在过去的半个世纪中,森林生态系统物质能量流动模型的发展使得生态学者对矿物元素的来源、去处、转换速率及其干扰破坏的影响有了进一步的认识.其中"春坝"假设提出在早春来临之际,林冠树木仍处于冬眠之时,林下地被植物先于林冠树木发芽开花,吸收土壤养分于植物体内,使得养分免于因雨水或雪水而造成的淋失而得以保留于森林生态系统中.本文对"春坝"假设的研究作一小结.现有的研究结果显示,早春地被植物对土壤氮的吸收量因不同生态系统而异;而土壤微生物对氮的吸收量远高于过去人们所认识的水平.早春地被物与土壤微生物都能在季节的变化中吸收土壤养分、共同起着"春坝"作用.两者所起作用大小因气候及生态系统的不同而有所改变.  相似文献

asCP, the unique green fluorescent protein-like nonfluorescent chromoprotein from the sea anemone Anemonia sulcata, becomes fluorescent ("kindles") upon green light irradiation, with maximum emission at 595 nm. The kindled protein then relaxes to a nonfluorescent state or can be "quenched" instantly by blue light irradiation. In this work, we used asCP mutants to investigate the mechanism underlying kindling. Using site-directed mutagenesis we showed that amino acids spatially surrounding Tyr(66) in the chromophore are crucial for kindling. We propose a model of the kindling mechanism, in which the key event is chromophore turning or cis-trans isomerization. Using site-directed mutagenesis we also managed to transfer the kindling property to the two other coral chromoproteins. Remarkably, most kindling mutants were capable of both reversible and irreversible kindling. Also, we obtained novel variants that kindled upon blue light irradiation. The diversity of photoactivated fluorescent proteins that can be developed by site-directed mutagenesis is promising for biotechnological needs.  相似文献

Based on striking parallels between the cell kinetics in the epidermal lesion of psoriasis and the proliferation of hair matrix keratinocytes during the anagen phase of the hair growth cycle, the hypothesis is proposed that both phenomena may share the same "switch-on" mechanism. Particular emphasis is placed on a comparison between the Koebner phenomenon in psoriasis and wounding-induced anagen hair growth. In discussing alternative theoretical models for the proposed common "switch-on" mechanism, some useful experimental tools are suggested. Research into the mechanisms which control epithelial proliferation in psoriasis and hair growth may provide new insights into other growth processes, such as embryonic organogenesis and neoplasia, in which similar epithelial-mesenchymal interactions play a pivotal role.  相似文献

In this article is presented the result of the experiments on mice-hybrids F1(CBA x C57B1/6), which indicates the presence of the reaction of "ischemia/reperfusion" for stem cells of two "critical" cell renewal systems of organism (bone marrow and intestinal epithelium) during the irradiation under the conditions of hypoxic radioprotector application. The additional injection of the source of nitric oxide radicals-sodum nitroprusside (SNT) to the mice right after the irradiation under the conditions of hypoxic protection by serotonin, resulted the substantial increase of the survival rate of hematopoietic stem cells (registered by the methods of endogenous and exogenous colony forming in spleen) and stem cells of intestinal epithelium (registered by the method of intestinal "microcolonies"). The similar radioprotective effect was also registered during the test of survival rate of mice under tests of "bone marrow" and "intestinal" forms of radiation lethality that is evidence of the importance of the realization of phenomenon "ischemia/reperfusion" in the reaction of whole organism on the acute radiation injury. As SNP weakens the manifestation apoptosis and necrosis through competition with active forms of oxygen (AFO) during the period of "reperfusion" on basis of the found out phenomenon experimental model for studying mechanisms of stem cells damage in vivo induced by AFO and for the search of the modifiers weakening or strengthening such damage can be developed.  相似文献

