The mucilage found in the stem pith of Actinidia deliciosa contains d-glucuronic acid, d-mannose, l-fucose, l-arabinose, and d-galactose in the molar ratios 1.0:1.5:2.0:4.0. The native, carboxyl-reduced, and partially acid-hydrolysed polysaccharides were subjected to methylation analysis. Partial acid hydrolysis of the methylated, carboxyl-reduced glucuronomannan core produced a series of methylated oligosaccharides which, as their alditol derivatives, were isolated by reverse-phase h.p.l.c. and characterised by e.i.- and f.a.b.-m.s. The data suggest that the polysaccharide contains a →4)-β-d-GlcpA-(1→2)-α-d-Manp-(1→ backbone with most of the d-mannosyl and d-glucosyluronic acid residues substituted through positions 3 with oligosaccharides containing l-arabinose, α-l-fucose, and β-d-galactose.  相似文献

The extracellular anionic polysaccharide produced by the bacterium Alcaligenes (ATCC 31555) contains l-mannose, l-rhamnose, d-glucose, and d-glucuronic acid in the molar ratios 1.0:4.5:3.1:2.3. Analysis of the methylated and methylated, carboxyl-reduced polysaccharide indicated terminal non-reducing rhamnose and mannose, (1→4)-linked rhamnose, (1→3)- and (1→3,1→4)-linked glucose, and (1→4)-linked glucuronic acid to be present in the ratios 1.0:0.8:2.1:2.2:2.0:2.2. Partial acid hydrolysis and base-catalysed β-elimination gave a series of oligosaccharides that were isolated as their alkylated alditol derivatives by reverse-phase h.p.l.c. and characterised by f.a.b.-m.s., e.i.-m.s., and 1H-n.m.r. spectroscopy. The repeating unit 1, excluding O-acyl groups, is proposed.

The structure of the native dextran produced by Streptococcus sanguis ATCC 10558 was analyzed by g.l.c.-m.s. of the methylated alditol acetates derived from the polymer. The results indicate that the polymer contains D-glucosyl residues substituted at C-6 or C-3, or both, as well as unsubstituted D-glucosyl residues. These data aially purified dextransucrase on sucrose. The proportion of D-glucosyl residues substituted at C-3 is diminished in this case. It is concluded that several enzymes are involved in the dextran synthesis.  相似文献

The polysaccharide secreted by Klebsiella aerogenes type 54 strain A3 was isolated, methylated, the ester carboxyl-reduced, and the product partially hydrolyzed. The resulting, partially O-methylated oligosaccharides were reduced and ethylated, and the mixture of products was fractionated by l.c. The l.c. fractions containing per-O-alkylated oligosaccharide-alditols were analyzed by e.i.-m.s. Pure per-O-alkylated oligosaccharide-alditols were also analyzed by 1H-n.m.r. spectroscopy. The products obtained by base-catalyzed degradation and subsequent ethylation of the per-O-methylated polysaccharide were fractionated by l.c. The main product isolated was analyzed by e.i.-m.s., c.i.-m.s., and 1H-n.m.r. spectroscopy. The results of these studies, in conjunction with results of analytical methods commonly used in the elucidation of polysaccharide structures, unambiguously characterized the primary glycosyl structure of the polysaccharide. Base-labile substituents, previously reported to be present in the polysaccharide, were not studied. Structure 1 revises, and complements, previously reported structures.

A method for methylation analysis of intact glycoproteins is described. Starting with intact glycoprotein, the oligosaccharides are methylated, hydrolyzed, reduced, and acetylated. The partially methylated alditol acetates are then separated from noncarbohydrate contaminants on a silica gel G column. Partially methylated hexitol acetates are eluted from the column with petroleum ether:ethyl acetate (1:1, vv) and partially methylated N-acetylhexosaminitol acetates are subsequently eluted with methanol. Analysis by gas-liquid chromatography/mass spectrometry of the partially methylated alditol acetates shows no interfering contaminants. This method circumvents the need to make pronase glycopeptides and avoids the pitfalls of other methylation procedures.  相似文献

