The lactobacilli probiotics maintain a normal vaginal biota and prevent disease recurrence. This microorganisms form a pellicle on the vaginal epithelium that acts as a biologic barrier against colonization by pathogenic bacteria. In this paper were realized assays of exclusion, competition, and displacement. For these test, vaginal epithelial cells, two strains of lactobacilli and pathogenic bacteria (Staphylococcus aureus, Streptococcus agalactiae and Listeria monocytogenes) were used. The lactobacilli strains showed a great capacity of adherence, with a mean of 83.5 ± 26.67 Lactobacillus fermentum cells and 56.2 ± 20.87 Lactobacillus rhamnosus cells per vaginal epithelial cells. L. fermentum and L. rhamnosus were able to reduce the adherence of S. aureus, S. agalactiae and L. monocytogenes in a significant level in this assay (P < 0.01). The lactobacilli used in this study protect the vaginal epithelium through a series of barriers and interference mechanisms. The aim of present study was to assess the ability of vaginal Lactobacillus strains, selected for their probiotic properties, to block the adherence of pathogenic microorganisms in vitro by displacement, competition, and exclusion mechanisms.  相似文献

The sequence of bacterial events that occurs during the colonization of the gastrointestinal tract may affect the future health of the host. A clear understanding of the colonization process of the human neonatal gut in developing countries is lacking because the few available studies were mostly performed using culture techniques. Using molecular approaches, this study analyzed the fecal microbiota of children of low socioeconomic status in São Paulo, Brazil, during their first year of life. We collected fecal samples of healthy children at 3, 6, and 12 months of life. Total DNA was extracted directly from feces, and the bacteria-specific primers 27F-1492R were used to construct 16S rRNA libraries. Clones were randomly selected and partially sequenced. The main phylogenetic groups identified at 3 months were Streptococcus, unidentified bacteria, and Escherichia. At 6 months, Escherichia remained predominant, while the unidentified bacterial population increased significantly. At 12 months, a more complex composition of fecal microbiota was observed, represented by unidentified bacteria and microorganisms found at low rates at earlier ages. The genus Escherichia remained the most abundant microorganism (34 % relative abundance and 75 % prevalence). Principal component analysis (PCA) revealed changes in the composition of the microbiota at 6 months and an increase of diversity at 12 months of life. Bifidobacterium was identified by quantitative PCR (qPCR) and showed a high incidence in the microbiota at 3 months. The present results corroborate the global observation of inter-individual variability with an early establishment of microbial complexity at the end of the first year of life and highlight the presence of the Escherichia as abundant in microbiota composition of this group of children.  相似文献

Abstract

Many cave-dwelling microorganisms have been studied, but little is known about whether genes related to the nitrogen cycle are present in this environment, particularly in sandstone caves. The profiles of nitrogen fixation (nifH) and archaeal and bacterial ammonia oxidation (amoA) genes were analyzed along a cave of metasandstone and limestone lithologies. Using polymerase chain reaction (PCR)-denaturing gradient gel electrophoresis and quantitative PCR to evaluate these genes, we analyzed the variations in the microorganisms and frequency of these genes along the caves. In both caves analyzed for nifH, different community profiles were observed, particularly in entrance samples. Quantitative PCR of the genes showed that samples from the entrance of the caves contained a larger number of copies of amoA. In addition, archaeal amoA was predominant compared to bacterial amoA. The present study demonstrated that the genes for nitrogen fixation and ammonia oxidation are present in both cave lithologies and that their frequency and the organisms present along the caves vary with factors such as the characteristics of the cave itself.  相似文献