Cortisone administered once at 100 mg/kg during the first 3 weeks of infection inhibited rapid expulsion. In rats immunized with an abbreviated infection (T/M regime) inhibition averaged approximately 50%, whereas in rats given a complete infection (C.I.) 14% inhibition occurred. Sensitivity to 400 rad whole-body irradiation was greatest 7 days before a challenge infection in all immune rats. Three days after beginning the T/M infection rats were highly susceptible to cortisone but only weakly so to irradiation. Rats immunized by C.I. were equally, but only weakly, susceptible to either cortisone or irradiation 3 days after infection. Acute administration of cortisone 1 or 4 hr prior to challenge did not inhibit rapid expulsion but 60% inhibition occurred when cortisone was given 24 hr prior to challenge. Inhibition of rapid expulsion by irradiation 7 days prior to challenge was not reversed by immune serum and irradiation did not affect antibody titer in treated rats. It was suggested that irradiation 7 days before challenge compromised the intestinal, and not the immunological, component of rapid expulsion. Differences in sensitivity of "early" and "late" rapid expulsion to irradiation and cortisone therapy provide further evidence of functional differences between these rejection processes.  相似文献

The mechanisms by which hyperthermophilic Archaea, such as "Pyrococcus abyssi" and Pyrococcus furiosus, survive high doses of ionizing gamma irradiation are not thoroughly elucidated. Following gamma-ray irradiation at 2,500 Gy, the restoration of "P. abyssi" chromosomes took place within chromosome fragmentation. DNA synthesis in irradiated "P. abyssi" cells during the DNA repair phase was inhibited in comparison to nonirradiated control cultures, suggesting that DNA damage causes a replication block in this organism. We also found evidence for transient export of damaged DNA out of irradiated "P. abyssi" cells prior to a restart of chromosomal DNA synthesis. Our cell fractionation assays further suggest that "P. abyssi" contains a highly efficient DNA repair system which is continuously ready to repair the DNA damage caused by high temperature and/or ionizing radiation.  相似文献

Intracoronary beta irradiation (use of beta radiation for intracoronary irradiation) is an effective method in reducing neointimal proliferation after successful angioplasty and stent implantation. However, long-term results may be influenced by absolute dose and by the homogeneity in dose distribution. In our study, we investigated dose perturbation due to the presence of a conventional guide wire during irradiation. The Galileo III centering catheter and P-32 beta source were used. The 55 MD GAF Chromic foil was positioned within a phantom made of PMMA. The dose distribution at cylindrical surfaces has been assessed using GAF Chromic dosimetric foil MD55 (Nuclear Associates, USA). Our study demonstrated the significant dose reduction of 46% in the most "shaded" area. The dose reduction to 80% or less occupy the 60 degrees sector. This phenomenon can cause progression of late restenosis. In conclusion, the results suggest that technical improvements in centering catheter construction should be made to eliminate the "shielding" effect of the guide wire.  相似文献

During the first entering into hibernation in the ground squirrels there are periodic cessations of the process, especially during the transition to moderate and deep torpidity ("critical" periods) which significantly increase energy expenditure in animals during the phase of entering. During this period, high amplitude near-the-hour fluctuations are observed in total duration of wakefulness and sleep, contractile muscle activity, oxygen consumption, heart rate, brain temperature, and thyroid hormone concentration in the blood; at the end of this period, as well as during the exiting from it the duration of deep slow wave sleep significantly increases, paradoxical sleep is not identified. The data obtained confirm a hypothesis of "kindling" homeostasis as neuro-hormonal mechanism of the increase in thermal range of the activity of functional systems of the organism during the entering into torpidity in homoiothermic animals.  相似文献

It has been shown, that the total X-ray irradiation in the dozes of 0.5 and 1 Gy influences on the content of lipid peroxidation products and enzymatic activity of antioxidant system in rat spleen and thymus cells. The influence of preparations "AMMIVIT" and "Ceruloplasmin" on these processes is investigated also. So, the animals feeding by the vitamin concentrate "AMMIVIT" have lead to increase of MDA level (a final product of lipid peroxidation) and the overactivity of some antioxidant enzymes in rat spleen and thymus cells. Injection of the preparation "Ceruloplasmin" to experimental animals up 1 hour before the irradiation has normalized LPO intensity and activity of AO enzymes.  相似文献

