The influence of hypoxia on the morphological characteristics of circulating erythrocytes of the scorpion fish Scorpaena porcus (Linnaeus, 1758) has been investigated in an in vivo experiment. Under a 4-h adaptation of the fish to the conditions of ranked hypoxia their erythrocytes demonstrated a number of consecutive reactions. The volume and the surface area of the red blood cells was reduced by 4–5% (p < 0.001) at an oxygen concentration of 2.6 mg/L (30% saturation of water with oxygen) and increased by 4% (p < 0.001) at a concentration of 1.3 mg/L (15% saturation), relative to the control values (normoxia: 7–8 mg/L). The observed reaction of erythrocytes coincided quantitatively and qualitatively (the order of events) with the results of the experiments we performed previously in vitro. Our study has shown that the physiology of the black scorpion fish is tolerant to hypoxia and allows autonomous functioning of red blood cells under conditions of oxygen deficit.  相似文献

Temperature measurements in a plastic tube isolated from external influences containing an erythrocyte suspension of the scorpion fish (Scorpaena porcus Linnaeus, 1758) showed that these red blood cells are able to generate heat. Heat release in the cell suspension was expressed by a linear temperature increase in the tube during the entire experiment. Addition of extracellular ATP (1 mg mL–1) caused the effect of a thermal shift: a sharp temperature rise in the cell suspension for 30–60 s. We believe that the heat release was caused by hydrolysis of extracellular ATP by membrane ecto-ATPase. Inhibition of ecto-ATPase activity through the addition of EDTA (1 mM) to the erythrocyte suspension led to complete blockage of heat release; the effect of the thermal shift ceased. We assume that thermal properties of red blood cells play an important role in blood hemodynamics, especially in providing the “non-Newtonian” properties of blood. The thermal phenomena observed in suspensions of fish erythrocytes open new scientific directions in exploring the capabilities of multifunctional extracellular ATP.  相似文献

Low-molecular-weight compounds with anticoagulant activity were isolated from the scorpion Heterometrus laoticus venom. The determination of the structure of the isolated compounds by nuclear magnetic resonance and mass spectrometry showed that one of the isolated compounds is adenosine, and the other two are dipeptides leucyl-tryptophan and isoleucyl-tryptophan. The anticoagulant properties of adenosine, which is an inhibitor of platelet aggregation, is well known, but its presence in scorpion venom is shown for the first time. The ability of leucyl-tryptophan and isoleucyl-tryptophan to slow down blood clotting and their presence in scorpion venom are also established for the first time.  相似文献

The phospholipid and the fatty acid composition of the main phospholipid families of erythrocyte plasma membranes was studied in two species of cartilaginous fish: the common thrasher (Raja clavata L.) and the common stingray (Dasyatis pastinaca) and in three bony fish species: the scorpion fish (Scorpaena porcus L.), the smarida (Spicara flexuosa Raf.), and the horse mackerel (Trachurus mediterraneus ponticus Aleev). It was shown that in the studied fish, 70.0–80.0% of all membrane phospholipids were composed of phosphatidylcholine and phosphatidylethanolamine. Phosphatidylserine, monophosphoinositide, and sphingomyelin were minor components whose content in the erythrocyte membrane fluctuated from 3.0% to 13.0%. The fatty acid phospholipids composition was represented by a large specter of acids. From saturated acids, basic for plasma membranes are palmitic (C16: 0) and stearic (C18: 0) acids. From unsaturated acids, the larger part belongs to mono-, tetra-, penta-, and hexaenoic acids in fish phospholipids. The calculation of the double bond index and of the unsaturation coefficient showed difference in the deformation ability of erythrocyte membranes of the studied fish.  相似文献

Abstract: Palaeoscorpius devonicus Lehmann, 1944 is known from only a single specimen, found in the Eschenbach Pit near Bundenbach in the Lower Devonian Hunsrück Slate of Germany. It is a key fossil, having been interpreted both as the most basal member of the Scorpiones and as one of the order’s most likely candidates for an aquatic mode of life. Prepared both ventrally and dorsally, some aspects of its morphology remain problematic. Here, with the aid of new techniques, including computed tomography, we present a re‐investigation of this scorpion’s anatomy and a new reconstruction, with a particular focus on the species’ original habitat. On the basis of the environmental interpretation of the Hunsrück Slate and the completeness of the specimen, previous authors concluded that P. devonicus was marine, but none offered convincing morphological evidence. Recent studies of the deposit’s environment suggest that the Hunsrück Sea was part of an intrashelf basin, relatively close to the coastline, and fossils of land plants show that terrestrial wash‐in occasionally occurred. Our revised interpretation of the fossil’s morphology demonstrates that the scorpion was most probably terrestrial. Internal mesosomal organs are interpreted as book lungs, but other terrestrial adaptations are lacking. The absence of both coxapophyses and gnathobases makes determining the scorpion’s feeding mechanism difficult. Interpreting the scorpion’s character states within a phylogenetic framework, especially the possible presence of book lungs, implies either that the plesiomorphic position of P. devonicus is no longer supported or that the development of book lungs had already taken place early in the scorpion lineage.  相似文献

