药用植物根系土壤可培养粘细菌的分离鉴定   总被引:1,自引:0,他引:1  
[目的]对药用植物根系土壤可培养粘细菌进行分离与鉴定,探讨药用植物根系土壤粘细菌资源多样性.[方法]采集华南植物园和南岭国家森林公园22种药用植物根系土,利用辅助菌诱导分离技术分离样品中可培养粘细菌;通过菌株显微形态和菌落形态观察,结合16S rDNA基因序列分析,确定分离粘细菌菌株的系统发育地位.[结果]分离获得50株粘细菌,分属于3个科,7个属,其中粘球菌属(Myxococcus)18株,珊瑚球菌属(Corallococcus)11株,孢囊杆菌属(Cystobacter)7株,原囊菌属(Archangium)8株,标桩菌属(Stigmatella)1株,软骨霉状菌属(Chondromyces)4株,匣状球菌属(Pyxidicoccus)1株.粘球菌属和珊瑚球菌属分布最为广泛.[结论]研究发现药用植物根系土壤可培养粘细菌与土壤pH和有机碳含量等环境因子有一定的相关性.粘细菌更适宜在有机质丰富、pH近中性的环境中生长;粘球菌属和珊瑚球菌属的菌株对pH适应性较强,而粘球菌属和孢囊杆菌属的菌株对土壤有机碳含量依赖性不强,在贫瘠土壤中仍有分布.本研究对后续粘细菌资源的开发和利用提供良好的实验材料和理论基础.  相似文献

【目的】从土壤样品中分离粘细菌,对其进行纯化、归类与鉴定,以丰富粘细菌菌种资源;对纯化得到的粘细菌进行抑菌、杀虫及抗肿瘤生物活性的分析,为粘细菌次级代谢产物的开发奠定基础。【方法】采用灭活大肠杆菌和滤纸片诱导子实体法从土样中分离粘细菌,结合形态观察、生理生化特征和16S r DNA基因序列同源性分析,对分离到的各菌株进行鉴定并归类。通过平板扩散法、昆虫口服毒性测试、肿瘤细胞毒性实验等方法,检测粘细菌代谢产物生物活性。【结果】共分离得到35株粘细菌,初步分类为:粘球菌属(Myxococcus)9株;珊瑚球菌属(Corallococcus)9株;侏囊菌属(Nannocystis)11株;堆囊菌属(Sorangium)6株。从其中纯化得到8株粘细菌,对其进行了鉴定并命名。发现了具有高效且广谱的肿瘤细胞毒性效应菌株S22,S51和S55也具同样的细胞毒性;另外,还发现具有肿瘤细胞毒性的菌株S20和S22对枯草芽胞杆菌和白色念珠菌也具有较好的抑菌活性。【结论】土壤中存在丰富的粘细菌资源,发现了具有肿瘤细胞毒性的弱小珊瑚球菌与具有抑菌和强抗肿瘤活性的大孢珊瑚球菌,粘细菌是活性天然产物与新药开发的重要资源。  相似文献

【目的】采用传统的纯培养技术,分离新疆阿克苏地区典型的盐碱地中的粘细菌,并初步分析盐碱地土壤中可培养粘细菌资源的多样性。【方法】采用传统的水琼脂法、滤纸法和改良的土壤浸出液法分离新疆阿克苏地区25份盐碱地的粘细菌。结合分析土样的酸碱度、含盐量、地理位置及其植被分布情况分析新疆阿克苏地区盐碱地粘细菌资源多样性。【结果】共分离到58株粘细菌,它们被鉴定为:粘球菌属(Myxococcus)33株;珊瑚球菌属(Corallococcus)14株;孢囊杆菌属(Cystobacter)6株;堆囊菌属(Sorangium)2株;侏囊菌属(Nannocystis)2株;多囊菌属(Polyangium)1株。其中粘球菌抗逆性强,分离的菌株数最多,在pH值7.5-8.5范围的盐碱地中普遍存在;其次为珊瑚球菌属;而侏囊菌属、多囊菌属的菌株较少见。【结论】新疆阿克苏地区盐碱地粘细菌多样性不高,可能受分离纯化方法、含盐量以及土壤性质影响较大。  相似文献

