Bone marrow (about 0.5 ml) from au erythropoietic region is freed of blood clots by washing 1-3 min in 1 μg/ml colchicine solution (2-3 ml) and then soaking 1-2 hr at 20-30° C in a second change. For mammalian or avian marrows, the colchicine is made up in phosphate-buffered (pH 7) physiological NaCl solution; for amphibian, Ringer's A solution. Next the specimens are soaked about 20 min in a hypotonic solution as follows: for mammalian, 1% Na-citrate; for avian, a 1:4 dilution of the buffered NaCl solution by distilled water; and for amphibian, Ringer's A-distilled water, 1:1. Then they are heated in a mixture of 2% orcein in 45% acetic acid and 1 N HCl, 9:1. Immediately after heating, squash preparations are made with 2% acetic-orcein in the usual manner. An alternative method is to dissociate the marrow cells by agitating after colchicine treatment. Then, recovering the cells between changes by low-speed centrifugation, to carry out the hypotonic treatment and subsequent fixation in Carnoy's solution I (alcohol acetic, 3:1) before drying the cells onto slides from the fixative. After thorough drying the slides may be stained 10-20 min in acetic orcein, or by other suitable technics.  相似文献

Abstract

The physiological response of Hylocomium splendens (Hedw) Br. Eur. to incubation in distilled water and solutions of phosphate, arsenate, selenite and selenate was investigated. Potassium efflux was found useful for comparing toxicity but was unreliable for short-term studies due to an initial rapid loss of potassium within ten minutes of immersion in any solution. This potassium loss could, under noninhibitory conditions, be regained by moss shoots. Selenite prevented re-accumulation of potassium lost in the first few minutes of incubation whilst arsenate promoted potassium loss throughout incubation. Photosynthetic oxygen evolution was progressively reduced under conditions of increasing arsenic uptake. Thus, photosynthetic rates were a more sensitive measure of anion toxicity than potassium efflux.

Electron microscopic observations revealed that incubation in distilled water had little effect on immature leaves of H. splendens whilst arsenate incubation induced loss of cytoplasmic electron density and swelling of chloroplast lamellae in immature tissue. Mature leaves, incubated in both arsenate and distilled water, showed changes in celluar morphology compared to non incubated tissue. These changes were not reflected in potassium efflux data. The results indicate that shortterm submersion is disruptive to the fine structure of moss shoots. This is discussed in terms of the design and interpretation of experiments involving toxic ions.  相似文献

In field-grown sugar-beet concentration of insoluble oxalatewas low in roots and high (about 12 per cent of ethanol insolublematerial) in leaves, and for a particular leaf the concentrationincreased continuously during its life. Of the insoluble oxalate,15–30 per cent was present as the magnesium salt and theremainder as the calcium salt. Oxalate contents of plants grownin culture solutions with nitrate as nitrogen source were similarto those of plants grown in soil, but when nitrogen was suppliedas ammonium sulphate or ammonium nitrate both soluble and insolubleoxalate were low. Plants grown in soil with regular additionsof ammonium sulphate or ammonium nitrate also had very low concentrationsof soluble oxalate although insoluble oxalate was only slightlylower than with nitrate nitrogen. Disks of root or leaf tissuewashed for several days in distilled water lost insoluble oxalatebut when washed in tap water insoluble oxalate increased morethan twofold. Addition of calcium and nitrate to the distilledwater caused an increase of insoluble oxalate, while additionof potassium caused a decrease. Use of 14C labelled oxalateand washing experiments showed that oxalate can be metabolizedby tissue disks and so is not necessarily a final product ofmetabolism. The accumulation of oxalate appears to be connectedwith the assimilation of nitrate and the preservation of thecation-anion balance of the plant.  相似文献

