1. The adenosine-triphosphatase activity of rat-brain microsomes was measured between 0° and 37°. The stimulatory effect of Na+ plus K+ on the Mg2+-dependent adenosine-triphosphatase activity decreased sharply with decreasing temperature and became negligible at 0°. An Arrhenius plot drawn from the experimental data showed two discontinuities: one at about 6° and the other at about 20°. 2. The increment in activity induced by Na+ plus K+ was more sensitive to oligomycin at lower than at higher temperatures, but the opposite was observed for ouabain. The action of oligomycin showed a biphasic character, since below a certain concentration it caused slight activation of Na+-plus-K+-activated adenosine triphosphatase. 3. Where oligomycin increased the activity of the enzyme, it also enhanced the accumulation of an acid-precipitable phosphorylated compound formed through the transfer of the γ-phosphate group of [32P]ATP to the enzyme system. Stimulatory concentrations of oligomycin did not interfere with K+-mediated dephosphorylation of the intermediate, though high concentrations of oligomycin counteracted the effect of K+. 4. The temperature profile of K+-stimulated microsomal phosphatase qualitatively resembled that of microsomal adenosine triphosphatase.  相似文献

Homogenates from the salt-excreting leaves of the mangrove Avicennia nitida were subjected to differential centrifugation and investigated for adenosine triphosphatase activities. At pH 6.75 a salt stimulation with peaks at three different sodium to potassium ratios could be demonstrated above the activity due to Mg2+ ions. The stimulation by sodium and potassium depends on the ionic strength of the test medium, higher salt concentrations being inhibitory. The plant system seems thus more complicated than the animal activities. Technically, this means that a search for (Na+ + K+)-activated ATPases in plants should be performed with a close spacing of Na:K ratios at several constant levels of salt. Literature data on the transport of Na+ and K+ indicate that the physiological situation is rather complex in plants.  相似文献

Summary Sodium- and potassium-dependent adenosine triphosphatase (Na+–K+-ATPase) is demonstrated in the branchial heart of Sepia officinalis L. by biochemical, cytochemical and autoradiographical methods. The biochemical data indicate the presence of Na+–K+-ATPase, shown by potassium and magnesium dependency and inhibition by ouabain. Cytochemically and autoradiographically, the enzyme is localized in the sarcolemma of the muscle cells. The positive reaction of the transparent cells (type I cells) is due to activity of alkaline phosphatases. The dark cells (type II cells) react negatively. In addition to the Na+–K+-ATPase, a magnesium-activated adenosine triphosphatase (Mg2+-ATPase) and a bicarbonate-stimulated ATPase (HCO 3 - -ATPase) are localized in the mitochondria.This study was supported by the Deutsche Forschungsgemeinschaft and is part of the doctoral dissertation  相似文献

1. The Ca2+-activated adenosine triphosphatase of heavy meromyosin is maximally stimulated by lower relative molar concentrations of phenylmercuric acetate than are required with myosin. 2. Stimulation of the Ca2+-activated adenosine triphosphatase of both heavy meromyosin and myosin by thiol reagents is markedly affected by ionic strength, the effects being greater with the former than with the latter. In particular, N-ethylmaleimide strongly inhibits the Ca2+-activated adenosine triphosphatase of heavy meromyosin at ionic strength below about 0·2. 3. The precise behaviour of the thiol reagents at low ionic strength is slightly modified by the age of the heavy meromyosin and myosin preparations. 4. Stimulation of the Mg2+-activated adenosine triphosphatase of heavy meromyosin by thiol reagents is relatively insensitive to ionic strength. 5. The adenosine triphosphatases of heavy meromyosin and myosin activated by potassium chloride in the absence of bivalent activators are inhibited by thiol reagents over the range of ionic strength at which stimulation occurs in the presence of calcium chloride as activator. 6. The modifying effects of potassium chloride and sodium chloride are qualitatively different when heavy-meromyosin adenosine triphosphatase is stimulated with phenylmercuric acetate. No such difference is observed when the enzyme is stimulated with N-ethylmaleimide.  相似文献

The inhibitory effect of ouabain on (Na+ + K+)-activated ATPase (Mg2+-dependent, (Na+ + K+)-activated ATP phosphohydrolase, EC 3.6.1.3) obtained from rat brain microsomal fraction was re-examined using a modified method to estimate the inhibited reaction velocity. This method involves a preincubation of a ouabain-enzyme mixture in the presence of Na+, Mg2+ and ATP to bring the ouabain-enzyme reaction to near equilibrium. The (Na+ + K+)-activated ATPase reaction was subsequently started by the addition of a KCl solution.  相似文献

