灭活的双歧杆菌对小鼠血清中细胞因子水平的影响   总被引:5,自引:2,他引:5  
目的:观察灭活的双歧杆菌对小鼠血清中I-—1β、IL-6和IFN-γ水平的影响。方法:小鼠腹腔注射环磷酰胺造成免疫低下动物模型,分别以新鲜BS肉汤培养基、耗尽培养上清(SCS)以及灭活的和活的双歧杆菌菌液进行灌胃。采用ELISA法检测血清中IL-1β、IL-6和IFN-γ的含量。结果:灭活的双歧杆菌与双歧杆菌活菌均可提高免疫低下小鼠血清中IL-1β、IL-6和IFN-γ的含量,二者的作用效果差异无显著性(P>0.05),SCS也具有一定的免疫促进作用,但与双歧杆菌活菌相比差异有显著性(P<0.05)。结论:灭活的双歧杆菌具有与双歧杆菌活菌相同或相近的免疫学活性,两者均可提高小鼠血清中细胞因子水平。  相似文献

目的 探讨青春双歧杆菌对1型糖尿病小鼠胰岛β细胞的保护作用.方法 给予1型糖尿病小鼠口服青春双歧杆菌,观察实验组和对照组体重的变化及糖尿病发病率.ELISA法测定血清、脾细胞上清IL-4和IFN-γ浓度,电镜观察胰岛超微结构变化.结果 15周龄时实验组体重(23.41±1.64)g, 而对照组为(22.31±1.32)g(P<0.05);实验组发病率低于对照组(P<0.05);实验组血清、脾细胞上清IL-4浓度明显高于对照组,而IFN-γ低于对照组(P<0.05);对照组残存胰岛的β细胞数目稀少,有核膜和内浆网扩张、核糖体脱颗粒和分泌颗粒空泡样变等超微结构异常,而实验组胰岛β细胞数显著多于对照组(P<0.05),且无上述超微异常.结论 青春双歧杆菌通过上调IL-4水平,降低IFN-γ水平,促使免疫平衡向Th2方向偏移,且有保护胰岛β细胞超微结构的作用,在一定程度上可预防和延缓NOD鼠糖尿病的发生.  相似文献

目的通过观察青春双歧杆菌对2型糖尿病模型大鼠肠道菌群的变化,和血清中总胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白(HDL-C)、超氧化物歧化酶(SOD)和丙二醛(MDA)的水平,探讨青春双歧杆菌对2型糖尿病模型大鼠肠道功能和脂质代谢的影响。方法采用青春双歧杆菌灌胃2型糖尿病模型大鼠,取粪便检查正常菌群,取血和脏器检测TC、TG、HDL-C、SOD和MDA含量。结果青春双歧杆菌导致肠道内双歧杆菌、乳杆菌的数量增加,而肠杆菌、肠球菌数量下降;TC、TG和MDA水平下降,而HDL-C和SOD水平升高。结论青春双歧杆菌具有改善2型糖尿病模型大鼠肠道功能和降血脂作用,与二甲双胍联合应用效果更佳。  相似文献

目的 探讨早期应用青春双歧杆菌对NOD小鼠1型糖尿病发病的影响.方法 给予NOD小鼠口服青春双歧杆菌,观察实验组和对照组(PBS)糖尿病发病率,HE染色观察胰岛炎,免疫组化检测胰岛Bcl-2和Bax的表达,RT-PCR测定TNF-α、IFN-γ和IL-10 mRNA表达.结果 实验组胰岛炎程度较对照组明显减轻(P<0.01),胰岛Bcl-2的表达高于对照组,而Bax的表达低于对照组(P<0.05);胰腺TNF-α、IFN-γ mRNA表达实验组明显低于对照组(P<0.05),而IL-10 mRNA表达差异无显著性;实验组发病率低于对照组(P<0.05).结论 青春双歧杆菌对NOD小鼠1型糖尿病有预防作用,其机制可能与调节Th1/Th2型细胞因子的免疫失衡有关.  相似文献