Here we describe recent breakthroughs in our understanding of microbial life in dry volcanic tephra (“soil”) that covers much of the surface area of the highest elevation volcanoes on Earth. Dry tephra above 6000 m.a.s.l. is perhaps the best Earth analog for the surface of Mars because these “soils” are acidic, extremely oligotrophic, exposed to a thin atmosphere, high UV fluxes, and extreme temperature fluctuations across the freezing point. The simple microbial communities found in these extreme sites have among the lowest alpha diversity of any known earthly ecosystem and contain bacteria and eukaryotes that are uniquely adapted to these extreme conditions. The most abundant eukaryotic organism across the highest elevation sites is a Naganishia species that is metabolically versatile, can withstand high levels of UV radiation and can grow at sub-zero temperatures, and during extreme diurnal freeze–thaw cycles (e.g. ??10 to +?30 °C). The most abundant bacterial phylotype at the highest dry sites sampled (6330 m.a.s.l. on Volcán Llullaillaco) belongs to the enigmatic B12-WMSP1 clade which is related to the Ktedonobacter/Thermosporothrix clade that includes versatile organisms with the largest known bacterial genomes. Close relatives of B12-WMSP1 are also found in fumarolic soils on Volcán Socompa and in oligotrophic, fumarolic caves on Mt. Erebus in Antarctica. In contrast to the extremely low diversity of dry tephra, fumaroles found at over 6000 m.a.s.l. on Volcán Socompa support very diverse microbial communities with alpha diversity levels rivalling those of low elevation temperate soils. Overall, the high-elevation biome of the Atacama region provides perhaps the best “natural experiment” in which to study microbial life in both its most extreme setting (dry tephra) and in one of its least extreme settings (fumarolic soils).  相似文献

The aim of this study was to survey microorganisms from the deep surface sediment samples collected from the Sea of Japan and to screen them for antimicrobial and antagonistic effects. Phylogenetic analysis revealed most isolates sharing 98–100 % sequence similarity to recognized species, including those recovered previously from marine or saline environments. Alteromonas, Halomonas, Marinobacter, Pseudoalteromonas, Salinicola, within the class Gammaproteobacteria, Sulfitobacter (Alphaproteobacteria), Bacillus, Paenibacillus and Paenisporosarcina (Firmicutes), Nocardiopsis and Streptomyces (Actinobacteria) occurred abundantly in all sediment samples. Antimicrobial screening revealed twenty three strains (13 %) capable to inhibit growth of one to eight test cultures and deep sediment isolates. Based on phylogenetic analysis mostly active strains belonged to the genera Bacillus, Brevibacillus, Nocardiopsis, Paenibacillus and Streptomyces. Antimicrobial substances (1–3) were isolated from strain Paenibacillus sp. Sl 79w showing a high inhibitory activity. On the basis of combined spectral analyses (IR, UV, 1H and 13C NMR) the compounds 1, 2 and 3 with [M + H]+ at 409.1 and 409.2 m/z, and with [M + Na]+ at 822.5 m/z were found to have a carbon skeleton of isocoumarin and peptide antibiotics, respectively. Our findings demonstrated that the deep surface sediments of the Sea of Japan represent an untapped source of diverse microorganisms capable of antimicrobial metabolite production.  相似文献

Phenol is a toxic aromatic compound used or produced in many industries and as a result a common component of industrial wastewaters. Phenol containing waste streams are frequently hypersaline and therefore require halophilic microorganisms for efficient biotreatment without dilution. In this study three halophilic bacteria isolated from different saline environments and identified as Halomonas organivorans, Arhodomonas aquaeolei and Modicisalibacter tunisiensis were shown to be able to grow on phenol in hypersaline media containing 100 g/L of total salts at a concentration of 3 mM (280 mg/L), well above the concentration found in most waste streams. Genes encoding the aromatic dioxygenase enzymes catechol 1,2 dioxygenase and protocatechuate 3,4-dioxygenase were present in all strains as determined by PCR amplification using primers specific for highly conserved regions of the genes. The gene for protocatechuate 3,4-dioxygenase was cloned from the isolated H. organivorans and the translated protein was evaluated by comparative protein sequence analysis with protocatechuate 3,4-dioxygenase proteins from other microorganisms. Although the analysis revealed a wide range of sequence divergence among the protocatechuate 3,4-dioxygenase family, all of the conserved domain amino acid structures identified for this enzyme family are identical or conservatively substituted in the H. organivorans enzyme.  相似文献

