Seeds of Cassia angustifolia Vahl., treated with various concentrations (0, 0.1, 0.15, 0.2 and 0.25 %) of Mancozeb, a broad-spectrum contact fungicide, were sown in field conditions to study the effect of the treatments on lipid peroxidation, proline accumulation and modulation of antioxidant system of seedlings obtained. Significant increase over the control was observed in treated plants for thiobarbituric acid-reactive substances content (up to 207 %), proline content (96 %) and total glutathione content (144 %), whereas the total ascorbate content decreased by 44 %. Increased enzymatic activity was recorded for ascorbate peroxidase (63 %), glutathione reductase (154 %) and superoxide dismutase (109 %), whereas catalase activity decreased by 58 % with 0.25 % Mancozeb treatment. The changes observed were dose-dependent, showing a strong correlation with the level of treatment.  相似文献

An efficient, rapid and reproducible plant regeneration protocol was successfully developed for Cassia angustifolia using nodal explants excised from 14-day-old aseptic seedlings. Of the two cytokinins, 6-benzyladenine (BA) and thidiazuron (TDZ) evaluated as supplements to Murashige and Skoog (MS) medium, TDZ at an optimal concentration of 5.0 μM was effective in inducing multiple shoots. The highest rate of shoot multiplication was achieved on MS medium supplemented with 5.0 μM TDZ and 1.0 μM indole-3-acetic acid (IAA) at pH 5.8. The regenerated shoots when subcultured on hormone free MS medium considerably increased the rate of shoot multiplication and shoot length by end of fourth subculture passage. Rooting was achieved on the isolated shoots using MS medium with 60 μM indole -3- butyric acid (IBA) and 1% activated charcoal for 1 week and subsequently transferring the shootlets to half strength MS liquid media without IBA and activated charcoal. The in vitro raised plantlets with well-developed shoot and roots were successfully established in earthen pots containing garden soil and grown in greenhouse.  相似文献

The two closely related ash species Fraxinus excelsior L. (common ash) and Fraxinus angustifolia Vahl (narrow-leaved ash) have a broad contact zone in France where they hybridize. However, little is known about the local structure of hybrid zone populations and the isolation mechanisms. We assessed the potential effect of floral phenology on the structure of a riparian ash hybrid zone population in central France. The distribution of flowering times was unimodal and lay between the flowering periods of the two species. Using microsatellite markers, we detected isolation by time, which has possibly originated from assortative mating. Multivariate analyses indicated that morphological variation is not distributed at random with respect to flowering times. Spatial autocorrelation analyses showed that temporal and spatial patterns were tightly linked. Interestingly, despite the fact that the population shows isolation by time, neighbourhood size and historical dispersal variance (sigma = 63 m) are similar to those detected in pure stands of F. excelsior where individuals flower rather synchronously and hermaphrodites are not the most frequent sexual type. Trees flowering at intermediate dates, which comprised the majority of the population, produced on average more flowers and fruits. We detected no significant differences in floral parasite infections relative to reproductive timing, although there was a tendency for late flowering trees to suffer from more gall attack. We discuss the impact of temporal variation in fitness traits and their possible role in the maintenance of the hybrid zone.  相似文献

Angiosperm and gymnosperm plants evolved from a common ancestor about 300 million years ago. Apart from morphological and structural differences in embryogenesis and seed origin, a set of embryogenesis-regulating genes and the molecular mechanisms involved in embryo development seem to have been conserved alike in both taxa. Few studies have covered molecular aspects of embryogenesis in the Brazilian pine, the only economically important native conifer in Brazil. Thus eight embryogenesis-regulating genes, viz., ARGONAUTE 1, CUP-SHAPED COTYLEDON 1, WUSCHEL-related WOX, S-LOCUS LECTIN PROTEIN KINASE, SCARECROW-like, VICILIN 7S, LEAFY COTYLEDON 1, and REVERSIBLE GLYCOSYLATED POLYPEPTIDE 1, were analyzed through semi-quantitative RT-PCR during embryo development and germination. All the eight were found to be differentially expressed in the various developmental stages of zygotic embryos, seeds and seedling tissues. To our knowledge, this is the first report on embryogenesis-regulating gene expression in members of the Araucariaceae family, as well as in plants with recalcitrant seeds.  相似文献

