The physical distribution of translocation breakpoints was analyzed in homoeologous recombinants involving chromosomes 1A, 1B, 1D of wheat and 1R of rye, and the long arms of chromosome 7S of Aegilops speltoides and 7A of wheat. Recombination between homoeologues was induced by removal of the Ph1 gene. In all instances, translocation breakpoints were concentrated in the distal ends of the chromosome arms and were absent in the proximal halves of the arms. The relationship between the relative distance from the centromere and the relative homoeologous recombination frequency was best explained by the function f(x)=0.0091e0.0592x. The pattern of recombination in homoeologous chromosomes was essentially the same as in homologues except that there were practically no double exchanges. Among 313 recombinant chromosomes, only one resulted from a double crossing-over. The distribution of translocation breakpoints in translocated arms indicated that positive chiasma interference operated in homoeologous recombination. This implies that the reduction of the length of alien chromosome segments present in translocations with wheat chromosomes may be more difficult than the production of the original recombinants.  相似文献

Distal location of chiasmata in chromosome arms is thought to be a consequence of the distal initiation of synapsis. Observations of meiotic behavior of a rye chromosome with an inverted arm show that patterns of chiasma distribution and frequency are also inverted; therefore, the patterns of synapsis and chiasma distribution are independent, and recombination frequency along a chromosome is position-independent and segment-specific. Since cases of random distribution of chiasmata and recombination are known in rye, a genetic mechanism must be present that licenses specific chromosome regions for recombination. Large differences in the metaphase I pairing of the inversion in various combinations of two armed and telocentric chromosomes confirm the major role of the telomere bouquet in early homologue recognition. However, occasional synapsis and chiasmate pairing of the distal regions of normal arms with the proximal regions of the inversion suggest that an alternative mechanism for juxtaposing of homologues must also be present. Synapsis in inversion heterozygotes was mostly complete but in the antiparallel orientation, hence defying homology, but non-homologues never synapsed. Instances of synapsis strictly limited to the chiasma-capable segments of the arm suggest that, in rye, both recombination-dependent and recombination-independent mechanisms for homologue recognition must be present.  相似文献

In a cytological analysis of the meiotic behaviour in PMCs of five hybrids between hexaploid triticale and durum wheat, Triticum turgidum L., chromosome association at meiotic first metaphase and the behaviour of rye univalents at first anaphase were analyzed. The chromosomes of the B genome, chromosomes 4A and 7A (disomic condition), and the seven rye chromosomes, could be distinguished by their C-banding pattern. No wheat-rye paring was detected at metaphase I. Rye univalents were observed as laggards which disjoined either predominantly equationaly (2R, 3R, 4R, 5R and 7R) or predominantly reductionaly (1R and 6R). Misdivision occurred in up to 3% of rye univalents.  相似文献

Summary The ph1b mutant in bread wheat has been used to induce homoeologous pairing and recombination between chromosome arm 1RL of cereal rye and wheat chromosome/s. A figure of 2.87% was estimated for the maximal frequency of recombination between a rye glutelin locus tightly linked to the centromere and the heterochromatic telomere on the long arm of rye chromosome 1R in the progeny of ph1b homozygotes. This equates to a gametic recombination frequency of 1.44%. This is the first substantiated genetic evidence for homoeologous recombination between wheat and rye chromosomes. No recombinants were confirmed in control populations heterozygous for ph1b. The ph1b mutant was also observed to generate recombination between wheat homoeologues.  相似文献

Introgressive hybridization is an efficient means to improve the genetic diversity of cultivated cereals, including triticale. To identify the triticale lines with Aegilops introgressions, genotyping was carried out with ten lines obtained by crossing hexaploid triticale with genome-substitution forms of the common wheat cultivar Aurora: Aurolata (AABBUU), Aurodes (AABBSS), and Aurotika (AABBTT). The genome composition of the triticale lines was studied by in situ hybridization, and recombination events involving Aegilops and/or common wheat chromosomes were assumed for nine out of the ten lines. Translocations involving rye chromosomes were not observed. Substitutions for rye chromosomes were detected in two lines resulting from crosses with Aurolata. Genomic in situ hybridization (GISH) with Ae. umbellulata DNA and molecular genetic analysis showed that chromosome 1R was substituted with Ae. umbellulata chromosome 1U in one of the lines and that 2R(2U) substitution took place in the other line. Fluorescence in situ hybridization (FISH) with the Spelt1 and pSc119.2 probes revealed a translocation from Ae. speltoides to the long arm of chromosome 1B in one of the two lines resulting from crosses with Aurodes and a translocation in the long arm of chromosome 7B in the other line. In addition, the pSc119.2 probe revealed chromosome 1B rearrangements in four lines resulting from crosses with Aurolata and in a line resulting from crosses with Aurotika. The lines were tested for main productivity parameters. A negative effect on all productivity parameters was demonstrated for Ae. umbellulata chromosome 2U. The overwinter survival in all of the lines was similar to or even higher than in the original triticale cultivars. A substantial increase in winter resistance as compared with the parental cultivar was observed for the line carrying the T7BS-7SL translocation. The line with the 1R(1U) chromosome substitution seemed promising for the baking properties of triticale.  相似文献