Thirty-eight faecal samples from the Plateau zokor, Myospalax baileyi Thomas, collected in the Haibei Area, Qinghai Province, China, were examined for the presence of coccidia (Apicomplexa: Eimeriidae). Seventeen of 38 faecal samples (44.7%) were found to contain coccidian oöcysts representing four new species of Eimeria Schneider, 1875, and four of 17 (23.5%) infected zokors were concurrently infected with two or three of these eimerian species. The sporulated oöcysts of Eimeria myospalacensis n. sp. are ovoidal, 9.5–17.0 × 8.0–13.0 (mean 13.0 × 10.4) μm; a polar granule is present, oöcyst residuum is absent; sporocysts are ovoidal, 4.5–7.5 × 3.0–5.0 (mean 6.3 × 4.2) μm and have both a Stieda body and residuum. Oöcysts of Eimeria fani n. sp. are ellipsoidal to cylindroidal, 12.5–16.0 × 8.0–11.0 (mean 14.6 × 9.9) μm; a polar granule is present, but micropyle and residuum are lacking; sporocysts are ovoidal, 4.5–7.5 × 3.0–5.3 (mean 6.7 × 4.4) μm; a residuum and a Steida body are present. Oöcysts of Eimeria baileyii n. sp. are ellipsoidal, 15.0–23.0 × 12.0–18.0 (mean 18.2 × 13.7) μm; a polar granule is present but oöcyst residuum is absent; sporocysts are ovoidal, 8.0–11.0 × 5.0–7.0 (mean 9.5 × 5.9) μm and have both a Stieda body and residuum. Oöcysts of Eimeria menyuanensis n. sp. are ovoidal, 12.5–21.0 × 11.0–18.0 (mean 17.1 × 14.6) μm, with a distinct micropyle c.2.5 μm wide; a polar granule is present but a residuum is absent; sporocysts are ovoidal, 8.0–12.0 × 5.0–7.0 (mean 10.2 × 6.4) μm, and have both a Stieda body and residuum.  相似文献

Two new species of Eimeria Schneider, 1875, from emerald tree skinks, Lamprolepis smaragdina (Lesson) are described from specimens collected in Papua New Guinea (PNG) and the Philippines. Oöcysts of Eimeria nuiailan n. sp. from the only L. smaragdina from PNG are ovoidal, with a smooth, colourless, bi-layered wall, measure 23.7 × 19.1 μm, and have a length/width (L/W) ratio of 1.3; both micropyle and oöcyst residuum are absent, but a fragmented polar granule is present. Sporocysts are ovoidal to ellipsoidal, 11.9 × 7.0 μm, L/W 1.7, and the wall is composed of two valves joined by a longitudinal suture; neither Stieda nor sub-Stieda bodies are present; a sporocyst residuum is present as a compact mass of granules. Sporozoites are elongate, 14.6 × 2.6 μm, and contain anterior and posterior refractile bodies with a nucleus between them. Oöcysts of Eimeria auffenbergi n. sp. from L. smaragdina collected in the Philippines are ovoidal, with a smooth, colourless, bi-layered wall, measure 19.9 × 15.8 μm, L/W 1.3; both micropyle and oöcyst residuum are absent, but one to four polar granules are present. Sporocysts are ovoidal to ellipsoidal, 10.3 × 5.8 μm, L/W 1.8, and the wall is composed of two valves joined by a longitudinal suture; neither Stieda nor sub-Stieda bodies are present; a sporocyst residuum is composed of dispersed granules.  相似文献

This study investigated the biomass, lipid production, fatty acid content, and other nutrients present in microorganisms by using four culture methods: (1) photoautotrophic pure Chlorella vulgaris cultures (PP); (2) heterotrophic pure C. vulgaris cultures (PH); (3) mixed cultures of Rhodotorula glutinis and C. vulgaris under photoautotrophic conditions (MP); and (4) heterotrophic mixed cultures (MH). The microorganisms in MP culture showed the optimum growth condition and lipid production. Among the cultures, MP yielded the highest number of cells and biomass (5.9?×?105 cells/mL and 0.523 g/L, respectively). Furthermore, lipid production in MP culture was 114.22 mg/L, which is 136 % higher than that in MH culture (48.22 mg/L). Considering the higher contents of palmitic acid (C16:0) at 24.65 %, oleic acid (C18:1) at 56.34 %, and protein at 42.39 g/100 g in the MP culture than in other cultures, we proposed that MP could be used effectively to support the growth of microorganisms. This method could also be used as a potential approach for biodiesel production.  相似文献

