The soil microbial population of a coke oven site was investigated in order to evaluate its potential for bioremediation. The study was carried out in soil samples with distinct polynuclear aromatic hydrocarbon (PAH) contamination levels, comparing the population profiles constituted by total heterotrophic and PAH-utilizing strains. Isolation of degrading strains was performed with phenanthrene or pyrene as sole carbon sources. The ability to degrade other PAHs, such as anthracene, fluorene and fluoranthene was also investigated. The results showed a reduction of 30% in species diversity and microbial density drops one order of magnitude in contaminated samples. Furthermore, the number of PAH-utilizing colonies was higher in the contaminated area and about 20% of the isolates were able to degrade phenanthrene and pyrene, while this value decreased to 0.15% in uncontaminated samples. Three PAH-degrader strains were identified as: CDC gr. IV C-2, Aeromonas sp. and Pseudomonas vesicularis. The ability of these strains to degrade other PAHs was also investigated.  相似文献

Pyrene and fluoranthene, when supplied as the sole carbon source, were not degraded by Burkholderia sp. VUN10013. However, when added in a mixture with phenanthrene, both pyrene and fluoranthene were degraded in liquid broth and soil. The amounts of pyrene and fluoranthene in liquid media (initial concentrations of 50 mg l−1 each) decreased to 42.1% and 41.1%, respectively, after 21 days. The amounts of pyrene and fluoranthene in soil (initial concentrations of 75 mg kg−1 dry soil each) decreased to 25.8% and 12.1%, respectively, after 60 days. None of the high molecular weight (HMW) polycylic aromatic hydrocarbons (PAHs) tested adversely affected phenanthrene degradation by this bacterial strain and the amount of phenanthrene decreased rapidly within 3 and 15 days of incubation in liquid broth and soil, respectively. Anthracene also stimulated the degradation of pyrene or fluoranthene by Burkholderia sp. VUN10013, but to a lesser extent than phenanthrene. The extent of anthracene degradation decreased in the presence of these HMW PAHs.  相似文献

Microorganisms obtained from a contaminated experimental soil were found to reductively dechlorinate the polychlorobiphenyls (PCBs) of ex-commercial Fenclor 54 and of a synthetic mixture of single congeners, under laboratory anaerobic conditions. The dechlorination rate and extent tended to increase as the chlorination degree of F 54 congeners increased. Several penta-chlorinated congeners temporarily accumulated during the final period of incubation. Dechlorination occurred primarily from the meta and para positions while ortho-sustituted congeners accumulated in the medium during incubation. The dechlorination pattern observed with these unacclimated microorganisms in both PCB mixtures could be only partially compared to patterns reported in the literature. The low product yield deriving from reductive dechlorination of PCBs, i.e. di-and tri-chlorinated biphenyls, and the slow rate of PCB biotransformation can be attributed to a lower dehalogenation capability of artificially contaminated soil microorganisms and, perhaps, also to the inadequacy of the adopted anaerobic medium. The behaviour of some organic micropollutants in the abiotic compartments water, particulate matter and bottom sediments of the Scheldt estuary, S.W. Netherlands, was studied between 1986 and 1989. Special attention was given to two individual PolyChlorinated Biphenyl congeners (PCB 52 and PCB 153), two Polycyclic Aromatic Hydrocarbons (PAHs) fluoranthene and benzo(a)pyrene and γ-HCH. Dissolved PCBs and γ-HCH behave conservatively during estuarine mixing. The concentration of γ-HCH in the fresh-water is extremely high in spring compared to the other seasons. Dissolved fluoranthene does not behave conservatively. The concentration of dissolved fluoranthene in the freshwater endmember shows a seasonal dependence with highest concentrations in winter and lowest in summer. Benzo(a)pyrene could hardly be detected in the dissolved phase with the methods used. PCBs and PAHs in particulate matter and sediments behave essentially conservatively as a result of the mixing of riverine particulates with high, and marine particulates with low organic micropollutant contents. Particulate fluoranthene is removed at low salinities under (nearly) anoxic conditions probably by microbial degradation. The rivers Scheldt and Rupel appear to be the major sources for the compounds studied. However, in particular the organic micropollutant contents of the sediments indicate that important emissions along the estuary were or are still present. Measurements of individual PCBs and PAHs in the <63 μm fraction of a dated sediment core from a salt marsh in the eastern part of the Scheldt estuary show that the recent input of PCBs and PAHs into the estuaryis probably 2–3 times lower than the maximum input in the mid-1960s. The PCB profile shows postdepositional congener-selective mobilization caused by advective transport. The PAH assemblage is remarkably uniform in the sediment core. The PAH ratios indicate that combustion of coal is the main source of PAHs in the Scheldt estuary.  相似文献

