Abstract: The largest aggregations of harbor seals (Phoca vitulina) in Alaska, USA, haul out on floating ice in tidewater glacial fjords. Seals use these fjords in peak numbers during the critical periods of pupping, breeding, and molting when visits by tour ships also peak. Documented and suspected declines of harbor seals in fjords with rising vessel traffic underscore the need to better understand possible impacts, particularly in areas where ship visits have risen substantially in the past 2 decades. We examined the interruption of haul-out bouts of harbor seals due to approaching cruise ships in Disenchantment Bay, Alaska. We conducted observations from cruise ships and focused on disturbance of seals as evidenced by seals flushing into the water from the floating ice on which they rested. We investigated rate of flushing in relation to vessel distance, approach angle, group size, and seal type (mother, pup, or other). Using a survival-regression analysis, we found that the risk of disturbing harbor seals increased when ships approached within 500 m; seals approached as close as 100 m were 25 times more likely to enter the water than seals 500 m from a ship. Seals were 4 times more prone to enter the water when ships were approaching directly rather than passing abeam. Seals responded similarly regardless of group size or seal type. Energetic models indicated a potential to disrupt energy balance and cause thermal stress in disturbed pups if they spent >50% of their time in ice-chilled water. Studies at non-glacial sites suggest that pups spend 40–70% of their time in the water. Voluntary guidelines for approaching seals in Alaska recommend that cruise ships approach ≥91 m (100 yards), a distance at which we show 90% of seals would flush into the water. Our findings indicate a need to develop regulations to maintain a 500-m separation between cruise ships and seals in all Alaskan glacial fjords.  相似文献

Mobile Harbor (MH) is a movable floating platform with a container handling system on board so that it can load/discharge containers to/from an anchored container ship in the open sea. As with typical quay crane operation, an efficient schedule for its operation is a key to enhancing its operational productivity. A MH operation scheduling problem is to determine a timed sequence of loading/discharging tasks, assignment of MH units to each task, and their docking position, with an objective of minimizing the makespan of a series of incoming container ships. A mixed integer programming model is formulated to formally define the problem. As a practical solution method to the problem, this paper proposes a rule-based algorithm and a random key based genetic algorithm (rkGA). Computational results show that the rkGA method produces a better-quality solution than the rule-based method, while requiring longer computation time.  相似文献

We examine the extent to which the assumptions underlying line transect sampling are satisfied in shipboard surveys of crabeater seals ( Lobodon carcinophaga ) hauled out on the Antarctic pack-ice. Measurement of the perpendicular distance of seal groups from the ship with an electronic inclinometer fitted to a rifle stock was unbiased. Crabeater seals showed little movement in response to the approaching ship. Movement away from the ship by seals close to the ship's track was partially responsible for a relative lack of sightings close to the transect-line, but otherwise had little effect on the sighting histogram. Minor deviations from the transect direction to avoid running over seals violated the assumption of uniform distribution of groups, and contributed to a relative lack in sightings close to (<40 m) the transect-line. We estimate that 5%-10% of seal groups close to the transect-line were not sighted by bridge observers prior to passing abeam of the ship, but most of these missed groups were likely to have been sighted some distance behind-abeam. Shipboard transects provided a biased sample of four environmental features known to be related to crabeater seal abundance because of logistical difficulties in the ship traversing along straight transects through thick ice. Calculation of transect length L from successive GPS locations was mildly sensitive to the frequency of locations. We provide analytical recommendations to reduce or eliminate the effect of assumption violation when present and hence minimize bias in abundance estimation.  相似文献

Tidewater glacial fjords support the largest populations of harbor seals (Phoca vitulina richardii) in Alaska and are a prime destination for tour ships. Chronic disturbance from ships, however subtle, could impact long‐term population stability. We examined variation in abundance and distribution of harbor seals on floating ice in Disenchantment Bay, Alaska, a tour ship destination for over a century with near daily visitation by ships in the spring/summer over the last decade. Counts of seals by aerial transect showed a sharp decline in May, prior to pupping and the first ships arriving; counts rebounded by the end of June remaining high until August. Seal distribution and abundance peaked in 5–7 tenths ice cover; total area of ice cover showed no effect. Despite regular flushing of seals by ships, we found no broad‐scale patterns in seal abundance and distribution that could be explained by ship presence. We cannot rule out mechanisms of long‐term disturbance, difficult to detect and that might explain notable differences with other, similar sites. Population declines at disturbed glacial sites and the still rising popularity of vessel‐based tourism indicate a need for individual‐based studies on how seals respond to the dynamics of glacial ice environments and human‐caused stresses.  相似文献