Summary Sodium- and potassium-dependent adenosine triphosphatase (Na+–K+-ATPase) has been demonstrated in the branchial heart appendage (pericardial gland) of Sepia officinalis L. by biochemical, cytochemical and autoradiographical methods. The biochemical data indicate the presence of Na+–K+-ATPase, judging from the potassium dependency and, with some restrictions, the inhibition by ouabain. Cytochemically and autoradiographically, the enzyme could be localized on the cytoplasmic surfaces of the lateral plasma membranes and the basal membrane infoldings (basal labyrinth) of the folded epithelium of the branchial heart appendage. The pdocytes of the peripheral zone of the organ reacted negatively. In addition to the Na+–K+-ATPase, a magnesium-activated adenosine triphosphatase (Mg2+-ATPase) was demonstrated in the folded epithelium, localized mainly in the mitochondria but also at the brush border and in the apical intercellular space, whereas a bicarbonate-stimulated ATPase (HCO 3 -ATPase) was present only in the mitochondria.This study was supported by the Deutsche Forschungsgemeinschaft  相似文献

SODIUM-potassium-activated, magnesium-dependent, adenosine triphosphatase (Na+, K+, Mg2+-ATPase) is widely accepted as an essential factor in sodium transport1 and observations on fish substantiate this view. There are concurrent increases, for example, of both Na+, K+, Mg2+-ATPase activity and osmoregulatory sodium transport2, in the intestinal mucosae3,4 and the gills3,5 of euryhaline teleosts during adaptation to seawater. Furthermore, the gills of stenohaline seawater teleosts, which actively secrete sodium, exhibit higher Na+, K+, Mg2+-ATPase activity than the gills of stenohaline freshwater teleosts, which do not actively secrete sodium3,5. Na+, K+, Mg2+-ATPase therefore seems to be important in maintaining tissue osmolarity well below that of seawater. It is disquieting to report therefore that Na+, K+, Mg2+-ATPase activity in the intestinal mucosae and gills of marine teleosts is inhibited by the organochlorine insecticide DDT. This observation may help to clarify the unexplained sensitivity of teleosts to DDT6.  相似文献

Washing of excised corn (Zea mays L., variety WF9×M14) root tissue is accompanied by an increase in (Mg2+ + K+)-stimulated adenosine triphosphatase. This is the adenosine triphosphatase described by Fisher, Hansen, and Hodges as positively correlated with ion accumulation rates. The increase in activity is confined to the microsomal fraction. A close parallel exists between increases in adenosine triphosphatase and phosphate absorption, and they respond similarly to inhibitors of RNA and protein synthesis. However, the amplitude of change is much smaller in adenosine triphosphatase. Possible reasons for this discrepancy are discussed.  相似文献

Purified plasma membranes from several transplantable human tumors exhibit very high Mg2+-dependent ATPase activities. Three types of Mg2+-dependent ATPases can be demonstrated: (1) an ouabain sensitive Na+, K+-ATPase, which is a minor component of the tumor plasma membrane ATPase, (2) a Mg2+-activated ATPase, which is a non-specific nucleoside triphosphatase, and (3) an ATPase activity stimulated by Na+ (or K+) alone. In three human melanomas, only the first two activities are found. In an astrocytoma and an oat cell carcinoma, all three activities are found. In the same two tumors, the plasma membrane Mg2+-ATPase is also stimulated by Con A. The relationship of these ATPases are discussed.  相似文献

The effect of certain biochemical parameters on transendothelial fluid transport has been studied. Cellular ATP level and (Na+ + K+)-activated as well as Mg2+-activated ATPase activities were measured by ultramicrotechniques using individual rabbit corneal endothelium after they had been subjected to in vitro perfusion with solutions fully supplemented or deficient singly or severally in glucose, adenosine and glutathione (GSH). With the complete medium, the transport system operates in vitro for approx. 6 h. Deletion of glucose alone, glucose and adenosine or glucose, adenosine and GSH brings about a cessation of fluid transport after 3.5 h, 2 to 2.5 h and 0.5 to 1 h, respectively. A marked decrease (62%) of the endothelial ATP level, however, occurs only when all metabolites are omitted. The favorable effect of GSH on transport activity is attributable to its capacity to sustain cellular ATP rather than to protect the functionality of (Na+ + K+)-activated ATPase. Adenosine, in the presence of GSH, maintains normal ATP levels and, additionally, exerts a protective effect on Mg2+-activated ATPase and possibly also on (Na+ + K+)-activated ATPase.  相似文献

