Previous work on chlorophyll-deficiency mutations in pea andbarley has shown that a significant increase in mutations isinduced by storing seeds under various conditions which leadto losses of viability to about 50 per cent. The work here showsthat a detectable increase in mutation frequency is also associatedwith much smaller losses of viability. Pea seeds were storedat 35 °C and 16.5 per cent moisture content for 40 and 57d when viability fell from 99 to 93 and 82 per cent, respectively.At the same time mutation frequency (percentage of seeds containingrecessive point mutations) increased from 1.62 per cent in thecontrol treatment to about 3 to 4 per cent. Barley seeds at15.5 per cent moisture content were stored at 50 °C for42 and 54 h, and at 35 °C for 28 and 39 d. During theseageing treatments viability fell from 98 to 75, 26, 93 and 48per cent respectively and the mutation frequency increased fromzero to between about 0.3 to 0.9 per cent. In both species theinduction of mutation by ageing treatments was significant butthe differences between the various ageing treatments were not.It is concluded that there is probably no safe threshold lossof viability which completely avoids mutation, and these resultssupport the view that for genetic conservation seeds shouldbe stored under conditions which minimise loss of viability. Over 50 per cent germination has been obtained from Elaeis guineensisform pisifera seeds stored in unaerated sterile distilled waterfor 6 months. The moisture levels of the seeds and excised embryoswere of the same order (20–30 and 60–70 per cent,respectively) as those of fully imbibed fresh seeds. The implicationsof an apparent lower oxygen requirement by seeds stored underwater as against germinating seeds are discussed in the contextof the successful storage. The Production and Distribution of Dry Matter in Maize after Flowering   总被引:3,自引:0,他引:3  
An experiment in which different groups of leaf laminae wereremoved, or ears shaded, shortly after silking showed that mostof the dry-matter increase after flowering was produced by upperleaves. The top five, the middle four, and the bottom six laminaeaccounted, respectively, for 26 per cent, 42 per cent, and 32per cent of the leaf area duration (D) of the laminae afterflowering; the estimated contributions of the three groups todry-matter production by the laminae after flowering were about40 per cent, 35–50 per cent, and 5–25 per cent,respectively. The sheaths provided about one-fifth of the totalleaf area and probably contributed about one-fifth, and laminaefour-fifths, of the total dry matter produced after flowering.The contribution from photosynthesis by the ear was negligible,presumably because its surface area was only 2 per cent of thatof the leaves. Leaf efficiency (dry matter produced per unitarea) decreased greatly from the top to the base of the shoot.When laminae were removed, the grain received a larger fractionof the dry matter accumulated after flowering, less dry matterremained in the stem, and the photosynthetic efficiency of theremaining leaves was apparently increased. When alternate laminae were removed at the time of silking (half-defoliation)D was decreased by 40 per cent, and the subsequent productionof dry matter decreased nearly proportionately, so that netassimilation rate (E) was not affected but grain dry weightwas decreased by only 32 per cent. At the final harvest, thegrain of half-defoliated plants constituted 80 per cent of thedry matter accumulated after flowering, compared with 65 percent for intact plants. Stem weight decreased from two weeksafter flowering in half-defoliated plants, but remained nearlyconstant in intact plants. When pollination was prevented andno grains formed, E during the first month after flowering wasunaffected; the dry matter that would have passed into the grainaccumulated in the stem and husks, not in the leaves. The decrease in stem weight caused by defoliation suggests thatpreviously stored dry matter was moved to the grain. That suchmovement is possible was shown by keeping prematurely harvestedshoots in the dark for two weeks with their cut ends in water;the dry weight of the grain increased and that of the stem,laminae, husks, and core decreased. Nevertheless, dry-matterproduction after flowering was more than sufficient for graingrowth, and previous photosynthesis probably contributed littleto the grain.  相似文献

Seeds of the Smooth-leafed Elm (Uimus carpinifolia) and of thetropical forest tree Terb (Terminalia brassii) were stored hermeticallyand sampled at intervals for periods of up to two years. Bothspecies possess ‘orthodox’ seed (increasing longevityis observed as either moisture content or temperature are reduced)within the temperature ranges from — 13 to 52°C (Elm)and from —4 to 42°C (Terb) and within the moisturecontent ranges from 3 to 19 per cent (Elm) and from 5 to 14per cent (Terb) on a fresh weight basis. Elm seed stored at—75°C showed the expected relationship between longevityand moisture content, but did not differ significantly in longevityfrom seed kept at — 13°C when moisture contents wereheld constant. Probit analysis of the relationship between germinationpercentage and time was performed for each