Lipopolysaccharide (LPS) treatment of rats suppresses CYP 4F4 and 4F5 expression by 50 and 40%, respectively, in a direct fashion occurring in the liver. This contention is borne out by essentially parallel dose-dependent changes observed upon treatment of rat hepatocyte cultures with LPS. An alternate avenue of triggering the inflammatory cascade is traumatic brain injury by controlled cortical impact. Such injury brings about a dramatic change in the expression of CYP 4F4 and 4F5 mRNA which reaches its greatest effect 24 h after impact compared with sham-operated but uninjured controls. At time points after 24 h the expression of both isoforms increases dramatically reaching highest levels at 2 weeks post-injury. These changes in mRNA expression are mirrored by changes in protein expression. The results are consistent with the notion that immediately after injury concentrations of leukotriene and prostaglandin mediators are elevated by decreased CYP 4F concentrations. As time after injury increases those conditions reverse. Increased CYP 4F expression leads to diminished concentrations of leukotriene and prostaglandin mediators and then to recovery and repair.  相似文献

This study examined mRNA levels and cell surface expression of IL-4 receptor (IL-4R) in murine T and B cells after incubation with IL-4. Northern blot analysis of mRNA levels of T cells isolated from mesenteric lymph nodes and spleen revealed that IL-4 induced a transient augmentation of IL-4R mRNA in a dose-dependent manner. Maximal levels of mRNA were detected as early as 5 h after initiation of culture. These data were complemented by studies examining the cell surface expression of IL-4R using an anti-IL-4R mAb. Resting T and B lymphocytes express IL-4R (T greater than B) and incubation of these cells with exogenous IL-4 increased IL-4R expression to a maximum after 24 h. This effect was abolished after addition of anti-IL-4 antibody. Continuous incubation of T cells in the presence of high concentrations of IL-4 resulted in a down-regulation of IL-4R expression. Addition of the protein synthesis inhibitor cycloheximide blocked the induced increases in IL-4R expression, indicating the requirement for de novo protein synthesis. Both the levels of mRNA and cell surface expression of IL-4R were not affected by addition of exogenous IL-2, and IL-4 regulation of IL-4R expression was not influenced by the immunosuppressive drug cyclosporin A. These data demonstrate that in T and B cells, IL-4 induces a transient up-regulation of IL-4 mRNA levels that is subsequently reflected in increased numbers of IL-4R displayed on the cell surface. This regulation of IL-4R expression by IL-4 provides an important mechanism for amplification of IL-4-dependent activation pathways.  相似文献

Dihydroavenanthramide D (DHAvD) is a synthetic analog to naturally occurring avenanthramide, which is the active component of oat. Previous study demonstrates that DHAvD strongly inhibits activation of nuclear factor-kappa B (NF-κB), which is a major component in cancer cell invasion. The present study investigated whether DHAvD can modulate MMP-9 expression and cell invasion in MCF-7 human breast cancer cells. MMP-9 expression and cell invasion in response to 12-O-tetradecanoylphorbol-13-acetate (TPA) was increased, whereas these inductions were muted by DHAvD. DHAvD also suppressed activation of mitogen-activated protein kinase (MAPK), and MAPK-mediated nuclear factor-kappa B (NF-κB) and activator protein-1 (AP-1) activations in TPA-treated MCF-7 cells. The results indicate that DHAvD-mediated inhibition of TPA-induced MMP-9 expression and cell invasion involves the suppression of the MAPK/NF-κB and MAPK/AP-1 pathways in MCF-7 cells. DHAvD may have potential value in breast cancer metastasis.  相似文献

To investigate the mechanisms of eosinophil recruitment in allergic airway inflammation, we examined the effects of interleukin (IL)-4, a Th2-type cytokine, on eotaxin and monocyte chemoattractant protein-4 (MCP-4) expression in human peripheral blood mononuclear cells (PBMCs; n = 10), in human lower airway mononuclear cells (n = 5), in the human lung epithelial cell lines A549 and BEAS-2B, and in human cultured airway epithelial cells. IL-4 inhibited eotaxin and MCP-4 mRNA expression induced by IL-1 beta and tumor necrosis factor-alpha in PBMCs but did not significantly inhibit expression in epithelial cells. Eotaxin and MCP-4 mRNA expression was not significantly induced by proinflammatory cytokines in lower airway mononuclear cells. IL-1 beta-induced eotaxin and MCP-4 protein production was also inhibited by IL-4 in PBMCs, whereas IL-4 enhanced eotaxin protein production in A549 cells. In contrast, dexamethasone inhibited eotaxin and MCP-4 expression in both PBMCs and epithelial cells. The divergent effects of IL-4 on eotaxin and MCP-4 expression between PBMCs and epithelial cells may create chemokine concentration gradients between the subepithelial layer and the capillary spaces that may promote the recruitment of eosinophils to the airway in Th2-type responses.  相似文献

