Four new bridging ligands containing two di-2-pyridylamine subunits have been synthesised. The coordination chemistry of these, along with one previously reported ligand, has been investigated through reactions with various silver(I), copper(II) and palladium(II) reactants. Typically, reaction of these ligands with silver(I) salts gave 1-D coordination polymers, within which the ligands act as divergent bridging units, while reaction with copper(II) and palladium(II) precursors provided discrete dinuclear complexes with chelating bidentate subunits. The complexes were characterised by a combination of elemental analysis, NMR spectroscopy and X-ray crystallography. The silver coordination polymers displayed interesting variations in supramolecular architecture, attributed to weak secondary interactions, such as Ag?π-bonding.  相似文献

The synthesis of three bis(thiosemicarbazone) compounds formed by the reaction of benzil with either thiosemicarbazide, 4-methyl-3-thiosemicarbazide or 4-phenyl-3-thiosemicarbazide are reported. The compounds were characterised by NMR spectroscopy, mass spectrometry and in the case of benzil bis(4-methyl-3-thiosemicarbazone) and benzil bis(4-phenyl-3-thiosemicarbazone) by X-ray crystallography. Attempts to purify benzil bis(thiosemicarbazone) and benzil bis (4-methyl-3-thiosemicarbazone) by recrystallisation resulted in the isolation of cyclised products that were characterised by X-ray crystallography. The 3 bis(thiosemicarbazone) compounds were used to synthesise both Cu(II) and Cu(I) complexes. The copper(II) complexes were formed by the reaction of the proligands with copper(II) acetate which gave neutral copper(II) complexes in which the thiosemicarbazone is doubly deprotonated, acting as a dianionic ligand. The copper(II)-benzil bis(4-phenyl-3-thiosemicarbazonato) complex was characterised by X-ray crystallography to show the copper in an essentially square planar N2S2 environment. The copper(I) complexes were synthesised by reacting the bis (thiosemicarbazone) ligands with [Cu(CH3CN)4]PF6 to give cationic complexes. The copper(I)-benzil-bis(thiosemicarbazone) complex was characterised by X-ray crystallography which revealed that the complex was a dimeric dication. Each of the benzil bis(thiosemicarbazone) ligands act as a bidentate N,S donor to each copper(I) atom, forming an overall helical structure in which each copper atom is in a strongly distorted tetrahedral N2S2 environment. Electrochemical measurements show that the copper(II)-benzil bis(thiosemicarbazonato) complex undergoes a reversible reduction at biologically accessible potentials.  相似文献

The condensation of 7-amino-4-methyl-coumarin (1) with a number of substituted salicylaldehydes yielded a series of Schiff bases (2a2k) in good yields. Subsequent reaction of these ligands with copper(II) acetate yielded Cu(II) complexes (3a3k) and some were characterised using X-ray crystallography. All of the free ligands and their metal complexes were tested for their anti-Candida activity. A number of the ligands and complexes exhibited anti-Candida activity comparable to that of the commercially available antifungal drugs, ketoconazole and Amphotericin B.  相似文献

Metallothioneins (MTs) are metal-chelating peptides that play an active role in zinc homeostasis. The participation of metal ligands other than cysteines and the presence of secondary structure elements in metal-MT complexes are fairly unknown, especially in nonvertebrate MTs. Here, four Zn(II) complexes of invertebrate MTs (mollusc, insect, nematode, and echinoderm) and the Zn(II)-MT complex of the mammalian MT1 isoform, heterologously synthesized in E. coli, were studied by analytic and spectroscopic techniques. By Raman and circular dichroism spectroscopy, new structural informations were obtained. The five analyzed MT isoforms consist largely of beta-turns with the near exclusion of alpha-helical segments. Raman spectroscopy was revealed as an useful tool, providing information about the state of the cysteine sulfur atoms (metal coordinated and oxidized), the participation of histidine in metal coordination, and the molecular environment of tyrosine residues. In all the five Zn(II)-MT studied samples, acid-labile sulfide anions were found as nonproteic ligands, since sulfide-containing and sulfide-devoid species coexisted in the corresponding preparations. Significantly, Raman bands useful as markers of sulfide bridging ligands were identified. Overall, this work illustrates how the combination of analytical and spectroscopic techniques can be a very informative approach for the analysis of in vivo-synthesized metal-MT complexes, providing new data on the metal binding behavior of MTs from the most diverse organisms.  相似文献

