The detailed effect on the light-harvesting apparatus of three different wild-type strains of Rhodopseudomonas acidophila in response to changes in both light-intensity and temperature have been investigated. In all three strains at high light-intensities (160 mol s m2 and above) the only LH2 antenna complex synthesised is the B800–850 complex. In strains 7050 and 7750 as the light-intensity is lowered the B800–850 complex is gradually replaced by another type of LH2 the B800–820 complex. However, at no light-intensities studied is this changeover complete when the cells are grown at 30°C. If however, the light-intensity is lowered at temperatures below 25°C with strain 7750 there is a complete replacement of the B800–850 complex by the B800–820 complex. At all light-intensities and temperatures tested, strain 10050 only synthesised the B800–850 complex. Strain 7050 also responded to changes in light-intensity by altering its carotenoid composition. At high light-intensity the major carotenoids were rhodopin and rhodopin-glucoside, while at low light-intensities the major ones were rhodopinal and rhodopinal-glucoside. This change in carotenoid content started to occur at rather higher light-intensities than the switchover from B800–850 to B800–820.  相似文献

A methanol-utilizing phototrophic bacterium, strain M402, was isolated from surface water of an acidic hot spring. The isolated strain was identified as Rhodopseudomonas acidophila from its morphological and physiological characters. Profiles of the utilization of non-aromatic compounds as carbon sources by this strain were in good agreement with those of some strains of R. acidophila reported by Pfennig [J. Bacteriol., 99, 597 (1969)]. However, strain M402 was found to be capable of utilizing vanillic acid, vanillin, vanillyl alcohol, ferulic acid, veratric acid, syringic acid, syringal-dehyde and benzyl alcohol as carbon sources under anaerobic-light conditions. Although Pfennig did not refer to these abilities of his strains, these notable characters of strain M402 seem to be additional new characters of R. acidophila.  相似文献

A chemolithoautotrophic type of metabolism, which was hitherto unknown for purple nonsulfur bacteria, was demonstrated by growth experiments using Rhodopseudomonas capsulata Kb1 and Rhodopseudomonas acidophila 10050. These strains were able to grow in a mineral medium in the dark at the expense of H2, O2, and CO2. A minimum doubling time of 9 h was obtained for R. capsulata under an atmosphere containing less than 15% oxygen; higher oxygen concentrations suppressed autotrophic but not chemoorganotrophic growth. Oxygen sensitivity of chemoautotrophically growing cells of R. acidophila was even more pronounced, whereas cells growing chemotrophically on methanol almost tolerated the oxygen concentration of air. Highest oxygen sensitivity of growth of R. acidophila was observed with formate as substrate. The growth yield of cultures grown semiaerobically in the dark on methanol was 0.23 g dry cell material per g methanol consumed.  相似文献

Summary
  • 1 The planktonic food web in extremely acidic mining lakes is restricted to a few species that are either acidophilic or acidotolerant. Common metazoans inhabiting acidic mining lakes with a pH below 3 include rotifers in the genera Cephalodella and Elosa.
  • 2 The life history response of Cephalodella acidophila to three environmental key factors, pH (2, 3.5, 5.0 and 7.0), temperature (10, 17.5 and 25 °C) and food concentration (10 000, 35 000 and 50 000 algal cells per mL), was investigated in a full factorial design using life‐table experiments.
  • 3 The effect of each of the three environmental variables investigated on the rotifer life cycle parameters (life span, fecundity and population growth rate) differed. C. acidophila is a stenoecious species with a pH optimum in the range 3–4 and a comparably high food threshold. Combining the laboratory results with field data, we conclude that C. acidophila is severely growth limited in its natural habitat. However, low pH alone is not harmful as long as temperatures are moderate to warm and food is abundant.

