be positive. The use of NRB10 as a direct enrichment medium increased the percentage recovery to 78.0% of the total positive samples. Pre-enrichment in buffered peptone water allowed the recovery of salmonellas from a total of 41 samples whereas direct enrichment recovered them from only 32. No significant difference was demonstrated in the efficiencies of RB10 and NRB10 in recovering salmonellas after pre-enrichment in buffered peptone water. Three selective agars were used; no significant difference in their ability to recover salmonellae was demonstrated.  相似文献

Comparisons of selenite cystine (SC) and TT enrichment broths for detecting salmonellas were made with pure culture suspensions, with samples of naturally or artificially contaminated foods and with poultry feed. Selenite cystine recovered higher numbers of salmonellas from pure cultures and ground beef, while TT broth recovered higher numbers from pork sausage and poultry feed. Differences in the recovery of salmonellas from other food products appeared to be insignificant. The use of both SC and TT is thus recommended for maximal recovery of these organisms.  相似文献

Rectal swabs and faeces were used in the regular sampling for salmonellas and Arizonas from a heavily-infected population of a marsupial, the quokka ( Setonix brachyurus ). The media used were strontium selenite A and strontium chloride B enrichment broths, with subculture onto modified bismuth sulphite agar and deoxycholate citrate agar. A study of sampling, enrichment, sub-culture and colony selection procedures produced an optimal scheme giving high yields but consistent with reasonable economy of time and materials. A three-swab sample was taken and inoculated into the two enrichment media, and with each enrichment subjected to three subcultures. The absolute efficiency of this procedure was greater than 80% (and confirmed by a serological method), compared with only 67% for a single swab in a single enrichment. Recovery of some serotypes depended on the media used; e.g. Arizonas could not be recovered satisfactorily from strontium chloride B enrichment. Faeces samples were found to be greatly superior to rectal swabs for detecting salmonellas and arizonas but they were less convenient in field studies. In a comparison of rectal swabs and faeces samples where the actual concentration of salmonellas was known, it was found that the efficiency of rectal swabs approached 100% if there were more than 103 salmonellas/g faeces, but this declined to approximately 50% if there were 102-103 salmonellas/g faeces and only 25% if there were less than 102 salmonellas/g faeces. A new statistical procedure was introduced for comparing the number of isolations from two methods, and this should be of use in similar methodological studies.  相似文献

AIMS: The prevalence and types of salmonella in broiler chickens during transportation and during slaughter and dressing were studied. This was part of a comprehensive investigation of salmonellas in two UK poultry companies, which aimed to find the origins and mechanisms of salmonella contamination. METHODS AND RESULTS: Salmonellas were isolated using cultural methods. Serovars of Salmonella detected during rearing were usually also found in a small proportion of birds on the day of slaughter and on the carcasses at various points during processing. There was little evidence of salmonellas spreading to large numbers of carcasses during processing. Many serovars found in the feedmills or hatcheries were also detected in the birds during rearing and/or slaughter. Transport crates were contaminated with salmonellas after washing and disinfection. CONCLUSIONS: Prevalence of salmonellas fell in the two companies during this survey. A small number of serovars predominated in the processing plants of each company. These serovars originated from the feed mills. Reasons for transport crate contamination were: (1) inadequate cleaning, resulting in residual faecal soiling; (2) disinfectant concentration and temperature of disinfectant too low; (3) contaminated recycled flume water used to soak the crates. SIGNIFICANCE AND IMPACT OF THE STUDY: Efforts to control salmonella infection in broilers need to concentrate on crate cleaning and disinfection and hygiene in the feed mills.  相似文献

