Lipid-protein particles ranging from 20 to 250 nm in diameter have been isolated from the cytosol of carnation petals by flotation centrifugation and also by ultrafiltration. The cytosolic lipid-protein particles resemble oil bodies, lipid-protein particles found in oil-bearing seeds, in that they contain triacylglycerol, are circumscribed by phospholipid that is not organized in a bilayer, appear to be derived from membranes and can be isolated by flotation. However, the cytosolic particles are distinguishable from oil bodies in that triacylglycerol is not the dominant lipid. Indeed, they contain a spectrum of lipids in addition to phospholipids and triacylglycerol including free fatty acids, sterol and wax esters, phosphatidic acid and diacylglycerol. These same lipids are present in corresponding microsomal membranes as well, but in much smaller proportions relative to phospholipid. The lipid-protein particles from carnation petals contain a 17-kDa protein that is of similar size to oil body oleosin, but does not cross-react with anti-oleosin antibodies. The data indicate that these cytosolic particles are structurally and chemically similar to oil bodies and are consistent with the notion that their genesis may be a means of removing destabilizing lipids from membrane bilayers.  相似文献

The control of ion concentration in the cytosol and the accumulation of ions in vacuoles are thought to be key factors in salt tolerance. These processes depend on the establishment in vacuolar membranes of an electrochemical H+ gradient generated by two distinct H+-translocating enzymes: a H+-PPase and a H+-ATPase. H+-lrans locating activities were characterized in tonoplast-enriched membrane fractions isolated by sucrose gradient centrifugation from sunflower ( Helianthus annuus L.) roots exposed for 3 days to different NaCl regimes. The 15/32% sucrose interface was enriched in membrane vesicles possessing a vacuolar-type H+-ATPase and a H+-PPase, as indicated by inhibitor sensitivity, pH optimum, substrate specificity, ion effects kinetic data and immunolabelling with specific antibodies. Mild and severe stress did not alter the pH profile, ion dependence, apparent Km nor the amount of antigenic protein of either enzyme. Saline treatments slightly increased K+-stimulaied PPase activity with no change in ATPase activity, while both PPi-dependent and NO3-sensitive ATP-dependent H+ transport activities were strongly stimulated. These results are discussed in terms of an adaptative mechanism of the moderately tolerant sunflower plants to salt stress.  相似文献

Corn ( Zea mays L.) plasma membranes from KI-washed microsomal fractions were further purified by isopycnic sucrose density centrifugation. An examination of separated fractions indicated that vesicles with nitrate-insensitive proton transport copurified with fractions containing vanadate-sensitive ATPase activity. The ATPase in purified plasma membrane was reconstituted into liposomes by a detergent dilution technique using deoxycholate. The reconstituted ATPase exhibited characteristics similar to those of the native enzyme. However, reconstituted preparations showed an enhanced sensitivity to vanadate, a diminished phosphatase activity and a high specific rate of ATP-dependent H+-transport. Apparent Ki values of reconstituted and native enzymes with respect to vanadate were 20 and 50 μ M , respectively; the KJ value of the H+-pumping of reconstituted ATPase was 30 μ M. The proton pumping of reconstituted vesicles could be discharged rapidly by p -trifluoromethoxyphenyl hydrazone (FCCP), hexokinase and vanadate. The hydrolysis of Mg-ATP by both native and reconstituted ATPases obeyed simple Michaelis-Menten plots with a Km between 0.5 and 0.6 m M. The reconstituted ATPase retained a pH profile similar to that of native enzyme with a maximum of pH 6.5.  相似文献

Two monovalent ion porters, the putative Na+/H+ antiporter (NapA) of Enterococcus hirae and the putative K+/H+ antiporter (KefC) of Escherichia coli, are similar in sequence throughout their hydrophobic domains. These two proteins, which comprise a novel family of transporters unrelated to the previously characterized Na+/H+ exchangers of E. coli (NhaA and NhaB) are proposed to function by essentially the same mechanism.  相似文献

Plasma membrane vesicles with H+-ATPase activity were purified from 8-day-old oat ( Avena sativa L. cv. Brighton) roots using an aqueous polymer two-phase system. Of several detergents tested, only lysophosphatidylcholine solubilized the H+-ATPase in an active form. Solubilization of the H+-ATPase with lysophosphatidylcholine was possible in the absence of glycerol, but the ATPase activity decreased about 4–5 times as rapidly in the absence as in the presence of 30% (w/v) glycerol. The solubilized enzyme was further stabilized by ATP and protons. Addition of 1 m M ATP to the plasma membranes halted inactivation of the H+-ATPase. Even in the absence of polyol compounds and ATP, the enzyme was stable for hours at relatively low pH with an optimum around pH 6.7 at room temperature. The curve for the stability of soluble H+-ATPase as a function of pH closely resembles the pH curve for the activity of the H+-ATPase. This suggests that binding of protons to transport sites may stabilize the soluble H+-ATPase in an enzymatically active form.  相似文献

