Eleusine (Poaceae) includes six diploid and three polyploid species and has three basic chromosome numbers, x=8, 9 and 10. The species are annual as well as perennial and all are wild except E. coracana, which is cultivated for grain and fodder in Africa and the Indian subcontinent. Eleusine coracana and E. africana have the same genome and chromosome number (2n=36). Eleusine indica and E. floccifolia are identified as two genome donors to these polyploid species. Eleusine kigeziensis is the third polyploid species of the genus with 2n=38. Its genome may have come from E. jaegeri and from one of the species with x=9, most probably from E. indica. Eleusine indica, E. tristachya, E. floccifolia and E. intermedia with x=9 and two polyploid species, E. coracana and E. africana, are closely related and there is free genetic flow between them. Eleusine multiflora with x=8 is significantly different in morphology and at genomic level from other species. Eleusine jaegeri with x=10 is morphologically similar to E. indica, however, more information is needed to ascertain its position in the genus. Eleusine coracana, which is commonly called finger millet, is a potential and nutritious crop for the increasing population of the world, particularly in arid and semi-arid regions. It can also serve as a gene pool for various important characters and disease resistant genes. Phylogenetic relationships in the genus Eleusine (Poaceae: Chloridoideae) were investigated using nuclear ITS and plastid trnT-trnF sequences. Separate and combined data sets were analyzed using parsimony, distance, and likelihood based methods, including Bayesian. Data congruence was examined using character and topological measures. Significant data heterogeneity was detected, but there was little conflict in the topological substructure measures for triplets and quartets, and resolution and clade support increased in the combined analysis. Data incongruence may be a result of noise and insufficient information in the slower evolving trnT-trnF. Monophyly of Eleusine is strongly supported in all analyses, but basal relationships in the genus remain uncertain. There is good support for a CAIK clade (E. coracana subsp. coracana and africana, E. indica, and E. kigeziensis), with E. tristachya as its sister group. Two putative ITS homeologues (A and B loci) were identified in the allotetraploid E. coracana; the 'B' locus sequence type was not found in the remaining species. Eleusine coracana and its putative 'A' genome donor, the diploid E. indica, are confirmed close allies, but sequence data contradicts the hypothesis that E. floccifolia is its second genome donor. We investigated spikelet development in four distantly related species of the grass tribe Andropogoneae to determine whether spikelet development and the formation of unisexual florets are uniform throughout the tribe. We studied development in Bothriochloa bladhii, Coelorachis aurita, Heteropogon contortus, and Hyparrhenia hirta, and compared these with Panicum, a member of the sister tribe Paniceae. Many aspects of spikelet development in the species we have studied correlate with what is already known for Tripsacum and maize (both Andropogoneae), despite variation in how unisexual florets are distributed on the plant. The formation of unisexual spikelets is also uniform. All florets initiate both pistil and stamen primordia. In florets destined to be male, cell death occurs in the subepidermal layers of the gynoecium after the formation of a gynoecial ridge. In florets destined to be female, there is no apparent cell death in the stamens, but growth ceases after anther formation. The similarity in spikelet development and the formation of unisexual florets point to a common genetic mechanism for sex determination throughout the Andropogoneae and possibly the entire Panicoideae. Use of a cell death pathway to cause gynoecial abortion may be the basis of one morphological character that defines the subfamily. 相似文献

Intermatings between sexual diploids (2n=36) and facultative apomictic tetraploids (2n=4n=72) of Tripsacum dactyloides var. dactyloides, (L.), L., have been generated. Subsequent exchange and addition of genetic material between the two cytotypes, via a backcrossing program, provides one method for the manipulation of genetic elements involved in apomictic reproduction (diplospory) of this species.Identification and development of fertile triploids, and their innate tendency to exhibit a high frequency of unreduced egg cells which remain receptive to fertilization by a sperm nucleus, allows for the unique development of BIII(2n=3x+n) tetraploids by genome accumulation. The resultant BIIIprogeny provide new breeding stock for investigating the inheritance and generation of such materials also allows for the occurrence of genetic recombination between sexual diploids and apomictic tetraploids. Potential use of fertile triploids, their tendency for genome accumulation and their use in exploiting apomictic tetraploid germplasm resources are also discussed.All programs and services of the U.S. Department of Agriculture are offered on a nondiscriminatory basis without regard to race, color, national origin, religion, sex, age, marital status, or handicap. 相似文献

