运用NCBI中的数据库消减杂交(Digital Differential Display,DDD)分析方法,从小鼠睾丸组织中分离了一个含有C2HC/C3H结构的新型锌指蛋白基因——ZIM74(GenBank登录号:AY350709)。通过推导和进一步的RT-PCR实验证实:该基因含4个外显子,gDNA在染色体上跨度29869bp,定位于小鼠染色体18D1。cDNA编码一个含347个氨基酸的新蛋白,带有C2HC/C3H结构域。Northern杂交结果显示:该基因含有2.37kb大小的唯一转录本,主要在睾丸中强表达,卵巢中有表达,而在其他组织中该基因无表达。结果提示:Zfp474基因对精于发生和卵母细胞的发育可能起重要作用。  相似文献

锌指蛋白是一类重要的转录因子, 广泛参与植物的生长发育和胁迫应答过程。文章使用生物信息学方法从水稻中预测并克隆了一个TFⅢA型锌指蛋白基因ZFP207(GenBank 登陆号: AK063147.1), 该基因包含一个567 bp的开放阅读框, 编码一条188个氨基酸组成的多肽,其编码产物的预测分子量为20.72 kDa, 等电点为9.67。锌指蛋白ZFP207含有一个典型的TFⅢA型锌指结构, 并且在C端含有一个可能具有转录抑制功能的EAR-motif, 但不具有任何已知的核定位信号。系统发生分析结果表明, ZFP207与植物中已经发现的单锌指蛋白亲缘关系较近。通过RT-PCR方法分析了ZFP207基因在成株期水稻不同组织中的表达, 发现ZFP207在茎和叶中表达量较高, 而在穗和根中表达量较低。在酵母中的转录激活实验结果显示, ZFP207在酵母中不具有转录激活功能。  相似文献

A novel C2H2-type zinc finger protein gene, ZFP15, was cloned from rice by RT-PCR approach. The ZFP15 gene encodes a protein of 144 amino acid residues with a predicted molecular mass of 15 kDa. The ZFP15 protein comprises two C2H2-type zinc finger domains, a putative nuclear localization signal (NLS) at its N-terminus but the DLN-box identified in all reported plant C2H2-type zinc finger proteins was not found. A homology search revealed that ZFP15 gene was localized within a cluster of C2H2-type zinc finger genes in BAC clone OJ1754_E06 mapped on chromosome 3. All three members in the cluster encoded proteins showed high identities in amino acids and might contribute to a co-regulation. The RT-PCR assay revealed that ZFP15 mRNA was not regulated by cold, salt, drought and ABA stresses, though CRT/DRE and ABRE elements were found in the promoter region of ZFP15 gene. The expression profiling also showed that ZFP15 mRNA was expressed with a lower level in leaves and roots, but not detected in stems. Besides, ZFP15 was shown to accumulate much more in flowering spike than in immature spike. Thus, ZFP15, as the first characterized C2H2-type zinc finger protein in rice, might play a regulatory role on rice spike development.  相似文献

A cDNA for the gene ZFP182, encoding a C2H2-type zinc finger protein, was cloned from rice by RT-PCR. ZFP182 codes an 18.2 kDa protein with two C2H2-type zinc finger motifs, one nuclear localization signal and one Leu-rich domain. The DLN-box/EAR-motif, which exists in most of plant C2H2-type zinc finger proteins, does not exist in ZFP182. The expression analysis showed that ZFP182 gene was constitutively expressed in leaves, culms, roots and spikes at the adult rice plants, and markedly induced in the seedlings by cold (4 degrees C), 150 mM NaCl and 0.1 mM ABA treatments. The approximate 1.4 kb promoter region of ZFP182 gene was fused into GUS reporter gene and transformed into tobacco. The histochemical analysis revealed that GUS expression could not be detected in transformed tobacco seedlings under normal conditions, but strongly observed in tobacco leaf discs and the vascular tissue of roots treated with NaCl or KCl. Expression of ZFP182 in transgenic tobacco and overexpression in rice increased plant tolerance to salt stress. These results demonstrated that ZFP182 might be involved in plant responses to salt stress.  相似文献

Eukaryotic LIM domain proteins contain zinc finger forming motifs rich in cysteine and histidine that enable them to interact with other proteins. A cDNA clone isolated from an adult schistosome cDNA library revealed a sequence that coded for a novel class of proteins bearing 6 LIM domains and an N-terminal PET domain, SmLIMPETin. Phylogeny reconstruction of SmLIMPETin and comparison of its sequence to invertebrate homologues and to the vertebrate four-and-a-half LIM domains protein family (FHLs), uncovered a novel LIM domain protein family, the invertebrate LIM and PET domain protein family (LIMPETin). Northern blots, RT-PCR and Western blot showed that SmLIMPETin gene was less expressed in sexually mature adult females compared to sexually immature adult females and sexually mature and immature adult males, and not expressed in schistosomula.  相似文献

In order to identify genes regulating meiosis, a mouse spermatocyte cDNA library was screened for sequences encoding proteins with C2H2-type zinc finger motifs which are typically expressed by the Drosophila Krüppel gene. Three new cDNAs were isolated, and they were designated CTfin33, CTfin51, and CTfin92. Among them, CTfin51 was selected for further study. The deduced amino acid sequence revealed seven zinc finger motifs in its C-terminal region. Northern blot and in situ hybridization showed CTfin51 mRNA expression in spermatocytes after the pachytene stage and in early stage round spermatids of prepuberal and adult males. Immunocytochemical staining with an antiserum against beta-gal-CTfin51 fusion protein was localized within nuclei of spermatocytes and spermatids. Oocyte nuclei after the pachytene stage also were immunoreactive for CTfin51 protein. Immunoblots revealed a band at M(r) 75,000 in protein extracts from the testis and the ovary. These results suggest that the CTfin51 gene encodes a DNA-binding regulatory protein functionally associated with meiosis in both male and female gametogenesis.  相似文献

We have isolated the DNMT3L gene that is related to the cytosine-5-methyltransferase 3 (DNMT3) family. The gene is located on chromosome 21q22.3 between the AIRE and the KIAA0653 genes and spans approximately 16 kb of genomic sequence. The encoded protein of 387 amino acids has a cysteine-rich region containing a novel-type zinc finger domain that is conserved in DNMT3A and DNMT3B but also in ATRX, a member of the SNF2 protein family. The novel domain, called an ADD (ATRX, DNMT3, DNMT3L)-type zinc finger, contains two subparts: a C2C2 and an imperfect PHD zinc finger. Expression of the DNMT3L mRNA was not detectable by Northern blotting; however, RT-PCR amplification revealed that it is expressed at low levels in several tissues including testis, ovary, and thymus.  相似文献

数据库消减杂交就是利用NCBI中的Unigene数据库中大量的数据资源,收集各种细胞或组织的基因表达谱进行两两比较或多重比较,获得两组文库间有统计学意义的差异表达基因。运用NCBI中的数据库消减杂交分析方法,从人睾丸组织中分离了一个含有C2H2结构的新型锌指蛋白基因-ZNF474(GenBank登录号:AY461732).通过推导和进一步的RT—PCR实验证实:该基因含2个外显子,gDNA在染色体上跨度30065bp,定位于人染色体5q23.1-q23。2.cDNA编码一个含364个氨基酸的新蛋白,分子质量是40.3kDa,等电点为9.59。Northem杂交结果显示:该基因含有2.37kb大小的唯一转录本,主要在睾丸中很强表达,卵巢中有弱表达,而其他组织中该基因无表达.结果提示:ZNF474基因对精子发生和卵母细胞的发育可能起重要作用、  相似文献