Three rye-specific repeated sequences, pSc10C, pSc20H and R173-1, were used to design sequence-specific anchored primers. These primers and 16 restriction site-specific adaptor primers were used in all possible combinations to establish sequence-specific amplified polymorphic (SSAP) markers for the 1RS chromosome arm of rye in a wheat background. Thirty 1RS-specific SSAP markers were detected in 19 primer combinations. Along with six markers localised previously on 1RS, 26 of the SSAP markers were mapped genetically in wheat genotypes carrying recombinant 1BL.1RS translocations. A clear decrease in recombination frequency from distal to proximal regions was observed. Wheat-rye addition lines for the 1R chromosome with different-sized deletions of the short arm were used to physically localise these markers. Physical mapping suggested an even distribution of the SSAP markers along the total length of the 1RS chromosome arm.Communicated by J.W. Snape  相似文献

Summary The substitution patterns of rye chromosomes in hexaploid triticale × wheat F2 hybrids, along with the transmission patterns of rye chromosomes through egg cells and pollen when several of the F1 hybrids were test crossed to triticale and wheat were investigated. The data indicated that the rye chromosome transmission through both the egg and pollen was random in number and in composition. The test crosses suggested that it was best to use wheat pollen for the transmission of rye chromosomes through the egg cells of the F1 hybrids and triticale egg cells for the transmission of rye chromosomes through F1 hybrid pollen. A deviation from random segregation in the F2 and the transmission rate was observed for rye chromosomes 1R, 4R/7R, and 6R. The transmission rates of 1R and 6R varied depending on the direction in which the cross was made. The results also indicated that there was little or no compensation between the R- and D-genomes and that the chromosomes of these two genomes appeared to be transmitted independently of each other.  相似文献

Wide hybrids have been used in generating genetic maps of many plant species. In this study, genetic and physical mapping was performed on ph1b-induced recombinants of rye chromosome 2R in wheat (Triticum aestivum L.). All recombinants were single breakpoint translocations. Recombination 2RS-2BS was absent from the terminal and the pericentric regions and was distributed randomly along an intercalary segment covering approximately 65% of the arm's length. Such a distribution probably resulted from structural differences at the telomeres of 2RS and wheat 2BS arm that disrupted telomeric initiation of pairing. Recombination 2RL-2BL was confined to the terminal 25% of the arm's length. A genetic map of homoeologous recombination 2R-2B was generated using relative recombination frequencies and aligned with maps of chromosomes 2B and 2R based on homologous recombination. The alignment of the short arms showed a shift of homoeologous recombination toward the centromere. On the long arms, the distribution of homoeologous recombination was the same as that of homologous recombination in the distal halves of the maps, but the absence of multiple crossovers in homoeologous recombination eliminated the proximal half of the map. The results confirm that homoeologous recombination in wheat is based on single exchanges per arm, indicate that the distribution of these single homoeologous exchanges is similar to the distribution of the first (distal) crossovers in homologues, and suggest that successive crossovers in an arm generate specific portions of genetic maps. A difference in the distribution of recombination between the short and long arms indicates that the distal crossover localization in wheat is not dictated by a restricted distribution of DNA sequences capable of recombination but by the pattern of pairing initiation, and that can be affected by structural differences. Restriction of homoeologous recombination to single crossovers in the distal part of the genetic map complicates chromosome engineering efforts targeting genes in the proximal map regions.  相似文献

Summary Several studies have indicated a noncorrespondence between genetic and physical distances in wheat chromosomes. To study the physical distribution of recombination, polymorphism for C-banding patterns was used to monitor recombination in 67 segments in 11 B-genome chromosome arms of Triticum turgidum. Recombination was absent in proximal regions of all chromosome arms; its frequency increased exponentially with distance from the centromere. A significant difference was observed between the distribution of recombination in physically short and physically long arms. In physically short arms, recombination was almost exclusively concentrated in distal segments and only those regions were represented in their genetic maps. In physically long arms, while a majority of the genetic distance was again based upon recombination in distal chromosome segments, some interstitial recombination was observed. Consequently, these regions also contributed to the genetic maps. Such a pattern of recombination, skewed toward terminal segments of chromosomes, is probably a result of telomeric pairing initiation and strong positive chiasma interference. Interference averaged 0.81 in 35 pairs of adjacent segments and 0.57 across the entire recombining portions of chromosome arms. The total genetic map lengths of the arms corresponded closely to those expected on the basis of their metaphase-I chiasma frequencies. As a consequence of this uneven distribution of recombination there can be a 153-fold difference (or more) in the number of DNA base pairs per unit (centiMorgan) of genetic length.  相似文献