The synthesis and X-ray structures of copper(II) complexes of the bidentate ligands, N-(4-oxo-5,5-diphenyl-4,5-dihydro-1H-imidazol-2-yl)-N′-phenylguanidino, 2-uanidinobenzimidazolo and N-(4-oxo-3-phenyl-1,3-diazaspiro[4.4]non-1-en-2-yl)guanidino, are reported. These complexes, which possess potential doublet (DA) or triplet (DAD) hydrogen bonding motifs, can form supramolecular structures based on synthons involving hydrogen bonding or phenyl embraces. The changes in supramolecular structure resulting from small changes in ligand structure, as well as from the use of different solvents for their crystallisation, are examined. The structures adopted are compared with others reported previously for complexes of related ligands.  相似文献

The systematic investigation of the copper(II) complexes of tripeptides Xaa-Xaa-His, Xaa-His-Xaa and His-Xaa-Xaa, where Xaa=Gly or Ala was performed by combined pH-metry, spectrophotometry, CD and in part EPR spectroscopy. The matrix rank analysis of the spectral data revealed the number of the coloured and optically active species as a basis for the solution speciation. A critical evaluation on the speciation and solution structure of the complexes formed is presented on the basis of their d-d band optical activity. The replacement of a Gly residue with the chiral Ala amino acid allowed us to gain decisive information on the solution structure of the complexes by CD spectroscopy. It was shown that the tripeptides with histidine in the third position formed CuH(-2)L species with (NH(2), 2N(-), ImN - where Im stands for imidazole) coordination sphere as a major species, and only the macrochelated CuL complexes as minor species around pH 5.0. In copper(II)-Xaa-His-Xaa tripeptide systems the CuH(-1)L (NH(2), N(-), ImN) is the most stable species at physiological pH, but the vacant fourth site around copper(II)ions is offered for further deprotonation, most probably resulting in mixed hydroxo species at low (<5 x 10(-4)M) metal ion concentrations, while a tetrameric complex is dominant when the copper concentration exceeds 3 x 10(-3)M. The histamine type coordination mode in CuL and CuL(2) complexes of His-Xaa-Xaa ligands predominates at low pH. The structural consequences drawn from the CD spectra for the mono and bis parent complexes were supported by theoretical calculations. CD spectra strongly suggest the participation of the imidazole nitrogen both in the Cu(2)H(-2)L(2) and CuH(-2)L complexes.  相似文献

Platinum(II) complexes have been demonstrated to form covalent bonds with sulfur-donating ligands (in glutathione, metallothionein and other sulfur-containing biomolecules) or coordination bonds with nitrogen-donating ligands (such as histidine and guanine). To investigate how these compounds interact with cysteine proteases, we chose terpyridine platinum(II) (TP-Pt(II)) complexes as a model system. By using X-ray crystallography, we demonstrated that TP-Pt(II) formed a covalent bond with the catalytic cysteine residue in pyroglutamyl peptidase I. Moreover, by using MALDI (matrix-assisted laser desorption/ionization) and TOF-TOF (time of flight) mass spectrometry, we elucidated that the TP-Pt(II) complex formed a covalent bond with the active-site cysteine residue in two other types of cysteine protease. Taken together, the results unequivocally showed that TP-Pt(II) complexes can selectively bind to the active site of most cysteine proteases. Our findings here can be useful in the design of new anti-cancer, anti-parasite or anti-virus platinum(II) compounds.  相似文献

Four new ternary complexes of Cu(II) containing aspartic or glutamic acids and methionine or cysteine as ligands were studied both in solution and in solid state. The solution study was performed using potentiometry, UV-Vis spectroscopy, electron paramagnetic resonance (EPR) spectroscopy, and electrochemistry. The stability constants of the ternary complexes were determined by potentiometry. The order of the values for the stability constants was CuGluMet (complex 4) < CuAspMet (complex 2) < CuGluCys (complex 3) < CuAspCys (complex 1), with complex 1 ≈ complex 3 ? complex 2 ≈ complex 4. This behavior was confirmed by the electronic spectra, EPR parameters and cyclic voltammetry. The compounds were synthesized in the solid state and characterized by thermogravimetry as well as vibrational and EPR spectroscopy. The results found in solid state are in agreement with the data found in solution studies where methionine, aspartic and glutamic acids coordinate to the copper(II) ion using the nitrogen atom of the amino group and one oxygen atom of the carboxylate group and cysteine coordinates through the nitrogen and sulfur atoms. The EPR data suggest that the complexes are square planar and that the complexes with cysteine as one of the ligands have some distortion.  相似文献

