Ornithodorin, isolated from the blood sucking soft tick Ornithodoros moubata, is a potent (Ki = 10(-12) M) and highly selective thrombin inhibitor. Internal sequence homology indicates a two domain protein. Each domain resembles the Kunitz inhibitor basic pancreatic trypsin inhibitor (BPTI) and also the tick anticoagulant peptide (TAP) isolated from the same organism. The 3.1 A crystal structure of the ornithodorin-thrombin complex confirms that both domains of ornithodorin exhibit a distorted BPTI-like fold. The N-terminal portion and the C-terminal helix of each domain are structurally very similar to BPTI, whereas the regions corresponding to the binding loop of BPTI adopt different conformations. Neither of the two 'reactive site loops' of ornithodorin contacts the protease in the ornithodorin-thrombin complex. Instead, the N-terminal residues of ornithodorin bind to the active site of thrombin, reminiscent of the thrombin-hirudin interaction. The C-terminal domain binds at the fibrinogen recognition exosite. Molecular recognition of its target protease by this double-headed Kunitz-type inhibitor diverges considerably from other members of this intensely studied superfamily. The complex structure provides a model to explain the perplexing results of mutagenesis studies on the TAP-factor Xa interaction.  相似文献

Galactaric acid, C6H10O8, (CAS Reg. No. 526-99-8), is triclinic, P1, with cell dimensions at ?147° [and 20°], a = 4.900(1) [4.918(1)], b = 5.728(1) [5.816(1)], c = 6.784(1) [6.849(1)] Å, α = 92.32(2) [92.31(2)], β = 93.74(2) [94.16(2)], γ = 93.08(2) [93.49(2)]°, V = 189.5 Å3, Z = 1, Dx = 1.831 [1.800], Dm = [1.790] g.cm?3, molecular symmetry I. The structure was solved by the direct method, MULTAN, and refined to R = 0.034, Rw = 0.039 for 787 reflections with FObs > 3σ(Fobs). The crystal structure has a system of strong, intermolecular hydrogen-bonds, which accounts for the high crystal density and low solubility in water.  相似文献

 The pH dependence of the dynamic quenching of the luminescence from Tb(III) and Eu(III) tris(pyridine-2,6-dicarboxylate≡DPA) chelates by the title proteins is studied. For Tb(DPA)3 3– also the quenching by the Lys 14→Glu and Lys99→Glu mutants of cytochrome c-550 (cytc-550) is investigated. The rate constants for quenching of the electronically excited Λ and Δ enantiomers of the luminophore by equine cytochrome c show a sharp decrease upon increasing the pH from 7 to 10, which can be described phenomenologically by deprotonation of a single acidic group with pK a of 9.2±0.1 for Eu and 9.4±0.1 for Tb. These values are similar to that found for the alkaline transition of the protein. The alkaline conformer(s) of the protein at pH>10 is found to be a very inefficient quencher of the lanthanide luminescence. For Tb, but not for Eu, a significant lowering of the degree of enantioselectivity (E q) in the quenching is found along with a reduction of the quenching rates. For cytc-550, the decrease of the quenching rate constants with increasing pH is described by pK a=9.8±0.1 and for the two mutants the same value is obtained. For the cytc-550 proteins the change of the quenching rates does not correlate with the alkaline transition, for which a pK a of 11.2 has been reported by other workers. For all proteins, the reduction of the quenching rates at high pH is ascribed to a reduction of the binding affinity of the excited lanthanide complex to the surface area of the protein near the exposed heme edge, caused by deprotonation of (presumably) several lysine residues. We describe the 2.3 ? (1 ?=0.1?nm) X-ray structure of α1m (α1-microglobulin), an abundant protein in human blood plasma, which reveals the β-barrel fold typical for lipocalins with a deep pocket lined by four loops at its open rim. Loop #1 harbours the residue Cys34 which is responsible for covalent cross-linking with plasma IgA. A single disulfide bond between Cys72 and Cys169 connects the C-terminal segment to the β-barrel, as in many other lipocalins. The exposed imidazole side chains of His122 and His123 in loop #4 give rise to a double Ni2+-binding site together with a crystallographic neighbour. The closest structural relatives of α1m are the complement protein component C8γ, the L-prostaglandin D synthase and lipocalin 15, three other structurally characterized members of the lipocalin family in humans that have only distant sequence similarity. In contrast with these, α1m is initially expressed as a bifunctional fusion protein with the protease inhibitor bikunin. Neither the electron density nor ESI-MS (electrospray ionization MS) provide evidence for a chromophore bound to the recombinant α1m, also known as 'yellow/brown lipocalin'. However, the three side chains of Lys92, Lys118 and Lys130 that were reported to be involved in covalent chromophore binding appear to be freely accessible to ligands accommodated in the hydrophobic pocket. A structural feature similar to the well-known Cys-Pro haem-binding motif indicates the presence of a haem-binding site within the loop region of α1m, which explains previous biochemical findings and supports a physiological role in haem scavenging, as well as redox-mediated detoxification.  相似文献

