群体遗传结构上的差异是遗传多样性的一种重要体现,对群体遗传结构的研究已有较久的历史,而其中的基因流研究近些年来越来越受到重视。它对群体遗传学、进化生物学、保护生物学、生态学有着极其重要的作用。虽然传统的群体遗传学能估测基因流大小,但它的精确性还有很大局限性。随着生物技术的进步,对基因流的研究逐渐向分子水平过渡,应用蛋白质电泳技术、分子标记技术(RAPD、RFLP、VNTR、ISSR、DNA测序等)方法对群体间基因流的流动水平进行了深入细致的研究。本文综述了群体遗传结构的几种模式:陆岛模式、海岛模式、阶石模式、距离隔离模式、层次模式,以及在群体遗传结构的几种模式基础上的基因流的研究方法、作用、地位和近些年来研究者的研究成果,并指出了这些方法的局限性。Abstract: The difference of population genetic structure is one of the important embodiments of genetic diversity. There is a long history of the study of population genetic structure, and the study of gene flow of population genetic structure is aroused more and more importance. It has an important effect on population genetics, evolution biology, conservation biology and ecology. Although the level of gene flow is estimated by traditional population genetics, there is a large restriction in its precision. With the development of biological technology, the methods of the research on gene flow reach the molecular level. Methods of protein electrophoresis and molecular markers (RAPD, RFLP, VNTR, ISSR and mitochondrial DNA) are used to research gene flow among populations. This paper introduces not only some models of population genetic structure: Continent-Island Model, Island Model, Stepping-Stone Model, Isolation-By-Distance Model and Hierarchical Model; but also the study methods, function and role of gene flow is based on models of population genetic structure, research achievements in recent years and the restriction of the methods.  相似文献

从1998~2001年调查了内蒙古自治区鄂尔多斯市、锡林郭勒盟、巴彦淖尔盟的蒙古族、汉族6个群体的拇指类型、环食指长、指甲类型、足趾长等4项人类群体遗传学指标,共获有效资料2760例。研究表明:(1)6个群体的直型拇指出现率均在60%以上,且均无性别间明显差异;(2)6个群体环指长率均远高于食指长率,有4个群体存在性别间显著差异;(3)各群体均表现出长型指甲最多、方型次之、扁型最少;(4)6个群体均多为拇趾长型,且各群体均无性别间显著差异;(5)4项指标之间无明显相关关系。 Abstract:From 1998 to 2001,a sample of 2760 Mongol and Han individuals was investigated in 3 areas (Eerduosi city,Xilinguole and Bayannaoer leagues) from Inner Mongolia.Each individual was studied on 4 genetic characters (thumb type,palmar digital formular,fingernail type and plantar digital formular).The result is as follows:(1) The frequencies of the straight thumb were over 60%,and there were no significant sexual differences in all of 6 groups.(2) The frequencies of the ring-finger-longer were much higher in 6 groups,and there were significant sexual differences in 4 groups.(3) The frequencies of the long-shaped fingernail were the highest in all groups,the squat-shaped were the lowest.(4) The big toes were longer than the second toes in most people of 6 groups,but without significant sexual differences in each groups.(5) There were no significant correlations between 4 characters.  相似文献

