Eggplants (Solanum melongena L. cv. Bonica) were grown in a glasshouse during summer under natural light with one unbranched shoot or one shoot with 3 to 4 branches and with or without fruit in quartz sand buffered and not buffered with 0.5% CaCO3 (w : v), respectively. Nutrient solutions supplied contained nitrate or ammonium as the sole nitrogen source. Compared with nutrient solutions containing nitrate (10 mM), solutions containing ammonium (10 mM) caused a decrease in net photosynthesis of eggplants during early stages of vegetative growth when grown in quartz sand not buffered with CaCO3. The decrease was not observed before leaves showed interveinal chlorosis. In contrast, net photosynthesis after bloom at first increased more rapidly in eggplants supplied with ammonium than with nitrate nitrogen. However, even in this case, net photosynthesis decreased four weeks later when ammonium nutrition was continued. The decrease was accompanied by epinasty and interveinal chlorosis on the lower leaves and later by severe wilting, leaf drop, stem lesions, and hampered growth of stems, roots, and fruits. These symptoms appeared later on plants not bearing fruits than on plants bearing fruits. If nutrient solutions containing increasing concentrations of ammonium (0.5–30 mM) were supplied after the time of first fruit ripening, shoot growth and set of later flowers and fruits were promoted. In contrast, vegetative growth and reproduction was only slightly affected by increasing the concentration of nitrate in the nutrient solutions. In quartz sand buffered with CaCO3 ammonium nutrition caused deleterious effects only under low light conditions (shade) and on young plants during rapid fruit growth. If eggplants were supplied with ammonium nitrogen before bloom, vegetative growth was promoted, and set of flowers and fruit occurred earlier than on plants supplied with nitrate. Furthermore, the number of flowers and fruit yield increased. These effects of ammonium nutrition were more pronounced when plants were grown with branched shoots than with unbranched shoots. The results indicate that vegetative and reproductive growth of eggplants may be manipulated without causing injury to the plants by supplying ammonium nitrogen as long as the age of the plants, carbohydrate reserves of the roots, quantity of ammonium nitrogen supplied, and pH of the growth medium are favourable. Kinetics of cell death and the production of dissolved organic carbon (DOC) were investigated in Anabaena flos-aquae (Lyngb.) Bréb grown on three different N sources (N2nitrate, and ammonium) in a phosphorus (P)-limited chemostat. The fraction of live cells in the total population increased as growth rate increased with decreasing P limitation. Cell death was less in nitrate and ammonium media than in N2. The specific death rate (γ), when calculated as the slope ofv?1x vs. D?1, where vxand D are live cell fraction (or cell viability) and dilution rate, respectively, was 0. 0082 day?1 in N2and 0.0042 day?1 in nitrate. The slope of the plot in ammonium culture was not significant; however, the value of the live cell fraction was within the range for the NO?3culture. The fraction of live vegetative cells in N2 culture was constant at all growth rates and the increase in the overall live cell fraction with growth rate was due entirely to an increase in live heterocysts. Live heterocysts comprised 3.5% of the total cells at a growth rate of 0.25 day?1 and increased to 6.3% at 0.75 day?1 with the ratio of live heterocysts to live vegetative cells linearly increasing with growth rate. The fraction of live vegetative cells was invariant in nitrate cultures us in N2cultures. The live heterocysts fraction also increased with growth rate in nitrate cultures, along with the live heterocysts : live vegetative cells ratio, but the level was lower than in N2cultures. DOC released from dead cells increased inversely with growth rate in N2from 36.4% of the total DOC at a growth rate of 0.75 day?1 to 54.15% at 0.25 day?1. The contribution of cell death to the total DOC production in nitrate and ammonium media was significantly less than that under N2DOC from dead cells consisted mainly of high-molecular-weight compounds, whereas DOC excreted from live cells was largely of low molecular weight.  相似文献