Taxonomic composition and variations in density and biomass of the plankton community in the Öregrundsgrepen, a shallow coastal area, were investigated from June 1972 to November 1973. The phytoplankton biomass was large in spring but small during the rest of the year. The spring bloom was dominated by diatoms and dinoflagellates, especially byThalassiosira spp. which were also important during other seasons. Small forms, such asCryptomonas spp.,Rhodomonas spp. and monads, dominated during summer. Blue-green algae were never of any major importance. During the summer, the trophogenic layer exceeded 10 m in thickness. The metazoan fauna was of lower diversity than the plankton flora. The dominating species, the copepodsAcartia bifilosa andEurytemora affinis, constituted on the average 83% of the standing crop. The low salinities, 5–6 S, were regarded as the principal pertinent limiting factor. The metazoan fauna reached large biomass values from July to October. The protozoan fauna (in the case of ciliates), obtained biomass maxima during the spring bloom. It is suggested that the Öregrundsgrepen represents an area of elevated productivity within a region of low overall production, presumably due to local upwelling. From June 1972 to May 1973, the average biomasses were: phytoplankton 0.464 g C m–2, ciliates 0.040 g C m–2, copepod nauplii 0.010 g C m–2, micro-rotifers 0.004 g C m–2, and mesozooplankton (larger than 0.2 mm) 0.312 g C m–2. It is estimated that about than 60% of the phytoplankton production is consumed by the microzooplankton (<0.2 mm).  相似文献

We calculate here the Raman frequencies of the lattice modes A(A g ), B(B 2g ) and C(B 1g B 3g ) as a function of pressure at room temperature for the solid phases (II, III and III’) of benzene. This calculation is performed using volume data through the mode Grüneisen parameter. It is found that our calculated frequencies of those lattice modes increase with increasing pressure, as expected. Calculated frequencies are in good agreement with the measurements of the three lattice modes for the solid phases studied in benzene.  相似文献

The kinetics of the oxidation of bromide ions by the bis(2,2′-bipyridine)manganese(III) ion have been investigated over a range of acid concentrations for several temperatures. Initial rates of reaction are measured and from their variation with [MnIII] and [Br−] it is concluded that the observed order in [Mn(bipy)23+aq] is one and that in [Br] is intermediate between zero and one. It is shown kinetically that intermediate complexes between MnIII and Br ions are involved and from the variation of the rate constant with acidity it is concluded that the decomposition of Mn(bipy)2Br2+aq is definitely involved as a rate controlling step and that the decomposition of the protonated complex Mn(bipy)(bipyH+)Br3+aq is also probably involved as a rate controlling step. The kinetics and mechanism for this oxidation are compared with those found for other cations complexed with bipyridine, for aqua-cations and other complexes of CoIII.  相似文献

The crystal structure of a regenerated form of (1→3)-α-d-glucan, obtained by solid state deacetylation of the triacetate derivative, has been determined by combined X-ray diffraction analysis and stereochemical model refinement. The structure crystallizes in an orthorhombic unit cell with parameters a = 16.46 A?, b = 9.55 A? and c (fibre repeat)=8.44 Å, and space group P212121. The chain conformation is nearly completely extended and is very close to a 2/1 helix, even though the dimer residue is the crystallographic repeat unit. An intramolecular O(2)  O(4)′ hydrogen bond stabilizes the conformation and extensive intermolecular hydrogen-bonding abilizes the packing. The resulting structure is sheet-like, with an alternating polarity of chain directions within the sheet. In its sheet-like character, extensive hydrogen-bonding, and insolubility in water, this polymorph of (1→3)-α-d-glucan resembles regenerated cellulose. The reliability of the structure analysis is indicated by the X-ray residual R=0.206.  相似文献

The IR and Raman spectra of Cs(SO2), K(SO2) and Na(SO2) molecules were studied by 32S/34S and 16O/18O isotopic substitution technique. These molecules have a planar ring configuration of C2v symmetry with the OSO angle equal to 109°±5° and the SO bond length of 0.149±0.001 nm. The alkali metal atom interacts symmetrically with the oxygen atoms of the SO2 group. The doubling observed for the vibrations of Cs(S16O18O) was attributed to a matrix effect.  相似文献

