Carbohydrate levels and activities of enzymes associated with starch, sucrose and sorbitol metabolism were assayed in leaves of peach [ Prunus persica (L.) Batsch cv. Redhaven] of different ages, in order to examine developmental changes in leaf carbohydrate metabolism. Dry matter, soluble protein, chlorophyll and the activities of key enzymes of the reductive pentose phosphate pathway increased during leaf development. The levels of leaf carbohydrates, especially sorbitol and starch, also increased. Changes of starch levels were related to increases in the activities of enzymes associated to starch metabolism, such as ADPglucose-pyrophosphorylase (E.C. 2.7.7.27) and amylase (E.C. 3.2.1.1. plus E.C. 3.2.1.2). The activities of enzymes involved in sucrose and sorbitol degradation decreased during leaf development, whereas the activities of aldose-6-phosphate reductase (E.C. 1.1.1.200) and cytosolic fructase-1,6-bisphosphatase (E.C. 3.1.3.11) increased. In contrast, the activity of sucrose-phosphate synthase (E.C. 2.4.1.14) did not vary in a significant manner. The results suggest that the ability to synthesize and utilize both sucrose and sorbitol changes as peach leaves mature, and also that there are differences in metabolism of these two transport sugars during leaf development.  相似文献

Simple sequence repeats (SSRs) have proven to be highly polymorphic, easily reproducible, codominant markers. However, developing an SSR map is very time consuming and expensive, and most SSRs are not specifically linked to gene loci of immediate interest. The ideal situation would be to combine a high-throughput, relatively inexpensive mapping technique with rapid identification of SSR loci in mapped regions of interest. For this reason, we coupled the high-throughput technique of AFLP mapping with subsequent direct targeting of SSRs identified in AFLP-marked regions of interest. This approach relied on the availability of peach bacterial artificial chromosome (BAC) library resources. We present examples of using this strategy to rapidly identify SSR loci tightly linked to two important, simply inherited traits in peach (Prunus persica (L.) Batsch): root-knot nematode resistance and control of the evergrowing trait. Shoots were regenerated from the proximal region of immature cotyledons (with the embryonic axis removed) of Prunus persica (peach) and from the same area in mature cotyledons of P. domestica (plum) and P. cerasus (sour cherry) on MS medium containing (in mgl-1) thiamine-HCl, 0.4; nicotinic acid, 0.5; pyridoxine-HCl, 0.5; sucrose, 25 000; and 0.7% agar. The medium was supplemented with 0.0–2.5 M indole-butyric acid and 5–12.5 M thidiazuron. Cultures were incubated at 24 °C under 16h photoperiod. Shoots regenerated adventitiously over a broad range of thidiazuron concentrations and 2.5 M indole-butyric acid in 35 days. The presence of the embryonic axis inhibited the development of shoots. Regenerated shoots of peach and plum were rooted on half-strength MS inorganic semi-solid medium with 2.5–5.0 M indole-butyric acid. Yields of 106–108 peach mesophyll cells and protoplasts · gfw-1 were obtained depending on factors such as digesting enzymes, and leaf size. Onozuka R-10 (2%) in combination with Macerase (0.5%) was found best for protoplast isolation and mediocre for cell isolation among several enzyme combinations tested. Viability was 90% for protoplasts and 60% for cells. Pectolyase Y23 was found to be ineffective in our investigation. Small leaves, 4–10 mm in length, were a superior source for protoplast isolation than medium or big expanded leaves, 22–30 mm in length. The high yields of protoplasts could be obtained only when keeping the ratio of leaf biomass to volume of digesting enzyme solution under 20 mg ml-1. Purification of protoplasts on a sucrose gradient yielded about 107 protoplasts · gfw-1, however, the preparation was still contaminated by intact cells. Protoplasts were cultured under different growth regulators and physical conditions. Subcellular localization of indole-3-acetic acid (IAA) in leaf cells of peach ( Prunus persica [L.] Batsch 'Hakuho') was investigated using imuno-gold electron microscopy. The distribution pattern of the gold particles, which detected IAA, changed as cells matured. The most prominent feature was the accumulation of the gold label in the chloroplasts and mitochondria of the parenchyma cells of opened leaves. Throughout the development, the cytosol, nuclei, and cell wall were labelled, although the level was low and no significant changes occurred. The density of colloidal gold at each stage of leaf development was well correlated with the analytical data obtained by high-performance liquid chromatography (HPLC).  相似文献

