Eucalyptus camaldulensis Dehnh. seedlings inoculated with Pisolithus tinctorius (Pers.) Coker & Couch and Thelephora terrestris Ehrl. per Fr. were grown in well watered soil (s –0.03 MPa) or subjected to a long-term soil water stress of up to –1.0 MPa over 13-week period in a glasshouse. After 13 weeks, all seedling containers were watered to field capacity and then water was withheld from the E. camaldulensis seedlings to induce a short-term drought. Diurnal measurements of seedling photosynthesis rate (A), leaf stomatal conductance (g) and leaf water potential (p) were completed before, during, and after the short term drought. Although they were growing in an equal soil volume, photosynthesis rate (A), leaf stomatal conductance and leaf water potential (p) of larger seedlings with P. tinctorius ectomycorrhizae were similar to those of smaller seedlings colonized with T. terrestris during the short-term drought period. Seedlings inoculated with Pisolithus tinctorius maintained higher photosynthesis rates over the course of the short-term drought. Thus, P. tinctorius ectomycorrhizae appear to be more efficient than those of T. terrestris in assisting seedlings to maintain gas exchange and photosynthesis under limited soil moisture conditions.  相似文献

Summary The technique of15N isotope dilution was used to verify that nitrogen was fixed and transferred to the plant byKlebsiella pneumoniae strain Pp in association withPoa pratensis orTriticum aestivum. Surface sterilized, sprouting seeds were inoculated withK. pneumoniae and grown in sand in modified Leonard jars. Potassium nitrate enriched with15N was used to provide N concentrations ranging from 10–40 mg Nl–1 nutrient solution. After 10–18 weeks the shoots and roots were analyzed separately for dry matter, N content, total N, and atom %15N excess. The acetylene reduction technique was used to test for the presence of N2-fixing organisms on the roots. The data from15N isotope dilution demonstrated that up to 33.8% of N in the shoots ofP. pratensis and 15.9% in those ofT. aestivum were derived from associative N2 fixation byK. pneumoniae. In most experiments the dry matter yield, N content, and total N yield of the shoots ofP. pratensis were increased byK. pneumoniae inoculation, whereas inoculation had no significant effect on the dry matter yield, N content or total N of the shoots ofT. aestivum.  相似文献

A range of known biocontrol or plant growth-stimulating species ofTrichoderma orGliocladium were grown on peat-bran substrate to yield between 5×107–3×1010 colony forming units (cfu's)g–1 substrate after 14 days growth. Inocula were incorporated into peat:sand potting compost infested withRhizoctonia solani to give 7–8 × 104 cfu's of antagonist g–1 compost and assessed for biological control activity using lettuce seedlings. Six of the eight antagonists decreased daming-off and three of these consistently increased yield in comparison withR. solani treatment alone.Subsequently, peat-bran inoculum ofT. harzianum isolate TH1 was incorporated at 0.5% w/v intoR. solani infested potting compost. Both autoclaved and nonautoclaved inoculum ofT. harzianum TH1 decreased disease and increased yield. Incorporation of ethyl acetate-extracted autoclaved inoculum ofT. harzianum TH1 resulted in similar levels of biocontrol and improved plant growth as did incorporation of nonautoclaved and autoclavedT. harzianum TH1 inoculum. The need to standardize inocula and controls is emphasized.  相似文献

Castanopsis fissa Rehd. & Wils. is widely distributed from the tropics to the temperate regions of China and Japan and is an important forest component in Hong Kong. Pot-grown C. fissa seedlings inoculated with vegetative mycelial inocula of seven ectomycorrhizal fungi for 20 weeks were analysed for growth performance and mineral nutrient uptake of N, P, K and Ca. Shoot growth stimulation in all fungal treatments generally occurred in the first 4–8 weeks of seedling development. Uptake of P was generally enhanced by all fungi inoculated. Seedlings inoculated with Pisolithus tinctorius (Pers.) Coker and Couch and Cenococcum geophilm (Sow.) Fredinard et Winge, which colonized 22% and 33% of roots respectively, exhibited growth stimulation. The results indicate that P. tinctorius and C. geophilum are suitable for use in large-scale nursey inoculation.  相似文献

