Endophytic actinobacteria isolated from Artemisia annua were characterized and evaluated for their bioactivities. A total of 228 isolates representing at least 19 different genera
of actinobacteria were obtained and several of them seemed to be novel taxa. An evaluation of antimicrobial activity showed
that more isolates possessed activity towards plant pathogens than activity against other pathogenic bacteria or yeasts. High
frequencies of PCR amplification were obtained for type I polyketide synthases (PKS-I, 21.1%), type II polyketide synthases
(PKS-II, 45.2%) and nonribosomal peptide synthetases (NRPS, 32.5%). The results of herbicidal activity screening indicated
that 19 out of 117 samples of fermentation broths completely inhibited the germination of Echinochloa crusgalli. This study indicated that endophytic actinobacteria associated with A. annua are abundant and have potentially beneficial and diverse bioactivities which should be pursued for their biotechnical promise. 相似文献

A procedure for isolation of intact forespores from sporulating Bacillus megaterium cells was developed. The cells were digested with lysozyme and made to release free forespores from the protoplasts by disruption of the cytoplasmic membrane with sonication in phosphate buffer containing 10% glycerol. The suitability of the procedure was confirmed by recovery of dipicolinic acid in the isolated forespores and an electron microscopic observation. The fine structure of the forespores prepared at 6 hr (t6) after initiation of sporulation was similar to that of mature spores, except that the cortex layer and primordial cell wall were thinner and the core was larger. The density, determined by density gradient centrifugation, of the forespores isolated at t6, t10, t12, and mature spores was estimated to be 1.2783, 1.2875, 1.2861, and 1.2858, respectively. The isolated forespores at t6 and t8 were extremely heat labile (D80 of 9.5 and 21.5 min, respectively) relative to mature spores (D80 of 277.8 min). These forespores were also less resistant to organic solvents. Germination of the forespores as well as mature spores was induced by KNO3, D-glucose, and L-leucine. Forespores at t6 were more sensitive to KNO3-induced germination than those at t10, t12, and mature spores when measured by reduction in the optical density of cell suspension. 相似文献

Three new pentacyclic triterpenoid saponins (1–3), together with medicagenic acid (4) were isolated and purified from 70% EtOH extract of Medicago sativa L. by different column chromatographic and semi-preparative HPLC. Their structures were established by direct interpretation of their spectral data, mainly HR-ESI-MS, 1D-NMR, 2D-NMR, and chemical methods, as well as comparison with literature data. Additionally, all isolates were evaluated for their neuroprotective activities against H2O2-induced damage in human neuroblastoma SHSY5Y cells. As a results, compounds 1 and 2 (67.14% and 73.05%) exhibited potent neuroprotective activities. These findings provide new insights into developing better treatment of neurodegenerative diseases for M. sativa in the future. 相似文献

A filamentous division mutant, PV302, of Bacillus megaterium QM B1551 was isolated while screening for sporulation-defective mutants after nitrosoguanidine mutagenesis. Both phase-contrast and electron microscopy revealed that the mutant produced small spherical cells as well as filaments. It also accumulated large amounts of poly-beta-hydroxybutyrate. Poly-beta-hydroxybutyrate accounted for 16% of the dry weight of the mutant strain even after 28 h growth. In comparison to the parental strain, the division mutant also showed both an inability to sporulate and a reduced growth rate. All these phenotypes transduced together. Revertants gained the ability to sporulate, divide, and grow normally. Transductional mapping of the mutation, designated div-1, established a new linkage group for B. megaterium consisting of div-1 and the pyrimidine biosynthesis genes pyrD BCF. The spherical cells were separated from filaments by sucrose gradients and were tested for nucleic acid content and viability. The purified spherical cell fraction contained one-fifth the amount of DNA per mg protein as compared with the filamentous cell fraction and was shown to contain both non-viable minicells and some cells capable of growing after a lag of about 4 h. This suggests that the mutation not only causes defects in septum placement and sporulation, but may possibly affect DNA partitioning. 相似文献

Germination bioassay was carried out to test the biological activity of Achillea santolina L. (ASAE), Artemisia monosperma Del. (AMAE), Pituranthus tortuosus L. (PTAE) and Thymus capitatus L. (TCAE) aqueous extracts (collected from Taif region, KSA) on germination percentage (GP), plumule (PL) and radicle (RL) lengths (mm) besides seedling dry weight (SDW) (mg/seedlings) of Medicago polymorpha L. The inhibitory effect of P. tortuosus was insignificant compared to the other three donor species which attained the strongest allelopathic potential in the following order: A. santolina > A. monosperma > T. capitatus. Growth experiment using crude powder of the four donor species was conducted to examine their effects on leaf area index (LAI), photosynthetic pigments, total available carbohydrates (TAC) and total protein (TP). It is worth mentioning that each of the four donor species crude powders mixed with clay loam soil appeared to have a great inhibitory allelopathic effect on LAI, total photosynthetic pigment and chlorophyll a (Chl a) while carotenoids exhibited a slight increase with the application of the four donor species crude powders. TAC and TP were significantly decreased with increasing the crude powder concentrations while a slight decrease was recorded for carbon/nitrogen (C/N) ratio. There is possibility of using these allelochemicals directly or as structural leads for the discovery and development of environmentally friendly herbicides to control weeds. The study recommended that these species must be phytochemically examined in future for their allelochemicals in order to provide information on the possibilities of using one or more of these species as bioherbicides. 相似文献

