【背景】2013年11月在甘肃省兰州市皋兰县的日光温室秋冬茬甜瓜种植棚发现黄萎症状的甜瓜植株,病株率约为1%。【目的】明确甜瓜黄萎病的病原。【方法】采用组织分离法进行病原菌分离;通过科赫氏法则(Koch’s法则)明确分出病菌的致病性;采用形态学和分子生物学方法对病原菌进行种类鉴定。【结果】分离得到轮枝菌属真菌8株,轮枝菌属真菌的病株分出率达100%;2个代表性菌株GLTG-2和GLTG-5(显微特征相似但菌落形态和生长速率不同),在温度18-24℃及昼/夜光周期为11.5 h/12.5 h的试验条件下,人工接种可引起甜瓜苗矮化、枯萎;接种后40 d,枯死株率分别为70%和40%;BLASTn分析结果显示,菌株GLTG-2的rDNA-ITS序列与Verticillium dahliae菌株MRHf7的序列相似性达99.78%,菌株GLTG-5的rDNA-ITS序列与V.dahliae菌株MRHf7和Vd414的序列相似性达100.00%。【结论】引起甜瓜黄萎病的病原菌被鉴定为大丽轮枝菌(V. dahliae),这是大丽轮枝菌引起甜瓜黄萎病在我国和亚洲地区的首次报道。  相似文献

大丽轮枝菌是一种土传性植物病原真菌,可侵染多种植物并引发黄萎病。目前,人们关于大丽轮枝菌的侵染和致病机制的了解还很不深入。本文通过敲除大丽轮枝菌编码丝氨酸/苏氨酸的蛋白激酶基因VdSCH9,阐明了其在大丽轮枝菌生长发育及致病过程中的作用。SCH9基因在酵母中的表达与cAMP-PKA途径和TOR信号通路相关,对酵母的生长、压力响应和寿命等有重要作用。大丽轮枝菌VdSCH9敲除突变体的生长速率显著下降,菌落边缘菌丝更为稀疏,菌丝分枝减少,对棉花植株为害的平均病情指数为56.6,显著低于野生型和互补突变体的平均病情指数90.5和82.8,对茄子植株为害的平均病情指数为65.9,也显著低于野生型和互补突变体的平均病情指数91.1和89.8。另外,敲除突变体对于高渗透压、氧化还原压力、细胞膜和细胞壁完整性等压力条件的敏感性增强。因此,VdSCH9对于大丽轮枝菌的生长、压力响应及致病力均有重要作用。  相似文献

Verticillium dahliae infection leads to Verticillium wilt in cotton and other dicotyledon crops. To reduce the loss of economic crops, more attention has been focused on the key genes involved in pathogenicity of this soil‐borne plant fungal pathogen. Sho1 encodes a conserved tetraspan transmembrane protein which is a key element of the two upstream branches of the HOG‐MAPK pathway in fungi. Sho1 is required for full virulence in a wide variety of pathogenic fungi. In this study, sho1 mutant in V. dahliae (designated ΔVdsho1) was generated by Agrobacterium tumefaciens‐mediated transformation. ΔVdsho1 strain was highly sensitive to menadione (at concentration of 120 μm ) and hydrogen peroxide (at concentration of 250 μm ), displayed delayed spore germination and reduced spore production compared with the wild type and the complemented strains. During infection of host cotton plants, ΔVdsho1 exhibited impaired ability of root attachment and invasive growth. Results from the present work suggest that VdSho1 controls external sensing, virulence and multiple growth‐related traits in V. dahliae and might serve as a potential target for control of Verticillium wilt.  相似文献

The karyotype profile of Verticillium dahliae was resolved by pulse-field gel electrophoresis. It revealed 6 chromosomal bands that corresponded to 7 chromosomes as shown by RFLP analysis using as probe the telomeric consensus sequence (AACCCT)(5). The number of chromosomes was further verified by the sensitivity of the hybridization signals to Bal31 digestion and the exclusion of interfering mitochondrial DNA signals. The corresponding sizes of the seven separated chromosomes were 6.7, 5.6, 4.1, 3.4, 3.1, 3.1 and 2.4Mb, raising the total genomic size of the fungus to approximately 28.4Mb. Twenty five homologous V. dahliae genes obtained either from randomly sequenced clones or PCR amplification were used as hybridization probes and were located onto the seven chromosomes.  相似文献

