目的:采用胰蛋白酶制备霞水母ACE抑制肽,并以响应面法优化酶解肽制备工艺,通过超滤分离获得具有ACE抑制作用的酶解肽活性部位。方法:单因素实验以ACE抑制率为指标,考察温度、酶解时间、pH、加酶量,进一步通过响应面法优化酶解工艺,以不同截留分子量的纤维膜进行分离。结果:胰蛋白酶水解霞水母制备ACE抑制肽的最佳工艺条件为:酶解温度:49.6℃,加酶量为0.69%,pH为7.8,酶解时间2.0h,酶解产物的平均ACE抑制率为56.79%,与预测值57.20%的相对误差为0.41%,经过超滤分离获得分子量1k Da的酶解产物,ACE抑制活性最高,其IC50为66.58μg/ml。结论:确定霞水母酶解肽的制备工艺,其中分子量1kDa的部分为主要效应物质基础。  相似文献

以百尾参多糖脱色率、多糖损失率为指标,通过单因素和正交试验研究活性炭含量、时间、温度和pH对多糖脱色效果的影响,优化百尾参多糖最佳脱色条件;对Sevage法、三氯乙酸(TCA)法、蛋白质等电点法以及酶法4种脱蛋白方法进行比较,选择较好的脱蛋白方法。建立环磷酰胺免疫低下小鼠模型,研究百尾参多糖的免疫调节活性。结果表明活性炭脱色的最佳条件是活性炭加入量1%、温度60 ℃,时间45 min、pH 4.5;酶法为4种方法中较好的脱蛋白方法。百尾参多糖能加速恢复环磷酰胺致小鼠免疫器官萎缩,也能剂量依赖性地提高小鼠血清中的白介素-2(IL-2)、肿瘤坏死因子α(TNF-α)等细胞因子含量,表明百尾参多糖对环磷酰胺的免疫抑制具有保护作用。  相似文献

本研究旨在探讨活性炭对熟地多糖提取液脱色的工艺条件。在单因素试验基础上,采用正交试验,以多糖脱色率(%)和多糖剩余率(%)为指标,用紫外-可见分光光度法测定,确定了熟地多糖活性炭脱色的最优工艺的参数。结果发现:活性炭添加量对脱色效果的影响最大,其次为温度,再次为时间。最佳脱色条件为脱色温度60℃,活性碳添加量5%,吸附时间30 min,在此条件下,脱色率为98.20%,多糖剩余率为84.89%。从而表明活性炭对熟地多糖脱色工艺可行,简便,有效。  相似文献

通过实验比较了不同吸附剂的脱色效果及其在脱色过程中引起的氨基酸损失率,选择活性炭作为微生物菌体蛋白质水解溶液脱色的吸附剂。对影响其脱色效果的几个主要因素进行研究,并通过正交实验分析优化后得到脱色条件为:吸附剂用量1.5 g(每50 mL蛋白提取液),脱色温度55℃,pH值5和脱色时间1 h。  相似文献

将TiO2光催化剂负载于活性炭,然后将活性炭装填于冷电弧-光催化-吸附集成反应器中,用该反应器对豆粕氨基酸进行脱色。以豆粕氨基酸的脱色率和氨基酸保留率为指标,探讨处理电压、处理时间、溶液pH和活性炭填充量4个因素对脱色效果的影响。在单因素试验的基础上,利用二次回归正交旋转组合设计建立回归模型方程确定最佳工艺条件,并对方程进行回归分析和系数检验。结果表明:回归方程在#=0.01水平上高度显著。各因素对脱色效果的影响程度大小依次为:溶液pH>活性炭填充量>处理时间>处理电压。最佳脱色条件为处理电压20kV、处理时间11min、溶液pH 3.6和活性炭填充量63g,此条件下氨基酸溶液脱色率为94.8%,保留率为83.1%。  相似文献

以多糖含量和得率为指标,采用正交试验法对陕北红枣中多糖的提取与分离工艺进行优选。酶加水提取的最佳工艺为:①温度70℃、酶添加量3.0%、时间2h;②温度70℃、酶添加量2.0%、时间4h。分离的最佳工艺为:分级沉淀多糖时采用浓缩液:无水乙醇(v/v)1:3效果最好;当采用活性炭脱色时,活性炭用量为7g/L时,既达到了脱色的目的,多糖的损失率又较小。  相似文献