Six doubled-haploid (DH) lines, derived by anther culture from octoploid triticale x wheat hybrids, were characterized using cytological, biochemical and molecular techniques. Lines varied in their wheat and rye genome composition, and were either wheat-rye chromosome multiple addition lines or had spontaneous substitutions and/or wheat-rye translocations. Most of the lines contained a pair of 4R chromosomes, whereas 1R or 7R were present in others. The results are similar to those previously obtained with hexaploid triticale x wheat crosses and indicate that it is possible to produce alien (wheat/rye) addition, substitution, and translocation lines directly from the anther culture of intergeneric hybrids.  相似文献

Summary Plants were regenerated from cultured immature embryos of two pairs of sister lines of triticale (X Triticosecale) cvs Rosner and Drira and five sister lines of rye (Secale cereale). The triticale lines differ in heterochromatic content of a particular rye chromosome (6R or 7R), while the rye lines differ in only one heterochromatic band. Variation in morphogenetic response was present between the triticale cultivars and between the rye lines. One of the rye lines (7RL+ +) showed a distinctive superior response in terms of somatic embryogenesis. These findings are discussed in relation to factors affecting morphogenetic response and genetic stability in culture.  相似文献

Presence of genes in gene-rich regions on wheat chromosomes has been widely reported. However, there is a lack of information on the precise characterization of these regions with respect to the distribution of genes and recombination. We attempted to critically analyze the available data to characterize gene-rich regions and to study the distribution of genes and recombination on wheat homoeologous group 6 chromosomes which are a reservoir of several useful genes controlling traits of economic importance. Consensus physical and genetic linkage maps were constructed for homoeologous group 6 using physical and genetic mapping data. Five major gene-rich regions were identified on homoeologous group 6 chromosomes, with two on the short and three on long arm. More than 90% of marker or gene loci were present in these five gene-rich regions, which comprise about 30% of the total physical chromosomal length. The gene-rich regions were mainly present in the distal 60% regions of the chromosomes. About 61% of the total loci map in the most distal regions which span only about 4% of the physical length of the chromosome. A range of sub-microscopic regions within each gene-rich region were also identified. Comparisons of the consensus physical and genetic linkage maps revealed that recombination occurred mainly in the gene-rich regions. Seventy percent of the total recombination occurred in the two most distally located regions that span only 4% of the physical length of the chromosomes. The relationship of recombination to the gene-rich region is not linear with distance from the centromere, especially on the long arm. The kb/cM estimates for group 6 chromosomes ranged from 146 kb in the gene-rich to about 10 Mb in the gene-poor region. In wheat (Triticum aestivum L.), the crossover (CO) frequency increases gradually from the centromeres to the telomeres. However, little is known about the factors affecting both the distribution and the intensity of recombination along this gradient. To investigate this, we studied in detail the pattern of CO along chromosome 3B of bread wheat. A dense reference genetic map comprising 102 markers homogeneously distributed along the chromosome was compared to a physical deletion map. Most of the COs (90%) occurred in the distal subtelomeric regions that represent 40% of the chromosome. About 27% of the proximal regions surrounding the centromere showed a very weak CO frequency with only three COs found in the 752 gametes studied. Moreover, we observed a clear decrease of CO frequency on the distal region of the short arm. Finally, the intensity of interference was assessed for the first time in wheat using a Gamma model. The results showed m values of 1.2 for male recombination and 3.5 for female recombination, suggesting positive interference along wheat chromosome 3B.  相似文献