In the Mediterranean basin, Verticillium Wilt of Olive (VWO) is diffused throughout its range of cultivation, causing severe yield losses and tree mortality. The disease was reported in almost all the Mediterranean and Middle East countries, and in Lebanon it is of increasing significance also on many valuable crops. The disease has already been reported on potato, peach and almond in the Bekaa valley; however, to date no information is available about the incidence of VWO and the inoculum density of Verticillium dahliae microsclerotia in soil of the main agricultural areas of Lebanon. Results from the present investigations demonstrate a high V. dahliae frequency in soils (75.3%), coupled with a mean soil inoculum density of 17.0 MS g?1, clearly indicating a great impact on the production of susceptible hosts in Lebanon, mainly in Bekaa region. Molecular method to assess the microsclerotia inoculum density in soil allowed the detection of a higher frequency of infested soils, as compared with the traditional plating, thus confirming its higher sensitivity. The overall Verticillium wilt prevalence in the inspected olive orchards was 46.2%, and the frequency of V. dahliae‐infected trees was 25.7%. The widespread presence of V. dahliae in all olive growing areas of Lebanon enforces the adoption of measures aimed at reducing the soil inoculum density before any new olive plantation, and the use of strong phytosanitary regulations to improve the certification schemes of propagating material.  相似文献

[目的]从棉花黄萎病真菌Verticillium dahliae中克隆木聚糖酶基因,并在毕赤酵母中进行异源表达,研究酶学性质.[方法]通过多序列比对设计简并引物,扩增出真菌V. dahliae木聚糖酶基因片段,再采用基因组步行PCR技术获得全长木聚糖酶基因序列.经BLAST比对并结合GT-AG原则分析,该基因含有一个大小为63 bp的内含子,利用DpnI介导的缺失方法对含内含子的全长木聚糖酶基因进行剪接,获得该基因的全长cDNA.将克隆到的cDNA在毕赤酵母GS115进行了表达,重组酶经纯化后进行酶学性质分析.[结果]BLAST比对显示,该cDNA推测的氨基酸序列和已知木聚糖酶的最高一致性为72%.测得该酶最适反应温度为45℃,最适反应pH值为6,在pH5-9维持50%以上的活性,对山毛榉材木聚糖具有最好的水解效果.Mg2 和Ca2 对酶有激活作用,分别提高了33.7%和16.6%,EDTA,β-巯基乙醇和NaN3对酶的活性基本没有影响,Tween-80和DMSO使酶活性提高了28.4%和12.8%.[结论]本文从引起棉花黄萎病的真菌V. dahliae中克隆到的木聚糖酶基因是在GenBank上登录的第一个来自棉花黄萎病真菌的木聚糖酶基因序列.本文所用的克隆方法可以高效的从植物病原真菌和白腐真菌克隆只含一个内含子的11家族的新木聚糖酶基因,避免了摸索原始菌株酶表达诱导条件,检测酶的活性等繁琐的操作.酶学性质分析显示该酶在低聚木糖的制备,面包改良上有潜在的应用价值.  相似文献

大丽轮枝菌微菌核的萌发条件及致死温度   总被引:2,自引:0,他引:2  
微菌核是大丽轮枝菌在土壤中的主要存活结构和初侵染源,在土壤中可存活14年之久,其数量及存活状况直接影响着大丽轮枝菌型黄萎病的发生为害程度。以棉花黄萎病菌Verticillium dahliae XJ2008菌株为试材,研究了微菌核萌发的最佳条件、致死温度及土壤温度对微菌核存活的影响。结果表明,20℃、pH8.0是微菌核萌发的最佳条件。大丽轮枝菌微菌核具有很强的耐高温特性,随着处理时间的延长,微菌核的萌发率呈下降趋势,在55℃及以上处理时其萌发率下降迅速,而在50℃及以下处理时其萌发率下降相对较慢,在55℃处理360min可使微菌核完全致死,而在40℃、45℃和50℃处理1,440min,仍然有少量的微菌核存活,但其萌发率随着时间的推移呈下降趋势。土壤微菌核模拟试验结果表明,土壤温度对微菌核有很强的致死作用,40℃条件下处理4d后土壤中的微菌核已全部死亡。该研究结果为通过覆膜增温防治作物黄萎病提供了理论依据。  相似文献

利用硫酸铵沉淀、(A)KTA explorer 10蛋白纯化仪、非变性电泳、割胶电洗脱等方法,从大丽轮枝菌(Verticillium dahliae)发酵液中分离纯化出一种蛋白激发子,经SDS-PAGE电泳检测单一条带,相对分子量为20 kD.该蛋白激发子能够诱导烟草的过敏反应,处理6h后,处理部位出现水溃状,24h后出现坏死斑.该激发子可以诱导烟草细胞在较短时间内产生防卫反应信号分子H2O2和NO,并引起活性氧爆发.  相似文献