storage environment,yielding a slope from which the standard deviation of the distributionof seed deaths over time () was calculated. Standard deviationvalues were used in turn to determine the values of constantsin a viability equation which had previously been applied toseed of barley, chickpea, cowpea and soybean. The equation,which gave a good fit to the results obtained, can be used topredict viability for seed in storage over a wide range of environmentalconditions. Some limitations to the applicability of the viability equationwere defined. At 22 per cent and higher moisture contents Elmseed survived longer than predicted. Furthermore, all Elm andTerb seed was killed quickly on placing in —75°C at22 and 20 per cent moisture content respectively, but high viabilitywas retained for several days at 19 and 17 per cent respectively.Practical implications of the results are discussed. Wheat seeds (Triticum durum) were stored under both hermeticand aerobic conditions at 25 °C with moisture contents from15 to 33 per cent. Under hermetic storage, seeds lost viabilitymore rapidly the higher the moisture content, whereas in aerobicstorage, seed longevity was enhanced as the moisture contentwas increased from 24 to 31 per cent, and over this range ofmoisture content the seeds survived longer under aerobic thanhermetic storage. On the contrary, an apparent reversal of thistrend occurred when moisture content was increased above 31per cent. The possibility that the changes in longevity occurring at highermoisture contents might be due to the activation of seed metabolismwas supported by the enhanced incorporation of [3H]leucine intoTCA insoluble material (indicating increased protein synthesis)and the reduced leakage of glucose (indicating increased membranerestitution) when seeds were allowed to achieve higher moisturecontents during the prehydration period. The highest level ofseed activation was found in seeds preconditioned to about 31per cent moisture content. Moreover, these seeds, when subsequentlystored under aerobic conditions, maintained a higher rate ofprotein synthesis and lower membrane permeability during thestorage period than seeds at lower water contents. It is suggested that seeds stored at a sufficient hydrationlevel in the presence of oxygen can sustain an effective metabolismduring extended storage, thus permitting the repair of cellulardamage. However, it might be possible that at about 33 per centmoisture content seed could suffer from an excessive advancementof metabolism. Loss of seed viability in lettuce (Lactuca sativa L.) duringstorage is associated with an increase in the frequency of cellsin the surviving seeds showing chromosome damage during firstmitoses. The relation is linear when probit of the frequencyof aberrant cells is plotted as a function of probit percentagenormal germination. The slope of the relation, however, variesaccording to moisture content so that the proportion of aberrantcells for any given loss of germination increases with decreasein moisture content over the range 13.0–5.5 per cent.At 3.3 per cent moisture content, however, the proportion ofaberrations was no greater than at 5.5 per cent moisture content;and at 18.1 per cent moisture content the proportion was noless than at 13.0 per cent moisture content. Despite these differences,the increase in chromosomal aberrations per unit time for agiven temperature was always less the lower the moisture content.Diplontic selection markedly reduced the frequency of chromosomalaberrations and eliminated the differences in these frequenciesbetween the different storage treatments. But even after fiveweeks' growth, root tips from aged seed still contained abouttwice as many aberrant cells as compared with similar root tipsderived from the original seed stock. Studies on the frequencyof recessive mutations indicated that excessive amounts of heritablemutations were not present in the progenies of aged seed, evenwhen stored at moisture contents as low as 5.5 per cent. Allthis and other evidence reinforces the view that orthodox seedsfor genetic conservation should be stored at not more than about5 per cent moisture content, and that even lower moisture contentsare worth considering. The results also emphasise the need formaintaining a high regeneration standard, i.e. the percentageto which seed viability is allowed to fall during storage beforethe seed stock is regenerated. To help in understanding the mechanisms of dormancy in wheat(Triticum aestivum L.), brief drying of intact ears has beenexamined as a technique for rapid imposition of dormancy. Beforenatural grain desiccation at 40 d post anthesis (dpa), wholeplants were moved for 15 to 20 d to ‘wet’, humidconditions (90–100% RH) or to ‘dry’ conditions(35–40% RH). The duration of subsequent dormancy was atleast doubled by the ‘dry’ treatment, however, onlyif the grain was allowed to dry before 50 dpa. In the ‘wet’ears there was a low level (up to 22%) in-ear germination by55 dpa and the remaining, non-sprouted grain no longer becamedormant on drying. Brief (4 d) drying episodes could also preventsprouting and induce some dormancy, but only over a 4 to 10d window of sensitivity before 50 dpa. Grain drying when