A new coccidian species of the genus Caryospora Léger, 1904 (Protozoa, Apicomplexa, Eimeriidae) is reported from the colubrid snake host Philodryas olfersii Lichtenstein at a coastal area in the State of Rio de Janeiro, south-eastern Brazil. Oöcysts of Caryospora olfersii n. sp. are spherical to sub-spherical, 33.1 × 31.2 μm, with smooth, colourless, three-layered wall, c.1.4; middle layer lightly striated. Micropyle, oöcyst residuum and polar granule are all absent. Sporocysts are ovoid, 22.9 × 17.4 μm on average, with one extremity in the shape of a short neck. Stieda body present, 3.2 × 1.3 μm, sub-Stieda body present, homogeneous, larger than Stieda body, 4.5 × 1.7 μm. Sporozoites are inserted in a bulky sporocyst residuum.  相似文献

Isospora frontalis n. sp. oöcysts are spherical to sub-spherical, 27.9 × 26.9 μm, with a smooth, bi-layered wall c.1.4 μm thick. A micropyle and an oöcyst residuum are absent, but polar granules are present. Sporocysts are elongate ellipsoidal, 19.6 × 11.1 μm. The Stieda body is knob-like and the substieda body is slight. The sporocyst residuum is composed of scattered granules, and the sporozoites are vermiform with a refractile body and a nucleus. Isospora teresopoliensis n. sp. oöcysts are spherical to sub-spherical, 25.7 × 24.3 μm, with a smooth, bi-layered wall 1.3 μm thick. A micropyle, oöcyst residuum and polar granule are absent. Sporocysts are ovoid, 18.8 × 11.2 μm. The Stieda body is nipple-shaped and the substieda body is large and prominent. The sporocyst residuum is composed of scattered granules, and the sporozoites are large and elongate, with a refractile body and nucleus. Oöcysts of Isospora chanchaoi n. sp. are spherical to sub-spherical or ovoid, 24.2 × 22.0 μm, with a smooth, bi-layered wall c.1.2 μm thick. Both a micropyle and an oöcyst residuum are absent, but one or two polar granules are present. The sporocysts are ellipsoidal, 16.1 × 10.3 μm. The Stieda body is nipple-shaped and the substieda body is small but prominent. The sporocyst residuum forms a compact mass of granules, and the sporozoites are large and elongate, with a refractile body and a nucleus.  相似文献

Snow is not only abiotic component of cryosphere but also a true functional ecosystem. In the past were the Vysoké Tatry Mts, the highest mountains of the Carpathians, the most studied mountain area in the Europe in terms of cryoflora. The aim of this study was to investigate the occurrence of snow microorganisms and its environmental conditions also in other high mountains of Slovakia. Due to climatic conditions of high altitudes, snow patches persist in deep concave relief shapes until late of June. Therefore these conditions are suitable for snow microorganisms. Sampling of snow microorganisms was carried out from May to September of 2013 and 2014. In selected mountains were mostly found snow alga cf. Chloromonas nivalis and we found also the presence of individuals of cyanobacteria, microscopic fungi, ciliates, rotifer and tardigrade. Recorded occurrence of thaw–froze cycles during august and high intensity of photosynthetically active radiation (PAR) (max. 2133 μmol.m?2.s?1) could be considered as stress factors this harsh environment. The pH values of taken samples were slightly acidified, in some cases neutral. The conductivity of water from melted snow had high coefficient of variation (values characteristic for distilled water until values 160 μS.cm?1). The chemical analyses determined the higher content of N-NO3 in analysed samples of melted snow in compare with content of N-NH4 and P-PO4. Despite these facts paper confirms potential of Slovak high mountains as suitable place for life of snow microorganisms.  相似文献

