脂肪酸不仅是细菌细胞膜组分,还是许多生物活性物质的合成原料。不饱和脂肪酸(unsaturated fatty acid, UFA)具有更低的相变温度,是细菌调节细胞膜流动性的重要分子,因此UFA合成途径是重要的抗菌药物筛选靶点。细菌可利用厌氧途径合成UFA,其中模式生物大肠杆菌利用经典的FabA-FabB途径合成UFA,但不同细菌中UFA合成的厌氧途径具有多样性,相关催化酶类也不尽相同;细菌还可以利用需氧途径合成UFA,利用脂肪酸脱饱和酶直接将饱和脂肪酸(saturated fatty acid, SFA)转化为不饱和脂肪酸,而不同脱饱和酶会生成不同结构的UFA,在逆境耐受、致病力等多方面发挥重要作用;细菌还可以利用单加氧酶,将脂肪酸合成途径中癸酰酰基载体蛋白(acyl carrier protein, ACP)转化为顺-3-癸烯酰ACP,并最终合成UFA。细菌脂肪酸合成相关的其他酶类在UFA合成或不同种类UFA调节中也发挥着重要作用。本文系统地总结了细菌UFA合成途径与相关酶类的多样性研究进展,旨在为进一步了解细菌UFA合成机制,并以此为靶点开发抗菌药物等方面提供理论支撑。  相似文献

Results from our previous studies demonstrated that activation of Toll-like receptor 4 (Tlr4), the lipopolysaccharide (LPS) receptor, is sufficient to induce nuclear factor kappaB activation and expression of inducible cyclooxygenase (COX-2) in macrophages. Saturated fatty acids (SFAs) acylated in lipid A moiety of LPS are essential for biological activities of LPS. Thus, we determined whether these fatty acids modulate LPS-induced signaling pathways and COX-2 expression in monocyte/macrophage cells (RAW 264.7). Results show that SFAs, but not unsaturated fatty acids (UFAs), induce nuclear factor kappaB activation and expression of COX-2 and other inflammatory markers. This induction is inhibited by a dominant-negative Tlr4. UFAs inhibit COX-2 expression induced by SFAs, constitutively active Tlr4, or LPS. However, UFAs fail to inhibit COX-2 expression induced by activation of signaling components downstream of Tlr4. Together, these results suggest that both SFA-induced COX-2 expression and its inhibition by UFAs are mediated through a common signaling pathway derived from Tlr4. These results represent a novel mechanism by which fatty acids modulate signaling pathways and target gene expression. Furthermore, these results suggest a possibility that propensity of monocyte/macrophage activation is modulated through Tlr4 by different types of free fatty acids, which in turn can be altered by kinds of dietary fat consumed.  相似文献

The lipid composition of cell membranes exerts a crucial influence on cell physiology. Indeed, one double bond triggers membrane fluidity, essential for cell functionality, but additional double bonds increase the susceptibility to peroxidation, which produces reactive compounds that impair the viability of cells. It has therefore been suggested, but never tested in an extensive comparative context, that the composition of membrane fatty acids has been optimized during evolution. A similar prediction has been made for fatty acid chain length, on which susceptibility to peroxidation also depends. Here I tested for stabilizing selection on fatty acid composition by evaluating the fitting of the single stationary peak (SSP) model of evolution to a large data set from 107 species of birds, against alternative evolutionary models. I found that across‐species variation in average chain length and in the proportion of monounsaturated fatty acids (MUFAs), but not in the proportion of polyunsaturated (PUFAs) nor saturated (SFAs) fatty acids, was better explained by SSP models than by other models. Results show optimum values of fatty acid chain length and proportion of MUFAs of 18 C atoms and 25.5% mol, respectively, the strength of stabilizing selection being particularly high in chain length. This is the first evidence of evolutionary optimization in fatty acid composition, suggesting that certain values may have been selected because of their adaptive capacity to minimize susceptibility to lipid peroxidation.  相似文献

The degree to which fatty acids modulate brain function beyond periods of rapid brain growth is poorly understood. Nevertheless, recent evidence suggests that dietary fatty acid composition influences numerous behaviors including body temperature regulation, pain sensitivity, feeding behavior including macronutrient selection, and cognitive performance. Importantly, alterations are observed in the absence of essential fatty acid (EFA) deficiency, beyond periods of rapid brain development, and at levels similar to those consumed by the North American population. Data suggest that the content of saturated fatty acids (SFAs), and not that of the EFAs, may be the important component of dietary fat mediating macronutrient selection and cognition under these experimental conditions. Yet, a direct role of SFAs in modulating brain functions has not been elucidated. A discussion of potential mechanisms which may directly involve the central nervous system, or may indirectly influence central processes via peripheral pathway(s) is presented.  相似文献