Lignosulfonate (LS) is a lignin-based polymer obtained as a by-product from paper industry, which may have potential as an amendment with macronutrients. We studied effects of LS on the interaction between Scots pine (Pinus sylvestris L.) seedlings and hypocotyl cuttings and the ectomycorrhizal (ECM) fungusPisolithus tinctorius (Pers.) Coker and Couch. The experiments were performed in vitroon the MMN agar medium containing Fe–LS chelate at the concentrations of 0, 5, 10 and 25 mg/L. Inoculation with P. tinctoriusincreased root growth of the seedlings. Fe–LS enhanced P. tinctorius induced formation of lateral roots and had a dose-dependent positive effect on the establishment of mycorrhizas on the seedlings. The growth of the fungal mycelium was improved by Fe–LS, which might cause faster and more intensive contact with the roots and, thus, better root growth and mycorrhiza formation. P.tinctorius enhanced also adventitious root formation and subsequent root growth of the hypocotyl cuttings but without any synergistic effect with Fe–LS. Our study with P. tinctorius and Scots pine in vitro indicates that a low-cost by-product Fe–LS, obtained from paper industry, may be a potential tool to improve the efficiency of fungal inoculations, thus, facilitating the early interaction between an ECM fungus and host seedling.  相似文献

Biofilms are a natural form of cell immobilization that result from microbial attachment to solid supports. Biofilm reactors with polypropylene composite-supports containing up to 25% (w/w) of various agricultural materials (corn hulls, cellulose, oat hulls, soybean hulls or starch) and nutrients (soybean flour or zein) were used for ethanol production. Pure cultures ofZymomonas mobilis, ATCC 31821 orSaccharomyces cerevisiae ATCC 24859 and mixed cultures with either of these ethanol-producing microorganisms and the biofilm-formingStreptomyces viridosporus T7A ATCC 39115 were evaluated. An ethanol productivity of 374g L–1 h–1 (44% yield) was obtained on polypropylene composite-supports of soybean hull-zein-polypropylene by usingZ. mobilis, whereas mixed-culture fermentations withS. viridosporus resulted in ethanol productivity of 147.5 g L–1 h–1 when polypropylene composite-supports of corn starch-soybean flour were used. WithS. cerevisiae, maximum productivity of 40 g L–1 h–1 (47% yield) was obtained on polypropylene composite-supports of soybean hull-soybean flour, whereas mixed-culture fermentation withS. viridosporus resulted in ethanol productivity of 190g L–1 h–1 (35% yield) when polypropylene composite-supports of oat hull-polypropylene were used. The maximum productivities obtained without supports (suspension culture) were 124 g L–1 h–1 and 5 g L–1 h–1 withZ. mobilis andS. cerevisiae, respectively. Therefore, forZ. mobilis andS. cerevisiae, ethanol productivities in biofilm fermentations were three- and eight-fold higher than suspension culture fermentations, respectively. Biofilm formation on the chips was detected by weight change and Gram staining of the support material at the end of the fermentation. An obligate fungus Albugo candida (Pers. ex Lév.) Ktze. (race unidentified) was successfully grown on host callus tissues of Brassica juncea cv. Varuna. Of the various type of diseased explants used, young (green) hypertrophied inflorescence axis bearing non-erumpent zoosporangial blisters allowed the fungus to multiply asexually over the host calli on modified MS-medium (Murashige and Skoog, 1962). The dual cultures were maintained up to 6–8 subcultures without loss of viability of zoosporangia on MS-medium supplemented with 10.0 mg L–1 IBA, 0.05 mg L–1 kinetin, 25.0 mg L–1 AA, 1.0 mg L–1 biotin, 1.0 mg L–1 thiamine-HCl and 1.0 g L–1 casein hydrolysate. The fungus grew only on the callus cells and not axenically on the medium. Summary The symbiotic association of the water fernAzolla with the blue-green algaAnabaena azollae can fix 30–60 kg N ha–1 per rice cropping season. The value of this fixed N for rice production, however, is only realized once the N is released from theAzolla biomass and taken up by the rice plants. The availability of N applied asAzolla or as urea was measured in field experiments by two15N methods. In the first,Azolla caroliniana (Willd.) was labelled with15N in nutrient solution and incorporated into the soil at a rate of 144 kg N ha–1. The recovery ofAzolla-N in the above ground parts of rice [Oryza sativa (L) cv. Nucleoryza] was found to be 32% vs. 26% for urea applied at a rate of 100 kg N/ha; there was no significant difference in recovery. In the second, 100 kg N/ha of15N-urea was applied separately or in combination with either 250 or 330 kg N ha–1 of unlabelledAzolla. At the higher rate, the recovery ofAzolla-N was significantly greater than that of urea. There was a significant interaction when both N sources were applied together, which resulted in a greater recovery of N from each source in comparison to that source applied separately. Increasing the combined urea andAzolla application rate from 350 kg N ha–1 to 430 kg N ha–1 increased the N yield but had no effect on the dry matter yield of rice plants. The additional N taken up at the higher level of N application accumulated to a greater extent in the straw compared to the panicles. Since no assumptions need to be made about the contribution of soil N in the method using15N-labelledAzolla, this method is preferable to the15N dilution technique for assessing the availability ofAzolla-N to rice. Pot trials usingAzolla stored at –20°C or following oven-drying showed that both treatments decreased the recovery of N by one third in comparison to freshAzolla.  相似文献

