The oxygen evolving complex of Photosystem II undergoes four light-induced oxidation transitions, S0-S1,…,S3-(S4)S0 during its catalytic cycle. The oxidizing equivalents are stored at a (Mn)4Ca cluster, the site of water oxidation. EPR spectroscopy has yielded valuable information on the S states. S2 shows a notable heterogeneity with two spectral forms; a g = 2 (S = 1/2) multiline, and a g = 4.1 (S = 5/2) signal. These oscillate in parallel during the period-four cycle. Cyanobacteria show only the multiline signal, but upon advancement to S3 they exhibit the same characteristic g = 10 (S = 3) absorption with plant preparations, implying that this latter signal results from the multiline configuration. The fate of the g = 4.1 conformation during advancement to S3 is accordingly unknown. We searched for light-induced transient changes in the EPR spectra at temperatures below and above the half-inhibition temperature for the S2 to S3 transition (ca 230 K). We observed that, above about 220 K the g = 4.1 signal converts to a multiline form prior to advancement to S3. We cannot exclude that the conversion results from visible-light excitation of the Mn cluster itself. The fact however, that the conversion coincides with the onset of the S2 to S3 transition, suggests that it is triggered by the charge-separation process, possibly the oxidation of tyr Z and the accompanying proton relocations. It therefore appears that a configuration of (Mn)4Ca with a low-spin ground state advances to S3.  相似文献

Mitochondria from respiring cells were isolated under anaerobic conditions. Microscopic images were largely devoid of contaminants, and samples consumed O2 in an NADH-dependent manner. Protein and metal concentrations of packed mitochondria were determined, as was the percentage of external void volume. Samples were similarly packed into electron paramagnetic resonance tubes, either in the as-isolated state or after exposure to various reagents. Analyses revealed two signals originating from species that could be removed by chelation, including rhombic Fe3+ (g = 4.3) and aqueous Mn2+ ions (g = 2.00 with Mn-based hyperfine). Three S = 5/2 signals from Fe3+ hemes were observed, probably arising from cytochrome c peroxidase and the a3:Cub site of cytochrome c oxidase. Three Fe/S-based signals were observed, with averaged g values of 1.94, 1.90 and 2.01. These probably arise, respectively, from the [Fe2S2]+ cluster of succinate dehydrogenase, the [Fe2S2]+ cluster of the Rieske protein of cytochrome bc 1, and the [Fe3S4]+ cluster of aconitase, homoaconitase or succinate dehydrogenase. Also observed was a low-intensity isotropic g = 2.00 signal arising from organic-based radicals, and a broad signal with g ave = 2.02. Mössbauer spectra of intact mitochondria were dominated by signals from Fe4S4 clusters (60–85% of Fe). The major feature in as-isolated samples, and in samples treated with ethylenebis(oxyethylenenitrilo)tetraacetic acid, dithionite or O2, was a quadrupole doublet with ΔE Q = 1.15 mm/s and δ = 0.45 mm/s, assigned to [Fe4S4]2+ clusters. Substantial high-spin non-heme Fe2+ (up to 20%) and Fe3+ (up to 15%) species were observed. The distribution of Fe was qualitatively similar to that suggested by the mitochondrial proteome.  相似文献

Biotin synthase (BioB) is an S-adenosylmethionine radical enzyme that catalyzes addition of sulfur to dethiobiotin to form the biotin thiophane ring. In vitro, Escherichia coli BioB is active for only one turnover, during which the [2Fe-2S]2+ cluster is destroyed, one sulfide from the cluster is incorporated as the biotin thiophane sulfur, while Fe2+ ions and the remaining S2− ion are released from the protein. The present work examines the fate of the protein following the loss of the FeS clusters. We examine the quaternary structure and thermal stability of active and inactive states of BioB, and find that loss of either the [4Fe-4S]2+ or [2Fe-2S]2+ clusters results in destabilization but not global unfolding of BioB. Using susceptibility to limited proteolysis as a guide, we find that specific regions of the protein appear to be transiently unfolded following loss of these clusters. We also examine the in vivo degradation of biotin synthase during growth in low-iron minimal media and find that BioB is degraded by an apparent ATP-dependent proteolysis mechanism that sequentially cleaves small fragments starting at the C-terminus. BioB appears to be resistant to degradation and capable of multiple turnovers only under high-iron conditions that favor repair of the FeS clusters, a process most likely mediated by the Isc or Suf iron-sulfur cluster assembly systems.  相似文献

