Imaging modalities that can be used intra-operatively do not provide sufficient details to confidently locate the abnormalities and critical healthy areas that have been identified from high-resolution pre-operative scans. However, as we have shown in our previous work, high quality pre-operative images can be warped to the intra-operative position of the brain. This can be achieved by computing deformations within the brain using a biomechanical model. In this paper, using a previously developed patient-specific model of brain undergoing craniotomy-induced shift, we conduct a parametric analysis to investigate in detail the influences of constitutive models of the brain tissue. We conclude that the choice of the brain tissue constitutive model, when used with an appropriate finite deformation solution, does not affect the accuracy of computed displacements, and therefore a simple linear elastic model for the brain tissue is sufficient.  相似文献

We employ a structurally-motivated phenomenological formulation to identify biomechanical experiments which can be used to determine a vascular constitutive relation directly from data. Large deformations, nonlinear material behavior, load-dependent anisotropy, material heterogeneity and incompressibility are accounted for in the analysis. For purposes of illustration, we outline a procedure for studying elastic arteries wherein the behavior of the media and adventitia is considered separately. This general approach for identifying vascular constitutive relations can be applied to any vessel or airway, however, and should provide certain advantages over previous microstructural or purely phenomenological formulations.  相似文献

Using the three-dimensional morphological data provided by computed tomography, finite element (FE) models can be generated and used to compute the stiffness and strength of whole bones. Three-dimensional constitutive laws capturing the main features of bone mechanical behavior can be developed and implemented into FE software to enable simulations on complex bone structures. For this purpose, a constitutive law is proposed, which captures the compressive behavior of trabecular bone as a porous material with accumulation of irreversible strain and loss of stiffness beyond its yield point and softening beyond its ultimate point. To account for these features, a constitutive law based on damage coupled with hardening anisotropic elastoplasticity is formulated using density and fabric-based tensors. To prevent mesh dependence of the solution, a nonlocal averaging technique is adopted. The law has been implemented into a FE software and some simple simulations are first presented to illustrate its behavior. Finally, examples dealing with compression of vertebral bodies clearly show the impact of softening on the localization of the inelastic process.  相似文献

High-level production of human leptin by fed-batch culture of recombinant Escherichia coli using constitutive promoter system was investigated. For the constitutive expression of the obese gene encoding human leptin, the strong constitutive HCE promoter cloned from the D-amino acid aminotransferase gene of Geobacillus toebii was used. To develop an optimal host-vector system, several different recombinant E. coli strains were compared for leptin production. In flask cultures, E. coli FMJ123, which is a rpoS mutant strain, showed the highest level of leptin production (41% of total proteins). By comparing the expression levels of leptin in several different rpoS- and rpoS+ strains, it could be concluded that rpoS mutation positively affected constitutive production of leptin. For the large-scale production of human leptin, fed-batch cultures of recombinant E. coli FMJ123 were carried out using three different feeding solutions--chemically defined, yeast extract-containing, and casamino acid-containing feeding solutions. Among these, the use of casamino acid-containing feeding solution allowed production of leptin up to 2.1 g/L, which was 2.1- and 1.8-fold higher than that obtained with chemically defined and yeast extract-contained feeding solutions, respectively. These results suggest that the HCE promoter can be used for the efficient production of leptin, and most likely other recombinant proteins, in a constitutive manner.  相似文献

The interaction of rhodopsin and transducin has been the focus of study for more than 30 years, but only recently have efforts to purify an activated complex in detergent solution materialized. These efforts have used native rhodopsin isolated from bovine retina and employed either sucrose density gradient centrifugation or size exclusion chromatography to purify the complex. While there is general agreement on most properties of the activated complex, subunit stoichiometry is not yet settled, with rhodopsin/transducin molar ratios of both 2/1 and 1/1 reported. In this report, we introduce methods for preparation of the complex that include use of recombinant rhodopsin, so as to take advantage of mutations that confer constitutive activity and enhanced thermal stability on the protein, and immunoaffinity chromatography for purification of the complex. We show that chromatography on ConA-Sepharose can substitute for the immunoaffinity column and that bicelles can be used instead of detergent solution. We demonstrate the following: that rhodopsin has a covalently bound all-trans-retinal chromophore and therefore corresponds to the active metarhodopin II state; that transducin has an empty nucleotide-binding pocket; that the isolated complex is active and dissociates upon addition of guanine nucleotide; and finally that the stoichiometry corresponds reproducibly to a 1/1 molar ratio of rhodopsin to transducin.  相似文献