大兴安岭地区粘细菌资源的多样性及其生物活性   总被引:1,自引:0,他引:1  
【背景】粘细菌是一类具有社会性行为的高等原核生物,其代谢产物具有丰富、多样、新颖的生物活性,是筛选天然药物的良好资源,具有很大的研究开发应用价值。【目的】分析大兴安岭地区粘细菌资源的多样性;分离纯化可培养的粘细菌,分析其抗菌活性并从中筛选对马铃薯晚疫病菌具有拮抗作用的菌株。【方法】通过变性梯度凝胶电泳(DGGE)技术对从大兴安岭地区采集的15个土样中粘细菌资源的多样性进行分析,利用兔粪诱导、大肠杆菌划线诱导和滤纸诱导3种方法从土样中分离可培养的粘细菌,结合形态观察、生理生化特征和16S rRNA基因序列分析确定这些菌株的分类地位,通过平板对峙法对其进行抗菌活性分析。【结果】DGGE的分析结果显示,15个土样中共鉴定到13个属的粘细菌,基本覆盖了大部分已知的粘细菌种属以及部分未分类的粘细菌,表现出了丰富的多样性。共分离得到88株菌,从中得到22株纯菌,经鉴定属于2个属(粘球菌属与珊瑚球菌属)8个种(黄色粘球菌、橙色粘球菌、变绿粘球菌、珊瑚粘球菌、具枝粘球菌、大孢珊瑚球菌、弱小珊瑚球菌和珊瑚状珊瑚球菌)。抗菌结果显示:22株纯菌均表现出可以对一种甚至多种指示菌产生抗性,其中19株抑制大肠杆菌的生长,14株抑制马铃薯晚疫病菌的生长,8株抑制金黄色葡萄球菌的生长,13株抑制酿酒酵母菌的生长,7株抑制枯草芽孢杆菌的生长。【结论】内蒙古大兴安岭地区蕴藏着丰富的粘细菌资源,粘球菌属及珊瑚球菌属可能为该地区粘细菌菌群中的优势菌。分离纯化出的粘细菌菌株均表现出抑制一种甚至多种指示菌生长的活性,其中64%的纯菌对马铃薯晚疫病菌产生抗性,具有进一步研究的潜在价值。  相似文献

【目的】利用被捕食细菌诱导粘细菌形成子实体的方法分离新疆盐碱地粘细菌,并初步分析被捕食菌与被诱导粘细菌之间关系。【方法】从盐碱地分离出16株对粘细菌具有不同诱导作用的被捕食细菌,将其用以分离新疆阿克苏地区盐碱地粘细菌。【结果】对25个盐碱地土样的粘细菌进行分离,共分离到55株粘细菌,结合分子生物学手段和形态学特征,初步将其归属为粘球菌属、珊瑚球菌属、Pyxidicoccus、孢囊杆菌属和侏囊菌属及6株无法纯化的疑似粘细菌。供试的16株被捕食细菌诱导粘球菌属均具有较好效果,而Pyxidicoccus和孢囊杆菌只能由革兰氏阳性菌诱导。【结论】与传统分离方法比较,采用多种被捕食细菌分离粘细菌其出菌率明显提高,数量增加,且易观察和挑取,有利于后续纯化。不同粘细菌的捕食菌谱及捕食嗜好不同,增加被捕食细菌筛选范围可能会提高粘细菌的分离效率。本研究采用被捕食菌分离粘细菌的方法为粘细菌的分离提供了新思路与途径。  相似文献