Electronic structures of chalcogenide-bridged binuclear clusters of vanadium and niobium with the {M2(μ-Q2)2}4+, {M2(μ-Q)2}4+ and {M2(μ-Q)(μ-Q)2}4+ cores (Q = S, Se, Te) have been studied by density functional theory methods. In the vanadium clusters, the V-V distances are calculated to be in the range of 2.766-3.193 ?, whereas in the niobium clusters the calculated Nb-Nb bond lengths fall in the range of 2.881-3.380 ?, in accordance with the experimentally determined values. The calculated M-M bond distances generally decrease in the order {M2(μ-Q2)2}4+ > {M2(μ-Q)2}4+ > {M2(μ-Q)(μ-Q)2}4+ (M = V, Nb, Q = S, Se). The calculated enthalpies of formation for the V clusters are higher than for the corresponding Nb clusters. On the other hand, the M2Q2 clusters have always higher enthalpies of formation than the M2Q3 species, and also (with the exception of M = V, Q = S) higher values of enthalpy of formation than for the M2Q4 species. The hardness η of the niobium clusters are higher than that of the vanadium analogs, except for the [V2S2(SH2)8]4+ case. The enthalpies ΔH298 and the free energies ΔG298 for the reactions of hydrogen addition to the [V2(μ-S2)2(H2O)8]4+ and the [Nb2(μ-S2)2(H2O)8]4+ clusters at constant pressure are −121.75 and −59.73 kJ/mol for the vanadium cluster, and 13.97 and 75.15 kJ/mol for the niobium cluster.  相似文献

The in-situ formed hydrazone Schiff base ligand (E)-N′-(2-oxy-3-methoxybenzylidene)benzohydrazide (L2−) reacts with copper(II) acetate to a tetranuclear open cubane [Cu(L)]4 complex which crystallizes as two symmetry-independent (Z′ = 2) S4-symmetrical molecules in different twofold special positions with a homodromic water tetramer. The two independent (A and B) open- or pseudo-cubanes with Cu4O4 cores of 4 + 2 class (Ruiz classification) each have three different magnetic exchange pathways leading to an overall antiferromagnetic coupling with J1B = J2B = −17.2 cm−1, J1A = −36.7 cm−1, J2A = −159 cm−1, J3A = J3B = 33.5 cm−1, g = 2.40 and ρ = 0.0687. The magnetic properties have been analysed using the H = −Σi,jJij(SiSj) spin Hamiltonian.  相似文献

4 S4]3 +  and the reduced [Fe4S4]2 +  clusters in the high-potential iron protein I from Ectothiorhodospira halophila were measured in a temperature range from 5 K to 240 K. EPR measurements and 57Fe electron-nuclear double resonance (ENDOR) experiments were carried out with the oxidized protein. In the oxidized state the cluster has a net spin S = 1/2 and is paramagnetic. As common in [Fe4S4]3 +  clusters, the M?ssbauer spectrum was simulated with two species contributing equally to the absorption area: two Fe3 +  atoms couple to the “ferric-ferric” pair, and one Fe2 +  and one Fe3 +  atom give the “ferric-ferrous pair”. For the simulation of the M?ssbauer spectrum, g-values were taken from EPR measurements. A-tensor components were determined by 57Fe ENDOR experiments that turned out to be a necessary source of estimating parameters independently. In order to obtain a detailed agreement of M?ssbauer and ENDOR data, electronic relaxation has to be taken into account. Relaxing the symmetry condition in a way that the electric field gradient tensor does not coincide with g- and A-tensors yielded an even better agreement of experimental and theoretical M?ssbauer spectra. Spin-spin and spin-lattice relaxation times were estimated by pulsed EPR; the former turned out to be the dominating mechanism at T = 5 K. Relaxation times measured by pulsed EPR and obtained from the M?ssbauer fit were compared and yield nearly identical values. The reduced cluster has one additional electron and has a diamagnetic (S = 0) ground state. All the four irons are indistinguishable in the M?ssbauer spectrum, indicating a mixed-valence state of Fe2.5 +  for each. Received: 15 February 1999 / Accepted: 31 August 1999  相似文献