We employed an associative learning paradigm to test the hypothesis that exercise hyperpnea in humans arises from learned responses forged by prior experience. Twelve subjects undertook a "conditioning" and a "nonconditioning" session on separate days, with order of performance counterbalanced among subjects. In both sessions, subjects performed repeated bouts of 6 min of treadmill exercise, each separated by 5 min of rest. The only difference between sessions was that all the second-to-penultimate runs of the conditioning session were performed with added dead space in the breathing circuit. Cardiorespiratory responses during the first and last runs (the "control" and "test" runs) were compared for each session. Steady-state exercise end-tidal PCO(2) was significantly lower (P = 0.003) during test than during control runs for both sessions (dropping by 1.8 +/- 2 and 1.4 +/- 3 Torr during conditioning and nonconditioning sessions, respectively). This and all other test-control run differences tended to be greater during the first session performed regardless of session type. Our data provide no support for the hypothesis implicating associative learning processes in the ventilatory response to exercise in humans.  相似文献

Episodic memory, which depends critically on the integrity of the medial temporal lobe (MTL), has been described as "mental time travel" in which the rememberer "jumps back in time." The neural mechanism underlying this ability remains elusive. Mathematical and computational models of performance in episodic memory tasks provide a specific hypothesis regarding the computation that supports such a jump back in time. The models suggest that a representation of temporal context, a representation that changes gradually over macroscopic periods of time, is the cue for episodic recall. According to these models, a jump back in time corresponds to a stimulus recovering a prior state of temporal context. In vivo single-neuron recordings were taken from the human MTL while epilepsy patients distinguished novel from repeated images in a continuous recognition memory task. The firing pattern of the ensemble of MTL neurons showed robust temporal autocorrelation over macroscopic periods of time during performance of the memory task. The gradually-changing part of the ensemble state was causally affected by the visual stimulus being presented. Critically, repetition of a stimulus caused the ensemble to elicit a pattern of activity that resembled the pattern of activity present before the initial presentation of the stimulus. These findings confirm a direct prediction of this class of temporal context models and may be a signature of the mechanism that underlies the experience of episodic memory as mental time travel. ? 2012 Wiley Periodicals, Inc.  相似文献

The influence of sublethal doses of ionizing radiation on cuticular structures of the reproductive apparatus of the large fruit-tree tortrix moth was studied. The variability of cuticular teeth of the aedeagus and cuticular projections of the antrum was studied in laboratory cultures after irradiation and in the control. After irradiation of the pupae, numerous protuberances or irregularly arranged small denticles were observed over the entire cuticle of the genitalia. Additional teeth appeared on the aedeagus. The lateral tooth of the aedeagus was more strongly affected by irradiation than the apical one. The influence of irradiation is mostly manifested within the first 24 h of pupa formation. The functional significance of teeth of the aedeagus and projections of the antrum during mating is considered. Based on the variability of aedeagus teeth and external cuticular projections at the antrum inflexion, the applicability of the “lock-and-key” hypothesis to evolution of copulation mechanism in the large fruit-tree tortrix moth is considered.  相似文献

According to the "stop-and-go" hypothesis of slow axonal transport, cytoskeletal and cytosolic proteins are transported along axons at fast rates but the average velocity is slow because the movements are infrequent and bidirectional. To test whether this hypothesis can explain the kinetics of slow axonal transport in vivo, we have developed a stochastic model of neurofilament transport in axons. We propose that neurofilaments move in both anterograde and retrograde directions along cytoskeletal tracks, alternating between short bouts of rapid movement and short "on-track" pauses, and that they can also temporarily disengage from these tracks, resulting in more prolonged "off-track" pauses. We derive the kinetic parameters of the model from a detailed analysis of the moving and pausing behavior of single neurofilaments in axons of cultured neurons. We show that the model can match the shape, velocity, and spreading of the neurofilament transport waves obtained by radioisotopic pulse labeling in vivo. The model predicts that axonal neurofilaments spend approximately 8% of their time on track and approximately 97% of their time pausing during their journey along the axon.  相似文献