Candidate genes are sequenced genes of known biological action involved in the development or physiology of a trait. Twenty-one putative candidate genes were designed after an exhaustive search in the public databases along with an elaborate literature survey for candidate gene products and/or regulatory sequences associated with enhanced drought resistance. The downloaded sequences were then used to design primers considering the flanking sequences as well. Polymerase chain reaction (PCR) performed on 10 diverse cultivars that involvedJaponica, Indica and local accessions, revealed 12 polymorphic candidate genes. Seven polymorphic candidate genes were then utilized to genotype 148 individuals of CT9993 × IR62266 doubled haploid (DH) mapping population. The segregation data were tested for deviation from the expected Mendelian ratio (1:1) using a Chi-square test (<1%). Based on this, four candidate genes were assessed to be significant and the remaining three, as non-significant. All the significant candidate genes were biased towards CT9993, the female parent in the DH mapping population. Single-marker analysis strongly associated (<1%) them to different traits under both well-watered and low-moisture stress conditions. Two candidate genes,EXP15 andEXP13, were found to be associated with root number and silicon content in the stem respectively, under both well-watered and low-moisture stress conditions  相似文献

Antigenotoxic properties and the possible mechanisms of water extracts from Cassia tora L. (WECT) treated with different degrees of roasting (unroasted and roasted at 150 and 250 degrees C) were evaluated by the Ames Salmonella/microsome test and the Comet assay. Results indicated that WECT, especially unroasted C. tora (WEUCT), markedly suppressed the mutagenicity of 2-amino-6-methyldipyrido(1,2-a:3':2'-d)imidazole (Glu-P-1) and 3-amino-1,4-dimethyl-5H-pyrido(4,3-b)indole (Trp-P-1). In the Comet assay performed on human lymphocytes, WECT exhibited significant protective effect on Trp-P-1-mediated DNA damage followed the order of unroasted (55%) > roasted at 150 degrees C (42% ) > roasted at 250 degrees C (29%). Pre-treatment of the lymphocytes with WEUCT resulted in 30% repression of DNA damage. However, no significant effect on excision-repair system was found during DNA damage expression time in post-treatment scheme (p>0.05). WEUCT showed 84% scavenging effect on oxygen free radicals generated in the activation process of mutagen detected by electron paramagentic resonance system. Two possible mechanisms were considered: (1) neutralization the reactive intermediate of Trp-P-1; and (2) protecting cells directly as an antioxidant that scavenge the oxygen radicals from the activation process of mutagen. The individual anthraquinone content in extracts of C. tora was measured by HPLC. Three anthraquinones, chrysophanol, emodin and rhein, have been detected under experimental conditions. The anthraquinone content decreased with increased roasting temperature. Each of these anthraquinones demonstrated significant antigenotoxicity against Trp-P-1 in the Comet assay. In conclusion, our data suggest that the decrease in antigenotoxic potency of roasted C. tora was related to the reduction in their anthraquinones.  相似文献

Four popular mulberry cultivars (Morus indica L. cvs.V-1, MR-2, S-36 and K-2) were assessed for drought tolerance with an integration of selective approaches. The potted plants were subjected to two watering treatments for 75 days: control pots were watered up to 100% field capacity (FC) and stressed pots were maintained at 25–30% FC. Net photosynthetic rate (Pn), stomatal conductance (gs), transpiration rate (E) and instantaneous water use efficiency (WUEi) were the key parameters to assess photosynthetic gas exchange performance. Drought caused marked down-regulation in leaf gas exchange in all cultivars (cvs) except V-1 which maintained better Pn, gs, E and higher WUEi under severe water deficit. All the four cvs also showed differential antioxidative responses under water stress. Higher concentrations of carotenoids, ascorbic acid, glutathione, α-tocopherol and proline were observed in the leaf extracts of V-1, while minimum accumulation of those metabolites was recorded with K-2 and S-36. An endogenous loss of α-tocopherol and higher lipid peroxidation were encountered in K-2, S-36 and MR-2, whereas V-1 showed minimum lipid peroxidation under water deficit regimes. Comparative morpho-anatomical analysis revealed a well-developed root system and a better anatomical architecture in V-1 which could further contribute tolerance during drought stress.  相似文献