In this work, we evaluate the abilities of the plants Brassica juncea, Avena sativa, Brachiaria decumbens, and Medicago sativa to uptake polychlorinated biphenyls (PCBs) and induce degradation of soil microorganisms from contaminated soil. Removal of PCBs 44, 66, 118, 153, 170, and 180 was evaluated in both rhizospheric and nonrhizospheric soils. Microbial and bphA1 gene quantifications were performed by real-time PCR. The PCB concentrations in plant tissues and soil were determined, and a fluorescein diacetate (FDA) hydrolysis assay was used to measure microbial activity in soil. The removal percentages for all PCB congeners in planted soil versus unplanted control soil were statistically significant and varied between 45% and 63%. PCBs 118, 153, 138, and 170 were detected in Brachiaria decumbens roots at different concentrations. In planted soil, an increase in the concentration of bacteria was observed compared to the initial concentration and the concentration in unplanted control soil; however, no significant differences were identified between plants. The number of copies of the bphA1 gene was higher in rhizospheric versus non- rhizospheric soil for all plants at the end of the experiment. However, alfalfa and oat rhizospheric soil showed significant differences in the copy number of the bphA1 gene. In general, the concentration of fluorescein in the rhizospheric soil was greater than that in the nonrhizospheric soil. Although the plants had a positive effect on PCB removal, this effect varied depending on the type of PCB, the plant, and the soil.  相似文献

The ability of birch (Betula pendula) and mulberry (Morus rubra), cultivated either separately or together with perennial ryegrass (Lolium perenne), to enhance the biodegradation of 15 selected polycyclic aromatic hydrocarbons (PAHs) in long-term contaminated soil was studied in a greenhouse experiment. The microecosystems (MESs) were cultivated for either 12 or 18 months at a natural photoperiod. The fact that the soil chosen for cultivation had been contaminated for over 50 years was expected to be the main factor limiting biodegradation. Extracts of both planted and unplanted soil were analyzed using HPLC. After 1 year of cultivation, the overall content of the investigated compounds had declined to 50%. The concentrations of fluoranthene and pyrene, the PAHs originally present in the soil in the highest concentrations (103.5 and 83.3 mg/kg, respectively), had decreased to 28.0 and 18.0 mg/kg, respectively. In addition, other compounds were successfully degraded, including even benzo[a]pyrene. Benzo[ghi]perylene and indeno[1,2,3-cd]pyrene were the only PAHs remaining and were almost entirely undegraded. Because few significant differences were found between the various types of MESs studied, degradation can primarily be attributed to the presence of degrading microorganisms in the soil.  相似文献

Mycobacterium sp. PYR‐1, which was previously shown to mineralize several individual polycyclic aromatic hydrocarbons (PAHs), simultaneously degraded phenanthrene, anthracene, fluoranthene, pyrene and benzo[a]pyrene in a six‐component synthetic mixture. Chrysene was not degraded significantly. When provided with a complex carbon source, Mycobacterium sp. PYR‐1 degraded greater than 74% of the total PAH mixture during 6 d of incubation. Mycobacterium sp. PYR‐1 appeared to preferentially degrade phenanthrene. No significant difference in degradation rates was observed between fluoranthene and pyrene. Anthracene degradation was slightly delayed but, once initiated, proceeded at a constant rate. Benzo[a]pyrene was degraded slowly. Degradation of a crude mixture of benzene‐soluble PAHs from contaminated sediments resulted in a 47% reduction of the material in 6 d compared with that of autoclaved controls. Experiments using an environmental microcosm test system indicated that mineralization rates of individual 14C‐labeled compounds were significantly lower in the mixtures than in equivalent doses of these compounds alone. Mineralization of the complete mixture was estimated conservatively to be between 49.7 and 53.6% and was nearly 50% in 30 d of incubation when all compounds were radiolabeled. These results strengthen the argument for the potential application of Mycobacterium sp. PYR‐1 for bioremediation of PAH‐contaminated wastes.  相似文献