Summary Na+, K+, Mg++-activated adenosine triphosphatase and K+, Mg++-activatedp-nitrophenyl phosphatase prepared from a membrane fraction of bovine cerebral cortex were studied with regard to the manner of their activation by phospholipids, using phosphatidyl serine, lysolecithin, monodecyl and didecyl phosphates. The kinetic and chromatographic studies suggested the following. (1) When the enzyme proteins bind the phospholipids in a proper ratio, they attain the optimum activation. (2) The binding causes a simple conversion of the enzymes from an inactive form to a fully activated form. (3) The lipids in both micellar form and molecular dispersion activate the enzymes. (4) Of the proteins contained in the enzyme preparation, only a group of proteins possessing the ATPase and the phosphatase activities bind phospholipids, and the amount of the bound lipids is limited.  相似文献

1. After removal of tropomyosin and troponin from the `natural'' actomyosin complex, the adenosine triphosphatase activity of the resulting `desensitized'' actomyosin is stimulated to the same extent by various bivalent cations with an ionic radius in the range 0·65–0·99å when tested at optimum concentration of the metal ion in the presence of 2·5mm-ATP at low ionic strength and pH7·6. Under identical conditions the adenosine triphosphatase activity of myosin alone is stimulated to an appreciable extent only by Ca2+ (ionic radius 0·99å). 2. Tropomyosin narrows the range of size of the stimulatory cations by inhibiting specifically the adenosine triphosphatase activity of `desensitized'' actomyosin when stimulated by Ca2+ or the slightly smaller Cd2+ (ionic radius 0·97å). Tropomyosin has no effect on the adenosine triphosphatase activity of `desensitized'' actomyosin when stimulated by the smaller cations, nor on the Ca2+-activated adenosine triphosphatase activity of myosin alone. 3. The adenosine triphosphatase activity of the `natural'' actomyosin system (containing tropomyosin and troponin) stimulated by the smallest cation, Mg2+ (ionic radius 0·65å), is low when the system is deprived of Ca2+ but high in the presence of small amounts of Ca2+. This sensitivity to Ca2+ seems to be a unique feature of the Mg2+-stimulated system. 4. The changes in specificity of the myosin adenosine triphosphatase activity in its requirement for bivalent cations caused by interaction with actin, tropomyosin and troponin primarily concern the size of the metal ions. The effects on enzymic properties of myofibrils due to tropomyosin and troponin can be demonstrated at low and at physiological ionic strength.  相似文献

1. Homogenates of neural lobes of bovine pituitary glands were fractionated by differential and density-gradient ultracentrifugation and the distribution of adenosine triphosphatase (ATPase) activity was studied. It was shown that all the activity was membrane-bound. 2. On the basis of ionic requirements the ATPase activity was grouped into three categories: (a) Mg2+-dependent, (b) Ca2+-dependent and (c) Mg2++Na++K+-dependent (ouabain-sensitive) ATPases. The activity in the absence of bivalent cations was negligible. The ratio between the activities of the three ATPases varied between the different subcellular fractions. 3. Preincubation of the subcellular fractions with deoxycholate increased the activity of the Mg2++Na++K+-dependent enzyme, whereas the Mg2+- and Ca2+-activated ATPases were either unaffected or slightly inhibited. Triton X-100 solubilized the Mg2+- and Ca2+-ATPases; however, the activity of the Mg2++Na++K+-ATPase was abolished by the concentration of Triton X-100 used. 4. All the subfractions displayed unspecific nucleotide triphosphatase activity towards GTP, ITP and UTP. These substrates inhibited the hydrolysis of ATP by all three ATPases. ADP also inhibited the ATPases. 5. Polyacrylamide-gel electrophoresis of extracts containing the Mg2+- and Ca2+-dependent ATPase activity solubilized by Triton X-100 revealed the presence of two enzymes; one activated by either Mg2+ or Ca2+ and the other activated only by Ca2+. 6. In sucrose density gradients the distribution of vasopressin was different from that of all three types of ATPases. It is therefore suggested that the neurosecretory granules do not possess ATPase activity.  相似文献

