Welfare‐state retrenchment, transnational migration and the integration of immigrants have been accompanied by a growing ethnicization and racialization of welfare‐state politics in the United States and Germany. Although ethnicization and racialization may have other, deeper causes, immigration and its effects on social policies and provisions has become one of the main targets of dissent in the politics of West European and North American welfare states. While in the United States these developments have reinforced race‐class cleavages that have existed since the emergence of her welfare state, German welfare‐state politics has evolved from being primarily class‐based to class‐ethnicity divides in recent years. These cleavages have disadvan‐taged social democratic positions that tried to bridge protection of native workers and humanitarian obligations towards refugees but have supported nationalist‐populist trends. Cosmopolitan‐liberal positions face the formidable task of developing institutional mechanisms for handling conflicts in ethnically pluralist'welfare states.  相似文献

Steady-state and time-resolved absorption and fluorescence spectroscopic experiments have been carried out at room and cryogenic temperatures on aggregated and unaggregated monomeric and trimeric LHCII complexes isolated from spinach chloroplasts. Protein aggregation has been hypothesized to be one of the mechanistic factors controlling the dissipation of excess photo-excited state energy of chlorophyll during the process known as nonphotochemical quenching. The data obtained from the present experiments reveal the role of protein aggregation on the spectroscopic properties and dynamics of energy transfer and excited state deactivation of the protein-bound chlorophyll and carotenoid pigments.  相似文献

Background  

Cattail (Typha domingensis) has been spreading in phosphorus (P) enriched areas of the oligotrophic Florida Everglades at the expense of sawgrass (Cladium mariscus spp. jamaicense). Abundant evidence in the literature explains how the opportunistic features of Typha might lead to a complete dominance in P-enriched areas. Less clear is how Typha can grow and acquire P at extremely low P levels, which prevail in the unimpacted areas of the Everglades.  相似文献

We have studied the spatiotemporal distribution of coherent EEG links in 10 healthy right-handed subjects under the conditions of comfortable wakefulness with a straight position of the head and with the head voluntarily turned to fixed left and right positions. The computations were made separately in each standard range of EEG frequencies (Δ, θ, α, and β). The results were compared with the data obtained during studies of right-handed patients with right-sided (RS, n = 9) and left-sided (LS; n = 8) forms of spasmodic torticollis (ST). It has been shown that, upon a voluntary right turn of the head, the number of strong and moderate coherent EEG links in healthy subjects increases considerably more than upon a left turn. This may be accounted for by the greater functional importance of the control of the right part of the visual space as the area of manipulation of the dominant hand. In patients with RS ST, the number of intra- and interhemispheric strong and moderate coherent EEG links increased in all four frequency ranges, as compared to healthy subjects, upon the right turn of the head. In patients with LS ST, the spatial structure of the bioelectric potential field of the brain is less reorganized upon the voluntary left turn of the head as compared to the level of coherent EEG links in healthy subjects. Presumably, this phenomenon reflects a stronger insufficiency of neurophysiological mechanisms of postural control and compensation for its disturbances in patients with the LS form of ST, which results in a more severe disability as compared to patients with right-sided ST.  相似文献

Novobiocin (0.05 μg/ml) reduced the growth rate of cultures of Escherichiacoli strain DK6 by about a factor of 2. The lag in appearance of β-galactosidase-forming capacity was extended from 50 sec to 85 sec by the drug. This appeared to be the result of a reduced rate of nascent mRNA elongation.  相似文献