The production of aboveground tissue of three alder species (Alnus crispa (Ait.) Pursh,A. rugosa (Du Roi) Spreng. andA. glutinosa (L) Gaertn.) on four sites ranged from 0.4 t ha–1 yr–1 to 4.0 t ha–1 yr–1 after four growing seasons. Large differences were observed among the four sites studied and among species. Soil nutrient levels affected the biomass production and foliar symptoms of P and Mg deficiency occurred withA. crispa andA. rugosa. Because of their poor aboveground biomass production (0.4–1.4 t ha–1 yr–1),A. crispa andA. rugosa should be used mainly as nurse trees. For its higher potential for biomass production (up to 4.0 t ha–1 yr–1), and its apparent higher ability to use P and Mg on deficient sites,A. glutinosa should be used preferably toA. crispa andA. rugosa for the production of biomass.  相似文献

Summary Eight thermophilic fungi were tested for production of mannanases and galactanases. Highest mannanase activities were produced byTalaromyces byssochlamydoides andTalaromyces emersonii. Mannanases from all strains tested were induced by locust bean gum except in the case ofThermoascus aurantiacus, where mannose had a greater inducing effect. Locust bean gum was also the best inducer of -mannosidase and galactanase except in the case ofT. emersonii where galactose was a better inducer of both these enzymes. Highest mannanase activity was produced byTalaromyces species when peptone was used as nitrogen source whereas sodium nitrate promoted maximum production of this enzyme byThielavia terrestris andT. aurantiacus. The pH optima of mannanases from the thermophilic fungi were in the range 5.0–6.6 and contrasted with the low pH optimum (3.2) of the enzyme fromAspergillus niger. Galactanases had pH optima in the range 4.3–5.8. The mannanase fromT. emersonii and the galactanase fromT. terrestris were most thermostable, each retaining 100% activity for 3 h at 60°C.  相似文献