利用太谷核不育基因构建的遗传变异丰富的基础群体DNS2,进行了连续5轮歧化选择。本论文从不同的世代中,选择了10个子群体进行RAPD分析。采用7个引物扩增出116个位点,从基因频率和表型带两个角度的分析都表明,群体具有丰富的遗传变异。整个群体总的多态位点百分率达88.79,总杂合度为0.3143。子群体内(间)遗传距离的结果显示:子群体内的遗传差异小于子群体间的遗传差异;各选择子群体与未选群体间都有较大的遗传距离;随着选择轮次的增加,株高选择子群体间的遗传距离逐渐增大;对同一性状进行选择的子群体间世代内(间)平均遗传距离小于对不同性状进行选择的子群体内(间)的遗传距离。RAPD分子聚类结果显示出对同一性状进行选择的子群体聚在一起,反映了对株高选高的选择效果比较明显。 Abstract:A base population was established through multi-parent random crossing by using Taigu dominant male-sterile wheat,and then five cycles of 2-way selection for four quantitative characters were conducted.The dynamic changes of genetic structures in the open-pollinated wheat population were examined by RAPD technique.Seven primers in RAPD analysis amplified 116 sites.The results of gene frequencies and phenotypic bands showed abundant genetic variations existed in the population.The percentage of polymorphic loci was 8879,and the average heterozygosity was 0.3143 in the whole population.The genetic distance of RAPD showed as follows :① The genetic distance within a subpopulation was lower than that between every two subpopulations.② Each subpopulation had considerable divergence compared with unselected population.③ The genetic distances between the subpopulations which were selected for plant height gradually increased accompanied with the selection.④The genetic distance between subpopulations which were selected for the same character was lower than that were selected for different characters both in the same generation and among different generations.The cluster results of RAPD genetic distance demonstrated that the subpopulations selected for the same character going to one cluster.It also showed that the selective effect of increasing plant height was obvious.  相似文献

小麦异交群体选择分化的RAPD分析   总被引:3,自引:1,他引:2  
用RAPD分子标记技术及数量遗传分析手段对小麦异交群体趋异选择4代得到的各子群体的遗传关系进行了分析,并对两种结果进行了相关分析。采用11个引物扩增出134个位点,RAPD分析结果表明,群体具有丰富的遗传变异,整个群体的多态位点百分率达0.8134,杂合度达0.3006;子群体间遗传分化较小,相对基因分化系数Gst为0.2310,固定指数平均0.2120,标准遗传距离为0.1044±0.048,表明遗传变异多数来自群体之内。而数量遗传分析发现,选择性状进展都与预期方向相符,主成分遗传距离大部分达显著水平,说明选择使群体发生了相当程度的分化。对两种遗传距离进行聚类分析表明,二种信息间关系各类群成员多不一致。其原因可能在于:数量性状是人工选择的目标,RAPD是基因组随机序列的扩增,二者之间的相关与引物选择有关。 Abstract:The genetic relations of some subpopulations in the fourth generation of selection in outbreeding population of winter wheat were examined by RAPD techniques and quantitative analysis. The correlations of the two results were examined.For RAPD analysis, 11 primers were adopted, and 134 sites were amplified. The results showed that abundant genetic variations existed in the populations. In the whole population, the percentage of polymorphic sites (P) is 0.8134, and the averaged heterozygosity is 0.3006. However,genetic differentiation among subpopulations is small. The relative gene differentiation (Gst) is 0.2310,fixed index averages 0.2120, and the standard genetic distance between subpopulations is 0.1044±0.048.All above shows that most of the genetic variations existed within populations.The cluster results from RAPD analysis and principal component distances showed that similarity relations were only found in small part of the subpopulations. The reason for the results may be that the genes concerned for quantitative traits are selected directly,while the genes involved in RAPD analysis are only random samples of the whole genomes.  相似文献

研究广东少数民族群体GALNS基因StuI位点的遗传多态性以及该位点等位基因片段传递的规律,为今后的连锁分析打下基础。采用PCR-RFLP方法,对72例无血缘关系的健康广东少数民族个体的144条染色体和3个家系9位成员的18条染色体进行检测,然后用χ2检验进行统计学处理。等位基因片段D1的频率为0.70, D2为0.30,杂合率为29%,D1、D2的传递规律与理论上预计的完全符合。广东少数民族群体中StuI位点具有多态性,其基因频率(D1和D2)与国外高加索群体的有显著差别,与日本群体及中国南方汉族群体的则无显著差别;而杂合率与高加索群体及日本群体的均有显著差异,但与中国南方汉族群体的则无显著差异。 Abstract:To investigate the genetic polymorphism of the StuI site in the GALNS gene from a national minority population in Guangdong and to study the mode of transmission of alleles,PCR-RFLP was used to analyze 144 chromosomes from 72 Guangdong unrelated healthy national minority individuals,and the genotypes of members in three families.To compare the frequencies and heterzygosity between Guangdong national minority people and Caucasians,Japanese and Chinese Han people by using χ2 test.The frequency of allele D1(295bp) was 0.70,allele D2(138 plus 157 bp)0.30,the heterozygosity was 29%.The genotypes of each member of all families detected were completely agreement with the theorical assessment.The site of StuI in the GALNS gene from national minority population in Guangdong has polymorphism.There is significant difference between Guangdong national minority population and Caucasians in Western countries,but no significant difference was found between Guangdong national minority population and Japanese and Chinese Han population.In addition,there is significant difference between Guangdong national minority population and Caucasians and Japanese in the heterzygosity,but no significant difference between Guangdong national minority population and Chinese Han population.The transmission of alleles was completely in agreement with the Mendelian genetic law.  相似文献