Studies have been carried out in which growth patterns of a Salmonella sp. and competing micro-organisms, especially other Enterobacteriaceae, were followed during pre-enrichment in buffered peptone water (BPw) and subsequent selective enrichment in tetrathionate broth (TBB). Pre-enrichment cultures were inoculated with minced meat and three reference samples containing nalidixic acid-resistant salmonellas. Irrespective of their initial numbers in BPw, Enterobacteriaceae increased to 108/ml or more. During incubation in TBB at 43C, numbers of lactose-positive Enterobacteriaceae decreased in most enrichments which resulted in a positive salmonella isolation, but remained constant in the majority of those that did not. Levels of lactose-negative Enterobacteriaceae did not decrease in most salmonella-positive tests, but did so in half of the salmonella-negative ones. In the salmonella-positive tests the numbers of salmonellas had increased to 103–107/ml in BPw and after transfer to TBB slowly reached 104/ml or more. In all cases the numbers of salmonellas exceeded those of the competing flora on brilliant green agar (BGA). In the salmonella-negative tests the numbers of salmonellas had increased less in BPw and decreased in most of the TBB enrichments. In none of these negative tests did the numbers of salmonellas exceed those of the competing flora on BGA. Escherichia coli dominated in most of the salmonella-negative tests. The results suggest more influence of lactose-positive than lactose-negative Enterobacteriaceae on the detection of salmonellas. The effect of competing microorganisms seems to depend not only upon their initial numbers, but also upon the types that can interact with salmonellas during selective enrichment.  相似文献

Three sandwich-ELISAs, two of which are commercially available (Tecra and Locate), and one developed at the Veterinary Sciences Division, Stormont and a 3-step culture protocol, were compared for the detection of salmonella in 1000 animal specimens. Eight hundred and fifty of these were new submissions and the remainder were frozen portions from specimens previously shown to contain salmonellas by culture. The incidence of ELISA false-negative and false-positive results was highest for the Stormont and Locate kits respectively although the differences in sensitivity and specificity between the three ELISAs was not statistically significant. On 16 occasions all ELISA methods indicated the presence of salmonellas when none were isolated by initial culture, eight of these specimens contained salmonellas when reinvestigated by culture.  相似文献

The Removal of Salmonellas in Conventional Sewage Treatment Processes   总被引:2,自引:2,他引:0  
The numbers of salmonellas in raw sewage entering a treatment plant varied hourly and diurnally; their peak concentration preceded the peak influent waste water flow into the plant by about 2 hours. Salmonellas were detected in all raw influent samples collected from 2 sewage works and the mean population level at the daily peak period was 3000 organisms/1. On average. 70–80% of the salmonellas were removed during primary sedimentation when upwards of 74–84% of solids were removed. Biological treatment and secondary sedimentation removed a further 70–100% of the pathogen. Compared with the activated sludge process the trickling filters were less efficient in removing salmonellas and they were adversely affected by increased loading following rainfall. Considering the whole treatment process, the Guildford works with its activated sludge treatment removed an average of 99–83% salmonellas while the Woking works with its trickling filter plant removed an average of 93–04% of the organisms. The large variance in the numbers of salmonellas in the final effluent from the trickling filters suggest that greater emphasis should be placed on the actual quality of the effluent rather than on percentage removal efficiencies.  相似文献

Sewage sludge was monitored for the presence of salmonellas after it had been dumped on a pasture so that it formed a number of extensive, thick pats. Although salmonellas were isolated from the sewage sludge, and also from grass and soil beneath the sludge for up to 94 days, they were not isolated from the faeces of grazing horses or surrounding drainage ditches. The results are discussed with regard to the possible hazards resulting from the use of sewage sludge on pasture.  相似文献