Continuous ethanol fermentations were performed in duplicate for 60 days withZymomonas mobilis ATCC 331821 orSaccharomyces cerevisiae ATCC 24859 in packed-bed reactors with polypropylene or plastic composite-supports. The plastic composite-supports used contained polypropylene (75%) with ground soybean-hulls (20%) and zein (5%) forZ. mobilis, or with ground soybean-hulls (20%) and soybean flour (5%) forS. cerevisiae. Maximum ethanol productivities of 536 gL–1 h–1 (39% yield) and 499 gL–1 h–1 (37% yield) were obtained withZ. mobilis on polypropylene and plastic composite-supports of soybean hull-zein, respectively. ForZ. mobilis, and optimal yield of 50% was observed at a 1.92h–1 dilution rate for soybean hull-zein plastic composite-supports with a productivity of 96gL–1h–1, whereas with polypropylene-supports the yield was 32% and the productivity was 60gL–1h–1. With aS. cerevisiae fermentation, the ethanol production was less, with a maximum productivity of 76gL–1h–1 on the plastic composite-support at a 2.88h–1 dilution rate with a 45% yield. Polypropylene-support bioreactors were discontinued due to reactor plugging by the cell mass accumulation. Support shape (3-mm chips) was responsible for bioreactor plugging due to extensive biofilm development on the plastic composite-supports. With suspensionculture continuous fermentations in continuously-stirred benchtop fermentors, maximum productivities of 5gL–1h–1 were obtained with a yield of 24 and 26% withS. cerevisiae andZ. mobilis, respectively. Cell washout in suspensionculture continuous fermentations was observed at a 1.0h–1 dilution rate. Therefore, for continuous ethanol fermentations, biofilm reactors out-performed suspension-culture reactors, with 15 to 100-fold higher productivities (gL–1h–1) and with higher percentage yields forS. cerevisiae andZ. mobilis, respectively. Intergeneric hybridizations were made betweenT. elongatum, and twoPsathyrostachys and fiveLeymus species. The seed set obtained onT. elongatum ×Leymus hybrids ranged from 5.65% to 20.00%, depending onLeymus species. The seed set obtained onT. elongatum ×Psathyrostachys hybrids ranged from 16.07% to 19.70%. Meiotic pairing at metaphase-I in JN diploid hybrids ofT. elongatum ×Psathyrostachys species revealed a very low level homology between the basic J and N genomes, and further demonstrated that the two genomes are quite diverged. Chromosome pairing in theT. elongatum ×Leymus secalinus hybrid averaged 15.19 univalents + 2.62 rod bivalents + 0.26 ring bivalents + 0.02 trivalents, suggesting that the partial Je chromosomes ofT. elongatum has homology withLeymus secalinus genomes.L. secalinus might have 3–4 chromosomes originating from Je genome.  相似文献

Changes in protein biosynthesis were examined during the early stages of differentiation of Eucalyptus grandis-Pisolithus tinctorius ectomycorrhizas by two-dimensional polyacrylamide gel electrophoresis of 35S-labelled proteins. Three distinct isolates of P. tinctorius Coker & Couch were chosen based on the rate of ectomycorrhizal formation (i.e. infectivity) with E. grandis W. Hill ex Maiden. The isolate H506 was not able to induce mycorrhiza, isolate 441 showed moderate infectivity and isolate H2144 exhibited a very high infectivity. Mycorrhiza were produced in vitro in a system where seeds were germinated in the presence of fungal mycelium and exudates. The non-mycorrhizal isolate caused no changes in root protein biosynthesis as analyzed by two-dimensional polyacrylamide gel electrophoresis, whereas drastic alterations in protein biosynthesis were observed from initial contact with the aggressive mycobionts. During mycorrhizal development, there was a marked inhibition of plant polypeptides synthesis, enhanced accumulation of some fungal polypeptides and the emergence of symbiosis-specific polypeptides, the so-called ectomycorrhizins. The major changes were observed in a group of fungal acidic polypeptides (apparent molecular weight 28–32 kDa) including the ectomycorrhizin E32. These polypeptides first appeared at contact and their synthesis increased during mycorrhizal formation, suggesting a role in mycorrhizal development, most likely as structural proteins. In vitro weathering of phlogopite by ectomycorrhizal fungi   总被引:1,自引:0,他引:1  
Oxalate accumulation in external medium under hyphal mats of two ectomycorrhizal species is strongly stimulated (1.7 to 35 fold) by a simultaneous depletion of available K+ and Mg2+. Pisolithus tinctorius strain 441 accumulates oxalate both on NH4–N and on NO3–N whereas Paxillus involutus strain COU only accumulates oxalate on NO3–N. On NO3–N, under a simultaneous K+ and Mg2+ deficiency, P. involutus COU is a very active oxalate producer compared to P. tinctorius 441. The present results could explain the various mineralogical evolutions of a phlogopite mica previously recorded under P. involutus COU or P. tinctorius 441 and suggest a key role for fungal oxalic acid during mineral weathering in response to nutrient deficiency.  相似文献

