Antifertility effects of busulfan were evaluated using adult coyotes. In addition, antifertility effects of PMHI were evaluated in adult males. Adult males and females were alloted randomly to the following treatments: (1) untreated control, (2) a single oral dose of 3 mg busulfan/kg of body weight (BW) or (3) two oral doses of 3 mg busulfan/kg BW given nine days apart. The untreated males were used as controls in both experiments. Additional male coyotes were allotted randomly to PMHI treatments as follows: (1) a single oral dose of 2 mg PMHI/kg BW, or (2) two oral doses of 1 mg PMHI/kg BW given seven days apart. Blood samples were taken from females and serum analyzed for progesterone by radioimmunoassay. Males were hemicastrated (left testicle) 30 days after onset of treatment. The right testicle was removed 30 days later. Testes and epididymides were fixed in 10% buffered formalin and prepared for histologic examination. For females developing corpus luteum (CL), the maximum peak progesterone concentration for those given two doses of busulfan was less (P<0.05) than that for untreated controls and single-dose females (16.9, 22.4 and 26.3 ng/ml, respectively). The double treatment of busulfan prevented more females (4 of 10) from developing CL (P<0.08) than controls (0 of 7) or single-dose females (1 of 9). None of the busulfan-treated male coyotes had histologic evidence of spermatogenesis 60 days after the onset of treatment. The oral dose or doses of PMHI did not result in complete degeneration of seminiferous tubules. Busulfan given orally did not cause any adverse reactions, but orally administered PMHI often induced vomiting within 18 min after treatment. We conclude that busulfan is capable of affecting male and female reproductive parameters, but PMHI appears to have little effect on spermatogenesis in coyotes when given orally in a single- or a double-dose. 相似文献

Conventionally housed 130-160 g adult male Japanese quail were given gossypol acetic acid (gossypol) im at 25 mg/kg in 0.5 ml of 10% EtOH for 12 and 24 days (Groups 1 and 2), respectively. One day after treatment was terminated they were allowed to mate with laying females individually for 20 days. Fertility was 0% from mating of the Group 1 birds on days 1-2 and increased to 25, 35, 55 and 65% on days 3-6 after cessation of gossypol treatment. At day 11, fertility was 80 vs 84% in controls, whereas hatchability was 70% for both. By comparison, eggs from Group 2 mated quail were infertile for up to 20 days after the termination of gossypol treatment. In a parallel experiment, the percent testes to body weight ratio in control and 7, 14, 21, and 28-day gossypol-treated quail was 2.5, 2.2, 1.8, 0.5, and 0.2%, respectively. In 12 vs 24-day treated birds, 7, 14, 21, and 28 days after gossypol treatment, the ratios were 1.0 vs 0.5%, 2.0 vs 0.8% and 2.8 vs 1.9%, respectively. The decreased fertility and hatachability, and testicular atrophy resulting from gossypol given to male quail was dose-time related. Furthermore, the androgen-dependent cloacal gland was drastically reduced in size by the treatment with gossypol. The mode of action of gossypol in male quail is different than it is in mammals in that the testicular size of mammals remains unchanged with long-term gossypol treatment. Dry leaf powder of A. paniculata, when fed orally to male albino rats, at a dose level of 20 mg powder per day for 60 days, resulted in cessation of spermatogenesis, degenerative changes in the seminiferous tubules, regression of Leydig cells and regressive and/or degenerative changes in the epididymis, seminal vesicle, ventral prostate and coagulating gland. There was reduction in the weight and fluid content of the accessory glands. The treatment also resulted in accumulation of glycogen and cholesterol in the testis, and increased activities of lactate dehydrogenase in testis and alkaline phosphatase in testis and ventral prostate. The results suggest antispermatogenic and/or antiandrogenic effect of the plant. 相似文献