Rat and rabbit brains containing surgical lesions of 5-10 days' duration were fixed in 10% formalin (neutralized with calcium carbonate) for 1 week to 6 months. Frozen sections (15-20 n) were rinsed and then soaked 7 minutes in a 1.7% solution of strong ammonia in distilled water. Subsequent treatment was as follows: rinse; 0.05% aqueous potassium permanganate 5-15 minutes; 0.5% aqueous potassium metabisulfite, 2 changes of 2.5 minutes each; wash thoroughly in 3 changes distilled water; 1.5% aqueous silver nitrate, 0.5-1.0 hr.; 1% citric acid, 5-10 sec.; 2 changes distilled water; 1% sodium thiosulfate, 30 see.; 3 changes distilled water. Each section is then processed separately. Ammoniacal silver solution (450 mg. silver nitrate in 10 ml. distilled water; add 5 ml. ethanol; let cool to room temperature; add 1 ml. strong ammonia water and 0.9 ml. of 2.5% aqueous sodium hydroxide), 0.5-1.0 min. with gentle agitation. Reduction of about 1 minute is accomplished in: distilled water, 45 ml.; ethanol, 5 ml.; 10% formalin, 1.5 ml.; 1% citric acid, 1.5 ml. Rinsing; 1% sodium thiosulfate, 10 sec.; thorough washing followed by dehydration through graded alcohol and 3 changes of xylene or toluene complete the staining process. Normal nerve fibers are slightly stained to unstained, degenerating fibers, black. The treatment in potassium permanganate is critical since too little favors overstaining of normal fibers and too much abolishes staining of degenerating fibers.  相似文献

A slowly activating potassium current was expressed in Xenopus oocytes by injection of RNA transcribed from a rat kidney cDNA clone. Hypotonic solutions (160 mOsmol/l; control was 220 mOsmol/l) increased the current by increasing the rate of activation and by decreasing the depolarization needed to activate the current. This effect of hypotonicity was not observed in calcium-free solution, but was unaffected by staurosporine or the calmodulin antagonist W7. Cytochalasin D reduced the current and prevented the increase by hypotonic solution. The results suggest that the increase in this potassium current by hypotonic solution might result from calcium entry and changes in the actin network.  相似文献

Changes in the volume of fiber water in hypotonic and hypertonic Ringer's solution were determined for the sartorius, stomach, and cardiac muscle of the frog using two methods. Loss of water in hypertonic solutions was nearly the same in all muscles, but swelling in hypotonic solutions was greatest in the sartorius, smallest in the heart. For the sartorius the deviation from the properties of an osmometer can be accounted for by a loss of electrolyte and by assuming that a small part of the fiber water is bound, but this appears insufficient to explain the behavior of stomach and cardiac muscle in hypotonic solutions. In very dilute solutions of CaCl2 and MgCl2 a large difference in concentration of electrolytes is maintained between the fibers and the medium. Under these conditions divalent cations, accumulating in the fibers, produce a change in physical properties which indicates increased internal cross-linking. It is suggested, therefore, that swelling is limited as in a gel and that a considerable hydrostatic pressure may develop within the fibers.  相似文献

Preparation of isolated large intestine of the frog was filled with Ringer's solution diluted with distilled water (1:5) and was placed into the glass with normal Ringer's solution. The preparation was weighed within every 30 min and the osmotic permeability was determined for water of the mucous and serous layers of the intestine. Then one of the peptides was added to Ringer's solution and the experiment continued. It is stated that bombesin, neurotensin, encephalins, substance P, somatostatin, pituitrin are able to change liquid absorption from the large intestine cavity when the concentration of Ringer's solution in the cavity and from its serous surface is the same. Bombesin and neurotensin inhibited while encephalins stimulated liquid absorption and these effects depended on the transport of ions. Liquid absorption by the osmotic gradient decreased using bombesin, substance P and increased using somatostatin. More complex peptide-peptide relations are observed if using pituitrin and other peptides. cAMP is shown to participate in bombesin effects.  相似文献