鳕鱼碎肉酶解液脱腥脱苦的研究   总被引:4,自引:0,他引:4  
为去除鳕鱼碎肉酶解液的腥苦味,分别比较了粉末活性炭吸附、酵母发酵和环状糊精包埋法的脱腥脱苦效果。采用单因素试验、正交试验等方法,研究了温度、时间、pH和脱腥剂用量对脱腥脱苦的影响,并得出了各脱腥剂的最佳脱腥脱苦条件。结果表明,活性炭和酵母粉联合去苦腥味效果最优:鳕鱼碎肉酶解液经质量分数1%酵母粉35℃下发酵1 h,然后加入质量分数0.5%的活性炭,在40℃、pH 5.0条件下吸附40 min,制得的酶解液基本上无苦腥味,且活性回收率可达91.81%。以上结果为鳕鱼碎肉酶解液的进一步应用奠定了技术基础。  相似文献

为了提高L-鸟氨酸发酵液的脱色效率,对其脱色工艺开展研究。首先通过树脂的选择性吸附确定L-鸟氨酸发酵液中色素的化学性质,随后结合颗粒活性炭的物理结构和零电荷点分析,优选出在接近中性条件下具有优良脱色性能的颗粒活性炭。在静态条件下考察pH、温度、时间和活性炭用量等因素对其脱色性能的影响。在此基础上,考察活性炭层析柱对L-鸟氨酸发酵液的动态脱色工艺和基于两步解吸法的活性炭再生工艺,单柱可动态脱色处理45倍床层体积(BV)的发酵液,脱色率达97%以上,脱色液呈无色透明状,L-鸟氨酸损失率低于1%,活性炭再生效果保持稳定。  相似文献

塔拉多糖是一种半乳甘露聚糖胶,对于我们具有非常重要的应用价值。本实验主要对塔拉提取物中的塔拉多糖进行脱色工艺化研究;在单因素实验的基础上,对活性炭颗粒质量、脱色时间、脱色温度以及脱色次数这四种因素进行正交优化实验,其最佳脱色实验参数为:活性炭颗粒0.6 g,脱色45 min,脱色温度45℃脱色次数3次,脱色率可以达到50.21%,同时多糖类保留率为90.39%。  相似文献

以脱色率、多糖保留率和细胞抑制率3个指标,从D301、D303、D315、D900树脂筛选出脱色效果较好的树脂;采用MTT法检测,脱色后的山茱萸粗多糖抗HL-60细胞活性提高;对D303树脂,通过单因素和正交试验对其进行优化,确定最优工艺条件是时间7 h、温度50℃、样品浓度15 mg/m L,脱色率和多糖保留率分别为79.71%、81.12%。大孔树脂脱色工艺的研究,为山茱萸药物的研发提供了一定实验基础。  相似文献

The present work is focused on efficient immobilization of polygalacturonase on polyethylene matrix, followed by its application in apple juice clarification. Immobilization of polygalacturonase on activated polyethylene and its use in apple juice clarification was not reported so far. Aspergillus niger Van Tieghem (MTCC 3323) produced polygalacturonase when grown in modified Riviere's medium containing pectin as single carbon source by fed-batch culture. The enzyme was precipitated with ethanol and purified by gel filtration chromatography (Sephacryl S-100) and immobilized onto glutaraldehyde-activated polyethylene. The method is very simple and time saving for enzyme immobilization. Various characteristics of immobilized enzyme such as optimum reaction temperature and pH, temperature and pH stability, binding kinetics, efficiency of binding, reusability and metal ion effect on immobilized enzymes were evaluated in comparison to the free enzyme. Both the free and immobilized enzyme showed maximum activity at a temperature of 45 degrees C and pH 4.8. Maximum binding efficiency was 38%. The immobilized enzyme was reusable for 3 cycles with 50% loss of activity after the third cycle. Twenty-four U of immobilized enzyme at 45 degrees C and 1 h incubation time increased the transmittance of the apple juice by about 55% at 650 nm. The immobilized enzyme can be of industrial advantage in terms of sturdiness, availability, inertness, low price, reusability and temperature stability.  相似文献