The seasonal cycle of biomass and tissue composition of Ulva rigida C. Agardh, in relation to nitrogen availability in the water column, was studied in 1991-1992 in the Sacca di Goro, a highly eutrophic lagoon in the Po River Delta (Italy). Nitrate uptake rates and storage capacity were also determined in laboratory experiments. The seasonal growth of U. rigida was related to the seasonal trend of nitrogen concentration in the water column. U. rigida biomass increased exponentially during spring and attained peaks of about 300-400 g dry mass (DM) m−2 in June. As biomass increased, U. rigida depleted nitrate in the water column. Thallus nitrate reserves also declined from 100 μmol N (g DM)−1 to almost undetectable levels, and total thallus nitrogen declined from 4% to 2.5% DM and 1.25% DM in 1991 and 1992, respectively. During summer, U. rigida decomposition increased, and organic nitrogen concentrations in the water column increased. The uptake experiments demonstrated an inverse relationship between thallus nitrate content and nitrate uptake rates. A modified Michaelis-Menten equation that accounts for thallus nitrate fit the uptake data well. U. rigida can accumulate up to about 400-500 μmol nitrate (g DM)−1 in cellular reserves. U. rigida in the Sacca di Goro has higher Km and lower Vmax/Km ratios for nitrate uptake than other chlorophycean species, indicating a low efficiency of uptake at low nitrate concentrations. This low uptake efficiency, and the ability to exploit N availability by storing cellular nitrate pools in excess of immediate growth needs, may represent a physiological response to an eutrophic environment where nitrate is in large supply for most of the year.  相似文献

The long-day flowering of Lemna perpusilla 6746 on an SH inhibitor-containingmedium was inhibited by the application of ammonium ion to themedium. Ammonium ion not only suppressed long-day flowering,but relieved the inhibition of vegetative growth caused by theinhibitors. Nitrite, casamino acids, glutamine and asparaginehad a similar effect, suggesting that the inhibition of long-dayflowering by ammonium ion is not a direct effect of the ion.Most amino acids, with the exception of glutamate and aspartate,also prevented long-day flowering, but their effects on vegetativegrowth varied. No qualitative differences in amino acid compositionwere observed among plants cultured on media containing nitrate,nitrite or NH44NO3as the sole nitrogen source. However, theamounts of free and total amino acids werehigher in plants fedwith nitrite or NH4NO3 than in those fed with nitrate. Thissuggests that the inhibition of long-day flowering by ammoniumand nitrite can be ascribed to increased nitrogen metabolism. Though decreased activity by SH inhibitors of nitrate reductase(SH enzyme) is assumed to result in long-day flowering by loweringthe nitrogen metabolism, lowering the nitrogen level in M mediumdid not bring about floral initiation in the absence of SH inhibitors. The contents of gibberellic acid (GA3), glucose and starch, and dry mass in four nodes of apical parts of male and female thalli of Chara tomentosa were measured. The GA3 concentration was about 4 times higher in male than in female thallus and about 10 times higher in the first node than in the next three nodes of both thalli. The apical part of male thallus contains 2.5 times less glucose and 1.7 times less starch than female one. The average amount of dry mass of male and female thallus was similar: 9.9 and 9.8 % of fresh mass. The findings of this investigation demonstrate the interdependence of gibberellic acid and sugars in development of male and female Chara tomentosa thalli.  相似文献

