The moisture content/probit viability relationship for storedseeds of Zizania palustris L. and Spartina anglica C. E. Hubbardwas linear and independent of the rate of embryo drying. Theseresults provide firm evidence of recalcitrant storage physiologyin these taxa. Preliminary tests strongly suggest that freshseeds of Porteresia coarctata (Roxb.) Tateoka are also intolerantof desiccation In Z. palustris apparent differences in desiccation tolerancebetween individuals can be partly explained by wide variationin individual embryo moisture contents during desiccation. Long-termstorage experiments in solutions of polyethylene glycol 6000(PEG) suggest that the actual variation in desiccation toleranceis confined to a narrow range of embryo water potentials inthe range –2 to –3 MPa. Seeds (caryopses) of North American wild rice (Zizania palustrisvar. interior), a temperate aquatic grass, have been thoughtto require storage at low temperatures and high moisture contentsto preserve viability. The seeds are also deeply dormant atmaturity and require up to 6 months of stratification to breakdormancy. We report here that wild rice seeds can retain viabilityat moisture contents 30% (f. wt. basis) for up to 6 monthsat temperatures as high as 30 °C, and for at least 1 yearat temperatures below 20 °C. Dormancy is not broken at temperaturesabove 10 °C, but subsequent stratification requirementsare unaffected by prior warm storage. Cold storage is thereforenot required to maintain viability of wild rice seeds, but isnecessary to break dormancy. Hydrated wild rice seeds can befrozen to –10 °C without damage, but dormancy is notlost at subfreezing temperatures. These results provide newoptions for long-term storage of wild rice seeds. Seed growth characteristics of Aesculus hippocastanum were examinedin detail during development from about 70 to 140 d after anthesis(DAA), mainly in 1988 and 1989. Mean fresh and dry weights increasedfor both the axis and the whole seed up to the time of peakseed fall at 135 DAA with no cessation before fruit abscission.Water per seed increased up to 100 DAA, after which no furtherincrease occurred; moisture content declined for the embryonicaxis and whole seed respectively from above 75 and 65% at 95DAA to 65 and 50% at 130 DAA. At fruit shedding in 1990 waterpotential values of -1·2, -2·6 and -1·1MPa were observed for the testa, cotyledon and axis tissuesrespectively; relevant sorption isotherms are presented. Decreases in seed moisture content during development were accompaniedby increases in desiccation tolerance and in germinability,both reaching their maximum at the time of peak seed fall. Atmaturity, only about 10% viability was retained on drying seedto 20% moisture content; it is confirmed that the seeds are'recalcitrant'. The exact relationship between moisture contentand germination during development was dependent on the deptof dormancy, as judged by the period of chilling required; eachduration of chilling at 2°C within the range 3-12 weeksyields a curve of sigmoid shape. No germination occurred at26°C without chilling, but nearly full germination can beobserved for samples collected at 6 weeks before maximum seedfall with 12 weeks chilling. Embryos excised from fresh seed had a moisture content of about37% (fr. wt.), 7% above that of the whole seed: mainly as aresult of the dryness of the seed coat. When cultured for 14d at 26°C embryo growth ranged from unorganized to fullyorganized. Embryos which had been directly surface-sterilizedexhibited lower fully organized growth than aseptically excisedembryos. During desiccation of the whole seed at 15°C and 15% r.h.moisture loss from the embryo was slow until the embryo reacheda mean critical moisture content for fully organized growthin vitro of about 30%. After this point the rate of moistureloss increased, quickly diminishing the moisture content differentialbetween the embryo and the whole seed. No such shoulder in theembryo desiccation curve was observed when using a low viability(7%) seed batch. When dried below 30% moisture content, suchaseptically excised embryos increasingly exhibited unorganizedgrowth in vitro, with only root meristem survival (46%) at 13%moisture content. In contrast, a much higher level of root meristem survival (84%of control) was observed when aseptically excised embryos wererapidly desiccated to 13% moisture content, using a flow ofsterile air. However, the benefits of rapid drying were lostif the embryos were directly surface-sterilized prior to desiccation.Furthermore, 80% of aseptically excised embryos which survivedrapid desiccation to 20% moisture content exhibited root meristemsurvival following subsequent cryopreservation. Seeds of Araucaria hunsteinii K. Schum. dried quicker at 29°Cthan at 19°C and quicker with the seed-coat removed thanwhen intact; seeds enclosed in polyethylene bags increased inmoisture content. At 15°C, seeds in a flow