Potassium currents through the somatic membrane of giant neurons ofHelix pomatia in normal (10 mM Ca) Ringer's solution and low-calcium (1 mM Ca) solution were studied by the voltage clamp method. With a decrease in the Ca concentration to 1 mM peak potassium conductance versus membrane, potential curves and inactivation curves were shifted along the voltage axis in the negative direction by about 10 mV. Inactivation of the delayed potassium current was slowed in low Ca solution. The effect of a decrease in external calcium concentration on volt-ampere and inactivation characteristics increased with a rise in external pH. These effects of a low Ca concentration on potassium mechanisms of the giant neuron somatic membrane can be attributed to changes in the negative surface potential in the region of the potassium channels.A. Soil patches on opposite sides of Pseudoroegneria spicata plants in the field were treated with either distilled water or a nutrient solution containing N, P, or K. Roots from these enriched and control patches were tested three days later for their capacities of ammonium, phosphate, and potassium uptake. When phosphate was augmented in the enriched patches, rates of phosphate uptake increased significantly, but not rates of ammonium or potassium uptake. When the enriched patches were augmented with nitrogen, uptake capacities of both ammonium and potassium increased significantly (mean increases of up to 88% and 71% for ammonium and potassium, respectively). Potassium augmentation did not lead to increased soil-available K and, correspondingly, did not induce changes in the capacity for uptake of K, N, or P. The potential importance of nutrient uptake kinetics in the exploitation of nutrient-rich soil patches is discussed.  相似文献

1. A study of the calcium equilibrium in isolated frog muscle has been attempted. 2. When sartorius muscles were immersed in Ca(45) Ringer's solution, the surface phase took up the Ca(45) in about 1 minute; the extracellular water space and connective tissue in about 30 minutes; and the intracellular space in about 300 minutes. 3. The percentages of total calcium in the whole muscle immersed in Ringer's solution was as follows: 10 per cent in the surface phase; 12 per cent in the extracellular water space; 17 per cent in the dry connective tissue; 24 per cent in the intracellular space; and 37 per cent as non-exchangeable calcium. 4. The exchange constants of isolated frog sartorius muscle to calcium has been determined. The flux of intracellular calcium in the steady state was approximately 0.8 mM/(liter hr). 5. It appears that there is a calcium pump pushing calcium out of the cell against an electrochemical gradient of about 4 cal./mM of calcium. However, since the flux is low, the maximum energy required per hour to pump calcium out of the cell against this high gradient is only about 2 cal./kg. muscle or about 1 per cent of the resting energy.  相似文献

Summary Red blood cells of the marine teleost,Opsanus tau (oyster toadfish), were characterized as to their normal hemoglobin, ion and water contents. Cells were exposed to ouabain containing, hyposmotic salt solutions (osmolarity reduced to 2/3 of normal) in which the cation or anion composition was varied. It was found that the initial cell volume expansion due to water influx was independent of the anion present. However, a secondary volume reduction was dependent on the presence of chloride or bromide anions. During volume reduction, cellular potassium and chloride ion contents fell by about equal amounts. Potassium loss was commensurate to the total amount of potassium ions detected extracellularly about 1.5h after the initial osmotic shock. No major changes were seen in the cellular sodium ion contents. When chloride ions within the cells and in the suspending medium were replaced by nitrate, iodide or thiocyanate, the cells failed to return to volumes close to those of isosmotically suspended controls, and the cellular potassium content also remained constant. In hypotonic potassium chloride the cells failed to extrude potassium chloride and water, and hence retained their expanded volume. Neither potassium loss nor volume decrease occurred in cells swollen in hypotonic sodium chloride media containing furosemide or 4,4 diisothiocyano-2,2-stilbene-disulfonic acid (DIDS). These two compounds are known inhibitors of monovalent cation cotransport and anion self exchange, respectively, in mammalian red cells. Hence toadfish red cells respond to osmotic swelling primarily by activation of an ouabain-insensitive, chloride dependent potassium transport system which is sensitive to inhibition by furosemide and DIDS.  相似文献

