0-4℃低温处理黄豆、绿豆、红小豆3种作物幼苗,它们的生长均明显受抑制,其中红小豆和绿豆从处理的第一天起生长就完全受抑;黄豆幼苗生长虽也受抑,但仍有缓慢生长现象。比较3种作物根系中POD、COD、ATPase同工酶,其中COD未观察到有明显变化,但POD和ATPase同工酶变化明显,0-4℃低温下3种幼苗生长受抑与根系及同工酶的变化呈一定的相关性。  相似文献

过去所得的实验表明,植物中ATPase活性变化与耐冷性有关。春小麦和番茄对低温是非常敏感的,当将这两种植物的幼苗在低温下培养时,质膜ATPase活性下降甚至消失。相反,在冬小麦中,当幼苗在低温下处理时,质膜ATPase活性增加.为了研究ATPase同工酶变化与植物耐冷性之间的关系,以小麦等四种植物为材料,在0-1℃低温下处理2天、2周和4周,然后检测幼苗根系内的ATPase同工酶,结果发现,在0-1℃培养时,幼苗根系内ATPase同工酶谱带减少,ATPase同工酶谱带变化与植物幼苗耐冷性呈一定相关性。ATPase同工酶谱带变化可能是受冷害所致。  相似文献

研究外源ABA对低温胁迫的水稻幼苗酯酶同工酶的影响。结果表明,水稻幼苗经7℃-10℃低温处理6d,长势明显不降,与其相应的酯酶同工酶也发生变化。外施ABA提高了水稻幼苗的抗寒能力,同样在酯酶同工酶酶谱上也有相应改变。  相似文献

本研究对乌鳢9种组织内的LDH、MDH和ATPase同工酶作了分析,结果表明,乌鳢各组织内的LDH、MDH有明显的组织特异性,LDH由两个基因编码。骨骼肌中的s-MDH也有两个基因编码。ATPase同工酶存在于乌鳢的多种组织中,其基因编码数有待进一步探讨。  相似文献

探讨了24-表油菜素内酯(EBR)对低氧胁迫黄瓜幼苗根系有氧呼吸同工酶表达的影响.结果表明:低氧胁迫增强了异柠檬酸脱氢酶(IDH)、琥珀酸脱氢酶(SDH)、苹果酸脱氢酶(MDH)及苹果酸酶(ME)同工酶的表达,并产生了一些新的条带;低氧下施用10-3 mg·L-1的外源EBR处理后6、9d时IDH、MDH同工酶的表达分别比单纯低氧处理提高了52.8%、13.6%及39.1%、11.3%,ME同工酶的表达在处理3d时比单纯低氧处理提高了11.6%,SDH同工酶表达6、9d时则分别比单纯低氧处理下降了42.9%和36.1%.可见,低氧胁迫下营养液添加EBR可调节黄瓜根系IDH、SDH、MDH及ME同工酶的表达,进而有利于缓解低氧胁迫对黄瓜幼苗根系的伤害.  相似文献

以耐冷性存在明显差异的栽培黄瓜‘长春密刺’和‘北京截头’为材料,研究了冷胁迫下C a2 、钙离子螯合剂(EGTA)和钙调素拮抗剂(CPZ)对低温胁迫黄瓜幼苗叶片3种抗氧化酶活性及POD同工酶的影响.对保护酶活性的研究结果表明:C aC l2、EGTA和CPZ处理均能引起3种抗氧化酶活性变化,但变化幅度因材料而异.胁迫处理中C a2 能提高2种材料的叶片抗氧化酶活性,减少其在低温逆境中的下降幅度,而EGTA和CPZ处理降低了2种材料的抗氧化酶活性,加大了低温逆境中的下降幅度.POD同工酶研究发现,C aC l2延缓冷胁迫中同工酶条带的丢失,EGTA和CPZ处理则使同工酶条带丢失提前,但材料不同对酶条带丢失时间也有影响.  相似文献

