The determination of the enzymatic activity of the yeasts has been applied to the identification of species, specially that ofCandida albicans. In order to know its usefulness in species of clinical interest, we have tested the commercial system API ZYM (Bio Mérieux) on 500 isolated strains of different organic samples, belonging to eight genera and twenty species. All the strains showed positivity to Phosphatase alcaline, Esterase (C4), Esterase lipase (C8), Leucine arylamidase and Phosphatase acid, and negativity to Lipase (C14), Trypsin, Chymotrypsin, -galactosidase, -glucoronidase, -manosidase and -fucosidase. Fourteen enzymatic activity patterns were obtained considering the substrates with variable results for the whole of the strains: Valine arylamidase, Cystine arylamidase, Naphthol-AS-BI-phosphohydrolase, -galactosidase, -glucosidase, -glucosidase and N-acetyl--glucosaminidase. In the majority of the species, the enzymatic profile did not have very specific results since it is usually shared by more than one species.C. albicans is that which presents the greatest number of enzymatic variations, some of these are similar to those of other common clinical species, such asCandida krusei, Candida parapsilosis andCandida tropicalis. The natural abundance of the nitrogen isotope 15, 15N, was analysed in leaves of 23 subarctic vascular plant species and two lichens from a tree-line heath at 450 m altitude and a fellfield at 1150 m altitude close to Abisko in N. Sweden, as well as in soil, rain and snow. The aim was to reveal if plant species with different types of mycorrhizal fungi also differ in their use of the various soil N sources. The dwarf shrubs and the shrubs, which in combination formed more than 65% of the total above-ground biomass at both sites, were colonized by ericoid or ectomycorrhizal fungi. Their leaf 15N was between–8.8 and–5.5 at the heath and between–6.1 and –3.3 at the fellfield. The leaf 15N of non- or arbuscular mycorrhizal species was markedly different, ranging from –4.1 to –0.4 at the heath, and from –3.4 to+2.2 at the fellfield. We conclude that ericoid and ectomycorrhizal dwarf shrubs and shrubs utilize a distinct N source, most likely a fraction of the organic N in fresh litter, and not complexed N in recalcitrant organic matter. The latter is the largest component of soil total N, which had a 15N of –0.7 at the heath and +0.5 at the fellfield. Our field-based data thus support earlier controlled-environment studies and studies on the N uptake of excised roots, which have demonstrated protease activity and amino acid uptake by ericoid and ectomycorrhizal tundra species. The leaves of ectomycorrhizal plants had slightly higher 15N (fellfield) and N concentration than leaves of the ericoids, and Betula nana, Dryas octopetala and Salix spp. also showed NO inf3 sup- reductase activity. These species may depend more on soil inorganic N than the ericoids. The 15N of non- or arbuscular mycorrhizal species indicates that the 15N of inorganic N available to these plants was higher than that of average fresh litter, probably due to high microbial immobilization of inorganic N. The 15N of NH inf4 sup+ -N was +12.3 in winter snow and +1.9 in summer rain. Precipitation N might be a major contributer in species with poorly developed root systems, e.g. Lycopodium selago. Our results show that coexisting plant species under severe nutrient limitation may tap several different N sources: NH inf4 sup+ , NO inf3 sup- and organic N from the soil, atmospheric N2, and N in precipitation. Ericoid and ectomycorrhizal fungi are of major importance for plant N uptake in tundra ecosystems, and mycorrhizal fungi probably exert a major control on plant 15N in organic soils.  相似文献

