From a wild type strain Challis of the group H streptococcus, greening (Challis α) and β-hemolytic (Challis β) colonies were isolated on horse blood agar. Both colonies formed greening on sheep blood agar, and no significant differences were found in their biological, serological and chemical analyses. They, however, showed clear differences on the transformability. Transformability, the producibility of competence-provoking factor (CPF) and competency which have been reported on the Challis strain were all found in Challis β strain. On the other hand, Challis α strain did not produce antibiotic-resistant transformants with the addition of CPF, and could not produce CPF even when the cells were cultured under various conditions of incubation or treated with lysozyme or detergents. The transformabilities of antibiotic-resistant mutants obtained from the Challis β strain were lower than those of the original Challis β strain, as pointed out by other investigators, while the Challis α strain became transformable on antibiotic resistance only when it acquired streptomycin resistance. In the Challis β strain and the antibiotic-resistant mutants of Challis α strain, the separate markers of streptomycin, penicillin, tetracycline, mitomycin C, as well as the combinations of these markers were found to be transformed at the highest rate in the strains having transformation of streptomycin resistance. The findings are discussed with respect to incorporation of deoxyribonucleic acid into recipient cells and to the reports of other workers.  相似文献

Galactose-negative mutants of the group H Streptococcus strain Challis were obtained by treatment with nitrosoguanidine. Enzyme assays of extracts of these mutants revealed that 12 of the mutants were lacking one of the enzymes of the Leloir pathway. Thus, the Leloir pathway is the major means of galactose metabolism in strain Challis. In addition, uridyl diphosphate galactose pyrophosphorylase, a permease function, and at least one other function are required for the utilization of galactose. The enzymes of the Leloir pathway are induced by galactose and fucose; no compounds which act as repressors of these enzymes have been found, although the system appears to be sensitive to catabolite repression. Transformation was used to map the mutants. The genes for galactose-1-phosphate uridyl transferase and glucose-4-epimerase appear to be closely linked. Within the transferase gene, six mutations have been mapped. The permease function and the undetermined functions are not linked to the Leloir pathway.  相似文献

The binding of streptococcal competence factor (CF) was found to be specific for a strain of streptococcus which was capable of undergoing competence induction. Three other streptococcal strains which could not be induced to competence, did not bind CF. CF binding was independent of time, temperature, age of the culture, and type of growth media employed. Several observations indicated that the receptor sites for CF are located in the bacterial membrane: (i) the retention of CF by spheroplasts, (ii) the binding of CF by isolated membrane fractions, and (iii), the degradation of CF binding capacity of membranes by different chemicals and enzymes.  相似文献

Acidified phenol extracts prepared from competent cultures of a group H Streptococcus strain Wicky made competent with competence factor derived from cultures of another group H Streptococcus, strain Challis, showed a difference in polyacrylamide-gel protein patterns when compared to extracts prepared from noncompetent cultures of strain Wicky. The prominent single protein band difference did not appear when Wicky cells were simultaneously treated with competence factor and chloramphenicol, an inhibitor of the development of competence. Chloramphenicol had no effect on transformation nor the appearance of the "new" protein band when added to fully competent cells. This new protein, which is associated with the appearance of competence, seems to be synthesized as a result of induction by competence factor; its exact role, however, is as yet unknown.  相似文献

A chemically defined medium is desirable for nutritional studies and is frequently necessary for biochemical investigations. Several defined media are available for use with tissue and cell cultures from dicotyledonous plants. A fully defined medium has now been developed for cell suspension cultures from sugarcane. Prior to this, the only medium successfully used for cell cultures of monocotyledonous plants was a modification of Straus' synthetic medium (used to grow cell suspensions of corn). Cell suspension cultures from sugarcane stalk parenchyma, originally established in complex media containing coconut milk or yeast extract, can be grown in this synthetic medium, which consists of inorganic salts, vitamins, sucrose, 2.4-dicliloropheuoxyacetic acid, and a mixture of 13 amino acids. The most important of the amino acids are arginine aspartic acid, and glutamic acid. This simplified medium wilt aid in the investigation of the unusual and important role of arginine in sugar-cane growth and metabolism.  相似文献

