Eucalyptus hemicellulosic hydrolysate used for xylitol production by Candida guilliermondii FTI20037 was previously treated either with ion-exchange resins or with activated charcoal adsorption combined with pH adjustment, in order that acetic acid, furfural and hydroxymethylfurfural could be removed. The best results for xylitol yield factor (0.76 g/g) and volumetric productivity (0.68 g/(l h) were attained when a three-fold concentrated hydrolysate was treated with ion-exchange resins. Using activated charcoal combined with pH adjustment for treating a three-fold concentrated hydrolysate resulted in a xylitol yield factor of 0.40 g/g and a volumetric productivity of 0.30 g/(l h). This same treatment applied to a six-fold concentrated hydrolysate resulted in a xylitol yield factor of 0.66 g/g and a volumetric productivity of 0.50 g/(l h).  相似文献

Candida guilliermondii FTI 20037 cells were entrapped in Ca-alginate beads and used for xylitol production from sugarcane bagasse hemicellulosic hydrolysate in a stirred tank reactor (STR). Screening design and response surface methodologies were used to determine adequate cultivation conditions for this fermentation system. Quadratic models were fitted to the experimental data by regression analysis, considering the yield (YP/S) and the productivity (QP) of the xylose-to-xylitol bioconversion as dependent variables. Using a five-fold concentrated hydrolysate, air flowrate of 1.30 l/min, agitation speed of 300 rpm, initial cell concentration of 1.4 g/l and value 6.0 for the initial pH of the fermentation medium resulted in a xylitol production of 47.5 g/l after 120 h of fermentation, corresponding to a YP/S of 0.81 g/g and to a QP of 0.40 g/l h.  相似文献

To increase the NAD(P)H-dependent xylitol production in recombinant Saccharomyces cerevisiae harboring the xylose reductase gene from Pichia stipitis, the activity of glucose 6-phosphate dehydrogenase (G6PDH) encoded by the ZWF1 gene was amplified to increase the metabolic flux toward the pentose phosphate pathway and NADPH regeneration. Compared with the control strain, the specific G6PDH activity was enhanced approximately 6.0-fold by overexpression of the ZWF1 gene. Amplification in the G6PDH activity clearly improved the NAD(P)H-dependent xylitol production in the recombinant S. cerevisiae strain. With the aid of an elevated G6PDH level, maximum xylitol concentration of 86 g/l was achieved with productivity of 2.0 g/l h in the glucose-limited fed-batch cultivation, corresponding to 25% improvement in volumetric xylitol productivity compared with the recombinant S. cerevisiae strain containing the xylose reductase gene only.  相似文献

固定在多孔聚氨酯载体中的热带假丝酵母(Candida tropicalis), 可有效地利用玉米芯半纤维素水解液生产木糖醇。在摇瓶条件下, 采用分批发酵方式, 确立了适宜的发酵工艺参数为: 接种量7%, 聚氨酯加入量1.0 g/100 mL, 温度30°C, 初始pH值6.0, 分段改变摇床转速进行溶氧调节, 其中0~24 h 为200 r/min; 24 h~46 h为140 r/min。聚氨酯固定化提高了菌体对发酵抑制物的耐受力, 固定化细胞密度高, 发酵性能稳定, 发酵产率和体积生产速率都有所提高。水解液未经脱色与离子交换便可转化成木糖醇, 大幅降低了成本, 显示了良好的应用前景。固定化细胞连续重复进行12批次21 d的发酵, 木糖醇得率平均为67.6%, 体积生产速率平均为1.92 g/(L·h)。  相似文献