目的探讨DLL4和Notch4在膀胱尿路上皮癌中的表达及其临床意义。方法收集福建医科大学附属第一医院2008-2010年有完整临床病理资料的膀胱尿路上皮癌存档蜡块85例和27例癌旁组织,采用组织芯片免疫组织化学法检测膀胱尿路上皮癌和癌旁组织DLL4和Notch4的表达水平。对DLL4和Notch4的表达进行半定量分析,并用SPSSl3.0软件对各组免疫组织化学反应阳性率采用x。检验或Fisher精确概率法分析。结果(1)DLL4和Notch4在膀胱尿路上皮癌中呈高表达,癌旁组织中呈低表达,差异有显著性(P〈O.05)。(2)Notch4和DLL4在膀胱尿路上皮癌中的表达与患者性别、年龄、临床分期、病理分级及初复发均无显著相关性(P〉0.05)。结论DLL4和Notch4在膀胱尿路上皮癌中的高表达与膀胱尿路上皮癌血管生成密切相关,阻断DLL4-Noteh4信号通路有望抑制膀胱尿路上皮癌的发生发展。  相似文献

The chemokine receptor CXCR4 is involved in the growth and metastasis of tumor cells. However, the expression of its ligand, the chemokine CXCL12, in tumors and its role in regulating the accumulation of immune cells within the tumors is not clear. Using ELISA and immunohistochemistry we found that CXCL12 is expressed in the majority of nonsmall cell lung cancer tissue sections obtained from stage IA to IIB nonsmall cell lung cancer patients undergoing operation. Histopathologic examination of these sections indicated that high CXCL12 expression correlated with increased tumor inflammation. In addition, disease recurrence rates in a subgroup of adenocarcinoma patients showed a tendency to correlate with high CXCL12 expression in the tumor. Isolation of adenocarcinoma-infiltrating immune cells demonstrated an increase in the percentage of CD4+CD69+CXCR4+ T cells as compared with normal lung tissue. About 30% of these cells expressed the regulatory T cell markers CD25high and FoxP3. The percentage of CD8 T cells within the tumor did not change, however; the percentage of NK and NK T cells was significantly reduced. In correlation with CXCR4 expression, CD4 T cells showed increased migration in response to CXCL12 compared with CD8 T cells and NK cells. Overall, these observations suggest that CXCL12 expression may influence tumor progression by shaping the immune cell population infiltrating lung adenocarcinoma tumors.  相似文献

Previously, we have shown that overexpression of 4-hydroxy-2-nonenal (HNE)-detoxifying enzyme glutathione S-transferase A4-4 (hGSTA4-4) in human lens epithelial cells (HLE B-3) leads to pro-carcinogenic phenotypic transformation of these cells [R. Sharma, et al. Eur. J. Biochem. 271 (2004) 1960-1701]. We now demonstrate that hGSTA4-transfection also causes a profound change in the expression of genes involved in cell adhesion, cell cycle control, proliferation, cell growth, and apoptosis, which is consistent with phenotypic changes of the transformed cells. The expression of p53, p21, p16, fibronectin 1, laminin gamma1, connexin 43, Fas, integrin alpha6, TGFalpha, and c-jun was down-regulated, while the expression of protein kinase C beta II (PKCbetaII), c-myc, cyclin-dependent kinase 2 (CDK2), and TGFbeta was up-regulated in transfected cells. These results demonstrate that HNE serves as a crucial signaling molecule and, by modulating the expression of genes, can influence cellular functions.  相似文献

腺病毒E4启动子结合蛋白-4(E4BP4)是哺乳动物细胞核内的一种碱性亮氨酸拉链(bZIP)型转录因子,参与调控细胞的存活和增殖。前期研究表明,它在孕第5天的小鼠着床位点有明显的高表达。本文分别应用Northem blot、in situ杂交、Western blot和免疫组织化学技术,对E4BP4基因在小鼠妊娠初始期子宫、着床期胚胎着床位点和非着床位点的表达情况进行了研究。观察发现:在小鼠妊娠初始期,E4BP4基因在子宫组织中的表达逐步上调;至胚胎着床期间,其在胚胎着床位点的表达水平进一步提高,并明显高于非着床位点;该基因的表达不依赖于胚胎,人工蜕膜化可诱导其表达:E4BP4 mRNA和E4BP4蛋白分子都主要分布于子宫腔周围的基质细胞和蜕膜细胞。上述结果提示E4BP4基因可能通过促进着床位点基质细胞的增殖和抑制蜕膜细胞的凋亡而参与胚胎着床过程的调控。  相似文献