Summary 600,000 seedlings ofAlnus crispa were inoculated with a 111 mixture of theFrankia strains ACN1 AG , AGN1 exo AG and MGP10i. After 3 successive inoculations and screenings, one individual, AC-4, was selected as non-nodulating (Nod) with Frankiae. This selected individual AC-4 (Nod) and two other clones ofA. crispa, AC-2 and AC-5, known for their ability to nodulate (Nod+) and two other clones ofA. crispa, AC-2 and AC-5, known for their ability to nodulate (Nod+) withFrankia werein vitro propagated. The different clones ofA. crispa in culture required different kinds and concentrations of sugar during the in vitro multiplication and rooting stages. Nodulation tests using 7Frankia strains indicated that the clone AC-4 (Nod) was non-nodulating with 6 of the 7Frankia strains tested. One strain,Frankia ANNI, isolated from one unique nodule produced on the mother-plant AC-4, induced 38% of the AC-4 plantlets to nodulate but with a number of nodules 10 to 20 times less than the clones AC-2 (Nod+) and AC-5 (Nod+). Morphological observations of the roots of AC-4 (Nod) indicated that this clone had few and abnormally short root hairs.  相似文献

Summary Experiments, performed withPinus pinaster cloned shoots submitted to an auxin treatment (NAA 10–6 M, 18 days), demonstrated that rooting abilityin vitro persists over 5 successive induction cycles (through out a 9-month period). Rooting ability needs a permanent synthesis of auxin synergists which activate the metabolism of cell dedifferentiation and root primordium initiation. Agar culture permitted intense meristem initiation, but prevented active root elongation. In the presence of a mycorrhizal fungus,Pisolithus tinctorius orHebeloma cylindrosporum, roots resumed growth and short lateral root formation was stimulated. These two phenomena induced by fungal association improve the quality of the root systems required to facilitate successful transplantation from test-tubes to field conditions.  相似文献

