A transect along the axis of the headwaters of a tidal estuary was sampled for microbial, nutrient, and physical parameters. Chlorophylla averaged 42g 1–1 and phytoplankton comprised an estimated 80% of the total microbial biomass as determined by adenosine triphosphate (ATP). Bacterial concentrations ranged from 0.3–53.9×106 cells ml–1 and comprised about 4% of the total living microbial biomass. Bacterial production, determined by3H-methyl-thymidine incorporation was about 0.05–2.09× 109 cells 1–1 h–1, with specific growth rates of 0.26–1.69 d–1. Most bacterial production was retained on 0.2m pore size filters, but passed through 1.0m filters. Significant positive correlations were found between all biomass measures and most nutrient measures with the exception of dissolved inorganic nitrogen nutrients where correlations were negative. Seasonal variability was evident in all parameters and variability among the stations was evident in most. The results suggest that bacterial production requires a significant carbon input, likely derived from autotrophic production, and that microbial trophic interactions are important.  相似文献

Published field data for lupins grown in a deep sandy soil in the wheatbelt of south-western Australia were used to test the predictive ability of a model of three-dimensional root growth. The model has the capacity to simulate the growth of individual root sections in response to the supply and demand for water and nitrate. N mineralisation was not modelled explicitly, but was accounted for through the use of a seasonally variable mineralisation input derived from the field data. Simulated nitrogen and water contents and root length densities in the soil profile agreed well with observed profiles, although all were slightly under-predicted. A sensitivity analysis revealed that model predictions were most sensitive to the drained upper limit values (v/v) and the mineralisation rates (gN m–3 s–1) incorporated as external inputs to the model, along with the unit rate of N2 fixation (mol nodule–1 s–1) and unit root growth rates (m mol–1 s–1) which are physiological parameters previously calibrated for lupins. The amount of nitrate leached was predicted well. Spatial plots of nitrate leaching were a close inverse of the root length density plots, with the highest nitrate leaching below the inter-plant zones, and the least nitrate leaching directly below each plant. These results suggest that the root distribution of a legume species such as lupin can have an effect on the leaching of nitrate to depth. It may thus be possible to reduce the total amount of nitrate leached under lupin crops by investigating factors such as the spatial deployment of roots, planting densities and intercropping.  相似文献

Age and growth of the whiskery shark, Furgaleus macki, from southwestern Australia were examined using vertebral ageing and tag-recapture data. The readability of bands on the vertebral centra varied markedly between individuals. Four readers were used to make band counts, with the most experienced reader having the lowest index of average percent error and the highest level of agreement with final counts. Marginal increment analysis indicated that opaque bands form in January. With parturition occurring from August to October, size data suggests that the first band is probably formed 15–17 months after birth. The age at maturity was estimated to be 4.5 years for males, and 6.5 years for females. The oldest male was 10.5 years, and oldest female was 11.5 years. Von Bertalanffy growth parameters for males were L =121.5cm fork length, K=0.423 year–1, t 0=–0.472 years, were L =120.7cm fork length, K=0.369 year–1, t 0=–0.544 years for females, and were L =118.1cm fork length, K=0.420 year–1, t 0=–0.491 years for combined sexes. Data from a tag recapture study were analysed using a maximum likelihood method to verify the estimates of growth parameters from vertebral ageing. Von Bertalanffy growth parameters from the tag recapture study were L =128.2cm fork length, K=0.288 year–1, t 0=–0.654 years. The two methods of estimating growth parameters produced similar results, with rapid growth until approximately 5 years of age, after which there was little increase in length.  相似文献

Based on a review of N2O field studies in Europe, major soil, climate and management controls of N2O release from agricultural mineral soils in the European Union have been identified. Data for these N2O emission drivers can easily be gathered from statistical services. Using stepwise multivariate linear regression analysis, empirical first order models of N2O emissions have been established which allow – in contrast to existing large-scale approaches – a regionally disaggregated estimation of N2O emissions at sub-national, national and continental level in the temperate and boreal climate regions of Europe. Arable soils showed lower mean and maximum emissions in oceanic temperate climate (Temperate West) than in pre-alpine temperate and sub-boreal climate (Sub-boreal Europe). Therefore, two separate regression models were developed. Nitrous oxide emissions from arable soils the Temperate West amount to an average flux rate below 2 kg N2O-N ha–1 yr–1 and rarely exceed 5 kg N2O-N ha–1 yr–1. They are modelled by the parameters fertiliser, topsoil organic carbon and sand content. In Sub-boreal European arable soils, N2O emissions vary in a much wider range between 0 and 27 kg N2O-N ha–1 yr–1 in dependence of available nitrogen, represented in the model by fertiliser and topsoil nitrogen content. Compared to existing methods for large scale inventories, the regression models allow a better regional fit to measured values since they integrate additional driving forces for N2O emissions. For grasslands, a fertiliser-based model was established which yields higher emission estimates than existing ones. Due to an extreme variability, no climate, soil nor management parameters could be included in the empirical grasslands model.  相似文献