【背景】黏细菌是一类具有多细胞群体行为特征的高等原核生物,其对植物病原真菌和细菌的捕食特性使其在植物病害防治方面具有重要的应用潜力。【目的】探究乌鲁木齐天山大峡谷原始森林可培养黏细菌的多样性并分析其抗菌活性,为发掘黏细菌生防菌株奠定基础。【方法】以天山大峡谷原始森林采集的土样和腐木为分离材料,采用兔粪诱导法和被捕食菌诱导法从中分离纯化黏细菌菌株,结合形态学观察、生理生化测定和16S rRNA基因序列分析确定其分类地位,并以6种植物病原真菌[大丽轮枝菌(Verticillium dahliae)、尖孢镰刀菌萎蔫专化型(Fusarium oxysporum f. sp. vasinfectum)、拟轮枝链孢霉(Fusarium verticillioides)、立枯丝核菌(Rhizoctonia solani)、黄色镰刀菌(Fusarium culmorum)、细极链格孢菌(Alternaria tenuissima)]和1种植物病原细菌[梨火疫病菌(Erwinia amylovora)]为靶标菌,通过平板对峙法和菌苔捕食法测定其抗菌活性。【结果】从采集的样品中分离出70株菌株,经纯化后获得36株黏细菌纯培养物。经鉴定隶属于4个属,黏球菌属(Myxococcus) 30株、孢囊杆菌属(Cystobacter) 3株、珊瑚球菌属(Corallococcus) 2株和原囊菌属(Archangium) 1株。抗菌活性分析显示,本研究获得的36株黏细菌至少对2种植物病原真菌有抗菌活性,表现出广谱的抗真菌活性,初步筛选出一株菌株NSE37-1兼具广谱和高效抗真菌活性;供试的15株黏细菌对梨火疫病菌均具有捕食活性,初步筛选出一株对梨火疫病菌具有较强捕食能力的黏细菌菌株NSE25。【结论】天山大峡谷可培养黏细菌资源比较丰富,黏球菌属是该地区可培养黏细菌菌群中的优势菌。分离纯化出的黏细菌菌株均表现出广谱的抗植物病原菌活性,具有进一步研究和开发的潜在价值。  相似文献

目的:研究塔里木河天然胡杨林部分地区可培养细菌的生态分布。方法:通过塔里木河胡杨林采样,可培养菌分离及16S rDNA序列鉴定。结果:从3种不同样品(水样、土样和胡杨树杆分泌物)中分离筛选了22株细菌,其中17株菌为革兰氏阳性菌,5株为革兰氏阴性菌。根据生理生化特征与16S rDNA序列分析结果表明,其中15株菌属于芽孢杆菌属,4株属于不动杆菌属、假单胞菌属、动性球菌属和Agrococcus属各含有1个分离株。结论:塔里木河胡杨林可培养微生物中芽孢杆菌比较丰富,其中有3个可能的新种。  相似文献

以MS基本培养基添加蕙兰菌根浸出液制成的培养基进行分离培养的方法,从野生蕙兰(Cymbidium faberi)根部首次分离到内生细菌。经过分离纯化培养获得纯菌株27株。经过16S rDNA基因序列测序,并与GenBank数据比对,其相似性均在98%以上,分析鉴定结果表明,存活的22株菌可分为8属14种。分别隶属于伯克氏菌属(Burkholderia)、假单胞菌属(Pseudomonas)、芽胞杆菌属(Bacillus)、Leifsonia属、贪食菌属(Variovorax)、欧文氏菌属(Erwinia)、Duganella属和不动杆菌属(Acinetobacter)。对这些菌株进行分离培养及鉴定有助于理解兰花与微生物之间的相互作用关系,为开发利用这些微生物开辟新的思路。  相似文献

通过分离陕西秦岭地区羊肚菌根系土样中的黏细菌,建立黏细菌的分离方法,分析羊肚菌根系土样中黏细菌的多样性。采集自陕西商洛地区种植羊肚菌根系土样,采用不同诱导方法分离土样中的黏细菌,比较不同方法的出菌率,通过菌落形态、显微照片对黏细菌进行观察,并结合16S rRNA序列,分析得到的黏细菌种类。采用活的大肠埃希菌作为诱导底物分离出的黏细菌数量最多,本次实验从羊肚菌根系土样中共分离纯化出17株黏细菌,包括5个属,其中黏球菌属(Myxococcus)12株,珊瑚球菌属(Corallococcus)2株,孢囊杆菌属(Cystobacter)1株,原囊菌属(Archangium)1株,标桩菌属(Stigmatella)1株。结果显示羊肚菌根系土壤有黏细菌存在,其中以黏球菌属居多,为后续探讨黏细菌与羊肚菌之间是否存在互生关系提供参考。  相似文献