以'新泰密刺'黄瓜为材料,采用营养液栽培方法,对根际低氧胁迫下黄瓜幼苗根系SOD、POD和CAT同工酶进行了分析.结果显示,与对照相比,单纯低氧胁迫处理的黄瓜幼苗根系SOD和CAT同工酶活性先降低再升高, 而POD同工酶活性则持续升高;营养液增施4 mmol·L-1 CaCl2明显缓解了低氧胁迫对黄瓜植株的伤害, 其SOD、POD和CAT同工酶活性接近对照水平;与单纯低氧胁迫相比,营养液增施50 μmol·L-1 LaCl3显著抑制了幼苗根系SOD、POD和CAT同工酶活性的升高;营养液增施20 μmol·L-1三氟拉嗪(TFP)引起植株根系SOD、POD和CAT同工酶表达量的剧烈波动,随胁迫时间的延长SOD和CAT同工酶均先迅速升高后又迅速降低,而POD同工酶活性则迅速降低.研究表明,外源Ca2+增加了Ca2+向黄瓜植株体内的运输,促进了低氧逆境胁迫信号向体内的传递,提高了根系保护酶的表达量及其活性氧清除水平,从而增强了黄瓜植株耐低氧胁迫的能力.  相似文献

将草地早熟禾(Poa pratensis L.)巴润(baron)干种子分别进行蒸馏水和硼酸处理后利用返回式卫星搭载,以未搭载干种子作为对照.对不同处理植株过氧化物同工酶(POD)和酯酶同工酶(EST)酶谱进行分析.结果表明,空间条件处理引起部分草地早熟禾植株POD和EST的差异,在酶带位置、数量、强弱等方面与对照相比出现了一定的变化,说明空间条件作用后草地早熟禾种子后代植株产生了一定变异.  相似文献

Roots of wheat seedlings (Triticum aestivum L. cv. Weibulls Starke) were cooled (+1°C) for 24 h while the shoots were kept at 25°C. The treatment induced an increased water deficit in the leaves. Fresh weight, dry weight, and the uptake and distribution of potassium and calcium were measured before and after cooling. Growth, measured both as fresh weight and dry weight increase, was reduced during the cold treatment. Afterwards (at 20°C), growth recovered to nearly pre-stress rates. Analysis of the potassium fluxes in and out of the roots by 86Rb techniques showed that influx, and to a lesser extent efflux, were inhibited at low temperature. The result was a net potassium uptake rate of one-third that of unstressed plants. After the cooling period the potassium influx increased to the rate of control plants. The potassium efflux increased to one and one-half times the rate of unstressed wheat so that net uptake was negative. The increase in potassium efflux was explained by a higher permeability of the root cell membranes after cooling. The net uptake of calcium was reduced to one-third by root cooling. Contrary to potassium uptake, calcium uptake increased under post-stress conditions, partly due to a low efflux rate. During root cooling there was a redistribution of dry matter from the leaves down towards the lower part of the shoot. Afterwards the original distribution of dry matter was reestablished. The net flow of potassium and calcium followed a similar pattern as dry matter, suggesting a growth-regulated flow.  相似文献

采用电镜观察和聚丙烯酰胺垂直板电泳法,研究了GB对低温胁迫下黄瓜幼苗叶绿体超微结构和SOD、POD同工酶的影响。结果表明,低温胁迫下,GB可使黄瓜幼苗叶绿体膜结构保持完好,而对照的叶绿体基粒破坏严重;与对照相比,GB处理对SOD、POD同工酶谱带数目均无影响,但POD同工酶的P2(Rf0.24)、P3(Rf0.30)、P4(Rf0.36)酶带的活性增加,SOD同工酶的S1(Rf0.14)、S5(Rf0.58)酶带活性较高,保持了黄瓜幼苗体内相对较高的抗氧化酶活性。GB可保护膜的完整性和稳定性,从而提高黄瓜幼苗的抗冷性。  相似文献

