A nickel(II) complex with 6-(p-chlorobenzoyl)-5,7,12,14-tetramethyl-benzo[b]-1,4,8,11- tetraazacyclo[14]tetradecine was synthesized and characterized by measurements of NMR, IR and an electronic spectra. The X-ray crystal structure shows that the coordination geometry around the Ni atom is a square planar with the unsymmetrical tetraaza[14]annulene.  相似文献

The reaction of [Ag2(κ2-P,P′-DPEphos)2(μ-OTf)2] (1) (DPEphos = bis(2-(diphenylphosphino)phenyl]ether) with 1,10-phenanthroline (phen) and 4,4′-bipyridine in equimolar ratios afford, respectively, the mononuclear complex [Ag(κ2-P,P′-DPEphos)(phen)][OTf] (2) and the coordination polymer [Ag(κ2-P,P′-DPEphos)(μ-4,4′-bpy)]n[OTf]n (3). In complex 3, the silver atoms are bridged by 4,4′-bipyridine units to form a zigzag metallopolymer.  相似文献

Gastrin-releasing peptide receptors (GRP-Rs, also known as bombesin 2 receptors) are overexpressed in a variety of human cancers, including prostate cancer, and therefore they represent a promising target for in vivo imaging of tumors using positron emission tomography (PET). Structural modifications of the non-peptidic GRP-R antagonist PD-176252 ((S)-1a) led to the identification of the fluorinated analog (S)-3-(1H-indol-3-yl)-N-[1-[5-(2-fluoroethoxy)pyridin-2-yl]cyclohexylmethyl]-2-methyl-2-[3-(4-nitrophenyl)ureido]propionamide ((S)-1m) that showed high affinity and antagonistic properties for GRP-R. This antagonist was stable in rat plasma and towards microsomal oxidative metabolism in vitro. (S)-1m was successfully radiolabeled with fluorine-18 through a conventional radiochemistry procedure. [18F](S)-1m showed high affinity and displaceable interaction for GRP-Rs in PC3 cells in vitro.  相似文献

The reaction, at 25 °C in methanol, between [Pd(SeCN)4]2− and bis(diphenylphosphino)methane (dpm) has been found to produce cyano(selenocyanato) [diphenyl(diphenylphosphinomethyl) phosphine selenide]palladium(II), [Pd(dpmSe)(CN)(SeCN)], wherein the cyanide group is trans to an Se atom which has been inserted into one PdP bond, and the selenocyanate group is trans to the unchanged diphenylphosphino group. The structure has been confirmed by the results of a single crystal X-ray diffraction study. The structural isomer, [Pd(dpm)- (SeCN)2], of the foregoing complex has also been prepared by the reaction of Pd(C2H3O2)2 with dpm, followed by reaction with KSeCN. Heating the [Pd(dpm)(SeCN)2] isomer converts it into [Pd- (dpmSe)(CN)(SeCN)]. A mechanism is proposed for the isomerization which involves an intramolecular selenium atom insertion.  相似文献

Asymmetric trigonal-bipyramidal Zn(II) complex 1 formed by 2-[bis(2-aminoethyl)amino]ethanol (L) was found to be able to promote the cleavage of supercoiled plasmid DNA pBR322 to the nicked and linear DNA via a hydrolytic manner but only in neutral Tris-HCl buffer, no cleavage was observed in HEPES or NaH2PO4/Na2HPO4 buffer. However, the copper complex 2 of L, possessing the similar coordination geometry, can only promote DNA cleavage via an oxidative mechanism in the presence of ascorbic acid. ESI-MS study implies that complex 1 exist mainly as [Zn(L)]2+/[Zn(L-H)]+ in neutral Tris-HCl buffer. Moreover, there is no discriminable species for complex 1 in HEPES or NaH2PO4/Na2HPO4 buffer. A phosphate activation mechanism via phosphate coordinating to Zn(II) center of [Zn(L)]2+/[Zn(L-H)]+ to form the stable trigonal-bipyramidal structure is proposed for the hydrolytic cleavage promote by complex 1. For complex 2, the abundance of [Cu(L)Cl]+ is higher than that of [Cu(L)]2+/[Cu(L-H)]+ in Tris-HCl buffer. The lower phosphate binding/activating ability of Cu(II) in complex 2 may be the origin for its incapability to promote the hydrolytic DNA cleavage. However, the readily accessible redox potential of Cu(II) makes complex 2 promote the oxidative DNA cleavage. Although the DNA cleavage promoted by complex 1 has no specificity, trigonal-bipyramidal Zn(II) complexes formed by asymmetric tripodal polyamine with ethoxyl pendent should be a novel potential model for practical artificial nuclease.  相似文献

