The inaccessibility of mammalian organogenesis stage embryos has precluded their widespread use in embryological and teratological studies. As organogenesis occurs during the last 1.5 days of the 10. 7 days of gestation in the stripe-faced dunnart (Sminthopsis macroura), the aim of the present study was to investigate whether day 9 and day 10 embryos and fetuses could be grown to term in vitro. High glucose Dulbecco's modified Eagle's medium with 10% fetal calf serum (FCS) supported embryonic growth for various periods of time, some to within 5 h of the predicted time of parturition. A roller culture system maintained at 35 degrees C was used to incubate organogenesis stage embryos (n = 43). Nine unincubated (control) embryos were either fixed for microscopic analysis or frozen for microprotein determination. The results of the present study indicate that with some optimization of the culture conditions (increasing oxygen in the gas phase in the culture tubes, replacing FCS with rat serum), it might be possible for organogenesis stage S. macroura embryos to be grown to term. A scoring scheme for assessing morphological development was devised for use as a standard in staging organogenesis stage embryos. This scheme reflects the highly compressed schedule of developmental events that occurs mainly during day 9 of gestation in S. macroura embryos. In comparison, during embryogenesis in Didelphis virginiana these developmental events occur from day 8 to day 10.5 of gestation, and birth occurs on day 13. 相似文献

Seasonal changes in the number and distribution of spermatozoa in males, and annual changes in the distribution of litters and embryos in females were examined in the iteroparous dasyurid marsupial, Sminthopsis macroura , in captivity. Total number of sperm in the testis (0.53 × 106 sperm/testis) and epididymidis (0.54 × 106 sperm/epididymidis) were extremely low when compared with those in other marsupials and eutherian mammals. Testicular sperm production and epididymal sperm reserves were high between May and October and declined to a minimum in March. These changes reflected monthly changes in testicular and epididymal weight and testis morphology. Data on changing epididymal sperm distribution suggest that sperm storage in the cauda epididymidis is limited and that few sperm are required for successful insemination. Litters were born between June and January, with most litters occurring between July and October. Second pregnancies occurred between October and January, with a peak in December. The data indicate that the timing of mating activity and litter production by S. macroura correspond very closely with the period of maximum sperm production by males. The synchrony of these events contrasts dramatically with that of similar-sized semelparous dasyurid species. It is hypothesized that testicular failure prior to the mating season, copulatory behaviour, and possibly male die-off in dasyurid marsupials are related to the degree of competition between males for mates and, hence, population density and environmental predictability. These data suggest that intermale sperm competition is affected by the periods of female receptivity and the length of sperm storage in the female reproductive tract. Fundamental differences in the reproductive strategies of iteroparous and semelparous dasyurid marsupials are discussed. 相似文献

Summary We examined the effect of cycles of 12 h warm (35 ± 2 °C) and 12 h (21 ± 2 °C) ambient temperature (Ta) upon the circadian activity rhythms of stripe-faced dunnarts, Sminthopsis macroura, free-running in conditions of constant dark (DD) or constant light (LL). It was hypothesized that dunnarts would entrain to the temperature cycles (TaHLs) or show perturbations of period, and that LL would act synergistically with the TaHLs in these effects. Under DD, 2 of 6 animals showed clear entrainment to the TaHLs. Other animals exhibited changes of period () and heavy negative masking of activity during the warm fraction of the TaHLs. Under LL, 3 of 12 animals entrained to the TaHLs. It was concluded that Ta is a significant though weak Zeitgeber for S. macroura compared to light. It is possible that TaHLs entrain homeotherm activity rhythms by altering the rhythm of body temperature, which is usually tightly coupled to activity.Abbreviations

Ovulation occurs in Sminthopsis macroura approximately 160 hr after administration of 1.3 IU PMSG, and yields significantly more oocytes than does spontaneous ovulation (P = 0.001). Germinal vesicle (GV)-stage oocytes have a thin cortical rim of microfilaments, which is disrupted by exposure to cytochalasin D. After GV breakdown, the first meiotic spindle forms subcortically and parallel to the oolemma. It rotates during anaphase and telophase to extrude the first polar body. This rotation is associated with a local cortical concentration of microfilaments, which is extruded in the first polar body. The second meiotic spindle is orthogonal to the surface, and extrusion of the second polar body is not associated with obvious local changes in cortical actin, resulting in a polar body containing little polymerized actin. The sites of second polar body emission and sperm entry are always in the half of the oocyte opposite the concentrating yolk mass, and are within 60° of each other in most oocytes. During the concentration and eccentric movement of the yolk, microfilaments condense around it. During yolk expulsion, these microfilaments become continuous with those located subcortically. During early cleavage, the cytocortex of the zygote, but not of the extruded yolk mass, stains heavily for polymerised actin. Multiple sites of pericentriolar material are detectable in the cytoplasm of some secondary unfertilized oocytes which, in the presence of taxol, generate large cytasters and pseudospindle structures. After fertilization, a large aster is formed in association with the sperm entry point and serves as the center of an extensive cytoplasmic network of microtubules which surrounds but does not enter the yolk mass. Taxol treatment generates small cytasters within this meshwork and promotes selective stabilization of some periyolk microtubules opposite to the sperm aster. © 1995 Wiley-Liss, Inc. 相似文献