Oral administration of 70% methanolic extract of T. cordifolia stem to male rats at the dose level of 100 mg/rat/day for 60 days did not cause body weight loss but decreased the weight of testes, epididymis, seminal vesicle and ventral prostate in a significant manner. Sperm motility as well as sperm density were reduced significantly which resulted in reduction of male fertility by 100%. The stem extract brought about an interference with spermatogenesis. The round spermatids were decreased by 73.12%. However, the population of preleptotene and pachytene spermatocytes were decreased by 47.60% and 52.85% respectively, followed by secondary spermatocytes (48.10%). Leydig cell nuclear area and mature Leydig cell numbers were significantly reduced when compared with controls. Serum testosterone levels showed significant reduction after Tinospora extract feeding. Seminiferous tubule diameter, Leydig cell nuclear area as well as cross sectional surface area of Sertoli cells were reduced significantly when compared to controls. Biochemical parameters i.e. protein, sialic acid, glycogen contents of testes decreased significantly. Seminal vesicular fructose also depleted whereas, testicular cholesterol was elevated significantly followed by a reduction in testosterone levels. Gossypol acetic acid was administered orally (30, 60, 90 and 120 mg/kg/day) on Days 1-5 post coitum to mature female rats. At autopsy on Day 10, pregnancy in most treated animals (6/7 and 6/8) was blocked at high doses (90 and 120 mg/kg/day respectively). As the daily dose decreased to 60 mg/kg/day half (4/8) were not pregnant. However, at a lower dose (30 mg/kg/day), or at a single dose of 200 mg/kg at Day 1 p.c., pregnancy was not blocked. The concentrations of progesterone in the serum of these females were significantly decreased except at the low dose. The numbers of implantation sites in the treated females that did remain pregnant were similar to those in control females except at the dose of 120 mg/kg/day. Gossypol did not retard the development of the preimplantation embryo or cavitation. The Pontamine Blue test revealed that the drug did not interfere with the initiation of implantation. We suggest that gossypol has an antifertility effect in the female rat because it is luteolytic and disrupts post-implantation development. 相似文献

In the present study, the effect of oral administration of Melia azedarach Linn. (dharek) seed extract on fertility index, uterine weight and various histological and biochemical parameters of uterus were studied in the adult cyclic Wistar rats. Average number of embryos and implantation losses in the pregnant animals treated with dharek seed extract was also studied. The extract was prepared using a flash evaporator at 35 degrees C and dissolved in olive oil to prepare doses on per kg body weight basis. The results indicated a reduction in fertility index and average number of embryos in mated rats treated with the dharek extract. Pre-implantation, post-implantation and total prenatal mortalities were increased in rats treated with dharek seed extract during early (D1-D7) and late (D7-D18) stages of gestation period at doses of 5, 10 and 20 mg kg(-1) body wt day(-1). Histological studies showed a significant reduction in myometrial thickness, uterine gland diameter, luminal diameter of uterine glands and luminal epithelial cell height in rats treated with dharek seed extract at 1mg kg(-1) body wt day(-1) for 18 days. Pits and folds in luminal epithelial, mitotic activity in luminal and glandular epithelial cells of uterus were observed to be absent. Biochemically, a significant increase in protein and glycogen contents was observed. Thus, in conclusion, the application of this plant extract in rodent control programme may help to elevate the socioeconomic status of the society. 相似文献

The antifertility activity and toxicity of alpha-chlorohydrin and seven aromatic ketal derivatives were investigated in male rats. At a dose of 5 mg/kg injected intraperitoneally each day for 14 days, alpha-chlorohydrin and the methoxy benzaldehyde derivative (compound 2) produced complete infertility. The benzaldehyde derivative (compound 1) was 89% effective and the other five compounds 71-25% effective. All compounds except the least effective antifertility agent, the methylbenzaldehyde derivative (compound 3), reduced the motility of sperm recovered from the epididymis. None of the compounds caused a decrease in body or testes weight but some increased adrenal weight. 相似文献

We demonstrated that the decreased response to acetylcholine observed in aorta of male and female spontaneously hypertensive rats is corrected after sustained (15 days) reduction of blood pressure levels by losartan. In order to verify if the same occurs in resistance vessels, vascular diameter changes induced by topical application of acetylcholine and bradykinin (endothelium-dependent vasodilators) and sodium nitroprusside (endothelium-independent vasodilator) to mesenteric arterioles studied in vivo, in situ were determined in rats treated with losartan for 24 h (acute) or 15 days (chronic). Rats that presented similar reduction (in %) of the blood pressure levels after losartan treatment were chosen. Sodium nitroprusside induced similar responses in losartan-treated and untreated male or female SHR. Whereas in female SHR, losartan corrected the diminished arteriolar response to endothelium-dependent vasodilators after acute and chronic treatment, in male SHR this correction only occurred after chronic treatment. Thus, losartan corrected the endothelial dysfunction more easily in female than in male SHR and independently of the normalization or the magnitude of the reduction of the blood pressure levels. In an attempt to explain the difference, we evaluated the losartan effect on nitric-oxide synthase (NOS) activity and angiotensin II AT1 and AT2 receptor gene expression in these animals. In male and female SHR, NOS activity and AT1 receptor expression were not altered by acute or chronic treatment. On the other hand, AT2 receptor expression was augmented only in female SHR by these treatments. Therefore, augmented AT2 receptor expression, but not alteration of NOS activity or AT1 receptor expression, might explain the difference observed. 相似文献

Antifertility effect of sympathomimetic drugs on male rats when applied locally to the vas deferens.