Here, we report the establishment of an efficient, in vitro, shoot organogenesis, regeneration system for soybeans [Glycine max (L.) Merr.]. Mature soybean seeds were soaked for 24 h, the embryonic tips were collected and cultured on MSB5 medium supplemented with 3.5 mg l–1 N6-benzylaminopurine (BAP) for 24 h, and explants were transferred to MSB5 medium supplemented with 0.2 mg l–1 BAP and 0.2 mg l–1 indolebutyric acid. Use of embryonic tips yielded a higher regeneration frequency (87.7%) than regeneration systems using cotyledonary nodes (40.3%) and hypocotyl segments (56.4%) as starting materials. Regenerated embryonic tips were inoculated with Agrobacterium tumefaciens strain EHA105, which contains the binary vector pCAMBIA2301, and cultured for 20 h. Our results showed that the T-DNA transfer efficiency reached up to 78.2% and the transformation efficiency reached up to 15.8%. Nutrient concentrations in current and 1-year-old needles were analyzed annually for 5 years after application of hardened wood ash in 1–4-year-old Norway spruce (Picea abies (L.) Karst.) stands within a range of climate and fertility gradients. At each site, 3000 kg ha–1 hardened wood ash of two types, Nymölla and Perstorp, was applied in a randomized block design. Wood ash Nymölla contained 12 kg ha–1 P, 30 kg ha–1 K, 891 kg ha–1 Ca, 72 kg ha–1 Mg and wood ash Perstorp contained 12 kg ha–1 P, 60 kg ha–1 K, 486 kg ha–1 Ca, and 60 kg ha–1 Mg. The ash was intended to compensate for nutrients removed at the preceding harvest when logging residues were collected and removed from the site (whole-tree harvesting). The climate gradient included four climate zones throughout Sweden and each of these included a fertility gradient of three sites classified according to their ground vegetation type. There were no effects on nutrient concentrations in the needles 1 year after the application of wood ash. Five years after ash application, the concentrations of P, K and Ca in current and 1-year-old needles were higher than in the control plots. The results were consistent over all stands, irrespective of climate zone and fertility status. P and K concentrations were higher in spruce needles from plots treated with Perstorp wood ash, whereas Ca concentrations were higher in those of Nymölla treated plots. Analyses across all study sites revealed a treatment effect in terms of increased ratios of P:N, K:N and Ca:N in 1-year-old needles. The ratio P:N tended to increase with time in the Perstorp wood ash treatment compared with the control. The needle concentrations of Mg and S were not affected by the ash applications. The increase in needle nutrient concentrations after application of hardened wood ash suggests that wood ash recycling could be used in order to replace nutrients removed at whole-tree harvesting.  相似文献

Nitrate reduction was studied in the dinoflagellatePeridinium cinctum collected from extensive algal blooms in Lake Kinneret (Israel).Among several methods tested for the preparation of cell free extracts, only the use of a ground-glass tissue culture homogenizer was found to be efficient. The assimilatory nitrate reductase ofP. cinctum was located in a particulate fraction. In this respect,P. cinctum did not behave like other eukaryotes, such as green algae, but as a prokaryote. Nitrite reductase activity was found in the soluble fraction.Nitrate reductase used NADH as a preferable electron donor; it reacted also with NADPH but only to give 16.5% of the NADH dependent rate. Methyl viologen and benzyl viologen could also serve as electron donors, with rates higher than the NADH dependent activity (3–6 times and 1.5–3 times, respectively). The Km of nitrate reductase for NADH was 2.8×10–4 M and for NO3-1.9×10–4 M. Flavins did not stimulate the activity, nor was ferricyanide able to activate it. Carboxylic anions stimulated nitrate reductase activity 3–4 fold, an effect which was not mimicked by other anions.Chlorate, azide and cyanide were competitive inhibitors ofP. cinctum, nitrate reductase withK i values of 1.79×10–3 M, 2.1×10–5 M and 8.9×10–6 M respectively.  相似文献

Summary Resinicium bicolor (Alb. & Schw. ex Fr.) Parm. [=Odontia bicolor (Alb. & Schw. ex Fr.) Bres.] is an outcrossing resupinate basidiomycete associated with root and butt rots of trees, but is itself only very weakly pathogenic. The distribution of genets among every spruce stump in a 70-year-old 1250 m2 spruce stand was analysed using somatic incompatibility testing. R. bicolor was present on 40% of 8-to 10-year-old stumps. Nineteen genets were found occupying 32 stumps; yielding probabilities of colonisation following establishment by basidiospores of 0.20–0.24 and by mycelial extension or dispersal of 0.16–0.20. The probability of colonisation decreased with increasing distance from a point of establishment. R. bicolor responded to both enrichment and destructive disturbances by the formation of an extensive cord system which enabled it to colonise discontinuously distributed resources and to overgrow fungi adjacent to it in a single resource unit, including Heterobasidion annosum.  相似文献