目的自星花金龟是我国北方常见腐食性昆虫,为了利用其腐食性行为转化处理有机垃圾、畜禽粪便及农作物秸杆等农业有机废弃物资源,并探索其中肠消化的微生态机制,而对其肠道细菌进行系统研究。方法从白星花金龟成虫肠道中无菌操作分离获得5株细菌,对其菌体形态、培养性状、染色反应、生理生化反应等进行鉴定及数量测定。结果白星花金龟成虫肠道的5个细菌菌株分别属于气球菌属(Aerococcus)、短状杆菌属(Brachybacterium)、棍状杆菌属(Clavibacter)、李斯特菌属(Listeria)、皮杆菌属(Dermabacter),以皮杆菌属和棍状杆菌属数量最多,分别为1.3×10^7和1.0×10^7;其次为短状杆菌属和气球菌属,分别为8×10^6和5×10^6;李斯特菌属数量最少,为7×10^5。结论各菌株之间细菌数量存在明显差异,以皮杆菌属细菌数量最多,分离未获得真菌和放线菌。  相似文献

Five ballistoconidiogenous yeast strains, isolated from plant leaves at Cuc Phuong National Forest of Ninh Binh, Vietnam, were assigned to the genus Kockovaella based on morphological and chemotaxonomical characteristics. They represent four new species based on analyses of 18S rDNA sequence, sequences of internal transcribed spacer regions, and DNA-DNA reassociation experiments. Four new species, Kockovaella calophylli (1 strain), Kockovaella cucphuongensis (2 strains), Kockovaella litseae (1 strain), and Kockovaella vietnamensis (1 strain) are proposed for these strains.  相似文献

The growth, morphology, and life cycle of two marine myxobacterial isolates, halotolerant Myxococcus fulvus strain HW-1 and halophilic Haliangium ochraceum strain SMP-2, were studied as models to determine the living patterns of myxobacteria in the ocean. The growth, morphology, and development of halotolerant strain HW-1 shifted in response to salinity. The optimal seawater concentration for growth of HW-1 was 0 to 80% (salinity, 0.1 to 2.9%), and the strain grew poorly in media with a salinity of more than 4%. The cells became shorter as the seawater concentration increased. The fruiting body structure was complete only on agar prepared with low concentrations of seawater or salts (less than 60% seawater; salinity, 2.1%), and rudimentary structures or even simple cell mounds appeared as the seawater concentration increased. In contrast, the halophilic strain SMP-2 was unable to grow without NaCl. The cell length and the morphology of the fruiting body-like structure did not change in response to salts. In seawater liquid medium, the cells of both strains were confirmed to be able to form myxospores directly from vegetative cells, but they could not do so in medium containing a low seawater concentration (10% or less). HW-1 cells from medium containing a high concentration of seawater grew independent of cell density, while cells from medium containing a low concentration of seawater (10% or less) showed density-dependent growth. SMP-2 cells showed density-dependent growth under all salinity conditions. The results suggest that the halotolerant myxobacteria are the result of degenerative adaptation of soil myxobacteria to the marine environment, while the halophilic myxobacteria form a different evolutionary group that is indigenous to the ocean.  相似文献