A thermotolerant yeast capable of fermenting xylose to xylitol at 40°C was isolated and identified as a strain of Debaryomyces hansenii by ITS sequencing. This paper reports the production of xylitol from D-xylose and sugarcane bagasse hemicellulose by free and Ca-alginate immobilized cells of D. hansenii. The efficiency of free and immobilized cells were compared for xylitol production from D-xylose and hemicellulose in batch culture at 40°C. The maximum xylitol produced by free cells was 68.6 g/L from 100 g/L of xylose, with a yield of 0.76 g/g and volumetric productivity 0.44 g/L/h. The yield of xylitol and volumetric productivity were 0.69 g/g and 0.28 g/L/h respectively from hemicellulosic hydrolysate of sugarcane bagasse after detoxification with activated charcoal and ion exchange resins. The Ca-alginate immobilized D. hansenii cells produced 73.8 g of xylitol from 100 g/L of xylose with a yield of 0.82 g/g and volumetric productivity of 0.46 g/L/h and were reused for five batches with steady bioconversion rates and yields.  相似文献

Xylitol is a well-known sugar substitute with low-calorie and anti-cariogenic characteristics. An effort of biological production of xylitol from xylose was made in repeated fed-batch and cell-recycle fermentations of recombinant Saccharomyces cerevisiae BJ3505/δXR harboring the xylose reductase gene from Pichia stipitis. Batch fermentation with 20 g/l xylose and 18 g/l glucose resulted in 9.52 g/l dry cell mass, 20.1 g/l xylitol concentration and approximately 100% conversion yield. Repeated fed-batch operation to remove 10% of culture broth and to supplement an equal volume of 200 g/l xylose was designed to improve xylitol production. In spite of a sudden drop of cell concentration, an increase in dry cell mass led to high accumulation of xylitol at 48.7 g/l. To overcome loss of xylitol-producing biocatalysts in repeated fed-batch fermentation, cell-recycle equipment of hollow fiber membrane was implemented into a xylitol production system. Cell-recycle operation maintained concentration of the recombinant cells high inside a bioreactor. Final dry cell mass of 22.0 g/l, 116 g/l xylitol concentration, 2.34 g/l h overall xylitol productivity were obtained in cell-recycle fermentation supplemented with xylose and yeast extract solution, which were equivalent to 2.3-, 5.8- and 3.8-fold increases compared with the corresponding values of batch-type xylitol production parameters.  相似文献

A variety of raw materials have been used in fermentation process. This study shows the use of rice straw hemicellulosic hydrolysate, as the only source of nutrient, to produce high added-value products. In the present work, the activity of the enzymes xylose reductase (XR); xylitol dehydrogenase (XD); and glucose-6-phosphate dehydrogenase (G6PD) during cultivation of Candida guilliermondii on rice straw hemicellulosic hydrolysate was measured and correlated with xylitol production under different pH values (around 4.5 and 7.5) and initial xylose concentration (around 30 and 70 g l(-1)). Independent of the pH value and xylose concentration evaluated, the title of XD remained constant. On the other hand, the volumetric activity of G6PD increased whereas the level of XR decreased when the initial xylose concentration was increased from 30 to 70 g l(-1). The highest values of xylitol productivity (Q (P) approximately 0.40 g l(-1)) and yield factor (Y (P/S) approximately 0.60 g g(-1)) were reached at highest G6PD/XR ratio and lowest XR/XD ratio. These results suggest that NADPH concentrations influence the formation of xylitol more than the activity ratios of the enzymes XR and XD. Thus, an optimal rate between G6PD and XR must be reached in order to optimize the xylitol production.  相似文献

The effect of dilution rate on the production of lactic acid from whey permeate by Lactobacillus helveticus has been investigated. In the first chemostat of a two-stage system, total conversion (98.1%) and maximum lactic acid concentration (43.7 g l−1) were obtained at a dilution rate (DItot) of 0.06 h−1. Maximum volumetric productivities of lactic acid (8.27 g l−1 h−1) and biomass (1.90 g l−1 h−1) occurred at DItot of 0.40 h−1. The fraction of -lactate in the product was found to increase with dilution rate and reached a maximum of 66% at the same dilution rate. The maximum specific growth ratemax) on this medium was 0.7 h−1. A YATP (max) value of 22.4 g dry weight (mol ATP)−1 and a maintenance coefficient of 8.0 mmol ATP (g dry weight h)−1 were determined. The second stage, in series with the first, confirmed these results and further showed that the total residence time could be reduced by 50%, compared with a single chemostat for the same nearly complete level of substrate conversion.  相似文献