Polycyclic Aromatic Hydrocarbons (PAHs) are diverse organic contaminants released into the environment by both natural and anthropogenic activities. These compounds have negative impacts on plants growth and development. Although there are many reports on their existence in different parts of plant, their uptake and translocation pathways and mechanisms are not well understood yet. This paper highlights the uptake, translocation and accumulation of PAHs by wheat, sunflower and alfalfa through an experimental study under controlled conditions. Seeds were cultivated in a soil containing 50 mg/kg of phenanthrene and fluorene and their concentrations in plants roots and shoots were determined using a gas chromatograph after 7 and 14 days. The results showed that phenanthrene and fluorene concentrations in the treated plants were increased over the time. PAHs bioavailability was time and species dependent and generally, phenanthrene uptake and translocation was faster than that of fluorene, probably due to their higher Kow. Fluorene tended to accumulate in roots, but phenanthrene was transported to aerial parts of plants.  相似文献

以菲和芘为多环芳烃(PAHs)代表物,采用水培体系研究了黑麦草(Lolium multiflorum Lam)对水中PAHs的吸收作用,重点研究了植物吸收菲和芘的时间动态.水中菲和芘起始浓度分别为1.00mg/L和0.12mg/L.0~288h内,黑麦草根和茎叶中菲和芘含量均先快速增加而后降低,积累量不断增大,植物根系和茎叶富集系数则先快速升高而后趋于稳定.茎叶中菲和芘含量、茎叶对菲和芘的富集系数比根低1~3数量级,积累量也明显小于根系.黑麦草根系对水中芘有更强的富集能力,其根系富集系数比菲大85%~179%;而其茎叶对菲的富集作用则略强.菲和芘在植物体内有明显的传导作用.0~288h,传导系数(TF)先显著升高而后趋于恒定;但实验条件下,菲和芘的TF值均很小,分别不高于0.031和0.009,且芘的TF值明显小于菲,表明供试植物对芘的传导能力更弱.  相似文献

A mixture of six polyaromatic hydrocarbons (naphthalene, phenanthrene, fluoranthene, pyrene, chyrysene and benzo[a]pyrene), varying in size from 2 to 5 rings, was dissolved in dodecane, and used as the delivery phase of a partitioning bioreactor. Two species of Sphingomonas were then used individually, and as a consortium, to determine which of the PAHs were degraded. Only low molecular weight PAHs (naphthalene, phenanthrene and fluoranthene) were degraded by the individual strains, but the consortium degraded all substrates either to completion or near completion.  相似文献

Soil and sediment samples from New Orleans have been collected and analyzed for contamination by 16 polycyclic aromatic hydrocarbons (PAHs) and 8 trace metals. Total PAH contents were found to vary from 40 μ g/kg to 40,000 μ g/kg, and concentrations of total metals varied in the range of 80 mg/kg and 7600 mg/kg. Source analysis of PAHs using diagnostic concentration ratios such as phenanthrene/anthracene and fluoranthene/ pyrene indicated that PAHs found at elevated concentrations in New Orleans soils and sediments were of pyrolytic origins. Spearman rank bivariate correlation analysis revealed significant correlations between soil PAHs and metals (r = 0.80, p < 0.0001) and between sediment PAHs and metals (r = 0.62, p < 0.05), suggesting common pollution sources for the two groups of environmental pollutants. Strong correlations were also found between Pb and Zn in soils (r = 0.93, p < 0.0001) as well as in sediments (r = 0.65, p < 0.05).  相似文献

The present study aims at analyzing the degradation of polycyclic aromatic hydrocarbons (PAHs) at acidic conditions (pH = 2) by acidophilic Stenotrophomonas maltophilia strain AJH1 (KU664513). The strain AJH1 was obtained from an enrichment culture obtained from soil samples of mining area in the presence of PAH as sole sources of carbon and energy. Strain AJH1was able to degrade low (anthracene, phenanthrene, naphthalene, fluorene) and high (pyrene, benzo(e)pyrene and benzo(k)fluoranthene) molecular weight PAHs in acidophilic mineral salt medium at pH 2, with removal rates of up to 95% (LMW PAH) and 80% (HMW PAH), respectively. In addition, strain AJH1 treated petroleum wastewater with 89 ± 1.1% COD removal under acidic condition (pH 2) in a continuously stirred reactor. Acidophilic S. maltophilia strain AJH1, hence holds the promise as an effective degrader for biological treatment of PAHs contaminated wastewater at acidic pH.