The regulatory mechanism of state transitions was studied in Chlamydomonas reinhardtii (C.r.) wild type (WT) as well as mutant strains deficient in the photosystem I (PSI) or the photosystem II (PSII) core. Time-resolved fluorescence measurements were obtained on instantly frozen cells incubated beforehand in the dark in aerobic or anaerobic conditions which leads to state 1 (S1) or state 2 (S2). WT data contains information on the light-harvesting complex (LHC) connected to PSI and PSII. The mutants' data contain information on either LHCII-LHCI-PSI or LHCII-PSII, plus information on LHC antennas devoid of a PS core. In a simultaneous analysis of the data from all strains under S1 or S2 conditions a unified model for the excited state dynamics at 77 K was created. This yielded the completely resolved LHCII-LHCI-PSI and LHCII-PSII dynamics and quantified the state transitions. In WT cells the fraction of light absorbed by LHCII connected to PSII decreases from 45% in S1 to 29% in S2, while it increases from 0% to 16% for LHCII connected to PSI. Thus (16/45 =) 36% of all LHCII is involved in the state transition. In the mutant strains deficient in the PSI core, the red most species peaking at 716 nm disappears completely, indicating that this far red Chl pigment is located in the PSI core. In the mutant strain deficient in the PSII core, red shifted species with maxima at 684 and 686 nm appear in the LHCII antenna. LHCII-684 is quenched and decays with a rate of (310 ps)? 1.  相似文献

A new method of assay for the A6-desaturation of linoleic acid was developed. This method, which uses HPLC for separation of the fatty acid substrate and product, exhibited a lower coefficient of variation (0.3%) than the reported TLC method (3.5%) [l], and avoided the step of methylation of the saponified fatty acid substrate and product. Using this new method of assay, the kinetics of the Δ6-desaturase in a multi-enzyme system were analysed. A number of factors that could have striking effects on desaturase kinetics were investigated, including the effect of (i) endogenous microsomal linoleic acid on total substrate concentration, and (ii) the pre-reaction catalysed by acyl-CoA synthetase and competing reactions catalysed by lysophospholipid acyltransferase and acyl-CoA hydrolase. Endogenous free linoleate in the hepatic microsomes was found to be 2.9 ± l.0 AM (0.5 mg microsomal protein/ml), which was comparable to added substrate concentrations (l.8 to 7.9 μM). The kinetics of the Δ6-desaturase were dissected from the kinetics of the above mentioned pre-reaction and competing reactions through a combination of experimental approaches and computer modeling. From computer modeling, a Km and Vmax of l.5 μM and 0.063 nmol/min were calculated for the Δ6-desaturase, compared to Km and Vmax of 10.7 μM and 0.08 nmol/min calculated directly from data uncorrected for endogenous substrate. It was concluded that lysophospholipid acyltransferase, acyl-CoA synthetase and endogenous linoleic acid significantly affect the kinetic measurements of hepatic microsomal Δ6-desaturase. These results have implications for kinetic analyses of all desaturases in microsomal systems.  相似文献

Mitochondrial Complex II (succinate:ubiquinone oxidoreductase) is purified in a partially inactivated state, which can be activated by removal of tightly bound oxaloacetate (E.B. Kearney, et al., Biochem. Biophys. Res. Commun. 49 1115–1121). We crystallized Complex II in the presence of oxaloacetate or with the endogenous inhibitor bound. The structure showed a ligand essentially identical to the “malate-like intermediate” found in Shewanella Flavocytochrome c crystallized with fumarate (P. Taylor, et al., Nat. Struct. Biol. 6 1108–1112) Crystallization of Complex II in the presence of excess fumarate also gave the malate-like intermediate or a mixture of that and fumarate at the active site. In order to more conveniently monitor the occupation state of the dicarboxylate site, we are developing a library of UV/Vis spectral effects induced by binding different ligands to the site. Treatment with fumarate results in rapid development of the fumarate difference spectrum and then a very slow conversion into a species spectrally similar to the OAA-liganded complex. Complex II is known to be capable of oxidizing malate to the enol form of oxaloacetate (Y.O. Belikova, et al., Biochim. Biophys. Acta 936 1–9). The observations above suggest it may also be capable of interconverting fumarate and malate. It may be useful for understanding the mechanism and regulation of the enzyme to identify the malate-like intermediate and its pathway of formation from oxaloacetate or fumarate.  相似文献