The effect of inoculum level on xylitol production byCandida guilliermondii was evaluated in a rice straw hemicellulose hydrolysate. High initial cell density did not show a positive effect in this bioconversion since increasing the initial cell density from 0.67 g L–1 to 2.41 g L–1 decreased both the rate of xylose utilization and xylitol accumulation. The maximum xylitol yield (0.71 g g–1) and volumetric productivity (0.56 g L–1 h–1) were reached with an inoculum level of 0.9 g L–1. These results show that under appropriate inoculum conditions rice straw hemicellulose hydrolysate can be converted into xylitol by the yeastC. guilliermondii with efficiency values as high as 77% of the theoretical maximum.  相似文献

A detoxification method using activated charcoal with concentrated rice straw hemicellulosic hydrolysate improved the conversion of xylose to xylitol by the yeast Candida guilliermondii by 22%. This was achieved when the hydrolysate:charcoal ratio was 40 g g–1, resulting in removal of 27% of phenolic compounds. Under this condition, the xylitol yield factor (0.72 g g–1) and volumetric productivity (0.61 g l–1 h–1) were close to those attained in a semi-defined medium simulating hydrolysate sugars.  相似文献

A brewery spent-grain hemicellulosic hydrolysate was used for xylitol production by Debaryomyces hansenii. Addition of 6 g yeast extract/l increased the xylitol yield to 0.57 g/g, and productivity to 0.51 g/l h that were, respectively, 1.4 -and 1.8-times higher than the values obtained with non-supplemented hydrolysate. When corn steep liquor was combined with 3 g yeast extract/l, the highest xylitol yield, 0.58 g/g, was obtained with a similar productivity.  相似文献

The effect of overall oxygen mass transfer coefficient (kLa) on the conversion of xylose to xylitol by Candida guilliermondii FTI 20037 was investigated in batch experiments. Rice straw hemicellulose hydrolysate obtained by acid hydrolysis was employed as a xylose-rich medium. The results showed that this bioconversion strongly depended on the aeration rate. The maximum volumetric productivity (0.52 g/l hу) and the highest xylitol yield (0.73 g/g) were achieved at an overall oxygen mass transfer coefficient of 15 hу. Under these conditions 80% efficiency in relation to theoretical yield was attained.  相似文献

The production of xylitol from concentrated synthetic xylose solutions (S(o) = 130-135 g/L) by Debaryomyces hansenii was investigated at different pH and temperature values. At optimum starting pH (pH(o) = 5.5), T = 24 degrees C, and relatively low starting biomass levels (0.5-0.6 g(x)/L), 88% of xylose was utilized for xylitol production, the rest being preferentially fermented to ethanol (10%). Under these conditions, nearly 70% of initial carbon was recovered as xylitol, corresponding to final xylitol concentration of 91.9 g(P)/L, product yield on substrate of 0.81 g(P)/g(S), and maximum volumetric and specific productivities of 1.86 g(P)/L x h and 1.43 g(P)/g(x) x h, respectively. At higher and lower pH(o) values, respiration also became important, consuming up to 32% of xylose, while negligible amounts were utilized for cell growth (0.8-1.8%). The same approach extended to the effect of temperature on the metabolism of this yeast at pH(o) = 5.5 and higher biomass levels (1.4-3.0 g(x)/L) revealed that, at temperatures ranging from 32-37 degrees C, xylose was nearly completely consumed to produce xylitol, reaching a maximum volumetric productivity of 4.67 g(P)/L x h at 35 degrees C. Similarly, both respiration and ethanol fermentation became significant either at higher or at lower temperatures. Finally, to elucidate the kinetic mechanisms of both xylitol production and thermal inactivation of the system, the related thermodynamic parameters were estimated from the experimental data with the Arrhenius model: activation enthalpy and entropy were 57.7 kJ/mol and -0.152 kJ/mol x K for xylitol production and 187.3 kJ/mol and 0.054 kJ/mol x K for thermal inactivation, respectively.  相似文献