利用离体抑菌圈法从茄子根际土壤筛选出8株对黄萎病菌大丽轮枝菌(Verticillium dahliae)抑菌效果明显的芽孢杆菌,其中F53菌株分泌几丁质酶和-β1,3-葡聚糖酶,对所检测的12种植物病原菌具明显的抑制作用,其发酵液热、酸碱稳定性强,用F53菌株制成的菌剂对茄子黄萎病盆栽试验防效为52.4%~75.8%,初步鉴定F53菌株为环状芽孢杆菌(Bacillus circulans)。  相似文献

  • GFP‐tagged V. dahliae isolates with different pathogenic abilities were used to analyse the colonisation and infection of V. dahliae in the roots and leaves of different upland cotton cultivars, the relationships among infection processes, the immune responses and the resistance ability of different cultivars against V. dahliae.
  • Here, we report a new infection model for V. dahliae in upland cotton plants. V. dahliae can colonise and infect any organ of upland cotton plants and then spread to the entire plant from the infected organ through the surface and interior of the organ.
  • Vascular tissue was found to not be the sole transmission route of V. dahliae in cotton plants. In addition, the rate of infection of a V. dahliae isolate with strong pathogenicity was notably faster than that of an isolate with weak pathogenicity. The resistance of upland cotton to Verticillium wilt was related to the degree of the immune response induced in plants infected with V. dahliae. These results provide a theoretical basis for studying the mechanism underlying the interaction between V. dahliae and upland cotton. 微丝在真菌生长发育、胞质分裂等生命过程中具有重要功能。通过农杆菌介导遗传转化方法,将荧光mCherry标记微丝的表达载体pSULPH-Lifeact-mCherry转入大丽轮枝菌(Verticillium dahliae Kleb.)野生型V592,获得稳定的微丝荧光标记菌株V592/Lifeact-mCherry,并检测了其生物学表型和孢子萌发、菌丝生长等过程中的微丝荧光动态变化。结果表明:微丝荧光标记菌株的菌落形态、生长速率、产孢量、萌发率等表型与野生型没有显著差异;且可以观察到微丝荧光信号在分生孢子和菌丝的顶端及隔膜都有清晰定位,同时对该菌株隔膜形成过程微丝动态观察发现,微丝参与胞质分裂进程中肌动球蛋白收缩环CAR (Contractile actomyosin ring)的形成。微丝荧光标记菌株可用于微丝在真菌发育中的动力学研究,这为深入研究微丝在真菌发育及致病过程中的作用机制提供理论与实践支撑。  相似文献

In August 1994 verticilliosis-like symptoms were noticed in a maturing crop of sunflower at IACR-Rothamsted. Wilting plants with chlorotic areas on some leaves, and dark areas of microsclerotia at the base of stems, were first observed on 11 August. As the plants matured, dark stripes were also observed on the stems. Many stems became brittle and later collapsed. Infected xylem showed a brown discolouration, later turning to black, and microsclerotia developed. Verticillium dahliae was isolated from all parts of the plant.
In general, once symptoms were present, the incidence of verticilliosis-affected plants increased, and the disease developed, more rapidly in early maturing varieties than in the later maturing types. At harvest, incidence ranged between 24.5% and 89.8%, depending on variety. Early maturing varieties appeared to be more susceptible than later maturing types.
Isolates of V. dahliae were cultured in the laboratory. Sunflower plants were artificially inoculated with these isolates and developed verticilliosis symptoms when grown under controlled environmental conditions.  相似文献

Verticillium wilt of olive (Olea europaea L.), caused by Verticillium dahliae, is nowadays the most serious olive disease in Spain. The disease increments are being observed particularly in young olive plantations, favoured by several factors including inadequate cultural practices and crop production intensification, such as irrigation. Thus, three olive orchards affected by Verticillium wilt, with disease incidence ranging 30–50%, were selected to determine if the drip irrigation could favour the increase of pathogen in soil. Pathogen in soil was quantified in wet zones around the drippers and in dry zones out of them. Inoculum density in all experiments was higher in wet than in dry areas. After 4 months of watering, soil pathogen population increased considerably in wet and dry areas but inoculum density remained higher in the wet soil.  相似文献