We report the first isolation and sequencing of genomic BAC clones containing the marsupial milk protein genes Whey Acidic Protein (WAP) and Early Lactation Protein (ELP). The stripe-faced dunnart WAPgene sequence contained five exons, the middle three of which code for the WAPmotifs and four disulphide core domains which characterize WAP. The dunnart ELPgene sequence contained three exons encoding a protein with a Kunitz motif common to serine protease inhibitors. Fluorescence in situ hybridization located the WAPgene to chromosome 1p in the stripe-faced dunnart, and the ELPgene to 2q. Northern blot analysis of lactating mammary tissue of the closely related fat-tailed dunnart has shown asynchronous expression of these milk protein genes. ELPwas expressed at only the earlier phase of lactation and WAPonly at the later phase of lactation, in contrast to beta-lactoglobulin (BLG) and alpha-lactalbumin (ALA) genes, which were expressed in both phases of lactation. This asynchronous expression during the lactation cycle in the fat-tailed dunnart is similar to other marsupials and it probably represents a pattern that is ancestral to Australian marsupials. 相似文献

A study of spermatozoa in the isthmus of the oviduct and of the surrounding epithelial cells in the dasyurid marsupial, Sminthopsis crassicaudata, was carried out. At least 10% of the ejaculated spermatozoa probably populate the isthmus region, where many come to reside in crypts until around the time of ovulation. Ultrastructural observations of spermatozoa in this region indicated that they had intact acrosomes and were identical in their morphology with those in the cauda epididymidis. After ovulation spermatozoa rapidly disappeared, some of which may be phagocytosed by the cells lining the crypts. These epithelial cells were also found to have many large, electron-dense granules at the time of sperm storage, but the contents did not appear to be released until the zygotes passed along the tract. The secretory activity of these cells may thus relate more to the production of the shell membrane that comes to surround the zygote than to the cells performing a nutritive or protective function for the spermatozoa during their period of storage within the female reproductive tract. 相似文献

Marsupials are potentially excellent models for the study of gastrulation because of their superficial embryonic area (EA), post-gastrulation implantation and their potential to provide information about the evolution of gastrulation. Very few studies have examined this developmental period in marsupials. Using an established developmental timetable, peri-gastrula stage Sminthopsis macroura blastocysts were collected and described in detail by observations on live blastocysts and by the use of histological and immunohistochemical techniques on fixed blastocysts. Gastrulation in S. macroura shares several aspects common to that of both eutherian mammals and birds, but in terms of tissue arrangement and conceptus form, is more similar to the chick than other mammals. Two methods of culturing EA explants flat without their shell were devised. The techniques will markedly increase the number of possible experimental manipulations, which previously were limited by the presence of blastocyst investments. Exposure of fractions of explants of round, morphologically uniform pre-gastrula stage EA to growth factors or signaling molecules implicated in vertebrate gastrulation suggests that like the chick and mouse, the marsupial epiblast is patterned prior to gastrulation. Of all factors tested, basic fibroblast growth factor (bFGF) had the most prominent effect, promoting cell differentiation, and possible mesoderm formation. Data from explant culture suggests that similar to the chick and mouse, limited specification precedes the onset of gastrulation. 相似文献

The fluid that surrounds the embryo in the uterus contains important nourishing factors and secretions. To maintain the distinct microenvironment in the uterine lumen, the tight junctions between uterine epithelial cells are remodeled to decrease paracellular movement of molecules and solutes. Modifications to tight junctions between uterine epithelial cells is a common feature of pregnancy in eutherian mammals, regardless of placental type. Here we used immunofluorescence microscopy and western blot analysis to describe distributional changes to tight junctional proteins, claudin‐1, ‐3, ‐4, and ‐5, in the uterine epithelial cells of a marsupial species, Sminthopsis crassicaudata. Immunofluorescence microscopy revealed claudin‐1, ‐3, and ‐5 in the tight junctions of the uterine epithelium of S. crassicaudata during pregnancy. These specific claudins are associated with restricting passive movement of fluid between epithelial cells in eutherians. Hence, their function during pregnancy in S. crassicaudata may be to maintain the uterine luminal content surrounding developing embryos. Claudin‐4 disappears from all uterine regions of S. crassicaudata at the time of implantation, in contrast with the distribution of this claudin in some eutherian mammals. We conclude that like eutherian mammals, distributional changes to claudins in the uterine epithelial cells of S. crassicaudata are necessary to support pregnancy. However, the combination of individual claudin isoforms in the tight junctions of the uterine epithelium of S. crassicaudata differs from that of eutherian mammals. Our findings suggest that the precise permeability of the paracellular pathway of the uterine epithelium is species‐specific. 相似文献