It has already been shown that the mutant Leu94Gly of horse cytochrome c exists in a molten globule (MG) state. We have carried out studies of reversible folding and unfolding induced by LiCl of this mutant at pH 6.0 and 25 °C by observing changes in the difference molar absorption coefficient at 402 nm, the mean residue ellipticity at 222 nm, and the difference mean residue ellipticity at 409 nm. This process is a three-state process when measured by these probes. The stable folding intermediate state has been characterized by far- and near-UV circular dichroism, tryptophan fluorescence, 8-anilino-1-naphthalenesulfonic acid binding, and dynamic light scattering measurements, which led us to conclude that the intermediate is a premolten globule (PMG). Analysis of the reversible unfolding transition curves for the stability of different states in terms of the Gibbs free energy change at pH 6.0 and 25 °C led us to conclude that the MG state is more stable than the PMG state by 5.4 ± 0.1 kcal mol−1, whereas the PMG state is more stable than the denatured (D) state by only 1.1 ± 0.1 kcal mol−1. A comparison of the conformational and thermodynamic properties of the LiCl-induced PMG state at pH 6.0 with those of the PMG state induced by NaCl at pH 2.0 suggests that a similar PMG state is obtained under both denaturing conditions. Differential scanning calorimetry measurements suggest that heat induces a reversible two-state transition between MG and D states.  相似文献

Biodiesel is a clean-burning fuel produced from grease, vegetable oils, or animal fats. Biodiesel is produced by transesterification of oils with short-chain alcohols or by the esterification of fatty acids. The transesterification reaction consists of transforming triglycerides into fatty acid alkyl esters, in the presence of an alcohol, such as methanol or ethanol, and a catalyst, such as an alkali or acid, with glycerol as a byproduct. Because of diminishing petroleum reserves and the deleterious environmental consequences of exhaust gases from petroleum diesel, biodiesel has attracted attention during the past few years as a renewable and environmentally friendly fuel. Since biodiesel is made entirely from vegetable oil or animal fats, it is renewable and biodegradable. The majority of biodiesel today is produced by alkali-catalyzed transesterification with methanol, which results in a relatively short reaction time. However, the vegetable oil and alcohol must be substantially anhydrous and have a low free fatty acid content, because the presence of water or free fatty acid or both promotes soap formation. In this article, we examine different biodiesel sources (edible and nonedible), virgin oil versus waste oil, algae-based biodiesel that is gaining increasing importance, role of different catalysts including enzyme catalysts, and the current state-of-the-art in biodiesel production. The cardiometabolic syndrome, associated with increased cardiovascular disease risk in the industrialized world, is estimated to affect one in four adults. Although the mechanisms linking obesity and cardiovascular disease remain unclear, research continues to unravel the molecular pathways behind this pandemic. Adipose tissue has emerged as a metabolically active participant in mediating vascular complications, serving as an active endocrine and paracrine organ secreting adipokines, which participate in diverse metabolic processes. Among these adipokines is adiponectin, which seems to possess antiatherogenic and anti-inflammatory effects and may be protective against cardiovascular disease development. The current review describes the pathophysiology of adiponectin in atherosclerotic disease.  相似文献

Through a comparison of privatization programs in two physics institutes, this article explores the ways in which scientists in the Siberian science city of Akademgorodok adapted to the low levels of state funding available to them in the 1990s. Scientists transformed structures that were available under socialism into hybrid state-private ventures. Rather than ‘freeing’ Russian science from its former dependence on the state, however, these changes have reconfigured, and in some cases even strengthened, the relationship between state power and the production of knowledge. Seeing ‘the state’ as it is constituted in Russian scientists' discourse challenges Western models of the autonomy of science.  相似文献