of air driedquicker than seeds in a box with silica gel, which in turn driedquicker than seeds in a box with no desiccant. No loss of germinationability occurred on drying fresh seed from 53 to about 32 percent moisture content (fresh weight basis); during further desiccationthe percentage germination was related to percentage moisturecontent in the form of a sigmoid curve, culminating in a completefailure to germinate at approximately 14 per cent moisture content.A consistent relationship was observed for all treatments andthe mean critical moisture content for seed death (failure togerminate) was near 23 per cent. Excised embryos grew on 1 percent agar but died if previously subjected to 14 h of desiccationat 15°C. In contrast, no relationship was found between germination andmoisture content of A. cunninghamii D. Don on desiccation from21 to 7 per cent moisture content. Possible causes for the observeddifference in response to desiccation are discussed and methodsfor seed storage are considered. To help in understanding the mechanisms of dormancy in wheat(Triticum aestivum L.), brief drying of intact ears has beenexamined as a technique for rapid imposition of dormancy. Beforenatural grain desiccation at 40 d post anthesis (dpa), wholeplants were moved for 15 to 20 d to ‘wet’, humidconditions (90–100% RH) or to ‘dry’ conditions(35–40% RH). The duration of subsequent dormancy was atleast doubled by the ‘dry’ treatment, however, onlyif the grain was allowed to dry before 50 dpa. In the ‘wet’ears there was a low level (up to 22%) in-ear germination by55 dpa and the remaining, non-sprouted grain no longer becamedormant on drying. Brief (4 d) drying episodes could also preventsprouting and induce some dormancy, but only over a 4 to 10d window of sensitivity before 50 dpa. Grain drying when properlytimed not only arrests development but, perhaps through damagingeffects of drying, causes dormancy. Biochemical properties, i.e. endogenous abscisic acid, proline,sugars, respiration, adenosine phosphates and adenylate energycharge, and growth and moisture content were measured duringthe development of seeds of Machilus thunbergii. As dry matteraccumulated in the embryo during development, moisture content,ABA, proline, respiration and sugars all declined. At maturity,the dry mass of the seeds failed to attain a plateau beforethe period of natural seed shedding; the axis and cotyledonsreached moisture contents of 58 and 45%, respectively. Dryingof immature seeds at 73% relative humidity and 25 °C for30 d resulted in a complete loss of viability at all developmentalstages tested with the exception of mature seeds that were ableto tolerate a 5% decrease in moisture content before germinationdeclined. ABA was detected in all embryos tested, with a maximum value16.·16 µg g-1 d. wt about midway through development.Although the presence of ABA induced no tolerance to desiccationof mature seeds, it did coincide with decreased content of waterin the developing seeds and decreased respiration. Desiccationdamage of M. thunbergii seeds occurred when moisture contentwas still high (45%) and this damage was not related to theabsence of oligosaccharides in the mature seeds. We concludethat developing embryos and mature seeds of M. thunbergii haveproperties common to many recalcitrant seeds, with seeds beingsensitive to desiccation at all stages, having a prominent ABApeak, little proline, lacking oligosaccharides, and specifically,little dormancy and a moderate rate of respiration of matureseeds (0·9 µmol O2 min-1 g-1 f. wt). Adenosinetriphosphate content and energy charge decreased from stagefour to stage eight of seed development, then increased againto 103 nmol g-1 d. wt and 0·73, respectively, in matureseeds. Effects of 2 °C chilling on the threshold moisture contentsand water potentials for various physiological processes wereestimated forAesculus hippocastanumL. seed. Seed harvested atthe time of maximum seed fall exhibited a dual response to drying:partial drying from near 50% to 32–40% moisture contentprogressively increased germination percentage (at 16 °C)up to various peak values; further desiccation was detrimental,confirming that the seeds are ‘recalcitrant’. Themoisture content for optimum germination was increased by atleast 10% as the chilling period was raised from 0 to 9 weeks.A negative linear relationship was found between log10mean timeto germinate and probit final germination, regardless of pre-treatment,indicating that partial desiccation and chilling are interchangeablein promoting germination of hydrated seed. For nearly fullyhydrated seeds, increasing the chilling period from 6 to 26weeks increased the viability-loss onset point for desiccationinjury from near 40% to about 48% moisture content without alteringthe drying rates of seed tissues. Extending moist chilling invarious seed lots from 0 to 26 weeks decreased subsequent longevityat 16 °C. For 