A virus causing a severe disease of tomatoes in Argentina was identified as a strain of eggplant mosaic virus (EMV). It resembles the type, Abelia latent and various Andean potato latent strains of EMV in its host range and its transmissibility at allow rate by Epitrix sp. It differs from these strains, however, serologically and in some of its cytopathic effects. In serological agar gel double diffusion tests it proved to be closely related to the tomato white necrosis isolate of EMV studied by Barradas (1983) in Brazil.  相似文献

1991年从沈阳地区茄子花叶病叶片中分离到了SY-Is分离物。经汁液摩擦接种7个科16种植物,这一分离物可浸染4个科8种植物,但不侵染黄瓜、两葫芦和蚕豆,也不能经桃蚜和萝卜蚜传播。该分离物体外抗性的致死温度(TIP)为90-95℃;稀释限点(DEP)为10 ̄(-6)-10 ̄(-7);体外保毒期(L)为一个月以上。光学显微镜下可见晶状胞质内含体。叶滴法电镜观察可见大小为±300×18nm的杆状病毒粒子。该病病叶汁液与烟草花叶病毒(TMV)普通株抗血清呈明显阳性反应。用TMV的一对引物进行多聚酶链反应(PCR),可扩增出一特异性核酸片段。根据上述实验结果,我们认为引起沈阳地区茄子花叶病的毒原种类为TMV,是茄子花叶病的新毒原。  相似文献

The virus causing mosaic of muskmelon in the Punjab is transmitted through seed, sap and aphids but not through beetle, whitefly, fungi or contact. It systemically infected Nicotiana tabacum (var. “White Burley” and CTRI-Special), N. glutinosa, N. rustica and Capsicum annuum besides various cucurbit hosts when inoculated mechanically. The virus gave positive reaction with the antiserum of cucumber mosaic virus and the particles are spherical in shape. The virus has been identified as a distinct strain of cucumber mosaic virus and is designated as muskmelon strain of cucumber mosaic virus (CMV-mst.).  相似文献

Calanthe mild mosaic potyvirus (CalMMV), a previously undescribed virus found in several locations in Japan, causes mild leaf mosaic and flower colour breaking of Calanthe plants. CalMMV was mechanically transmitted only to Calanthe sp., Phalaenopsis sp. and Tetragonia expansa of 50 plant species tested and was transmitted by the aphid Myzus persicae in a nonpersistent manner. The virus has flexuotis particles about 764 nm long and induced the formation of intracellular cytoplasmic cylindrical inclusions. The virus particles contain a single poly-peptide of 32.0 kDa and a single RNA of mol. weight 3.1 × 106. As determined by immuno-electron microscopy, CalMMV is distantly related to the Japanese isolate of dendrobium mosaic potyvirus (DeMV-J), but it showed no serological relationship to any of seven other potyviruses. The sequence of the 3'-terminal 1306 nucleo-tides of the viral genome was determined. The coat protein (CP) coding sequence is predicted to be 804 nucleotides in length, encoding a protein of 268 amino acids with a calculated mol. weight of 30 389. The 3' noncoding region is 169 nucleotides long and is followed by a polyadenylate tract. The amino acid sequence of the CP of CalMMV was 73% homologous to that of DeMV-J, but less than 66% to other potyviruses.  相似文献

Cucumber green mottle mosaic virus may spread in bottlegourd under field conditions through soil contaminated with infected plant debris followed by contact. No seed transmission was noticed in bottlegourd (Lagenaria siceraria) or vegetable marrow (Cucurbita pepo) although pollen grains and cotyledons from infected bottlegourd flowers or seeds, respectively, contained negliginle amounts of virus. Cucumber leaf beerles (Raphidopalpa fevicollis) are probable vectors since their regurgitated fluid and excretes contained infective virus particles. No vector fungi were found in the soil around infected bottlegourd plants.  相似文献

Schizaphis graminum (Rondani) is proved to be an additional vector of maize mosaic virus (MMV). The pH range for the infectivity of the virus in extracted juice is found to be from 4.4 to 9.0, the optimum being 5.6 to 7.2. Effect of certain chemicals on the virusin vitro has also been studied. Cross protection between MMV and Sugar-cane mosaic virus (SMV) indicated positive results. It has been concluded on the basis of similar physical properties, tolerance towards certain chemicals, host range, symptomatology, aphid vectors and positive immunological tests, that MMV and SMV are related viruses.  相似文献