The autolysis of trypsin and α-chymotrypsin is accelerated in the presence of colloidal silica and glass surfaces. It is proposed that adsorption of the enzymes (favoured by electrostatic factors) results in a conformational change that renders the adsorbed enzyme more susceptible to proteolytic attack. Although the adsorbed enzymes are more susceptible to proteolysis, their activity towards low-molecular-weight substrates is not affected, indicating a relatively minor conformational change on adsorption. The rates of autolysis in solution (i.e. in `inert' vessels) are second-order for both trypsin and α -chymotrypsin, with rate constants of 13.0mol−1·dm3·s−1 for trypsin (in 50mm-NaCl at pH8.0 at 25°C) and 10.2mol−1·dm3·s−1 for α-chymotrypsin (in 0.1m-glycine at pH9.2 at 30°C). In glass vessels or in the presence of small areas of silica surface (as colloidal silica particles), the autolysis of both trypsin and α-chymotrypsin can show first-order kinetics. Under these conditions, saturation of the surface occurs and the fast surface proteolytic reaction controls the overall kinetic order. However, when greater areas of silica surface are present, saturation of the surface does not occur, and, since for a considerable portion of the adsorption isotherm the amount adsorbed is approximately proportional to the concentration in solution, second-order kinetics are again observed. A number of negatively charged macromolecules have been shown similarly to increase the rate of autolysis of trypsin: thus this effect, observed initially with glass and silica surfaces, is of more general occurrence when these enzymes adsorb on or interact with negatively charged surfaces and macromolecules. These observations explain the confusion in the literature with regard to the kinetics of autolysis of α-chymotrypsin, where first-order, second-order and intermediate kinetics have been reported. A further effect of glass surfaces and negatively charged macromolecules is to shift the pH–activity curve of trypsin to higher pH values, as a consequence of the effective decrease in pH in the `microenvironment' of the enzyme associated with the negatively charged surface or macromolecule.  相似文献

2′-Deoxy-5′-guanosinemonosphoric acid (B) reacts with cis-[Pt(NH3)2(OH2)2]2+ in two steps to form the cis-[Pt(NH3)2B2]y+ ion. In the first step 2′-d-5′- GMPH2 reacts some ten times faster than 5′-GMPH2 does. Rate constants, ΔH#, ΔS# and ΔV# are very similar for the two bases in the second reaction. It is proposed that the product in the first step contains no water and is cis-[Pt(NH3)2B]x+ in which the nucleobase is bidentate bonding through both N(7) of guanine and an oxygen atom of the phosphate group.  相似文献

Stretch induced activation and release induced deactivation of single glycerol-extracted insect flight muscle fibres were investigated. The results are interpreted to indicate that the muscle length controls the number of acting cross bridges, whereas their attachment-detachment kinetics in mainly determined by the state of strain of the cross bridges. It is concluded that the net detachment rate of the cross bridges is enhanced if the muscle is released thereby “unloading” the cross bridges. This behaviour of the unloaded cross bridge is a basic postulation of most of the molecular muscle contraction models.
  1. The delayed tension rise induced by stretches of different amplitudes could be restored to the level before the stretch by a release to the initial length.
  2. The delayed tension decrease induced by a release of moderate (up to δL=1.5% L i)amplitude is quantitatively restored within the delayed increase induced by the restretch to the initial length.
  3. Stiffness, which decreased during the delayed tension drop after release, is restored during a delayed stiffness increase effected by a restretch to the initial length.
  4. The rate and the extent of the stiffness drop after release increased with increasing amplitude of the release and with increasing temperature.
  5. After the deactivation, i.e., after tension and stiffness achieved a new steady level after the release, the attached cross bridges are already in the same state of strain as they were before the release. This finding is interpreted to indicate that within the deactivation phase all cross bridges attached prior the release are replaced by cross bridges attached after the release.
  6. The rate of tension and stiffness decay after release does not depend on the absolute muscle length but on the amplitude of the release which induced the deactivation.