Type 1 diabetic patients with varying severity of kidney disease were investigated to create multimetabolite models of the disease process. Urinary albumin excretion rate was measured for 3358 patients with type 1 diabetes. Prospective records were available for 1051 patients, of whom 163 showed progression of albuminuria (8.3-year follow-up), and 162 were selected as stable controls. At baseline, serum lipids, lipoprotein subclasses, and low-molecular weight metabolites were quantified by NMR spectroscopy (325 samples). The data were analyzed by the self-organizing map. In cross-sectional analyses, patients with no complications had low serum lipids, less inflammation, and better glycemic control, whereas patients with advanced kidney disease had high serum cystatin-C and sphingomyelin. These phenotype extremes shared low unsaturated fatty acids (UFAs) and phospholipids. Prospectively, progressive albuminuria was associated with high UFAs, phospholipids, and IDL and LDL lipids. Progression at longer duration was associated with high HDL lipids, whereas earlier progression was associated with poor glycemic control, increased saturated fatty acids (SFAs), and inflammation. Diabetic kidney disease consists of diverse metabolic phenotypes: UFAs, phospholipids, IDL, and LDL may be important in the subclinical phase, high SFAs and low HDL suggest accelerated progression, and the sphingolipid pathway in advanced kidney injury deserves further research.  相似文献

The effect of the carbon to nitrogen (C/N) ratio of the medium and the aeration rate on the lipid content and fatty acid composition ofChlorella sorokiniana was investigated using heterotrophic, batch culture. Both parameters had a significant effect. A C/N ratio of approximately 20, was found to indicate a change from carbon to nitrogen limitation forC. sorokiniana. Cell lipid content was at a minimum at this value and increased at both higher and lower C/N values. Low C/N ratios favoured a high proportion of trienoic fatty acids at the expense of monoenoic acids. Aeration enhanced cell growth, fatty acid yield and the synthesis of unsaturated dienoic and trienoic fatty acids, but reduced cell lipid content. The results demonstrate that the fatty acid composition and lipid content of heterotrophically-grown microalgae can be favourably manipulated by varying culture conditions.  相似文献

This study was designed to examine the effect of thermal acclimation on the lipid composition of fat depot organs the liver and kidneys of larval sea lamprey, Petromyzon marinus. We found that 21 °C-acclimated larvae possessed lower total lipid amounts in the liver (39% lower) and kidneys (30% lower) than 13 °C-acclimated larvae. Relatively lower lipid contents in the liver and kidneys of 21 °C-acclimated lamprey primarily resulted from a reduction in stored lipid reserve, triacylglycerol, but not the structural lipid, phospholipid. Compared to 21 °C-acclimated larvae, 13 °C-acclimated larvae were found to possess fewer saturated fatty acids (SFAs) and more unsaturated fatty acids (USFAs) in renal triacylglycerol and phospholipid classes, while there were no significant differences in the SFAs and USFAs of hepatic triacylglycerol, phospholipid, cholesteryl ester, fatty acid, and monoacylglycerol classes. Fewer SFAs, found in the kidney triacylglycerol of 13 °C-acclimated lamprey, were due to lower 12:0 and 14:0 fatty acids, but those in the renal phospholipid class were characterized by fewer 14:0, 15:0, and 16:0 fatty acids. More USFAs in renal triacylglycerol, as indicated by a higher unsaturation index, primarily resulted from higher polyunsaturated fatty acids (18:2ω6, 18:3ω3, and 18:4ω3); whereas, in the renal phospholipid class, this was a result of higher monoenes (18:1, 20:1, and 22:1ω9) and ω3 polyunsaturated fatty acids (18:4ω3). These data suggest that the influence of thermal acclimation on the lipid composition of lamprey fat depot organs depends on tissue and lipid class.  相似文献