Clusters [MoS4Ag3(PPh3)3{S2P(OPri)2}] (1), [WS4Ag3(PPh3)3{S2P(OPri)2}] (2) and [WOS3Ag3(PPh3)3{S2P(OPri)2}] (3) were synthesized by the reaction of (NH4)2MoS4/(NH4)2WS4, (NH4)2WOS3 with Ag[S2P(OPri)2]. Their structures have been characterized by X-ray diffraction. The clusters consist of a distorted tetrahedral MS4 (or MOS3) (M = Mo, W) with three Ag atoms and three sulfur atom bridges (Fig. 1), and resemble roughly that of cubane-like clusters. The nonlinear optical (NLO) properties were studied with an 8 ns pulsed laser at 532 nm. Its optical response to the incident light exhibits good optical absorptive and refractive effects, with α2 = 1.56 × 10−10 m W−1, n2 = 3.87 × 10−17 m2 W−1 for cluster 1; α2 = 1.33 × 10−10 m W−1n2 = 6.52 × 10−17 m2 W−1for cluster 2; and α2 = 2.54 × 10−10 m W−1, n2 = 4.07 × 10−17 m2 W−1 for cluster 3 for a 1.56 × 10−4 mol dm−3 CH2Cl2 solution.  相似文献

The pentagonal bipyramidal high-spin iron(II) complex, [(TPA2C(O)NHtBu)Fe(CF3SO3)]+, is shown to exhibit a high-anisotropy ground state, with fits to dc magnetization data providing an axial zero-field splitting parameter of D = − 7.9 cm−1. The utility of this compound as a building unit is demonstrated, as its reaction with [ReCl4(CN)2]2− affords the cyano-bridged dinuclear cluster (TPA2C(O)NHtBu)FeReCl4(CN)2. dc magnetic susceptibility measurements reveal intracluster ferromagnetic exchange interactions between FeII and ReIV centers, with J = +3.0 cm−1, giving rise to a spin ground state of S = 7/2. Moreover, fits to dc magnetization data obtained for the FeRe cluster show the presence of strong axial anisotropy, with D = −2.3 cm−1. Finally, variable-frequency ac susceptibility measurements reveal the onset of slow magnetic relaxation at low temperature, suggesting that the FeRe cluster is a single-molecule magnet.  相似文献

We have employed EPR and a set of recently developed electron nuclear double resonance (ENDOR) spectroscopies to characterize a suite of [2Fe?C2S] ferredoxin clusters from Aquifex aeolicus (Aae Fd1, Fd4, and Fd5). Antiferromagnetic coupling between the FeII, S?=?2, and FeIII, S?=?5/2, sites of the [2Fe?C2S]+ cluster in these proteins creates an S?=?1/2 ground state. A complete discussion of the spin-Hamiltonian contributions to g includes new symmetry arguments along with references to related FeS model compounds and their symmetry and EPR properties. Complete 57Fe hyperfine coupling (hfc) tensors for each iron, with respective orientations relative to g, have been determined by the use of ??stochastic?? continuous wave and/or ??random hopped?? pulsed ENDOR, with the relative utility of the two approaches being emphasized. The reported hyperfine tensors include absolute signs determined by a modified pulsed ENDOR saturation and recovery (PESTRE) technique, RD-PESTRE??a post-processing protocol of the ??raw data?? that comprises an ENDOR spectrum. The 57Fe hyperfine tensor components found by ENDOR are nicely consistent with those previously found by M?ssbauer spectroscopy, while accurate tensor orientations are unique to the ENDOR approach. These measurements demonstrate the capabilities of the newly developed methods. The high-precision hfc tensors serve as a benchmark for this class of FeS proteins, while the variation in the 57Fe hfc tensors as a function of symmetry in these small FeS clusters provides a reference for higher-nuclearity FeS clusters, such as those found in nitrogenase.  相似文献

