An electron microscopic study of the mature megagametophyte in Zea mays   总被引:4,自引:0,他引:4  
With light microscopy maize megagametophytes stained with Alcian blue-periodic acid-Schiff (AB-PAS) reveal acid or neutral polysaccharides in various cell walls. Comparative fine structural studies were made of permanganate- or OsO4-fixed material. Organelle distribution is random in the vacuolate and multinucleate antipodal cells; organelles are abundant; starch is scarce. Antipodal cell walls have large openings forming several syncytia. Some walls are papillate. In the central cell (primary endosperm cell) a thin peripheral layer of cytoplasm surrounds the large vacuole; organelle number is moderate; starch is abundant. The central cell wall is also papillate adjacent to the antipodals and around the egg apparatus. In the synergids organelle distribution is non-random; nuclei and numerous organelles occupy the micropylar cytoplasm of each synergid; vacuoles dominate the chalazal cytoplasm of these cells. The filiform apparatus stains with AB-PAS and is composed of both lightly and darkly stained amorphous material. In the egg, organelle distribution is perinuclear with vacuoles proximal to the micropyle; mitochondria are large, abundant and polymorphic; starch is abundant. Nucleolar diameter is five times greater in the central cell and egg than in the antipodal cells and ten times greater than in the synergids. Plasmodesmata occur in all cell walls within the gametophyte, but none appear in the gametophyte wall itself. It is suggested that the antipodals and synergids might be secretory, the latter probably being involved in pollen tube attraction, and that stored metabolites in the central cell and egg cytoplasm support rapid increase in metabolism following fertilization.  相似文献

Summary Localization of polysaccharides in the freeze-substituted, Eponembedded ovaries of Paspalum longifolium prior to pollination was carried out by periodic acid-Schiff's (PAS), periodic acid-thiosemicarbazide-silver proteinate (PA-TSC-AgPr) and periodic acid-thiosemicarbazide-osmium (PA-TSC-OsO4) reactions. The specificities of these three reactions were also studied. These three reactions are all effective for light microscopic demonstration of polysaccharides in the filiform apparatus, starch grains in the cells and PAS substance in the micropylar region. Nonspecific staining of the nucleoli of the egg and polar nuclei was observed in the PAS reaction. The PA-TSC-AgPr reaction is very specific for polysaccharides but its overall reaction takes a much longer period of time than the PAS reaction. The PA-TSC-OsO4 reaction colors the cytoplasm and nuclei of most cells and therefore stains of the cell walls, especially those of the egg cell and synergids, do not stand out clearly. The synergid cytoplasm contains some amorphous polysaccharides and thus it colors even in PAS and PA-TSC-AgPr preparations. In the mature embryo sac, the egg and central cell as well as antipodals are vacuolated but the two synergids have no visible vacuoles under light microscope. Each synergid has a prominent filiform apparatus at the micropylar end, which stains intensely in all three preparations. The walls of the central cell and antipodals adjacent to the nucellar cells have many inward papillae which are also intensely stained in all three preparations. Starch grains are abundant in the ovary wall and usually absent in the nucellus and integuments. They are present in the egg, central cell and antipodals, but not in the two synergids.  相似文献

Abstract

Cyto-embryological study and geographical distribution of CIRSIUM CASABONAE Lam. et DC. — The mature megagametophyte of Cirsium Casabonae Lam. et DC. (Compositae) is eight nucleate of the normal or Polygonum type.

In the nucellus there is only one megaspore mother cell. After meiosis the three micropylar megaspores degenerate. The three antipodal cells start to degenerate as soon as the gametophyte reaches maturity. Microsporogenesis is regular.

The chromosome number of Cirsium Casabonae Lam. et DC. is 2n = 32. This shows that also in Europe, as in America, there is at least one species of Cirsium with basis number × = 16.

The study of the geographical distribution of Cirsium Casabonae Lam. et DC. shows it to be an Atlantic-West Mediterranean endemism.  相似文献

Megasporogenesis and megagametogenesis were examined in Glycine max with light, fluorescence, and electron microscopy. Megasporogenesis results in a linear tetrad of four megaspores. Megagametophyte development is of the Polygonum type, with the functional chalazal megaspore undergoing three successive mitotic divisions to produce an eight-nucleate, seven-celled mature megagametophyte. The central cell becomes packed with starch. At fertilization, the antipodals are degenerate, the polar nuclei have fused, starch is diminished, and the egg occupies most of the micropylar portion of the megagametophyte. Several pollen tubes were occasionally observed at each micropyle, yet only one was involved in fertilization. Pollen tube entry occurs through a slightly reduced, viable synergid cell. Endosperm development precedes embryo growth. These results describing normal development allow important comparison with genetic mutants of soybean that affect female fertility.  相似文献