Six peach and six nectarine cultivars were evaluated for the phenolic content in their pulp and peel tissues. Chlorogenic acid, catechin, epicatechin, rutin and cyanidin-3-glucoside were detected as the main phenolic compounds of ripened fruits. The concentration was always higher in peel tissue, with average values ranging from 1 to 8 mg g−1 dry weight (DW) depending on cultivar. Of the tested varieties, the white-flesh nectarine 'Silver Rome' emerged as the cultivar with the highest amount of total phenolics. Phenolic compounds were also profiled during fruit growth and ripening in the yellow nectarine cv. 'Stark Red Gold', which showed a decreasing concentration during fruit development in both peel and pulp tissues. Average amounts of total phenolics were approximately 25 mg g−1 DW 60 days after full bloom and decreased to 3 mg g−1 DW at ripening in pulp tissue. Differences among peel and pulp composition show the different dietetic and antioxidant potential of fruits consumed unpeeled and peeled.  相似文献

Fruit ripening in Prunus persica involves a number of physiological changes, being one of the most significant the mesocarp softening in melting varieties. In order to get a better understanding of the molecular processes involved in this phenomenon, the protein accumulation patterns in firm and soft fruit of three peach and two nectarine melting flesh varieties were assessed using 2D gel analysis. A General Linear Model (GLM) two-way analysis of variance determined that 164 of the 621 protein spots analyzed displayed a differential accumulation associated with the softening process. Among them, only 14 proteins changed their accumulation in all the varieties assessed, including proteins mostly involved in carbohydrates and cell wall metabolism as well as fruit senescence. The analysis among varieties showed that 195 and 189 spots changed within the firm and soft fruit conditions, respectively. Despite the changes in relative abundance in the spot proteins, the proteome is conserved among varieties and during the transition from firm to soft fruit. Only two spots proteins exhibited a qualitative change in all the conditions assessed. These results are in agreement with the notion that Prunus persica commercial varieties have a narrow genetic background.  相似文献

Cuticular and cellular components of leaves of Citrus aurantifolia Swingle were tested for toxicity to Gloeosporium limetticola Clausen, responsible for the withertip disease. Cuticular waxes isolated from mature, resistant leaves showed little activity. A dimethoxy furocoumarin, isopimpinellin, isolated from the cellular components, was highly toxic, and ester and acidic fractions obtained from both young susceptible and mature resistant leaves showed some activity. The contents in the leaves and the toxicities in vitro of the active constituents suggest that they may contribute, to an important degree, to the defence of the leaf against G. limetticola.  相似文献

Stomatal behaviour and the amount and synthesis of epicuticular waxes were studied in maize leaves of various degrees of chilling resistance in order to investigate the rapid desiccation characteristic of chilling injury. In contrast to the resistant variety (KSC 360), the sensitive line (Lye) failed to close the stomata during chilling treatment. Both, the amount of epicuticular waxes and their degree of labeling by 1-14C-acetate were much higher in the resistant than in the sensitive leaves. Chilling-sensitivity was also related to a suppressed conversion of free fatty acids into free primary alcohols.  相似文献