Objective

The control of gait requires executive and attentional functions. As preterm children show executive and attentional deficits compared to full-term children, performing concurrent tasks that impose additional cognitive load may lead to poorer walking performance in preterm compared to full-term children. Knowledge regarding gait in preterm children after early childhood is scarce. We examined straight walking and if it is more affected in very preterm than in full-term children in dual-task paradigms.

Study design

Twenty preterm children with very low birth-weight (≤ 1500 g), 24 preterm children with birth-weight > 1500 g, and 44 full-term children, born between 2001 and 2006, were investigated. Gait was assessed using an electronic walkway system (GAITRite) while walking without a concurrent task (single-task) and while performing one concurrent (dual-task) or two concurrent (triple-task) tasks. Spatio-temporal gait parameters (gait velocity, cadence, stride length, single support time, double support time), normalized gait parameters (normalized velocity, normalized cadence, normalized stride length) and gait variability parameters (stride velocity variability, stride length variability) were analyzed.

Results

In dual- and triple-task conditions children showed decreased gait velocity, cadence, stride length, as well as increased single support time, double support time and gait variability compared to single-task walking. Further, results showed systematic decreases in stride velocity variability from preterm children with very low birth weight (≤ 1500 g) to preterm children with birth weight > 1500 g to full-term children. There were no significant interactions between walking conditions and prematurity status.

Conclusions

Dual and triple tasking affects gait of preterm and full-term children, confirming previous results that walking requires executive and attentional functions. Birth-weight dependent systematic changes in stride velocity variability indicate poorer walking performance in preterm children who were less mature at birth.  相似文献

A population of the Hawaiian coral-reef goby, Asterropteryx semipunctata, was sampled over a 12-month period to determine basic demographics and reproductive parameters. The sexes did not differ in median length or weight, although the largest males were considerably longer and heavier than the largest females. Overall adult sex ratio was 1:1; monthly sex ratios did not differ from unity except in June, when there was a significant female bias. Minimum age at maturity (17.5–19mm SL) was estimated to be 4.5–5 months after hatching. Nearly all fish over 22mm SL were mature. Mature females were found in all months of the year, and females that showed evidence of recent or imminent spawning were collected in every month except December. Gonadal analyses indicated a peak in breeding during the summer (May–July) and minimal spawning during the winter (January–February). Between 20% and 30% of females showed evidence of having spawned within the 24-h period prior to collection; therefore, it was estimated that females spawned, on average, at least once every five days and perhaps as frequently as every three days. Mean batch fecundity was 708 eggs (± 418), and was not well predicted by standard length, body weight, or somatic condition. Relative batch fecundity (mean = 1.44 oocytes mg–1 somatic wet weight) varied seasonally, with higher values in spring and summer (April–July) than in fall and winter (September–January). Reproductive parameters are compared and contrasted with those of other gobiid fishes to elucidate general differences between temperate and tropical species.  相似文献

Mathematical model parameters for the methanogenic degradation of propylene glycol were estimated in a sequential manner by means of an optimization technique. Model parameters determined from an initial experimental data set using one bioreactor were then verified with the results from a second bioreactor. The proposed methodology is a useful tool to obtain model parameters for continuous flow reactors with completely mixed regime. Reproduction and growth of the vermilion snapper, Rhomboplites aurorubens, were studied in Trinidad and Tobago. The smallest individual caught measured 145mm total length (TL) and all fish appeared to be mature. It was not possible to precisely determine size at first maturity due to the use of macroscopic techniques. The smallest spent male and female measured 181 and 211mm TL respectively, suggesting a size at first maturity below these sizes. Spawning occurred throughout the year, with a period of peak spawning from about June to November in the rainy season when river runoff increased. Sagittal otolith sections were used for age determination and the opaque ring, which was counted as the annulus, was deposited from January to May in the dry season. A total of 11 age groups between the ages of 2–12 years (155–505mm total length) were found. The von Bertalanffy growth parameters were: L=532mm, K=0.13y–1, and t0=–0.17, where L is the asymptotic length, K is the growth coefficient and t0 is the theoretical age at zero length. The relationship between weight (WT) and length (TL) was WT=3.43×10–5 TL2.82. Vermilion snapper in this study area appears to grow slower and attain a smaller asymptotic length, but has a longer lifespan than found in populations in higher latitudes. This may be attributed to different levels of exploitation, which may be higher in the latter areas.  相似文献