Summary Investigations were made on the rye chromosome constitution and on the presence of telomeric heterochromatin in rye chromosomes of the 26 most widely and 24 most narrowly adapted triticale strains. Among widely adapted lines, 22 (85%) had a complete rye genome and four triticales only had chromosomal R-D genome substitutions. Twenty-three (96%) of the 24 most narrowly adapted triticales had substitutions between the chromosomes of the R and D genomes. The most widely adapted triticales accumulated fewer modified rye chromosomes in comparison to narrowly adapted lines. They had from one to three rye chromosomes with heterochromatic deletions: 46% of widely adapted lines had two modified rye chromosomes; 34% had three modified rye chromosomes, and 19% had a single modified rye chromosome. In widely adapted strains, the 1R, 4R, 5R and 6R modified chromosomes were observed; they were present in 80%, 73%, 50% and 11% of the cases, respectively. The most narrowly adapted triticales had from two to four modified rye chromosomes: 58% of the strains had three modified rye chromosomes; 29% had four modified rye chromosomes and 12% had two modified rye chromosomes. The modified 4R and 5R chromosomes were present in all of these lines. The 1R (modified), 6R (modified) and 7R (modified) were found in 83%, 25% and 16%, respectively, of the narrowly adapted strains.Results support the previous observations (Pilch 1980b) that a wide adaptation of hexaploid triticales is associated with the presence of the full potential of rye genome, and that it is independent of the amount of telomeric heterochromatin possessed by rye chromosomes.  相似文献

In many organisms, homologous pairing and synapsis depend on the meiotic recombination machinery that repairs double-strand DNA breaks (DSBs) produced at the onset of meiosis. The culmination of recombination via crossover gives rise to chiasmata, which locate distally in many plant species such as rye, Secale cereale. Although, synapsis initiates close to the chromosome ends, a direct effect of regions with high crossover frequency on partner identification and synapsis initiation has not been demonstrated. Here, we analyze the dynamics of distal and proximal regions of a rye chromosome introgressed into wheat to define their role on meiotic homology search and synapsis. We have used lines with a pair of two-armed chromosome 1R of rye, or a pair of telocentrics of its long arm (1RL), which were homozygous for the standard 1RL structure, homozygous for an inversion of 1RL that changes chiasma location from distal to proximal, or heterozygous for the inversion. Physical mapping of recombination produced in the ditelocentric heterozygote (1RL/1RL(inv)) showed that 70% of crossovers in the arm were confined to a terminal segment representing 10% of the 1RL length. The dynamics of the arms 1RL and 1RL(inv) during zygotene demonstrates that crossover-rich regions are more active in recognizing the homologous partner and developing synapsis than crossover-poor regions. When the crossover-rich regions are positioned in the vicinity of chromosome ends, their association is facilitated by telomere clustering; when they are positioned centrally in one of the two-armed chromosomes and distally in the homolog, their association is probably derived from chromosome elongation. On the other hand, chromosome movements that disassemble the bouquet may facilitate chromosome pairing correction by dissolution of improper chromosome associations. Taken together, these data support that repair of DSBs via crossover is essential in both the search of the homologous partner and consolidation of homologous synapsis.  相似文献

The relationship between the chromosomal location of heterochromatin C-bands and of four non-homologous repeated sequence families constituting 8 to 12% of total rye DNA has been investigated in chromosomes of rye (Secale cereale) by in situ hybridisation. Three rye varieties, a set of rye disomic additions to wheat and a triticale were studied. Only centromeric and nucleolar organizer region (NOR) associated C-bands failed to display hybridisation to at least one of the sequences and many telomeric blocks of heterochromatin contained all four repeated sequence families. Both between-variety differences in the chromosomal distribution of repeated sequences, and intravarietal heterozygosities were frequently noted and are probably widespread. — Previously reported deletions of heterochromatin from King II rye chromosomes added to the Holdfast wheat complement were correlated with deletions of some, but not all, of the highly repeated sequence families. A previously unreported loss of some families from King II rye chromosome 4R/7R in a Holdfast wheat genetic background was detected. This loss was not associated with complete deletion of a C-band. A deletion has also probably occurred from the short arm telomere of 4R/7R in the triticale variety Rosner. It is suggested that the families of repeats in rye telomeric heterochromatin which are absent from wheat are selected against in the wheat genetic background.  相似文献

Summary Genetic mapping of polymorphic C-bands allows direct comparisons between genetic and physical maps. Eleven C-bands and two seed storage protein genes on chromosome 1B, polymorphic between Langdon durum and four accessions of T. dicoccoides, were used to study the distribution of recombination along the entire length of the chromosome. Recombination in the short arm was almost completely restricted to the satellite, two-thirds of the arm's length from the centromere; the Gli-B1 gene was found to be tightly linked to the telomeric C-band. In the long arm, the distal 51.4% of the arm accounted for 88% of recombination; the proximal half of the arm accounted for the remaining 12%. While the amount of crossing-over differed significantly between the four T. dicoccoides 1B chromosomes, there were no significant differences in the relative distributions of crossing-over along the chromosome. Consequently, the genetic maps obtained from the four individual T. dicoccoides chromosomes were combined to yield a consensus map of 14 markers (including the centromere) for the chromosome.  相似文献
