A theoretical analysis was performed to determine the number of fractions a multidisperse, immunomagnetically labeled cell population can be separated into based on the surface marker (antigen) density. A number of assumptions were made in this analysis: that there is a proportionality between the number of surface markers on the cell surface and the number of immunomagnetic labels bound; that this surface marker density is independent of the cell diameter; and that there is only the presence of magnetic and drag forces acting on the cell. Due to the normal distribution of cell diameters, a "randomizing" effect enters into the analysis, and an analogy between the "theoretical plate" analysis of distillation, adsorption, and chromatography can be made. Using the experimentally determined, normal distribution of cell diameters for human lymphocytes and a breast cancer cell line, and fluorescent activated cell screening data of specific surface marker distributions, examples of theoretical plate calculations were made and discussed.  相似文献

A rapid, simultaneous double-staining procedure using fluorescein diacetate (FDA) and propidium iodide (PI) is described for use in the determination of cell viability in cell suspension. Air-dried slide preparations can be made from the cell suspensions so that an accurate estimate of the viability of the cells in the original suspension can be made up to 1 week later. Viable cells fluoresce bright green, while nonviable cells are bright red. Furthermore, when FDA-PI staining is compared to trypan blue dye exclusion as a method to determine cell viability, FDA-PI is found to be more consistent over prolonged periods of exposure to the dyes. Therefore, double staining with FDA-PI is a rapid, convenient, and reliable method to determine cell viability.  相似文献

A possibility of tissue metaplasia (transformation of one cell type into another) during limb regeneration in lower vertebrates has been a matter of vivid arguments over the last decades. These discussions are rather irreconcilable in character mainly due to the lack of reliable cell markers which permit to follow all the stages of cell transformation during metaplasia. The final conclusions can be made only if any artifacts of cell labelling are excluded. Latest findings obtained using nuclear and cytoplasmic markers are presented which suggest that many data interpreted previously as a convincing proof of metaplasia may be a consequence of the involvement of nondifferentiated cells in regeneration. Molecular biological approaches are believed to be most promising for the solution of disputable problems of tissue metaplasia. However, recent findings about actin gene hypomethylation are insufficient for any final conclusions about the possibility of metaplasia during limb regeneration. The answer to many unsolved questions of developmental biology can be made only when combined use is made of modern methods of cell and molecular biology.  相似文献

During vertebrate gastrulation, a relatively limited number of blastodermal cells undergoes a stereotypical set of cellular movements that leads to formation of the three germ layers: ectoderm, mesoderm and endoderm. Gastrulation, therefore, provides a unique developmental system in which to study cell movements in vivo in a fairly simple cellular context. Recent advances have been made in elucidating the cellular and molecular mechanisms that underlie cell movements during zebrafish gastrulation. These findings can be compared with observations made in other model systems to identify potential general mechanisms of cell migration during development.  相似文献

The authors studied comparatively the endometrium and endometrosis cell cultures of 35 women before and (in part of the group) after the synthetic progestin treatment. Methods for cultivating the endometrium and endometriosis cells were developed. A normal endometrium cell culture was found to be made up by epithelioid cells, while the cell culture from the endometriosis focus exhibited mostly a mixed cell population. The analysis of the internal endometriosis cell cultures before and after the infecundin treatment failed to show any clear distinction in respect to the cell growth and morphology. Endometriosis cells were found to possess low mitotic activity which made the kariotype studies difficult. the data obtained are the result of the first effort to cultivate endometriosis cells.  相似文献

Cell motility is an essential biological action in the creation, operation and maintenance of our bodies. Developing mathematical models elucidating cell motility will greatly advance our understanding of this fundamental biological process. With accurate models it is possible to explore many permutations of the same event and concisely investigate their outcome. While great advancements have been made in experimental studies of cell motility, it now has somewhat fallen on mathematical models to taking a leading role in future developments. The obvious reason for this is the complexity of cell motility. Employing the processing power of today’s computers will give researches the ability to run complex biophysical and biochemical scenarios, without the inherent difficulty and time associated with in vitro investigations. Before any great advancement can be made, the basics of cell motility will have to be well-defined. Without this, complicated mathematical models will be hindered by their inherent conjecture. This review will look at current mathematical investigations of cell motility, explore the reasoning behind such work and conclude with how best to advance this interesting and challenging research area.  相似文献

