Algal growth on organic compounds as nitrogen sources   总被引:19,自引:0,他引:19  
Two experimental series were run to evaluate the potential of algal development on dissolved organic nitrogen (DON) compounds as the sole source of nitrogen (N) nutrition. Monocultures of several common Lake Kinneret algae (Pediastrum duplex, Synechococcus sp., Microcystis aeruginosa, Aphanizomenon ovalisporum and Cyclotella sp.) were incubated for 3 weeks in the laboratory with different inorganic (NH4+, NO3-) or organic (hypoxanthine, urea, guanine, ornithine, glucosamine, lysine) nitrogen sources. Even though the cultures were not axenic, marked differences were observed in algal growth response. Pediastrum, Cyclotella and Aphanizomenon grew well on most N sources, and cyanobacterial growth and yield were consistently greatest when the urea was the only N source. We also followed algal growth and eventual species dominance in batch samples of GF/F-filtered lake water, supplemented with orthophosphate and different inorganic or organic N compounds and inoculated with concentrated lake phytoplankton. Although no clear impact on phytoplankton growth (as chlorophyll concentration) was observed, in seven out of 11 experiments we could discern changes in the algal species that became dominant in flasks with different organic and inorganic N sources. Our results are consistent with the proposition that components of the DON pool are not only an important potential, direct or indirect N source for phytoplankton, but also that different algal species can exploit these sources with varying capabilities so that different N substrates may selectively stimulate the development of dominant algal species.   相似文献

Growth of rice callus tissue is discouraged when methionineis excluded from CMAA medium. While determining the methionineelimination effect, the amino acid interrelationships amongmethionine, lysine, threonine and isoleucine in the nutritionof the callus tissue were found. Poor growth, found in cultureson methionine deficient media was seen only when the media containedboth threonine and lysine, simultaneously. The substitutionof homoserine for methionine was also observed. Determination of free amino acid composition in tissues revealedthat free methionine was barely detectable in tissues grownwith sufficient amounts of threonine and lysine. When the concentrationof either threonine or lysine was reduced, the free methioninecontent of the tissue increased. When the methionine deficientmedium was supplemented with homoserine, the free methioninein the tissue increased, although the tissue retained a considerableamount of free threonine and lysine. Cultivation of tissue onan isoleucine deficient medium resulted in a significant decreasein free threonine content. These experimental results suggest that the biosynthetic pathwayto methionine is cooperatively inhibited by threonine and lysine,and that threonine decomposition is inhibited by its end productisoleucine. Rice callus was initiated from cultured anthers and was maintained on medium containing ammonium as sole nitrogen source and supplemented with either a buffer or an organic acid. Plant regeneration also was obtained on this medium, but the morphogenetic capacity of the callus culture declined as rapidly as on a medium containing a mixture of ammonium and nitrate. The results of the studies suggest that organic acids provide an additional function beyond that of buffering the medium during growth on ammonium.  相似文献

Epiphytic bromeliads have no contact with the pedosphere, so they need to draw their nutrients from the atmosphere as well as from the host tree and animal debris. Terrestrial bromeliads, like Ananas comosus, generally depend on the soil as their main nutrient source. The aim of this study was to investigate and compare some aspects of the nitrogen metabolism of two bromeliads with different growth habits: Ananas comosus, a terrestrial bromeliad, and Vriesea gigantea, an epiphytic tank bromeliad. Nitrogen-starved plants were grown in vitro for 3, 7, 15, 30, and 60 days, either with 5 mmol L−1 ammonium [(NH4)2SO4] or urea as the sole nitrogen source. When NH4+ was supplied to the plants, it stimulated a faster increase of chlorophyll content in A. comosus than in V. gigantea. In the presence of urea, after 15 days of the plants in culture, there was a significant increase in tissue free-NH4+ and total amino acids for V. gigantea only. V. gigantea presented a higher level of total free amino acids than A. comosus when nitrogen was supplied to the plants. Asparagine was the main amino acid accumulated in both bromeliads when plants were transferred to the medium with nitrogen. When the ratio of the main individual free amino acids between the bromeliads grown in NH4+ and urea was compared, values such as 7.2 for asparagine, 5.3 for glutamate, and 1.8 for aspartate in A. comosus, and values such as 2.3 for asparagine, 1.1 for glutamate and 0.7 for aspartate in V. gigantea were observed, demonstrating that the last is more efficient in assimilating urea. The results prompted us to support the idea that V. gigantea, an epiphytic tank bromeliad, is better adapted to absorb and assimilate organic nitrogen, such as urea, while A. comosus, a terrestrial plant, is better adapted to inorganic nitrogen forms, such as ammonium. Growth rate, maximum dry weight yield, and carbohydrate utilization were measured with pith callus derived from Nicotiana tabacum L. var. Wisconsin No. 38. Maximum tissue dry weights increased as carbohydrate (either glucose or sucrose) in the medium was increased. The time at which maximum growth was obtained coincided with the time at which carbohydrate was exhausted from the medium. The addition of carbohydrate to the medium before the end of log phase growth supported that amount of additional growth which would have been obtained if all of the carbohydrate had been added to the medium prior to planting the tissue. Maximum obtainable dry weights at logarithmic growth rates greater than 0.16 doubling per day depended on the amount of carbohydrate in the medium and not on a particular hormonal regime.  相似文献