Oligosaccharides, obtained by digestion of a xyloglucan from the cell walls of Phaseolus coccineus with cellulase, have been isolated by gel filtration. Oligosaccharides containing 2–6 residues accounted for ~57% of the hydrolysate, with larger oligosaccharides (d.p. ~10) and partially degraded xyloglucan accounting for ~32% of the polymer. The major glycosidic linkages were determined by methylation analysis. Methylated penta- and hexa-saccharide alditols were isolated by reverse-phase h.p.l.c. and characterised by e.i.-m.s. and f.a.b.-m.s. Methylated derivatives of the di-, tri-, and tetra-saccharide alditols were examined by g.l.c.-m.s. in the e.i. and c.i. modes. A structure based on these results is proposed.  相似文献

Cell wall glycoproteins from Chlamydomonas reinhardii and the glycopeptides produced by the action of thermolysin were subjected to standard methylation analysis. GC-MS of the methylated alditol acetates revealed short oligosaccharides some of which show branching. O-glycosidically linked galactofuranosyl residues are present. The asymmetric distribution of the major O-glycosidic linkages is also reported.  相似文献

The polysaccharide of the mucin secreted by the leaves of Drosera capensis is composed of l-arabinose, d-xylose, d-galactose, d-mannose, and d-glucuronic acid in the molar ratio of 3.6:1.0:4.9:8.4:8.2. For structural elucidation, methylation analysis using g.l.c. and g.l.c.-m.s. was performed on the native, the carboxyl-reduced, and the degraded polysaccharides. Partial hydrolysis, periodate oxidation, chromium trioxide oxidation, and uronic acid degradation were also performed on the native and carboxyl-reduced polysaccharides. Partial hydrolysis of the native and carboxyl-reduced polysaccharides gave various oligosaccharides that were characterized and suggest a structure containing a d-glucurono-d-mannan backbone having a repeating unit → 4)-β-d-GlcpA-(1 → 2)-α-d-Manp-(1 →. l-Arabinose and d-xylose are present as nonreducing furanosyl and pyranosyl end-groups, respectively, both attached to O-3 of d-glucuronic acid residues of the backbone. d-Galactose is present as non-reducing pyranosyl end-group linked to O-3 of d-mannose residues.  相似文献

The extracellular anionic polysaccharide produced by the bacterium Agrobacterium radiobacter (ATCC 53271) contains D-galactose, D-glucose, and pyruvic acid in the molar ratio 2:15:2. Analysis of the methylated polysaccharide indicated the presence of terminal, non-reducing glucosyl, 3-, 4-, 6-, 2,4-, and 4,6-linked glucosyl residues, 3-linked 4,6-O-[(S)-1-carboxyethylidene]glucosyl residues, and 3-linked galactosyl residues. Partial acid hydrolysis of the methylated polysaccharide, followed by reduction with NaB2H4 and then O-ethylation, gave a mixture of alkylated oligoglycosyl alditols that were separated by reversed-phase h.p.l.c. and analyzed by 1H-n.m.r. spectroscopy, g.l.c.-m.s., and glycosyl-linkage composition analysis. Smith degradation of the polysaccharide gave three diglycosyl alditols that were separated by semi-preparative, high-pH anion-exchange chromatography, and were analyzed by 1H-n.m.r. spectroscopy, g.l.c.-m.s., and glycosyl-linkage composition analysis. The polymer obtained by NaBH4 reduction of the periodate-oxidized polysaccharide was methylated, and the noncyclic acetals were hydrolyzed with aq. 90% formic acid to generate a mixture of partially O-methylated mono- and di-glycosyl alditols. The partially O-methylated oligoglycosyl alditols were O-ethylated. The resulting alkylated oligoglycosyl alditols were separated by reverse-phase h.p.l.c. and then characterized by 1H-n.m.r. spectroscopy, g.l.c.-m.s., and glycosyl-linkage composition analysis. The results from the studies described here provide strong evidence that the acidic polysaccharide secreted by A. radiobacter (ATCC 53271) has a heptadecasaccharide repeating unit.  相似文献