A silvi-herbal trial was conducted in the hot arid region of India to study the performance of a shrub (Cassia angustifolia) in combination with different tree species. The study area was frequented by frost. Leaf yield of C. angustifolia under different treatments was estimated. It was found that the shrubs produced a significantly higher yield of leaves in the vicinity of the tree species as compared to the shrubs at a far distance from the trees. This was perhaps because of more protection of crops near the canopy of the plants during frosts. The yield was not much affected by the variation of tree species indicating that the effect of tree-shrub combination was not profound. C. angustifolia as inter-crop provides support to the farming system by way of conferring stability and generating assured income.  相似文献

Proteinaceous extract obtained from Cassia occidentalis seeds with purification fold of 3.91 and 82.7% of bovine trypsin inhibitory activity was assessed at different concentrations (25, 50, 100, 200, 400 and 800 μg/ml) against Spodoptera litura. Assay of larval feeding suggested proteinaceous extract to be toxic as prepupal (80.16%) and pupal mortalities (100%) along with growth deterrent effect with only 16.71% pupation was observed at 800 μg/ml. Fifty per cent mortality (LC50) was observed at 132.91 μg/ml. Also the inhibitor affected fecundity, longevity and percentage of egg hatching. Nutritional indices were adversely affected as both efficiency of conversion of ingested and digested food decreased while approximate digestibility and metabolic cost increased. In vitro studies on proteolytic enzymes of S. litura revealed inhibition of trypsin and chymotrypsin in lumen and faecal matter at all tested concentrations. Also proteinaceous extract inhibited mycelial growth in Fusarium oxysporum, Alternaria brassicicola and Alternaria alternata at 100 μg/ml.  相似文献

Due to severe water resource shortage, genetics of and breeding for DT (drought tolerance) in rice (Oryza sativa L.) have become one of the hot research topics. Identification of grain yield QTLs (quantitative trait loci) directly related to the DT trait of rice can provide useful information for breeding new drought‐resistant and water‐saving rice varieties via marker‐assisted selection. A population of 105 advanced BILs (backcross introgression lines) derived from a cross between Zhenshan97B and IRAT109 in Zhenshan97B background were grown under drought stress in a field experiment and phenotypic traits were investigated. The results showed that in the target interval of RM273‐RM255 on chromosome 4, three main‐effect QTLs related to panicle length, panicle number, and spikelet number per panicle were identified (LOD [logarithm of the odds] > 2.0). The panicle length‐related QTL had two loci located in the neighboring intervals of RM17308‐RM17305 and RM17349‐RM17190, which explained 18.80% and 20.42%, respectively, of the phenotypic variation, while the panicle number‐related QTL was identified in the interval of RM1354‐RM17308, explaining 11.47% of the phenotypic variation. As far as the spikelet number per panicle‐related QTL was concerned, it was found to be located in the interval of RM17308‐RM17305, which explained 28.08% of the phenotypic variation. Using the online Plant‐GE query system, a total of 13 matched ESTs (expressed sequence tags) were found in the target region, and of the 13 ESTs, 12 had corresponding predicted genes. For instance, the two ESTs CB096766 and CA765747 were corresponded to the same predicted gene LOC_Os04g46370, while the other four ESTs, CA754286, CB000011, CX056247, and CX056240, were corresponded to the same predicted gene LOC_Os04g46390.  相似文献