用同步电泳分型的方法,对中国内蒙古鄂伦春族、鄂温克族、达斡尔族人506份血痕红细胞酶EAP、ADA、AK1遗传多态性进行了研究,被调查群体中均未检出上述三种酶型的变异型。被调查群体三种酶型基因频率范围分别为EAPA 0.2139~0.2280;ADA10.9500~0.9596;AK1 1.0~0.9963。 Abstract:In the paper the phenotype freguencies of the EAP,ADA and AK1 have been investigated by the emthod of typing of EAP-ADA-AK on the samemixed starch/agarose gel electrophresis among 3 nationalities in Inner Mongolia.(Oroqen,Ewenki and Dawuor.) 波尔山羊杂交后代及其亲本随机扩增多态DNA研究   总被引:24,自引:4,他引:20  
利用随机扩增多态DNA技术研究了波尔山羊、唐山奶山羊、青龙本地山羊以及波尔山羊与这两个山羊群体杂交后代共计128个山羊个体的随机扩增多态DNA。结果表明:(1)总群体平均遗传多样性指数(Hsp)为0.6974,群体遗传分化指数为0.9706,山羊群体间平均遗传距离指数(0.1314~0.2052)明显大于群体内的相应值(0.0582~0.1440),上述结果说明,所研究山羊群体不仅具有较为丰富的遗传多样性,而且其核基因组遗传变异主要存在于群体间。(2)山羊群体间的分子聚类关系与各群体间的亲缘关系基本一致。 Abstract:The random amplified polymorphic DNA(RAPD)of 128 individuals was studied,which were from Boer goat, Tangshan dairy goat,Qinglong native goat and their hybrids crossbred with Boer goat.The average index of genetic polymorphism for whole population(Hsp)and the index of genetic differentiation were 0.6974 and 0.9706,respectively.The average index of genetic distance between populations(0.1314~0.2052)was significantly higher than that within populations(0.0582~0.1440).All of these indicated that the genetic polymorphism was not only abundant,but also the genetic variation was mainly existed between goat populations.The molecular dendrogram among goat populations was in accord with their genetic relationship.  相似文献