Cold resistance in insects has traditionally been measured in terms of survival following a stress, but alternative methods are increasingly being used because of their relevance to the ecology of organisms and their utility in characterizing variation among species, populations and individuals. One such method capable of discriminating among Drosophila species and conspecific Drosophila populations from different environments is adult chill coma recovery time, the time taken for adults to become active again after being knocked down by a cold stress. Here we characterized the chill coma response of D.melanogaster in detail. Adults were exposed to a range of temperatures and stressful periods prior to measuring recovery. Recovery from chill coma in D.melanogaster was biphasic; as flies were stressed under cooler temperatures, recovery times leveled off and then decreased before sharply increasing again as mortality starts to occur. This biphasic response has previously been observed in D.subobscura where it has a somewhat different shape. A second mechanism therefore acts at relatively lower temperatures to ameliorate the effects of the cold stress. When D.melanogaster were reared at 19 and 25 °C for two generations, the shape of the curve relating temperature to recovery time was similar, but flies from the warmer temperature had longer recovery times and showed responses that leveled off and then decreased at relatively higher temperatures. As exposure time to cold stress was increased, recovery times also increased except at mild stress levels. Chill coma recovery in D.melanogaster is a complex trait and likely to reflect multiple underlying components.  相似文献

It is shown that a tetrapeptide fragment of defensin does not alter the phospholipid composition in the membranes of CHO-K1 cells but regulates the fatty acid composition of phosphatidylcholine, phosphatidylethanolamine (PEA), phosphatidylserine (PS), and phosphatidylinositol (PI). Incubation of the cells in the presence of this tetrapeptide resulted in modification of unsaturated fatty acid composition in the studied phospholipids. The content of monoenoic (mainly C18 : 1ω9) and/or dienoic (C18 : 2ω6) fatty acids increased, while the level of polyenoic fatty acids decreased. It was found that in the polyenoic fatty acid group of the PEA, PS and PI molecules, the ω3-/ω6-acid ratio decreased mainly due to the lower content of long-chain ω3-acids with 20 and/or 22 carbonic atoms. The possible role of this peptide in inhibition of the activity of Δ6- and Δ5-desaturases involved in the synthesis of long-chain polyenoic fatty acids, the quantitative alteration of which in phospholipids influences physicochemical parameters in cell membranes, is discussed.  相似文献

Microalgal fatty acid composition: implications for biodiesel quality   总被引:3,自引:0,他引:3  
The fuel properties of microalgal biodiesel are predicted using published microalgal fatty acid (FA) compositions and predictive fuel models. Biodiesels produced from the microalgae investigated are predicted to have extremely poor oxidative stabilities and the majority also have poor cold-flow properties. The cetane number in most cases is out of specification, but less so than the oxidative stability and cold flow. These findings support the idea that feedstocks rich in monounsaturated fatty acids (MUFAs) are desirable for biodiesel but the composition of the saturated fatty acids (SFAs) is also shown to be of great importance. There is an apparent relationship between algal class and the percentage of FAs represented by MUFA. This potentially allows for the identification of high-MUFA algal classes, or at least provides some basis for researchers to make initial selections of target classes for bioprospecting. Comparisons of FA groups between algal classes also show that the SFAs of Mediophyceae contain significantly higher proportions of C14:0, which is in contrast to the normally abundant C16:0 and the Mediophyceae therefore have better cold-flow characteristics than other classes with similar total SFA contents. Certain particularly promising cases for biodiesel production are presented as species level examples of feedstocks that are close to satisfying the biodiesel standards and to further illustrate the challenges that remain. Variation in FA composition as a response to changes in certain environmental variables forms another important facet to feedstock selection and is briefly considered, with suggestions for further research.  相似文献

The effect of ambient temperature on the composition of intracellular fatty acids and the release of free fatty acids (FFA) into a medium by cyanobacterium Spirulina platensis and eukaryotic microalgae, Chlorella vulgaris and Botryococcus braunii, was studied using their batch cultures. It was found that all the species studied, regardless of their taxonomic status, responded to the temperature regime by similar changes in their intracellular fatty acid composition: the relative content of more unsaturated fatty acids decreased with the elevation of temperature. At the same time, in the prokaryote, this temperature shift blocked, first of all, the elongation of 16:0 to 18:0 and then their further desaturation. In eukaryotes, the change in the desaturation of dienoic to trienoic fatty acids was the most pronounced process. The ratio of dienoic to trienoic fatty acids remained almost unchanged in S. platensis. The relative content of extracellular unsaturated FFA increased in the prokaryotic organism S. platensis at a higher temperature. But no significant changes in the composition of extracellular unsaturated FFA were detected in eukaryotic algae upon temperature elevation.  相似文献