In this study, we examined whether a satisfactory infrageneric grouping in Eremopyrum (Ledeb.) Jaub. & Spach can be achieved with the help of cluster analysis based on external vegetative and floral morphological characters of specimens either gathered from the field or borrowed from various herbaria. Twenty morphological characters were chosen and scored for 36 populations representing the genus. The data were subjected to numerical taxonomic analyses, and a phenogram was obtained by applying Gower general similarity index. The results of this study suggest that there are basically five species in Turkey, namely E. triticeum (Gaertner) Nevski, E. orientale (L.) Jaub. & Spach, E. distans (K. Koch) Nevski, E. bonaepartis (Sprengel) Nevski, and E. confusum Melderis. For the first time a new variety, E. bonaepartis (Sprengel) Nevski. var. aristatum Cabi & Do?an, is proposed along with two new combinations: E. bonaepartis (Sprengel) Nevski. var. sinaicum (Steudel) Cabi & Do?an and E. confusum Melderis var. sublanuginosum (Drob.) Cabi & Dogan. An account of Eremopyrum in Turkey is given, including a key to the species and varieties, and a general indication of their distribution. 相似文献

Apomixis is a particular mode of reproduction that allows progeny formation without meiosis and fertilization. Eulaliopsis binata, a tetraploid apomictic species, is widely used for making paper, rope and mats. There is great potential for fixation of heterosis in E. binata due to autonomous endosperm formation in this species. Although most of its embryo sac originates from nucellus cells, termed apospory, we observed sexual reproduction initiation in 86.8 to 96.8% of the ovules, evidenced by callose deposition on the walls of cells undergoing megasporogenesis. However, only 2-3% mature polygonum-type sexual embryo sacs were confirmed by embryological investigation. Callose was not detected on aposporous initial cell walls. The aposporous initial cells differentiated during pre- and post-meiosis of the megaspore mother cell, while the sexual embryo sac degenerated at the megaspore stage. DNA content ratio of embryo and endosperm in some individuals was 2C:3C, based on flow cytometry screening of seed, similar to that of normal sexual seed. These results confirm that apomictic E. binata has conserved sexual reproduction to a certain degree, which may contribute to maintaining genetic diversity. The finding of sexual reproduction in apomictic E. binata could be useful for research on genetic mechanism of apomixis in E. binata. 相似文献

Callus was successfully initiated on root, mesocotyl and leaf base segments of 3- to 4-day-old seedlings of ragi (Eleusine coracana Gaertn.). 2,4-D along with casein hydrolysate for Murashige and Skoog's basal medium was found to be most effective for callus initiation and maintenance. Mesocotyl and leaf base tissue derived calli gave shoot buds in medium in which the 2,4-D concentration was lowered. 相似文献

Some have postulated that highland Mexican maize was derived from an ancient high-altitude teosinte and that later introgression between the two taxa occurred. We used scanning electron microscopy to examine the inflorescence development in both the tassel and ear of a high-altitude Toluca teosinte. One of the most interesting observations was the presence of atypical multiranked orthostiches in the central spike of some male Toluca teosinte inflorescences. Most tassels exhibited a central spike with a pure, four-ranked, tetrastichous phyllotaxy or an intermediate (distichous/tetrastichous) phyllotaxy. A few A(1) tassels had a more typical distichous (two-ranked) central spike. Most ears showed the two-rank condition expected for teosintes. However, three ears displayed an intermediate (distichous/tristichous or distichous/ tetrastichous) phyllotaxy and one ear was tetrastichous. Our analysis of spikelet and floret development in all Toluca inflorescences revealed a pattern similar to that in landrace and U.S. maize, as well as to their close relatives, the teosintes. We suggest that this investigation may reveal inflorescence development in a natural maize-teosinte hybrid. This study further supports our hypothesis that both maleness and femaleness in the Zea inflorescences are derived from a common developmental pathway and underpins a proposal that andropogonoid grasses share a common pattern of inflorescence development. 相似文献

Summary High numbers of embryos are difficult to obtain in liquid cultures of sweet potato (Ipomoea batatas (L.) Lam.) because discrete cell aggregates, produced through calli fragmentation, do not support embryo growth. In an effort to demonstrate that embryo development is possible from discrete cell aggregates, we compared embryo formation from cell aggregates 250–355 μm in diameter cultured either in suspension in liquid medium, on agar solidified medium, or immobilized on alginate beads floated in liquid medium. Embryos were initiated but remained arrested in their globular stage on cell aggregates cultured in suspension. Embryos developed to the torpedo stage from cell aggregates cultured on solidified medium and from cell aggregates anchored on alginate beads. Thus, embryos continued to develop beyond the globular stage when a structural polarity, which led probably to the establishment of a physiological polarity, was created. The production of sweet potato embryos in liquid culture can be improved by using alginate beads or culture conditions and protocols leading to the release during calli fragmentation of polarized individual cell aggregates.