Summary Corynebacterium glutamicum was observed to undergo several morphological and ultrastructural changes during a shift in dilution rate when grown in phosphate-limiting continuous culture. At 0.1 h–1 the cell culture appeared homogeneous and the average diameter of cells and the cell length was approximately 0.7 m and 1–1.5 m respectively. At 0.04 h–1 there was essentially no change in these readings, but at this dilution rate there was a significant proportion of cells that measured three times the original length. At six residence times the elongated cells were increasing in number, but these changes occurred without diminishing the system's performance. Some changes in cell ultrastructure were observed during the shift in dilution rate. Thus, the presence of polyphosphate granules and of glycogen appeared in the cytoplasm of producing cells (0.04 h–1). The reproductibility of tank experiments concerning unicellular marine algal development was analyzed by means of parallel experiments with cultures ofThalassiosira rotula andSkeletonema costatum, using large flexible plastic tanks under semi-natural conditions. The tanks (3–4 m3, 4–5 m deep) were exposed in the German Bight at a station in the outer harbour of helgoland. The water was obtained from the open North Sea in towable tanks; it was filtered (plate filter), enriched with nitrate (20–30 gat dm–3), phosphate (1.3–2.3 gat dm–3) and silicate (15–23 gat dm–3)-nearly natural springtime concentrations in this area-and inoculated with 103–105 cells dm–3. The water was mixed with non-metal stirring equipment. Within 5 days, concentrations of 106–107 cells dm–3 in an exponential growth phase were obtained. In experiments withT. rotula a parallel development was achieved in spite of some contamination by surrounding water. This is the case for nearly all parameters analyzed (nutrient salts, phytoplankton, bacteria, C, N and particulate carbohydrates). The heterotrophic bacteria, which were determined by means of the plate method, reached concentrations of up to 106 (T. rotula) and 105 (S. costatum) CFU cm–3, respectively. They showed a consistent retrograde development at diatom concentrations above a certain level. The crop did not increase again until the diatoms had reached the stationary phase. During exponential growth ofT. rotula (G=8.9–11.7 h) a partially synchronous cell division was observed. There were also rhythms with respect to cell size (pervalvar axes) and chain length (number of cells). For the experiments withS. costatum (G=10–11.4 h) diurnal variations of cell size and chain length occurred. The present results indicate acceptable reproducibility of algal development and related phenomena in enclosed water bodies.  相似文献