Utilising the 15N dilution technique the relationship between the proportion of N derived from N2 fixation and relative abundance of ureides in xylem sap was evaluated for Phaseolus vulgaris L. cv. Mokcham during vegetative and reproductive development. In order to establish calibration curves for time integrated estimates of N2 fixation, plants were raised in sand culture during the dry season in northern Thailand and continuously supplied with a N-free nutrient solution or the same solution amended with 0, 3, 6 or 9 mol m–3 nitrate. Large changes in plant dependence on N2 fixation were concomitantly reflected by corresponding alterations in N solutes in xylem sap. Regression analyses of the data suggested high correlations between relative ureide content and N2 fixation, but different slopes and line intercepts indicated the requirement for the use of calibration curves established for different phases of the development of the plant. Largest age related differences were noted between vegetative and reproductive development. Judging from 95% confidence limits, utilisation of appropriate calibrations can reduce errors of the technique to close to ±5%.A second experiment, involving similarly cultivated plants exposed to different sources of mineral N, indicated an effect of ammonium on xylem sap composition. This implies that calibrations, in which N2 fixation is regulated only by applications of various concentrations of nitrate, may lead to errors in situations where a major proportion of the plant available soil nitrogen fraction is present in the form of ammonium.  相似文献

Nitrate N is a major inhibitor of the soybean/Bradyrhizobium symbiosis in legumes and although this inhibition has been studied for many years, as yet no consensus has been reached on the specific and quantitative interactions between nitrate and ammonium supply and N2 fixation. The effect of nitrate and ammonium supply on plant growth, nodulation and N2 fixation capacity during the full growth cycle was investigated in both greenhouse and growth chamber experiments with three soybean genotypes. The results show that a high concentration of mineral N (10 mM), either as nitrate or ammonium or ammonium nitrate significantly suppressed nodule number, nodule dry weight and total N2 fixed per plant of nodulated soybeans. However, lower mineral N concentrations, either 1 mM or 3.75 mM significantly enhanced nodule number, nodule dry weight and total N2 fixed per plant, while specific nodulation (nodule dry weight g–1 root DW, SNOD) and specific N2 fixation (total N2 fixed g–1 root DW, SNF) were significantly reduced, particularly at the early vegetative growth stage V4, compared to the treatment with N2 fixation as the only N source, in both growth chamber and greenhouse experiments. Therefore, we suggest that SNOD or SNF might be better indicators to express the suppressing effect of mineral N addition on nodule performance and N2 fixed. Our studies also showed that ammonium alone was the more efficient N source than either ammonium nitrate or nitrate for soybean, as it resulted in higher biomass accumulation, nodule dry weight, total N accumulation and total N2 fixed by 23, 20, 18 and 44%, respectively, compared to NO3 as the N source.  相似文献

Tomato plants (Lycopersicon esculentum Mill. cv. Counter) were grown in 12-L polyethylene containers in aerated and CaCO3-buffered nutrient solutions containing different concentrations of complete stock solutions with either nitrate (stock solution N) or ammonium (stock solution A) as the only nitrogen source (X1 = standard concentration with 5 mM NO3 -N or NH4 +-N, and X3, X5.5, X8 and X11 = 3, 5.5, 8, 11 times the standard concentration), or a mixture of both stock solutions (N:A ratio = 100:0, 75:25, 50:50, 25:75, 0:100) at moderate nutrient concentration (X3). Total dry matter production and fruit dry weight were only slightly affected by increasing nutrient concentration if nitrate was supplied as the sole nitrogen source. Compared to nitrate, ammonium nitrogen caused a decrease in total dry weight (32–86% between X1 and X11), but led to an increase in both total dry weight and fruit dry weight (11% and 30%) at low concentration if supplied in addition to nitrate nitrogen (N:A ratio = 75:25). Dry matter partitioning in plants was affected by the strength of the nutrient solution, but even more by ammonium nitrogen. Fruits accumulated relatively less dry matter than did the vegetative parts of tomato plants when supplied with nutrient solutions containing ammonium as the only nitrogen source (fruit dry weight to total dry weight ratio 0.37 and 0.15 at low and high nutrient concentration), while nitrate nitrogen rather supported an increase in dry matter accumulation in the reproductive organs (fruit dry weight to total dry weight ratio 0.39–0.46). The water use efficiency (WUE) was only slightly affected by the strength of the nutrient solutions containing nitrate nitrogen (2.9–3.4 g DW (kg H2O)–1), while ammonium nitrogen led to a decrease in WUE from 2.4 to 1.3 g DW (kg H2O)–1at low (X1) and high (X11) nutrient concentration, respectively. The proline content of leaves fluctuated (0.1–5.0 mol (g fresh weight)–1) according to nutrient concentration and global radiation, and reflected enhanced sensitivity of plants to these potential stress factors if ammonium was the predominant N source supplied. It was concluded, that proline is a reliable indicator of the environmental stress imposed on hydroponically grown tomato plants.  相似文献