To appraise the usefulness of the enzyme-linked immunosorbent assay (ELISA) technique for examining the serological diversity of slow-growing rhizobia, twelve diverse strains from three countries were examined with four antisera. Eleven of the strains were from the cowpea miscellany, and the twelfth was a Rhizobium japonicum strain. Some cowpea strains showed no antigenic relatedness with each other while others were closely related, and some showed a greater affinity with the R. japonicum strain than with other cowpea strains. All of the strains showed antigenic homology to an isolate from a wild Arachis sp., while two strains isolated from adjacent plants of the same cultivar had little homology. These patterns ofrelatedness and diversity clearly demonstrated the utility of the ELISA method, and so it was used to examine 53 strains isolated from cowpeas grown at three West African locations, Maradi (Niger), Ibadan and Onne (Nigeria). Broad ranges of serological diversity were found in the rhizobia at each location, moreover each population had its own general characteristics. Maradi strains were highly reactive with the five antisera used, Onne strains less so, and Ibadan strains even less so. ELISA reactivity correlated with colony morphology but not with nodulation potential.  相似文献

Forty-six strains of tempe-forming Rhizopus species were screened for their ability to grow on raffinose as the sole carbon source. Six of the strains showed good growth and sporulation. These isolates were one Rhizopus oligosporus, one Rhizopus microsporus var. chinensis, three Rhizopus oryzae and one Rhizopus stolonifer. These six moulds and R. oligosporus strain NRRL 2710 were investigated for their metabolism of the raffinose family of -galactoside carbohydrates. Degradation experiments were performed in submerged culture in a medium containing soybean -protein, sodium phytate and either stachyose, raffinose or melibiose. R. oryzae and R. stolonifer completely consumed the tested carbohydrates as carbon source. R. microsporus var. chinensis failed to hydrolyse the -galactosidic bonds of raffinose, stachyose or melibiose, whereas it was able to use sucrose and the fructose moiety of raffinose or stachyose for growth. R. oligosporus NRRL 2710 was unable to hydrolyse any of the tested carbohydrates. The results of the oligosaccharide degradation experiments could be verified during tempe production from soybeans with the selected fungal species.  相似文献

Summary A constitutive -galactosidase has been localized in the cytosol of thermoacidophilic bacterium Caldariella acidophila. Cells have been entrapped in polyacrylamide gel with full retention of enzymic activity; no activity decrease is observed after 8 months of storage. Enzyme properties in entrapped cells are similar to those of the free enzyme. A 73% hydrolysis of lactose has been achieved in a continuous system on a 2 ml entrapped cell column operating at 70°C; half life in these conditions is 30 days.In this paper we report preliminary data on immobilization of cells of Caldariella acidophila, an extreme thermophilic bacterium having a constitutive -galactosidase (EC 3.2.1.23) activity.  相似文献

Cells of Chlamydomonas acidophila Negoro, isolated from three soils with different available copper contents (74, 80, and 87 μg·g?1), were assayed for their responses to copper. Soil pH ranged from 3.3–3.9. Responses were evaluated using algistatic assays involving five day exposure to copper concentrations from 0.1–100 mg·L?1 at pH 3.8 and 6.6 in defined liquid media. Interspecies and intraspecies comparisons were made between the soil isolates and laboratory strains of C. reinhardtii and C. acidophila, respectively. Algistatic copper concentrations of soil isolates were 20–125 times greater than those of the laboratory strain of C. reinhardtii. Concentrations of 0.1 mg Cu·L?1, or greater, killed the laboratory strain of C. acidophila. Soil isolates of C. acidophila appeared to be copper tolerant; however, there was no conclusive evidence to indicate that the level of copper tolerance in the soil isolates was positively correlated with the level of available copper in the soil.  相似文献