properlytimed not only arrests development but, perhaps through damagingeffects of drying, causes dormancy. In seeds of Trifolium repens, T. pratense, and Lupinus arboreus,the hilum is a hygroscopically activated valve in the impermeableepidermis of the testa. When relative humidity was low the fissurein the hilum opened permitting the seed to dry out; when therelative humidity was high the fissure closed obstructing theabsorption of moisture. During seed-ripening the moisture contentfell readily to approximately 25 per cent., and thereafter moreslowly until the epidermis became impermeable at approximately14 per cent, moisture content. Further drying of the seed tookplace only by diffusion of water vapour through the hilum. ‘Hard’seeds tended to have a moisture content in equilibrium withthe lowest relative humidity to which they had been exposed.They absorbed moisture under conditions of gradually increasingrelative humidity such that the hilar fissure remained open.The duration of the impermeable condition increased with thedegree of desiccation brought about by loss of water throughthe hilum.  相似文献

Spring wheat was grown to maturity in three growth rooms providing:(a) 18 h of light at 20° C and 6 h of darkness at 15°C (hot long days, HL); (b) 18 h of light at 15° C and 6h of darkness at 15° C (cold long days, CL); (c) 14 h lightat 20° C and 10 h of darkness at 15° C (hot short days,HS). Plants were moved between environments at spikelet initiationand anthesis, so dividing the growth period into three. Meanlengths in days of these periods in the different environmentswere: Period 1: HL 16, CL 18, HS 25; Period 2: HL 42, CL andHS 61; Period3: HL 53, CL 83, HS 63. The length of periods 2and 3 also depended on previous treatments. Grain dry weight was affected by environmental differences inall periods and effects in successive periods were additive.Compared with HL, CL or HS in period I before initiation increasedgrain yield by 6 per cent by increasing grain number per ear,HS in period 2 between initiation and anthesis decreased itby 24 per cent by decreasing the number of grains per spikeletand the proportion of spikelets that contained grain; CL inperiod 2 increased it by 21 per cent by increasing the numberof ears; CL in period 3 after anthesis increased it by 16 percent because leaves died later; HS in period 3 decreased itby 14 per cent because there was less radiation and hence lessphotosynthesis. Dry weight of shoot and root at maturity wasincreased by CL or HS in periods 1 or 2, and increased by CLand decreased by HS in period 3. The effects on final yieldof treatment during periods 1 and 2 were the consequence ofsimilar effects already produced at anthesis, and shoot androot dry weight changed little during period 3. The effects of environmental differences on grain dry weightcould not be explained by differences in leaf-area durationafter anthesis (D3), except that CL in period 3 increased bothyield and D3 but not proportionately, so that, as with HS inthe same period, grain: leaf ratio was decreased. Environmentaldifferences in periods 1 and 2 appeared to affect grain weightby altering the capacity of the ear to accumulate carbohydrates,determined by the number of grains per ear, rather than by alteringthe supply of carbohydrates, determined by D3. Seed of three chickpea (Cicer arietinum L.), three cowpea [Vignaunguiculata (L.) Walp.] and four soya bean [Glycine max (L.)Merr.] cultivars were hermetically stored for up to 2 yearsin various constant environments which included temperaturesfrom —20 to 70 °C and moisture contents (fresh weightbasis) from 5 to 25 per cent. In all cases the survival curvescould be described by negative cumulative normal distributions.The longevity of the various seed lots differed but the valueof the standard deviation (the reciprocal of which gives theslope of the survival curve when percentage germination is transformedto probit) was the same for all cultivars within a species whenstored under similar conditions. Within each species the relativeeffects of moisture and temperature on longevity did not differsignificantly between cultivars. In all three species therewas a negative logarithmic relationship between seed moisturecontent and longevity, but the relative effect of moisture contentdiffered between the species: differences in the longevity ofsoya bean seed as a function of moisture content were less thanfor either cowpea or chickpea. The relative effect of temperatureon seed longevity did not differ between the three species,and the seed of all three species showed increasing temperaturecoefficients for the change in rate of loss of viability withincrease in temperature. The complete pattern of loss in viabilityin all three species can be described by a single equation whichwas developed for barley and has also been shown to apply toonion seed. The constants applicable to the three grain legumeshave been calculated so that it is now possible to predict percentageviability of any seed lot of these species after any storageperiod under a very wide range of storage conditions. Germination of Triticum durum Desf. ‘Cappelli’ caryopsesstored in hermetically-sealed containers at 10°C or -20°Cwas analysed. Caryopses