The sialidase of swine influenza A viruses of N1 and N2 subtypes, isolated from 1930 to 1992, was studied for substrate specificity with ganglio-series, lacto-series type II and GM3 gangliosides containing Neu5Ac2-3Gal, Neu5Gc2-3Gal and Neu5Ac2-6Gal linkages. All viral sialidases tested showed that the activity for hydrolysing substrates with Neu5Ac2-3Gal was higher than the activities with Neu5Gc2-3Gal and Neu5Ac2-6Gal linkages. When GM1b, GM3 and sialylparagloboside were used as substrates, the earliest strain (A/Wisconsin/15/30 H1N1, isolated in 1930) showed the activity ratio of Neu5Ac2-6Gal to Neu5Ac2-3Gal to be 0.13:0.2, and the ratio Neu5Gc2-3Gal/Neu5Ac2-3Gal to be 0.19:0.37, while those strains isolated from 1978 to 1992 exhibited ratios of 0.29:0.58 for Neu5Ac2-6Gal/Neu5Ac2-3Gal and 0.51:0.76 for Neu5Gc2-3Gal/Neu5Ac2-3Gal. Despite the large number of studies of nitrate metabolism in plants, it remains undetermined to what extent this key plant system is controlled by overall plant N nutrition on the one hand, and by the nitrate ion itself on the other hand. To investigate these questions, V max for nitrate uptake (high-affinity range), and nitrate reductase (NR) mRNA and activity, were measured in roots of N-limited barley (Hordeum vulgare L. cv. Golf) grown under conditions of constant relative addition of nitrate, with the seminal roots split between two culture compartments. The total amount of nitrate added per unit time (0.09·d-1) was distributed between the two root parts (subroots) in ratios of 1000, 982, 955, 9010, 8020, and 5050. These nitrate-addition ratios resulted in nitrate fluxes ranging from 0 to 23 mol nitrate·g-1 DW root·h-1, while the external nitrate concentrations varied between 0 and 1.2 M. The apparent V max for net nitrate uptake showed saturation-type responses to nitrate flux maintained during preceding growth. The flux resulting in half-maximal induction of nitrate uptake was approximately 4 mol nitrate·g-1 DW root·h-1, corresponding to an external nitrate concentration of 0.7 M. The activity of NR and levels of NR mRNA did not saturate within the range of nitrate fluxes studied. None of the parameters studied saturated with respect to the steady-state external nitrate concentration. At the zero nitrate addition — the 0%-root — initial uptake activity as determined in short-term 15N-labelling experiments was insignificant, and NR activity and NR mRNA were not detectable. However, nitrate uptake was rapidly induced, showing that the 0%-root had retained the capacity to respond to nitrate. Summary The total nitrogen and nitrate turnovers of three plots of plant communities typical of cattle resting sites in the Bavarian Alps were investigated. Annual net N mineralisation of the soil and N content of the above-ground phytomass at maximum development are low in the Senecio cordatus dominated community W at 1,450 m altitude (80 and 57 kg N/ha respectively) and highest in the nearly pure Rumex alpinus stand K (1,240 m) (156 and 148 kg N/ha). The subalpine mixed Rumex alpinus community S (1,790 m) is intermediate with low mineralisation but high N content in the aboveground parts (69 and 128 kg N/ha). In general the mineralisation values are below those from other comparable plant communities. The main component of N mineralisation is nitrification in all three plots.The nitrate content in the phytomass at full development increases from 0.1 g/m2 in W to 0.3 g/m2 in S and to 1.0 g/m2 in K. With one exception (Adenostyles alliariae) no species showed a real nitrate accumulation in the leaf laminae towards the end of the growing season.  相似文献

Fowl plague virus, strain Dutch, was metabolically labeled withd-[2-3H]mannose, or withd-[6-3H]glucosamine, and the small subunit (HA2; 0.8 mg in total) of the viral hemagglutinin was isolated by preparative sodium dodecylsulfate-polyacrylamide gel electrophoresis. After proteolytic digestion, the radioactive oligosaccharides were sequentially liberated from the glycopeptides by treatment with different endo--N-acetylglucosaminidases and with peptide:N-glycosidase or, finally, by hydrazinolysis. In this manner, four groups of glycans could be obtained by consecutive gel filtrations and were subfractionated by HPLC. The structures of the individual oligosaccharides were analyzed by micromethylation, by acetolysis or by digestion with exoglycosidases. Summary The eye lens-crystallins in cow and chicken are encoded by a family of at least six genes. In order to assess the distribution of the corresponding genes among other vertebrates we hybridized -crystallin sequences (A2, A3/A1, A4, B1, B2, B3), isolated from a bovine lens cDNA library, to Southern blots on whichEcoR1-digested chromosomal DNA was blotted from different vertebrate species. These included human, chimpanzee, calf, rat, pigeon, duck, monitor lizard, toad, trout, and lamprey. Positive hybridization signals were found in the representatives of virtually all classes of vertebrates. The basic B-crystallins gave hybridization signals in more species than the acidic A ones. In monitor lizard and toad the weakest hybridization signals for basic crystallin probes were found. For acidic crystallin probes the distribution pattern was more simple; among cold-blooded vertebrates a signal for A2 was found in trout and lamprey, for A4 in trout, and for A3/A1 only in toad. The results demonstrate that the duplications leading to the -crystallin gene family occurred before or during the earliest stages of vertebrate evolution.  相似文献