本文对扁豆(Dolichos lablab)成熟胚囊的超微结构进行了研究,在成熟胚囊中,卵细胞和助细胞仅在珠孔端1/3有细胞壁,靠近合点端,卵细胞一助细胞,卵细胞-中央细胞,助细胞-中央细胞之间没有细胞壁存在,相邻细胞的质膜靠在一起,在卵细胞和中央细胞的质膜间,有些地方存在中等电子密度的物质,卵细胞的细胞质中含有很多的线粒体和质体,内质网和高尔基体较少,助细胞的珠孔端有一复杂的丝状器,靠近珠孔端的细胞质中有很多管状的内质网,表明助细胞可能具有分泌功能,在助细胞的合点端,含有丰富的粗糙内质网,助细胞和卵细胞的质膜之间有很多囊泡状的结构,中央细胞内含有丰富的线粒体,高尔基体和内质网,中央细胞的壁向内形成突起,在周缘细胞质中含有丰富的脂滴。  相似文献

Studies of ultrastructure and ATPase localization of the mature embryo sac in Vicia faba L. show that the egg cell has no cell wall at thechalazal end, it has a chalazally located nucleus and a large micropylar vacuole. There are many nuclear pores in the nuclear membrane. The cytoplasm is restricted around the nucleus. Dictyosome and mitochondria are few. There are some starch grains and lipid grains in the egg cytoplasm. There are no obvious differences between two synergids. No cell wall is seen at the chalazal end either, but there are some vesicles which project to vacuole of the central cell and fuse with its vacuolar membrane. Plasmodesmata connections occur within the synergid wall where it is adjacent to the central cell. The synergid has a micropylarly located nucleus and a chalazal vacuole, the nucleus is irregularly shaped. The synergid cytoplasm is rich in organelles. The filiform aparatus is of relatively heterogeneous structure. The central cell is occupied by a large vacuole and its cytoplasm is confined to a thin layer along the empryo sac wall, but is rich in various organelles, starch grains and lipid bodies. Nucleolar vacuoles are often present two polar nuclei. The nuclear membranes of two polar nuclei have partly fused. ATPase reactive product was located obviously at the endoplasmic reticulum in cytoplasm of the egg cell and central cell. The embryo sac wall consists of different density of osmiophilic layer. There are some wall ingrowths in chalazal region of the embryo sac. The long-shaped and cuneate cells of chalazal region are peculiar. Special tracks of ATPase reactive products are visible at their intercellular space which may be related to transportation of nutrients.  相似文献

 The structure of and the calcium distribution in the ovary-ovule transmitting tracts of Crocus were examined by using conventional and antimonate fixation methods for electron microscopy, and fluorescence microscopy as well. The ovarian locules are bordered with epidermal cells and transmitting cells. The epidermal cells are extensively cutinized, and the cytoplasm is rich in the organelles and vesicles prevalently engaged in thickening the exposed wall. The transmitting cells have uncutinized walls with numerous labyrinthine ingrowths, and the cytoplasm is rich in organelles and vesicles engaged in the secretion of the extracellular matrix (e.m.). The micropylar tissue of the ovule consists of largely vacuolated cells with scarcely structured cytoplasm, the e.m. appears to be of gametophytic origin. Both the ovary and ovule e.m. have a glycosidic and proteinic nature, and are rich in loosely bound Ca2+ ions. In the embryo sac, Ca2+ ions are concentrated in one synergid. The results are discussed in relation to the guided growth of pollen tubes. Summary The ultrastructure of the antipodals ofAconitum vulparia Rchb. was studied in mature embryo sacs. Antipodal cell wall thickness varies in different parts of the cells. The antipodals resemble transfer cells with distinctly marked wall ingrowths which are particularly well developed in the chalazal part and between the antipodals. A few plasmodesmata occur in the cell wall between the antipodals and the central cell. The cytoplasm is rich in ribosomes which occur free or bound to the membranes of the well developed endoplasmic reticulum. Only in the micropylar region of the cells are some larger vacuoles found. The antipodals contain numerous mitochondria, plastids and apparently active dictyosomes. Vesicles with electron dense contents, microbodies, multivesicular bodies as well as lipid droplets and small multiple concentric cisternae are also present in the cytoplasm. The giant endopolyploid nuclei have lobed outlines, especially at the chalazal side of the nuclei.Ultrastructural features, especially the occurrence of numerous free ribosomes and the development of extensive rough endoplasmic reticulum, suggest high metabolic activity in the growing and differentiating antipodals of this species.  相似文献

