Four cyclic derivatives of des-Arg9[Leu8]bradykinin have been obtained by classical methods of peptide chemistry. They are cyclo-(-X-Arg-Pro-Pro-Gly-Phe-Gly-Pro-Leu-), where X=Lys or none, and cyclo-(Y-Arg-Pro-Pro-Gly-Phe-Ser-Pro-Leu-), where Y= Lys or Orn. Peptide bonds have been formed by the pentafluorophenylester method, and cyclization has been carried out in a diluted dioxane solution with 40% yield. Subsequent cleavage of protecting groups was made by treatment with hydrogen fluoride. The products obtained were purified by droplet counter-current chromatography. These substances liberate histamine from the rat mast cells comparably to bradykinin and fail to produce myotripic and vascular effects.  相似文献

Two conformationally constrained enkephalin analogs were synthesized by substitution of cysteines in positions 2 and 5 and oxidative disulfide bond formation. In the guinea pig ileum assay the obtained cyclic analogs, [D-Cys2-L-Cys5]enkephalinamide and [D-Cys2-D-Cys5]enkephalinamide, showed potency ratios of 37.9 ± 0.8 and 73.3 ± 0.9, respectively, relative to [Met5]enkephalin. The extremely high potency of the analogs was shown to be a consequence of the conformational restrictions introduced by cyclization. Rat brain membrane binding studies with [3H]naloxone and [3H](D-Ala2, D-Leu5)enkephalin as radiolabels revealed a moderate preference of both analogs for μ-receptors over δ-receptors. Furthermore, the cystine-containing analogs were shown to be highly resistant to enzymatic degradation.  相似文献

A number of substituted bradykinin analogs were prepared in which the proline in position 3 was replaced by analogs of proline. All of the bradykinin analogs, with the exception of l-azetidine-2-carboxyl3-bradykinin showed significant ability to inhibit prolyl hydroxylase activity. Addition of an l-glutamyl residue to the amino terminus of 3,4-dehydro-l-prolyl3-bradykinin and trans-4-hydroxy-l-prolyl3-bradykinin resulted in competitive inhibitors of increased effectiveness with Ki, values approximately 10?4m. One of the peptides, l-3,4-dehydro-l-prolyl3-bradykinin, appeared to serve as a substrate for prolyl hydroxylase.  相似文献

N alpha-protected amino acid methylthiomethyl esters (MTM) were obtained in good yields under mild conditions using the "ButBr/Me2SO" reagent. Selective removal of the N-protecting group was achieved in HCl/anhydrous ethyl ether and the MTM ester hydrochlorides were successfully used in the synthesis of dipeptides.  相似文献

An efficient synthesis of a Pk trisaccharide with a functionalized side arm at the reducing end for conjugation to other molecules is presented. Construction of the Pk trisaccharide with a high alpha-selectivity was achieved in high yield by coupling a reactive galactosyl phosphite donor with a lactosyl acceptor.  相似文献

Cyclo-epsilon-(L-lysine1, glycine6-bradykinin) (CLGB) and cyclo-epsilon-kallidin have been synthesised in solution. To prepare linear precursors, fragment condensation (3 + 3 or 4) + 3 was used. Peptide bond formation, including cyclization, was carried out mainly through intermediate pentafluorophenyl esters. After purification on silicagel, protected cyclopeptides were obtained with a 50 to 60% yield. The protecting groups were eliminated by treatment with hydrogen fluoride in the presence of anisole. CLGB and CK were purified by droplet countercurrent chromatography and by reversed-phase HPLC, respectively.  相似文献

The conformation in aqueous solution of several alpha-aminoisobutyric acid (AIB)-containing analogs of bradykinin (BK) has been probed by complementary CD and 1H n.m.r. measurements. The conclusion reached is that substitution of AIB for Pro2 and/or Pro3 in BK stabilizes a degree of beta-turn conformation in the N-terminal tetrapeptide moiety of the resulting analogs. Changing the solvent from water to DMSO or TFE further enhances the contribution of particular hydrogen bonded structures to the time-averaged conformation of these peptides. Bradykinin and [AIB7]-BK adopt similar hydrogen bonded conformations in TFE, apparently with a contribution from a beta-turn involving their common Arg1-Pro2-Pro3-Gly4 moiety. The contrasting biological activities of BK and its AIB-analogs are considered in terms of the conformational analogy between the AIB-residue and cis' Pro and the propensity for a beta-turn at the N-terminus of the peptide.  相似文献

In this study, we evaluated 20 of our previously synthesized peptides on isolated rat uterus by Holton's procedure with minor modifications, and compared their activity with that assessed previously by their ability to inhibit vasodepressor response to exogenous bradykinin (BK) in conscious rats. We used [D-Arg(0), Hyp(3), Thi(5, 8), (D-Phe)(7)]BK, the B(2) antagonist of Vavrek and Stewart as a model when designing our analogs. We observed that, in the case of the rat uterus test, the activity of peptides modified by acylation of the N-terminus with various bulky groups depends substantially on the chemical character of the substituent. We also learned that, contrary to previous examples, acylation of the N-terminus of antagonists, which contain a sterically restricted fragment in the C-terminal part, may not improve their antagonistic potencies. Besides an improved characterization of a series BK analogs, our studies have provided new information on the structure-activity relationship, which in turn may be of value in the design of more potent and selective bradykinin antagonists. The results of our studies appear to support the hypothesis of others about the presence of different subtypes of B(2) receptors in rat uterus and blood vessels.  相似文献