Summary In this paper we describe current fluctuations in the mammalian epithelium, rabbit descending colon. Pieces of isolated colon epithelium bathed in Na+ or K+ Ringer's solutions were studied under short-circuit conditions with the current noise spectra recorded over the range of 1–200 Hz. When the epithelium was bathed on both sides with Na+ Ringer's solution (the mucosal solution contained 50 m amiloride), no Lorentzian components were found in the power spectrum. After imposition of a potassium gradient across the epithelium by replacement of the mucosal solution by K+ Ringer's (containing 50 m amiloride), a Lorentzian component appeared with an average corner frequency,f c=15.6±0.91 Hz and a mean plateau valueS o=(7.04±2.94)×10–20 A2 sec/cm2. The Lorentzian component was enhanced by voltage clamping the colon in a direction favorable for K+ entry across the apical membrane. Elimination of the K+ gradient by bathing the colon on both sides with K+ Ringer's solutions abolished the noise signal. The Lorentzian component was also depressed by mucosal addition of Cs+ or tetraethylammonium (TEA) and by serosal addition of Ba2+. The one-sided action of these K+ channel blockers suggests a cellular location for the fluctuating channels. Addition of nystatin to the mucosal solution abolished the Lorentzian component. Serosal nystatin did not affect the Lorentzian noise. This finding indicates an apical membrane location for the fluctuating channels. The data were similar in some respects to K+ channel fluctuations recorded from the apical membranes of amphibian epithelia such as the frog skin and toad gallbladder. The results are relevant to recent reports concerning transcellular potassium secretion in the colon and indicate that the colon possesses spontaneously fluctuating potassium channels in its apical membranes in parallel to the Na+ transport pathway.  相似文献

Soluble and insoluble oxalate and insoluble calcium were measured in the leaves of Phaseolus vulgaris. The plants were grown in nutrient solutions with two different concentrations of calcium. Two developmental stages of the leaves were studied. Although the content of insoluble calcium differs widely according to leaf age and growth conditions, the percentage bound in crystals is nearly the same in all cases. In the growing leaves, concentrations of total oxalate are independent of calcium supply, thus, showing that the known rise in numbers of crystals, and of cells containing them, is not induced via oxalate biosynthesis. Fully expanded leaves contain more oxalate when grown in a nutrient solution with higher calcium concentration. Amounts of oxalate in percent of dry weight are similar to those given in the literature for other legume leaves.  相似文献

Potassium transport in microvessels isolated from rat brain by a technique involving density gradient centrifugation was studied in HEPES buffer solutions of varying osmolarity from 200 to 420 mosmols, containing different concentration of sodium chloride, choline chloride, or sodium nitrate. The flux of 86Rb (as a tracer for K) into and out of the endothelial cells was estimated. Potassium influx was very sensitive to the osmolarity of the medium. Ouabain-insensitive K-component was reduced in hypotonic medium and was increased in medium made hypertonic with sodium chloride or mannitol. Choline chloride replacement caused a large reduction in K influx. Potassium influx was significant decrease when nitrate is substituted for chloride ion in isotonic and hypertonic media, whereas a slight decrease was found in hypotonic medium. The decrease of K influx in the ion-replacement medium is due to a decrement of the ouabain-insensitive component. Potassium efflux was unchanged in hypotonic medium but was somewhat reduced in hypertonic medium. The marked effect of medium osmolarity on K fluxes suggests that these fluxes may be responsible for the volume regulatory K movements. The possible mechanism of changes of K flux under anisotonic media is also discussed.  相似文献