The structures of thiacalix[2]thianthrene, p-tert-butylthiacalix[2]thianthrene and their complexes with Zn2+, Cd2+ and Hg2+ were obtained using B3LYP/LanL2DZ and HF/LanL2DZ calculations. The structures of the most stable conformers of thiacalix[2]thianthrene and p-tert-butylthiacalix[2]thianthrene optimized at either the B3LYP/LanL2DZ or HF/LanL2DZ level are in good agreement with their corresponding X-ray crystallographic structures. The binding energies of cations, Zn2+, Cd2+ and Hg2+ to thiacalix[2]thianthrene and to p-tert-butylthiacalix[2]thianthrene conformers, and the thermodynamic properties of their associations were obtained. The relative selectivities of both thiacalix[2]thianthrene and thiacalix[2]thianthrene conformer are in same order: Zn2+ >> Hg2+ > Cd2+.   相似文献

The coordination of the ligands bis(pyrazol-1-yl)acetate (bpza) and bis(3,5-dimethylpyrazol-1-yl)acetate (bdmpza) to rhenium(VII) was investigated. The compounds [(bpza)ReO3] and [(bdmpza)ReO3] were synthesised by reaction of bpza and bdmpza with perrhenic acid with the loss of one water molecule. The new complex [(bdmpza)ReO3] was characterised by single-crystal X-ray analysis. It has a monomeric structure with a distorted octahedron for the [N,N,O]ReO3 central core.  相似文献

Human rhinoviruses (HRVs) are the most common cause of viral respiratory infections and their complications. So far, no anti-viral agent has been approved for prevention or treatment of HRV infections. Pursuing our researches on small molecules with anti-rhinovirus activity, in this paper we describe the synthesis and in vitro anti-HRV 1B and 14 properties of new [2-(2H-chromen-3-yl)vinyl]pyridines and 3-[2-(pyridinyl)vinyl]-4H-chromen-4-ones. Generally, the synthesized compounds interfered with the replication of both serotypes at the micromolar or submicromolar concentrations. Preliminary results on their mechanism of action, performed on selected (E)-2-[2-(2H-chromen-3-yl)vinyl]pyridine, indicate an interference with the early step(s) of HRV 1B and 14 replication, probably at the uncoating level.  相似文献

Two new ruthenium(II) complexes of [Ru(bpy)(pp[2,3]p)2](ClO4)2 and [Ru(phen)(pp[2,3]p)2](ClO4)(2) (bpy=2,2'-bipyridine, phen=1,10-phenanthroline, pp[2,3]p=pyrido[2',3':5,6]pyrazino[2,3-f][1,10]phenanthroline) have been synthesized and characterized by elemental analysis and 1H NMR spectra. The calf thymus DNA-binding properties of the two complexes were investigated by UV-visible and emission spectroscopy, competitive binding experiments with ethidium bromide and viscosity measurements. The results indicate that the two complexes intercalate between the base pairs of the DNA tightly with intrinsic DNA-binding constants of 3.08 x 10(6) and 6.53 x 10(6) M(-1) in buffered 50 mM NaCl, respectively, which are much larger than 6.9 x 10(5) M(-1) for [Ru(bpy)2(pp[2,3]p)](ClO4)2 containing two ancillary ligands of bpy.  相似文献

A new class of amidoalkyl dibenzofuranols and 1H-benzo[2,3]benzofuro[4,5-e][1,3]oxazin-3(2H)-ones was synthesized in very good yields through polyphosphoric acid supported on silica (PPA-SiO2) catalyzed one-pot three component condensation of 2-dibenzofuranol; aromatic aldehydes and acetamide or benzamide or urea under solvent free conditions. At 125 °C the reaction led to the formation of amidoalkyl dibenzofuranols 5a-k where as at 160 °C cyclization take place to give oxazin-3(2H)-one analogues 6a-e. Screening all the 16 compounds for in vitro antimycobacterial activity against Mycobacterium tuberculosis H37Rv (MTB) resulted 1-((4-chlorophenyl)(2-hydroxydibenzo[b,d]furanyl)methyl)urea 5h; 1-((4-bromophenyl)(2-hydroxydibenzo[b,d]furanyl)methyl)urea 5i; 1-phenyl-1H-benzo[2,3]benzo furo[4,5-e][1,3]oxazin-3(2H)-one 6a (MIC 3.13 μg/mL) and 1-(4-chlorophenyl)-1H-benzo[2,3]benzofuro[4,5-e][1,3]oxazin-3(2H)-one 6b; 1-(4-bromophenyl)-1H-benzo[2,3]benzofuro [4,5-e][1,3]oxazin-3(2H)-one 6c (MIC 1.56 μg/mL) as most active antitubercular agents.  相似文献