A procedure for the isolation and purification of competence factor produced in a defined medium by group H streptococci, strain Challis-6, is presented. Partial characterization and chemical analysis of the product are described. The procedure yields competence factor of high purity, as shown by homogeneity in electrofocusing, by electrophoresis in sodium dodecyl sulfate polyacrylamide gels, and by chemical analysis. The data indicate that competence factor is a small, dialyzable, highly basic compound. It is free from lipids, phosphorus, and carbohydrates, and is colorless and thermoresistant. Its biological activity is destroyed by trypsin but not by deoxyribonuclease, ribonuclease, lipase, or lysozyme. Its high isoelectric point of above pH 11.0 suggests that competence factor may be a protamine or a polymer of basic amino acids. The possibility that a polyamine may be an integral part of the polypeptide molecule has not been excluded.  相似文献

Details in the fermentation of oxytetracycline in a synthetic medium with Streptomyces rimosus have been presented. In these studies, an organic nitrogen source was shown to be essential for the production of significant amounts of antibiotic activity. Of the amino acids tested, aspartic acid, proline, threonine, valine, and beta-alanine were utilized well for both growth and antibiotic production. Markedly different fermentation patterns were observed with aspartic acid and beta-alanine. Glycerol and glucose supported antibiotic yields superior to those found with other carbohydrates tested. Short chain organic acids were not effectively utilized for growth in the absence of a readily fermentable carbohydrate.  相似文献

Competence factor (CF) was purified 600-fold from culture filtrates of competent group H streptococcus, strain Challis, by fractionation on a carboxymethyl-Sephadex column. The preparation did not contain orcinol- or diphenylamine-positive material, and CF activity was destroyed by trypsin. An examination of its chemical composition revealed 10 amino acids, muramic acid, glucosamine, and glucose.  相似文献

A protein-free chemically defined medium for cultivation of pathogenic Leptospira was developed. The medium permitted continued serial subculturing of 9 serogroups (52 strains) of the 12 serogroups (61 strains) tested. Growth was initiated from small inocula, and the growth rate and maximal cell yields were similar to those on serum-containing media. The nutritional requirements of serogroups L. canicola, L. pomona, and L. grippotyphosa were studied in a basal medium composed of inorganic salts, a fatty acid, vitamin B(12), and thiamine. All strains tested utilized ammonium chloride as the sole nitrogen source. A fatty acid, vitamin B(12), and ferrous ions were essential. Growth was stimulated by thiamine, potassium, and calcium ions.  相似文献

A nutritional study was made of five strains of Bacillus coagulans obtained from various culture collections. These five strains were descendants of two original isolates; three had been derived from one parent culture in years past and the other two were transfers from another parent culture. Therefore, the five cultures should have represented two distinct groups of genetically identical cultures. Three of the strains obtained from one culture collection had become methyl red-negative and sorbitol-negative and had gained abilities to hydrolyze gelatin and ferment arabinose. Nutritional requirements of the five cultures, determined at 37, 45, and 55 C, differed considerably among strains; however, thiamine and biotin were required by all cultures at all temperatures. Aspartic acid was stimulatory at 37 C and was required at 45 C; folic acid, basic amino acids, and certain other nutrilites were required at 55 C. Adenine supplementation was necessary for two strains at 55 C to prevent autolysis; this phenomenon is discussed. The response of these organisms to both serine and the basic amino acids at the three growth temperatures seems especially significant. The media devised for the growth of the five strains of B. coagulans used in this study permit excellent growth at three incubation temperatures.  相似文献

Defined media that promote the initiation and undifferentiated growth of callus derived from stem explants of four cultivars of cassava, Manihot esculenta Crantz, are described. Growth rates and yields of cassava callus after 4 weeks of culture are shown to be comparable to those of callus of Nicotiana tabacum L. cv. Wisconsin No. 38. Nitrogen sources of ammonium nitrate or of ammonium chloride plus succinate supported growth of all four cultivars. Sucrose was superior to glucose as a carbon source. The cassava cultivars differed in their response to increasing concentrations of sucrose between 0.5% (w/v) and 3%, two of them increasing in dry matter with increasing sucrose concentrations of up to 3%. When cultured in the light on defined media that contained higher ratios of cytokinin to auxin, callus of the latter two cultivars turned green. Roots but not shoots differentiated from the callus of all cultivars. The influence of hormone concentrations, sucrose level, and nitrogen source on greening and root formation is summarized.  相似文献

