Using the medaka, Oryzias latipes (orange-red variety), the mechanisms of action on leucophores of adenosine and adenine nucleotides, including cyclic AMP, were studied. All these substances were found to be very effective in dispersing leucosomes. Their pigment-dispersing action was antagonized by methylxanthines. These substances did not aggregate leucosomes. It was concluded that leucophores possess specific binding sites for adenosine, i.e. adenosine receptors, on the cell membrane, which mediate leucosome dispersion. Further, it was shown that even the action of an established intracellular second messenger, cyclic AMP, is primarily manifested through the receptors.  相似文献

The pattern of adrenergic innervation to scale chromatophores of the wild-type medaka, Oryzias latipes, was examined by autoradiography with 3H-norepinephrine and found for the first time to be changed reversibly during prolonged background adaptation. In scales of the medaka, which was adapted to a black background for 10-15 days, a great number of melanophores and dense networks of varicose fibers were observed: many fibers built up a radial plexus around each melanophore. However, the dense distribution of varicose fibers disappeared with a decrease in the number of melanophores during long-term adaptation to a white background. As to the changes in the innervation pattern to amelanotic melanophores of the medaka, orange-red variety, a similar result was obtained. Although the increase in the number of leucophores was observed in the medaka adapted to a white background, no exact plexuses of labeled fibers were confirmed around leucophores. From these results, it is concluded that the density of chromatic nerve fibers changes in parallel with the variation of the number of melanophores during prolonged background adaptation.  相似文献

In whitish parts of teleost skin, the coloration is attributed to a light scattering phenomenon within light-reflecting chromatophores, namely leucophores and iridophores, which contain high refractive index materials in their cytoplasmic organelles, leucosomes and light-reflecting platelets, respectively. Previous chemical examinations revealed that guanine is a major constituent of the materials in the platelets of the iridophores, while, in leucophores, the detailed chemical nature of the materials contained in the leucosomes has not been reported. Here, using liquid chromatography-tandem mass spectroscopy, we investigated the chemical features of materials eluted from scales, larvae, and single chromatophores of the medaka. Results of the liquid chromatography-tandem mass spectroscopy suggested that uric acid is a major constituent of the high refractive index materials in medaka leucophores and is a unique marker to investigate the presence of leucophores in the fish. The whitish appearance of the medaka leucophores may be attributed to the light-scattering phenomenon in leucosomes, which contain highly concentrated uric acid.  相似文献

Two cyclic AMP-independent protein kinases from rat liver nuclei were inhibited competitively by adenosine and a variety of its analogues: cardycepin, tubercidin, 6-mercaptopurine riboside, 6-methylaminopurine riboside, 6-dimethylaminopurine riboside, and 2'-deoxyadenosine. Neither enzyme was inhibited by 6-methoxypurine riboside. Kinase NI was more sensitive to cordycepin, tubercidin, 6-methylaminopurine riboside,, 2'-deoxyadenosine, and adenosine than was kinase NII. Although the effects of all analogues tested were reversed by increasing the concentration of ATP, kinases NII and NI exhibited marked differences in their affinities for adenosine. The results presented here suggest that 1) differences in the catalytic properties of nuclear protein kinases can be detected by inhibitor studies and 2) modifications in the purine ring and sugar moiety of an adenosine analogue can alter its ability to inhibit nuclear protein kinases.  相似文献

Delayed-type mutations induced by radiation have recently been demonstrated in various somatic-cell systems. Such mutations are thought to result from the transmission of genetic instability through many cell divisions subsequent to a single exposure to ionizing radiation. Here, we have examined whether 'transgenerational' delayed-type mutations can arise during embryonic development of the medaka fish as a result of exposing the sperm and spermatids of live fish to 137Cs gamma-radiation. To do this, we made use of a sensitive specific-locus test (SLT) for the medaka that we have recently developed. Because the medaka has a transparent egg membrane and embryo body, both visible mosaics and whole-body mutations can be detected during development at an early-expressed pigmentation locus. When wild-type +/+ males were gamma-irradiated and then mated with wl/wl females, the frequency of F1 embryos with both wild-type orange leucophores (wl/+) and mutant-type white leucophores (wl/wl*) (mosaic mutants) was about 5.7x10(-3)/Gy. The frequency of embryos with only white leucophores (whole-body mutants) was about 1.3x10(-3)/Gy. These results suggest that delayed mutations frequently arise in medaka fish embryos that have been fertilized with irradiated sperm. Some possible mechanisms involved in the generation of these delayed mutational events (including genomic instability in the early embryos) are discussed.  相似文献