Most aphids develop a cyclic parthenogenesis life-cycle. After several generations of viviparous parthenogenetic females, it follows a single annual generation of sexual individuals, usually in autumn, that mate and lay the sexual eggs. Shortening of photoperiod at the end of the summer is a key factor inducing the sexual response. With the survey here reported we aimed at identifying a collection of candidate genes to participate at some point in the cascade of events that lead to the sexual phenotypes. Following a suppression subtractive hybridization methodology (SSH) on the model aphid Acyrthosiphon pisum, we built and characterised two reciprocal cDNA libraries (SDU and SDD) enriched respectively in genes up-regulated or down-regulated by short photoperiod conditions that lead to the sexual response in this aphid species. A total of 557 ESTs were obtained altogether representing 223 non-overlapping contigs. 29% of these were new sequences not present in previous aphid EST libraries. BLAST searches allowed putative identification of about 54% of the contigs present in both libraries. Relative quantification of expression through real-time quantitative PCR demonstrated the differential expression in relation with the photoperiod of 6 genes (3 up-regulated and 3 down-regulated by shortening the day length). Among these, expression of a tubulin gene, two cuticular proteins and a yet unidentified sequence along the day-night cycle was further investigated. Implications for current studies on gene regulation of the dichotomy sex vs. parthenogenesis in aphids are discussed.  相似文献

Results of the spatial distribution of fine roots are reported from a Norway spruce (Picea abies (L.) Karst.) in SW Sweden stand subjected to drought (D) and ammonium-sulphate application (NS). The sampling was carried out by excavating monoliths in segments of 0.5 × 0.5 × 0.1 m to a depth of one meter. Root data also included in the study were obtained by excavating whole trees and soil coring.The data suggest a fairly deep distribution pattern of fine roots (< 1 mm in diameter) in the study area compared with other forest sites in SW Sweden. The weight fraction of living fine roots in the LFH-horizon amounted to 53, 36 and 55% of the total fine-root biomass and 12, 30 and 32% of the total fine-root necromass (dead fine roots) in the control, D and NS-treatment areas respectively. Drought seemed to result in a redistribution of fine roots to deeper mineral soil horizons. Ammonium sulphate application led to the reverse, viz, a concentration of fine roots to the LFH-horizon. A significantly smaller fine-root necromass was indicated in the LFH-horizon of the control areas compared with both the D and NS-treatment areas, suggesting a high mortality of fine roots in these areas. A heavy dry matter fraction accumulates in roots > 1 mm in diameter and in stumps. These root fraction, were frequently found between the trees, although the stump constitutes an important fraction in terms of dry weight.  相似文献

Abstract

The carotenoid content of edible plant storage organs is a key nutritional and quality trait. Although the structural genes that encode the biosynthetic enzymes are well characterised, much less is known about the factors that determine overall storage organ content. In this study, genome-wide QTL mapping, in concert with an efficient ‘genetical genomics’ analysis using bulked samples, has been employed to investigate the genetic architecture of potato tuber carotenoid content. Two diploid populations of Solanum tuberosum Group Phureja were genotyped (AFLP, SSR and DArT markers) and analysed for their tuber carotenoid content over two growing seasons. Common to both populations were QTL that explained relatively small proportions of the variation in constituent carotenoids and a major QTL on chromosome 3 explaining up to 71 % of the variation in carotenoid content. In one of the populations (01H15), a second major carotenoid QTL was identified on chromosome 9, explaining up to 20 % of the phenotypic variation. Whereas the major chromosome 3 QTL was likely to be due to an allele of a gene encoding β-carotene hydroxylase, no known carotenoid biosynthetic genes are located in the vicinity of the chromosome 9 QTL. A unique expression profiling strategy using phenotypically distinct bulks comprised individuals with similar carotenoid content provided further support for the QTL mapping to chromosome 9. This study shows the potential of using the potato genome sequence to link genetic maps to data arising from eQTL approaches to enhance the discovery of candidate genes underlying QTLs.  相似文献