Cheap package cruises have become very popular in Great Britain, but the ships used for these cruises are often not suitable for elderly, handicapped, and mentally unfit people. The cruises run to very tight schedules with many strenuous shore excursions, and do not necessarily constitute restful holidays at sea. Many passengers who embark on these cruises are suffering from pre-existing diseases, which may become exacerbated during the voyage. Lipid droplets (LDs) are neutral lipid-rich organelles involved in many cellular processes. A well-known example is their accumulation in leukocytes upon activation by pro-inflammatory stimuli such as lipopolysaccharides (LPS) derived from gram-negative bacteria. A role of LDs and LD-associated proteins during inflammation in the brain is unknown, however. We have now studied their dynamics and regulation in microglia, the resident immune cells in the brain. We find that LPS treatment of microglia leads to the accumulation in them of LDs, and enhancement of the size of LDs. This induction of LDs was abolished by triacsin C, an inhibitor of triglyceride biosynthesis. LPS strongly activated c-Jun N-terminal kinase (JNK) and p38 MAPK stress signaling pathways and increased the expression of LD-associated protein perilipin-2 (ADRP) in a time-dependent manner. Immunostaining showed that perilipin-2 in LPS-treated microglia predominantly colocalized with LDs. Inhibitors of p38 α/β (SB203580) and PI3K/Akt pathway (LY294002), but not that of JNK (SP600125), reduced LPS-induced LD accumulation and eliminated the activating effect of LPS on perilipin-2. In addition, cytosolic phospholipase A(2) (cPLA(2)-α), a key enzyme for arachidonic acid release, colocalized with LPS-induced LDs. These observations suggest that LDs may play an important role in eicosanoid synthesis in activated microglia; they provide a novel insight into the regulation of LDs in inflammatory cells of the brain and point to a potential role of p38 α/β in LPS-induced LD accumulation. Collectively, our findings imply that LD formation and perilipin-2 induction could be microglial biomarkers of inflammation in the central nervous system.  相似文献

Abstract: To determine the spatial resolution of Global Positioning System (GPS) receiver data, rigorous testing is essential. We tested performance of the Lotek 3300 GPS collar for medium-sized mammals (Lotek Engineering, Inc., Newmarket, ON, Canada). To mimic real wildlife monitoring situations, we performed both static (stationary receiver) and mobile tests, placing the receiver collar on a dog. We compared fix locations of the mobile receiver with the actual trajectory described by a portable Trimble high-precision GPS. We determined performance in relation to habitat type and leaf cover. Location error was habitat-dependent, with the best results in open habitat and much poorer ones in forest, particularly coniferous-dominated forest. For both static and mobile tests, location accuracy was higher when the number of satellites contacted was high and when the residual positional dilution of precision (PDOP) value was low. However, location error was highly variable, even for a given PDOP value and a given number of satellites contacted. Finally, mobile collars performed less well than their static counterparts, presumably because of frequent changes of GPS position and orientation.  相似文献

North Atlantic right whales were extensively hunted during the whaling era and have not recovered. One of the primary factors inhibiting their recovery is anthropogenic mortality caused by ship strikes. To assess risk factors involved in ship strikes, we used a multi-sensor acoustic recording tag to measure the responses of whales to passing ships and experimentally tested their responses to controlled sound exposures, which included recordings of ship noise, the social sounds of conspecifics and a signal designed to alert the whales. The whales reacted strongly to the alert signal, they reacted mildly to the social sounds of conspecifics, but they showed no such responses to the sounds of approaching vessels as well as actual vessels. Whales responded to the alert by swimming strongly to the surface, a response likely to increase rather than decrease the risk of collision.  相似文献