目的:初步探讨类风湿关节炎大鼠免疫功能紊乱的机制。方法:采用酶联免疫吸附法(ELISA)检测类风湿关节炎大鼠血清中Th1细胞分泌的细胞因子IFN-γ和Th2细胞分泌的细胞因子IL-4的水平。结果:类风湿关节炎大鼠血清中IFN-γ的水平与正常组相比明显升高(p<0.01),IL-4的水平明显降低(p<0.01)。结论:在类风湿关节炎大鼠血清中Th1/Th2细胞分泌的细胞因子水平存在着明显失衡,两者以Th1细胞分泌细胞因子占优势,这可能与其发病机制密切相关。  相似文献

目的通过对脾肾阳虚型溃疡性结肠炎(UC)模型大鼠血清及结肠组织中IL-1、IL-6、TNF-α及IFN-γ表达水平的检测,探讨它们在UC发生发展过程中的作用。方法采用灌服大黄水煎液+肌肉注射氢化可的松并结合TNBS(2,4,6-三硝基苯磺酸)+乙醇灌肠建立脾肾阳虚型UC动物模型。将60只大鼠随机分为空白组、脾肾阳虚型UC模型7、14d及21d组,采用酶联免疫法检测各组大鼠血清及结肠组织中IL-1、IL-6、TNF-α及IFN-γ的含量。结果与空白组比较,脾肾阳虚型UC模型组大鼠血清及结肠组织中IL-1、IL-6、TNF-α及IFN-γ含量明显升高(P0.05);尤以模型21d组最为明显。结论促炎性细胞因子IL-1、IL-6、TNF-α及IFN-γ在脾肾阳虚型UC发病过程中起重要作用。  相似文献

目的建立诱发性2型糖尿病小鼠模型,并将其与自发性2型糖尿病小鼠db/db进行比较分析。客观评价两种2型糖尿病小鼠模型,为糖尿病研究中动物模型的选择与实际应用提供实验依据。方法高脂饲料喂养C57BL/6J小鼠4周,腹腔连续3次注射STZ,建立诱发性2型糖尿病小鼠模型。感染后4周,大体肉眼观察小鼠的肝脏、肾脏,测定糖耐量,血清生化指标及血清细胞因子IL-2、IL-4、IL-6、IFN-γ、TNF-α、IL-17、IL-10表达量,将其与同龄的自发性2型糖尿病小鼠db/db进行比较分析。结果肉眼观察发现,两组模型小鼠的肝脏、肾脏与对照组均具有明显差异。糖耐量分析中,两组模型小鼠与对照组小鼠各时间点的血糖值均具有统计学差异(P〈0.05),耐糖功能低下,两组模型小鼠间血糖值无统计学差异。血液生化指标中,与对照组小鼠相比,两组模型小鼠GLU、CHOL、LDLC明显升高(P〈0.05);两组模型小鼠相互比较,诱发性2型糖尿病小鼠血脂水平较高(P〈0.05)。免疫指标比较显示:除IL-2外,两组模型小鼠血清中细胞因子水平均较对照组小鼠明显升高(P〈0.05),而db/db小鼠血清中细胞因子表达较诱发性糖尿病小鼠高,其中IL-6、IFN-γ、TNF-α具有显著性差异(P〈0.05)。结论两组2型糖尿病模型小鼠均在一定程度上模拟了人类糖尿病患者症状,但由于糖尿病产生的原因不同而存在着一定的差异,研究者可根据实际需要参照相关数据进行选择。  相似文献

本试验旨在研究金线莲多糖(ARP)对免疫抑制小鼠脾淋巴细胞体外增殖、NO及细胞因子IL-2、IL-6、IFN-γ分泌水平的影响。MTT法检测小鼠脾淋巴细胞体外增殖;Griess法检测NO分泌水平;ELISA法检测细胞因子IL-2、IL-6、IFN-γ的含量。结果显示,与对照组比较,ARP在50~400μg/m L可明显促进免疫抑制小鼠脾淋巴细胞体外增殖(P0.01),促进NO分泌(P0.01),促进细胞因子IL-2、IL-6和IFN-γ分泌(P0.05,P0.01)。以上结果提示ARP能提高免疫抑制小鼠脾淋巴细胞体外免疫活性,其作用机制可能与促进免疫抑制小鼠脾淋巴细胞增殖,促进NO产生以及提高IL-2、IL-6、IFN-γ的分泌水平有关。  相似文献