The classical methods of peptide chemistry have been employed to synthesize loop-shaped derivatives of bradykinin and polisteskinin, Lys-Lys-Lys-[cyclo (9----1 epsilon), Lys1, Gly6]bradykinin and Lys-Lys-Lys-Leu-Arg-Gly[cyclo (9----1 epsilon)Lys1, Gly6] bradykinin. In the course of synthesis, the linear "tail" fragments were attached to partially deblocked cyclopeptide. Protective groups were removed by treating with hydrogen fluoride, the end products were purified using reversed-phase and ion exchange chromatography. Biological experiments in vivo have revealed that the two compounds elicit a prolonged hypotensive effect in rats which is characteristic of cyclic bradykinin analogues. With the latter compound, a decrease in arterial pressure is preceded by a brief hypertensive action. The loop-shaped analogues are slightly myotropic when applied to rat uterus preparations in vitro.  相似文献

A series of milnacipran analogs containing a heteroaromatic group were synthesized and studied as monoamine transporter inhibitors. Many compounds exhibited higher potency than milnacipran at NET and NET/SERT with no significant change in lipophilicity. For example, compound R-26f was about 10-fold more potent than milnacipran with IC(50) values of 8.7 and 26nM at NET and SERT, respectively.  相似文献

We have synthesized three analogs of the potent vasodilator peptide bradykinin, ArgProProGlyPhe SerProPheArg (BK), containing dehydrophenylalanine (ΔzPhe) in place of the phenylalanyl residues at positions 5 and/or 8. The analogs, [ΔzPhe5]BK, [ΔzPhe8]BK, and [ΔzPhe5,8]BK, were assayed for their effects on isolated smooth muscle tissues and on the systemic arterial blood pressure of rats. In these assays [ΔzPhe5]BK showed considerably high biological activities, particularly in terms of its blood pressure-lowering effects, being over 23 times more potent than BK when given intravenously. [ΔzPhe8]BK was less potent than BK and [ΔzPhe5,8]BK had effects comparable to those of BK. All three synthetic analogs appear to be more resistant than BK to enzymic degradation during passage through the pulmonary vascular bed.  相似文献

We describe a reversible immobilization method for carboxyl group containing haptens that makes the repeated usage of a BIAcore biosensor chip possible. Haptens which are immobilized according to the surface thiol method can be removed completely from the sensor surface again by a reducing step. In the first part of our study, analogues of the herbicides 2,4-dichlorophenoxyacetic acid and 2,4,5-trichlorophenoxyacetic acid were immobilized in succession to a biosensor surface of a BIAcore surface plasmon resonance instrument according to the thiol coupling method. Direct kinetic analysis of these ligands to a polyclonal anti-2,4-dichlorophenoxyacetic acid antibody were performed using these biosensor surfaces. In the second part of the study, different amounts of 2,4-dichlorophenoxyacetic acid were sequentially immobilized onto the same biosensor surface in order to generate a calibration plot for 2,4-dichlorophenoxyacetic acid. Using this plot, the quantitative detection of the herbicide down to a concentration of 0.1 microg/mL, the maximum admissible concentration of pesticides in drinking water, is possible.  相似文献

N-Acetyl-S-farnesyl-L-cysteine (AFC) is the minimal substrate for the enzyme isoprenylcysteine carboxyl methyltransferase (Icmt). A series of amide-modified farnesylcysteine analogs were synthesized and screened against human Icmt. From a 23-membered library of compounds, six inhibitors were identified and evaluated further. The adamantyl derivative 7c was the most potent inhibitor with an IC(50) of 12.4 microM.  相似文献

Three analogs of bradykinin (BK) containing l-3,4-dehydroproline (l-Δ3Pro) at positions 2, 3, or 7 of the BK sequence have been synthesized by the solid-phase technique and assayed for their effects on isolated smooth muscle tissues and on the systemic arterial blood pressure of rats. In these assays, (l-Δ3Pro2)-BK and (l-Δ3Pro3)-BK are as potent as BK itself, and each appears to be more resistant to enzymic degradation than BK itself during passage through the pulmonary circulation. (l-Δ3Pro7)-BK has approximately 25% of the potency of BK and appears to be more susceptible to pulmonary degradation than is BK. The circular dichroism (CD) spectra for the three analogs in water are similar in profile between 250 and 200 nm, although with different intensities, and more closely parallel the CD spectrum of BK in trifluoroethanol (TFE) than in water. The CD spectra in TFE show changes for all three analogs from the aqueous spectra. The TFE CD spectra of (l-Δ3Pro3)-BK and (l-Δ3Pro7)-BK are similar in profile to the spectrum of BK in TFE, whereas the spectrum of (l-Δ3Pro2)-BK is considerably different from that of BK. The spectroscopic changes can be interpreted in terms of a change in polyproline-like character or β-fold formation, presumably imposed by the greater rigidity of the dehydroproline ring.  相似文献