Sulfate-dependent degradation of glycolate was studied with a new sulfate-reducing bacterium, strain PerGlyS, enriched and
isolated from marine anoxic sediment. Cells were gram-negative, motile rods with a DNA G+C content of 56.2±0.2 mol%. Cytochromes
of theb- andc-type and menaquinone-5 were detected. A sulfite reductase of the desulforubidin-type was identified by characteristic absorption
maxima at 279, 396, 545, and 580 nm. The purified desulforubidin is a heteropolymer consisting of three subunits with molecular
masses of 42.5 (α), 38.5 (β), and 13 kDa (γ). Strain PerGlyS oxidized glycolate completely to CO2. Lactate, malate, and fumarate were oxidized incompletely, yielding more sulfide and less acetate than expected for typical
incomplete oxidation of these substrates. Part of the acetate residues formed was oxidized through the CO-dehydrogenase pathway.
The biochemistry of glycolate degradation was investigated in cell-free extracts. A membrane-bound glycolate dehydrogenase,
but no glyoxylate-metabolizing enzyme activity was detected; the further degradation pathway is unclear.

Citrate synthase was purified to homogeneity from a Gram-positive bacterium (Bacillus megaterium) for the first time. The Mr of the native enzyme was determined to be 84 000 (S.E.M. +/- 5000). Sodium dodecyl sulphate/polyacrylamide-gel electrophoresis and gel filtration in guanidinium chloride revealed a single protein species of Mr 40 300 (S.E.M. +/- 4400), indicating a dimeric enzyme. This dimeric structure was confirmed by cross-linking the native enzyme with dimethyl suberimidate and with glutaraldehyde, followed by electrophoretic analysis. The enzyme follows Michaelis-Menten kinetics with respect to both substrates, acetyl-CoA and oxaloacetate, and is sensitive to non-specific inhibition by a range of adenine nucleotides. In both molecular and catalytic properties the citrate synthase closely resembles the enzyme from eukaryotic sources and contrasts markedly with the larger, hexameric, enzyme from Gram-negative bacteria. 相似文献

A novel bacterium, strain BMP-1(T), was isolated from a continuous wastewater treatment culture system operating with a bacterial consortium. Cells of the isolate were Gram-variable, aerobic, moderately halotolerant, motile and endospore-forming rods. Strain BMP-1(T) grew chemolithoautotrophically by oxidation of thiosulfate to sulfate with a growth yield of 1.07 g protein mol(-1) of thiosulfate consumed. DNA G+C content was 43.8 mol%. Its cell wall had peptidoglycan based on m-diaminopimelic acid, and the major component of fatty acid was C(15 : 0). The 16S rRNA gene analysis showed that strain belongs to the genus Bacillus, sharing a 99.5% of sequence similarity with Bacillus jeotgali CCM 7133(T). DNA-DNA hybridization between the isolate of this study and this strain was 44%. Thus, the inclusion of strain BMP-1(T) in the genus Bacillus is suggested as a novel species and the name Bacillus thioparus sp. nov. (Type strain BMP-1(T)=BM-B-436(T)=CECT 7196(T)) is proposed. A proteolytic activity present in spores of Bacillus megaterium has previously been implicated in the initiation of hydrolysis of the A, B, and C proteins which are degraded during spore germination. Four mutants of B. megaterium containing 20 to 30% of the normal level of spore proteolytic activity have been isolated. Partial purification of the protease from wild-type spores by a reviewed procedure resulted in the resolution of spore protease activity on the A, B, and C proteins into two peaks--a major one (protease II) and a minor one (protease I). The protease mutants tested lacked active protease II. All of the mutants exhibited a decreased rate of degradation of the A, B, and C proteins during spore germination at 30 degrees C, but degradation of the proteins did occur. Degradation of the A, B, and C proteins during germination of the mutant spores was decreased neither by blockade of ATP production nor by germination at 44 degrees C. Initiation of spore germination was normal in all four mutants, and all four mutants went through outgrowth, grew, and sporulated normally in rich medium. Similarly, outgrowth of spores of two of the four mutants was normal in minimal medium at 30 degrees C. In the two mutants studied, the kinetics of loss of spore heat resistance and spore UV light resistance during germination were identical to those of wild-type spores. This indicates that the A, B, and C proteins alone are not sufficient to account for the heat or UV light resistance of the dormant spore. 相似文献