The life history of Coleochaete scutata Bréb. was analyzed by Feulgen microspectrophotometry, a technique measuring DNA content in individual nuclei. By correlating nuclear DNA content with morphological structures or stages in the life history, changes in ploidy level are revealed. The microspectrophotometric study confirmed the earlier reports of a haploid vegetative thallus with mitotic division restricted primarily to the margin of the thallus. In the mitotic cycle the G1 (pre-synthesis) phase is longer in duration than the synthesis find G2 (post-synthesis) phases. Oogamous sexual reproduction results in resistant oospores which attain DNA levels of 2C 8C (1C being the DNA level of gamete nuclei).  相似文献

Nitrate, ammonia, urea, and glycine were compared as nitrogen sources for Acetabularia mediterranea. Cells grew normally in media containing nitrate or urea, while cells did not grow at all when the same amount of N was supplied as ammonium ion. The utilization of glycine remains questionable. Cells in medium without added N (NDM) increased in length and some formed reproductive caps. The whorls of vegetative cells showed considerable hypertrophy in NDM and in glycine. This hypertrophy was due to the elongation of only the first-(a1) and second- (a2) order articles. When cut, the basal portion of cells without added N regenerated new apices with whorls. The development of these whorls was inversely proportional to the NO2 concentration. Analyses showed that the intracellular nitrogen pool in young cells and regenerating bases was very small, about 1/10 of that of fully grown cells. Therefore we suggest that trace amounts of N contaminants in the medium supported growth and development, the uptake of which was facilitated by the hypertrophied whorls, under N-limited conditions.  相似文献

A vegetative clone ofUlva lactuca L. was selected for mass culture and nutrient uptake experiments with fish pond wastewater. Growth rates of over 55 g dry wt. d?1 per 6001(1 m2) tank were obtained. Growth rate was linked to stocking density, tank flushing rates and aeration induced thallus movement. The plants could not survive on the macronutrients provided by a weekly pulse of wastewater. A continuous supply of fish pond wastewater was required to maintain good growth rates. An ‘uncoupling’ of growth rate and thallus nitrogen content was observed. The plants were able to store nitrogen from a pulsed ammonium supply and allot the nitrogen reserves to new tissue growth. Plants with slower growth rates or a continuous supply of ammonium had higher thallus nitrogen content.Ulva efficiently removed up to 85% of the ammonium from fish pond wastewater in darkness or light independently of temperature fluctuations.  相似文献

Various levels of protection against x-irradiation damage in bacteriophage T1 may be obtained by the addition of inorganic salts to the aqueous virus suspensions during irradiation. The highest survival values are obtained with the nitrite salts, and their protective power is attributed primarily to their function as reducing agents. The nitrate ion shows greater protection than the corresponding sulfate or chloride ions. This may be due in part to the lower energy level of the nitrate ion, by reason of resonance. Since greater expenditure of incident energy is required to raise the ion from the ground state, the energy thus dissipated may be ineffective in the inactivation of virus particles. The ammonium salts exhibit protection of a different order of magnitude from that of the metallic salts. It is postulated that NH4+ protects in a threefold way: (a) dehydration, (b) reduction, in which the ammonia is oxidized to nitrite and the nitrite to nitrate, and (c) stabilization of the virus protein. Metallic salts likewise protect, but a point of maximum protection is reached in lower concentrations than in the case of the ammonium salts. After this maximum protection is reached, there is a rapid decline in survival with increased concentration. This prevents protection of the order of magnitude that can be obtained with the ammonium salts. It is postulated that a specific cationic interaction with the phage may be responsible for the decreased protection. Bacteriophage is protected during x-irradiation by an alkaline pH, in the case of NH4OH. This protection could not be produced with NaOH, presumably because of the greater hydrolysis of the protein components of the virus particle in solutions of NaOH, whereas NH4OH stabilizes the protein.  相似文献