26-week-chilled seeds longevity (the periodto lose one probit of germination) differed above and belowa threshold moisture content of 48%. It remained constant inthe moisture-content range 48–38%, but increased progressivelyas moisture content was raised above 48%. This threshold moisturecontent coincided with the value above which chilled seed pre-germinatedin storage. The onset and development of both the ability to germinate andto tolerate rapid enforced desiccation were investigated duringthe development and maturation of seeds of bean (Phaseolus vulgahsL.) at different temperatures and also after different slow-dryingtreatments. The onset of germinability occurred when seeds wereless than half-filled in the absence of both a post-ovule abscissionprogramme and water loss from the seeds. Maximum ability togerminate normally and maximum tolerance to rapid enforced desiccationto 14–16% moisture content did not occur until 2–23d and 6–23 d after mass maturity (end of the seed-fillingperiod), respectively. The slow-drying of immature seeds for7 d ex planta before rapid enforced desiccation increased theability to germinate and stimulated the onset of desicationtolerance. Holding seeds moist for 7 d (during which time moisturecontent declined by <5%) had similar effects, but seed germinationafter rapid enforced desiccation was consistently greater inseeds first dried slowly than held moist. Comparisons betweenseeds less than half-filled dried slowly ex planta and fullseeds undergoing maturation drying in planta showed that a similar(slow) rate of water loss over a 7 d period had a similar effecton the subsequent ability of seeds to tolerate rapid enforceddesiccation. Thus, neither a post-ovule abscission programmenor loss of water were required for the onset of the abilityto germinate in developing bean seeds, but both were requiredfor the development of the ability to germinate and resistanceto solute leakage, when rehydrated, after rapid enforced desiccation. The effects of different drying methods on desiccation toleranceand longevity in seeds of foxglove (Digitalis purpurea L.) wereassessed from just prior to mass maturity (when seeds have attainedmaximum dry weight), and at intervals during the post-abscissionphase of development. Tolerance of drying under seed conservationconditions (15% relative humidity, RH, and 15 °C), was acquiredclose to mass maturity at 36 d after flowering (DAF). Increasesin desiccation tolerance were induced when drying was delayedfor 4 d by placing seeds in a near-saturated atmosphere (approx.100% RH), or if seeds were pre-dried for 7 d at either approx.32% or approx. 73% RH. Irrespective of the drying treatment, seed longevity increasedthroughout the sampling period, i.e. beyond the point of massmaturity and throughout the post-abscission phase, up to thepoint of incipient natural dispersal. At each developmentalstage, delayed drying or pre-drying led to an increase in seedlongevity under controlled ageing conditions compared with seedsdried directly under seed conservation conditions. Increasesin longevity were apparent as increases in the estimates forthe intercept of transformed seed survival curves (Ki) and forthe standard deviation of the normal distribution of seed lifespans,and also in the mean time to death of individuals in storage,consistent with a continuation of ripening events. Over 50 per cent germination has been obtained from Elaeis guineensisform pisifera seeds stored in unaerated sterile distilled waterfor 6 months. The moisture levels of the seeds and excised embryoswere of the same order (20–30 and 60–70 per cent,respectively) as those of fully imbibed fresh seeds. The implicationsof an apparent lower oxygen requirement by seeds stored underwater as against germinating seeds are discussed in the contextof the successful storage. It has been suggested that rate of desiccation can influencethe expression of recalcitrant behaviour in seeds, thus complicatingthe task of determining which seeds are truly recalcitrant.The objective of this study was to see if variable rates ofdesiccation influenced such behaviour inQuercus nigra L., atree seed known to be recalcitrant. Acorn moisture content, leachate conductivity, and germinationwere determined at various times during desiccation at threerates at 27 and 40°C. Moisture contents and germinationdecreased as the severity of desiccation increased. Leachateconductivity increased slightly but was not a sensitive indicatorof loss of viability. The critical (lethal) moisture contentfor these acorns was 10–15%, although rehydration within48 h of reaching this level prevented death in about 25% ofthe acorns. At 27°C any desiccation treatment that producedlosses of 30–50mg of moisture per g of acorn dry weightper day should be suitable as a test for recalcitrance in thegenus. In seeds of Trifolium repens, T. pratense, and Lupinus arboreus,the hilum is a hygroscopically