The ontogeny of the peach seed and its organs correlates wellwith the stages of growth of the pericarp of peach fruit. Theinitial stages of rapid f.wt (FW I) and d.wt (DW I) increasecoincide with the period of rapid f.wt increase of the wholeseed and rapid size increase of the endosperm respectively.The period of slow f.wt increase of the fruit (FW II) coincideswith the period of rapid size increase of the embryo. The seed and pericarp compete strongly for assimilates throughoutfruit development and the seed is the weaker competitor. Thereis no period however when the growth of the seed or one of itsorgans inhibits the sink strength of the pericarp to allow theseed to develop. The data presented indicate that the periodsof high nutrient demand (DW I and DW III) are created in thepericarp in response to a stimulatory signal from the seed andthe period of low nutrient demand (DW II) results from the absenceof this signal after the endosperm reaches full size.  相似文献

BACKGROUND AND AIMS: Prunus, subgenus Padus, exhibits two completely different calcium oxalate crystal macropatterns in mature leaves. Foliar macropattern development has been described previously in P. virginiana, representing one version. Prunus serotina, in the group exhibiting the second macropattern, is described here. The goal was to describe developmental details for comparison with P. virginiana, and to extend the sparse current knowledge of crystal macropatterns. METHODS: Leaves at various developmental stages were removed from local trees and from herbarium specimens. Early leaf stages and freehand leaf and stem sections were mounted directly in aqueous glycerine; larger leaves were processed whole or in representative pieces in household bleach, dehydrated in alcohol/xylol, and mounted in Permount. Crystals were detected microscopically between crossed polarizers. KEY RESULTS: Bud scales have a dense druse population. Druses appear first at the stipule tip and proliferate basipetally but soon stop forming; growing stipules therefore have a declining density of druses. Druses appear at the tip of leaves <1 mm long, then proliferate basipetally in the midrib. Lamina druses appear in the distal marginal teeth of leaves 3 cm long; from here they proliferate basipetally and towards midrib along major veins. In about two-thirds-grown leaves (6-9 cm length) druses are all adaxial to veins of most orders; a shift occurs then to formation of prisms, which appear first abaxial to, then all around, veins. Mature leaves have virtually all prisms encrusting all major veins, more sparsely along smaller minor veins. Late season leaves form epitactic crystals on existing prismatics. CONCLUSIONS: The developing and mature macropattern of P. serotina is almost the reverse of the pattern described previously in P. virginiana, and shows that two closely related species can develop radically different modes of crystallization. The few detailed macropattern studies to date reveal striking variations that indicate a new level of organization that must be integrated with the anatomical, physiological and molecular approaches that have been dominant so far.  相似文献