The craniofacial haft resists forces generated in the face during feeding, but the importance of these forces for the form of the craniofacial haft remains to be determined. In vivo bone strain data were recorded from the medial orbital wall in an owl monkey (Aotus), rhesus macaques (Macaca mulatta), and a galago (Otolemur) during feeding. These data were used to determine whether: the interorbital region can be modeled as a simple beam under bending or shear; the face is twisting on the brain case during unilateral biting or mastication; the interorbital "pillar" is being axially compressed during incisor loading and both axially compressed and laterally bent during mastication; and the interorbital "pillar" transmits axial compressive forces from the toothrow to the braincase. The strain data reveal that the interorbital region cannot be modeled as a anteroposteriorly oriented beam bent superiorly in the sagittal plane during incision or mastication. The strain orientations recorded in the majority of experiments are concordant with those predicted for a short beam under shear, although the anthropoids displayed evidence of multiple loading regimes in the medial orbital wall. Strain orientation data corroborate the hypothesis that the strepsirrhine face is twisted during mastication. The hypothesis that the interorbital region is a member in a rigid frame subjected to axial compression during mastication receives some support. The hypothesis that the interorbital region is a member in a rigid frame subjected to lateral bending during mastication is supported by the epsilon1/absolute value epsilon2 ratio data but not by the strain orientation data. The timing of peak shear strains in the medial orbital wall of anthropoids does not bear a consistent relationship to the timing of peak shear strain in the mandibular corpus, suggesting that bite force is not the only external force influencing the medial orbital wall. Strain orientation data suggest the existence of two distinct loading regimes, possibly associated with masseter or medial pterygoid contraction. Regardless of the loading regime, all taxa showed low strain magnitudes in the medial orbital wall relative to the anterior root of the zygoma and the mandibular corpus. The strain gradients documented here and elsewhere suggest that, in anthropoids at least, local effects of external forces are more important than a single global loading regime. The low strain magnitudes in the medial orbital wall and in other thin bony plates around the orbit suggest that these structures are not optimally designed for resisting feeding forces. It is hypothesized that their function is to provide rigid support and protection for soft-tissue structures such as the nasal epithelium, the brain, meninges, and the eye and its adnexa. In contrast with the face of Otolemur, which appears to be subjected to a single predominant loading regime, anthropoids may experience different loading regimes in different parts of the face. This implies that the anthropoid and strepsirrhine facial skulls might be optimized for different functions.  相似文献

Pea seeds (Pisum sativum L.) from the seed lot with 80% germinability were separated in fractions according to room temperature phosphorescence: strong seeds were assigned to fraction I, and weak seeds formed to fraction II. During imbibition, the seeds of fraction II exhibited twofold higher rates of water uptake and experienced hypoxia. Some of these seeds suffocated from hypoxia, and other seeds produced seedlings with morphological defects (such seeds were considered incapable of germination). One week after irradiation with the dose of 3 Gy, germination percentage decreased to 45%, which was caused by the increase of number of weak seeds. The germinability of seeds subjected to gamma-irradiation at doses of 7 and 10 Gy was similar to that of control seeds. In these sub-lots, there appeared so-called "improved" seeds, which were similar to non-irradiated seeds in terms of phosphorescence level, the rate of water uptake and germination percentage. It was shown with the use of PCMB that the difference in the rates of water uptake by seeds of fraction I and II depended on the permeability of cell membranes. The permeability was determined by the state of aquaporins ("open"-"closed"). The experiments with phosphatase inhibitor (NaF) shown that in seeds irradiated with dose of 3 Gy (fraction II), the mechanism of aquaporins closing was broken (phosphatase was inactivated). In "improved" seeds (after irradiation with dose of 10 Gy), aquaporins were closed irreversibly in air-dry state, when aquaporin dephosphorylation was unlikely. It was concluded that the abnormal increase (following the initially decrease) in germination of pea seeds after irradiation can be explained without invoking the hypothesis on hyper function of reparatory mechanism of at low doses of irradiation.  相似文献

Psychophysiological data support the concept that migraine is the result of cortical hypersensitivity, hyperactivity, and a lack of habituation. There is evidence that this is a brain-stem related information processing dysfunction. This cortical activity reflects a periodicity between 2 migraine attacks and it may be due to endogenous or exogenous factors. In the few days preceding the next attack slow cortical potentials are highest and habituation delay experimentally recorded during contingent negative variation is at a maximum. These striking features of slow cortical potentials are predictors of the next attack. The pronounced negativity can be fed back to the patient. The data support the hypothesis that a change in amplitudes of slow cortical potentials is caused by altered habituation during the recording session. This kind of neurofeedback can be characterized as empirically based because it improves habituation and it proves to be clinically efficient.  相似文献