1. The mechanism of the action of atropine, which is known to accelerate the dispersion response of fish melanophores, was examined by use of various receptor antagonists.2. The effects of atropine were found to be independent of adenosine receptors, beta-adrenoceptors and MSH receptors on the melanophore membrane.3. Analogs of atropine, such as scopolamine, also had a potent pigment-dispersing effect on melanophores, whereas the quaternary ammonium derivatives, which are positively charged molecules, had only a small effect.4. These results suggest that the possible site of atropine action is within the chromatophores themselves.5. In addition to the melanosome-dispersing effect, atropine caused a shift in the spectral peak of reflected light toward shorter wavelengths and the dispersion of leucosomes in the motile iridophores of the blue damselfish and in the leucophores of the medaka, respectively.  相似文献

Actions of the adrenergic beta-2 agonists, salbutamol and terbutaline, and the beta-1 antagonists, metoprolol and atenolol, were examined on denervated melanophores and leucophores of a teleost, Oryzias latipes. Beta-2 agonists depressed the pigment-aggregation response of melanophores to norepinephrine, while beta-1 antagonists inhibited the dispersion response of leucophores to isoproterenol but not the melanophore response. These findings suggest that adrenergic receptors mediating pigment dispersion in melanophores are beta-2 and those of leucophores are beta-1. The possible relations between receptor mechanisms and the responses of chromatophores are discussed.  相似文献

The adenine derivatives adenosine 5'-triphosphate (ATP), adenosine 5'-diphosphate (ADP), adenosine 5'-monophosphate (AMP) and adenosine (AD), in concentrations of 10(-3)M and 10(-4)M caused significant and dose-related modifications in the basal acid secretion from isolated whole rat stomach. The first three purine derivatives, ATP, ADP and AMP significantly increased the spontaneous acid secretion. On the other hand, AD caused a significant reduction in the basal acid secretion. When ATP, ADP, AMP and AD were assayed in the presence of the adrenergic and cholinergic blocking agents, ergotamine, 10(-6)M, propranolol, 5 X 10(-7)M and atropine, 10(-6)M, all these purine derivatives, including AD, caused a significant increase in the basal acid secretion.  相似文献

Human malaria infected erythrocytes show a dramatic increase in adenosine deaminase activity in vitro. Using recently developed culture techniques, adenosine deaminase-deficient human erythrocytes were infected in vitro with the major human pathogen Plasmodium falciparum. Adenosine deaminase activity was undetectable in the uninfected host red cells, but increased by 2-fold over normal levels in these cells with an 8% parasitemia. The enzyme in these cells appeared unique in that its activity was markedly elevated over that of other parasite purine enzymes, was not cross-reactive with antibody against human erythrocyte adenosine deaminase, and though inhibited competitively by deoxycoformycin was relatively insensitive to erythro-9-(2-hydroxy-3-nonyl) adenine. The use of adenosine deaminase-deficient erythrocytes for the in vitro cultivation of Plasmodium provides a unique system for the study of parasite enzyme and allows further insight into the purine metabolism of the intraerythrocytic malaria parasite.  相似文献

An adenosine (phosphate) deaminase from the squid liver had much lower activity for 5′-deoxyadenosine than that for adenosine, 2′-, or 3′-deoxyadenosine. 3′-IMP and inosine as well as purine riboside and adenine competitively inhibited the deamination of adenosine 3′ phenylphosphonate by the enzyme, but 5′-AMP and 5′-IMP did not. The enzyme deaminated the 5′-hydroxyl terminal adenosine residue in dinucleotides and trinucleotide, but not the 3′-hydroxyl terminal one in dinucleotides. The 5′-hydroxyl group of the ribose moiety was necessary for the substrate binding and catalytic activity of the squid enzyme. These results indicated that the recognition of ribose moiety in the substrate by the squid enzyme might be intermediate between those by adenosine deaminase and adenosine (phosphate) deaminase from microorganisms.  相似文献