activated valve in the impermeableepidermis of the testa. When relative humidity was low the fissurein the hilum opened permitting the seed to dry out; when therelative humidity was high the fissure closed obstructing theabsorption of moisture. During seed-ripening the moisture contentfell readily to approximately 25 per cent., and thereafter moreslowly until the epidermis became impermeable at approximately14 per cent, moisture content. Further drying of the seed tookplace only by diffusion of water vapour through the hilum. ‘Hard’seeds tended to have a moisture content in equilibrium withthe lowest relative humidity to which they had been exposed.They absorbed moisture under conditions of gradually increasingrelative humidity such that the hilar fissure remained open.The duration of the impermeable condition increased with thedegree of desiccation brought about by loss of water throughthe hilum.  相似文献

Seeds of four cultivars of arabica coffee (Coffea arabica L.)were tested for germination following hermetic storage for upto 12 months at several different combinations of temperaturesbetween –20 °C and 15 °C and moisture contentsbetween 5% and 10% (wet basis). Most of the seeds from one cultivarwithstood desiccation to between 5% and 6% moisture content,a seed water potential of approximately –250 MPa, butthose of the remaining three cultivars were much more sensitiveto desiccation damage. Moreover, in all four cultivars, seedlongevity at cool and sub-zero temperatures, and at low moisturecontents did not conform with orthodox seed storage behaviour:viability was lost more rapidly under these conditions thanat either warmer temperatures or higher moisture contents. Theresults confirm that coffee seeds fail to satisfy the definitionsof either typical orthodox or recalcitrant seed storage behaviour.These results, therefore, point to the possibility of a thirdcategory of storage behaviour intermediate between those oforthodox and recalcitrant seeds. Investigations were undertaken on the desiccation and freezingsensitivity of recalcitrant seeds of three species: tea [Camelliasinensis (L.) O. Kuntze], cocoa (Theobroma cacao L.) and jackfruit(Artocarpus heterophyllus Lamk.). All species showed changesin the physiological characteristics, desiccation and freezingsensitivity of both the seed and the embryonic axes with increasingseed maturity. Fully mature seeds of tea, cocoa and jackfruitsurvived desiccation to 24, 35 and 31% moisture content, respectively,but at these moisture levels seeds were not able to toleratefreezing in liquid nitrogen (-196 °C). Some survival ofcryopreservation was, however, achieved for excised embryonicaxes of partially and fully-mature tea and jackfruit seeds afterdrying to 14% moisture content; cocoa axes were totally freezingsensitive at all three stages of physiological maturity studied.Biochemical investigations on fully mature axes after desiccationand freezing showed that the decline in viability with moisturelevel was associated with increased leachate conductivity, lipidperoxidation products and/or soluble carbohydrates. How Texas wild rice, Zizania texana, became isolated in the San Marcos River of Central Texas, hundreds of kilometres from other wild rice populations is not known. Zizania seeds are intolerant of short-term desiccation. Seeds desiccated at 14% relative humidity (RH) and 75% RH do not survive after only 5-6 d and 2-3 wk of drying. Water loss is rapid and reaches a maximum at the time of seed death due to drying. And although all Zizania seeds germinate well following a long, cold dormancy period, Z. texana seeds readily germinate in the isothermic water (22°C) of the San Marcos River and Springs without an obligate, cold dormant period. Within 30-60 d of collection, Z. texana seeds germinate in substantial numbers, unlike seeds of Z. palustris, which require a long, cold dormant period. The Texas population of Z. texana may represent a relict population of a once more widely dispersed wild rice population, since the San Marcos springs probably have never gone dry.  相似文献

The germinability, vigour, and desiccation tolerance of muskmelon(Cucumis melo L. cv. Top Mark) seeds was studied in relationto changes in seed water content during development within thefleshy fruit. Seed water content (fresh weight basis) declinedfrom 91% to 42% between 10 d and 35 d after anthesis (DAA) (whenmaximum dry weight was attained), then declined more slowlyto a minimum of 35% at 50 DAA before increasing again to 43%at 65 DAA. Fresh intact seeds were first germinable at 25 to30 DAA and attained maximum germination percentages at 45 DAA.Between 15 and 35 DAA, cotyledons, hypocotyls, radicles andepicotyls of isolated embryos (testa and perisperm enveloperemoved) sequentially developed the ability to grow when incubatedon water. Dehydration to water contents less that those attainedwithin the fleshy fruit is not a requirement