利用叶绿体微卫星(cpSSR)分子标记对日本国内的7个日本落叶松(Larix kaempferi)群体遗传结构进行了研究。11对cpSSR引物中筛选出的3对多态性引物,共产生10个长度不同片段,在197个样品中组合出现10个不同的单倍型(haplotype)。各群体的单倍型差异较大。cpSSR基因座揭示了日本落叶松的遗传变异:平均等位基因数A=3.33,平均有效等位基因数NE=1.20,基因多样度HE=0.17,群体间变异占总群体变异的5.37%,遗传变异主要来自于群体内个体间。Abstract: Genetic structure of seven populations in Larix kaempferi in Japan was studied by use of cpSSR markers. Ten different length fragments in and ten different kinds of haplotypes were reduced in 197 samples based on 3 pairs of polymorphic primers screened from 11 pairs of primers. There were significant variant haplotypes among the populations. The genetic variation in the populations of Larix kaempferi was detected by using cpSSR with the number of average loci A=3.33, the number of average efficient loci NE=1.20, gene diversity HE=0.17 and 5.37% variation from different populations. 采用水平切片淀粉凝胶电泳的方法,对分布于我国5省的8个库蚊复组(Culex pipiens complex)野生群体的遗传多样性进行研究,分析了4个酶系统7个基因座(ME、MDH-1、MDH-2、MDH-3、GPD、EST-2、EST-3)的酶谱资料。结果显示:(1)群体内存在不同程度的遗传变异(He为0.098~0.41);(2)较低的基因流水平(Nm=0.64)使遗传漂变起主要作用,造成群体之间的遗传分化(Gst=0.303),而总群体的遗传多样性相对富集于群体之内(Hs/Dst=2)。(3)库蚊群体的遗传结构属于距离隔离模式。(4)群体间的遗传一致性(或遗传距离)反映出群体间的遗传分化程度,也表明与地理位置存在对应关系。Abstract: Eight field populations of Culex pipiens complex collected from five provinces (Guangdong, Henan, Shandong, Beijing and Yunnan) in 2001 were used to study genetic diversity by starch gel electrophoresis. Data from seven loci (ME、MDH-1、MDH-2、MDH-3、GPD、EST-2、EST-3) of four isozymes were analyzed by software Biosys2.0 and FSTAT(Version 2.9.3). The results were as follows: (1) The values of He (from 0.098 to 0.41) indicated genetic variabilities of different degree in populations.(2)The low level of gene flow (Nm=0.64) could not prevent genetic drift to cause the gene differentiation between populations. The genetic diversity between populations attributed to the genetic diversity of total populations is small (Gst =0.303), and the great part is accumulated within populations (Hs/Dst=2). (3) The genetic structure of Culex pipiens complex population was the isolation-by-distance model. (4) The genetic identity (or genetic distance) revealed the scale of genetic differentiation between populations which related to the collection sites.  相似文献

利用微卫星(SSR)标记对我国4个省内的5个栓皮栎(Quercus variabilis Bl.)天然群体的遗传多样性进行了研究。16对SSR标记揭示了栓皮栎丰富的遗传多样性:等位基因数(A)平均8.4375个,有效等位基因数(Ne)平均为5.9512个,平均期望杂合度(He)0.8059,Nei多样性指数(h)为0.8041。栓皮栎自然分布区中心地带的群体具有较高的遗传多样性,而人为对森林的破坏将降低林木群体的遗传多样性。栓皮栎群体的变异主要来源于群体内,群体间分化较小,遗传分化系数仅为0.0455。此外,栓皮栎群体间的遗传距离与地理距离之间存在显著的正相关。这些遗传信息为栓皮栎遗传多样性的保护和利用提供了一定依据。Abstract: Genetic diversity of five Quercus variabilis natural populations in four provinces of China was studied with microsatellite (SSR) markers. A relatively high level of genetic diversity was detected in Q. variabilis species with 16 polymorphic microsatellite loci. Average number of alleles (A) and effective number of alleles (Ne) were 8.4375 and 5.9512 respectively. The mean expected heterozygosity (He) was 0.8059 and Nei diversity index (h) was 0.8041. Higher diversity was found with the populations from the central range of the species in contrast to those from peripheral areas and human activities might decrease the genetic diversity of populations. The majority of genetic variation occurred within populations, which could be concluded from the low coefficient of genetic differentiation (Fst=0.0455). In addition, significant correlation was found between geographical distance and genetic distance. All these results present a basis to the conservation and utilization of genetic diversity of Quercus variabilis.  相似文献

本文提出亚群体分析法,在二个衍生于杂交稻推广组合的F2群体中,挑选均匀分布于连锁图谱的DNA标记作为固定因子,分别根据每个固定因子的基因型将F2群体分成三类亚群体:母本型(I型)、父本型(II型)和杂合型(III型)。在大量III型亚群体中,杂合度与产量和穗数呈显著正相关。在表现这种相关性的III型亚群体内分析产量QTL和穗数QTL,发现超显性作用是研究组合杂种优势的主要遗传基础。 Abstract:Two F2 populations were each derived from a commercial hybrid and used for identification of genetic factors contributing to heterosis of rice.DNA markers distributing evenly in the linkage maps were selected as fixing factors.The F2 populations were divided into three types of sub-populations based on the genotypes of each fixing factor:maternal type (Type I),paternal type (Type II) and heterozygous type (Type III).In a large number of Type III sub-populations,significant correlations were observed between heterozygosity and grain yield,and between heterozygosity and number of panicle.QTL analysis in Type III sub-populations showing such correlations indicated that over-dominance was the major genetic basis of heterosis in the two crosses.  相似文献