Ten Escherichia coli strains isolated from five different aquatic environments representing three distinct geographical regions of India showed significantly high levels of tolerance to the inorganic form of mercury, i.e., mercuric chloride (HgCl2). MRD14 isolated from the Dal Lake (Kashmir) could tolerate the highest concentration of HgCl2, i.e., 55 g/mL, and MRF1 from the flood water of the Yamuna River (Delhi) tolerated the lowest concentration, i.e., 25 g/mL. All ten strains revealed the presence of a plasmid of approximately 24 kb, and transformation of the isolated plasmids into the mercury-sensitive competent cells of E. coli DH5 rendered the transformants resistant to the same concentration of mercury as the wild-type strains. Mating experiments were performed to assess the self-transmissible nature of these promiscuous plasmids. The transfer of mercury resistance from these wild-type strains to the mercury-sensitive, naladixic acid-resistant E. coli K12 (Flac+) strain used as a recipient was observed in six of the nine strains tested. Transconjugants revealed the presence of a plasmid of approximately 24 kb. An evaluation of the mechanism of mercury resistance in the three most efficient strains (MRG12, MRD11, and MRD14) encountered in our study was determined by cold vapor atomic absorption spectroscopy (CV-AAS), and it was noted that resistance to HgCl2 was conferred by conversion of the toxic ionic form of mercury (Hg++) to the nontoxic elemental form (Hg0) in all three strains. Rhodopseudomonas acidophila strain 7050 achieved balanced growth when provided with either asparagine or glutamine as nitrogen source. Under these growth conditions R. acidophila synthesized a mixed amidase which exhibited similar activity (223–422 nmol/min·mg protein) against either nitrogen source. Determination of the free intracellular amino acid pools show that deamidation of asparagine and glutamine resulted in elevated levels of both aspartate and glutamate. Cell-free extracts of R. acidophila showed significant aminotransferase activity, particulary glutamine-oxaloacetate aminotransferase (89.7–209.3 nmol/min·mg protein), glycine oxaloacetate aminotransferase (135–227 nmol/min ·mg protein), alanine glyoxylate aminotransferase (66.3–163.2 nmol/min·mg protein) and serineglyoxylate aminotransferase (57.1–68.4 nmol/min ·mg protein). Short term labelling experiments using 14C-glyoxylate show that glycine plays an important role in amino nitrogen transfer in R. acidophila and that the enzymes for the metabolism of glyoxylate via glycine, serine and hydroxypyruvate were present in cell-free extracts. Lipopolysaccharides (LPS) from a number of purple nonsulfur bacteria and of phylogenetically related species were analyzed for the presence of sialic acid by gas chromatography/mass spectrometry. Species and strains of the genera Rhodobacter, Rhodopseudomonas, Rhodomicrobium, Rhodospirillum, Rhodocyclus and Rhodopila were investigated, sialic acid, however, was found only in the genus Rhodobacter. It occurs in strains of Rhodobacter capsulatus, R. sphaeroides, R. sulfidophilus and R. veldkampii. All these species belong to the -3 subgroup of purple bacteria as defined by 16S rRNA catalogues. Approximately equimolar ratios of sialic acid and of 2-keto-3-deoxy-octonate (KDO) were found in isolated LPSs. Sodium deoxycholate gel electrophoresis of these LPS-samples also suggested a location of sialic acid in the LPS core region. Two Arctic species of Raphidonema, a weakly filamentous green alga, were compared on the basis of their growth and morphology response to varied environmental factors. The strain of R. nivale was isolated from snow and R. sempervirens from soil collected in Svalbard. Morphological variability was evaluated in both strains, with emphasis laid on pleiomorphism of R. nivale. Growth characteristics in varied temperature and light were compared. Field observations were performed in summer 2003 at Werenskiold glacier, southwest Spitsbergen. In both strains, temperature and light were found to affect the shape and size of the cells, and in R. nivale also the number of cells in filament, probably by modifying growth and division rates. The strain of R. nivale grew best at 12°C and 200 mol m–2 s–1, R. sempervirens at 18°C and 50 mol m–2 s–1. In snow, R. nivale only appeared after katabatic wind had brought soil on the snow surface, and its concentrations decreased very rapidly. Cells of R. nivale in snow showed great signs of damage. On the basis of obtained results, it is suggested here that R. nivale is a soil species well adapted to soil environment, and only occasionally brought on snow.  相似文献