During the visual process, light absorption in the 11-cis retinylidene chromophore leads to a rapid cis-trans-isomerization which initiates the phototransduction step. Important spectroscopic properties of this chromophore can be derived from polarized UV-absorption spectra of crystalline 11-cis-retinal if a parallel X-ray structure analysis is performed. Several questions about the relation between molecular geometry and spectroscopic behavior could not be answered from these spectra. All crystal forms of 11-cis-retinal contain this molecule in its 6-s-cis-ring conformation. For the retinal homologue, -8-apocarotenal (APC), however, two crystal forms with different ring conformation can be grown. The spectrum of -APC (6-s-cis) shows a vibronic structure whereas that of -APC (6-s-trans) is diffuse but has a distinct shoulder on the low energy side of the main band. This S-band is typical for retinal spectra and has been ascribed to a transition into a 1A g -* -state. The appearance of the S-band is not correlated with a 6-s-cis-conformation as suggested by the retinal spectra but is due to intermolecular interactions: -APC has a dense dimer packing and a strong electrostatic interaction between the -electron systems. This might cause the forbidden 1A g -* -transition. On the other hand, this interaction is missing in the loose and polar packing of -APC which favors vibration in the polyene chain. Summary The ultrastructure of the compound eyes of 13 amphipod species has been investigated. An amphipod type of compound eye can be characterized by the constellation and consistency of a number of morphological features, most of which are also found in other compound eyes. The amphipod eye falls into four sub-categories (types). The ampeliscid type has a tripartite aberrant lens eye; the lysianassid type has a reduced or no dioptric apparatus and a hypertrophied rhabdom; the hyperid type possesses a large number of ommatidial units with long crystalline cones and dark instead of reflecting accessory pigment; and finally, the gammarid type can be interpreted as a generalized amphipod type. The lysianassid type is adapted to low light intensities and demonstrates convergent development with the compound eyes of other deep-sea crustaceans. The ampeliscid type is more similar to the gammarid type. The absorption and magnetic circular dichroism spectra of the transitions 3H43F2, 3H5 3F3, 3F4 and 3H6 of (Cp-d5)3UCl have been measured at room and low temperatures. Preliminary crystal field calculations allowed the assignment of many transitions. On the basis of the crystal field eigenfunctions and eigenvalues obtained from these calculations, the temperature dependence of the paramagnetic susceptibility was calculated. For an orbital reduction factor of k=0.93, a satisfactory agreement of experimental and calculated values was achieved.  相似文献

Otoferlin (Otof), whose genetic mutations cause profound deafness in humans, is a protein composed of at least six C2 domains, which are known as Ca2+-binding and phospholipid-binding regions. Mammalian ferlin proteins are proposed to act in membrane fusion events, with Otof being specifically required for exocytosis in auditory hair cells. Ferlin C2 domains exhibit a rather low level of sequence similarity to those of synaptotagmins, protein kinase C isoforms, or phospholipases. Here, we report the crystal structure of the N-terminal C2 domain of Otof (C2A) at 1.95-Å resolution. In contrast to previous predictions, we found that this C2 domain is complete with eight β-strands. Comparing the structure of Otof C2A to those of other C2 domains revealed one top loop in Otof to be significantly shorter. This results in a depression of the surface, which is positively charged for the Otof C2A domain, and contrasts with the head-like protrusion surrounded by a negatively charged “neck” typically found in other C2 domains. Isothermal titration calorimetry and circular dichroism spectroscopy studies confirmed that Otof C2A is unable to bind Ca2+, while the synaptotagmin-1 C2A domain exhibited Ca2+ binding under the same conditions. Furthermore, floatation assays revealed a failure of Otof C2A to bind to phospholipid membranes. Accordingly, no positively charged β-groove-like surface structure, which is known to bind phosphatidylinositol-4,5-bisphosphate in other C2 domains, was found at the respective position in Otof C2A. Taken together, these data demonstrate that the Otof C2A domain differs structurally and functionally from other C2 domains.  相似文献

tRNAs are aminoacylated with the correct amino acid by the cognate aminoacyl-tRNA synthetase. The tRNA/synthetase systems can be divided into two classes: class I and class II. Within class I, the tRNA identity elements that enable the specificity consist of complex sequence and structure motifs, whereas in class II the identity elements are assured by few and simple determinants, which are mostly located in the tRNA acceptor stem. The tRNA(Gly)/glycyl-tRNA-synthetase (GlyRS) system is a special case regarding evolutionary aspects. There exist two different types of GlyRS, namely an archaebacterial/human type and an eubacterial type, reflecting the evolutionary divergence within this system. We previously reported the crystal structures of an Escherichia coli and of a human tRNA(Gly) acceptor stem microhelix. Here we present the crystal structure of a thermophilic tRNA(Gly) aminoacyl stem from Thermus thermophilus at 1.6? resolution and provide insight into the RNA geometry and hydration.  相似文献