Eight oxy-bridged dinuclear copper(II) complexes with catecholase-like sites, [Cu(L1)X]2 (HL1 = 1-diethylaminopropan-2-ol, X=N3- 1, NCO- 2, and NO2- 3), [Cu(L2)X]2 (HL2=N-ethylsalicylaldimine, X=NO3- 4, Cl- 5, N3- 6, NCS- 7), and [Cu(L3)]2(ClO4)2, 8 (HL3=N-(salicylidene)-N'-(2-pyridylaldene)propanediamine) have been prepared and characterized. The single crystal X-ray analysis show that the structures of complexes 6 and 8 are dimeric with two adjacent copper(II) atoms bridged by pairs of micro-oxy atoms from the L2 and L3 ligands. Magnetic susceptibility measurements in the temperature range 4-300 K indicate significant antiferromagnetic coupling for 4, 5 and 7 and ferromagnetic coupling for 6 between the copper(II) atoms. The catecholase activity of complexes for the oxidation of 3,5-di-tert-butylcatechol by O2 was studied and it was found that the complexes with the bond distance of Cu(II)...Cu(II) located at 2.9-3.0 A show higher catecholase activity.  相似文献

A 1,4-disubstituted dibenzofuran derivative of 1,4,7,10-tetraazacyclododecane (cyclen), L1, has been prepared by the direct reaction of cyclen and chloroacetyldibenzofuran and the mono-substituted derivative, L2, by reaction of chloroacetyldibenzofuran and 1,4,7-tris(t-butoxycarbonyl)-1,4,7,10-tetraazacyclododecane followed by deprotection with trifluoroacetic acid. The ligands were characterized by 1H and 13C NMR spectroscopy, IR spectroscopy and mass spectrometry. The reaction of the 1,4-disubstituted dibenzofuran cyclen, L1, with Cu(ClO4)2·6H2O in methanol yielded crystals of [CuL1](ClO4)2·MeOH·1/2H2O that were suitable for single crystal structural analysis. The X-ray structure confirmed that the 1,4-disubstituted dibenzofuran cyclen had been formed. The copper(II) coordination sphere in the complex cation, [CuL1]2+, is occupied by four nitrogen atoms from the macrocycle and an amide oxygen donor from one dibenzofuran pendant group. As is typical for copper(II)-cyclen complexes, the Cu(II) centre sits above the plane of the macrocycle nitrogen towards the oxygen donor, in this case by 0.5 Å. Fluorescence emission studies indicate that coordination of the macrocycle to either copper(II) or zinc(II) results in a decrease in emission with respect to the emission of the pure ligand.  相似文献

The synthesis and characterization of the copper(II) complexes of a series of tetradentate, pentadentate and hexadentate aminopyridine ligands that contain ethylenediamine and/or propylenediamine groups are described. The ligands include: 1,12-bis(2-pyridyl)-2,5,8,11-tetraazadodecane, TRIEN-pyr; 1,13-bis(2-pyridyl)-2,5,9,12-tetraazatridecane, DIEN-PN-pyr; 1,14-bis(2-pyridyl)-2,6,9,13-tetraazatetradecane, DIPN-EN-pyr; 1,15-bis(2-pyridyl)-2,6,l0,14-tetraazapentadecane, TRIPN-pyr; 1,9-bis(2-pyridyl)-2,5,8-triazanonane, DIEN-pyr; 1,11-bis(2-pyridyl)-2,6,10-triazaundecanenane, DIPN-pyr; 1,6-bis(2-pyridyl)-2,5-diazahexane, EN-pyr; and 1,7-bis(2-pyridyl)-2,6-diazaheptane, PN-pyr. The following methods were used to determine the binding geometries of the copper(II) complexes in the solid, solution, and gas phases: magnetic susceptibility measurements, absorption spectroscopy, EPR spectroscopy, electrochemistry, and electrospray ionization mass spectrometry. An X-ray structure was determined for the DIPN-pyr complex. The solid state structures were all found to be monomeric Cu(II) complexes with the coordination number set by the denticity of the ligand while the solution structures of all of the complexes except those with TRIPN-pyr and DIPN-pyr were found to be square pyramidal or elongated octahedral. The TRIPN-pyr and DIPN-pyr complexes showed considerable trigonal bipyramidal distortions. The gas phase data showed that the substitution of 6-membered for 5-membered chelate rings helped the ligand span more coordination sites. The TRIEN-pyr complex was 4- or 5-coordinate compared to the 5- or 6-coordination seen with the other three hexadentate ligands, and the DIPN-pyr complex was weakly 5-coordinate as compared to the 4-coordinate DIEN-pyr complex. The preferred structures of the ligands were consistent with their electrochemical behavior which showed the stability of the Cu(II) complex decreased in the order: DIPN-EN-pyr, TRIEN-pyr, DIEN-PN-pyr > DIEN-pyr > DIPN-pyr > TRIPN-pyr >  PN-pyr > EN-pyr.  相似文献