Abstract Effect of ammonium on in vivo activity of nitrate reductase in roots, shoots and leaves of maize (Zea mays L.) seedlings was studied in relation to light/dark conditions and EDTA supply. Supply of 5 mM (NH4)2SO4 increased the steady state level of enzyme only in leaves and in light, while it had no effect in roots and shoots and in the dark. The substrate induction of enzyme was also little affected by 1 to 10 mM (NH4)2SO4 in roots and shoots. In the leaves the activity in the dark was either inhibited (minus EDTA) or stimulated (plus EDTA) by 5 to 10 mM (NH4)2SO4. The activity was stimulated in the light also in the presence of EDTA at higher concentrations of ammonium. When different concentrations of ammonium were supplied without any exogenous nitrate in the light, the enzyme activity increased at low concentration and was either inhibited or unaffected at higher concentrations depending upon the tissue used. Supply of EDTA with ammonium modified its effect to some extent. It is suggested that the effect of ammonium on nitrate reductase activity depends upon the tissue used and the effective concentration of the ammonium.  相似文献

The effect of some ammonium salts on nitrate reductase (NR) level, onin vivo nitrate reduction and on nitrate content was followed in the presence of nitrate in the medium, under changing experimental conditions, in excisedPisum sativum roots, and their effect was compared with that of KNO3, Ca(NO3)2 and NaNO3 at 15 mM NO3 - concentration, i.e. at a concentration which considerably exceeded the level of saturation with nitrate with respect to nitrate reductase. The effect of ammonium salts on NR level is indirect and changes from a positive one to a strongly negative one which is dependent on the time of action of the salt, on the presence of other cations, on pH of the solution of the ammonium salt and on the nature of the anion of the ammonium salt. A positive effect on the enzyme level can be observed in the presence of other cations than NH4 + at suitable concentrations of those ammonium salts, the solutions of which have their pH values in the acid region (i.e. NH4H2PO4, (NH4)2SO4 and NH4NO3). However their positive effect is independent of the presence of NH4 + ions, and it is obviously the result of an increased concentration of H+ ions. A clear-cut negative effect on NR level can be observed after 24 h in one-salt NH4NO3 solution where NH4 + is not balanced with other cations and thus certainly can adversely influence many metabolic processes, and in the solutions containing neutral (pH 6.2) and dibasic ammonium phosphates in which dissolved undissociated ammonia [(NH3). (H2O) which can also affect many metabolic processes incl. proteosynthesis] probably has a toxic influence. Thein vivo nitrate reduction is always depressed in excised pea roots in the presence of ammonium salts in the medium, regardless of the level of nitrate reductase. Under the described conditions, no relationship could be established between the enzyme level and the so-called metabolic NO3 - pool (i.e. NO2 - production under anaerobic conditions), nor between NR level and the total nitrate content in the roots. One-salt solutions of NaNO3, Ca(NO3)2 and KNO3 exert different effects on the level of nitrate reductase and on the content of NO3 - in the roots, but the in vivo NO3 - reduction shows the same trend as NR level in the roots influenced by these salts. Cl- ions, supplied in NH4C1, depress both NR level and NO3 - content in the roots at higher concentrations, but they do not significantly affect the in vivo nitrate reduction in comparison with other ammonium salts. These results indicate that NR level,in vivo nitrate reduction, and nitrate uptake can be regulated in pea roots independently of each other.  相似文献