Tonoplast, ion antiport activities are critical to ion homeostasis and sequestration in plants. The biochemical properties of these activities, and the enzymes that catalyse them, are little characterized. Here we applied biochemical approaches to study some characteristics and to distinguish between Ca2+/H+ and Cd2+/H+ antiporter activities of tonoplast vesicles from non‐transformed, wild‐type plants. Solubilization and reconstitution of oat‐seedling (Avena sativa L.) root tonoplast vesicles resulted in about a 6‐fold loss of protein, about a 6‐fold enhancement of Cd2+/H+ antiport specific activity (at 10 µM Cd2+), and almost complete loss of Ca2+/H+ antiport activity. Similar results were found for vesicles from mature tobacco (Nicotiana tabacum) roots. Cd2+ concentration‐dependent proton efflux was similar and linear with both oat vesicles and proteoliposomes. In contrast, Ca2+ concentration‐dependent proton efflux of oat vesicles was easily observed while that with proteoliposomes was minimal and non‐linear. Cd2+ pre‐treatment of oat vesicles reduced verapamil inhibition of Cd2+/H+ activity and verapamil binding to vesicles, while Ca2+ pre‐treatment was much less protective of Ca2+/H+ activity and verapamil binding. Results show the usefulness of reconstitution, and also inhibitor/ion interaction assays for distinguishing between transporter activities in vitro, but they do not resolve the question of whether there are separate enzymes for Cd2+/H+ and Ca2+/H+. Our observation that solubilization and reconstitution have similar effects on both Cd2+/H+ and Ca2+/H+ activities of root tonoplast vesicles from immature oat and mature tobacco roots suggests that the transporters involved are similar in young and mature roots, and in roots of different species.  相似文献

Abstract Fusicoccin (FC)-stimulated K+ (86Rb) uptake and proton extrusion of maize (Zea mays) root apical segments were inhibited by pretreatment of 4-day-old seedlings with the herbicide Chlorsulfuron. In the range of Chlorsulfuron concentrations 0.01-10 mmol m?3, the percentage of inhibition was 15% at 0.01 mmol m?3 and progressively increased with Chlorsulfuron concentration up to 60% at 10 mmol m?3. At the maximum concentration tested (10 mmol m?3), the inhibition was evident after 1.5 h of pre-treatment. The binding of FC to microsomal fractions of root segments from Chlorsulfuron-pretreated seedlings was inhibited by 30%. It is suggested that Chlorsulfuron causes an alteration at the plasmalemma level involving the FC binding sites. The ineffectiveness of Chlorsulfuron in inhibiting FC-stimulaled K+ uptake when administered to excised segments, while inhibiting the enzyme acetolactate synthase, pointed out by Ray (1984) as the site of action of Chlorsulfuron in pea plants, suggests that the observed inhibition of K+ uptake and H+ extrusion is not induced by Chlorsulfuron inhibition of this enzyme. An alternative site of action of Chlorsulfuron is hypothesized in maize plants.  相似文献

Na+ transport across the tonoplast and its accumulation in the vacuoles is of crucial importance for plant adaptation to salinity. Mild and severe salt stress increased both ATP- and PPi-dependent H+ transport in tonoplast vesicles from sunflower seedling roots, suggesting the possibility that a Na+/H+ antiport system could be operating in such vesicles under salt conditions (E. Ballesteros et al. 1996. Physiol. Plant. 97: 259–268). During a mild salt stress, Na+ was mainly accumulated in the roots. Under a more severe salt treatment, Na+ was equally distributed in shoots and roots. In contrast to what was observed with Na+, all the salt treatments reduced the shoot K+ content. Dissipation by Na+ of the H+ gradient generated by the tonoplast H+-ATPase, monitored as fluorescence quenching of acridine orange, was used to measure Na+/H+ exchange across tonoplast-enriched vesicles isolated by sucrose gradient centrifugation from sunflower (Helianthus annuus L.) roots treated for 3 days with different NaCl regimes. Salt treatments induced a Na+/H+ exchange activity, which displayed saturation kinetics for Na+ added to the assay medium. This activity was partially inhibited by 125 μM amiloride, a competitive inhibitor of Na+/H+ antiports. No Na+/H+ exchange was detected in vesicles from control roots. The activity was specific for Na+. since K+ added to the assay medium slightly dissipated H+ gradients and displayed non-saturating kinetics for all salt treatments. Apparent Km for Na+/H+ exchange in tonoplast vesicles from 150 mM NaCl-treated roots was lower than that of 75 mM NaCl-treated roots, Vmax remaining unchanged. The results suggest that the existence of a specific Na+/H+ exchange activity in tonoplast-enriched vesicle fractions, induced by salt stress, could represent an adaptative response in sunflower plants, moderately tolerant to salinity.  相似文献