The reaction of [RuCl3(2mqn)NO] (H2mqn=2-methyl-8-quinolinol) with 2-chloro-8-quinolinol (H2cqn) afforded cis-1 [RuCl(2cqn)(2mqn)NO] (the oxygen of 2cqn is trans to the NO) (complex 1), cis-1 [RuCl(2cqn)(2mqn)NO] (the oxygen of 2mqn is trans to the NO) (complex 2) and a 1:1 mixture of cis-2 [RuCl(2cqn)(2mqn)NO] (the oxygen of 2mqn is trans to the NO) and cis-2 [RuCl(2cqn)(2mqn)NO] (the oxygen of 2cqn is trans to the NO) (complex 3). The reaction was compared with that of [RuCl3(2mqn)NO] with 8-quinolinol (Hqn) or 5-chloro-8-quinolinol (H5cqn). Photoirradiation reaction of complex 1 at room temperature in deaerated CH2Cl2 in the presence of NO gave trans-[RuCl(2cqn)(2mqn)NO] (the Cl is trans to the NO) and complex 2 with recovery of complex 1. The reaction was contrasted with that of cis-1 [RuCl(qn)(2mqn)NO] or cis-1 [RuCl(5cqn)(2mqn)NO]. The crystal structure of complex 1 was determined by X-ray diffraction. The reactions were examined under consideration of atomic charge of the phenolato oxygen in 8-quinolinol and its derivatives calculated at the restricted Hartree-Fock/6-311G** level.  相似文献

The ability of a metal-coordinated pyrazole to engage in hydrogen bonding has been explored by synthesis of the title complex, bis[3-([thiomethyl]methyl)pyrazole]copper(II) perchlorate (3). The coordination in 3 can be described as pseudo-octahedral, with two relatively tightly-bound 3-[(thiomethyl)methyl]pyrazole ligands occupying the equatorial plane, forming a [CuN(2)S(2)](2+) unit with the S donors mutually trans to each other. The axial positions are each filled by a weakly bound perchlorate counterion, one oxygen of which forms a hydrogen bond with the pyrazole N-H moiety on an adjacent [CuN(2)S(2)](2+) unit.  相似文献

A iron(II) complex of the linear hexadentate N6 ligand H2L2-3-2, [Fe(H2L2-3-2)](ClO4)2, was synthesized and the spin crossover properties were investigated, where H2L2-3-2 denotes bis[N-(2-methylimidazol-4-yl)methylidene-2-aminoethyl]propanediamine. The complex showed a gradual and reversible one-step spin crossover (SCO) between the high-spin (S = 2) and low-spin (S = 0) states at T1/2 = 208 K without hysteresis. The crystal structures were determined at 296 and 250 K (HS state), 230, 210, and 200 K (intermediate between the HS and LS states) and 150 and 110 K (LS state). The spin transition from 296 to 150 K accompanies with the conformation change of the chelate rings at the triamine moiety and the formation of the hydrogen bond network in the same space group of orthorhombic Pbcn (no. 60). However, in the LS state at 110 K, the space group changed from orthorhombic Pbcn at 150 K (Pcan when the same axial setting to 110 K was used) to monoclinic P21/a (no. 14) at 110 K, although no spin transition and no change of assembly structure between 150 and 110 K were observed. It give us an idea that the space group transformation is mainly related to the conformational thermodynamic stability of the chelate rings at the triamine moiety and is not directly correlated with the spin transition.  相似文献

The reaction of [Ru(CO)2Cl2]n with bis(2-pyridylmethyl)amine (bpma) in refluxing ethanol followed by anion exchange yields two products: cis,fac-[Ru(bpma)(CO)2Cl]PF6 (1a, 71%) and trans,fac-[Ru(bpma)(CO)2Cl]PF6 (1b, 29%). Reaction of 1a with AgBF4 in acetone, followed by acetonitrile and then anion exchange gave cis,fac-[Ru(bpma)(CO)2(CH3CN)](PF6)2 (2a). In the same way, 1b afforded trans,fac-[Ru(bpma)(CO)2(CH3CN)](PF6)2 (2b). Reaction of depolymerized [Ru(CO)2Cl2]n with bpma in ethanol at room temperature afforded cis,cis-[Ru(η2-bpma)(CO)2Cl2] (3). In refluxing ethanol, 3 was converted to cis,fac-[Ru(bpma)(CO)2Cl]Cl (1a-Cl). Heating 3 in chlorobenzene afforded 1b-Cl, exclusively; heating 3 in ethylene glycol gave mainly 1a-Cl. Heating 1a-Cl in ethanol resulted in no isomerization, but heating in chlorobenzene gave a mixture of 3 and 1b-Cl. Anion exchange for PF6 with 1a-Cl and 1b-Cl afforded 1a and 1b, respectively, whereas anion exchange for BPh4 afforded 1a-BPh4. Compounds 1a, 1b, 2a and 3 have been structurally characterized.  相似文献