A new anti-insect neurotoxin, AaH IT4, has been isolated from the venom of the North African scorpion Androctonus australis Hector. This polypeptide has a toxic effect on insects and mammals and is capable of competing with anti-insect scorpion toxins for binding to the sodium channel of insects; it also modulates the binding of alpha-type and beta-type anti-mammal scorpion toxins to the mammal sodium channel. This is the first report of a scorpion toxin able to exhibit these three kinds of activity. The molecule is composed of 65 amino acid residues and lacks methionine and, more unexpectedly, proline, which until now has been considered to play a role in the folded structure of all scorpion neurotoxins. The primary structure showed a poor homology with the sequences of other scorpion toxins; however, it had features in common with beta-type toxins. In fact, radioimmunoassays using antibodies directed to scorpion toxins representative of the main structural groups showed that there is a recognition of AaH IT4 via anti-beta-type toxin antibodies only. A circular dichroism study revealed a low content of regular secondary structures, particularly in beta-sheet structures, when compared to other scorpion toxins. This protein might be the first member of a new class of toxins to have ancestral structural features and a wide toxic range.  相似文献

Current predictions about the responses of species to climate change strongly rely on projecting altered environmental conditions on their distributions. In this study, we investigated the effects of future climate change scenarios on the potential distribution of 10 species of scorpions in north‐eastern Brazil in the context of their degree of specialisation to closed (Atlantic and Amazon Forests) and open (Caatinga and Cerrado) habitats. Scorpion species were classified as habitat specialists or generalists according to the IndVal index, and present and future species distribution models were prepared using minimum volume ellipsoids. According to IndVal, four species were classified as closed‐forest specialists (Ananteris mauryi, Tityus brazilae, Tityus pusillus and Tityus neglectus), four as open‐forest specialists (Jaguajir agamemnon, Jaguajir rochae, Physoctonus debilis and Bothriurus rochai), and two as generalists (Tityus stigmurus and Bothriurus asper). All species presented a drastic reduction in potential distribution, ranging from 44% to 72%, when compared with their current distribution. In addition, we found a reduction in scorpion species richness under future climate change scenarios. This finding has implications for scorpion conservation. Further, the results show that climate change may impact the composition of scorpion assemblages in north‐eastern Brazil, revealing important implications for human–scorpion interactions.  相似文献

Animals balance the risk of predation against other vital needs by adjusting their spatial behavior to match spatiotemporal variation in predation risk. To map this ‘landscape of fear’, prey use evolutionary rules of thumbs that are associated with the activity and hunting efficiency of predators. In addition, prey acquire perceptual information about the presence, identity and state of potential predators and use these cues to focus their acute anti‐predatory responses. Our goal was to explore if and how prey also use such perceptual information that decays with time to update their spatiotemporal risk assessment. We placed scorpions in freshly dug burrows and recorded the spatial activity and defense behavior of their isopod prey upon encountering the burrows straight after settling the scorpions and seven days later. To corroborate our understanding, we also examined the isopods’ detailed reactions towards deserted scorpion burrows. The isopods reacted defensively to scorpion burrows during their first encounter. After seven days, proportionally more isopods approached the scorpion burrows on their way out for foraging and fewer isopods encountered it on their way back. No changes in the spatial activity were observed towards deserted burrows. In addition, on the eighth day, more isopods entered the risky area near the scorpion burrows when leaving their own burrow than on the first encounter. The results suggest that isopods used predator cues to readjust the ‘landscape of fear’. Yet, rather than avoiding the dangerous areas altogether, the isopods implemented risky inspection behavior, validating whether the danger is actual. Our findings imply that inspection behavior toward predators can be used for future planning of prey spatial activity, offsetting possible ‘information decay costs’.  相似文献

All scorpion toxins from different 30 species are simply reviewed. A new classification system of scorpion toxins is first proposed: scorpion toxins are classified into three families (long-chain scorpion toxins with 4 disulfide bridges, short-chain scorpion toxins with 3 disulfide bridges, and intermediate-type scorpion toxins with 3 or 4 disulfide bridges). Intermediate-type scorpion toxins provide a strong proof for the conclusion that channel toxins from scorpion venoms evolve from a common ancestor. Common organization of precursor nucleotides and genomic sequence, similar 3-dimensional structure, and the existence of intermediate type scorpion toxins and functionally intercrossing scorpion toxins show that all scorpion toxins affecting ion channels evolve from the common ancestor, which produce millions of scorpion toxins with function-diversity.  相似文献