William Osler''s connections with the sea included a strong family history of seafaring, his own transatlantic crossings (of which there were at least 32) and the occasional use of nautical imagery in his inspirational writings. An unusual Oslerian connection with the sea emerged after his death in the form of a World War II Liberty ship. Through the SS William Osler and its sister ships, Osler was symbolically reunited with colleagues associated with the early days of the Johns Hopkins Hospital. The William Osler circumnavigated the globe in 1943 without engaging the enemy. She was then converted into an army hospital ship and renamed the USHS Wisteria.  相似文献

The lipid droplets (LDs) are intracellular organelles mainly dedicated to the storage and provision of fatty acids. To accomplish these functions the LDs interact with other organelles and cytosolic proteins. In order to explore possible correlations between the physiological states of cells and the protein composition of LDs we have determined and compared the proteomic profiles of lipid droplets isolated from the fat bodies of 5th-instar larvae and adult Manduca sexta insects and from ovaries. These LD-rich tissues represent three clearly distinct metabolic states in regard to lipid metabolism: 1) Larval fat body synthesizes fatty acids (FA) and accumulates large amounts as triglyceride (TG); 2) Fat body from adult insects provides FA to support reproduction and flight; 3) Ovaries do not synthesize FA, but accumulate considerable amounts of TG in LDs. Major qualitative and semi-quantitative variations in the protein compositions of the LDs isolated from these three tissues were observed by MS/MS and partially validated by immuno-blotting. The differences observed included changes in the abundance of lipid droplet specific proteins, cytosolic proteins, mitochondrial proteins and also proteins associated with the machinery of protein synthesis. These results suggest that changes in the interaction of LDs with other organelles and cytosolic proteins are tightly related to the physiological state of cells. Herein, we summarize and compare the protein compositions of three subtypes of LDs and also describe for the first time the proteomic profile of LDs from an insect ovary. The compositions and compositional differences found among the LDs are discussed to provide a platform for future studies on the role of LDs, and their associated proteins, in cellular metabolism.  相似文献

In September 1993, 26 live specimens of the small, delicate sea anemoneHaliplanella luciae (Verrill, 1898) were sampled from the hull of a ship docked in Hamburg Harbour. After a worldwide journey the ship had passed into the freshwater region of the river Elbe. Although the migratory potential of the species (supposed home region is Japan) is well known, its transport on ships' bottoms has never been documented. Behavioural traits enabling the anemone to settle on ships are discussed together with probable reasons whyHaliplanella luciae did not establish itself in the fauna of the German Bight.  相似文献

Brain-specific angiogenesis inhibitor 2 (BAI2) is a member of adhesion-G protein-coupled receptors (GPCRs). BAI2 is dominantly expressed in the brain and its physiological ligands and functions are still unclear. Adhesion-GPCRs, including BAI2, commonly have a long N-terminal extracellular region (ECR) containing the GPCR proteolysis site (GPS) and the cleavage of the ECR at the GPS domain is suspected to be important for their function. In this study, we analyzed the proteolytic processing of BAI2 and its activation mechanism. Several cleaved C-terminal fragments of BAI2 were identified in mouse hippocampus. We confirmed that mutation in the GPS domain caused inhibition of the proteolysis of BAI2, which indicated the possibility that BAI2 was cleaved at the GPS domain. The association of the ECR putatively cleaved at the GPS domain and the C-terminal seven-transmembrane (7TM) fragment was detected by co-immunoprecipitation. We also found that furin prohormone convertase cleaved BAI2 at another site in the ECR. Additionally, the C-terminal fragment cleaved at the GPS domain specifically activated the nuclear factor of activated T-cells (NFAT) pathway. These results suggest that BAI2 is a functional GPCR regulated by proteolytic processing and activates the NFAT pathway.  相似文献