【目的】通过体内外实验评估5种乳杆菌缓解牛乳β-乳球蛋白(BLG)过敏的作用,为今后筛选具有抗过敏活性的乳杆菌提供参考。【方法】首先体外分析5种活的/热致死的乳杆菌促进小鼠原代淋巴细胞分泌细胞因子(CK)IFN-γ和IL-4的水平,随后应用小鼠BLG过敏模型评估这5种乳杆菌抑制过敏的能力。将实验动物随机分为空白组、BLG致敏组和5种活的/热致死乳杆菌组。采用ELISA法检测各组小鼠淋巴细胞分泌Thl/Th2型CK的水平,并测定小鼠血清中总IgE和BLG特异性IgE的含量。【结果】在体外可促进淋巴细胞分泌IFN-γ、抑制IL-4,使其IFN-γ/IL-4比值(代表Thl/Th2细胞平衡)显著高于正常对照组(P<0.05)的乳杆菌,在体内实验中也能有效提高致敏小鼠淋巴细胞的IFN-γ/IL-4分泌率,并显著降低致敏小鼠血清中总IgE和BLG特异性IgE的水平(P<0.05)。相反,在体外的IFN-γ/IL-4比值较低的乳杆菌,不能缓解特异性IgE抗体介导的食物过敏反应。【结论】基于乳杆菌体外刺激小鼠原代淋巴细胞分泌Th1/Th2型CK的结果,可以预测菌株在体内具有可通过纠正Th2占优势的Th1/Th2细胞失衡,下调抗体分泌量,缓解小鼠BLG过敏症状的能力。  相似文献

目的探讨双歧杆菌对过敏性哮喘儿童外周血单核细胞(PBMC)来源的树突状细胞(DC)分泌IL-1β、IL-6、IL-10、IL-12、IL-23和IFN-γ的影响。方法从15例过敏性哮喘儿童和15例非哮喘儿童的外周血单个核细胞诱导生成未成熟DC,加入双歧杆菌后继续培养DC2d,用ELISA方法检测培养上清中IL-1β、IL-6、IL-10、IL-12、IL-23和IFN-γ的水平。结果双歧杆菌能明显刺激哮喘儿童DC分泌IL-12、IFN-γ,IL-1β及IL-6和非哮喘儿童DC分泌IL-12、IL-10、IL-1β及IL-23水平增高。结论双歧杆菌能够刺激过敏性哮喘儿童DC分泌IL-12和IFN-γ,可能改变Th2优势分化,纠正Th1/Th2失衡。同时双歧杆菌还能刺激哮喘儿童DC分泌IL-1p及IL-6增高,达到促进,Th17细胞分化的作用。  相似文献

Combinatory responses of proinflamamtory cytokines have been examined on the nitric oxide-mediated function in cultured mouse calvarial osteoblasts. Interleukin-1β (IL-1β) and tumor necrosis factor-α (TNF-α) induced iNOS gene expression and NO production, although these actions were inhibited by L-NG-monomethylarginine (L-NMMA) and decreased alkaline phosphatase (ALPase) activity. Furthermore, NO donors, sodium nitroprusside (SNP) and NONOate dose-dependently elevated ALPase activity. In contrast, transforming-growth factor-β (TGF-β) decreased NO production stimulated by IL-1β, TNF-α and interferon-γ (IFN-γ). iNOS was expressed by mouse calvarial osteoblast cells after stimulation with IL-1β, TNF-α, and IFN-γ. Incubation of mouse calvarial osteoblast cells with the cytokines inhibited growth and ALPase activity. However, TGF-β-treatment abolished these effects of IL-1β, TNF-α and IFN-γ on growth inhibition and stimulation of ALPase in mouse calvarial osteoblast cells. In contrast, IL-1β, TNF-α, and IFN-γ exerted growth-inhibiting effects on mouse calvarial osteoblast cells which were partly NO-dependent. The results suggest that NO may act predominantly as a modulator of cytokine-induced effects on mouse calvarial osteoblast cells and TGF-β is a negative regulator of the NO production stimulated by IL-1β, TNF-α and IFN-γ.  相似文献