CXCL16 is a transmembrane non-ELR CXC chemokine that signals via CXCR6 to induce aortic smooth muscle cell (ASMC) proliferation. While bacterial lipopolysaccharide (LPS) has been shown to stimulate CXCL16 expression in SMC, its effects on CXCR6 are not known. Here, we demonstrate that LPS upregulates CXCR6 mRNA, protein, and surface expression in human ASMC. Inhibition of TLR4 with neutralizing antibodies or specific siRNA interference blocked LPS-mediated CXCR6 expression. LPS stimulated both AP-1 (c-Fos, c-Jun) and NF-kappaB (p50 and p65) activation, but only inhibition of AP-1 attenuated LPS-induced CXCR6 expression. Using dominant negative expression vectors and siRNA interference, we demonstrate that LPS induces AP-1 activation via MyD88, TRAF6, ERK1/2, and JNK signaling pathways. Furthermore, the flavoprotein inhibitor diphenyleniodonium chloride significantly attenuated LPS-mediated AP-1-dependent CXCR6 expression, as did inhibition of NOX4 NADPH oxidase by siRNA. Finally, CXCR6 knockdown inhibited CXCL16-induced ASMC proliferation. These results demonstrate that LPS-TLR4-NOX4-AP-1 signaling can induce CXCR6 expression in ASMC, and suggest that the CXCL16-CXCR6 axis may be an important proinflammatory pathway in the pathogenesis of atherosclerosis.  相似文献

The murine tooth development is governed by sequential and reciprocal epithelial-mesenchymal interactions. Multiple signaling molecules are expressed in the developing tooth germ and interact each other to mediate the inductive tissue interactions. Among them are Sonic hedgehog (SHH), Bone Morphogenetic Protein-2 (BMP2) and Bone Morphogenetic Protein-4 (BMP4). We have investigated the interactions between these signaling molecules during early tooth development. We found that the expression of Shh and Bmp2 is downregulated at E12.5 and E13.5 in the dental epithelium of the Msx1 mutant tooth germ where Bmp4 expression is significantly reduced in the dental mesenchyme. Inhibition of BMP4 activity by noggin resulted in repression of Shh and Bmp2 in wild-type dental epithelium. When implanted into the dental mesenchyme of Msx1 mutants, beads soaked with BMP4 protein were able to restore the expression of both Shh and Bmp2 in the Msx1 mutant epithelium. These results demonstrated that mesenchymal BMP4 represents one component of the signal acting on the epithelium to maintain Shh and Bmp2 expression. In contrast, BMP4-soaked beads repressed Shh and Bmp2 expression in the wild-type dental epithelium. TUNEL assay indicated that this suppression of gene expression by exogenous BMP4 was not the result of an increase in programmed cell death in the tooth germ. Ectopic expression of human Bmp4 to the dental mesenchyme driven by the mouse Msx1 promoter restored Shh expression in the Msx1 mutant dental epithelium but repressed Shh in the wild-type tooth germ in vivo. We further demonstrated that this regulation of Shh expression by BMP4 is conserved in the mouse developing limb bud. In addition, Shh expression was unaffected in the developing limb buds of the transgenic mice in which a constitutively active Bmpr-IB is ectopically expressed in the forelimb posterior mesenchyme and throughout the hindlimb mesenchyme, suggesting that the repression of Shh expression by BMP4 may not be mediated by BMP receptor-IB. These results provide evidence for a new function of BMP4. BMP4 can act upstream to Shh by regulating Shh expression in mouse developing tooth germ and limb bud. Taken together, our data provide insight into a new regulatory mechanism for Shh expression, and suggest that this BMP4-mediated pathway in Shh regulation may have a general implication in vertebrate organogenesis.  相似文献

Oxidative stress and the accumulation of reactive oxygen specie (ROS) play a role in cancer cells developing an advanced, phenotypic signature that associates with metastasis and progression. Increased ROS concentrations are involved in promoting cancer development and metastasis by inducing expression of oncogenes, suppressing activity of anti-survival molecules and by activating various cell survival and proliferation signaling pathways. Oxidative stress is higher in the epithelium of cancer patients than patients without the disease, and antioxidant trials are currently being explored as a therapeutic option. However, studies have shown that ROS increases expression of CXCR4 in cancer and immune cells. CXCR4 expression in tumors strongly correlates to metastasis and poor prognosis. Fibroblast growth factor receptor (FGFR) signalling is important in the initiation and regulation of osteogenesis. Although mutations in FGFR1, 2, and 3 genes are known to cause skeletal deformities, the expression of FGFR4 in bony tissue remains unclear. We have investigated the expression pattern of FGFR4 in the neonatal mouse calvaria and compared it to the expression pattern in cultures of primary osteoblasts. Immunohistochemistry demonstrated that FGFR4 was highly expressed in rudimentary membranous bone and strictly localised to the cellular components (osteoblasts) between the periosteal and endosteal layers. Cells in close proximity to the newly formed osteoid (preosteoblasts) also expressed FGFR4 on both the endosteal and periosteal surfaces. Immunocytochemical analysis of primary osteoblast cultures taken from the same cranial region also revealed high levels of FGFR4 expression, suggesting a similar pattern of cellular expression in vivo and in vitro. RT-PCR and Western blotting for FGFR4 confirmed its presence in primary osteoblast cultures. These results suggest that FGFR4 may be an important regulator of osteogenesis with involvement in preosteoblast proliferation and differentiation as well as osteoblast functioning during intramembranous ossification. The consistent expression of FGFR4 in vivo and in vitro supports the use of primary osteoblast cultures for elucidating the role of FGFR4 during osteogenesis.  相似文献