A procedure was developed which demonstrates the expression of differential resistance in pine callus tissues to the fungal pathogen Phytophthora cinnamomi Rands. Callus tissues were maintained on a modified Murashige and Skoog medium with 10–5M 2,4-D and inoculated with hyphae of P. cinnamomi at 26°C in the dark. The number of intracellular hyphae was used as an index of resistance. The effect of nitrogenous sources like ammonium sulphate (AS), prilled urea (U), urea super granule (USG) and farm yard manure (FYM) was studied on the fresh biomass (FB) and acetylene reduction activity (ARA) ofAzolla pinnata, R. Brown (Bangkok isolate), grown as a dual crop with rice, and rice yield in three successive seasons. Irrespective of the N-sources and seasons, the FB and ARA of Azolla were observed to be maximum on 14th day after Azolla inoculation (DAI). The different N-sources had significant effect on the ARA and to a lesser extent on the FB of Azolla. The treatment without fertilizer-N (control) exhibited highest ARA, FB and total N-content of Azolla. These were inhibited to a lesser extent by USG and FYM, though used at higher rates of 75 kg N ha−1 and 90 kgN ha−1 respectively, compared to that by AS and U, used at lower rates of 45kg N ha−1 each.  相似文献

Tuber callus by subculture ofDioscorea deltoidea Wall, was grown with different sources of nitrogen on N-free Murashige and Skoog’s basal media supplemented with a high level of kinetin (1, 3, 6, 9 and 12 ml 1-1) and low concentration of auxin (0.01 mg 1-1 2,4-D) for shoot bud differentiation. Shoot and root differentiation was observed only in the case of ammonium nitrate. Other sources of nitrogen failed to produce shoot bud differentiation except in the case of ammonium citrate where tissues were slight green in colour and were recognizable as pro-embryo.  相似文献

研究了8种有机碳源和4种氮源对三角褐指藻(Phaeodactylum tricornutum)生长的影响.结果表明,三角褐指藻具有兼养生长的能力,碳浓度为5mmol/L和50mmol/L时,葡萄糖、果糖、乙酸钠和甘油对其生长有明显的促进作用,乳酸钠和乙醇抑制藻细胞生长,半乳糖和柠檬酸钠对其生长随有机碳浓度而异.甘油、葡萄糖和乙酸钠的适宜浓度范围分别为5 -800mmol/L、100 -400mmol/L和50 -300mmol/L.培养液中分别加入50mmol/L甘油、400mmol/L葡萄糖和150mmol/L乙酸钠,培养第14天,三角褐指藻的最大生物量分别为对照的1.45倍、1.25倍和1.11倍.甘油兼养生长的最优氮源是尿素,适宜浓度范围为0.88-8.8mmol/L.当尿素浓度为4.4mmol/L时,最高生物量可达1.31g/L.  相似文献

Summary Agrochemical resistant mutants of nitrogen fixing cyanobacteriumTolypothrix tenuis were isolated after MNNG mutagenesis. The mutants exhibited higher nitrogenase activity and released more quantities of extracellular nitrogenous, substances such as ammonia, indole acetic acid like substances and amino acids when compared to the parent. They also increased the available nitrogen status of the soil in rice culture. Significant increase in the growth and yield upon inoculation of these mutants into rice culture was observed in comparison with chemical nitrogen fertilizer urea, as well as the parent strain treament.  相似文献

Summary Growth of rice plants in a nitrogen-free medium was enhanced by inoculation with a nitrogenase-derepressed mutant (strain SA-1) of a cyanobacterium,Anabaena variabilis which excretes NH4 + into the medium. The growth rates of tobacco callus tissues on media containing 10−6 to 10 μm 6-(γ,γ-dimethylallylamino)purine (2iP) were measured. At concentrations of 10−4 μm and above growth rates were exponential and dependent on cytokinin concentration. At 2iP concentrations of 10−4 to 0.33 μm, the exponential rate was maintained for 4 to 5 doublings of fresh and dry weight. After this period a linear phase, resulting in approximately 1 doubling of weight, occurred. The growth of tissues on media containing higher than 0.33 μm 2iP was exponential for only about 15 days. At the end of this time, and well before they achieved half their final weight, they exhibited growth which was less rapid than logarithmic but more rapid than linear. Comparisons with zeatin, 6-benzylaminopurine and kinetin indicated that, although the maximum growth rates obtained with relatively high concentrations (0.1-1 μm) were similar, the naturally occurring cytokinins, 2iP and zeatin, promoted faster rates at lower concentrations (10−3-10−2 μm) than did 6-benzylaminopurine and kinetin.  相似文献