NLRP3 inflammasome plays a key role in Western diet–induced systemic inflammation and was recently shown to mediate long-lasting trained immunity in myeloid cells. Saturated fatty acids (SFAs) are sterile triggers able to induce the assembly of the NLRP3 inflammasome in macrophages, leading to IL-1β secretion while unsaturated ones (UFAs) prevent SFAs-mediated NLRP3 activation. Unlike previous studies using LPS-primed bone marrow derived macrophages, we do not see any ROS or IRE-1α involvement in SFAs-mediated NLRP3 activation in human monocytes-derived macrophages. Rather we show that SFAs need to enter the cells and to be activated into acyl-CoA to lead to NLRP3 activation in human macrophages. However, their β-oxidation is dispensable. Instead, they are channeled towards phospholipids but redirected towards lipid droplets containing triacylglycerol in the presence of UFAs. Lipidomic analyses and Laurdan fluorescence experiments demonstrate that SFAs induce a dramatic saturation of phosphatidylcholine (PC) correlated with a loss of membrane fluidity, both events inhibited by UFAs. The silencing of CCTα, the key enzyme in PC synthesis, prevents SFA-mediated NLRP3 activation, demonstrating the essential role of the de novo PC synthesis. This SFA-induced membrane remodeling promotes a disruption of the plasma membrane Na, K-ATPase, instigating a K+ efflux essential and sufficient for NLRP3 activation. This work opens novel therapeutic avenues to interfere with Western diet-associated diseases such as those targeting the glycerolipid pathway.  相似文献

To assess the trade‐offs associated with cold and heat tolerance, selection experiments were conducted on the rate of recovery from chill‐ and heat‐coma using Drosophila melanogaster. Flies were treated with cold and heat to induce coma, and those that showed rapid or slow recovery from coma were selected. The lines selected for rapid (or slow) recovery from chill‐coma also showed rapid (slow) recovery from heat‐coma, although such a correlation was not observed in the lines selected for the rate of recovery from heat‐coma. On the other hand, survival after cold was enhanced in both lines selected for rapid and slow recovery from chill‐coma, and survival after heat was enhanced in both lines selected for rapid and slow recovery from heat‐coma. It was assumed that cold and heat treatments to induce coma caused some damages to flies and those that were tolerant to cold or heat were unintentionally selected in the present coma‐based selection. Only a weak trade‐off was observed between survival‐based cold and heat tolerance. On the other hand, developmental time was prolonged and desiccation resistance, walking speed, and longevity were reduced in the lines selected for rapid and slow recovery from chill‐ and/or heat‐coma, suggesting that these resistance and life‐history traits are under trade‐offs with cold and/or heat tolerance. © 2008 The Linnean Society of London, Biological Journal of the Linnean Society, 2008, 95 , 72–80.  相似文献

Unesterified fatty acids were measured in mouse erythrocytes infected either with chloroquine-susceptible (CS) or with chloroquine-resistant (CR) lines of Plasmodium berghei. This work was undertaken to identify candidates for the lipid involved in ferriprotoporphyrin IX (FP) polymerization. Linoleic, oleic, palmitic, and stearic acids were quantified by gas chromatography/mass spectrometry. In total, they increased 4-fold with CS infections and 6-fold with CR infections. Treating infected mice with chloroquine did not affect the amounts of unesterified fatty acids in erythrocytes. Of the four fatty acids, only linoleic acid increased disproportionately to the total. It increased 16-fold for the CS line and 35-fold for the CR line. The method could detect monoglycerides but they were below the limit of detection. It could not detect diglycerides, triglycerides or phospholipids. Triglycerides and phospholipids have been tested previously, however, and found to be ineffective at promoting FP polymerization. Therefore, other than linoleic acid, the lipids most likely to be involved in FP polymerization are diglycerides. We tested dilinoleolyglycerol in the present work and found it to be an effective promoter of FP polymerization. These results suggest that linoleic acid or a diglyceride containing it has the critical role of promoting FP polymerization in malaria parasites.  相似文献