A pectin isolated from rapeseed, hulls by extraction with aqueous ammonium oxalate, had a degree of esterification of 83% and contained residues of hexuronic (mainly D-galacturonic) acid (76%), D-galactose (2–3%), L-arabinose (8–9%), D-xylose (2%), L-rhamnose (2–3%), and L-fucose (1%). Partial acid hydrolysis of the derived pectic acid furnished 2-O-(α-D-galactopyranosyluronic acid)-L-rhamnose, 4-O-(α-D-galactopyranosyluronic acid)-D-galacturonic acid and the polymer-homologous tri- and tetrasaccharides, and 4-O-(glucopyranosyluronic acid)-L-fucose. The cleavage products from the methylated pectin were examined by g.l.c. and the partially methylated alditol acetates from the methylated carboxyl-reduced polysaccharide by g.l.c.-mass spectrometry. Parallel methylation studies on lemon-peel pectin have established a close similarity between the two pectins.  相似文献

Partially methylated and acetylated 3-deoxyoctitols were prepared from derivatives of 3-deoxy-d-manno-2-octulosonic acid (KDO), and identified as the d-glycero-d-talo and d-glycero-d-galacto isomers by g.l.c.-m.s. Mono- and oligosaccharide derivatives of KDO were subjected in sequence to methylation, carboxyl-reduction, hydrolysis, carbonyl-reduction, and acetylation to yield 1,2,6-tri-O-acetyl-3-deoxyoctitol derivatives. Carboxyl-reduction and then methylation gave the series of 2,6-di-O-acetyl derivatives. Oligosaccharides with KDO at the reducing end, e.g., β-d-ribofuranosyl-(1→7)-KDO, α-l-glycero-d-manno-heptopyranosyl-(1→5)-KDO, and α-KDOp-(2→4)-KDO, yielded, after carbonyl-reduction, methylation, carboxyl-reduction, hydrolysis, and acetylation, the 1,7-, 1,5-, and 1,4-di-O-acetyl derivatives, whereas remethylation after carboxyl-reduction gave the 7-, 5-, and 4-O-acetyl derivatives of 3-deoxyoctitol. General rules for the fragmentation of 3-deoxyoctitols during e.i.-m.s. were established.  相似文献

The use of partially ethylated alditol acetates for the analysis by gas-liquid chromatography of the components of polysaccharides, and the glycosidic linkages of these components, is described. The derivatives are prepared by procedures analogous to those for the synthesis of partially methylated alditol acetates. Derivatization requires two successive ethylations and more-strenuous conditions of hydrolysis and reduction than for the methyl analogs. The partially ethylated alditol acetates are formed in nearly quantitative yield and give single, sharp peaks on gas chromatography. Retention-time data, relative to two internal standards, are given for 79 glycosidic linkage-isomers of mannose, galactose, glucose, arabinose, xylose, rhamnose, and fucose, on four g.l.c. columns. One of these columns is a newly developed, highly polar, capillary column. Direct comparisons of these retention times to retention times of partially methylated alditol acetates are made. The ethyl analogs are eluted sooner that the corresponding methyl derivatives, and the amount of this shift in elution time is dependent upon the number of alkyl groups in the derivative. This change in elution time allows separation of many polysaccharide components by g.l.c. that are not separable as their partially methylated alditol acetates. Others, separated as their O-methyl derivatives, are coeluted as their partially ethylated alditol acetates. The two derivatives thus provide excellent complementary procedures because of their differential chromatographic separation and because of the similarity of their preparation.  相似文献

Results are presented which demonstrate that the molar flame-responses of partially methylated partially ethylated alditol acetates should be calculated on an effective carbon response (e.c.r.) basis. The relative responses of 2,3,4,6-tetra-O-ethyl-D-glucitol 1,5-diacetate, 2,3,6-tri-O-ethyl-D-glucitol 1,4,5-triacetate, hexa-O-ethyl-D-glucitol, hexa-O-methyl-D-glucitol, α-D-galactopyranose pentaacetate were measured and compared to the predicted values from three theories: equal molar response, equal weight response, effective carbon response. The observed values agree very well (±0?6%) with the e.c.r.-calculated values. The other theories of relative response can result in as much as 100% error in quantitation. The e.c.r-calculated relative response-factors for all commonly found partially methylated partially ethylated alditol acetates are presented, and their use is suggested for accurate quantitation.  相似文献