ABSTRACT

Cave bears have disappeared from the Alps from different altitudes at different times. The temporal progression of the HDEL (Height Dependent Extinction Line) – a compilation of the geologically most recent radiocarbon dates per altitude level – is not consistent with the general cooling of the temperatures from about 45 ka BP. The cave bear sites of the Northern Alps with the most recent radiocarbon ages are not situated in the lowlands but in caves in altitudes of 1,500 m to 1,700 m above sea level (a.s.l.).

Cave bears fed almost exclusively on herbs and leaves. It was assumed that with the general cooling in the OIS 3 since about 45 ka BP also the migration of the alpine elements into the lowlands took place. It could be recognized that the populations in the lower situated cave bear site became earlier extinct than the cave bear population in the higher altitudes.

With new radiocarbon dates, done at the Curt-Engelhorn-Center Archaeometry at the Reiss-Engelhorn-Museen in Mannheim (Germany), the HDEL can be determined much more precisely and the causes of gradual extinction are also better understood.  相似文献

The effect of vertical physico-chemical stratification on the planktonic microbial community composition of the deep, hypersaline and heliothermal Lake Ursu (Sovata, Romania) was examined in this study. On site and laboratory measurements were performed to determine the physical and chemical variables of the lake water, and culture-based and cultivation-independent techniques were applied to identify the members of microbial communities. The surface of the lake was characterized by a low salinity water layer while the deepest region was extremely saline (up to 300 g/L salinity). Many parameters (e.g. photosynthetically active radiation, dissolved oxygen concentration, pH, redox potential) changed dramatically from 2 to 4 m below the water surface in conjunction with the increasing salinity values. The water temperature reached a maximum at this depth. At around 3 m depth, there was a water layer with high (bacterio) chlorophyll content dominated by Prosthecochloris vibrioformis, a phototrophic green sulfur bacterium. Characteristic microbial communities with various prokaryotic taxa were identified along the different environmental parameters present in the different water layers. Some of these bacteria were known to be heterotrophic and therefore may be involved in the decomposition of lake organic material (e.g. Halomonas, Idiomarina and Pseudoalteromonas) while others in the transformation of sulfur compounds (e.g. Prosthecochloris). Eukaryotic microorganisms identified by molecular methods in the lake water belonged to genera of green algae (Mantionella and Picochlorum), and were restricted mainly to the upper layers.  相似文献

The antimicrobial effect of OKCEL® H-D, a topical, absorbable hemostatic textile prepared from oxidized cellulose, was tested. Testing by dilution and diffusion methods was conducted on a spectrum of 27 select microorganisms, including also antibiotic-resistant strains. OKCEL® H-D showed inhibitory effects on nearly all tested bacteria. In testing using the dilution suspension method, the majority of bacteria showed decrease in cell density by 7–8 orders of magnitude after just 6 h of exposure. For clinical isolates of antibiotic-resistant strains, a reduction occurred after 24 h of exposure. In testing the antimicrobial effects of OKCEL® H-D by the dilution method was least effective on spore-forming Bacillus subtilis, for which no antimicrobial effect was detected after 48 h, and on Mycobacterium smegmatis, for which the number of cells decreased by four orders of magnitude only after 24 h. By the diffusion method, inhibition zones were recorded for nearly all test microorganisms except for Staphylococcus aureus, M. smegmatis, and Listeria monocytogenes. No growth beneath the tested OKCEL® H-D material was recorded, however, even for the latter-named bacteria strains, which attests to its good inhibitory effect.  相似文献