Constitutive and thermoinducible expression plasmids based on strong P(R),P(L) promoters from phage lambda were compared for production of TNF-alpha and its analogs under various conditions. Much higher accumulation of TNF was obtained in a constitutive system, so the wider applicability of such systems was studied. In constitutive systems, proteolytically susceptible proteins can be produced easily at low cultivation temperatures and the addition of expensive or toxic chemical inducers is not required. On the other hand, toxic proteins cannot be produced and selection pressure must be strictly maintained to ensure segregational stability of plasmids. Accumulation of TNF-alpha and various analogs at levels up to 25% of total soluble protein was repeatedly achieved, which was 2-3-fold higher than in a thermoinducible system. The stable behavior of the constitutive system in laboratory fermentors was also confirmed. We propose the constitutive system described here as a general model for many currently used expression systems containing strong but not completely repressed promoters. Such systems may be considered as constitutive ones with reduced promoter strengths, but still exhibiting all the intrinsic properties of constitutive expression systems. Although all modern expression systems are inducible, wider use of a constitutive system is evidently possible.  相似文献

The correlation between constitutive and induced resistance to herbivores in plants has long been of interest to evolutionary biologists, and various approaches to determining levels of resistance have been used in this field of research. In this study, we examined the relationship between constitutive and induced resistance to the diamondback moth, Plutella xylostella (L.) (Lepidoptera: Plutellidae), in 11 closely related species of wild crucifers. We assessed the survival, development, and reproduction of the test insects and calculated their intrinsic rate of increase as an indicator of constitutive and induced resistance for the plants. We used larvae of P. xylostella and jasmonic acid as elicitors of the induced response. We failed to find a correlation between constitutive and induced resistance in these crucifer plants when the induction of resistance was initiated by either herbivory or jasmonic acid application. Analysis of the results suggests that the failure to detect a relationship between the two types of resistance could be caused by flaws in measuring constitutive resistance, which was apparently confounded with induced resistance. We discuss the difficulties and pitfalls in measuring constitutive resistance and ways to improve the methodology in investigating the relationships between constitutive and induced resistance in plants.  相似文献

Although it has been suggested that induced and constitutive plant resistance should have different effects on insect herbivore population dynamics, there is little experimental evidence that plant resistance can influence herbivore populations longer than one season. We used a density-manipulation experiment and model fitting to examine the effects of constitutive and induced resistance on herbivore dynamics over both the short and long term. We used likelihood methods to fit population dynamic models to recruitment data for populations of Mexican bean beetles on soybean varieties with no resistance, constitutive resistance, or induced resistance. We compared model configurations that fit parameters for resistance types separately to models that did not account for resistance type. Models representing the hypothesis that the three resistance types differed in their effects on beetle dynamics received the most support. Induced resistance resulted in lower population growth rates and stronger density dependence than no resistance. Constitutive resistance resulted in lower population growth rates and stronger density dependence than induced resistance. Constitutive resistance had a stronger effect on both short-term beetle recruitment and predicted beetle population dynamics than induced resistance. The results of this study suggest that induced and constitutive resistance can differ in their effects on herbivore populations even in a relatively complex system.  相似文献

By using a combination of continuum and statistical mechanics we derive an integral constitutive relation for bio-artificial tissue models consisting of a monodisperse population of cells in a uniform collagenous matrix. This constitutive relation quantitatively models the dependence of tissue stress on deformation history, and makes explicit the separate contribution of cells and matrix to the mechanical behavior of the composite tissue. Thus microscopic cell mechanical properties can be deduced via this theory from measurements of macroscopic tissue properties. A central feature of the constitutive relation is the appearance of "anisotropy tensors" that embody the effects of cell orientation on tissue mechanics. The theory assumes that the tissues are stable over the observation time, and does not in its present form allow for cell migration, reorientation, or internal remodeling. We have compared the predictions of the theory to uniaxial relaxation tests on fibroblast-populated collagen matrices (FPMs) and find that the experimental results generally support the theory and yield values of fibroblast contractile force and stiffness roughly an order of magnitude smaller than, and viscosity comparable to, the corresponding properties of active skeletal muscle. The method used here to derive the tissue constitutive equation permits more sophisticated cell models to be used in developing more accurate representations of tissue properties.  相似文献