 In this study, megasporogenesis of the plant model Arabidopsis thaliana was investigated by electron microscopy for the first time. The data described here could constitute a reference for future investigations of Arabidopsis mutants. During the beginning of meiosis the megaspore mother cell shows a polarity created by unequal distribution of organelles in the cytoplasm. Plastids accumulate in the chalazal region and long parallel saccules of endoplasmic reticulum, small vacuoles and some dictyosomes are found in the micropylar region. Plasmodesmata are abundant in the chalazal cell wall. The nucleus is almost centrally localized and contains a prominent excentric nucleolus and numerous typical synaptonemal complexes. After the second division of meiosis the four megaspores are separated by thin cell walls crossed by numerous plasmodesmata and do not show significant cellular organization. The young functional megaspore is characterized by a large nucleus and a large granular nucleolus. The cytoplasm is very electron dense due to the abundance of free ribosomes and contains the following randomly distributed organelles: mitochondria, a few short saccules of endoplasmic reticulum, dictyosomes and undifferentiated plastids. However, there is no apparent polarity, except for the distribution of some small vacuoles which are more abundant in the micropylar region of the cell. 水稻(Oryza sativa L.)胚囊成熟时,卵细胞的合点端无细胞壁,核居细胞中部,细胞器集中在核周围,液泡分散于细胞周边区域。助细胞珠孔端有丝状器,合点端无壁,核位于细胞中部贴壁处,细胞器主要分布在珠孔端,液泡主要分布在合点端。开花前不久,一个助细胞退化。中央细胞为大液泡所占,两个极核靠近卵器而部分融合,细胞器集中在极核周围和靠近卵器处,与珠心相接的胚囊壁上有发达的内突。反足细胞多个形成群体,其增殖主要依靠无丝分裂与壁的自由生长,反足细胞含丰富活跃的细胞器,与珠心相接的壁上有发达的内突。开花后6小时双受精已完成,合子和两个助细胞合点端均形成完整壁。合子中开始形成多聚核糖体、液泡减小。退化助细胞含花粉管释放的物质,其合点端迴抱合子。极核已分裂成数个胚乳游离核,中央细胞中细胞器呈活化状态。反足细胞仍在继续增殖。讨论了卵细胞的极性、助细胞的退化、卵器与中央细胞间界壁的变化、反足细胞的分裂特点等问题。  相似文献

本文报道了竹节参(Panax japonicus C.A.Mey)雌配子体(胚囊)的发育过程。竹节参大孢子母细胞减数分裂产生线形排列的大孢子四分体。胚囊发育属蓼型,由合点端大孢子发育而成。游离核胚囊时期,胚囊珠孔端的细胞器种类和数量都较胚囊合点端多;胚囊合点端相邻的珠被细胞中有含淀粉粒的小质体,与胚囊珠孔端相邻的退化中的非功能大孢子中则有含淀粉粒的大质体和大类脂体。成熟胚囊中,反足细胞较早退化;极核融合成次生核;卵细胞高度液泡化,细胞器数量较少;助细胞则有丰富的细胞器和发达的丝状器。PAS反应表明,受精前的成熟胚囊中积累淀粉粒。次生核受精后,很快分裂产生胚乳游离核,到几十至数百个核时形成胚乳细胞。卵细胞受精后则要经过较长的休眠期。  相似文献

The synergid cells are located in the female gametophyte and are essential for angiosperm reproduction. During the fertilization process, a pollen tube grows into one of the synergid cells, ceases growth, ruptures, and releases its two sperm cells into this cell. The synergid cells produce an attractant that guides the pollen tube to the female gametophyte and likely contain factors that control arrest of pollen tube growth, pollen tube discharge, and gamete fusion. The synergid cells contain an elaborated cell wall at their micropylar poles, the filiform apparatus that likely plays a role in pollen tube guidance and pollen tube reception. Recent genetic, molecular, and physiological studies in Arabidopsis, maize, and Torenia have provided insights into synergid cell development and the control of pollen tube growth by the synergid cell.  相似文献