Places of refuge for ships in distress is a topic before the International Maritime Organization as a result of several recent well-publicized refusals by maritime authorities of coastal states to allow such ships to enter sheltered waters within national jurisdiction. The traditional right of refuge of the crew, ship, and cargo is pitted against threat perceptions held by coastal states resulting in a "not in my backyard" syndrome. Instances of modern state practice seem to restrict the right of refuge to a purely humanitarian dimension. There is a need to reevaluate the right of refuge and to establish a system of places of refuge on the basis of regional cooperation to counter the potential threat of stricken ships that are unable to effect necessary repairs in sheltered areas within national jurisdiction.  相似文献

Expanding upon the model of vessels of exploration as scientific instruments first proposed by Richard Sorrenson, this essay examines the changing nature of the ship as scientific space on expedition vessels during the late nineteenth century. Particular attention is paid to the expedition of H.M.S. Challenger (1872–1876) as a turning point in the design of shipboard spaces that established a place for scientists at sea and gave scientific legitimacy to the new science of oceanography. There was a progressive development in research vessel design from “ship as instrument” to “ship as laboratory” and changing spatial practices aboard these vessels were paralleled by changes in shipboard culture. I suggest that the “ship as laboratory” has now in turn been supplanted by a new model, the “ship as invisible technician”, as oceanographic research vessels deploy remote-sensing equipment and gather data that are no longer analyzed on board.  相似文献

Aim Some nations, and the International Maritime Organization, are moving towards requirements for managing ballast water to reduce the number of alien species transported and released. These and other measures will be most efficient when targeted at ships posing the greatest risks. Here, we analyse world‐wide ship movements and port environmental conditions to explore how these risk components differ across arriving ships. Location Global, with a case study of the Laurentian Great Lakes. Methods We gathered salinity and temperature data for all global shipping ports, and data for all global ship movements during a 12 ‐month period. We applied these data to the Laurentian Great Lakes to determine which global ports may donate new species to the Great Lakes via ship traffic, and which are most environmentally similar to the Great Lakes. Results We show that ships regularly travel to the Great Lakes from all major coastal, and many inland, regions of the world. Most global ports, and thus the species in them, are separated from the Great Lakes by no more than two ship voyages. Combined with a measure of environmental similarity among global ports, we identify ship routes likely to transport species adapted for survival in the Great Lakes and global regions that may be the source of increasing future invasions. Main conclusions The Great Lakes account for a small fraction of global shipping yet are closely connected to all other ports, and the species in them, by the shipping network. Our methods and data allow risks from individual ships to be ranked so that management activities can be targeted at ships most likely to introduce new invaders. Because our data sets are global, they could be applied to ship arrivals at any global port.  相似文献

Although caveolins normally reside in caveolae, they can accumulate on the surface of cytoplasmic lipid droplets (LDs). Here, we first provided support for our model that overaccumulation of caveolins in the endoplasmic reticulum (ER) diverts the proteins to nascent LDs budding from the ER. Next, we found that a mutant H-Ras, present on the cytoplasmic surface of the ER but lacking a hydrophobic peptide domain, did not accumulate on LDs. We used the fact that wild-type caveolin-1 accumulates in LDs after brefeldin A treatment or when linked to an ER retrieval motif to search for mutants defective in LD targeting. The hydrophobic domain, but no specific sequence therein, was required for LD targeting of caveolin-1. Certain Leu insertions blocked LD targeting, independently of hydrophobic domain length, but dependent on their position in the domain. We propose that proper packing of putative hydrophobic helices may be required for LD targeting of caveolin-1.  相似文献

We evaluated the utility of a satellite-linked GPS in obtaining location data from Pacific walruses (Odobenus rosmarus divergens). A unit was attached to one of the tusks of each of three adult male walruses in Bristol Bay, Alaska. The units were designed to relay GPS positions through the Argos Data Collection and Location System. The GPS was only minimally effective in obtaining location data. An average of only 5% of the attempts yielded a position, and only a small number of these were locations at sea. The paucity of successful attempts was probably due to infrequent and brief surfacings of the GPS, the proximity of cliffs to predominant haul-out sites in the study region, and the packing of animals when they were hauled out in herds. Argos was effective in relaying GPS positions in this study, but as GPS technology advances, and its application to marine mammal studies becomes more prevalent, it seems that the greatest challenge to the study of many species will be in data retrieval.  相似文献