The experiments were carried out on the IMR-32 human neuroblastoma and NIE-115 murine neuroblastoma cultured cells. Peculiarities of the ion channel expression and their correlation with the main morphological parameters characterizing neuronal differentiation were studied under conditions of incubation of the cells with 2-interferon and 2,5-oligoadenylate, 2–5A. Twenty-four hours after addition of 1000 U act./ml interferon to the culture medium, a 56% increase in the mean projective surface of the IMR-32 cells was observed, and after a nine-day-long exposition this increase was 132%, as compared with the control. Mean total length of the cellular protrusions nearly doubled after nine-day-long incubation. Morphological and electrophysiological properties of the N1E-115 murine neuroblastoma cells showed practically no changes after incubation with human 2-interferon. Cultivation of the IMR-32 human neuroblastoma cells in the presence of 2–5A evoked insignificant changes in their morphological parameters. By contrast, the mean total length of neurites of the N1E-115 neuroblastoma protrusion-supplied cells increased more than a factor of five after eight-day-long incubation with 2–5A in a 1.0 µM concentration, and at a 0.01 µM 2–5A concentration this increase was about fourfold. At the same time, the projective surface exhibited no significant changes either in the neurite-supplied or the neurite-free cellular subpopulations. Twenty-four hours after the incubation with human 2-interferon had been begun, the density of sodium current in the IMR-32 human neuroblastoma cells increased by 250% compared with the control. A similar effect was observed after the addition of 2–5A to the medium: the density of sodium current approximately doubled. Cultivation of the neurite-supplied N1E-115 murine neuroblastoma cells was followed by a gradual increase in the density of fast sodium current both in control and 2–5A-influenced samples, but in the latter case this increase was significantly faster. In the neurite-free cells, the density of sodium current was 27% higher after their 24-h-long incubation with 2–5A, as compared with the control values (11.05±0.9 and 8.7±0.9 pA/pF, respectively). Longer incubation resulted in a sharp decrease in the density of potassium current. Summary Quantitative variation in seven morphological characteristics (leaf length and width, leaf length/ width ratio, flower, petal and stomata length, and number of chloroplasts in guard cells) were studied in Petunia hybrida plants regenerated from anther tissue culture and belonging to four different classes of ploidy (2n, 2n–3n, 3n–2n, 4n–8n). Results showed that leaf size is not a good characteristic for discriminating between plants of different ploidy — flower and stomata characteristics being more adequate for this purpose. After applying stepwise discriminant analysis the association chloroplast number — leaf length/width ratio — petal length was verified to be more appropriate for the discrimination of ploidy classes.  相似文献

We tested the hypothesis that dopamine contributes significantly to the hydroxyl radical (OH)-induced striatal neurotoxicity caused by 3-nitropropionic acid (3-NP) in a rat model of Huntington's disease. Dopamine (10–100 μM) or 3-NP (10–1000 μM) individually caused a significant increase in the generation of hydroxyl radical (OH) in the mitochondria, which was synergistically enhanced when the lowest dose of the neurotoxin (10 μM) and dopamine (100 μM) were present together. Similarly, systemic administration of l-DOPA (100–250 mg/kg) and a low dose of 3-NP (10 mg/kg) potentiated OH generation in the striatum, and the rats exhibited significant decrease in stride length, a direct indication of neuropathology. The pathology was also evident in striatal sections subjected to NeuN immunohistochemistry. The significant changes in stride length, the production of striatal OH and neuropathological features due to administration of a toxic dose of 3-NP (20 mg/kg) were significantly attenuated by treating the rats with tyrosine hydroxylase inhibitor α-methyl-p-tyrosine prior to 3-NP administration. These results strongly implicate a major contributory role of striatal dopamine in increased generation of OH, which leads to striatal neurodegeneration and accompanied behavioral changes, in 3-NP model of Huntington's disease.  相似文献

Two types of time scales – 10-day intervals (D) and phenological phases (P) – were applied to environmental variables for the development of statistical regression models relating to the southwest pampa region of Argentina, with the aim of detecting the effects of weather on wheat yields for the period 1977–1999. The parameters were grouped as meteorological and processed variables and indices. The processed variables used were total soil water availability (SWA) and the ratio of actual evapotranspiration to potential evapotranspiration (), obtained from a water-balance model in which the moisture anomaly index (Z) and the Palmer drought severity index (PDSI) were calculated according to Palmers model. For these parameters it was possible to establish the times of year and the phenological phases with the best correlation to grain yield. The regression equation for meteorological variables on a 10-day scale provides one of the best fits. Using mixed parameters, the two models, D and P, give rise to a standard error of estimate of approximately 200 kg ha–1. Truncated models perform better on a P scale than on a D scale. The use of phenological stages improved yield assessment, particularly for those years with extreme meteorological conditions. The optimum models were tested and root-mean-square errors (RMSE) of 440 kg ha–1 and 470 kg ha–1 were obtained for P and D scales respectively.  相似文献

Summary Cells isolated from ascidian smooth muscle were about 1.5–2 mm in length. Each contained 20–40 nucle in proportion to cell length. The cytoplasm was characterized by the presence of an enormous quantity of glycogen particles, tubular elements of sarcoplasmic reticulum coupled to the cell membrane, and conspicuous contractile elements. Thick and thin filaments had diameters of about 14–16 nm and 6–7 nm, respectively. The population density of the thick filaments was much higher (mean 270/m2 filament area) than in vertebrate smooth muscles. The ratio of thick to thin filaments was about 16. All the thick filaments were surrounded by a single row of 5–9 thin filaments forming a rosette, and cross-bridges with periodicities of 14.5 and 29 nm were found between them. The contractile apparatus consisted of numerous myofibrils which were arranged nearly along the cell axis and were separated from each other by a network of 10-nm filaments. The myofibrils further consisted of many irregularly arranged sarcomerelike structures, each of which was comprised of a small group of thick and thin filaments with attached dense bodies.  相似文献