The T-type calcium channel inhibitor Mibefradil was reported to protect the heart from atrial remodeling, a key process involved in the development of atrial fibrillation and arrhythmias. Mibefradil is not a selective T-type calcium channel inhibitor and also affects the function of different ion channels. Our aim was to develop a selective T-type calcium channel inhibitor to validate the importance of T-type-related pharmacology in atrial fibrillation. Structural optimisation of a previously disclosed hit series focussed on minimising exposure to the central nervous system and improving pharmacokinetic properties, while maintain adequate potency and selectivity. This resulted in the design of N-[[1-[2-(tert-butylcarbamoylamino)ethyl]-4-(hydroxymethyl)-4-piperidyl]methyl]-3,5-dichloro-benzamide, a novel, selective, peripherally restricted chemical probe to verify the role of T-type calcium channel inhibition on atrial fibrillation protection.  相似文献

Gibberellins (GAs) are either required for, or at least promote, the growth of the pea (Pisum sativum L.) fruit. Whether the pericarp of the pea fruit produces GAs in situ and/or whether GAs are transported into the pericarp from the developing seeds or maternal plant is currently unknown. The objective of this research was to investigate whether the pericarp tissue contains enzymes capable of metabolizing GAs from [14C]GA12-7-aldehyde ([14C]GA12ald) to biologically active GAs. The metabolism of GAs early in the biosynthetic pathway, [14C]GA12 and [14C]GA12ald, was investigated in pericarp tissue isolated from 4-day-old pea fruits. [14C]GA12ald was metabolized primarily to [14C]GA12ald-conjugate, [14C]GA12, [14C]GA53, and polar conjugate-like products by isolated pericarp. In contrast, [14C]GA12 was converted primarily to [14C]GA53 and polar conjugate-like products. Upon further investigations with intact 4-day-old fruits on the plant, [14C]GA12 was found to be converted to a product which copurified with endogenous GA20. Lastly, [2H]GA20 and [2H]GA1 were recovered 48 hours after application of [2H]- and [14C]GA53 to pericarp tissue of intact 3-day-old pea fruits. These results demonstrate that pericarp tissue metabolizes GAs and suggests a function for pericarp GA metabolism during fruit growth.  相似文献

A study of the neurotropic, neuroprotective, and antioxidant action of the enantiomers and racemate of 2-[(3,7-dioxo-2,4,6,8-tetraazabicyclo[3.3.0]oct-2-yl)]-4-methylthiobutanoic acid synthesized in a stereoselective reaction of (R)-, (S)-, or (R,S)-N-carbamoylmethionine with 4,5-dihydroxyimidazolidine-2-one showed that only (+)-(S)-2-[(1S,5R)-(3,7-dioxo-2,4,6,8-tetraazabicyclo[3.3.0]oct-2-yl)]-4-methylthiobutanoic acid had neuroprotective properties. X-ray structure analysis showed that the predominating racemate of glycolurils is crystallized from aqueous solutions as a conglomerate. Antioxidant activity was not detected.  相似文献

A cytostatic, homo-aza-steroidal ester of [p-[bis-(2-chloroethyl) amino]phenyl]acetic acid (ASE) was reduced with NaB3H4 and [3H]ASE-treated DNA prepared in vitro. We found that: (1) ASE reacts preferentially with purines; (2) ASE decreases the thermal stability of the double helix upon binding to DNA; (3) [3H]ASE binding sites are clustered along the DNA molecules; (4) ASE binding sites probably represent oligo- or polypurine sequences.  相似文献

A novel series of 5(R)-[1,2,3]triazolylmethyl and (5R)-[(4-F-[1,2,3]triazolyl)methyl]oxazolidinones having various piperidine group were synthesized and evaluated antibacterial activity against clinically isolated resistant strains of Gram-positive and Gram-negative bacteria. The compound 12a having exo-cyanoethylidene group in the 4-position of piperidine ring was found to be two to threefold more potent than the linezolid against penicillin-resistant Staphylococcus pneumonia and Staphylococcus agalactiae, and also exhibited reduced MAO-B inhibitory activity.  相似文献