A modified lysine decarboxylase broth has been developed which could be used with a Bactometer M123 to differentiate salmonellas from other bacteria by the characteristics of the conductance detection curve. The medium was used in combination with a selenite cystine trimethylamine oxide dulcitol medium to screen 50 strains of salmonellas and 42 strains of other organisms to establish detection curve magnitude and rate values which could be used to identify curves specific to salmonellas. The combination of media detected all salmonellas tested except Salmonella pullorum . The two media were used to screen 100 inoculated product samples with the Bactometer instrument, in parallel with traditional plating procedures, and using various combinations of pre-enrichment and selective enrichment incubation periods. After 24 h pre-enrichment, the Bactometer system detected more positive samples than the conventional plating procedures after pre-enrichment and selective enrichment. It is considered that these media used in parallel in the Bactometer after conventional pre-enrichment could provide a 48 h screening procedure for salmonellas with a sensitivity comparable to present plating procedures.  相似文献

(1976) after pre-enrichment in buffered peptone water during the first 6 h was obtained by replacement of tryptone by soya peptone. The competing bacteria, i.e. those which grow on brilliant green agar and which may interfere with the isolation of salmonellas when Rappaport's medium (R10) incubated at 43°C is used for enrichment were inhibited or reduced in numbers when the normal amount of 5 g soya peptone/litre was used. When the amount was increased to 10 g/l, growth occurred, mainly of Enterobacter and Klebsiella species. The isolation of salmonellas was found to be largely dependent on the number and the ratio of their competitors. Every measure taken to reduce the number of competitive bacteria increases the possibility of isolating salmonellas. This explains the effect of improved selectivity of Rappaport's medium when small inocula are used. Rapid onset of growth of salmonellas by employing soya peptone introduces the possibility of using shorter incubation times than 48 h.  相似文献

Ninety-five salmonellas and 40 non-salmonellas were screened in the Bactometer using lysine-iron-cystine-neutral red (LICNR) in order to evaluate the selectivity of the medium for the detection of salmonellas. Results for blackening of the medium in the well (indicating hydrogen sulphide, H2S, production) and step size are presented. Five out of 95 salmonellas tested failed to produce blackening in the well, four of these five are known to be non-H2S producers. Although salmonellas generally gave a larger step value than non-salmonellas, this criterion could not be used to distinguish reliably the two groups of organisms.  相似文献

In a continuous biogas plant, receiving manure from 200 dairy cows and 400 calves and young stock, survival of salmonellas and Ascaris suum eggs was studied. The bacteria and parasite eggs were kept in filter sacs in the manure that had a temperature of 55°C. No viable salmonellas or Ascaris suum eggs could be found after 24h in the digester. Survival of salmonellas and Ascaris suum eggs was also studied in the manure pit where the manure was stored after digestion. The temperature in the manure pit varied between 22–27°C. Salmonellas survived 35 but not 42 days. On day 56, when the experiments had to be stopped, 60% of the Ascaris eggs were viable.  相似文献

Improvements in performance of a lysine conductance medium for the detection of salmonellas were achieved from a study of the effects of its various components. When sodium biselenite was included as an inhibitor for non-salmonella organisms conductance signals were depressed. The inclusion of sodium chloride reduced this toxicity and improved conductance responses. Increasing the pH to pH 7.0 prevented the medium becoming too acidic and inhibitory to salmonellas. The new medium detected 70-0% of salmonella-positive animal protein samples.  相似文献

Salmonella destruction efficiencies of 99% were obtained after 10 h aeration at 15°C in a laboratory model of the activated sludge process. This study demonstrated that, in a batch process, the removal of salmonellas occurred in three phases. (i) By 4 h, 90% of the original inoculum had disappeared from the activated sludge, probably due mainly to predation by ciliated protozoa. The remaining 10% was distributed between the liquid phase (approximately 90%) and the sludge floc (approximately 10%). (ii) During the next 2 h this situation was inverted so that, by 6 h, more than 80% of the remaining salmonellas were then adsorbed to floc, leaving less than 20% in liquid suspension. (iii) From 6 h onwards there was a much slower decline of the remaining salmonellas attached to floc. The addition of dioctyl sodium sulphosuccinate after 4 h, inactivated the ciliated protozoa populations and completely eliminated the continued reduction of salmonellas from activated sludge observed previously.  相似文献