were maintained in laboratory conditions(20 ± 4°C) prior to controlled storage, which began5 d, 240 d and 7 years after harvesting. In addition, after9 years of storage, one 10°C stored batch of caryopses andtwo -20°C stored batches were returned to laboratory conditions.Germination over time and seed longevity were investigated.Results showed that: (1) under laboratory conditions, caryopsesin relative (primary) dormancy at the beginning of storage hadlost dormancy after 45 d and germination ability was lost bythe end of year 7. (2) When stored at 10°C, relative dormancyin caryopses was lost within 1 year, but 100% germination abilitywas retained after 23 years of storage. (3) When stored at -20°C,caryopses that were dormant at the beginning of storage (5 dafter harvesting) maintained this condition for 23 years whilecaryopses which were placed in storage 240 d after harvesting,when relative dormancy had already been broken, maintained 100%germination ability. Caryopses returned to laboratory conditionsafter 9 years of storage at 10°C or -20°C showed thesame trend as caryopses maintained exclusively in laboratoryconditions since the time of harvesting. Caryopses removed from-20°C overcame relative dormancy in 50 d and maintainedgermination ability for roughly 7 years, while those removedfrom 10°C lost the ability to germinate by the end of thefifth year. Respiratory Loss of Recently Assimilated Carbon in Wheat   总被引:2,自引:0,他引:2  
A series of experiments was undertaken to assess the amountof respiration associated with the growth of wheat at differentstages. Plants (or in some cases just the flag leaf) were labelledwith 14CO2 and the amount of 14CO2 respired during the subsequent48 or 72 h was measured. The evolution of 14C, expressed asa percentage of the amount initially assimilated (referred toas the R/A value) was used as a measure of the overall efficiencyof dry matter production. Respiratory 14CO2 evolution from labelledplants was most rapid in the first 12 h after labelling, thereafterdeclining rapidly. Evolution was also more rapid following labellingsat the end of the light period (dusk) than at the beginningof it (dawn). The R/A values were greatest (42 and 50 per centrespectively for dawn- and dusk-labelled plants) for young plantsand least (13 and 28 per cent respectively) for plants duringmid grain filling. When flag leaves, as distinct from wholeplants, were labelled, R/A values were lower still (9 and 21per cent respectively), indicating that flag leaf assimilatewas used efficiently in grain production. The calculated minimum R/A for the formation of grain material(10 per cent protein, 90 per cent starch) was 6.2 per cent.That the experimentally determined values were greater thanthis is attributed to the turnover of carbon in enzymes, toother maintenance processes, and possibly to the operation ofthe pentose phosphate pathway of glucose oxidation. R/A valueswere lower in those plants labelled at the beginning than thoseat the end of the photoperiod. This was considered to be a consequenceof refixation of respiratory 14CO2 during the light. The higherR/A values found for young plants were considered to be a consequenceof the greater percentage of 14C translocated to the roots (rootsbeing unable to refix respired CO2) and of greater turnoverof enzymes associated with more active metabolism. The survival of seeds of lemon (Citrus limon L.), lime [C. arantifolia(Christm.) Swing.] and sour orange (C. aurantium L.) was examinedunder a wide range of constant moisture contents and temperatures.Seed longevity was increased by decreasing the moisture contentand temperature of the storage environment. Maximum viabilitywas maintained in the combination of storage conditions includingthe lowest moisture content (5 per cent) and lowest temperature(–20 °C) investigated. Isolated embryos of three contrasting barley genotypes werecultured in vitro on a range of media comprising 16 combinationsof sucrose (3–12 per cent) and 2, 4-D (0–8 mg 1–1)concentration. Cultures were incubated at a range of temperaturesfrom 5 to 25°C and were examined after 21 days when shootlengths as well as fresh and dry weights were recorded. Therelative influence on growth of increasing concentrations ofsucrose and 2, 4-D was investigated, as was their interactionwith the incubation temperature. The genotypes were found todiffer markedly in their response to these two media components,with each parameter of growth differentially affected. Freshly harvested seeds of Zizania palustris L. representingfour distinct stages of development were intolerant of desiccation.Rapid drying did not improve survival. The relationship betweenprobit viability and embryo moisture content for freshly harvestedmature seeds was identical to that described in an earlier publicationfor seeds previously stored fully imbibed at low temperaturesto remove dormancy. A small reduction in embryo moisture contentduring the first 3 d of drying at 15% r.h. and 15°C ledto a highly sigificant (P < 0.01) increase in the proportionof fresh seeds capable of germination in response to a rangeof dormancy breaking factors including giberellins. Seeds of two lettuce cultivars (Lactuca sativa L., cv. Meikoninginand cv. Grand