Abstract. Rates of proton extrusion and potassium (86Rb) influx by intact roots of barley ( Hordeum vulgare cvs . Fergus, Conquest and Betzes) plants were simultaneously measured in short-term (15min) experiments. The nature and extent of apparent coupling between these ion fluxes was explored by manipulating conditions of temperature, pH and cation composition and concentration during flux determinations. In addition, the influence of salt status upon these fluxes was examined. At low K+ concentrations (0.01 to 1 mol m−3), H+ efflux and K+ influx were strongly correlated in both low- and high-K+ roots, although K+: H+ exchange stoichiometries were almost consistently greater than 2:1. At higher concentrations (1 to 5 mol m−3), H+ efflux was either reduced or remained unchanged while K+ influxes increased. In the presence of Na2SO4, rates of H+ extrusion demonstrated similar cation dependence, although below 10 mol m−3 Na2SO4, H+ fluxes were generally 50% lower than in equivalent concentrations of K2SO4. These observations are considered in the context of current hypotheses regarding the mechanisms of k+/H+ exchange.  相似文献

Infection of a murine-spleen dendritic cell line by Listeria monocytogenes was found to induce cell death through apoptosis. To characterize the bacterial product(s) involved in induction of apoptosis, dendritic cells were infected with the L. monocytogenes EGD strain and several isogenic mutants deficient in the production of individual listerial virulence factors. The ability to induce cellular apoptosis was retained by all mutants tested, except the prfA and Δhly mutants, both of which are unable to produce listeriolysin. Apoptosis was also induced by purified listeriolysin suggesting that this protein directly induces apoptosis. Purified recombinant listeriolysins rendered either weakly haemolytic by a C-484 to S mutation, or non-haemolytic by a W-491 to A mutation exhibited little or no capacity to induce apoptosis, indicating that both activities are associated within the same protein region. Treatment with purified listeriolysin or L. monocytogenes infection also triggers apoptosis in explanted bone-marrow dendritic cells. Thus invasion of dendritic cells by L. monocytogenes, which results in cell death, may play an important role in the pathogenesis of listerial infections by impairing immune responses, hindering bacterial clearance and promoting spread of the infection.  相似文献

Abstract An alkaliphilic cyanobacterium characterized as a Synechocystis species was purified from a soil sample taken from a village in Java, Indonesia, by its preferential growth at elevated pH; it grew optimally at pH 9.5. Phosphorus nuclear magnetic resonance studies showed that the organism can maintain a ΔpH of over 2 pH units at an external pH of 10. It was observed that the viability of the organism in the dark was dependent on sodium ions. Evidence from experiments in which the extrusion of Na+ was measured from cells subjected to an alkali shock suggests that the organism possesses a Na+ / H+ electrogenic antiporter which is used for the maintenance of pH homeostasis.  相似文献

Nigericin stimulated the ATPase activity of tightly-sealed membrane vesicles prepared from Hevea brasiliensis Müll.-Arg. lutoïds in the presence of K+. This stimulation required a functioning membrane since it was membrane-bound and since it was not observed for the ATPase activity solubilized from the tonoplast by dichloromethane. The extent of nigericin-induced stimulation of tonoplast ATPase was proportional to the ΔpH collapsed by the ionophore in the presence of K+.  相似文献

Plantago species differ in their strategy towards salt stress, a major difference being the uptake and distribution of Na+ ions. A salt-sensitive ( Plantago media L.) and a salt-tolerant ( P. maritima L.) species were compared with respect to Na+/H+ antiport activities at the tonoplast. After exposure of the plants to 50 m M NaCl for 6 days isolated tonoplast vesicles of P. maritima showed Na+/H+ antiport activity with saturation kinetics and a Km of 2.4 m M Na+, NaCl-grown P. media and the control plants of both species showed no antiport activity. Selectivity of the antiport system for Na+ was high and was determined by adding different chloride salts after formation of a Δ pH in the vesicles. Specific tonoplast ATPase activities were similar in the two species and did not alter after exposure to NaCl stress.  相似文献