The food-web structure of the Arctic deep Canada Basin was investigated in summer 2002 using carbon and nitrogen stable isotope tracers. Overall food-web length of the range of organisms sampled occupied four trophic levels, based on 3.8 trophic level enrichment (15N range: 5.3–17.7). It was, thus, 0.5–1 trophic levels longer than food webs in both Arctic shelf and temperate deep-sea systems. The food sources, pelagic particulate organic matter (POM) (13C=–25.8, 15N=5.3) and ice POM (13C=–26.9, 15N=4.1), were not significantly different. Organisms of all habitats, ice-associated, pelagic and benthic, covered a large range of 15N values. In general, ice-associated crustaceans (15N range 4.6–12.4, mean 6.9) and pelagic species (15N range 5.9–16.5, mean 11.5) were depleted relative to benthic invertebrates (15N range 4.6–17.7, mean 13.2). The predominantly herbivorous and predatory sympagic and pelagic species constitute a shorter food chain that is based on fresh material produced in the water column. Many benthic invertebrates were deposit feeders, relying on largely refractory material. However, sufficient fresh phytodetritus appeared to arrive at the seafloor to support some benthic suspension and surface deposit feeders on a low trophic level (e.g., crinoids, cumaceans). The enriched signatures of benthic deposit feeders and predators may be a consequence of low primary production in the high Arctic and the subsequent high degree of reworking of organic material.  相似文献

Two sets of backcross matings were performed to test for linkage between genes coding for the Ia-like antigens (Ia) and the B erythrocyte antigens (Ea-B) of the chicken. Evidence is presented which indicates that the la antigens are determined by a single codominant locus and that theEa-B and Ia loci are on the same chromosome. Failure to detect a single recombinant between theEa-B and Ia loci out of 208 progeny suggests close linkage of the two genes with a map distance of up to about 2 centimorgans. The Ia genes are thus included in theB major histocompatibility complex of the chicken.  相似文献

Summary Two specific -N-acetylglucosaminyltransferases involved in the branching and elongation of mucin oligosaccharide chains, namely, a 1,6 N-acetylglucosaminylsaminyltransferase that transfers N-acetylglucosamine from UDP-N-acetylglucosamine to Gal3GalNAc-Mucin to yield Gal3(GlcNAc6)GalNAc-Mucin and a 3-N-acetylglucosaminyl transferase that transfers N-acetylglucosamine from UDP-N-acetylglucosamine to Gal3(GlcNAC6)GalNAc-mucin to yield GlcNAc3Gal3 (GlcNAc6)GalNAc-Mucin were purified from the microsomal fraction of swine trachea epithelium. The 1,6-N-acetylglucosaminyltransferase was purified about 21,800-fold by procedures which included affinity chromatography on DEAE columns containing bound asialo Cowper's gland mucin glycoprotein with Gal1,3GalNAc side chains. The apparent molecular weight estimated by gel filtration was found to be about 60 Kd. The purified enzyme showed a high specificity for Gal1,3GalNAc chains and the most active substrates were mucin glycoproteins containing these chains. The apparent Km of the 6-glucosaminyltrans-ferase for Cowper's gland mucin glycoprotein containing Gal1,3GalNAc chains was 0.53 µM; for UDP-N-acetylglucosamine, 12 µM; and for Gal 1,3GalNAc NO2ø, 4 mM. The activity of the 6-glucosaminyltransferase was dependent on the extent of glycosylation of the Gal3GalNAc chains in Cowper's gland mucin glycoprotein.The best substrate for the partially purified 3-Glucosaminyltransferase was Cowper's gland mucin glycoprotein containing Gal1,3(GlcNAc6)GalNAc side chains. This enzyme showed little or no activity with intact sialylated Cowper's gland mucin glycoprotein or derivatives of this glycoprotein containing GalNAc or Gal1,3GalNAc side chains.The radioactive oligosaccharides formed by these enzymes in large scale reaction mixtures were released from the mucin glycoproteins by treatment with alkaline borohydride, isolated by gel filtration on Bio-Gel P-6 and characterized by methylation analysis and sequential digestion with exoglycosidases. The oligosaccharide products formed by the 6- and 3-glucosaminyltransferases were shown to be Gal3(GlcNAC6) GalNAc and GlcNAc3 Gal3(GlcNAC6)GalNAc respectively.Taken collectively, these results demonstrate that swine trachea epithelium contains two specific N-acetylglucosaminyltransferases which catalyze the initial branching and elongation reactions involved in the synthesis of O-linked oligosaccharide chains in respiratory mucin glycoproteins. The first enzyme a 6-glucosaminyltransferase converts Gal3GalNAc chains in mucin glycoproteins to Gal3(GlcNAc6)GalNAc chains. This product is the substrate for a second 3-glucosaminyltransferase which converts the Gal3(GlcNAc6)GalNAc chains to GlcNAc3Gal(GlcNAc6)GalNAc chains in the glycoprotein. Direct gradient analysis was used to relate the structure of mangrove communities to soil nutrient gradients. The predominant cations in the alluvial soils were magnesium and calcium, the values ranging from 8.6±0.9 to 24.6±2.0me per 100g. Organic carbon was high in the soils, ranging from 3.5% to 10.4%. All soil nutrients varied seasonally, in response to wet and dry periods of the climate. Correlation of species with soil nutrients indicated the nutrient status of the soils to be best defined in terms of calcium for the A stratum (species > 3m tall), calcium/postassium for the B stratum (species 1–3m tall) and magnesium for the C stratum (species < 1m tall). Ecological group classification shows that Avicennia africanaand Nypa fruticansoccurred almost exclusively at the highest calcium values of 20.8 and 17.6me per 100g, respectively. Acrostichum aureumand Sesuvium portulacastrumare insensitive to the magnesium gradient. The analysis revealed an overlapping range of occurrences for most of the species although with varying ecological optima along the gradient.  相似文献