The estimation of several cellular biophysical parameters must be made in order to mathematically predict optimal cryopreservation protocols. These parameters include total cell volume, osmotically inactive volume, cell surface area, and relative water and solute permeabilities. Recent attention has been paid to the determination of the osmotically inactive volume and, specifically, an argument was made suggesting that this volume was incorrectly determined in the literature [4]. Analysis of growth and division often involves measurements made on cell populations, which tend to average data. The value of single cell analysis needs to be appreciated, and models based on findings from single cells should be taken into greater consideration in our understanding of the way in which cell size and division are co-ordinated. Examples are given of some single cell analyses in mammalian cells, yeast and other microorganisms. There is also a short discussion on how far the results are in accord with simple models.  相似文献

Fish cell lines are relatively easy to culture and most have simple growth requirements that make cross contamination a potential problem. Cell line contamination is not an uncommon incident in laboratories handling more than one cell line and many reports have been made on cross contamination of mammalian cell lines. Although problems of misidentification and cross-contamination of fish cell lines have rarely been reported, these are issues of concern for cell culturists that can make scientific results and their reproducibility unreliable. Proper identification of cell lines is thus crucial and protocols for routine and rapid screening are preferred. Cytogenetic evaluation, DNA fingerprinting, microsatellite analysis and PCR methods have been published for inter-species identification of many cell lines, but discerning intra-species contamination has been challenging. More complex DNA fingerprinting and hybridization techniques coupled with isoenzyme analysis have been developed to discriminate intra-species contamination, however, these require complex and time consuming procedures to enable cell identification thus are difficult to apply for routine use. A simple proteomic approach has been made to identify several fish cell lines derived from tissues of the same or differing species. Protein expression signatures (PES) of the evaluated fish cell lines have been developed using 2-DE and image analysis. A higher degree of concordance was seen among cell lines derived from rainbow trout, than from other fish species. Similar concordance was seen in cells derived from the same tissues than from other tissues within the same species. These profiles have been saved in an electronic databank and could be made available to be used for discerning the origins of the various cell lines evaluated. This proteomic approach could thus serve as an additional, valuable and reliable technique for the identification of fish cell lines.  相似文献

The turgor pressure in cells of chickpea (Cicer arietinum L.) and faba bean (Vicia faba L.) seed coats was measured with a pressure probe. Measurements were made under in situ conditions by removing a section of wall from a pod, which remained attached to the plant, and exposing the intact seed. If the pod wall was removed and the turgor measurements made under ambient laboratory conditions of 50% to 70% relative humidity (RH), cell turgor pressure declined over time, typically reaching 0 MPa. If the pod wall was removed and the turgor measurements made under conditions of 100% RH, however, cell turgor pressure was stable over time, relatively uniform within the seed coat tissue, and was found to be 0.1-0.3 MPa for chickpea, and 0.1-0.2 MPa for faba bean. In both species there was a marked decline in cell turgor, beginning within about 60 s, when humidification was discontinued. The decline in cell turgor occurred regardless of the depth of the cell within the seed coat tissue, and this decline could be stopped, but not entirely reversed, when humidification was restored. An increase in cell turgor could also be caused by wetting of the seed. These responses indicate that a very rapid water exchange can occur within the seed coat tissue in situ. The rapid and, in some cases, relatively permanent loss of seed coat cell turgor in the absence of humidification raises serious concerns regarding desiccation artefacts which may be involved in the empty seed coat technique, often used to study seed carbon and water relations in grain legumes.  相似文献

These systems rely on conventional manufacturing techniques using organisms that are the result of either cell fusions or genetic engineering. A wealth of new techniques has allowed improvements and developments to be made in culture medium composition, cell modification, and bioreactor design and operation. The cytologic features of clear cell hepatocellular carcinoma are described. This tumor may cause a diagnostic dilemma since it resembles other clear cell tumors originating in the adrenals, kidneys and ovaries. However, clear cell hepatocellular carcinoma possesses some characteristic features that permit a cytologic diagnosis to be made by fine needle aspiration, thus contributing to proper management.  相似文献

Sickle cell disease is observed to occur in significantly high frequencies amongst the tribes of India. It has surged to the fore as an important public health problem among tribal groups, which needs serious attention. This paper presents the distribution of this abnormal genetic problem among scheduled tribes of India, in general and among those of Andhra Pradesh and Orissa states, in detail. Though the prevalence of sickle cell trait is high, the sickle cell disease cases are found to be very low, since all these surveys are made among adults. Most of the sickle cell disease cases might have expired during their early states. Hence, it is attempted to estimate the expected frequencies of disease cases from HbS gene frequencies. Estimations were also made by considering higher levels of inbreeding among these populations.  相似文献