The human erythrocyte membrane carriers for hexoses and nucleosides have several structural features in common. In order to assess functional similarities, the effects of adenosine derivatives on hexose transport and cytochalasin B binding sites were studied. Adenosine inhibited zero-trans uptake of 3-O-methylglucose half-maximally at 5 mM, while more hydrophobic adenosine deaminase-resistant derivatives were ten- to 20-fold more potent transport inhibitors. However, degradation of adenosine accounted for very little of this difference in potency. Hexose transport was rapidly inhibited by N6-(L-2-phenylisopropyl)adenosine at 5 degrees C in a dose-dependent fashion (EC50 = 240 microM), to lower the transport Vmax without affecting the Km. A direct interaction with the carrier protein was further indicated by the finding that N6-(L-2-phenylisopropyl)adenosine competitively inhibited [3H]cytochalasin B binding to erythrocytes (Ki = 143 microM) and decreased [3H]cytochalasin B photolabeling of hexose carriers in erythrocyte ghosts. The cross-reactivity of adenosine and several of its derivatives with the hexose carrier suggests further homologies between the carriers for hexoses and nucleosides, possibly related to their ability to transport hydrophilic molecules through the lipid core of the plasma membrane.  相似文献

The inhibition of 3H-noradrenaline (3H-NA) release by adenosine, and the possible involvement of purine receptors in the regulation of transmitter release in the portal vein were studied. The inhibitory effect of different concentrations of adenosine (10, 30, 100 and 300 microM) decreased with frequency of stimulation, but there was no marked concentration-dependence. Tetraethylammonium (TEA) enhanced the 3H-NA overflow induced by transmural stimulation. The adenosine-induced inhibition of 3H-NA overflow was antagonized by TEA. Transmural stimulation induced release of tritium from tissues prelabelled with either 3H-NA or 3H-adenine had a similar pattern of distribution. In contrast, when the rat portal vein was stimulated with (-) NA, the overflow of purine derivates was delayed and the maximum release was achieved 5 min later than the maximum induced by transmural stimulation. Phenoxybenzamine (PBA) increased 3H-NA overflow two-fold, but had no effect on the 3H-purine release induced by transmural stimulation. PBA reduced the 3H-purine release by exogenous (-) NA. These results indicate that in rat portal vein, the purine compounds have pre- and postjunctional origins and that the purine that modulates adrenergic neurotransmission might be of neuronal origin, possibly independent of adrenergic innervation.  相似文献

A new method to introduce a benzyl group onto the 2'-OH of purine ribonucleoside is described. Thus, 6-chloropurine 3'-O-benzoylriboside and its 5'-O-trityl congener were condensed with benzyl alcohol using the Mitsunobu reaction to give the 2'-O-benzyl derivative. The yields were varied from 4.6 to 62.9% depending on the solvent used. The product was converted to adenosine, indicating that the stereochemistry at C-2' is retained.  相似文献

The adenosine analogs [5'-N-ethylcarboxamideadenosine (NECA), 2-Chloro-adenosine (2-ClA), R-phenylisopropyladenosine (R-PIA), N6-cyclohexyl adenosine (CHA), and N6-cyclopentyladenosine (CPA)] produced both relaxation and contraction responses in isolated guinea-pig trachea. A concentration-related relaxation response was observed in trachea which were precontracted with either histamine or KC1. This response followed an order of analog potency that was indicative of the A2 receptor subtype (NECA greater than 2-ClA greater than R-PIA greater than CPA greater than CHA). Theophylline, an adenosine-receptor antagonist, blocked this relaxation response. In addition, a concentration-related contractile response was produced with adenosine analogs in those trachea that were not previously contracted. In contrast, the contractile response followed an analog potency indicative of the A1 receptor subtype (R-PIA greater than 2-ClA = CPA = CHA). This contractile response was not mediated by cholinergic, adrenergic or histaminergic receptors. 2-ClA induced a biphasic response, while NECA only relaxed these tissue under basal tone. Unlike the relaxation response, these contractile responses were not attenuated by theophylline, but were blocked by 1,3 dipropyl-8-(2 amino-4-chlorophenyl)xanthine (PACPX). These findings confirm the existence of two subpopulations of adenosine receptors in guinea pig trachealis muscle.  相似文献