We present a new constitutive formulation that combines certain desirable features of two previously used approaches (phenomenological and microstructural). Specifically, we assume that certain soft tissues can be idealized as composed of various families of noninteracting fibers and a homogeneous matrix. Both the fibers and the matrix are assumed to follow the gross deformation. Within the usual framework of pseudoelasticity, incompressibility, homogeneity, and the continuum hypothesis, a pseudostrain-energy function (W) is proposed wherein W is expressed in terms of matrix and fibrous contributions. Unlike phenomenological approaches where a W is usually chosen in an ad hoc manner, the present approach can be used to postulate reasonable forms of W based on limited structural information and multiaxial stress-strain data. Illustrative applications of the theory are discussed for visceral pleura and myocardium. Concise structurally motivated constitutive relations result, wherein load-dependent anisotropy, nonlinear material behavior, finite deformations, and incompressibility are accounted for.  相似文献

We measured a number of pigmentation and skin response phenotypes in a sample of volunteers (n=397) living in State College, PA. The majority of this sample was composed of four groups based on stated ancestry: African-American, European-American, Hispanic and East Asian. Several measures of melanin concentration (L*, melanin index and adjusted melanin index) were estimated by diffuse reflectance spectroscopy and compared. The efficacy of these measures for assessing constitutive pigmentation and melanogenic dose-response was evaluated. Similarly, several measures of erythema (a*, erythema index and adjusted erythema index) were compared and evaluated in their efficacy in measuring erythema and erythemal dose-response. We show a high correspondence among all of the measures for the assessment of constitutive pigmentation and baseline erythema. However, our results demonstrate that evaluating melanogenic dose-response is highly dependent on the summary statistic used: while L* is a valid measure of constitutive pigmentation it is not an effective measure of melanogenic dose-response. Our results also confirm the use of a*, as it is shown to be highly correlated with the adjusted erythema index, a more advanced measure of erythema based on the apparent absorbance. Diffuse reflectance spectroscopy can be used to quantify the constitutive pigmentation, melanogenic dose-response at 7 d and erythemal dose-response at both 24 h and 7 d postexposure.  相似文献

SR proteins are a conserved family of splicing factors that function in both constitutive and activated splicing. We reported previously that phosphorylation of the SR protein ASF/SF2 enhances its interaction with the U1 snRNP-specific 70K protein and is required for the protein to function in splicing, while other studies have provided evidence that subsequent dephosphorylation can also be required for SR protein function, at least in constitutive splicing. We now show that the phosphorylation status of ASF/SF2 can differentially affect several properties of the protein. In keeping with a dynamic cycle of phosphorylation-dephosphorylation during splicing, ASF/SF2 phosphorylation was found to affect interaction with several putative protein targets in different ways: positively, negatively or not at all. Extending these results, we also show that, in contrast to constitutive splicing, dephosphorylation is not required for ASF/SF2 to function as a splicing activator. We discuss these results with respect to the differential protein-protein interactions that must occur during constitutive and activated splicing.  相似文献

Constitutive and induced plant resistance against herbivores occurs throughout the plant kingdom, but little is known about the evolutionary relationship between these two types of resistances. We examined the relationships between constitutive and induced resistance to the diamondback moth, Plutella xylostella, in 11 wild species of crucifers, and analyzed the changes in volatiles associated with their expression in two species. We used larvae of P. xylostella and jasmonic acid (JA) as elicitors of the induced response. The level of resistance was estimated as the relative number of eggs laid on the plants by P. xylostella. Substantial variation in constitutive resistance was observed among the 11 crucifer species. When the plants were damaged by larvae, a negative correlation was found between constitutive and induced resistance. However, a positive correlation was detected between constitutive and induced resistance when the plants were treated by JA. The shift in resistance was associated with changes in the emission of volatiles. These results strongly suggest that (1) a trade-off occurs between constitutive and induced resistance in wild crucifers, and that (2) such a trade-off can be observed by treating the plants with a natural, but not an artificial, elicitor.  相似文献