Regulation of macrophage activity by T(H)1/2 cytokines is important to maintain the balance of immunity to provide adequate protective immunity while avoiding excessive inflammation. IFN-γ and IL-4 are the hallmark T(H)1 and T(H)2 cytokines, respectively. In avian species, information concerning regulation of macrophage activity by T(H)1/2 cytokines is limited. Here, we investigated the regulatory function of chicken T(H)1 cytokines IFN-γ, IL-18 and T(H)2 cytokines IL-4, IL-10 on the HD11 macrophage cell line. Chicken IFN-γ stimulated nitric oxide (NO) synthesis in HD11 cells and primed the cells to produce significantly greater amounts of NO when exposed to microbial agonists, lipopolysaccharide, lipoteichoic acid, peptidoglycan, CpG-ODN, and poly I:C. In contrast, chicken IL-4 exhibited bi-directional immune regulatory activity: it activated macrophage NO synthesis in the absence of inflammatory agonists, but inhibited NO production by macrophages in response to microbial agonists. Both IFN-γ and IL-4, however, enhanced oxidative burst activity of the HD11 cells when exposed to Salmonella enteritidis. IL-18 and IL-10 did not affect NO production nor oxidative burst in HD11 cells. Phagocytosis and bacterial killing by the HD11 cells were not affected by the treatments of these cytokines. Infection of HD11 cells with S.enteritidis was shown to completely abolish NO production regardless of IFN-γ treatment. This study has demonstrated that IFN-γ and IL-4 are important T(H)1 and T(H)2 cytokines that regulate macrophage function in chickens.  相似文献

IL-4 and IFN-γ are prototypical Th2 and Th1 cytokines, respectively. They reciprocally regulate a number of genes involved in Th1 vs Th2 immune balance. Using DD-PCR analysis, adenine nucleotide translocase (ANT) 3, an enzyme which exchanges ATP and ADP through mitochondrial membrane, has been identified as a novel target counter-regulated by IL-4 and IFN-γ. We have observed that IL-4 and IFN-γ each up-regulates ANT3 in T cells both at mRNA and protein levels, while cotreatment of IL-4 and IFN-γ counter-regulates ANT3 expression. In contrast, other isoforms of ANT were not affected by IL-4 or IFN-γ. Emplyoing transfection and overexpression of STAT6 and STAT1 in STAT-deficient cells, we demonstrate that induction of ANT3 by IL-4 and IFN-γ proceeds via pathways involving STAT6 and STAT1, respectively. Furthermore, regulation of ANT3 expression by IL-4 and IFN-γ correlated with the modulation T cell survival by these cytokines from dex-induced apoptosis. Considering the critical role of mitochondrial ANTs in energy metabolism and apoptosis, ANT3 regulation by IL-4 and IFN-γ may have a functional implication in cytokine-mediated T cell survival.  相似文献

目的通过复制轮状病毒(RV)肠道外感染乳鼠的动物模型,检测接种后乳鼠体内Th1/Th2平衡改变,对RV肠道外感染后机体免疫状态进行初步研究。方法48只乳鼠随机均分为3组:肠道外组、肠道内组和正常对照组。肠道外组通过腹腔注射猴RVSA11株,肠道内组灌胃等量RV悬液,对照组无特殊处理。分别在接种后第4天、第8天处死乳鼠,收集标本,观察心、肝、肾、肺等脏器病理变化,用ELISA法检测血清中IL-10和IFN-γ的表达。结果光镜下肠道外组乳鼠肾、肝、肺和脾脏出现病理改变。感染后第4天,肠道内、外组乳鼠血清IFN-γ水平均高于正常组,到第8天明显下降,基本达到基线水平;IL-10在肠道外组第4天增高,到第8天小幅下降,但仍然高于正常组;而肠道内组IL-10无明显改变。结论RV肠道外感染早期呈现Th1-Th2混合反应,而后期则以IL-10的表达为主,T细胞向Th2型免疫应答方向偏离,Th1/Th2细胞因子失衡机制可能是RV肠道外感染的重要因素。  相似文献