Nonexhaustive extraction (propanol, butanol, hydroxypropyl-β-cyclodextrin [HPCD]), persulfate oxidation and biodegradability assays were employed to determine the bioavailability of polycyclic aromatic hydrocarbons (PAHs) in creosote-contaminated soil. After 16 weeks incubation, greater than 89% of three-ring compounds (acenaphthene, anthracene, fluorene, and phenanthrene) and 21% to 79% of four-ring compounds (benz[a]anthracene, chrysene, fluoranthene, and pyrene) were degraded by the indigenous microorganisms under biopile conditions. No significant decrease in five- (benzo[a]pyrene, benzo[b+k]fluoranthene) and six-ring compounds (benz[g,h,i]perylene, indeno[1,2,3-c,d]pyrene) was observed. Desorption of PAHs using propanol or butanol could not predict PAH biodegradability: low-molecular-weight PAH biodegradability was underestimated whereas high-molecular-weight PAH biodegradability was overestimated. Persulfate oxidation and HPCD extraction of creosote-contaminated soil was able to predict three- and four-ring PAH biodegradability; however, the biodegradability of five-ring PAHs was overestimated. These results demonstrate that persulfate oxidation and HPCD extraction are good predictors of PAH biodegradability for compounds with octanol-water partitioning coefficients of < 6.  相似文献

Polychlorinated biphenyls (PCBs) are toxic and persistent compounds that are difficult to break down and biodegrade. Plant secondary metabolites (PSMs) on root exudates can act as inducers of the biphenyl catabolic pathway, enhancing PCB biodegradation. In this study, the authors evaluated the effect of root exudates and PSMs obtained from Avena sativa, Brachiaria decumbens, Medicago sativa, and Brassica juncea on the biodegradation of PCB 44, PCB 66, PCB 118, PCB 138, PCB 153, PCB 170, and PCB 180 by a microbial consortium isolated from the rhizosphere of plants grown on soil contaminated with Aroclor 1260. Microorganisms were identified as Pseudomonas sp. and Stenotrophomonas sp. based on their 16S rRNA sequence. The plant root exudates increased the degradation percentage of PCB 44, PCB 66, and PCB 118, which were used as carbon source by the microorganisms. Flavanone, flavone, isoflavone, 7-hydroxyflavanone, 7-hydroxyflavone, and 6-hydroxyflavone were the PSMs identified in the root exudates, which increased the degradation percentage of all seven PCB congeners; they were also used as growth substrates by microbial consortium. These results showed the importance of the interaction between plants and microorganisms for achieving the removal of persistent pollutants such as PCBs from soil.  相似文献

Ethoxyresorufin O-deethylase (EROD) activity was measured in the liver of dab (Limanda limanda) and flounder (Platichthys flesus) from the German Bight (southern North Sea) and compared with muscle and liver polychlorinated biphenyl (PCB) concentrations in an attempt to relate EROD activity to PCB body burden. In none of the different datasets (species-, tissue- or matrix-dependent) was a significant (P<0.05) correlation between PCB tissue contamination and EROD activity found. Yet EROD activity was significantly correlated with polycyclic aromatic hydrocarbons (PAH) levels (phenanthrene, fluoranthene, pyrene) in muscle tissue, indicating a possible dependence of EROD expression on other ubiquitous organic contaminants, thus making it a suitable biomarker for general pollution. Seventy-nine white rot strains were screened to determine if they had the potential for use in the degradation of oligocyclic aromates (PAHs) by measuring their dye-decoloration rate. Fourteen strains that were selected based on their dye-decoloration rate were then evaluated for the ability to tolerate various levels of PAHs spiked in agar medium. The ability of white rot fungi to degrade 3- or 4-ring PAHs (anthracene, phenanthrene, fluoranthene, pyrene) was determined. Two strains of Phanerochaete sordida (KUC8369, KUC8370) were possible PAHs degraders, degrading a significantly greater amount of phenanthrene and fluoranthene than the culture collection strain P. chrysosporium (a known PAHs degrader). The production of manganese peroxidase, the only extracellular ligninolytic enzyme detected during the cultivation, was evaluated.  相似文献