The octanuclear cyano-bridged cluster [(Tp)8Fe4Ni4(CN)12] · H2O · 24CH3CN (1) (Tp = hydrotris(1-pyrazolyl)borate) showing magnetic properties of single-molecule magnet has been synthesized by reaction of [fac-Fe(Tp)(CN)3] with {(Tp)Ni(NO3)} species formed from an equimolar reaction mixture of Ni(NO3)2 · 6H2O and KTp in MeCN. The X-ray analysis of 1 shows molecular cube structure in which FeIII and NiII ions reside in alternate corners. The average intramolecular Fe?Ni distance is 5.124 Å. Out-of-phase ac susceptibility and reduce magnetization measurements show that 1 is a single molecule magnet with ground spin state S = 6 and spin reversal energy barrier U = 14 K. Magnetic hysteresis loops were also observed by applying fast sweeping field.  相似文献

The reactions of mass-selected iron clusters Fen + (n=1-5) with dimethyl carbonate, (CH3O)2CO, are examined by means of Fourier-transform ion-cyclotron-resonance mass spectrometry. For the bare metal cation Fe+, loss of a methyl radical prevails which leads to the iron carbonate species FeOC(O)OCH3 +. For the corresponding Fen + clusters, this type of reaction is not observed anymore. Instead, the clusters show a strong tendency for a formal O-atom abstraction leading to the formation of the corresponding monoxide clusters FenO+ In addition, several bond activations of dimethyl carbonate are observed which markedly differ from the behavior of the mononuclear cation. Nevertheless, a mechanistic analysis implies that the initial steps are the same for bare Fe+ as well as small Fen + clusters.  相似文献

Four trinuclear Cu(II) complexes, [(CuL1)33-OH)](NO3)2 (1), [(CuL2)33-OH)](I)2·H2O (2), [(CuL3)33-OH)](I)2 (3) and [(CuL1)33-OH)][CuII3] (4), where HL1 (8-amino-4-methyl-5-azaoct-3-en-2-one), HL2 [7-amino-4-methyl-5-azaoct-3-en-2-one] and HL3 [7-amino-4-methyl-5-azahept-3-en-2-one] are the three tridentate Schiff bases, have been synthesized and structurally characterized by X-ray crystallography. All four complexes contain a partial cubane core, [(CuL)33-OH)]2+ in which the three [CuL] subunits are interconnected through two types of oxygen bridges afforded by the oxygen atoms of the ligands and the central OH group. The copper(II) ions are in a distorted square-pyramidal environment. The equatorial plane consists of the bridging oxygen of the central OH group together with three atoms (N, N, O) from the Schiff base. The oxygen atom of the Schiff base also coordinates to the axial position of Cu(II) of another subunit to form the cyclic trimer. Magnetic susceptibilities have been determined for these complexes over the temperature range of 2-300 K. The isotropic Hamiltonian, H = −J12S1S2 − J13S1S3 − J23S2S3 has been used to interpret the magnetic data. The best fit parameters obtained are: J = −54.98 cm−1, g = 2.24 for 1; J = −56.66 cm−1, g = 2.19 for 2;J = −44.39 cm−1, g = 2.16 for 3; J = −89.92 cm−1, g = 2.25 for 4. The EPR data at low temperature indicate that the phenomenon of spin frustration occurs for complexes 1-3.  相似文献

A mononuclear cobalt(II) complex, [Co(ac)2(H2O)2(MeIm)2], with heteroleptic coordination sphere possessing the {CoO2O′2N2} chromophore has been prepared and structurally characterized. The magnetic data down to 2 K show an enhanced magnetic anisotropy manifesting itself in a large zero-field splitting (ZFS) parameter. As a consequence, the magnetization deviates substantially from the Brillouin-function behavior. A fit to the zero-field splitting model gave the following set of magnetic parameters: D/hc = +95 cm−1, gx = 2.530, zj/hc = −0.078, χTIP = 16.7 × 10−9 m3 mol−1, (gz = 2.0). The Griffith-Figgis model and the Generalized Crystal-Field model lie beyond the spin-Hamiltonian formalism; they gave analogous, although not identical ZFS parameters: D/hc = 109 cm−1, and D/hc = 77 cm−1, respectively. The absorption spectrum taken in the FAR-IR region exhibits manifold absorption peaks referring to the transitions among the crystal-field multiplets of the parent 4A2g + 4Eg terms (D4h), originating in a crystal-field splitting of the octahedral 4T1g ground term.  相似文献