血管内皮素生长因子受体3(vascular endothelial growth factor receptor 3,VEGFR-3/FLT4)与黑色素瘤细胞的生长有关,其可能对毛色及黑色素生成有一定的调控作用。为探讨FLT4基因对羊驼毛色及黑色素生成的影响,本文采用免疫组织化学、qRT-PCR、Western印迹对FLT4在不同毛色羊驼皮肤毛囊中的表达进行了研究。结果显示,FLT4在不同毛色毛囊中外根鞘及毛乳头阳性表达不同,且棕色皮肤比白色皮肤阳性信号强;FLT4 mRNA在棕色羊驼皮肤的表达量明显高于白色羊驼皮肤 (P<0.01);FLT4蛋白在棕色皮肤中的表达量明显高于白色皮肤(P<0.01);为进一步研究FLT4对黑色素生成的影响;通过对体外培养的羊驼皮肤黑色素细胞中添加不同浓度的FLT4蛋白(0, 1, 10, 50, 100 ng/mL),与对照组相比,添加FLT4蛋白后酪氨酸酶(TYR)、酪氨酸相关蛋白-1(TYRP1)、受体酪氨酸激酶 c-KIT、小眼畸形相关转录因子(MITF)的 mRNA和蛋白质表达明显增加, 10 ng/mL组表达量最高(P<0.01);黑色素含量测定结果表明,不同浓度的FLT4蛋白处理的羊驼皮肤黑色素细胞的黑色素含量比对照组明显升高,添加FLT4蛋白10 ng/mL表达量最高(P<0.01)。由此可知,FLT4可以通过调节毛色相关基因的表达进而引起黑色素细胞中黑色素含量的变化。  相似文献

以实验室前期获得的转SlNAC4基因拟南芥(Arabidopsis thaliana)和野生型拟南芥为材料, 通过基因芯片技术检测其基因表达谱。结果表明, 与野生型拟南芥相比, 转SlNAC4基因拟南芥中差异表达的基因共有3 094个。 通过GO分析, 发现与非生物胁迫相关的差异基因共有195个, 与生长发育相关的差异基因共有47个, 其中包含MYB和WRKY转录因子基因。KEGG分析表明, 差异表达基因主要涉及植物激素信号转导和油菜素内酯合成等信号通路。进一步选择部分差异表达基因进行实时荧光定量PCR分析, 所得结果与芯片检测结果一致。该研究结果表明, SlNAC4可直接或间接地调控多个下游基因的表达, 进而调控植物的生长发育, 提高其抗逆性。  相似文献

Background

Robo4 is involved in hematopoietic stem/progenitor cell homeostasis and essential for tumor angiogenesis. Expression of Robo4 was recently found in solid tumors and leukemia stem cells. However, the clinical implications of Robo4 expression in patients with acute myeloid leukemia (AML) remain unclear.

Methods

We investigated the clinical and prognostic relevance of mRNA expression of Robo4 in bone marrow (BM) mononuclear cells from 218 adult patients with de novo AML. We also performed immunohistochemical staining to assess the Robo4 protein expression in the BM biopsy specimens from 30 selected AML patients in the cohort.

Results

Higher Robo4 expression was closely associated with lower white blood cell counts, expression of HLA-DR, CD13, CD34 and CD56 on leukemia cells, t(8;21) and ASXL1 mutation, but negatively correlated with t(15;17) and CEBPA mutation. Compared to patients with lower Robo4 expression, those with higher expression had significantly shorter disease-free survival (DFS) and overall survival (OS). This result was confirmed in an independent validation cohort. Furthermore, multivariate analyses showed that higher Robo4 expression was an independent poor prognostic factor for DFS and OS in total cohort and patients with intermediate-risk cytogenetics, irrespective of age, WBC count, karyotype, and mutation status of NPM1/FLT3-ITD, and CEBPA.

Conclusions

BM Robo4 expression can serve as a new biomarker to predict clinical outcomes in AML patients and Robo4 may serve as a potential therapeutic target in patients with higher Robo4 expression.  相似文献