NaCl Effects on Root Plasma Membrane ATPase of Salt Tolerant Wheat   总被引:1,自引:0,他引:1  
Wheat seedlings of a salt tolerant cultivar were grown hydroponically in presence and absence of 100 mM NaCl. Roots were harvested, and the plasma membrane (PM) fraction was purified. PM ATPase required a divalent cations for activity (Mg > Mn > Ca > Co > Zn > Ni > Cu), and it was further stimulated by monovalent cations (K > Rb > NH4 > Li > choline > Cs). The pH optima were 6.0 and 5.6 in absence and presence of 25 mM KCl, respectively. The enzyme was sensitive to vanadate and DCCD but insensitive to azide, oligomycine and nitrate. The enzyme displayed a high preference for ATP but was also able to hydrolyze other nucleotide tri- and diphosphates. The enzyme activity showed a simple Michaelis-Menten kinetics for the substrate Mg2+-ATP in both control and salt exposed roots. The polypeptide patterns of control and salt stressed PM fractions, detected by SDS-PAGE, were very similar. NaCl substantially reduced the PM ATPase specific activity, whereas it had little effect on the apparent Km for Mg2+-ATP. Since the root PM ATPase of salt sensitive and resistant genotypes responded similarly to salinity stress, it seems unlikely that the mechanism of salt tolerance in wheat is primarily based on differences in PM ATPase characteristics.  相似文献

After 48 hours at 2°C, hypocotyls from chill-sensitive Cucumis sativus seedlings showed a burst of O2 uptake. The alternative pathway became engaged to close to 45% full capacity during this postchilling respiratory burst. However, it only accounted for up to 50% of this increased respiratory O2 uptake. By 24 hours after chilling, when the seedlings were fully recovered from visible symptoms of chilling injury, the flux through the alternative pathway was back to the level (about 10%) found before chilling. Blocking chilling-induced ethylene production with aminoethoxyvinylglycine had no effect on this increased utilization of the alternative pathway.  相似文献

Muskmelon (Cucumis melo L.) plants were exposed to a 10°C chilling treatment for 72 hours, which induced leaf chilling injury symptoms (wilting, appearance of water-soaked areas, necrosis). Chilling caused an accumulation of starch, sucrose, hexoses (glucose and fructose), and certain amino acids (glutamate, aspartate, and citrulline) in source leaf tissues, but no accumulation of stachyose or other galactosyl-oligosaccharides occurred. Chilling also caused a general increase in sugar (stachyose, raffinose, sucrose) and amino acid content of the phloem sap, although rates of phloem transport were apparently reduced. Pretreatment of the leaves with a 20-milligram per liter abscisic acid (ABA) spray before chilling prevented the appearance of chilling injury symptoms. ABA pretreatment had little or no affect on sugar accumulation in leaf tissues but greatly reduced or eliminated the chilling-induced amino acid accumulation. Higher levels of aspartate and particularly of arginine were found in phloem saps from ABA-pretreated plants. The data indicate that changes in leaf metabolism caused by environmental stresses such as chilling may change the composition of cucurbit phloem sap. This raises the possibility that some of the deleterious effects of stress on sink tissues may, in part, be due to alterations in the nature of the assimilate supply.  相似文献

本文应用PAGE技术,分析了经秋水仙碱诱变的甜瓜的四个品种、一个新品系的13份材料和3个对照的POD、EST、MDH、ADH、GDH、SOD的同工酶.根据16份材料的谱带特征二态数字矩阵,用MEGA程序计算了它们间的Numberofdifference,Proportionofdifference和Kimura-2-parameterdistance遗传距离,分别用UPGMA和Neighbor-joining进行聚类分析.结果表明,甜瓜材料经秋水仙碱诱变均产生了一定的变异,品种内的遗传分化程度与形态性状和细胞学检测相符合.同工酶遗传变异的大小可以反映诱变材料变异大小,在一定程度上可为有效选择育种材料提供依据.  相似文献