采用JL-02多位点探针和Southern杂交对Wistar大鼠基因组进行了DNA指纹分析,并对国内最具有典型的6个地区9个Wistar大鼠群体内和群体间进行了DNA指纹分析比较。结果表明,DNA指纹图较好地反映了封闭群动物的遗传本质,具有良好的多态性。同一群体不同个体间Wistar大鼠遗传距离主要为0.2~0.6,将不同群体的Wistar大鼠DNA指纹图带进行分析表明,所有群体间的DNA指纹图的遗传距离在0.2~0.7。 Abstract:DNA fingerprinting of Wistar rat were studied with JL-02 Mulilocus probe and Southern hybridization,and comparing with different individuals of nine groups Wistar rat from six national classical area and different groups.It was indicated that DNA fingerprinting could reflect genetic material of outbred strain rat,and were more polymorphic.The genetic distances of Wistar rat were distributed for 0.2~0.6 among different individuals within same groups,and the genetic distances of Wistar rat were distributed for 0.2~0.7 among different groups.  相似文献

以广东徐闻金碧公司养殖场、广西北海营盘镇养殖场和南海水产研究所海南实验基地3个合浦珠母贝养殖群体为对象,利用8个微卫星位点M1、M2、M3、M4、M5、M6、M7、M8的引物进行了遗传多样性分析.结果表明:8个微卫星标记位点在3个养殖群体中共检测到58个等位基因,观测等位基因数为2~9个,平均有效等位基因数3.72~5.06,平均观察杂合度0.41~0.56,平均期望杂合度0.67~0.75,3个群体的平均多态信息含量PIC值为0.62~0.70,全部为高度多态(PIC≥0.5),表明这几个合浦珠母贝养殖群体目前仍具有较高的遗传多样性,遗传信息丰富,遗传变异大,可以作为良好的育种材料;在这3个养殖群体中,南海水产研究所海南实验基地的养殖群体的遗传多样性最高,广西北海营盘镇养殖群体遗传多样性最低,这一结果可以为今后选择育种、种质保护提供可资借鉴的资料.
Abstract:
By using eight mierosatellite loci (M1, M2, M3, M4, M5, M6, M7 and MS), the genetic diversity of three Pinctada fucata populations from the pearl farms in Xuwen of Guang-dong and Beihai of Guangxi, and from the experimental base of South China Sea Fisheries Re-search Institute in Hainan was studied. A total of fifty eight alleles of these eight microsatellite lo-ci were detected, among which, the observed allele number was 2-9, average effective allele number was 3.72-5.06, average observed population heterozygosity was 0. 41-0. 56, and aver-age observed expected heterozygosity was 0. 67-0. 75. All the three populations had a polymor-phie information content (PIC) of 0. 62-0.70, suggesting their high polymorphism (PIC > 0. 5). Among the three populations, the cultured population from the experimental base of South China Sea Fisheries Institute had the highest polymorphism, and that from Beihai of Guangxi had the lowest one. These results provided useful information for the selective breeding and germplast conservation of P. Fucata.  相似文献

采用水平淀粉凝胶电泳技术,对分布于金沙江流域的云南红豆杉天然群体的10种酶系统同工酶的遗传变异进行了研究。在谱带遗传分析的基础上确定了15个酶基因座及其等位基因。其中有14个酶基因座属多态,只有一个单态基因座(ME-3)。14个多态基因座中,4个基因座遗传变异小,对该天然群体的遗传变异贡献不大,其余10个基因座遗传变异丰富,对该天然群体的遗传变异贡献大。该天然群体具有明显丰富的遗传变异性,多态基因座比率P=0.933,等位基因平均数A=2.90,平均期望杂合度He=0.290。紫杉醇含量与群体遗传变异有着密切的关系。 Abstract:Genetic variation of ten isoenzymes was studied within population of Taxus yun nanensis Cheng et L. K. Fu in the Jinsha River Valley using the method of hor izo ntal starch gel electrophoresis.On the basis of banding analysis,eight enzyme sy stems , presumablly coded by fifteen isoenzyme loci and their alleles were score d , and fourteen were polymorphic with only one monomorphic locus(ME-3). Amon g them , four polymorphic loci with inevident genetic variation made little cont ribution to genetic variation of this population and other ten polymorphic loci with evident genetic variation made great contribution to genetic variation of this population . Isoenzyme data indicated high level of genetic variability in this population with P=0.933,A=2.90,He=0.290 . The taxol content had close r elation to genetic variation of population.  相似文献