A modification of reverse chromosome painting was carried out using genomic DNA from tumor cells as a complex probe for chromosomal in situ suppression hybridization to normal metaphase chromsome spreads. Amplified DNA sequences contained in such probes showed specific signals, revealing the normal chromosome positions from which these sequences were derived. As a model system, genomic DNAs were analyzed from three tumor cell lines with amplification units including the proto-oncogene c-myc. The smallest amplification unit was about 90 kb and was present in 16–24 copies; the largest unit was bigger than 600 kb and was present in 16–32 copies. Specific signals that co-localized with a differently labeled c-myc probe on chromosome band 8q24 were obtained with genomic DNA from each cell line. In further experiments, genomic DNA derived from primary tumor material was used in the case of a male patient with glioblastoma multiforme (GBM). Southern blot analysis using an epidermal growth factor receptor gene (EGFR) probe that maps to 7p13 indicated the amplification of sequences from this gene. Using reverse chromosome painting, signals were found both on band 7p13 and bands 12q13–q15. Notably, the signal on 12q13–q15 was consistently stronger. The weaker 7p13 signal showed co-localization with the major signal of the differently labeled EGFR probe. A minor signal of this probe was seen on 12q13, suggesting cross-hybridization to ERB3 sequences homologous to EGFR. The results indicate co-amplification of sequences from bands 12q13–q15, in addition to sequences from band 7p13. Several oncogenes map to 12q13–q15 providing candidate genes for a tumor-associated proto-oncogene amplification. Although the nature of the amplified sequences needs to be clarified, this experiment demonstrates the potential of reverse chromosome painting with genomic tumor DNA for rapidly mapping the normal chromosomal localization of the DNA from which the amplified sequences were derived. In addition, a weaker staining of chromosomes 10 and X was consistently observed indicating that these chromosomes were present in only one copy in the GBM genome. This rapid approach can be used to analyze cases where no metaphase spreads from the tumor material are available. It does not require any preknowledge of amplified sequences and can be applied to screen large numbers of tumors.  相似文献

Field research was conducted on four Atlantic Coastal Plain soils in the United States to evaluate response of corn (Zea mays L.) plants to Mn application. The soils under study were classified as either Aeric or Typic Ochraquults. Manganese application increased corn grain yields by an average of 1195 kg ha–1 on the four soils. The average grain yields on the soils were 7955 kg ha–1 for the control and 9150 kg ha–1 for the +Mn treatment. A Mitscherlich plant growth model was used to establish relationships between percent maximum grain yield and Mn concentration in the ear leaf at early silk (r=0.87, =0.01) and in the mature grain (r=0.58, =0.01). Based on 90% of maximum yield as the definition of the critical deficiency level, the critical Mn deficiency levels calculated with parameters from the Mitscherlich model were 10.6 mg kg–1 in the ear leaf and 4.9 mg kg–1 in the grain.  相似文献

Black seabream, Spondyliosoma cantharus, caught off the Canary Islands is characterized by a protogynous hermaphroditism. The size range of the catches is between 8 and 40cm, with a main distribution between 16 and 24cm. The mean length of the individuals shows an increase with increasing depth. Males:females ratio is unbalanced in favour of females (1:2.18). The reproductive season extends from late autumn to mid spring, with a peak in spawning activity in January–February. A dichromatism is found between males and females during the spawning season. Males reach maturity at a larger total length, 22.7cm (3 years old), than females, 17.3cm (2 years old). Morphometric relationship between length and mass for the whole population is described by the parameters: a=0.00732, and b=3.24747. Otoliths age readings indicate that the exploited population consists of eleven age groups (0–X years), including a very high proportion of individuals between 2 and 3 years old. The von Bertalanffy growth parameters for the whole population are: L=43.35cm, k=0.24 year–1, and t0=–0.11 year. The rates of total, natural and fishing mortality are 1.36, 0.52 and 0.84 year–1, respectively. The length at first capture is 16.8cm. The exploitation rate indicates that the stock is overfished.  相似文献