Three new species belonging to the genusCryptococcus have been described.C. kutzingii was among the unidentified strains present in the collection of the Centraalbureau voor Schimmelcultures. The type strain had been isolated from a medlar fruit.C. dimennae was isolated by di Menna from pasture plants in New Zealand.C. lactativorus was isolated from Antarctic sea water. Among the four methods for extracting extracellular polymeric substances (EPS) from Rhodopseudomonas acidophila (EDTA, NaOH, H2SO4, heating/centrifugation), EDTA extraction was found to be the most effective. The contents of the major components of EPS from R. acidophila, i.e., carbohydrate, protein and nucleic acid, were 6.5, 58.4 and 5.4 mg g–1 dry cells, respectively. The optimum extraction time was 1–3 h and the EDTA dosage was approximately 2.8 g g–1 dry cells. Under these conditions, no cell lysis was observed. The EPS content and the percentage of the three main components were greatly dependent on the extraction method. The intensity of absorption peaks for photosynthetic pigments in the UV–visible spectrum of bacteria remained unchanged prior to and after EDTA extraction; and no pigment peaks appeared in the EPS spectrum. This suggests that few cells were destroyed and lysis did not occur. UV–visible spectrum analysis, an easy and rapid technique, could be used to monitor cell lysis during EPS extraction from R. acidophila.  相似文献

S. panama 47 (antibiotic-sensitive, phage pattern A) was infected with R factors from a number of field strains of Enterobacteriaceae (Salmonella, Escherichia coli, Citrobacter, Enterobacter, Klebsiella andProteus) isolated from human and animal sources. These R factors could be grouped into 11 types i.e. R1 R11 on the basis of induced changes in the phage type of the recipient.R8 and R11 renderS. panama resistant to the phages A H:S. panama 47 (R3) and 47 (R6) adsorb the phages A F, but there is no phage multiplication: phages G and H are considered to be restricted and modified in these strains. The R factors R5 and R7 also exert restriction and modification on a number of the typing phages A H. The nature of the changes in phage pattern brought about by R4, R9 and R10 is not understood. R2 does not exert restriction (i.e. no change in phage pattern). The R factors were also investigated for the fi (fertility inhibition) and spp (restriction of phage ) markers.The R factors R3 R11 readily segregate, in the sense that the restriction and modification loci, and occasionally the Resistance Transfer Factor as a whole was frequently lost after R transfer. These 9 types of R factors were encountered infrequently in the present material.Resistance to tetracycline inS.panama is nearly always due to R factors of type R1. In other members of Enterobacteriaceae, notably inE.coli, R1 is less frequently found than R2.  相似文献

Fast growing strains of Rhizobium loti isolated from nodules of Lotus tenuis of the flooding Pampas of Argentina produced cellular (1–2)glucans having a higher degree of polymerization and more anionic substituents than (1–2)glucans accumulated by Agrobacterium tumefaciens cells. Inner membranes of R. loti contained a 235 kDa (1–2)glucan intermediate protein indistinguishable by polyacrylamide gel electrophoresis from the intermediate protein present in A. tumefaciens inner membranes. Incubation of inner membrannes of R. loti with UDP-Gle led to the formation of neutral (1–2)glucans with a higher degree of polymerization than glucans formed by A. tumefaciens inner membranes.Introduction in R. loti strains of plasmid pCD523 containing A. tumefaciens chvA and chvB virulence regions yielded strains that accumulated 4 times more cellular (1–2)glucans than wild type cells. This glucan was, regarding anionic substitution and degree of polymerization, indistinguishable from A. tumefaciens (1–2)glucans. Furthermore inner membranes of these R. loti exoconjugant cells contained higher levels of the 235 kDa (1–2)glucan intermediate protein and formed in vitro 8 times more neutral (1–2)glucan with a degree of polymerization corresponding to A. tumefaciens (1–2)glucan than inner membranes isolated from wild type cells.It was concluded that A. tumefaciens chvB gene is expressed in R. loti and determined the degree of polymerization of (1–2)glucan.Abbreviations Nod+ effective nodulation - Vir+ virulent - Vir- avirulent - Trpr trimethoprim resistence - Tcr tetracycline resistence - TCA trichloroacetic acid - PMSF phenyl methyl sulfonyl fluoride  相似文献