We report a new and improved pPZP vector (pPZP3425) for efficient plant transformation. This vector is derived from the widely used pPZP100 series of binary Agrobacterium vectors. One disadvantage of these vectors is the use of chloramphenicol resistance for selection in Escherichia coli and Agrobacteria. We have therefore included a kanamycin resistance gene for selection in Agrobacterium. Furthermore, the strong 35S CaMV promoter driving the plant resistance gene has been replaced by the weaker nos promoter because it has been shown that the 35S promoter driving the plant resistance marker can lead to ectopic expression of the transgene. During replacement of the 35S promoter, the NcoI site within the plant resistance gene has been removed, and NcoI can now be used for cloning purposes within the expression cassette which consists of an intron-containing gus gene driven by a strong constitutive promoter (35S promoter with doubled enhancer plus omega-element as translational enhancer). Thus, a single vector can conveniently be used for two purposes: (1) for overexpression of proteins by replacing the gus gene by the coding sequence of choice and (2) for creation of promoter:gus fusions by substituting the constitutive promoter by any other promoter. We demonstrate the usefulness of this vector for cloning a promoter:gus fusion and in planta transformation of Arabidopsis.  相似文献

A synthetic peptide patterned after the sequence of the inactivating "ball" domain of the Shaker B K(+) channel restores fast (N-type) inactivation in mutant deletion channels lacking their constitutive ball domains, as well as in K(+) channels that do not normally inactivate. We now report on the effect of phosphorylation at a single tyrosine in position 8 of the inactivating peptide both on its ability to restore fast channel inactivation in deletion mutant channels and on the conformation adopted by the phosphorylated peptide when challenged by anionic lipid vesicles, a model target mimicking features of the inactivation site in the channel protein. We find that the inactivating peptide phosphorylated at Y8 behaves functionally as well as structurally as the noninactivating mutant carrying the mutation L7E. Moreover, it is observed that the inactivating peptide can be phosphorylated by the Src tyrosine kinase either as a free peptide in solution or when forming part of the membrane-bound protein channel as the constitutive inactivating domain. These findings suggest that tyrosine phosphorylation-dephosphorylation of this inactivating ball domain could be of physiological relevance to rapidly interconvert fast-inactivating channels into delayed rectifiers and vice versa.  相似文献

Most animal cells are surrounded by a thin layer of actin meshwork below their membrane, commonly known as the actin cortex (or cortical membrane). An increasing number of studies have highlighted the role of this structure in many cell functions including contraction and locomotion, but modelling has been limited by the fact that the membrane thickness (about 1?μm) is usually much smaller than the typical size of a cell (10-100?μm). To overcome theoretical and numerical issues resulting from this observation, we introduce in this paper a continuum formulation, based on surface elasticity, that views the cortex as an infinitely thin membrane that can resists tangential deformation. To accurately model the large deformations of cells, we introduced equilibrium equations and constitutive relations within the Eulerian viewpoint such that all quantities (stress, rate of deformation) lie in the current configuration. A solution procedure is then introduced based on a coupled extended finite element approach that enables a continuum solution to the boundary value problem in which discontinuities in both strain and displacement (due to cortical elasticity) are easily handled. We validate the approach by studying the effect of cortical elasticity on the deformation of a cell adhering on a stiff substrate and undergoing internal contraction. Results show very good prediction of the proposed method when compared with experimental observations and analytical solutions for simple cases. In particular, the model can be used to study how cell properties such as stiffness and contraction of both cytoskeleton and cortical membrane lead to variations in cell's surface curvature. These numerical results show that the proposed method can be used to gain critical insights into how the cortical membrane affects cell deformation and how it may be used as a means to determine a cell's mechanical properties by measuring curvatures of its membrane.  相似文献

Most recent finite element models that represent muscles are generic or subject-specific models that use complex, constitutive laws. Identification of the parameters of such complex, constitutive laws could be an important limit for subject-specific approaches. The aim of this study was to assess the possibility of modelling muscle behaviour in compression with a parametric model and a simple, constitutive law. A quasi-static compression test was performed on the muscles of dogs. A parametric finite element model was designed using a linear, elastic, constitutive law. A multi-variate analysis was performed to assess the effects of geometry on muscle response. An inverse method was used to define Young's modulus. The non-linear response of the muscles was obtained using a subject-specific geometry and a linear elastic law. Thus, a simple muscle model can be used to have a bio-faithful, biomechanical response.  相似文献