An experiment has been conducted to study the effects of the inclusion of plant oil in rabbit diets. This study was aimed at evaluating the beneficial effects of the inclusion of maize oil (MO), rich in unsaturated fatty acids (UFAs), compared to palm oil (PO) containing saturated fatty acids (SFAs), on the meat fatty acid (FA) profile. As UFAs are susceptible to rancidity, Curcuma longa (CL), which is known for its antioxidant properties, was also added (3 g/kg) to the diet with two plant oils. CL contains curcuminoids, volatile oils, sugars, proteins, resins and polyunsaturated fatty acids (PUFAs). We also evaluated the influence of CL inclusion in the diet on the FA profile of the meat. Furthermore, the possibility of using these oil-enriched diets and the ability to assimilate CL in rabbits was evaluated by analysing the performance, carcass characteristics and meat quality. At the end of the experiment, there were no significant differences between the groups concerning the live weight, live weight gain, feed consumption, feed efficiency, carcass yield or the percentages of edible organs. The hind legs, forelegs, loins and abdominal wall, breast and ribs, skin and limbs and head were not affected by the oil type or by the inclusion of CL. The chemical composition, pH and oxidative status of the Longissimus dorsi muscle of the rabbits fed the experimental diets were not affected by the oil source or by the CL supplementation. Conversely, it has been shown that it is possible to modify the FA profile of rabbit meat and fat by dietary means. The SFA/PUFA ratio significantly decreased from -18% to -16% in the meat and from -25% to -23% in the perirenal fat of the rabbits fed diets containing MO without or with CL supplementation, respectively, compared to same tissues of the rabbits fed diets containing PO without or with CL supplementation, respectively. Similar trends were found for the atherogenic index, which decreased from -20% to -17% in the meat and from -26% to -23% in the perirenal fat, respectively, and the thrombogenic index, which decreased from -19% to -24% in the meat and from -24% to -23% in the perirenal fat, respectively. CL increased the α-Linolenic acid and PUFA n-3 contents and reduced the vaccenic acid content and the n-6/n-3 ratio in the meat of the rabbits fed the PO or MO diets.  相似文献

Fatty acid composition and structure in total lipids from the green above-ground parts of four alpine plants, Oxygraphis glacialis, Primula macrophylla, Rhodiola pamiroalaica, and Swertia marginata, were established by GC and GC-MS. A total of 55 fatty acids was detected, and 48 of them were identified. Ubiquitous palmitate, linoleate, and linolenate predominated in the lipids accounting for about 72-90% of the total fatty acids. At the same time, the latter contained numerous species, which were unusual for higher plants and included saturated odd-numbered n-acids (six C15-C25 species, 0.26-1.40%), saturated even-numbered very-long-chain n-acids (six C20-C30 species, 1.00-2.49%), iso-acids (nine C15-C26 species, 0.64-1.53%), anteiso-acids (four C15-C20 species, 0.08-1.57%), certain uncommon mono- and dienoic acids, as well as 16:3omega3, 18:3omega6, and 18:4omega3 acids that are absent from the most higher plants. Nine fatty acids were found here for the first time in higher plants and two may be new to science. The evidence on the unusual fatty acids is discussed with respect to their distribution in living organisms, pathways of biosynthesis, and chemotaxonomic role.  相似文献

Dietary polyunsaturated fatty acids (PUFA) play a key role in regulating delta-6 desaturase (D6D), the key enzyme for long-chain PUFA biosynthesis. Nevertheless, the extent of their effects on this enzyme remains controversial and difficult to assess. It has been generally admitted that C18 unsaturated fatty acids (UFAs) regulate negatively delta-6 desaturase (D6D). This inhibition has been evidenced in regard to a high glucose/fat free (HG/FF) diet used in reference. However, several nutritional investigations did not evidence any inhibition of desaturases when feeding fatty acids.Because the choice of the basal diet appeared to be of primary importance in such experiments, our goal was to reconsider the specific role of dietary UFAs on D6D regulation, depending on nutritional conditions. For that, sixteen adult Wistar rats were fed purified linoleic acid, α-linolenic acid or oleic acid, included in one of two diets at 4% by weight: an HG/FF or a high starch base (HS) where the pure UFAs replaced a mixed vegetable oil. Our results showed first that D6D specific activity was significantly greater when measured in presence of an HG/FF than with an HS/4% vegetable oil diet. Secondly, we found that linoleic and alpha-linolenic acids added to HG/FF reduced the specific activity of D6D. In contrast, when pure UFAs were added to an HS base, D6D specific activities remained unchanged or increased. Concordant results were obtained on D6D mRNA expression.Altogether, this study evidenced the importance of the nutritional status in D6D regulation by C18 UFAs: when used as control, HG/FF diet stimulates D6D compared with a standard control diet containing starch and 4% fats, leading to an overestimation of the D6D regulation by UFAs. Then, UFAs should be considered as repressors for unsaturated fatty acid biosynthesis only in very specific nutritional conditions.  相似文献