We manipulated the diversity of top predators in a three trophic level marine food web. The food web included four top benthic marine fish predators (black goby, rock goby, sea scorpion and shore rockling), an intermediate trophic level of small fish, and a lower trophic level of benthic invertebrates. We kept predator density constant and monitored the response of the lower trophic levels. As top predator diversity increased, secondary production increased. We also observed that in the presence of the manipulated fish predators, the density of small gobiid fish (intermediate consumers) was suppressed, releasing certain groups of benthic invertebrates (caprellid amphipods, copepods, nematodes and spirorbid worms) from heavy intermediate predation pressure. We attribute the mechanism responsible for this trophic cascade to a trait-mediated indirect interaction, with the small gobiid fish changing their use of space in response to altered predator diversity. In the absence of top fish predators, a full-blown trophic cascade occurs. Therefore the diversity of predators reduces the likelihood of trophic cascades occurring and hence provides insurance against the loss of an important ecosystem function (i.e. secondary production).  相似文献

Scorpion toxins have been the subject of many studies exploring their pharmacological potential. The high affinity and the overall selectivity to various types of ionic channels endowed scorpion toxins with a potential therapeutic effect against many channelopathies. These are diseases in which ionic channels play an important role in their development. Cancer is considered as a channelopathy since overexpression of some ionic channels was highlighted in many tumor cells and was linked to the pathology progression.Interestingly, an increasing number of studies have shown that scorpion venoms and toxins can decrease cancer growth in vitro and in vivo. Furthermore through their ability to penetrate the cell plasma membrane, certain scorpion toxins are able to enhance the efficiency of some clinical chemotherapies. These observations back-up the applicability of scorpion toxins as potential cancer therapeutics.In this review, we focused on the anti-cancer activity of scorpion toxins and their effect on the multiple hallmarks of cancer. We also shed light on effectors and receptors involved in signaling pathways in response to scorpion toxins effect. Until now, the anticancer mechanisms described for scorpion peptides consist on targeting ion channels to (i) inhibit cell proliferation and metastasis; and (ii) induce cell cycle arrest and/or apoptosis through membrane depolarization leading to hemostasis deregulation and caspase activation. Putative targets such as metalloproteinases, integrins and/or growth factor receptors, beside ion channels, have been unveiled to be affected by scorpion peptides.  相似文献

BmK AS is a β long-chain scorpion peptide from the venom of Buthus martensii Karsch (BmK). It was efficiently expressed as a soluble and functional peptide in Escherichia coli, and purified by metal chelating chromatography. About 4.2 mg/l purified recombinant BmK AS could be obtained. The recombinant BmK AS maintained a similar analgesic activity to the natural one in both the mouse-twisting test and hot-plate procedure. It also exhibited antimicrobial activity against both Gram-positive and Gram-negative bacteria. BmK AS is the first long-chain scorpion peptide reported to have antimicrobial activity, and is a valuable molecular scaffold for pharmacological research.  相似文献

Two neurotoxins, minax toxins 1 and 2, were isolated from venom of the scorpion Buthus minax from the Sudan. Molecular weights of 7000 and 6800 and 66 and 62 amino acids were found for minax toxins 1 and 2, respectively. Both toxins contain four disulfide bonds, 1 mol each of phenylalanine, histidine, and tryptophan, no free sulfhydryl groups, and no methionine. Both minax toxins 1 and 2 are basic polypeptides with isoelectric points of 8.2 and 9.0, respectively. There is a significant increase in the calcium content of rat hearts envenomated with minax toxins 1 and 2 or crude venom. This confirms earlier electron microscopic findings of calcium deposits in the heart following scorpion envenomation. There is a concomitant decrease in the calcium and phosphorus content of rat serum following envenomation. It seems that neither scorpion toxins nor scorpion venoms affect the mineral metabolism of the bone. The present investigation indicates that scorpion toxins have not only a neurotoxic action but also broader biological effects such as mineral metabolism.  相似文献

Cytogenetic and molecular analyses enabled identification of two cytotypes among individuals of the spotted scorpion fish Scorpaena plumieri from Margarita Island, Venezuela. Cytotype 1 was characterized by 48 subtelo‐acrocentric chromosomes and fundamental number (number of chromosome arms; FN) equalled 48, while cytotype 2 was characterized by two metacentric and 46 subtelo‐acrocentric chromosomes and FN was 50. These cytotypes also differed in the location of the ribosomal gene clusters and in the distribution of the constitutive heterochromatin. Moreover, fish from the cytotypes 1 and 2 were found to belong to distinct mitochondrial lineages. The presence of two S. plumieri cytotypes from two lineages separated by high genetic distance suggests that they correspond to sympatric cryptic species.  相似文献