Starting from methyl β-D-galactofuranoside, 3,5,6-tri-O-methyl-D-galactose (9) and 2,5,6-tri-O-methyl-D-galactose (16) were synthesized. The alditol acetates were prepared from 9 and 16, and their behavior in g.l.c. was compared. Mass spectra of the alditol acetates from 9 and 16 showed that these compounds gave fragmentations as expected. The alditol acetate from 16 was also prepared by an alternative route.  相似文献

Arabinogalactan-proteins (AGPs), found in the culture medium of suspension cells of Araucaria angustifolia grown in plant growth regulator-free and plant growth regulator-containing BM media, BM0 and BM2, respectively, were evaluated quantitatively and qualitatively. The concentrated extracellular fractions (CEFs), obtained from suspension cell cultures grown for 20 days in BM0 and BM2 media yielded two fractions, CEF-0 and CEF-2, respectively. CEF-0 and CEF-2 was submitted to selective precipitation using the β-glucosyl Yariv reagent (β-GlcY) to isolate AGPs for structural characterization; this yielded fractions designated CEF-0YPF and CEF-2YPF, respectively. The monosaccharide composition analysis established that samples were composed of Rha, Ara, Gal and uronic acid in a molar ratio 3:37:55:5 (CEF-0YPF) and 1:37:58:4 (CEF-2YPF), although trace amounts (<0.5 mol%) of Xyl were also found. Methylation analysis of CEF-YPF fractions showed similar results for both CEF-0YPF and CEF-2YPF, with non-reducing terminal units of Araf, Arap, Galp, Rhap and Xylp, as well as 3-O-substituted and 5-O-substituted Araf units and 3-O-substituted, 6-O-substituted and 3,6-di-O-susbtituted Galp units. The amino acid composition analysis established Ser, Ala, and Hyp as major amino acids in both samples. In conclusion, this investigation has shown that CEF-0YPF and CEF-2YPF contain macromolecules having typical AGP characteristics, including a Hyp/Ala/Ser-rich protein moiety, a (1  3) and/or (1  6) linked β-d-galactopyranosyl main chain substituted by Gal, Ara, Rha and Xyl residues, and binding affinity for β-GlcY and monoclonal anti-AGP antibodies.  相似文献

The lipo-oligosaccharide (LOS) from Campylobacter coli Labet 227 was extracted by aqueous phenol. After delipidation and gel chromatography, two oligosaccharides were isolated. The higher molecular weight material OS (I) which was estimated to contain six to seven sugar units was found to contain glucose, galactose, 2-acetamido-2-deoxyglucose, 2-acetamido-2-deoxygalactose and heptose. Analysis of the partially methylated alditol acetates by g.c.-m.s. revealed the presence of terminal hexoses, a 1.3-linked hexose, a terminal heptose, a 1,2,3-linked heptose as well as smaller quantities of a 1,3,4-linked heptose. 1H-n.m.r. spectra showed signals corresponding to six anomeric protons. The signals which corresponded to the methyl protons of the acetamido side chain confirmed that the acetamido forms of both amino sugars were present in OS (I). The lower molecular weight material OS (II) which was estimated to contain four sugar units was found to contain glucose, 2-acetamido-2-deoxy-galactose and very small quantities of heptose. It thus appears that OS (I) and OS (II) are the core oligosaccharides elaborated by this micro-organism. The possibility of a heterogeneous core is thus presented. The fatty acids present in the LOS were mainly 3-hydroxytetradecanoic acid, n-hexadecanoic acid and trace amounts of n-tetradecanoic acid and n-octadecanoic acid.  相似文献