The organization of isolated embryo sacs and eggs of Plumbago zeylanica was described before and after fertilization using microscopic cytochemistry and scanning electron microscopy. Major developmental events of fertilization, including preferential fertilization and early embryogenesis, are described in isolated embryo sacs. The two sperms, one unassociated with vegetative nucleus (Sua) and the other physically associated with the vegetative nucleus (Svn), fuse with nuclei of egg and central cell, respectively. The zygote divides asymmetrically to form a two-celled embryo, consisting of a massive suspensor occupying most of the micropylar portion of the embryo during early embryogenesis. Plastids are distributed in the perinuclear and micropylar regions of the egg cell and in cytoplasmic strands of the central cell before fertilization. Calcofluor white-positive fibrillar material in the filiform apparatus (presumed β-1,4 linked glucans) was investigated using scanning electron microscopy. The egg of P. zeylanica can easily be divided into three cytologically distinct regions: 1) perinuclear cytoplasm, 2) lateral cytoplasm, and 3) micropylar cytoplasm. Cytological differences are evident in the organization of the cell walls, general degree of vacuolization, and the distribution of heritable organelles, storage bodies, and microtubules. The present study supports the concept that the egg of P. zeylanica plays combined synergid and gamete functions.  相似文献

Embryological development of Lachnanthes caroliniana was studied utilizing standard anatomical techniques and SEM. Lachnanthes has a monocotyledonous anther wall development (endothecial cells with spiral secondary wall thickenings), successive microsporogenesis, and amoeboid (periplasmodial) tapetal development. Mature pollen grains are 2-nucleate with a proximal, fusiform generative cell. Ovules are initiated as 5–7 cylindrical primordia from a common placental base. Basal ovular swellings collectively contribute to the enlarged, peltate placenta. Mature ovules are pleurotropous, anatropous, bitegmic, and crassinucellate; the nucellus consists of a chalazal hypostase, radially elongate lateral cells, and a prominent micropylar nucellar cap. Megasporogenesis is successive, forming a linear tetrad of megaspores. Megagametogenesis is monosporic; the female gametophyte is of the Polygonum-type with relatively large, pyriform antipodals. Endosperm formation is helobial, resulting in the establishment of a ring of four thick-walled basal endosperm cells (the chalazal chamber) and numerous free nuclei (in the micropylar chamber). The mature cellular endosperm is filled with starch grains and has a chalazal cavity and a thick-walled peripheral layer. The discoid, peltately attached seeds have marginal wings derived by anticlinal divisions and buckling of the outer integument alone. Inner and middle cuticular layers are present in the seed coat. Lachnanthes is similar to all other investigated members of the Haemodoraceae in major embryological features. The significance of embryological evidence with regard to interfamilial classification is discussed. Future studies of ovule and seed development may prove valuable in phylogenetic studies in assessing the homology of placental, ovule, and seed morphology and anatomy.  相似文献

Papaver nudicaule placentae with attached ovules were dissected out of unpollinated gynoecia 1–3 days after anthesis, dusted with pollen, and cultured on modified Nitsch's growth medium at 23 C. Ovules were removed from expiants at 15, 24, 31, and 48 hr postpollination, fixed in GA-OsO4, embedded in Spurr's resin and sectioned (1.0 μm) for light microscopy. Placentae, 15 hr after pollination, were fixed and processed for scanning electron microscopy. Pollen germinates within 1 hr. Although most pollen tube growth appears random, there is directional growth toward the micropyle. The crassinucellate ovule contains an embryo sac consisting of three antipodals, two polar nuclei, and an egg apparatus composed of two synergids and a polarized egg having a large chalazal vacuole and micropylar nucleus. Pollen tube access into the megagametophyte is through a degenerate synergid, with fertilization occurring between 24 and 31 hr after pollination. Zygote establishment is accompanied by polarity reversal in which the nucleus assumes the chalazal position subtended by a large micropylar vacuole. Fertilized ovules normally develop into germinable seeds.  相似文献

Ultrastructure of the embryo sac lacking antipodals in prefertilization stages in Arabidopsis thaliana has been examined 2 hr before and 5 hr after manual cross pollination. The cytoplasm of both synergids before fertilization is rich in ribosomes, mitochondria, and rough endoplasmic reticulum, and also contains several microbodies and spherosomes. The filiform apparatus includes electron-dense material and a fibrous part. Many cortical microtubules appear in the filiform apparatus area. One of the two synergids degenerates before fertilization. The synergids, the egg cell, and central cell have a rich cytoskeleton of microtubules; only the synergids appear to contain microfilaments. At the chalazal end, the antipodals are initially present but degenerate by the time of pollination in most embryo sacs in the starchless line studied. The embryo sac is completely surrounded by a wall containing an electron-dense layer, separating it from the nucellus, including the chalazal end. When the antipodals have degenerated, the electron-dense layer disappears at the chalazal end only, and the wall between the central cell and the nucellus is homogeneous. Between the central cell and nucellar cells no plasmodesmata are found. The membranes of both antipodal cells at the chalazal end of the embryo sac appear sinuous, like those of transfer cells. The central cell has plastids preferentially distributed around the nucleus, but the other organelles are randomly distributed. The central cell in the embryo sac and the adjacent chalazal nucellar cells show a transfer-cell function in the embryo sac after the antipodals degenerate.  相似文献