Cowpea mosaic virus was one of the viruses isolated from Crotalaria juncea showing mosaic, distortion and puckering symptoms. The virus induced local necrotic lesions in Canavalia ensiformis, Cassia occidentalis, Nicotiana debneyi, N. occidentalis, N. repanda and N. sylvestris and some cultivars of Phaseolus vulgaris 4–6 days after inoculation. It induced local chlorotic spots, which later became necrotic, as well as systemic chlorotic spots, vein chlorosis, distortionand puckering in Chenopodium amaranticolor. Sap from systemically infected cowpea cv. ‘C20-55’was infective after dilution to 10?5 but not 10?6, after 10 min at 65°C but not 70°C, or after 4 days, but not 5 days, at a room temperature of 15–25°C. Infectious virus was recovered from fresh reproductive tissues of infected cowpea cvs ‘C20-55’and ‘Ife Brown’ plants but not after dehydration.  相似文献

从浙江省5个地区采集表现花叶症状的甘蔗病叶,用马铃薯Y病毒科简并引物做PCR扩增及测序鉴定.序列分析表明,5个甘蔗样品均含有高粱花叶病毒(SrMV),其中3个样品中还存在甘蔗花叶病毒(SCMV)的复合侵染.序列比较和系统进化树分析表明,浙江甘蔗样品中的SrMV序列彼此很相似,核苷酸同源性大于93%,与已报道的4个美国分离物在CP区域同源性很高,但是3′非编码区的同源性却仅为70%左右.SCMV欧洲和中国玉米分离物及美国、南非和澳大利亚甘蔗分离物分别形成两个远缘群体,浙江甘蔗分离物群体位于两者之间;群体间CP氨基酸序列同源性均大于80%.甘蔗和玉米上的SCMV差异明显,多为无义突变.  相似文献

Biological, serological and coat protein properties of a potyvirus (Poty-Rape) causing a mosaic disease of Brassica campestris and B. juncea in India were investigated. The virus readily infected 4 of the 5 plant species in the family Brassicaceae in which it induced severe systemic mosaic symptoms; it also induced chlorotic and necrotic local lesions in Chenopodium amaranticolor , but failed to infect 4 other species of Chenopodiaceae or 20 species of Amaranthaceae, Apiaceae, Canabinaceae, Compositae, Cucurbitaceae, Euphorbiaceae, Leguminosae and Solanaceae. The virus was transmitted in a non-persistant manner by Myzus persicae, Brevicoryne brassicae and Aphis gossypii. The Average size, of the virus particles in a purified preparation was 740 nm × 12 nm. SDS-PAGE analysis of the viral coat protein showed two major bands of approximately 37 kDa and 31 kDa, a pattern very similar to that of a reference isolate of turnip mosaic virus (TuMV) from the U.S. In Western-blot immunoassay, an antiserum to TuMV reacted with both the coat protein bands of the Poty-Rape islate and the reference TuMV, but not with the coat proteins of four other potyviruses. The high performance liquid chromatographic profile of tryptic peptides from the coat protein of Poty-Rape was found to be very similar to that of the reference TuMV, but differed substantially from those of four other potyviruses. The Poty-Rape isolate is considered to be a distinct strain of, TuMV.  相似文献

山东省玉米矮花叶病毒的生物学特性及基因组全序列测定   总被引:3,自引:0,他引:3  
对山东省玉米矮花叶病毒原分离物(SD)进行了寄主范围、血清学等普通生物学鉴定,测定了该病毒的基因组核苷酸全序列。该病毒基因组RNA由9596个核苷酸组成(不包括3’—polyA的长度),整个基因组按一个ORF编码一个3063个氨基酸的多聚蛋白。序列比较表明,该病毒分离物(SD)与玉米矮花叶病河南分离物(HN,EMBL登录号:AF94510)核苷酸全序列同源性最高,为98.2%,与已报道的甘蔗花叶病毒(SCMV)7个分离物同源性也高达79.5%-98.2%,但与玉米矮花叶病毒保加利亚分离物(MDMV—B8,AJ001691)和高梁花叶病毒萧山甘蔗分离物(SrMV—XoS,AJ310197)的同源性仅为67.8%和69.3%,与约翰逊草花叶病毒(226920,JGMV)差异最大。系统进化树分析也表明,该病毒与SCMV分离物位于同一进化簇,而与MDMV进化关系很远。  相似文献

The properties of a virus isolated from fluted pumpkin (Telfairia occidentalis) was investigated. It produced symptoms in some members of the Solanaceae and Leguminosae, and was transmitted nonpersistently by the aphid, Aphis spiraecola. On the basis of these alone, it is distinct from another previously described virus, Telfairia mosaic virus, which neither caused symptoms in members of these families nor was it transmitted by insects. Furthermore, the virus in crude sap or purified preparations reacted with antiserum to cucumber mosaic virus (CMV), but not with antisera to several common viruses in Nigeria. Electron microscopic examination revealed isometric particles of 29 × 1 nm in diameter. These properties confirmed that the virus is an isolate of CMV, and closely resembles the Y-strain in causing systemic mosaic symptoms in Vigna unguiculata. From infectivity and pathogenicity tests, it is concluded that it is the main cause of mosaic disease in fluted pumpkin.  相似文献