寄生蠕虫群体遗传学研究常用的遗传标记有等位酶、线粒体DNA、随机扩增多态性DNA或扩增性片段长度多态性和微卫星DNA等。应用这些遗传标记的研究表明,大多数寄生蠕虫群体遗传结构有不同水平的变异,这些变异的产生主要与寄生虫的生活史和群体生态、宿主的地理分布和环境等因素有关,并因此提出了有关遗传变异的一些假说。本文对寄生蠕虫群体遗传学的研究作一综述。 Abstract:Genetic markers including allozyme,mtDNA,RAPD/RFLP and micro DNA have been used in the research of helminth population genetics.Available data on helminth genetic variability have shown that most helminth populations exhibit different levels of genetic variation resulting mainly from the pattern of life cycle,geographical distribution and parasite-host interaction,and several hypotheses have been proposed to explain the genetic variation.  相似文献

利用模拟方法比较了5种方差组分估计方法(ANOVA,Henderson-Ⅲ,MLMT,R EMLMT和MIVQUE)对遗传参数的估计效果。结果表明:REMLMT法在各种情况下均能得到较好的参数估值,估计的准确度相对较高;ANOVA方法在小群体和不均衡资料情况下估计效果最差。同时,群体含量和结构能影响各种方法的估计效果, 群体含量较小或资料来源的公畜数太少,将导致遗传参数的估计误差增大,准确性降低。 Abstract:Five methods for estimating components of (Co) variance,including ANOVA,Henderson-III,MLMT,REMLMT and MIVQUE were compared using computer simulation.The results showed that REMLMT could obtain more accurate estimates for all data sets,while the estimates obtained by ANOVA always had greater deviations from the true values,especial for small and well-unbalanced data sets.Also,the effects of estimation were dependent on structure of population size,accuracy of estimates would be decreased when number of sires was too few or data contained small number of animals.  相似文献

中国南方汉族群体MPSI型Kpn I酶切位点的遗传多态性   总被引:1,自引:0,他引:1  
为研究中国汉族群体IDUA 基因Kpn I 酶切位点的遗传多态性以及该位点等位基因片段传递的规律, 采用PCR-RFLP技术, 对162例无血缘关系的健康中国汉人的324条 染色体进行检测,另又对5个家系16位成员进行同样的检测,然后用χ2检验进行统计学处理。结果表明,等位基因A1 频率为0.17,等位基因A2 频率为0.83,杂合率为29%;A1 、A2 的传递规律与理论上预计的完全符合。认为中国汉族群体IDUA 基因 Kpn I 酶切位点也具有遗传多态性, 并且与国外报道的无显著性差异;A1、A2 在世代中的传递完全符合孟德尔遗传规律。 Abstract:To investigate the genetic polymorphism of the Kpn I site in the α-L-iduronidase(IDUA) gene from a Han population in southern China and to study the mode of transmission of alleles, PCR-RFLP was used to analyze 324 chromosomes from 162 Chinese unrelated healthy Han individuals, and the analysis of the genotypes of 16 members in five families. To compare the frequencies and heterzygosity between Chinese Han population and Caucasians in Western by using χ2test. The frequency of allele 1 (450bp) was 0.17,allele 2 (390 plus 60 bp) 0.83, the heterozygosity was 29%.The genotypes of each member of all families detected was completely agreement with the theorical assessment. The locus of Kpn I in the IDUAgene from Han population has polymorphism. There is no significant difference between Chinese Han population and Caucasians in Western countries. The transmission of alleles was agreement with the Mendelian genetic law.  相似文献