Water permeability and composition of soluble cuticular lipids of isolated cuticular membranes from leaves of Citrus aurantium L. were investigated for 3 successive years. The average water permeability coefficient determined using 169 cuticular membranes was 1.09·10–7 cm s–1 with a standard deviation of 0.78·10–7 cm s–1. There were no significant differences in water permeability between years. Cuticular membranes are characterized by a great variability in water permeability both within and between years. Both water permeability of individual membranes and variability between membranes are shown to be determined by soluble cuticular lipids contained within the cuticular membranes. The soluble cuticular lipids of Citrus leaves are composed of fatty acids, primary alcohols, esters, and hydrocarbons. They occur in amounts of 9.84 g cm–2, which represents approx. 3% of the total mass of isolated cuticular membranes. The specific weight of cuticular membranes (365.4 g cm–1) and total amount of soluble cuticular lipids did not vary significantly between years. Significant differences were observed for the amounts and composition of the constituent classes of lipids. Six homologues comprise 86% of the fatty acids (C16; C18; C19; C21; C24; C26), 83% of the primary alcohols (C24; C26; C28; C30; C32; C34) and 88% of the esters (C36; C38; C40; C41; C42; C44). Eleven major homologues amount only to 62% of the total hydrocarbons (C16; C17; C18; C20; C26; C27; C29; C30; C31; C32; C33). Variability in the composition of soluble cuticular lipids between years was much smaller than variability of water permeability and, therefore, no relation between composition of soluble cuticular lipids and water permeability could be found. A study of the polyamine profile was carried out during zygotic embryo development in Prunus avium. Zygotic embryos were collected from 2 donor trees and sorted into 3 size classes: C1 [2.5 to 3.5 mm]; C2 [3.6 to 4.5 mm] and C3 [5.5 to 7 mm]. Evolution of the various polyamines was similar for the two donor trees. Changes in the relative amount of the various free polyamines were observed during zygotic embryo development. Agmatine and spermine levels increased from C1 to C3. Spermidine, the predominant polyamine, showed a two-fold decrease in C3 compared with C1 and C2; the evolution of putrescine was opposed, showing an increase in the last developmental stage. The putrescine/spermidine ratio could be a marker for these 3 developmental stages with a higher ratio in C3 compared with C1 and C2. Polyamine changes in cotyledons from class C1 were investigated during in vitro culture. A 10-day induction on a medium containing 2,4-dichlorophenoxyacetic acid (2,4-D) and kinetin caused a strong decline in free spermidine levels and a dramatic increase in free putrescine. The effect of light on peach leaf and flower bud break was examined. It was found that leafless dormant shoots were light-perceptive organs. Darkness, after light preconditioning during dormancy, reduced leaf bud opening; however, light was obligatory when the shoots were preconditioned in the dark. Relatively short exposures to light were sufficient to stimulate leaf bud break. Terminal buds were less inhibited by darkness than were laterals. Flower bud break was inhibited in light after dark preconditioning. The red region of the spectrum was found to be active; the phytochorome system seems to be involved in the light reactions, as the red light effect was reversible with subsequent far-red illumination. Supplementary light, producing long-day conditions, could partly compensate for insufficient chilling. A possible sequence of reactions in the plant is suggested.  相似文献

An n-hexane extract of fresh, mature leaves of Ludwigia adscendens, containing a thin layer of epicuticular waxes, has been analysed for the first time by TLC, IR and GC using standard hydrocarbons. The leaves contained 22 identified long chain (C15-C36) n-alkanes, accounting for 74.27% of the hydrocarbons present, and an unknown number of unidentified branched chain alkanes. The predominant n-alkane was C25 (11.02%), whilst C18 (7.62%), C20 (6.14%), C29 (5.36%) and C27 (5.29%) n-alkanes were moderately abundant: the C35 homologue was present only in minor amounts (0.22%).  相似文献

The potential for introgression of Prunus davidiana, a wild species related to peach, was evaluated with respect to problems of non-Mendelian segregation or suppressed recombination which often hamper breeding processes based on interspecific crosses. Three connected (F1, F2 and BC2) populations, derived from a cross between P. davidiana clone P1908 and the peach cultivar Summergrand were used. The intraspecific map of P. davidiana already established using the F1 progeny was complemented, and two interspecific maps, for the F2 and BC2 progenies, were built with a set of markers selected from the Prunus reference map. With the molecular data collected for the F2 map construction, regions with distorted marker segregation were detected on the genome; one third of all loci deviated significantly from the expected Mendelian ratios. However, some of these distorted segregations were probably not due to the interspecific cross. On linkage group 6, a skewed area under gametic selection was most likely influenced by the self-incompatibility gene of P. davidiana. Using anchor loci, a good colinearity between the three maps built and the Prunus reference map was demonstrated. Comparative mapping also revealed that homologous recombination occurred normally between P. davidiana and the Prunus persica genome. This confirmed the closeness of the two species. Higher recombination rates were generally observed between P. davidiana and P. persica than between Prunus amygdalus and P. persica. The consequences for plant breeding strategy are discussed. The three maps of the F1, F2 and BC2 progenies provide useful tools for QTL detection and marker-assisted selection, as well as for assessing the efficiency of the peach breeding scheme applied to introgress P. davidiana genes into peach cultivated varieties.  相似文献