The dominant selectable gene, Ecogpt, has been introduced, by the calcium phosphate precipitation technique, into normal human fibroblasts, along with the SV40 early region genes. In one transfectant clone, integration of these sequences into human chromosome 17 was demonstrated by the construction of human-mouse somatic cell hybrids, selected for by growth in medium containing mycophenolic acid and xanthine. A whole cell hybrid, made between the human transfectant and a mouse L cell, was used as donor of the Ecogpt-carrying human chromosome 17 to 'tribrids' growing in suspension, made by whole cell fusion between a mouse thymoma cell line, and to microcell hybrids made with a mouse teratocarcinoma cell line. Two tribrids contained karyotypically normal human chromosomes 17 and a small number of other human chromosomes, while a third tribrid had a portion of the long arm of chromosome 17 translocated to mouse as its only human genetic material. Two independent microcell hybrids contained a normal chromosome 17 and no other human chromosome on a mouse teratocarcinoma background. These experiments demonstrate the ability to construct human-mouse somatic cell hybrids using a dominant selection system. By applying this approach it should be possible to select for a wide range of different human chromosomes in whole cell and microcell hybrids. In particular, transfer of single human chromosomes to mouse teratocarcinoma cells will allow examination of developmentally regulated human gene sequences after differentiation of such hybrids.  相似文献

A survey of literature is made which indicates that various non-thermal influences may significantly affect cell thermostability in both directions. Therefore, special precautions should be taken to be sure that the observed changes in cell thermostability really represent strong negative correlation between the thermostability and the resting membrane potential levels in cells. This correlation implies that changes in cell thermostability, irrespectively of their origin (including those which account for thermal adaptation), are due to changes in the ionic asymmetry and permeability of cells.  相似文献

Summary An ultrastructural study has been made of a new cell type different from five known cell types in the anterior pituitary of normal mice. This new cell type is characterized by the special morphology of secretory granules. The immature form of the granules with high electron density in and around the Golgi area becomes vesiculated in the cell periphery. With glutaraldehyde-osmium fixation, however, the content of such vesiculated granules acquires electron density.In attempts to certify the independence of this cell type not related to the others and to elucidate its physiological function, a series of experiments has been made.After adrenalectomy the cell type exhibits a characteristic response represented by hypertrophy of the Golgi apparatus and prominent other cell organelles. Under the same condition the FSH-producing cell to be differentiated from the new cell type does not undergo any cytological changes. Instead, all FSH-producing cells show typical alterations at 100th day after orchidectomy, when the cell in question remains intact. Adrenalectomy following 100 day-orchidectomy causes a stimulative effect upon the new cell type. Furthermore, administration of hydrocortisone results in a hypofunctional state of this cell type.All these findings indicate that the new cell type identified in the present study is the site of ACTH production.  相似文献

A short clarifying view of how semiconductor quantum dots (QDs) can be made visible in tissue sections by autometallographic (AMG) silver enhancement and how the introduction of AMG enhanceable gold nanoparticles into isolated cells can be used to follow the fate of these marked cells in organisms and cell cultures. BACKGROUND: Metastasis of renal cell carcinoma to the vagina is rare, although it may be the first evidence of the existence of the primary tumor. CASE: A metastatic deposit of renal cell carcinoma in the vagina was diagnosed by cytology as clear cell adenocarcinoma, which was confirmed by biopsy. Radiographic and ultrasound examinations confirmed the renal site of origin, which was corroborated by immunohistochemistry of the biopsy specimen. CONCLUSION: When a cytologic diagnosis of vaginal clear cell adenocarcinoma is made, metastasis of renal cell carcinoma should be considered in the differential diagnosis.  相似文献

The p53 protein, like any other protein, cannot be made in the cell without RNA. And even once made, the p53 protein will be more rapidly degraded without the p53 RNA. Furthermore, the p53 RNA helps deciding which p53 isoform should be produced and under which cell conditions. Mutant p53 mRNA codes for an unstable and inactive protein. These matters are discussed in this article as well as the recent reports on p53 RNA mutations, interacting-proteins, 3′ processing and 5′–3′ loop.  相似文献