The adenosine triphosphatase (ATPase) system in worker honey-bee brains showed an increased activity of 57 per cent in Na+K+ATPase and 63 per cent in Mg2+ATPase from adult emergence to 7 days post-emergence. Mg2+ATPase activity remained about the same throughout the remainder of adult life, while Na+K+ATPase remained the same until the sixth week, when a decline occurred. The percentage mortality of the bees exceeded 90 per cent at the time of decline of Na+K+ATPase. The in vitro inhibition of Mg2+ATPase and Na+K+ATPase by 10 μM DDT was between 40 and 50 per cent and about 20 per cent, respectively. A somewhat greater sensitivity to DDT was determined in brains of older honey-bees.  相似文献

Golgi apparatus rich fractions from lactating bovine mammary gland had an Mg2+-dependent, Ca2+-stimulated adenosine triphosphatase. These Golgi apparatus fractions also accumulated Ca2+ in vitro. Accumulation of Ca2+ required ATP and could be abolished by treatment either with low concentrations of deoxycholate followed by ultrasound, or by heating at 100 °C for 10 min. The adenosine triphosphatase activity of Golgi apparatus was strongly stimulated by low concentrations of Ca2+ and moderately stimulated by high concentrations of K+. This activity was unaffected by Na+ and was not inhibited by ouabain. The pH optimum for the Mg2+-dependent hydrolysis of ATP was 7.5, the Km was 5 × 10−5 M and the activation energy was 6 000 calories/mole. This Mg2+-dependent adenosine triphosphatase activity was also found in rough endoplasmic reticulum, smooth microsomes and milk fat globule membrane, the latter membrane being derived directly from the apical plasma membrane. All of these membrane fractions had the ability to specifically accumulate Ca2+. Specific accumulation was highest with smooth microsomes and lowest with milk fat globule membrane with Golgi apparatus and rough endoplasmic reticulum being intermediate. These observations provide one plausible explanation for intracellular Ca2+ accumulation and secretion into milk. Further, these results help explain the ultrastructural observations of casein micelle formation in secretory vesicles elaborated by Golgi apparatus.  相似文献

Liver plasma membranes enriched in bile canaliculi were isolated from rat liver by a modification of the technique of Song et al. (J. Cell Biol. (1969) 41, 124–132) in order to study the possible role of ATPase in bile secretion. Optimum conditions for assaying (Na+ + K+)-activated ATPase in this membrane fraction were defined using male rats averaging 220 g in weight. (Na+ + K+)-activated ATPase activity was documented by demonstrating specific cation requirements for Na+ and K+, while the divalent cation, Ca2+, and the cardiac glycosides, ouabain and scillaren, were inhibitory. (Na+ + K+)-activated ATPase activity averaged 10.07 ± 2.80 μmol Pi/mg protei per h compared to 50.03 ± 11.41 for Mg2+-activated ATPase and 58.66 ± 10.07 for 5′-nucleotidase. Concentrations of ouabain and scillaren which previously inhibited canalicular bile secretion in the isolated perfused rat liver produced complete inhibition of (Na+ + K+)-activated ATPase without any effect on Mg2+-activated ATPase. Both (Na+ + K+)-activated ATPase and Mg2+-activated ATPase demonstrated temperature dependence but differed in temperature optima. Temperature induced changes in specific activity of (Na+ + K+)-activated ATPase directly paralleled previously demonstrated temperature optima for bile secretion. These studies indicate that (Na+ + K+)-activated ATPase is present in fractions of rat liver plasma membranes that are highly enriched in bile canaliculi and provide a model for further study of the effects of various physiological and chemical modifiers of bile secretion and cholestasis.  相似文献

A membrane fraction enriched with a magnesium-dependent, monovalent cation-stimulated ATPase was isolated from red beet (Beta vulgaris L.) storage roots by a combination of differential centrifugation, extraction with KI, and sucrose density gradient centrifugation. This fraction was distinct from endoplasmic reticulum, Golgi, mitochondrial, and possibly tonoplast membranes as determined from an analysis of marker enzymes. The ATPase activity associated with this fraction was further characterized and found to have a pH optimum of 6.5 in the presence of both Mg2+ and K+. The activity was substrate specific for ATP and had a temperature optimum near 40°C. Kinetics with Mg:ATP followed a simple Michaelis-Menten relationship. However the kinetics of K+-stimulation were complex and suggestive of negative cooperativity. When monovalent cations were present at 2.5 millimolarity, ATPase was stimulated in the sequence K+ > Rb+ > Na+ > Li+ but when the concentration was raised to 50 millimolarity, the sequence changed to K+ ≥ Na+ ≥ Rb+ > Li. The activity was not synergistically stimulated by combinations of Na+ and K+. The enzyme was insensitive to NaN3, oligomycin, ouabain, and sodium molybdate but sensitive to N,N′-dicyclohexylcarbodiimide, diethylstilbestrol, and sodium vanadate. Based on the similarity between the properties of this ATPase activity and those from other well characterized plant tissues, it has been concluded that this membrane fraction is enriched with plasma membrane vesicles.  相似文献