Local application of collars containing 25% methoxamine, 50% or 75% tyramine or 50% norephedrine to both vasa deferentia of rats caused a reduction in fertility but not in their ability to mate. A gradual return to fertility was seen in those animals which received the lower dose of tyramine or norephedrine, while the other treatments caused a permanent reduction in fertility. The cause of sterility was production of azoospermic ejaculates resulting from either a block in sperm transport in the vas deferens or from a deficiency in the ejaculatory mechanism. Only methoxamine caused a mechanical obstruction of the vas deferens, but it is possible that the other drugs caused a sustained spasm. An emission defect could have been due to transmitter depletion, receptor-specific desensitization or presynaptic alpha-adrenoreceptor-mediated inhibition. Spermatozoa from the cauda epididymidis were immotile following the treatments. 相似文献

The chemotherapeutic agent busulfan was tested for the induction of dominant lethal and specific-locus mutations in male mice. A dose of 5 mg/kg b.w. of busulfan induces dominant lethal mutations in spermatozoa. A dose of 20 mg/kg b.w. induces dominant lethal mutations in spermatozoa and spermatids. A total of 83,196 offspring were scored in the specific-locus experiments. Busulfan-induced specific-locus mutations were recovered in spermatozoa and spermatids, but not in spermatogonia. The sensitivity patterns for the induction of dominant lethal and specific-locus mutations by busulfan in germ cells of male mice are similar but not identical. 相似文献

The developmental prolactin-releasing effect of Tryptoline (T), Methoxytryptoline (MT) and Hydroxytryptoline (OHT) was examined comparatively in male and female rats. A single injection of T 15 mg/Kg increased serum prolactin in both sexes; the increase was significant from day 20 onwards. OHT evoked a sharp rise in 12 day-old rats and the releasing effect increased with age, both in males and females. No significant sex differences were observed in T or OHT treated rats. MT caused an increment in prolactin secretion in male rats and this action increased with age. The releasing effect of MT was not significant in females, even at 38 postnatal days. In adult animals, the tryptolines (15 mg/Kg) were able to increase serum prolactin in males and in females in diestrous; a dose of 5 mg/Kg of T was only effective in adult male rats. The prolactin-releasing effect was drastically reduced by orchidectomy and by ovariectomy. LH, FSH and TSH were not modified by any treatment. The present results show for the first time the ontogeny of the prolactin-releasing effect of tryptolines in male and female rats and that this effect depends on the presence of gonadal secretions in adults. 相似文献

Using the specific-locus method, the ability of 1-ethyl-1-nitrosourea (ENU) and procarbazine hydrochloride (procarbazine) to induce gene mutations in mouse oocytes was tested and confirmed. The sensitive stage for the induction of mutations in oocytes with 160 mg/kg of ENU is 2-4 weeks post treatment. The induced mutation frequency in this mating interval was 5.1 X 10(-7) mutations/locus/gamete/mg/kg. The induction of mutations by procarbazine occurred 8-33 weeks after treatment. The induced mutation frequency in this mating interval for the 400 mg/kg group was 0.4 X 10(-7) mutations/locus/gamete/mg/kg. One third of the induced mutations was lethal in homozygous condition in both experiments. ENU and procarbazine have a lower mutational response in oocytes than in stem-cell spermatogonia. 相似文献

Models of the evolution of gynodioecy assume that inbreeding affects male and female fertility equally and ignore quantitative variation in sex expression. The objectives of this study were to assess inbreeding effects, genetic background, and plant maturity on male and female fertility and the mechanism of male sterility inheritance for Nemophila menziesii (Hydrophyllaceae). Frequency of male-sterile flowers, number of anthers and ovules, and percentage of viable pollen were measured on plants from different pedigrees and five inbreeding levels (F = 0, 0.0625, 0.25, 0.5, and 0.75). Quantitative variation in male sterility was evident. As inbreeding increased, anther and ovule number decreased; the effect on anther number was greater than on ovule number. Pedigrees varied in number of male-sterile flowers and inbreeding effects. Frequency of male-sterile flowers was greatest among first flowers. No trade-off between male and female fertility was detected. A model attributing male sterility to a cytoplasmic locus and restoration to male fertility to a nuclear locus accounted for the distribution of complete sterility and hermaphroditism over the pedigrees. This study suggests that models of the evolution and maintenance of gynodioecy should allow for quantitative variation in male and female fertility components due to inbreeding, pedigree, and plant maturity. 相似文献