Summary Xylose-to-xylitol batch bioconversions from wheat straw hemicellulosic hydrolysate were carried out in Erlenmeyer flasks in order to assess the influence of medium composition (hydrolysate concentration, supplementation with ammonium sulphate, calcium chloride and rice bran extract, and initial pH) on xylitol production, productivity and yield. By using the screening design and the response surface methodologies, the statistically significant variables influencing the bioconversion were selected and linear models were fitted to the experimental data. According to the results, the best conditions to perform the bioconversion consisted in using a threefold concentrated hydrolysate supplemented with ammonium sulphate (1.0 g/l) and rice bran extract (5.0 g/l), whose pH was adjusted to 6.0 prior to inoculation. Under these conditions, a xylitol production of 24.17 g/l was observed after 72 h of fermentation, resulting in a productivity of 0.34 g/l h and in a bioconversion yield of 0.49 g/g.  相似文献

Rice straw was hydrolyzed into a mixture of sugars using diluted H(2)SO(4). During hydrolysis, a variety of inhibitors was also produced, including acetic acid, furfural, hydroxymethylfurfural, and lignin degradation products (several aromatic and phenolic compounds). To reduce the toxic compounds concentration in the hydrolyzate and to improve the xylitol yield and volumetric productivity, rice straw hemicellulosic hydrolyzate was treated with activated charcoal under different pH values, stirring rates, contact times, and temperatures, employing a 2(4) full-factorial design. Fermentative assays were conducted with treated hydrolyzates containing 90 g/L xylose. The results indicated that temperature, pH, and stirring rate strongly influenced the hydrolyzate treatment, temperature and pH interfering with all of the responses analyzed (removal of color and lignin degradation products, xylitol yield factor, and volumetric productivity). The combination of pH 2.0, 150 rpm, 45 degrees C, and 60 min was considered an optimal condition, providing significant removal rates of color (48.9%) and lignin degradation products (25.8%), as well as a xylitol production of 66 g/L, a volumetric productivity of 0.57 g/L.h, and a yield factor of 0.72 g/g.  相似文献

The operational conditions for xylitol production by fermentation of sugarcane bagasse hydrolysate in a fluidized bed reactor with cells immobilized on zeolite were evaluated. Fermentations were carried out under different conditions of air flowrate (0.0125-0.0375 vvm), zeolite mass (100-200 g), initial pH (4-6), and xylose concentration (40-60 g/L), according to a 2(4) full factorial design. The air flowrate increase resulted in a metabolic deviation from product to biomass formation. On the other hand, the pH increase favored both the xylitol yield (Y(P/S)) and volumetric productivity (Q(P)), and the xylose concentration increase positively influenced the xylitol concentration. The best operational conditions evaluated were based on the use of an air flowrate of 0.0125 vvm, 100 g of zeolite, pH 6, and xylose concentration of 60 g/L. Under these conditions, 38.5 g/L of xylitol were obtained, with a Y(P/S) of 0.72 g/g, Q(P) of 0.32 g/L.h, and cell retention of 25.9%.  相似文献

In this study we used the yeast Candida guilliermondii FTI 20037 immobilized by entrapment in Ca-alginate beads (2.5-3 mm diameter) for xylitol production from concentrated sugarcane bagasse hemicellulosic hydrolysate in a repeated batch system. The fermentation runs were carried out in 125- and 250-ml Erlenmeyer flasks placed in an orbital shaker at 30 degrees C and 200 rpm during 72 h, keeping constant the proportion between work volume and flask total volume. According to the results, cell viability was substantially high (98%) in all fermentative cycles. The values of parameters xylitol yield and volumetric productivity increased significantly with the reutilization of the immobilized biocatalysts. The highest values of xylitol final concentration (11.05 g/l), yield factor (0.47 g/g) and volumetric productivity (0.22 g/lh) were obtained in 250-ml Erlenmeyer flasks containing 80 ml of medium plus 20 ml of immobilized biocatalysts. The support used in this study (Ca-alginate) presented stability in the experimental conditions used. The results show that the use of immobilized cells is a promising approach for increasing the xylitol production rates.  相似文献