Summary This paper deals with the standardisation of a suitable method to determine urease activity in bottom deposits of sewage stabilisation ponds and activated sludge units.Standard conditions were established; pH, incubation time, sample quantity, temperature, and the results are presented graphically.As the method is not dependent on ammonia distillation under vacuo, it is therefore suitable for serial analysis.Results from tests using this method can be used to determine the urea degrading efficiency of purification systems like stabilisation ponds and activated sludge systems treating urea containing effluents.  相似文献

Broiler manure produced at large concentrated facilities poses risks to the quality of water and public health. This study utilizes broiler litter and cake as source materials for granular activated carbon production and optimizes conditions for their production. Pelletized manure samples were pyrolyzed at 700 degrees C for 1 h followed by activation in an inert atmosphere under steam at different water flow rates, for a period ranging from 15 to 75 min. Carbon physical and adsorptive properties were dependent on activation time and quantity of steam used as activant, yields varied from 18% to 28%, surface area varied from 253 to 548 m2/g and copper ion adsorption varied from 0.13 to 1.92 mmol Cu2+/g carbon. Best overall performing carbons were steam activated for 45 min at 3 ml/min. Comparative studies with commercial carbons revealed the broiler cake-based carbon as having the highest copper ion efficiency.  相似文献

The covalent immobilization of yeast invertase with glutaraldehyde at activated carbon, modified preliminarily by urea and dimethyl formamide treatment, has been established. Some physicochemical properties of the immobilized and native enzyme in water and water-organic solutions have been studied. Hydrolytic, as well as transferase enzyme characteristics have changed after immobilization. The optimal conditions for hydrolytic and transferase activity of immobilized invertase are pH 6.0 and 7.0, respectively. The optimum temperature for the immobilized enzyme is 30°C. The conversion degree of isoamyl alcohol depends on the substrate and enzyme concentrations in medium, holdup time and organic phase quantity in the reaction medium.  相似文献

The main factors that affect the large specific surface area (SSA) of the activated carbon from agricultural waste corn cobs were studied by chemically activated method with solution of KOH and soap which acted as surfactant. The experiment showed that not only the activation temperature, activation time and the mass ratio of KOH to the carbonized material, but also the activated methods using activator obviously influenced the SSA of activated carbon. The experimental operating conditions were as follows: the carbonized temperature being 450 degrees C and keeping time being 4 h using N2 as protective gas; the activation temperature being 850 degrees C and holding time being 1.2 h; the mass ratio of KOH to carbonized material being 4.0; the time of soaking carbonized material in the solution of KOH and soap being 30 min. Under the optimal conditions, the SSA of activated carbon from corn cobs reached 2700 m2/g. And the addition of the soap as surfactant may shorten the soaking time. The structure of the activated carbon prepared had narrow distribution of pore size and the micro-pores accounted for 78%. The advantages of the method described were easy and feasible.  相似文献

富集分离了除草剂二甲戊灵降解真菌,并研究了其降解特性,结果表明,真菌可以降解二甲戊灵,利用富集培养的方法从环境中分离到16株能降解二甲戊灵的真菌。其中10株真菌5d内对100mg·L^-1二甲戊灵的降解率大于60%,以其中3株生理耐受能力强、降解能力高的真菌为例,研究了外加碳源浓度、初始pH值、二甲戊灵浓度和培养温度对真菌生长量和降解能力的影响,此3株真菌经鉴定分别属于土生曲霉组(Aspergillus terreus)、长梗串孢霉属(Monilochaetes)和烟色曲霉组(Aspergillus furnigatus),在外加碳源浓度为0.5%~1.0%的范围内,真菌生长量和降解率达到最大;在中性培养液中,3株真菌的生长量大,降解能力强;在浓度为100mg·L^-1时降解率和生长量都比较大,而绝对去除量随二甲戊灵浓度的提高而增加,在500mg·L^-1时达到最大;真菌的生长和降解需要适宜的温度,20~30℃培养时,降解率和生长量最大,可为农药污染治理及生产污水处理提供理论依据。  相似文献