The effects of competence induction by competence factor (CF) on macromolecular synthesis in group H streptococcus strain Wicky were investigated. CF preparations (culture filtrates from competent group H streptococcus strain Challis) were either heated or partially purified to remove a bacteriocin. These preparations did not inhibit growth, although they induced high levels of competence in strain Wicky. The action of the CF preparations did not affect the overall rates of deoxyribonucleic acid and protein synthesis, but caused a reduction in the rates of ribonucleic acid (RNA) and peptidoglycan synthesis. When competence induction by CF was prevented, no alterations in RNA or peptidoglycan synthesis were observed, indicating that these changes are in fact related to the development of competence.  相似文献

The effects of various medium components on exoprotease production by Burkholderia pseudomallei were studied in order to understand the production of virulence factors by this pathogenic organism. FeSO(inf4), NaCl, and MgSO(inf4) do not significantly affect exoprotease activity, and CaCl(inf2) has only a slight influence. Conversely, ZnCl(inf2) plays a fundamental role since it drastically increases exoprotease production.  相似文献

A simple chemically defined medium was devised for exoprotein production by Bacillus brevis No. 47. About 2 mg/ml of proteins was produced in the synthetic medium containing 4% glucose and 1% ammonium sulfate. An essential component of fermentation medium was Ca salt which is required by this organism for assimilating glucose.

Studies on the effects of various medium components on protein production revealed that the conditions appropriate for growth are also suitable for protein accumulation. Some compounds, especially inhibitors of cell wall synthesis and certain detergents, were found to enhance protein production.  相似文献

Glucosylated deoxyribonucleic acid (DNA) from phages T4 and T6 competes poorly with homologous DNA causing only a slight decrease of transformation in Group H Streptococcus strain Challis. Other types of heterologous DNAs (Micrococcus luteus, Clostridium perfringens, Escherichia coli, calf thymus and non-glucosylated phage T6 DNA), in contrast to glucosylated T4 and T6 DNAs, compete with transforming DNA to the normal, high extent. These results indicate that as in transformation of Bacillus subtilis, the presence of glucose attached to 5-hydroxymethylcytosine in phage T6 DNA considerably decreases the interaction of such DNA with competent cells of the Challis strain. It also indicates that the guanine plus cytosine content of DNA is not decisive in determining its interaction with competent cells.  相似文献

STH(1), a streptocin elaborated by group H streptococcus strain Challis, is lethal for group H streptococcus strain Wicky and is produced maximally during the exponential growth phase of liquid medium cultures. Crude streptocin preparations are resistant to oxidation and display a biphasic pH stability (stability being maximal at pH 5.0 and 10.0). Survivor studies indicate that streptocin-mediated killing is a "one-hit" phenomenon and proceeds rapidly. The streptocin has been purified 50-fold with (NH(4))(2)SO(4) fractionation and Sephadex G100 chromatography and appears to exist in equilibrium between two molecular weight forms. Low ionic strength and neutral pH buffers favor the isolation of the 110,000 molecular weight form, whereas high ionic strength and alkaline pH conditions facilitate isolation of the 28,000 to 30,000 molecular weight form. These findings suggest an association-dissociation relationship between macromolecules of 28,000 to 30,000 molecular weight. Purified STH(1) has no "competence factor" (CF) activity. In addition, CF has no STH(1) activity and displays no inhibitory effect on exponential-phase Wicky cultures as determined by absorbancy measurements. It appears, therefore, that initiation of the competent state for transformation in strain Wicky is not necessarily accompanied by gross alterations in cell growth.  相似文献

A quasi-defined medium that supports the growth of Streptococcus agalactiae as pigmented colonies has been developed. The medium contains starch, a peptic digest of albumin, amino acids, nucleosides, vitamins, and salts. The presence of free cysteine, which could be replaced with other sulphur-containing compounds and to a lesser degree by reducing agents, was required for pigment formation.  相似文献