Two novel copper (II) substituted thiosemicarbazone Schiff base complexes [Cu(L1)(µ-SCN)]n(NO3)2 (1) and [Cu2(µ-SCN)(SCN)(L2)2](NO3) (2) have been synthesized by condensing substituted thiosemicarbazides like 4-methyl-3-thiosemicarbazide or 4-ethyl-3-thiosemicarbazide with 2-acetylpyridine. Both the metal complexes 1 and 2 are characterized using different spectroscopic techniques like IR, UV-Vis, ESR spectroscopy followed by elemental analysis, cyclic voltammetric measurement and single crystal X-ray structure analysis. X-ray crystal structure analysis reveal that complex 1 is polymeric while complex 2 is dimeric in nature. The coordination geometry around Cu(II) are square pyramidal in which thiosemicarbazone Schiff base ligand coordinate to the central Cu(II) atom in tridentate fashion. The prominent interaction patterns of 1 and 2 with CT-DNA were examined by employing electronic absorption and emission spectral titrations, cyclic voltammetry and viscosity measurements. All the results show that CT-DNA binds with both copper (II) complexes 1 and 2. Furthermore, protein binding ability in vitro of complexes 1 and 2 with both BSA and HSA were carried out using multispectroscopic techniques and a static quenching pattern was observed in both cases. Molecular docking study was employed to ascertain the exact mechanism of action of 1 and 2 with DNA and protein molecules (BSA and HSA). In vitro cytotoxicity activity of complexes 1 and 2 toward AGS and A549 was evaluated using MTT assay which demonstrates that both complexes 1 and 2 have superior prospectus to act as anticancer agents.

The coordination behaviour of the title ligand, 5-methyl-3-formylpyrazole N(4)-benzyl-N(4)-methylthiosemicarbazone(HMPz4BM), is reported with solid state isolation of copper(II) complexes, [Cu(HMPz4BM)X2] (X = Cl, Br, NO3, ClO4 and BF4) which have been spectroscopically and structurally characterised. I.r. data for the free ligand and its Cu(II) complexes indicate that HMPz4BM exhibits a neutral NNS tridentate function via the pyrazolyl nitrogen(tertiary), azomethine nitrogen and thione sulphur. Electronic spectral data are suggestive of a square pyramidal environment for the seemingly pentacoordinated Cu(II) species. E.s.r parameters (RT and LNT) of the reported copper(II) complexes are indicative of a dxx2–y2 ground state for the reported species. Cyclic voltammograms of Cu(II) complexes show a quasireversible CuII/CuIII couple and also an irreversible CuII/CuI couple. X-ray crystallography of a representative species, [Cu(HMPz4BM)(NO3)2] (C2/c, monoclinic ), has unambiguously documented the conjectural findings from i.r. data that coordinating sites of the title ligand are pyrazolyl (tertiary)nitrogen, azomethine nitrogen and the thione sulphur (NNS); and the oxygen of one of the nitrate ions has occupied the basal plane; the fifth coordination position has been occupied by the oxygen of another nitrate ion in a square pyramidal geometry. The antibacterial properties of the ligand and its copper(II) complexes studied on microorganism, Staphylococcus aureus have pointed out that most of the complexes have higher activities than that of the free ligand.  相似文献