Isospora canaria Box, 1975 (Protozoa, Apicomplexa, Eimeriidae) is reported and described from canaries Serinus canaria (Linnaeus) in southeast Brazil. Its oöcysts are subspheroidal to ellipsoidal, 24.4 × 22.2 μm, with smooth, bilayered wall, ~1.2 μm. The micropyle and the oöcyst residuum are absent, but a polar granule is present. The sporocysts are lemon-shaped, 17.6 × 10.6 μm. The Stieda body is nipple-like, and substieda body is prominent and homogeneous. The sporocyst residuum is composed of scattered granules. The sporozoite has anterior and posterior refractile bodies and a nucleus. The report of this coccidium recovered from exotic canaries in South America is relevant to native passerines, mainly to Carduelis yarrellii, which are listed as vulnerable species by IUCN (International Union for Conservation of Nature and Natural Resources).  相似文献

Comparisons of the behavior and ecology of primates living in intact and fragmented forest are critical to the development of conservation strategies for the many primate taxa threatened by habitat loss. From July 2009 to April 2010, we investigated the habitat use, ranging behavior, and diet of two groups of Boutourlini’s blue monkeys (Cercopithecus mitis boutourlinii), a subspecies endemic to western Ethiopia, whose habitats had experienced different levels of disturbance at Jibat Forest. Forest Group occupied primarily continuous tree-dominated forest with little human disturbance whereas Fragment Group inhabited a heavily degraded 2- to 3-km2 forest fragment nearly surrounded by farmland and connected tenuously to the continuous forest by a narrow corridor of riverine forest. Mean daily path lengths for both groups were nearly identical (Forest Group: 799 m; Fragment Group: 783 m) and exhibited little seasonal variability. The mean home range areas of Forest Group and Fragment Group were 72.0 and 61.2 ha, respectively. Forest Group (N?=?2232 feeding records) fed mostly on fruits (52.5 %), though they also ate animal prey (14.7 %), young leaves (11.1 %), shoots (8.7 %), and flowers (7.3 %). In contrast, fruits accounted for only 17.0 % of Fragment Group’s diet (N?=?2903 feeding records), with shoots (29.8 %), young leaves (17.1 %), animal prey (13.1 %), seeds (9.6 %), and flowers (6.8 %) also making substantial contributions to their diet. Only Fragment Group engaged in crop raiding, consuming seeds from barley and wheat extensively (33–41 % of diet) during 2 mo. Fragment Group (N?=?33) ate more plant species than Forest Group (N?=?24), though both groups exploited a small number of plant species relative to other subspecies of blue monkeys. Our study revealed that, like most other blue monkey subspecies, Boutourlini’s blue monkeys are quite flexible in the habitats they occupy as well as in their foraging habits. Despite this ecological flexibility, the long-term conservation of Boutourlini’s blue monkey is far from assured given its limited distribution, the rapidly growing human population, and the high rates of forest clearance in western Ethiopia.  相似文献

Bioaerosol particles including bacteria, fungi, and virus are originated from marine and terrestrial environments. The airborne microorganisms are transported for long distance through the free troposphere and are thought to influence the downwind ecosystems and human life. However, microbial communities in the free troposphere have not been understood in detail because the direct sampling of microbial cells at high altitude requires sophisticated sampling techniques. In this study, for the investigation of microbial species compositions in the free troposphere, air sampling using an aircraft was performed over the Noto Peninsula in Japan, where the tropospheric winds carry aerosol particles from continental areas. Two air samples were collected at 3,000 m on March 27, 2010, when air mass was carried from the Gobi Desert to Japan area. Microorganisms from one air sample grew in culture media containing up to 15 % NaCl, suggesting that halotolerant bacteria maintain their viabilities in the free troposphere. DGGE analysis revealed that the amended cultures were dominated by Bacillus subtilis, and the isolates obtained from the amended cultures were identical to B. subtilis. Furthermore, the 16S rDNA clone library (culture-independent survey) of the other air sample grew was composed of three phylotypes belonging to Firmicutes, Bacteroidetes, and Proteobacteria with the sequences of Firmicutes phylotype corresponding to that of the cultured B. subtilis sequence. Microscopic observation using FISH method indicated that B. subtilis particles occupied 80 % of total eubacterial particles on the mineral particles. The halotolerant bacteria identical to B. subtilis would dominate at high altitudes over Noto Peninsula where the prevailing westerly wind was blowing.  相似文献