Summary Dunaliella acidophila is an unicellular green alga which grows optimally at pH 0–1 while maintaining neutral internal pH. A plasma membrane preparation of this algae has been purified on sucrose density gradients. The preparation exhibits vanadatesensitive ATPase activity of 2 mol Pi/mg protein/min, an activity 15 to 30-fold higher than that in the related neutrophilic speciesD. salina. The following properties suggest that the ATPase is an electrogenic plasma membrane H+ pump. (i) ATP induces proton uptake and generates a positive-inside membrane potential as demonstrated with optical probes. (ii) ATP hydrolysis and proton uptake are inhibited by vanadate, diethylstilbestrol, dicyclohexylcarbodiimide and erythrosine but not by molybdate, azide or nitrate. (iii) ATP hydrolysis and proton uptake are stimulated by fussicoccin in a pH-dependent manner as found for plants plasma membrane H+-ATPase. Unusual properties of this enzyme are: (i) theK m for ATP is around 60 M, considerably lower than in other plasma membrane H+-ATPases, and (ii) the ATPase activity and proton uptake are stimulated three to fourfold by K+ and to a smaller extent by other monovalent cations. These results suggest thatD. acidophila possesses a vanadate-sensitive H+-ATPase with unusual features enabling it to maintain the large transmembrane pH gradient.  相似文献

Simple-sequence tandem repeat sequences in the 3 UTR of interleukin 5 (IL5)-receptor gene of human and mouse are polymorphic in their length among humans and different strains of mice. In 20 different human Epstein-Barr virus (EBV)-transformed cell lines, six alleles of IL5R could be distinguished. In the mouse, three different alleles are found. With the human-specific IL5R tandem repeat marker in human-rodent somatic cell hybrids, the IL5R gene was mapped to human Chromosome (Chr) 3 p25–p26. With the mouse-specific IL5R tandem repeat sequence in recombinant inbred strains of mice, the Il5r gene was mapped to the distal part of mouse Chr 6 close to the Raf-1 locus.  相似文献

Motile Aeromonas hydrophila strains were recovered from several freshwater sources by spread-plating water samples on starch-ampicillin agar, originally described as a medium for recovering Aeromonas hydrophila quantitatively from foods. Starch-ampicillin agar was compared with membrane Aeromonas medium and Rimler-Shotts medium for selectivity for, and recovery of, Aeromonas strains from freshwater. Thirty-four Aeromonas strains thus isolated were identified to species level by their phenotypic characteristics, and the Mol% G+C of representative strains was determined. Although resistant to 10 g of the vibriostatic agent 0/129, all these strains showed sensitivity to 150 g 0/129, which brings into question the use of this agent for distinguishing aeromonads from vibrios. The ability of these strains to produce extracellular virulence factors was generally similar to that reported for environmental strains isolated by other methods from various geographical locations within and beyond Australia. Ten of the 20 A. sobria strains, but none of the A. hydrophila or A. caviae strains, produced enterotoxin as shown by the suckling mouse test. Haemolysin was produced by 9/10 of the enterotoxigenic A. sobria strains and 2/9 A. hydrophila strains. Hemagglutinating activity was detected in 5/20 A. sobria and 7/9 A. hydrophila strains, and was inhibited by fucose and mannose, but not by galactose. The characteristics of these strains were comparable with those of Aeromonas strains isolated from other freshwater environments apart from their sensitivity to 0/129. Send offprint requests to: M. Summary Cells of thermophilic archaebacterium Caldariella acidophila have been trapped in chicken egg white by co-crosslinking with glutaraldehyde. The activity of cytosolic -galactosidase in trapped cells has been followed by varying temperature, pH, organic solvents.