In an effort to determine the subcellular localization of sodium- and potassium-activated adenosine triphosphatase (Na+, K+-ATPase) in the pseudobranch of the pinfish Lagodon rhomboides, this tissue was fractionated by differential centrifugation and the activities of several marker enzymes in the fractions were measured. Cytochrome c oxidase was found primarily in the mitochondrial-light mitochondrial (M+L) fraction. Phosphoglucomutase appeared almost exclusively in the soluble (S) fraction. Monoamine oxidase was concentrated in the nuclear (N) fraction, with a significant amount also in the microsomal (P) fraction but little in M+L or S. Na+, K+-ATPase and ouabain insensitive Mg2+-ATPase were distributed in N, M+L, and P, the former having its highest specific activity in P and the latter in M+L. Rate sedimentation analysis of the M+L fraction indicated that cytochrome c oxidase and Mg2+-ATPase were associated with a rapidly sedimenting particle population (presumably mitochondria), while Na+, K+-ATPase was found primarily in a slowly sedimenting component. At least 75% of the Na+, K+-ATPase in M+L appeared to be associated with structures containing no Mg2+-ATPase. Kinetic properties of the two ATPases were studied in the P fraction and were typical of these enzymes in other tissues. Na+, K+-ATPase activity was highly dependent on the ratio of Na+ and K+ concentrations but independent of absolute concentrations over at least a fourfold range.  相似文献

PbCl2 in micromolar concentrations stimulates phosphorylation of electroplax microsomal protein in the absence of Na+. Other divalent cations showed little or no such effect. The (Mg2+ + Pb2+)- and (Mg2+ + Na+)-dependent membrane-bound protein kinase activities in electroplax particulate preparations exhibit properties in common, including their acid stability, ouabain sensitivity, ATP specificity, and molecular size. It is concluded that the (Mg2+ + Pb2+)-dependent phosphoprotein is part of the Na+-, K+-dependent adenosine triphosphatase [(NaK)ATPase]. The Pb2+-dependent product, in contrast to the Na+-dependent one, is insensitive to K+ and the hydrolysis of ATP is thus inhibited.  相似文献

Liver plasma membrane phospholipid distribution, protein conformation, and 5′-nucleotidase, Mg2+-adenosine triphosphatase and (Na+ + K+)-adenosine triphosphatase specific activities, were shown to depend on pituitary status and treatment with bovine growth hormone.In whole liver homogenates, hypophysectomy produced a decrease in the proportion of phosphatidyl serine, lysophosphatidyl choline, and phosphatidic acid and diphosphatidyl glycerol and an increased proportion of phosphatidyl ethanolamine. The phospholipid distribution in liver plasma membranes was the same for normal and hypophysectomized rats. Plasma membranes obtained from bovine growth hormone-treated hypophysectomized rats had approximately 50%, more phosphatidyl serine than membranes obtained from untreated hypophysectomized or normal rats.Plasma membranes from hypophysectomized rats had 75% of the 5′-nucleotidase, the same level of (Na+ + K+)-adenosine triphosphatase, and twice the Mg2+-adenosine triphosphatase of membranes from normal rats. Twelve hours after administration of bovine growth hormone to hypophysectomized rats, (Na+ + K+)-adenosine triphosphatase had almost doubled and Mg2+-adenosine triphosphatase decreased by 50%. 5′-Nucleotidase remained unchanged. Twenty-four hours after bovine growth hormone administration, both (Na+ + K+)-adenosine triphosphatase and 5′-nucleotidase had increased. Mg2+-adenosine triphosphatase was 23% of the baseline level of untreated hypophysectomized rats. Treatment for 3 days or 5 days increased the 5′-nucleotidase 2-fold.Circular dichroism spectra of liver plasma membranes isolated from hypophysectomized rats consistently showed greater negative ellipticity in the far ultraviolet range (250-190 nm) than those from normal rats or rats treated with bovine growth hormone.  相似文献