The potential methane consumption activity was examined in various plant communities of a boreal Sphagnum-dominated Bakchar bog of West Siberia. In aerobic laboratory incubations, the peat consumed methane with the maximal rates varied from 17 to 153 nmol CH4 h–1g– 1.The highest oxidation took place in the peat from the cotton grass and dwarf shrub-cotton grass communities. The addition of different N-compounds inhibited CH4-uptake and was not a simple influence of shift in ionic balance (`salt effect'). The introduction of sodium chloride resulted in significantly weaker inhibition effect than the same amount of nitrite and nitrate salts. The inhibition occurred at NH4 +-N concentrations exceeding 100 mg kg–1, which was more than 200 times higher native N-content in peat. Communities with high CH4-uptake activity were more sensitive to ammonium. The inhibition by ammonium was non-competitive. The inhibition by ammonium was mainly due to the toxic action of nitrite and/or nitrate produced by nitrifiers. A strong positive correlation was found between the potential nitrifying activity and inhibition of CH4-uptake in ammonium-treated peat (R 2= 0.87). The oxidized N-compounds were more strong inhibitors than ammonium and their toxicity increased in the following range: NH4 +< NO2 < NO3 .  相似文献

Rates of exchanges of nitrate and ammonium across the sediment-water interface were measured in an inshore marine environment at Signy Island, South Orkney Islands, Antarctica, over 6 months from August 1991 to February 1992. The sediment was a source of ammonium to the water column but a sink of nitrate, although nitrate exchange rates were very variable. Concentration profiles of nitrate and ammonium in the sediment porewater corroborated the measured vertical exchanges. Bioturbation, by a largely amphipod benthic infauna which was confined to the top 2 cm of sediment, was investigated experimentally. Removal of bioturbation depressed sedimentary O2 uptake by 33% and sedimentary release of NH4 + by 50%. In contrast, in the absence of bioturbation, the removal of NO3 from the water column by the sediment increased in rate. The measured fluxes of ammonium and nitrate from the sediment did not match with the amount of nitrogen mineralised within the sediment, and urea may account for the difference. It is suggested that the export of nitrogen from the bottom sediment may be significant in sustaining primary production in the Antarctic inshore environment. Ammonium and urea are preferred to nitrate as a nitrogen source by phytoplankton. The nitrate concentrations in the sediment porewater were low, but a large pool of nitrate was identified in the top 0–2 cm layer, which was released by KCl extraction or by freezing of the sediment. This extractable pool of nitrate did not equilibrate with the soluble nitrate pool in the sediment, but seemed to be released from components of the benthic infauna, which were also largely confined to the top 0–2 cm. The physiological role of this nitrate is unknown.  相似文献

With a view to consider the increasing concern over nitrogen pollution in the aquatic environment, we investigated effects of nitrate (NO3) and nitrite (NO2) on the activity of dopaminergic neuron in zebrafish embryos and larvae. Both nitrate and nitrite exposure decreased the expression of tyrosine hydroxylase (TH) in dopaminergic neurons at 48 hpf. Only nitrite decreased the response to tactile stimulation at 72 hpf, whereas both nitrate and nitrite decreased the swimming activity at 6 dpf. When the embryos were exposed to nitrate or nitrite together with an estrogen receptor blocker (ICI 182,780), the decreases in TH expression and motor behavior caused by nitrate or nitrite alone were reversed suggesting the effects of nitrate and nitrite were mediated through estrogen receptor (ER). The result of co-incubation with an oxidoreductase inhibitor, diphenyleneiodonium, indicated the conversion to nitric oxide (NO) is likely to be responsible for the effects of nitrate and nitrite, which was further supported by the increased staining for NO after exposure. The present study demonstrates that nitrate and nitrite are neurotoxicants acting as an endocrine disruptor possibly through conversion to NO to downregulate the activity of dopaminergic neuron in early development of zebrafish.  相似文献