The general attributes of ecosystems are examined and a naturally occurring reference ecosystem is established, comparable with the isolated system of classical thermodynamics. Such an autonomous system with a stable, periodic input of energy is shown to assume certain structural characteristics that have an identifiable thermodynamic basis. Individual species tend to assume a state of least dissipation; this is most clearly evident in the dominant species (the species with the best integration of energy acquisition and conservation). It is concluded that ecosystem structure results from the antagonistic interaction of two nearly equal forces. These forces have their origin in the Principle of Most Action (least dissipation or least entropy production) and the universal Principle of Least Action. Most action is contingent on the equipartitioning of the energy available, through uniform interaction of similar individuals. The trend to Least action is contingent on increased dissipation attained through increasing diversity and increasing complexity. These principles exhibit a basic asymmetry. Given the operation of these opposing principles over evolutionary time, it is argued that ecosystems originated in the vicinity of thermodynamic equilibrium through the resonant amplification of reversible fluctuations. On account of the basic asymmetry the system was able to evolve away from thermodynamic equilibrium provided that it remained within the vicinity of ergodynamic equilibrium (equilibrium maintained by internal work, where the opposing forces are equal and opposite).At the highest level of generalization there appear to be three principles operating: i) maximum association of free-energy and materials; ii) energy conservation (deceleration of the energy flow) through symmetric interaction and increased homogeneity; and iii) the principle of least action which induces acceleration of the energy flow through asymmetrical interaction. The opposition and asymmetry of the two forces give rise to natural selection and evolution.  相似文献

The present study was undertaken to determine the effects of AT1 receptor blockade which occurred in response to losartan, on the extracellular matrix (ECM) degradation process in the Bio 14.6 (n = 12) and Bio 53.58 (n = 12) strains which are referred as models of hypertrophic and dilated cardiomyopathy, respectively. The administration of losartan (30 mg/kg/day) in hamsters from 10–20 weeks of age reduced the accumulation of the left ventricular collagen matrix in both of the Bio 14.6 and the Bio 53.58 strains. According to the RTPCR, the levels of mRNA for matrix metalloproteinase (MMP) and the tissue inhibitor of MMP (TIMP) were examined. MMP1, 2, 3, and 9 were more enhanced in both myopathic strains than in the control F1 strains. With losartan, the levels of MMP1, 2, 9, TIMP1 and 2 decreased in the both strains but those for MMP3 did not in Bio 14.6 strains. TIMP3 and 4 mRNA levels did not change in any of the experimental hamsters, whether treated or untreated with losartan. The Western blots also showed similar observations in the both strains as seen in mRNA expressions although MMP2 in the Bio 53.58 strains did not differ between treated and untreated with losartan. Although losartan has an inhibitory effect on collagen accumulation in the development of cardiomyopathy, MMPs (1, 2, 9) and TIMPs (1, 2) seem to be susceptible to responding to losartan in Bio cardiomyopathic hamsters.  相似文献