for developmentof germination capacity of muskmelon seeds. Seeds became tolerantof rapid desiccation after 25 DAA, and drying of immature seeds(25 to 40 DAA) increased their germination percentages uponsubsequent imbibition. Washing, drying, or washing followedby drying increased seedling vigour (root length) as comparedto fresh seeds, which had very poor vigour. Water absorptionisotherms were constructed to test whether changes in water-bindingcomponents were correlated with the development of desiccationtolerance. Isotherms for seeds older than 25 DAA fit well tothe D'Arcy/Watt model, which postulates the existence of high-affinity,low-affinity and multi-molecular water-binding sites. Desiccation-intolerantseeds younger than 25 DAA lacked the component of the absorptionisotherm characteristic of the high-affinity water-binding siteswhich have been hypothesized to confer desiccation tolerance.However, we were unable to determine whether the absence ofhigh-affinity binding characteristics was specifically relatedto desiccation intolerance or was artifactual due to the lossof volatiles when immature seed samples were dried at high temperatures. The longevity and desiccation tolerance of samples of seedsof a japonica rice (Oryza sativa L.) harvested serially duringdevelopment from plants grown in two temperature regimes, viz28/20 °C and 32/24 °C (12/12 h) were determined. Massmaturity (defined as the end of the seed-filling phase) occurred19·7 and 18·3 d after 50% anthesis, respectively.Longevity (determined at 40 °C with 15% moisture contentand quantified by the value of the constant Ki of the seed viabilityequation) improved during seed development and maturation until17 and 14 d after mass maturity in the cooler and warmer regimes,respectively, but declined thereafter. Changes in Ki with timewere similar in the two environments until mass maturity, butthe increase in Ki values after mass maturity was much greaterin the cooler regime. Tolerance of desiccation to low (4%) moisturecontents improved until 22 and 14 d after mass maturity in thecooler and warmer regimes, respectively, when maturation dryinghad reduced seed moisture contents naturally to 24 and 32% moisturecontent, respectively. Further delays to seed harvest reduceddesiccation tolerance, particularly in the warmer environment.Comparison among 15 samples of seeds harvested at differenttimes in the two environments showed a strong correlation (r= 0·947, P < 0·01) between longevity (Ki) anddesiccation tolerance (to 4% moisture content). The embryo dormancy shown in freshly harvested samples of Acervelutinum seeds is weakly established and very short-lived.Loss of this embryo dormancy occurred during post-harvest fruitstorage at either 5 or 17 C. In contrast, the dormancy of intactfruits and seeds was overcome only during storage at the lowertemperature. Removal of the cotyledons from embryos of freshlyharvested fruits allowed more rapid germination of the embryonicaxes, indicating that the cotyledons exert an inhibitory effect,although the axes still retained a measure of innate dormancy.The inhibitory effect of the cotyledons became less marked withincreasing duration of fruit storage, this loss of inhibitoryeffect occurring at both storage temperatures. Applied ABA stronglysuppressed germinative capacity in intact embryos and isolatedembryonic axes from freshly harvested fruits, but when ABA wasapplied to embryos of fruits that had been stored for variousperiods at 5 or 17 C, the inhibitory effect was first weakenedand then lost with increased storage. Although dormancy in the seeds of A. velutinum may be describedas intermediate between testa-imposed dormancy and true dormancy,it is perhaps more properly included in the former category. The responses of Q. robur L. fruits, seeds and embryonic axesto desiccation are characterized and discussed in relation tocurrent knowledge of recalcitrant seed behaviour. A relationshipbetween viability and seed moisture content is described. Thisrelationship was unaffected by rate of drying, year of harvestor presence of the pericarp. Desiccation sensitivity did notincrease with storage. Excised embryonic axes survived to lower moisture contents thanintact seeds. However, in the intact seed, loss of viabilityappeared to be determined by a critical moisture content inthe cotyledons. Consequently, the level of desiccation tolerancewithin the axis attached to cotyledons was not determined byaxis drying rate. A link is drawn between the difference in the desiccation toleranceof embryonic axes and of cotyledons, and estimates of theirdifferent levels of matrix-bound water. The results presentedare consistent with a critical moisture content for survivalwhich is determined by the loss of all free cellular water.This hypothesis takes account of the differential desiccationsensitivity of seed tissues and differences in desiccation tolerancebetween species.  相似文献