Rapids) were hermetically stored with constantmoisture contents ranging between 3.6 and 17.9 per cent (freshweight basis) at constant temperatures ranging between 5 and75 °C. The decline with time in percentage germination andpercentage normal seedlings was determined for each storagetreatment. The data were fitted to an equation which containsthe constants: K1, the probit of the initial percentage germinationor normal seedlings; KE, a species constant; CW, the constantof a logarithmic moisture term; CH, the constant of a lineartemperature term and CQ, the constant of a quadratic temperatureterm. Regression analysis of data from storage periods up to5.5 years at temperatures of 5–75 °C and seed moisturecontents of 3.6–13.6 per cent yielded the following values:KE= 8.218, CW=4.797±0.163, CH=0.0489±0.0050 andCQ=0.000365±0.000056. Although this equation consistentlyprovided a better fit, simplified equations, assuming eithera log-linear relationship between seed longevity and temperature,or a log-linear relationship between seed longevity and bothmoisture content and temperature, accounted for more than 94per cent of the variation at the restricted temperature rangeof 5–40 °C. Longevity of the same seed lots at sub-zero temperatures (–5,–10 and –20 °C) was studied in separate tests.Freezing damage, resulting in abnormal seedlings in the germinationtest, occurred at –20 °C when the moisture contentof the seeds exceeded 12 per cent. No decline in percentagenormal seedlings was observed after a storage period of 18 monthsor longer at –20 °C, provided the seed moisture contentdid not exceed 9.5 per cent. For seeds stored at –5 and–10 °C with 9.6–12.5 per cent moisture content,the observed rate of decline of percentage normal seedlingswas adequately predicted by the viability equation, using theabove values for the constants. This suggests that for low moisturecontents the viability equation can be applied to estimate longevityat sub-zero temperatures. Three-month ‘old’ and ‘fresh’ seedsof Talinum triangulare were subjected to various treatmentsto induce early and rapid germination. Scarification and activated carbon were the most effective treatmentsin improving total germination in fresh seeds, while the 3 and5 per cent thiourea treatments were most effective in improvingtotal germination in old seeds. Activated carbon, scarificationand 5 per cent thiourea treatments enhanced early germinationin both old and fresh seeds. Cumulative percentage germinationwas very high in fresh seeds after scarification or after treatmentwith activated carbon and 5 per cent thiourea, and lowest inseeds treated with 3 per cent thiourea and hot water. In oldseeds, highest cumulative percentage germination was obtainedwith 3 and 5 per cent thiourea treatments and scarification.Generally, higher germination was obtained with fresh seedsthan with old seeds. Partial seed-coat removal and treatment with 5 per cent thiouriaresulted in a higher rate of and cumulative percentage germinationcompared with seeds with the coat partially removed but nottreated with thiourea. Constantly high temperature (34 °C) increased both rateand total germination compared with seeds planted at room temperature(20–23 °C). Treatments that did not induce germinationwere 1 per cent thiourea, H2SO4, cold water soaking at roomtemperature, 6 per cent hydrogen peroxide and soil planting.These treatments effected less than 3 per cent germination. A 2 h soaking treatment in distilled water, or in aqueous solutionsof cysteine, potassium iodide, or sodium thiosulphate, had nosignificant effect (P > 0.25) on the subsequent longevityof lettuce seeds (Lactuca sativa L.) in two different storageenvironments. Neither did these treatments influence relationsbetween loss in germination and the frequency of chromosomalaberrations observed during first mitoses after storage. Incontrast partial hydration of lettuce seeds after storage byexposure to moist air (humidification) or to an osmoticum (priming)reversed some of the damage which resulted from ageing. Mostof the benefits occurred during the first 3 d of humidificationduring which seed moisture content rose to 34 per cent, or duringthe first 7 d of priming when seed moisture content increasedto 44 per cent. Both post-storage hydration treatments reducedthe frequency of chromosomal aberrations, increased the rateof root growth, and decreased the frequency of morphologicallyabnormal seedlings. Either treatment could be of practical use,but it is suggested that humidification is more convenient.Consideration should be given to adopting a humidification treatmentas standard practice following long-term seed storage for geneticconservation. The relationship between seed moisture content and seed longevityin sesame (Sesamum indicum L.) in hermetic storage at 50 °Cis logarithmic. The logarithmic relationship is maintained from15 per cent down to 2 per cent – the lowest moisture contenttested — but above 15 per cent this ‘air-dry’relationship no longer holds since further increase in seedmoisture content does not reduce longevity. Tentative estimatesof constant values for the improved seed viability equationare provided, and implications for long-term storage are discussed.