The effects of external K+, H+ and Ca2+ concentrations on the intracellular K+ concentration, [K+]i, and the K+-ATPase activity in 2-day-old mung bean roots [ Vigna mungo (L.) Hepper] were investigated. [K+]i, in mung bean roots was markedly decreased by external K+ or H+ stress and did not recover the initial value even after the stress was removed. This decrease in [K+]i, gradually disappeared with the addition of (Ca2+. Ca2+ may offset the harmful effects of ion stress. Ca2+ seems to have two effects on K+ transport; control of K+ permeability and activation of K+ uptake, although K+-ATPase activity was inhibited by Ca2+ concentrations higher than 10–4 M. We suggest that Ca2+ activates K+ uptake indirectly through the acidification of the cytoplasm.  相似文献

Abstract Buffering capacity and membrane H+ conductance were examined in seven Gram-negative species: Aquaspirillum serpens, Pseudomonas aeruginosa, Alcaligenes faecalis, Escherichia coli, Salmonella typhimurium, Proteus mirabilis and Aeromonas hydrophila . All strains of Enterobacteriaceae studied here showed a decrease in both parameters as the external pH increased, over the pH range studied. The other four species presented an increase in buffering capacity and membrane conductance to protons as the external pH increased from 5.5 to 7.0.  相似文献

White lupin ( Lupinus albus L.) is able to grow on soils with sparingly available phosphate (P) by producing specialized structures called cluster roots. To mobilize sparingly soluble P forms in soils, cluster roots release substantial amounts of carboxylates and concomitantly acidify the rhizosphere. The relationship between acidification and carboxylate exudation is still largely unknown. In the present work, we studied the linkage between organic acids (malate and citrate) and proton exudations in cluster roots of P-deficient white lupin. After the illumination started, citrate exudation increased transiently and reached a maximum after 5 h. This effect was accompanied by a strong acidification of the external medium and alkalinization of the cytosol, as evidenced by in vivo nuclear magnetic resonance (NMR) analysis. Fusicoccin, an activator of the plasma membrane (PM) H+-ATPase, stimulated citrate exudation, whereas vanadate, an inhibitor of the H+-ATPase, reduced citrate exudation. The burst of citrate exudation was associated with an increase in expression of the LHA1 PM H+-ATPase gene, an increased amount of H+-ATPase protein, a shift in pH optimum of the enzyme and post-translational modification of an H+-ATPase protein involving binding of activating 14-3-3 protein. Taken together, our results indicate a close link in cluster roots of P-deficient white lupin between the burst of citrate exudation and PM H+-ATPase-catalysed proton efflux.  相似文献

The activity of the H+-pyrophosphatase (H+-PPase) was characterized in microsomes from 24-h-old radish ( Raphanus sativus L., ev. Tondo Rosso Quarantino) seedlings, which are virtually devoid of the tonoplast H+-ATPase. The H+-PPase was localized to membranes which roughly comigrated with the plasma membrane in a sucrose density gradient, but clearly separated from plasma membrane when microsomes were partitioned in an aqueous dextran-polyethylene glycol two-phase system. The H+-PPase activity was strictly dependent on Mg2+ and on the presence of a monovalent cation (K+=Rb+=NH3+Cs+≫Na+Li+) and was insensitive to anions such as Cl−, Br−, NO3− and SO42-. It was inhibited by F−, imidodiphosphate and Ca2+. It had a pH optimum between pH 7.5 and 8.5 and was saturated by low concentrations of pyrophosphate (half saturation at 30 μ M pyrophosphate). All of these characteristics are identical to those reported for the tonoplast H+-PPase from various plant materials. The functional molecular weight of the H+-PPase, measured with the radiation-inactivation technique was 96 kDa.  相似文献

Abstract A method is described for the measurement of auxin-induced H+-efflux from small populations of Avena coleoptile segments. The method allows the simultaneous investigation of the kinetics of rapid auxin responses over a wide concentration range. IAA promoted linear rates of H+-efflux and the change in amplitude of response occurred mainly over a low, narrow concentration range (10–50 μmol m-3). The sigmoidal curve of best-fit to each set of dose-response data was determined using non-linear regression techniques, allowing the objective determination of characteristic tissue sensitivity parameters (RMIN, RMAX, KD and p). The sensitivity parameters for the auxin-type herbicide fluroxypyr are also presented as well as IAA parameters determined in the presence of abscisic acid and the ‘antiauxin’ PCIB. The interpretation of the parameter values and the potential use of sensitivity parameter analysis for the evaluation of theories concerning plant hormone action and interaction is discussed.  相似文献