A method based on peracetylated oxime (PAKO) derivatives has been developed for rapid g.l.c.-m.s. survey of ketoses. This derivatization procedure (and the chromatographic analysis of these derivatives) is identical to one previously employed to identify aldoses by means of peracetylated aldononitrile (PAAN) derivatives. The production of chemically different derivatives from the aldoses and ketoses by the same derivatization procedure greatly simplifies the chromatographic separation of the derivatives of the ketoses from those of the aldoses, and also results in distinctively different, mass-spectral fragmentation-pathways for the two sets of derivatives. Both the electron-impact (e.i.) and ammonia chemical-ionization (c.i.) mass spectra of PAKO derivatives have been examined. Extensive differences between the fragmentation-pathways of the PAAN and the PAKO derivatives have been observed both by e.i.m.s. and ammonia c.i.m.s. The g.l.c.-m.s. of these PAKO derivatives, in conjunction with various, isotopic variants of the derivatization process, can yield extensive structural information with regard to the starting saccharides associated with the known, or unknown, g.l.c. peaks. The g.l.c. and mass-spectral properties of highly O-methylated PAKO derivatives of d-fructose are compared, and contrasted, to those of the PAKO derivatives of non-O-methylated saccharides. The chromatographic properties of derivatives of oligosaccharides that result from the PAAN-PAKO derivatization procedure have also been studied.  相似文献

Suspension cultures of Echinacea purpurea have been established in MS medium supplemented with 2,4-D and an arabinogalactan-protein (AGP) was purified from the secreted soluble polymers by precipitation with ethanol, followed by precipitation with β-glucosyl Yariv reagent. It revealed typical features of AGPs: a high amount of polysaccharide (90% w/w) with the dominating monosaccharides galactose and arabinose and some glucuronic acid, and a small protein moiety (10% w/w) with the main amino acids Ala, Hyp, Glx, Ser, Asx and Thr. Linkage- and NMR-analyses showed the polysaccharide part to be composed of a branched core-polysaccharide of 3-, 6- and 3,6-linked Galp residues with terminal Araf, Arap, Galp and GlcAp residues. Compared to an AGP from pressed juice of the aerial parts of Echinacea purpurea, differences particularly in terminal arabinose mono- and oligosaccharides in arabinogalactan (AG) side branches could be detected. Testing of different AGP-antibodies with both AGPs confirmed the results of the analytical investigations. Binding of AGPs from plant and cell cultures to LM2, a monoclonal AGP-antibody reacting with a GlcA containing epitope, was comparable. The reactivity of a monoclonal antibody raised against the AGP from the plant recognizing a galactan epitope was also nearly similar with both AGPs. In contrast, polyclonal antibodies raised against the AGP from the plant and directed against an Araf-containing epitope of the AG side branches showed nearly no cross reactivity with the AGP from cell culture.  相似文献

A water soluble polysaccharide (RAP) was isolated and purified from Radix Astragali and its structure was elucidated by monosaccharide composition, partial acid hydrolysis and methylation analysis, and further supported by FT-IR, GC-MS and 1H and 13C NMR spectra, SEM and AFM microscopy. Its average molecular weight was 1334 kDa. It was composed of Rha, Ara, Glc, Gal and GalA in a molar ratio of 0.03:1.00:0.27:0.36:0.30. The backbone consisted of 1,2,4-linked Rhap, α-1,4-linked Glcp, α-1,4-linked GalAp6Me, β-1,3,6-linked Galp, with branched at O-4 of the 1,2,4-linked Rhap and O-3 or O-4 of β-1,3,6-linked Galp. The side chains mainly consisted of α-T-Araf and α-1,5-linked Araf with O-3 as branching points, having trace Glc and Gal. The terminal residues were T-linked Araf, T-linked Glcp and T-linked Galp. Morphology analysis showed that RAP took random coil feature. RAP exhibited significant immunomodulating effects by stimulating the proliferation of human peripheral blood mononuclear cells and enhancing its interleukin production.  相似文献

Arabinogalactan-protein, previously isolated from instant coffee powder of Coffea arabica, has been subjected to partial mild acidic and enzymatic hydrolyses. Separation of obtained mixtures by size exclusion and HPLC chromatographies afforded series of oligosaccharides, structure of which were studied by NMR spectroscopy. Mild acidic hydrolysis afforded oligosaccharides without any αAraf substituent while after enzymatic hydrolysis αAraf was found in di-, tri-, and tetrasaccharides. In all cases αAraf was a terminal substituent linked separately to O3, O6, and to both, O3 and O6, of βGal residues. Identification of di-, tri-, and tetrasaccharides containing α-Araf enabled to distinguish in the 1H NMR spectra αAraf signals linked to O6 and O3 of neighboring βGal unit. Composition of polymeric residues after enzymatic and mild acidic hydrolyses was also analyzed.  相似文献