As part of a program to explore the structural requirement of N-glycans in the carbohydrate-mediated biological interactions, N-linked pentasaccharide core structure was stereochemically modified in terms of glycosidic linkage. Three isomers, -D-Man-(13)-[-D-Man-(16)]--D-Man-(14)--D-GlcNAc-(14)--D-GlcNAc-L-Asn, -D-Man-(13)-[-D-Man-(16)]--D-Man-(14)--D-GlcNAc-(14)--D-GlcNAc-L-Asn, and -D-Man-(13)-[-D-man-(16)]--D-Man-(14)--D-GlcNAc-(14)--D-GlcNAc-L-Asn, were synthesized. Nitrate uptake in Chlorella saccharophila (Krüger) Nadson was found to be stimulated by blue light, leading to a doubling of the rate. In the presence of background red light (300 mol photons · m-2 · s-1), only 15–20 mol photons · m-2 · s-1 of blue light was sufficient to saturate this increased uptake rate. Incubation of Chlorella cells with anti-nitrate-reductase immunoglobulin-G fragments inhibited blue-light stimulation. However, ferricyanide (10 M) doubled and dithiothreitol (100 M) inhibited the stimulatory effect of blue light. Among the protein-kinase inhibitors used, only staurosporine (10 M) prevented the blue-light stimulation. Phosphatase inhibitors were without effect and sodium vanadate totally inhibited nitrate uptake, pointing to an involvement of the plasma-membrane ATPase. Preincubation of the cells with calmodulin antagonists or calcium ionophores did not significantly reduce blue-light stimulation of nitrate uptake. Conclusion Even a cursory look at the ethnographic literature on other Northwest Coast societies reveals some striking similarities with the Tlingit way of conceptualizing aristocrats as special persons. Thus the Kwakiutl referred to their chiefs as real or complete people, who were heavier than commoners. The Coast Tsimshian called their highest aristocrats real or ripe persons, in contrast to the low-ranking ones, who were described as unhealed or green. The Coast Tshimshian also referred to their chiefs as strong, heavy, and solid like a rock. The neighboring Gitksan contrasted the chiefs, described as people who were good and clean and stayed put, with the commoners, who were said to be dirty, ignorant, and always moving around. Because spirits of the dead liked to return to persons who were clean and showed respect by giving away wealth and feasts, there was considerable moral and practical pressure on the aristocrats to remain pure, train knowledgeable and clean heirs, and continue potlatching. Finally, among the Haida, rank was tied to a wider system of symbolic classification, associating aspects of food, space, clothing, ritual pollution and the ethic of industry with attributes of seniority.While some of the symbolic associations of aristocratic status are culture specific, others are present in several, if not all, of the NWC cultures. What we need is a comparative symbology of aristocratic status, which would combine the reanalysis of the existing ethnographic data with the introduction of some new materials that can still be obtained in the field. A sub-adult grey seal was caught and tagged with a satellite relay data logger at Tihällan (61°2800N, 17°2540E), Sweden, on 21 August 1992. We tracked it for 115 days during which time we recorded 841 locations, data on 3556 dives and 80 haulout periods. It travelled 730 km in the first 12 days to the southern Archipelago Sea. It stayed most of the remaining time within 25 km of a haulout site, with a major exception of an 8-day trip of 520 km. Dive shape and swimming activity were used to distinguish dive types. Square-shaped foraging dives were more common over the midday period. Haulout periods were more common during the night.  相似文献

Summary Four types of striated muscle fibers with distinctive ultrastructure were defined in the Atlantic hagfish (Myxine glutinosa, L.): white, intermediate, and red fibers of m. parietalis, and red fibers of m. craniovelaris.White fibers are thick, contain very few mitochondria and fat vacuoles, and possess distinct and separate myofibrils with thin Z-disks and distinct M-lines. Intermediate fibers are thinner, possess largely similar myofibrils that often are even better separated due to a higher content of fat vacuoles and especially mitochondria and glycogen granules. Red fibers of m. parietalis contain large amounts of mitochondria, fat vacuoles, and glycogen granules. Their myofibrils possess M-lines, and although branching more, the myofibrils of red fibers conform with a Fibrillenstruktur pattern like those of white and intermediate fibers. Red fibers of m. craniovelaris are very thin, possess many smaller fat vacuoles, and large amounts of mitochondria and glycogen granules. The myofibrils are significantly thinner than in m. parietalis fibers, run as quite independent well separated units, possess thicker Z-disks, and lack M-lines. Summary 1. Anaerobic amino-acid breakdown in a garden soil was accompanied by the formation of large quantities of volatile fatty acid and ammonia (plus indole in the case of tryptophan).2. Hydroxyproline, leucine, methionine, phenylalanine, proline, threonine, tryptophan, tyrosine, and valine were found to be resistant and decomposed more slowly than did alanine, arginine, aspartic acid, cystine, glutamic acid, glycine, histidine lysine, and serine. Casein hydrolysate was rapidly and completely deaminated.3. The slow rates of breakdown of the resistant acids when tested singly could not be attributed either to the effects of adsorption on the soil surface or to elimination of an essential Stickland reaction.4. The effect of glucose and of nitrate on anaerobic amino-acid metabolism have been tested. With the exception ofdl-methionine nitrate acted as an hydrogen acceptor in the anaerobic deamination of the resistant acids, glucose had no effect on their anaerobic metabolism.  相似文献