In the winter of 1981, 9 months after an intense fire in November 1980 in the recently logged Mumbulla State Forest, the numbers of a small population of Sminthopsis leucopus had slightly increased, while the common species of small mammals had declined sharply from the previous winter. The study reported in this paper aimed to identify the habitat of S. leucopus. Ninety study plots were divided equally among three classes of forest (unlogged forest, and forest logged in 1979 and 1980), between two aspects (north- and south-facing slopes), and among three topographic positions (ridge, midslope and gully). Plots were assessed on the basis of both floristic data and the structural components of ground, shrub and tree cover. The primary finding was that the habitat of S. leucopus was the treeless ridges and mid-slopes with less than 51% ground cover in the logged areas of burnt forest. The floristic analysis showed that the vegetation subcommunities restricted to gullies, or carrying a ferny ground layer of vegetation, were not selected by S. leucopus. An analysis of movements of male S. leucopus identified two classes. ‘Resident’ males occurred on logged ridges and midslopes characterized by a floristic subcommunity of open forest with a grassy understorey.‘Explorer’ males moved through a variety of ridge habitats, including unlogged forest, and were spread more widely through the floristic communities. The pattern of habitat selection identified in this study shows S. leucopus to be ecologically distinct from the other species of small mammals in the forest and thus in need of special consideration in management programmes if it is to be conserved. Under the current regime, the uniform treatment of the forest leads to widespread stands of dense regrowth to the exclusion of the species and is inimical to its survival. 相似文献

Formation of a placenta requires intimate contact between the embryonic and maternal uterine epithelia in early pregnancy. Contact is accompanied by a characteristic suite of changes to the plasma membranes of uterine epithelial cells, termed the plasma membrane transformation. The plasma membrane transformation occurs in eutherian mammals and in viviparous (live‐bearing) squamate reptiles, and may be fundamental to the evolution of viviparity in amniotes. Marsupials provide an excellent opportunity to test the generality of this phenomenon. Here, we present the first detailed study of the plasma membrane transformation in a marsupial. We combine electron microscopy and immunohistochemistry to describe morphological and molecular features of uterine epithelial cells during pregnancy in the fat‐tailed dunnart (Sminthopsis crassicaudata; Dasyuridae). Cell morphology changes dramatically in S. crassicaudata during pregnancy. Apical microvilli are replaced by irregular blunt projections, then by spiky projections postimplantation. Cell surfaces flatten and ciliated cells are lost. Junctional complexes between adjacent cells increase in depth, then decrease just before implantation, which is consistent with junctional protein localization in this region of the cell membrane. The uterine cellular changes in S. crassicaudata are consistent with a plasma membrane transformation, and support the idea that this phenomenon is fundamental to the evolution of viviparity in amniote vertebrates. Sminthopsis is the most speciose genus of living dasyurid marsupials and, along with its close relatives Antechinomys and Ningaui, constitutes the clade Sminthopsini. Phylogenetic relationships among the 23 species in this clade have been the subject of much morphological and molecular investigation, including a recent integration of penis morphology (in Sminthopsis) with molecular systematics. Several phylogenetic issues remain open, however, including the monophyly of Sminthopsis and branching order among early sminthopsin lineages. In this study, we revisit sminthopsin systematics with an expanded molecular data set, including new DNA sequences from mitochondrial (valine transfer-RNA and 16S ribosomal RNA) and nuclear (interphotoreceptor retinoid binding protein and beta-fibrinogen) loci, along with previously published sequences of cytochrome b, 12S ribosomal RNA, control region, and protamine P1. Our results again fail to establish the monophyly of Sminthopsis, but do provide a clearer resolution of early sminthopsin branching. Specifically, our phylogeny suggests three major groups of Sminthopsis species: S. longicaudata (perhaps the sister of Antechinomys); the Macroura species group of previous authors (S. crassicaudata, S. macroura, S. virginiae, S. douglasi, and S. bindi); and the remaining 13 species allied with the Murina species group. Our results depart from previous molecular findings by reuniting S. ooldea with the Murina group, while resolving S. psammophila as sister to the hairy-footed dunnarts (S. hirtipes and S. youngsoni). We suggest that this conflict traces to anomalous phylogenetic signal in previously published cytochrome b sequences. Penis morphology maps reasonably well onto our phylogeny, requiring parallel origination of only one of the ten morphotypes described for Sminthopsis. 相似文献