 The role of the cysteine thiolate ligand for the unusual copper coordination geometry in the blue copper proteins has been studied by comparing the electronic structure, geometry, and energetics of a number of small Cu(II) complexes. The geometries have been optimised with the density functional B3LYP method, and energies have been calculated by multiconfigurational second-order perturbation theory (the CASPT2 method). Most small inorganic Cu(II) complexes assume a tetragonal geometry, where four ligands make σ bonds to a Cu 3d orbital. If a ligand lone-pair orbital instead forms a π bond to the copper ion, it formally occupies two ligand positions in a square coordination, and the structure becomes trigonal. Large, soft, and polarisable ligands, such as SH and SeH, give rise to covalent copper-ligand bonds and structures close to a tetrahedron, which might be trigonal or tetragonal with approximately the same stability. On the other hand, small and hard ligands, such as NH3, OH2, and OH, give ionic bonds and flattened tetragonal structures. It is shown that axial type 1 (blue) copper proteins have a trigonal structure with a π bond to the cysteine sulphur atom, whereas rhombic type 1 and type 2 proteins have a tetragonal structure with σ bonds to all strong ligands. The soft cysteine ligand is essential for the stabilisation of a structure that is close to a tetrahedron (either trigonal or tetragonal), which ensures a low reorganisation energy during electron transfer. The 1:1 condensation of 1,2-diaminopropane and 1-phenylbutane-1,3-dione at high dilution gives a mixture of two positional isomers of terdentate mono-condensed Schiff bases 6-amino-3-methyl-1-phenyl-4-aza-2-hepten-1-one (HAMPAH) and 6-amino-3,5-dimethyl-1-phenyl-4-aza-2-hexen-1-one (HADPAH). The mixture of the terdentate ligands has been used for further condensation with pyridine-2-carboxaldehyde or 2-acetylpyridine to obtain the unsymmetrical tetradentate Schiff base ligands. The tetradentate Schiff bases are then allowed to react with the methanol solution of copper(II) and nickel(II) perchlorate separately. The X-ray diffraction confirms the structures of two of the complexes and shows that the condensation site of the diamine with 1-phenylbutane-1,3-dione is the same.  相似文献

A series of novel copper(II) complexes, L2Cu with newly synthesized 3,5--salicylaldimine (or 5--salicylaldimine) ligands derived from 2,4-di-tert-butyl phenol (or 4-tert-butyl phenol) and alkyl (aryl) amines have been prepared and their spectroscopic (IR, UV-Vis, ESI-MS), X-ray, magnetic and redox properties have been investigated. The X-ray crystallography analysis shows that all complexes are monomeric and their copper(II) centers are surrounded by phenolate oxygens and imine nitrogen atoms. Therefore, the coordination sphere around the copper atoms is N2O2 as seen in galactose oxidase active site. In addition, the geometric configurations of all complexes are square planar or slightly distorted square planar. The crystal system for all complexes is monoclinic, except for which is orthorhombic. The temperature dependence of magnetic susceptibility of complexes confirms the mononuclear structure of complexes. Oxidation of the Cu(II) complexes yielded the corresponding Cu(II)-phenoxyl radical species during the cyclic voltammetry experiments.  相似文献

Preparations of copper(II) and palladium(II) complexes of 4-amino-5-methylthio-3-(2-pyridyl)-1,2,4-triazole (L(1)) and the copper(II) complex of 1,4-dihydro-4-amino-3-(2-pyridyl)-5-thioxo-1,2,4-triazole (HL) are described. These complexes have been characterized by means of spectroscopy and microanalysis. Molecular structures of HL (1), [CuCl(2)(H(2)L)]Cl.2H(2)O (2a), cis-[CuCl(2)(L(1))] (3), and cis-[PdCl(2)(L(1))] (4) have been determined by single-crystal X-ray diffraction. The HL ligand acts as a N,S bidentate through the thioxo moiety and the exo-amino group whilst the ligand L(1) forms N,N coordination complexes through the pyridine and triazole nitrogen atoms. Speciation in solution of the systems Cu/HL and Cu/L(1) have been determined by means of potentiometry and spectrophotometry as well as for the Cu/L(1)/A (HA=glycine) system in order to determine species present at physiological pH. Antiproliferative activity of these complexes and their ligands was evaluated, using the AlamarBlue Assay, on normal human fibroblasts (HF) and human fibrosarcoma tumor (HT1080) cells. The copper compounds cis-[CuCl(2)(H(2)L)]Cl and cis-[CuCl(2)(L(1))] exerted significant antiproliferative activity of both normal and neoplastic cells; although dose-response experiments revealed that the HT1080 cell line was more sensitive to the tested drugs than normal fibroblasts.  相似文献

Five Ni(II) complexes of aroyl hydrazone ligands with 2,6-diacetyl pyridine monooxime are reported. X-ray crystal structure of the Ni(II) salicyloylhydrazone complex is also reported. In these complexes Ni(II) is in a distorted octahedral N4O2 coordination environment, with each of the two ligands coordinating through the pyridine nitrogen, imino-hydrazone nitrogen and the deprotonated oxygen of the hydrazone moiety. The iminooxime group remain uncoordinated in both the ligands and the planes containing the CH3-CN-OH groups are orthogonal to the adjacent pyridine rings. On excitation at 375 nm, the ligands as well as the Ni(II) complexes, show luminescence. However, the Ni(II) complexes have much lower quantum yield of emission than the free ligands.  相似文献