Salts of the Fe(III) spin crossover cation [FeIII(qsal)2]+ (qsalH = N-(8-quinolyl)salicylaldimine) and monoanions [MIII(pds)2] (M = Cu, Au; pds = pirazine-2,3-diselenolate) with formula [FeIII(qsal)2][MIII(pds)2] were prepared and characterized by single crystal X-ray diffraction and magnetic measurements. These two salts present magnetic properties essentially due to the FeIII centres in the high-spin state (S = 5/2), and do not have any spin transition.  相似文献

The crystal structure of the membrane-associated [NiFe] hydrogenase from Allochromatium vinosum has been determined to 2.1 Å resolution. Electron paramagnetic resonance (EPR) and Fourier transform infrared spectroscopy on dissolved crystals showed that it is present in the Ni-A state (> 90%). The structure of the A. vinosum [NiFe] hydrogenase shows significant similarities with [NiFe] hydrogenase structures derived from Desulfovibrio species. The amino acid sequence identity is ∼ 50%. The bimetallic [NiFe] active site is located in the large subunit of the heterodimer and possesses three diatomic non-protein ligands coordinated to the Fe (two CN , one CO). Ni is bound to the protein backbone via four cysteine thiolates; two of them also bridge the two metals. One of the bridging cysteines (Cys64) exhibits a modified thiolate in part of the sample. A mono-oxo bridging ligand was assigned between the metal ions of the catalytic center. This is in contrast to a proposal for Desulfovibrio sp. hydrogenases that show a di-oxo species in this position for the Ni-A state. The additional metal site located in the large subunit appears to be a Mg2+ ion. Three iron-sulfur clusters were found in the small subunit that forms the electron transfer chain connecting the catalytic site with the molecular surface. The calculated anomalous Fourier map indicates a distorted proximal iron-sulfur cluster in part of the crystals. This altered proximal cluster is supposed to be paramagnetic and is exchange coupled to the Ni3+ ion and the medial [Fe3S4]+ cluster that are both EPR active (S = 1/2 species). This finding of a modified proximal cluster in the [NiFe] hydrogenase might explain the observation of split EPR signals that are occasionally detected in the oxidized state of membrane-bound [NiFe] hydrogenases as from A. vinosum.  相似文献

Substitution reaction of fac-[FeII(CN)2(CO)3I] with triphenylphosphine (PPh3) produced mono phosphine substituted complex cis-cis-[FeII(CN)2(CO)2(PPh3)I]. Crystal structure of the product showed that carbonyl positioned trans- to iodide was replaced by PPh3. The substitution reaction was monitored by quantitative infrared spectroscopic method, and the rate law for the substitution reaction was determined to be rate = k[[FeII(CN)2(CO)2(PPh3)I]][PPh3]. Transition state enthalpy and entropy changes were obtained from Eyring equation k = (kBT/h)exp(−ΔH/RT + ΔS/R) with ΔH = 119(4) kJ mol−1 and ΔS = 102(10) J mol−1 K−1. Positive transition state entropy change suggests that the substitution reaction went through a dissociative pathway.  相似文献

The reaction of [FeII(H2O)6](BF4)2 with tris(2-pyridylmethyl)amine (TPyA) and triethylamine in methanol under aerobic conditions forms [(TPyA)FFeIIIOFeIIIF(TPyA)](BF4)2 · 0.5MeOH (1), in which each Fe(III) ion is coordinated to a TPyA and an F ion as well as an oxo ion (O2−) linking two Fe(III) ions. 1 has offset face-to-face π-π interactions between the dimers, and possesses a supramolecular network structure. The magnetic susceptibility of 1 can be fit with g = 2.0, J/kB = − 153 K (106 cm−1), and θ = − 0.3 K [H = − 2JSa · Sb]. These indicate that very strong antiferromagnetic interactions occur via the oxo bridge within the Fe(III) dimer and weak antiferromagnetic interactions between the dimers.  相似文献