The xylose conversion into by Candida guilliermondii was evaluated in sugar cane bagasse hemicellulosic hydrolysate. The effect of air flow rates of 0.4, 0.6 and 0.8 vvm cn xylitol formation was studied. In addition, inoculum previously adapted to the hydrolysate was also tested in the fermentation carried out at 0.6 vvm. The results showed that xylitol production depends markedly on the aeration rate and on the previous adaptation of the yeast to the hydrolysate. When the highest productivity of xylitol was 0.39 g/l × h. However, during the fermentation carried out at an air flow rate of 0.6 vvm with adapted inoculum, the productivity increased to 0.65 g/l × h. Furthermore, the adapted cells performed quite well in the presencel of acetic concentrations of about 4.5 g/l in the medium.  相似文献

This work evaluated the xylitol production from sugarcane bagasse hemicellulosic hydrolysate in a bubble column bioreactor using cells of the yeast Candida guilliermondii immobilized in calcium-alginate. The fermentation runs were performed according to a 22 full factorial design with three replicates at the center point in order to determine the effect of the variables: aeration rate (0.66–1.33 vvm) and immobilized system concentration (20–40% v/v), on the efficiency of xylose-to-xylitol conversion and on the xylitol volumetric productivity. The results indicated a significant influence of both variables on xylitol production. The highest conversion efficiency (41%) was attained using 1.33 vvm aeration rate and 40% immobilized system. Under these conditions, the volumetric productivity was 0.21 g l−1 h−1.  相似文献

土壤呼吸的温度敏感性(Q10)是陆地碳循环与气候系统间相互作用的关键参数。尽管已有大量关于不同类型森林Q10季节和年际变化规律的研究, 但是对Q10在区域尺度的空间变异特征及其影响因素仍认识不足, 已有结果缺乏一致结论。该研究通过整合已发表论文, 构建了中国森林生态系统年尺度Q10数据集, 共包含399条记录、5种森林类型(落叶阔叶林(DBF)、落叶针叶林(DNF)、常绿阔叶林(EBF)、常绿针叶林(ENF)、混交林(MF))。分析了不同森林类型Q10的空间变异特征及其与地理、气候和土壤因素的关系。结果显示, 1) Q10介于1.09到6.24之间, 平均值(±标准误差)为2.37 (± 0.04), 且在不同森林类型之间无显著差异; 2)当考虑所有森林类型时, Q10随纬度、海拔、土壤有机碳含量(SOC)和土壤全氮含量(TN)的增加而增大, 随经度、年平均气温(MAT)、平均年降水量(MAP)的增加而减小。气候(MATMAP)和土壤(SOCTN)因素间存在相互作用, 共同解释了33%的Q10空间变异, 其中MATSOCQ10空间变异的主要驱动因素; 3)不同类型森林Q10对气候和土壤因素的响应存在差异。在DNF中Q10MAP的增加而减小, 而其他类型森林中Q10MAP无显著相关性; 在EBF、DBF、ENF中Q10TN的增加而增大, 但Q10TN的敏感性在EBF中最高, 在ENF中最低。这些结果表明, 尽管Q10有一定的集中分布趋势, 但仍有较大范围的空间变异, 在进行碳收支估算时应注意尺度问题。Q10的主要驱动因素和Q10对环境因素的响应随森林类型而变化, 在气候变化情景下, 不同森林类型间Q10可能发生分异。因此, 未来的碳循环-气候模型还应考虑不同类型森林碳循环关键参数对气候变化的响应差异。  相似文献