IFN-γ-mediated inducible nitric oxide synthase (iNOS) expression is critical for controlling chlamydial infection through microbicidal nitric oxide (NO) production. Interleukin-17A (IL-17A), as a new proinflammatory cytokine, has been shown to play a protective role in host defense against Chlamydia muridarum (Cm) infection. To define the related mechanism, we investigated, in the present study, the effect of IL-17A on IFN-γ induced iNOS expression and NO production during Cm infection in vitro and in vivo. Our data showed that IL-17A significantly enhanced IFN-γ-induced iNOS expression and NO production and inhibited Cm growth in Cm-infected murine lung epithelial (TC-1) cells. The synergistic effect of IL-17A and IFN-γ on Chlamydia clearance from TC-1 cells correlated with iNOS induction. Since one of the main antimicrobial mechanisms of activated macrophages is the release of NO, we also examined the inhibitory effect of IL-17A and IFN-γ on Cm growth in peritoneal macrophages. IL-17A (10 ng/ml) synergizes with IFN-γ (200 U/ml) in macrophages to inhibit Cm growth. This effect was largely reversed by aminoguanidine (AG), an iNOS inhibitor. Finally, neutralization of IL-17A in Cm infected mice resulted in reduced iNOS expression in the lung and higher Cm growth. Taken together, the results indicate that IL-17A and IFN-γ play a synergistic role in inhibiting chlamydial lung infection, at least partially through enhancing iNOS expression and NO production in epithelial cells and macrophages.  相似文献

Although intraocular tumors reside in an immune-privileged site where immune responses are suppressed, some tumors are rejected. An example of this is the rejection of intraocular adenovirus-induced (adenovirus type 5 early region 1 [Ad5E1]) tumors in C57BL/6 mice. We previously identified an Ad5E1 tumor clone in which the rejection is IFN-γ dependent and culminates in the destruction of both the tumor and the eye. Although Ad5E1 tumors are not rejected when transplanted into the eyes of IFN-γ KO mice, they are rejected after s.c. transplantation. Thus, outside of the eye Ad5E1 tumors elicit a form of tumor immunity that is IFN-γ independent. In this article, we demonstrate that IFN-γ-independent s.c. rejection requires both CD4(+) and CD8(+) T cells. Furthermore, s.c. tumor rejection requires IL-17, which is produced by IFN-γ-deficient CD4(+) T cells in response to tumor Ags (TAs). Splenocytes from CD4-depleted IFN-γ KO mice produce significantly less IL-17 compared with splenocytes from isotype-treated IFN-γ KO animals in response to TAs. Furthermore, depletion of IL-17 decreases CTL activity against Ad5E1 tumor cells. In this model we propose that, in the absence of IFN-γ, CD4(+) T cells produce IL-17 in response to TAs, which increases CTL activity that mediates tumor rejection; however, this does not occur in the eye. IL-6 production within the eye is severely reduced, which is consistent with the failure to induce Th17 cells within the intraocular tumors. In contrast, the s.c. environment is replete with IL-6 and supports the induction of Th17 cells. Therefore, IFN-γ-independent tumor rejection is excluded from the eye and may represent a newly recognized form of ocular immune privilege.  相似文献

目的:探讨大麻素CBR2受体激动剂AM1241预处理对脂多糖(LPS)和γ-干扰素(IFN-γ)所致炎症反应对小胶质细胞活化和损伤的影响。方法:联用LPS和IFN-γ作为小胶质细胞损伤模型,将细胞分为Control组、AM1241组、LPS/IFN-γ组和AM1241+LPS/IFN-γ组;AM1241组和AM1241+LPS/IFN-γ组经AM1241预处理2h,LPS/IFN-γ组和AM1241+LPS/IFN-γ组用含LPS和IFN-γ的培养基培养24h。采用MTT法检测细胞代谢率,硝酸还原酶法检测细胞培养液中一氧化氮(NO)释放量,酶联免疫吸附剂测定细胞培养基中炎症因子释放量,倒置相差显微镜观察细胞形态。结果:与LPS/IFN-γ组相比,AM1241+LPS/IFN-γ组细胞代谢率明显升高(P〈0.05),NO、TNF-α、IL-1β和IL-10释放量明显减少(P〈0.05),活化和损伤程度明显减轻。结论:大麻素CBR2受体激动剂AM1241预处理可减轻LPS和IFN-γ对小胶质细胞的活化和损伤。  相似文献