As the depth of soil petroleum contamination can vary substantially under field conditions, a rhizotron experiment was performed to investigate the influence of endophyte, P. indica, on maize growth and degradation of petroleum components in a shallow and a deep-reaching subsurface layer of a soil. For control, a treatment without soil contamination was also included. The degree in contamination and the depth to which it extended had a strong effect on the growth of the plant roots. Contaminated soil layers severely inhibited root growth thus many roots preferred to bypass the shallow contaminated layer and grow in the uncontaminated soil. While the length and branching pattern of these roots were similar to those of uncontaminated treatment. Inoculation of maize with P. indica could improve root distribution and root and shoot growth in all three contamination treatments. This inoculation also enhanced petroleum degradation in soil, especially in the treatment with deep-reaching contamination, consequently the accumulation of petroleum hydrocarbons (PAHs) in the plant tissues were increased.  相似文献

Aerobic, mesophilic bacteria from coal tar–contaminated soil were analyzed for pyrene utilization capacity and identified by 16S ribosomal DNA sequencing as members of three genera: Bacillus spp., Pseudomonas sp., and Rhodococcus sp. The soil contained nine different hazardous polyaromatic hydrocarbons (PAHs): benzo[g, h, i]perylene, dibenzo[a, h]anthracene, indeno[1,2,3-c,d]pyrene, pyrene, acenaphthylene, fluorene, phenanthrene, benzo[k]fluoranthene, and benzo[b]fluoranthene. Bacillus spp. (PK-6) MTCC 1005 showed 56.4% utilization of pyrene (C16H10) (50 μg ml?1) in 4 days, with growth associated biosurfactant activity and resulted in the formation of five new intermediates: phenanthrene (C14H10), 9,10-diphenylphenanthrene (C26H18), 9-methoxyphenanthrene (C15H12O), 5,6,7,8-tetrahydro-1-naphthoic acid (C11H12O2), and 1,6,7-trimethylnaphthalene (C13H14). The results suggested that Bacillus spp. could be found suitable for practical field application for effective in situ PAH bioremediation.  相似文献

Out of a number of white-rot fungal cultures, strains ofIrpex lacteus andPleurotus ostreatus were selected for degradation of 7 three- and four-ring unsubstituted aromatic hydrocarbons (PAH) in two contaminated industrial soils. Respective data for removal of PAH in the two industrial soils byI. lacteus were: fluorene (41 and 67%), phenanthrene (20 and 56%), anthracene (29 and 49%), fluoranthene (29 and 57%), pyrene (24 and 42%), chrysene (16 and 32%) and benzo[a]anthracene (13 and 20%). In the same two industrial soilsP. ostreatus degraded the PAH with respective removal figures of fluorene (26 and 35%), phenanthrene (0 and 20%), anthracene (19 and 53%), fluoranthene (29 and 31%), pyrene (22 and 42%), chrysene (0 and 42%) and benzo[a]anthracene (0 and 13%). The degradation of PAH was determined against concentration of PAH in non-treated contaminated soils after 14 weeks of incubation. The fungal degradation of PAH in soil was studied simultaneously with ecotoxicity evaluation of fungal treated and non-treated contaminated soils. Compared to non-treated contaminated soil, fungus-treated soil samples indicated decrease in inhibition of bioluminescence in luminescent bacteria (Vibrio fischerii) and increase in germinated mustard (Brassica alba) seeds. The biphenyl-utilizing bacterial strain KBC101 has been newly isolated from soil. Biphenyl-grown cells of KBC101 efficiently degraded di- to nonachlorobiphenyls. The isolate was identified as Paenibacillus sp. with respect to its 16S rDNA sequence and fatty acid profiles, as well as various biological and physiological characteristics. In the case of highly chlorinated biphenyl (polychlorinated biphenyl; PCB) congeners, the degradation activities of this strain were superior to those of the previously reported strong PCB degrader, Rhodococcus sp. RHA1. Recalcitrant coplanar PCBs, such as 3,4,3,4-CB, were also efficiently degraded by strain KBC101 cells. This is the first report of a representative of the genus Paenibacillus capable of degrading PCBs. In addition to growth on biphenyl, strain KBC101 could grow on dibenzofuran, xanthene, benzophenone, anthrone, phenanthrene, naphthalene, fluorene, fluoranthene, and chrysene as sole sources of carbon and energy. Paenibacillus sp. strain KBC101 presented heterogeneous degradation profiles toward various aromatic compounds.  相似文献