Five strains were isolated which form a physiologically and phylogenetically coherent group of chlororespiring microorganisms and represent the first taxon in the Myxobacteria capable of anaerobic growth. The strains were enriched and isolated from various soils and sediments based on their ability to grow using acetate as an electron donor and 2-chlorophenol (2-CPh) as an electron acceptor. They are slender gram-negative rods with a bright red pigmentation that exhibit gliding motility and form spore-like structures. These unique chlororespiring myxobacteria also grow with 2,6-dichlorophenol, 2,5-dichlorophenol, 2-bromophenol, nitrate, fumarate, and oxygen as terminal electron acceptors, with optimal growth occurring at low concentrations (<1 mM) of electron acceptor. 2-CPh is reduced by all strains as an electron acceptor in preference to nitrate, which is reduced to ammonium. Acetate, H(2), succinate, pyruvate, formate, and lactate were used as electron donors. None of the strains grew by fermentation. The 16S ribosomal DNA (rDNA) sequences of the five strains form a coherent cluster deeply branching within the family Myxococcaceae within the class Myxobacteria and are mostly closely associated with the Myxococcus subgroup. With the exception of anaerobic growth and lack of a characteristic fruiting body, these strains closely resemble previously characterized myxobacteria and therefore should be considered part of the Myxococcus subgroup. The anaerobic growth and 9.0% difference in 16S rDNA sequence from those of other myxobacterial genera are sufficient to place these strains in a new genus and species designated Anaeromyxobacter dehalogenans. Six isolates of novel marine myxobacteria, designated strains SHK-1T, SMK-1-1, SMK-1-3, SMK-10, SKK-2, and SMP-6, were obtained from various coastal samples (mud, sands and algae) collected around Japan. All of the isolates had Gram-negative rod-shaped cells, motile by gliding and grew aerobically. They showed bacteriolytic action, fruiting body formation, and NaCl requirement for growth with an optimum concentration of 1.0-2.0% (w/v). In addition, divalent cationic components of seawater, such as Mg2+ or Ca2+, were also needed for growth. The major respiratory quinone was MK-7. The G+C content of genomic DNA ranged from 65.6 to 67.4 mol% (by HPLC). The isolates shared almost identical 16S rDNA sequences, and clustered with a recently described marine myxobacterium, Plesiocystis pacifica, as their closest relative on a phylogenetic tree (95.9-96.0% similarity). Physiological and chemotaxonomic differences between the new strains and strains of the genus Plesiocystis justify the proposal of a new genus. Therefore, we propose to classify the six isolates into a new taxon of marine myxobacteria with the name, Enhygromyxa salina gen. nov., sp. nov. By using baiting techniques and different purification methods, a high number of myxobacterial strains have been isolated as pure cultures from soil of different regions of China. Because myxobacterial cells do not disperse easily in liquid media, a medium containing an enzymatic hydrolysate of casein (CEH) medium have been used for purification and purity tests combined in a single step. The key method, in which isolates are reintroduced to sterile rabbit dung to induce fruiting bodies formation, facilitates purification of myxobacteria. Sterile rabbit dung pellets are used to mimic the natural growth substance of these organisms which has the advantage that characteristic fruiting bodies emerge, which is a key characteristics in the taxonomy of myxobacteria. In this study, the optimum program of isolation and purification of some myxobacteria strains has been established which will facilitate screening programs. Moreover, the development of fruiting body formation of strain BD20 (Chondromyces) and strain BD54 (Cystobacter) have been recorded in this study.  相似文献

[目的]为了探究武陵山放线菌多样性,以便从新放线菌菌株中发现新的潜在药物先导化合物.[方法]从武陵山采集280份土样,采用纯培养的方法,用4种培养基分离到1134株放线菌.选择其中30株代表菌进行了初步分类鉴定;以3株细菌和7株农作物致病真菌作为指示菌,检测其抑菌活性;利用特异性引物扩增的方法,检测是否具有聚酮合酶(PKS Ⅰ、PKSⅡ)基因、非核糖体多肽合成酶(NRPS)基因和多烯类化合物合成酶(CYP)基因.[结果]分离到的武陵山放线菌中,链霉菌占70%以上,还有小单孢菌等8个科13个属,其中有5个菌株是潜在的新种.选取的30株实验菌对细菌、真菌有不同程度的抗菌活性;其中含有4类化合物合成基因的菌株占23%~60%.[结论]武陵山原始森林土壤中,放线菌多样性很丰富,且存在很多未开发的稀有类群.有抑菌活性的菌株,可用于进一步的药物开发利用.  相似文献

Phenotypic and phylogenetic studies were performed on two myxobacterial strains, SMP-2 and SMP-10, isolated from coastal regions. The two strains are morphologically similar, in that both produce yellow fruiting bodies, comprising several sessile sporangioles in dense packs. They are differentiated from known terrestrial myxobacteria on the basis of salt requirements (2-3% NaCl) and the presence of anteiso-branched fatty acids. Comparative 16S rRNA gene sequencing studies revealed that SMP-2 and SMP-10 are genetically related, and constitute a new cluster within the myxobacteria group, together with the Polyangium vitellinum Pl vt1 strain as the closest neighbor. The sequence similarity between the two strains is 95.6%. Based on phenotypic and phylogenetic evidence, it is proposed that these two strains be assigned to a new genus, Haliangium gen. nov., with SMP-2 designated as Haliangium ochraceum sp. nov. (= JCM 11303(T) = DSM 14365(T)), and SMP-10 as Haliangium tepidum sp. nov. (= JCM 11304(T)= DSM 14436(T)).  相似文献