为进行阿特拉津(AT)污染的生物修复,从AT降解混合菌群中,经长期的交替液体摇瓶培养和平板划线分离,筛选到一株能完全降解AT的菌株SA1。经生理生化特征及16S rDNA序列分析,将该菌鉴定为假单胞菌属(Pseudomonas sp.)。与已报道的AT降解菌Pseudomonas sp.ADP不同,SA1能以AT为唯一碳源、氮源和能源生长,培养基中添加铵盐不抑制SA1的降解功能,而添加葡萄糖时,累积的氰尿酸会被快速降解。SA1生长的最适温度为37℃,最适pH值为7.0。SA1的静息细胞在10℃~40℃或pH值4~11时均能高效降解AT,比ADP降解具有更广的pH和温度范围,表明SA1降解菌株具有广阔的应用前景。SA1中AT降解基因为保守的atzABCD,并含有IS1071的tnpA基因片段,传代过程中降解基因会以一定频率丢失。 相似文献

A novel, strictly anaerobic, gram-negative, non-spore-forming, fusiform, rod-shaped bacterium having high dehydrodivanillin (DDV)-degrading activity was isolated from cow ruminal fluid. This strain degraded a range of six main lignin-related compounds such as DDV, ferulic acid, dehydrodiisoeugenol, guaiacoxyacetic acid, vanillin, and veratrylglycerol-beta-guaiacyl ether to the extent of 14 to 83% within 2 days under strictly anaerobic conditions. As DDV degradation intermediates, three aromatic compounds (dehydrodivanillic acid, vanillic acid, and 5-carboxyvanillic acid) and two alicyclic compounds (cyclohexanecarboxylic acid and cyclohexanol) were detected by thin-layer, high-performance liquid, and gas chromatography and mass spectrometry. The addition of 1% glucose and peptone in a synthetic medium stimulated growth of the strain but slowed down DDV degradation. The presence of 0.1% yeast extract increased both cell growth and DDV degradation. The growth yield in defined medium was 151.5 g (dry weight) of cells per mol of DDV utilized. Characterization of the strain indicated that it was distinct from known Fusobacterium and Clostridium species. The bacterium was easily induced to form protoplasts after treatment with either penicillin or lysozyme. The frequencies of protoplast formation and regeneration in the strain were 94 and 18%, respectively. 相似文献

mineral phosphate, and produced indoleacetic acid and siderophore. The heavy metal detoxification was studied in growing LRE07
cells. The strain bound over 65% of cadmium and 35% of zinc in its growing cells from single metal solutions 72 h after inoculation.
Besides the high removal efficiencies in single-ion system, an analogous removal phenomenon was also observed in multi-ions

N-Deoxyschizokinen, a novel siderophore, was isolated from stationary phase cultures of Bacillus megaterium ATCC 19213 and identified as 4-[(3(acetylhydroxyamino)propyl)amino]-2-[2-[(3-(acetylamino)propyl)amino]-2-oxoethyl]-2-hydroxy-4-oxo-butanoic acid. The siderophore was purified by HPLC and its structure determined using 1H and 13C NMR, 1H-1H COSY and electrospray mass spectroscopy. The monohydroxamate siderophore has the same carbon skeleton as schizokinen but the hydroxyl group on one hydroxamate is replaced by a hydrogen. Keratinolytic activity of Bacillus megaterium F7-1, a feather-degrading mesophilic bacterium 总被引:1,自引:0,他引:1
The aim of this study was to investigate environmental conditions affecting chicken feather degradation and keratinolytic enzyme production by Bacillus megaterium F7-1, a feather-degrading mesophilic bacterium. B. megaterium F7-1 degraded whole chicken feather completely within 7 days. The bacterium grew with an optimum at pH 7.0–11.0 and 25–40 °C, where maximum keratinolytic activity was also observed. The production of keratinolytic enzyme by B. megaterium F7-1 was inducible with feather. Keratinolytic enzyme production by B. megaterium F7-1 at 0.6% (w/v) skim milk was 468 U/ml, which was about 9.4-fold higher than that without skim milk. The amount of keratinolytic enzyme production depended on feather concentrations. The degradation rate of autoclaved chicken feathers by cell-free culture supernatant was 26% after 24 h of incubation, but the degradation of untreated chicken feathers was unsuccessful. B. megaterium F7-1 effectively degraded feather meal, duck feather and human nail, whereas human hair and sheep wool showed relatively low degradation rates. B. megaterium F7-1 presented high keratinolytic activity and was very effective in feather degradation, providing potential use for biotechnological processes of keratin hydrolysis. 相似文献