The ultrastructure of synergids of watermelon (Citrullus Lanatus L.) was investigated using transmission electron microscopy at following stages of embryo sacs: 1. Unpollination, on the first flowering day. 2. Unpollination, on 2nd day after anthesis (DAA). 3. Fertilization, on DAA 2. The synergids with distinct filiform apparatus at the micropylar end have abundant organelle, such as mitochondria, endoplasmic reticulum, and plastids in cytoplasm, which indicate that they are active on the first flowering day. No wall is present at the chalazal part of synergid, and there are some flocculent materials and vesicles in the spaces of cytoplasma membranes among synergid, egg cell and central cell in embryo sacs at the first and the second stages. On DAA 2, in unpollinated embryo sacs, the central large vacuole of synergid is divided into several smaller ones and the starch grains decrease in cytoplasm. There is no newly synthesized wall at the chalazal end of persistent synergid in fertilized embryo sacs. The contents of degenerated synergid, in the form of electron dense granules, are located in the wide space among central cell, zygote and persistent synergid, and some of them migrate into central cell through cytoplasma membrane. Therefore, it is deduced that the contents of synergid might serve as a nutrient supplement to the development of endosperm, but not embryo.  相似文献

A study of the egg apparatus of Quercus gambelii was made at both the light and the electron microscope levels. This investigation was concerned primarily with the changes that occur in these cells before and after the process of fertilization and what role, if any, is played by the synergids in this phenomenon. The synergids before fertilization are, on the basis of ultrastructure, healthy, intact, functional cells. They have numerous mitochondria, dictyosomes, endoplasmic reticulum, ribosomes, and a typical nucleus. A prominent filiform apparatus is present, but the cell wall only extends a short distance around the micropylar end of the cells. Just before fertilization, one of the synergids degenerates. This is the synergid that receives the pollen tube and its discharge, including both male gametes. Dictyosomes increase in number and activity in the other synergid (persistent synergid) after fertilization. Eventually a complete cell wall forms around both of the synergids. No plasmodesmata are present in these walls. The egg has numerous mitochondria, dictyosomes, endoplasmic reticulum, and ribosomes, both free in the cytoplasm and attached to the endoplasmic reticulum. Lipid bodies are characteristic of this cell. A cell wall is present only around the micropylar end of the egg. After fertilization, little change occurs in the zygote. The number and activity of the dictyosomes increase, apparently in correlation with cell wall formation. The number of lipid bodies increases. The zygote is approximately the same size as the egg. Plastids are scarce, and starch grains are typically absent from all cells of the egg apparatus. It is suggested that the synergids function in the secretion of chemotropic substances that guide the growth of the pollen tube. Comparisons are made between the egg apparatus of Quercus gambelii and that of the other plants studied thus far.  相似文献

Serially sectioned embryo sacs of Nicotiana tabacum were examined during fertilization events using transmission electron microscopy. After pollen tube discharge, the outer membrane of the sperm pair is removed, the two sperm cells are deposited in the degenerate synergid and the sperm cells migrate to the chalazal edge of the synergid where gametic fusion occurs. During fertilization, the male cytoplasm, including heritable organelles, is transmitted into the female reproductive cells as shown by: (1) the cytoplasmic confluence of one sperm and the central cell during cellular fusion, (2) the occurrence of sperm mitochondria (distinguished by ultrastructural differences) in the zygote cytoplasm and adjacent to the sperm nucleus, (3) the presence of darkly stained aggregates which are found exclusively in mature sperm cells within the cytoplasm of both female cells soon after cell fusion, and (4) the absence of any large enucleated cytoplasmic bodies containing recognizable organelles outside the zygote or endosperm cells. The infrequent occurrence of plastids in the sperm and the transmission of sperm cytoplasm into the egg during double fertilization provide the cytological basis for occasional biparental plastid inheritance as reported previously in tobacco. Although sperm mitochondria are transmitted into the egg/zygote, their inheritance has not been detected genetically. In one abnormal embryo sac, a pair of sperm cells was released into the cytoplasm of the presumptive zygote. Although pollen tube discharge usually removes the inner pollen-tube plasma membrane containing the two sperm cells, this did not occur in this case. When sperm cells are deposited in a degenerating synergid or outside of a cell, this outer membrane is removed, as it apparently is for fertilization.  相似文献