Summary Bullfrog tadpoles (Rana catesbeiana) were pre-exposed (7d) to 5 different solutions which varied in ionic composition and pH. Unidirectional sodium influx was then measured over a range of sodium concentrations (50–2000 M) and at 2 pH's (4.0 and 5.8). Tadpoles pre-treated in either distilled water or pH 4.0 soft water exhibited higher sodium influx than animals pre-treated in pH 5.8 soft water. Pre-exposure to solutions high in calcium or sodium reduced sodium influx. Tadpoles pre-treated in pH 4.0 soft water exhibited an increasedV max for sodium transport, whileK m was unchanged relative to tadpoles pre-treated in pH 5.8 soft water. In contrast,K m was increased andV max was unchanged in tadpoles pre-exposed to high concentrations of calcium or sodium. Within each pre-exposure treatment, influx was inhibited in pH 4.0 test water relative to pH 5.8 test water. Summary We describe the cannulated Rana pipiens, tadpole preparation that allows for stable recording in the tectum of the extracellular potential elicited by optic nerve stimulation. The largest components of the evoked tectal response consist of two previously identified waves and a major third, long-latency wave of long duration. These components were reversibly eleminated by perfusion of high magnesium/no calcium Ringer's solution or Ringer's solution containing cobalt chloride. In contrast, perfusion of high calcium/no magnesium Ringer's increased the amplitude and area of these components. We conclude that these components represent post-synaptic activity.Additionally, small, short-duration waves were identified as arising from the activity of retinal afferents. They consisted of a short-latency (3.1–7.6 ms) and a long-latency (12–23 ms) group. Waves belonging to both of these classes were still visible in both high magnesium/ no calcium Ringer's solution or Ringer's solution containing cobalt chloride and were unaffected by high calcium/no magnesium Ringer's. The average conduction velocities of the short- and long-latency groups matched the conduction velocities of, respectively, edge and convexity detectors in the adult. Activity of the glossopharyngeal nerve was recorded with bipolarsilver wire electrodes while taste stimuli were applied to thelingual surface in anesthetized mudpuppies. Taste stimuli wereinjected into a continuous stream of distilled water which wasrunning over the tongue, KCl, CaCl2 and LiCl2 at 0.4 M elicitedbrisk responses, as did HCl at 0.2 M and quinine at 6 x 10–4M. Sucrose, glucose and saccharin did not elicit responses.Twenty amino acids were surveyed for their ability to evokea response at 0.04 M: 1-arginine, 1-valine, 1-phenylalanine,1-tryptophan, 1-tyrosine, 1-glutamic acid, 1-lysine and histidinealways evoked responses, whereas other amino acids either didnot evoke responses or only occasionally evoked responses. Thesupernatants from solutions of minced worms and minnows andPurina Trout Chow were effective taste stimuli. Pre-adaptingthe tongue to Ringer's solution by running a continuous streamof Ringer's solution over it eliminated responses to quinineand decreased responses to NaCl. Pre-adapting the tongue to10–4 to 10–3 M amiloride, a potent sodium channelblocker, did not alter the responses to NaCl, LiCl, or othertaste stimuli.  相似文献

Muscle spindles were isolated from freshly removed cat lumbrical muscles in oxygenated Ringer's solution and placed in a solution containing 10 ml of 0.1% nitro blue tetrazolium and 10 ml of 0.2 M phosphate buffered sodium succinate, pH 7.6. Spindles were incubated in this solution at 37 C for 4-12 hr, returned to Ringer's for 30 min at room temperature, fixed in 10% formal-Ringer's for 30 min, and stored indefinitely in distilled water. With this technique the patterns of sensory innervation can be clearly visualized by the deposition of diformazan. The stained preparations may be mounted in glycerol and teased further for whole mount inspection or they may be embedded in Epon and serially sectioned for more detailed study.  相似文献

Cells derived from cultures of bone marrow or leucocytes were treated with hypotonic citrate solution, squashed in 45% acetic acid frozen with CO2 to allow removal of the cover glass without disturbing the smear, and stained by the following schedule: absolute alcohol, 5 min; coat with 0.2% parlodion and air dry; 70% alcohol, 5 min; distilled water, 5 min; stain 2-5 min in a mixture of 45 ml of a 0.3% solution of basic fuchsin in 5% phenol, 6 ml of glacial acetic acid, and 6 ml of 37% formaldehyde. Differentiate and dehydrate in absolute alcohol, clear in xylene and cover. The stain is durable for several weeks if slides are stored in darkness when not in use. Results resemble those obtained by Feulgen or aceto-orcein methods.  相似文献

The combined modalities of potassium arrest and local cardiac hypothermia were used for myocardial protection in 82 patients. The cardioplegic solution used was Ringer's lactate to which potassium chloride and sodium bicarbonate were added so that the final solution had a pH of 7.5 and 30 meq/liter potassium. The myocardium was cooled externally by cold Ringer's lactate at 4 °C and through coronary circulation by cold cardioplegic solution at 8 °C. The myocardial temperature was continuously monitored and kept between 12 and 18 °C. Moderate systemic hypothermia was used (26 to 30 °C). Eighty-two patients have been operated upon using this technique. Eighteen patients had single or double valve replacements, 4 had valve replacements with coronary bypass, and 60 had coronary bypass procedures. The operating conditions have been excellent and the myocardial protection offered by this technique has been good. Perioperative myocardial infarctions, as diagnosed by ECG and CPK (MB isoenzymes) and myocardial scans, were seen in 6 patients. In conclusion the combined modalities of potassium arrest and local cardiac hypothermia give excellent myocardial protection during cardiac surgery.  相似文献