1. The effects of purines on denervated melanophores of the medaka were studied under experimental conditions in which melanosomes were aggregated by norepinephrine or lithium ion beforehand.2. Adenosine and its derivatives caused melanosome dispersion; the order of potency for the series was; NECA > adenosine > ATP > 2-chloroadenosine > PIA > CHA > cyclic AMP.3. 8-Phenyltheophylline, a potent purinoceptor antagonist, blocked the effect of purines and caused a rightward shift of the adenosine and analog concentration-response curves.4. 8-Br cyclic AMP also caused melanosome dispersion but its action was not blocked by 8-phenyladenosine. Dibutyryl cyclic AMP, cyclic GMP, dibutyryl cyclic GMP, and 8-br cyclic GMP were all ineffective.5. The effect of adenosine was immediately eliminated by adenosine deaminase but, actions of NECA, AMP, ADP, ATP, and cyclic AMP were not.6. Forskolin, a potent activator of adenylate cyclase, mimicked the action of adenosine.7. It is concluded that adenosine and its derivatives mediate their melanosome-dispersing effect via a P1-purinoceptor that displays characteristics of the A2-subtype and that adenine nucleotides directly activate the A2-receptor without conversion to adenosine.  相似文献

In this paper we investigated the influence on affinity, selectivity and intrinsic activity upon modification of the adenosine agonist scaffold at the 3'- and 5'-positions of the ribofuranosyl moiety and the 2- and N6-positions of the purine base. This resulted in the synthesis of various analogues, that is, 3-12 and 24-33, with good hA3AR selectivity and moderate-to-high affinities (as in 32, Ki=27 nM). Interesting was the ability to tune the intrinsic activity depending on the substituent introduced at the 3'-position.  相似文献

We have demonstrated that in Chinese hamster ovary (CHO) cells, N6,O2'-dibutyryl adenosine cyclic 3':5'-monophosphate (dibutyryl cyclic AMP) has a remarkable morphogenetic effect in converting cells of a compact, epithelial-like morphology into a spindle-shaped, fibroblast-like form. Homogenates of CHO cells were found to contain two adenosine cyclic 3':5'-monophosphate (cyclic AMP) phosphodiesterase (EC 3.1.4.c) activities, which differ in apparent Km with respect to their substrate, cyclic AMP. These were designated cyclic AMP phosphodiesterase I, with a low Km of 2 to 5 muM and cyclic AMP phosphodiesterase II, with a high Km of 1 to 3 mM. Cyclic AMP phosphodiesterase I was competitively inhibited by N6-monobutyryl and dibutyryl cyclic AMP, with apparent Ki values of 40 to 60 muM and 0.25 to 0.35 mM, respectively. Experimental evidence demonstrates that the effect of exogenous dibutyryl cyclic AMP on cell morphology is a result of an increase in the endogenous level of cyclic AMP. This increase appears to be due largely to the inhibitory action of intracellular N6-monobutyryl cyclic AMP on cyclic AMP phosphodiesterase I, which results in a decreased rate of degradation of intracellular cyclic AMP.  相似文献

Formation and rephosphorylation of adenosine (adenosine cycling) was studied in isolated rat hearts during normoxia and under conditions of stimulated purine formation. Hearts were infused with an inhibitor of adenosine kinase (5-iodotubercidin, 2 microM). In addition, perfusions were carried out with or without acetate, which is converted into acetyl-CoA, with simultaneous breakdown of ATP to AMP and purines. We found a linear, concentration-dependent, increase in normoxic purine release by acetate (5-20 mM). Differences in total purine release with or without iodotubercidin were taken as a measure of adenosine cycling. In normoxic hearts, iodotubercidin caused a minor increase in purine release (2.7 nmol/min per g wet wt.). Acetate (12.5 mM) increased purine release by 4.9 nmol/min per g, and its combination with inhibitor gave a large increase, by 14.2 nmol/min per g. This indicates a strongly increased adenosine cycling rate during acetate infusion. However, no significant differences in purine release were observed when iodotubercidin was infused during hypoxia, anoxia or ischaemia. The hypothesis that adenosine cycling is near-maximal during normoxia was not confirmed. Increased myocardial adenosine formation appears to be regulated by the availability of AMP and not by inhibition of adenosine kinase. This enzyme mainly functions to salvage adenosine in order to prevent excessive loss of adenine nucleotides.  相似文献