This study investigated the effects of predation risk, dune position and microhabitat on foraging of the Lesser Hairy‐footed Dunnart Sminthopsis youngsoni, a small insectivorous marsupial, in the Simpson Desert of western Queensland. The intensity of foraging was assessed by establishing feeding stations (dishes containing mealworms) in open and bush microhabitats at three levels on sand dunes, and recording the numbers of prey taken by dunnarts from the stations after nightly bouts of foraging. Risk of predation was manipulated by provision of artificial illumination at alternate feeding stations on each of five occasions when trials were run. The numbers of mealworms left after feeding bouts varied inconsistently across trials, providing little evidence that dunnarts respond to habitat or predation risk while foraging. These results contrast sharply with studies of arid zone rodents, where foraging is usually sensitive to both predation risk and resource distribution. We suggest that S. youngsoni forages equally in all habitats of its sandridge environment, and experiences relatively low risk of predation whilst doing so. 相似文献

Embryos of Sminthopsis crassicaudata and Sminthopsis macroura were cultured for up to 96 hours during cleavage and early expansion of the blastocyst in Dulbecco's modified Eagle's medium (DMEG), DMEG containing 2.76 gm/liter sodium lactate (DMEGL), DMEG containing 3.5 gm/liter galactose (DMEGAL), DMEG containing 15 ng/ml progesterone (DMEGP) or 150 ng/ml progesterone (DMEGP10), and DMEGL containing 15 ng/ml progesterone (DMEGLP). The disappearance of sperm was used to indicate the time of ovulation (day 0). Fertilized eggs were found in the uterus at the end of day 1, four-cell stages at the end of day 2, and embryos completing the fourth division by the end of day 3 in S. macroura and day 4 in S. crassicaudata. Estimated developmental times in culture were similar to those obtained in vivo. In both species, the first two divisions take about 24 hours, cleavage is arrested for 24 hours or longer at the rounded four-cell stage, and the third and fourth divisions take a further 24 hours. The blastocyst expands during the next 24 hours in which time the fifth and sixth divisions occur. It was possible to culture embryos from S. macroura but not S. crassicaudata over the four-cell stage to early expanding blastocysts. DMEGAL did not support cleavage in culture. DMEG, DMEGL, DMEGP, DMEGP10, and DMEGLP all supported culture during cleavage and early blastocyst expansion. Blastocyst expansion was slightly enhanced using media containing sodium lactate. More embryos completed the fifth division and formed expanding blastocysts in DMEG, DMEGL, and DMEGLP. 相似文献

Male Antechinus stuartii (Marsupialia: Dasyuridae) undergo significant endocrinological changes during their life history. The endocrine changes are associated with complete male mortality. The accessory reproductive tract of males is important for seminal plasma production, and changes associated with the reproductive cycle were not described in detail by earlier studies. The present study sought to describe some of the seasonal changes in structure of the male accessory reproductive tract in relation to the known hormonal changes. The epididymis, prostate, and bulbourethral glands are relatively undifferentiated in February and May, a time when plasma concentrations of testosterone are known to be low. By July, considerable hypertrophy and differentiation of the accessory reproductive tract are observed. This is most obvious in the bulbourethral glands, which change from being indistinguishable from one another in February and May to being three large morphologically and histologically distinct glands in July. The hypertrophy and secretory activity continue into August, the breeding season. These findings correlate with the hormonal profiles found in other studies of A. stuartii. The small marsupial Antechinus stuartii experiences a synchronised life cycle that culminates in complete male mortality (within 3 weeks) following the 1 week mating
period in mid-August (late winter). There are pronounced physiological changes in male A. stuartii over the life cycle and renal function was assessed for correlation with these changes. Glomerular filtration rate and urine
and plasma electrolytes were determined in male and female A. stuartii in February, May, July and August. Females showed little change in glomerular filtration rate, except for pre-mating values
in August which decreased. In contrast, glomerular filtration rate of males decreased significantly in July and August. Plasma
sodium and chloride levels were higher in males than females and were higher in animals in July and August than in February
and May. Plasma potassium levels dropped in both males and females in July and August. Plasma osmolality was higher in animals
in February compared to animals from May and August. However, there were no significant sex or seasonal differences in urine
electrolytes, although urea concentration was higher in females than males. Urine osmolality was higher in both sexes in July
and August. There were no significant differences in total excretory rates of sodium, potassium or chloride between sexes
or between seasons. Many of the alterations in renal function are correlated with known physiological and hormonal profiles
in A. stuartii. This is the first observation of seasonal changes in glomerular filtration rate that are unrelated to dietary and water
stresses.