Lanthanide complexes are of great importance for their prospective applications in wide range of science and technology. Chiral lanthanide complexes can constitute stereo-discriminating probes in biological media, owing to the luminescent properties of the rare-earth ions. Sensitized emission with narrow bandwidth, having fast radiation rate and high emission quantum efficiency are the main perspective for synthesizing the complexes. Attention has been given on remarkable chirality with high dissymmetry factors (g = Δεext/εmax) of the complexes. For this purpose, beta-diketonato ligands with chiral BINAPO (1,1′-binapthyl phosphine oxide) ligand were chosen to achieve the goal. The complexes [Ln(TFN)3(S-BINAPO)](TFN = 4,4,4-trifluoro-1(2-napthyl)-1,3-butanedione), [Ln(HFT)3(S-BINAPO)] (HFT = 4,4,5,5,6,6,6-heptafluoro-1-(2-thienyl)-1,3-hexanedione) and [Ln(HFA)3(S-BINAPO)](hfa = hexafluoroacetylacetonate) (where Ln = Yb, Eu) were synthesized. The complex, [Eu(TFN)3(S-BINAPO)] gives strong red emission at 615 nm with narrow emission band (<10 nm) when excited by 465 nm light with quantum efficiency 86%. The dissymmetry factors (g = Δεext/εmax) corresponding to the 7F1 → 5D0 transition at 590 nm is 0.091 for [Eu(TFN)3(S-BINAPO)] and for [Yb(hfa)3(S-BINAPO)](hfa = hexafluoroacetylacetonate) corresponding to the 2F7/2 → 2F5/2 transitions is 0.12, are among the largest values for both Eu and Yb complexes to date, respectively. The Eu complexes, [Eu(HFT)3(S-BINAPO)] and [Eu(TFN)3(S-BINAPO)] are found to be spontaneously emissive, showing bright red emission, when placed in sunlight or even in the laboratory when light is switched on.  相似文献

Using the tetracyanometalate precursor [Fe(4,4′-dmbipy)(CN)4]- (4,4′-dmbipy = 4,4′-dimethyl-2,2′-bipyridine) as the building block, two new cyano-bridged one-dimensional heterobimetallic coordination polymers, [M(CH3OH)2Fe2(4,4′-dmbipy)2(CN)8]n (M = Cu, 1; Mn, 2), have been synthesized and structurally characterized. X-ray crystallography reveals that complexes 1 and 2 consist of heterobimetallic chains of squares, and the central MII ion is six-coordinated as an elongated distorted octahedral geometry. Magnetic studies show ferromagnetic coupling between FeIII and CuII ions in complex 1. Complex 2 exhibits ferrimagnetic behavior caused by the noncompensation of the local interacting spins (SMn = 5/2 and SFe = 1/2), which interact antiferromagnetically through bridging cyanide groups. magpack program has been employed to investigate the magnetic nature of squares chain structure.  相似文献

The mesostructured lamellar phases with the general formula [CnH2n + 1NH3]4Ge4S10 (n = 12, 14, 16, 18) were synthesized by metathesis of alkyl ammonium chloride surfactants and Na4Ge4S10 in aqueous medium. The crystal structures of the phases with n = 12 and 14 were determined by single-crystal X-ray diffraction; [C12H13NH3]4Ge4S10 crystallizes in monoclinic C2/c space group (a = 16.149(3), b = 46.576(9) c = 9.147(2) Å, β = 97.13(3)° and Z = 4) and [C14H29NH3]4Ge4S10 in the triclinic space group (a = 9.1280(6), b = 16.1992(1), c = 26.971(2) Å, α =73.370(1)°, β = 88.307(1)°, γ = 82.825(1)° and Z = 2). These compounds possess layers of adamantane [Ge4S10]4− anions separated by layers of deeply interpenetrated long chain alkylammonium molecules. Strong hydrogen bonding is observed between the terminal sulfur atoms of the [Ge4S10]4− clusters and H atoms of the NH3 groups of the surfactant molecules. Spectroscopic and thermal characterization of these compounds is reported.  相似文献
