对日本引进的爱知结球白菜[Brassica campestris ssp.pekinensis‘Aichi Hakusai']的组培再生株从染色体数目、形态特征及生化特性方面与实生株进行了比较.结果表明,再生株染色体数目与实生株相同,均为2n=20;再生株在移栽初期长势不齐,1个月后,生长趋于一致,形态指标均显著高于相同条件下栽培的实生株;两者叶片可溶性糖和可溶性蛋白质含量的变化趋势相似;可溶性蛋白质含量在移栽初期高,碳氮比(C/N)小,代谢类型主要是氮素代谢;随着生长发育的进行,叶面积增加,碳水化合物代谢加强,其可溶性糖含量增加,C/N比逐渐上升.说明爱知结球白菜的组培再生株在所测定的性状上保持了实生株的遗传稳定性.  相似文献

黄背栎(Quercus pannosa)是硬叶常绿阔叶林的代表树种之一, 具有较强的克隆萌生能力, 影响其种群更新和格局特征。通过对云南药山三个不同海拔(2900 m、3100 m、3300 m)的黄背栎种群分布格局展开调查, 采用点格局分析法对其不同海拔下克隆株与实生株的格局特征及其相互关系进行了研究。结果表明, 克隆株和实生株的种群密度随海拔递减, 实生苗在低海拔地段占优势。黄背栎实生株与克隆株在高海拔地段呈集群分布, 实生株在小尺度下(0—1 m)集群分布, 随尺度的增大逐渐过渡到随机分布; 低海拔处克隆株与实生株的空间关系呈显著正相关, 高海拔小尺度下二者为正相关, 大尺度下呈负相关。  相似文献

南方高丛蓝浆果‘南月’实生后代果实特性的变异分析   总被引:4,自引:0,他引:4  
对南方高丛蓝浆果(Vaccinium corymbosum hybrids)品种‘南月’(‘Southmoon’)实生后代的单果质量、果实横径、可溶性固形物含量、单果种子数、果色、口感以及蒂痕类型等果实特征的变异进行了分析。结果表明,‘南月’实生后代的单果质量为0.64~2.15g,果实横径为10.0~16.4mm;果实可溶性固形物含量为9.1%-15.8%,其中有37%的单株果实可溶性固形物含量在12.0%以上;单果种子数为3~93粒,有18%的单株单果种子数小于25粒;果实颜色以蓝紫色和黑蓝紫色为主,比例分别为50%和42%;果实口感以中等和好2个等级为主,比例分别为50%和40%。在对果实的主要特征和经济性状进行评估的基础上,初步筛选出21个‘南月’实生后代优良单株。  相似文献

利用广西南宁冬季无霜冻的气候特点,对陆地棉(Gossypium hirsutum L.)洞A细胞核雄性不育扦插株与宿生株及其杂交一代进行比较研究.结果表明:(1)1 a生扦插株开花较早,茎粗、主茎节间长度、铃重、子指4个性状显著好于实生株,但扦插株的种子产量显著低于实生株,其原因是果枝扦插形成的僵苗占扦插株的23.04%,而且扦插株全为不育株,实生株中则有50%左右的可育株;(2)2 a生扦插株性状与实生株无显著差异;(3)由扦插繁殖的不育株和种子繁殖的不育株配制的杂交F1代的产量与纤维品质无显著差异.因此认为,扦插繁殖陆地棉核不育株用于多年杂交制种是可行的,选择营养枝扦插是陆地棉核不育株扦插繁殖需要注意的关键问题. 宿生株及其杂交一代进行比较研究.结果表明:(1)1 a生扦插株开花较早,茎粗、主茎节间长度、铃重、子指4个性状显著好于实生株,但扦插株的种子产量显著低于实生株,其原因是果枝扦插形成的僵苗占扦插株的23.04%,而且扦插株全为不育株,实生株中则有50%左右的可育株;(2)2 a生扦插株性状与实生株无显著差异;(3)由扦插繁殖的不育株和种子繁殖的不育株配制的杂交F1代的产量与纤维品质无显著差异.因此认 ,扦插繁殖陆地棉核不育株用于多年杂交制种是可行的,选择营养枝扦插是陆地棉核不育株扦插繁殖需  相似文献

从甜味成分种类及含量的不同,研究甜菊(SteviarebaudianaBertoni)实生群体和单株无性系的变化,从中选育优质甜味成分含量高的良种。主要结果:(1)在实生群体中,丰产株型(圆纺锤形株)占7.3%,其中,优质甜味成分R-A含量超过St含量的R-A型株占10.96%,它们的R-A含量变幅为3.3~12.0%。(2)R-A型良种J-2单株无性系在繁殖达2000万株时,叶片大小和含甙量均存在极显著差异,R-A含量变幅为4.5~12.2%。(3)从R-A含量为3.86%的实生群体中选出R-A含量为7.04~12.03%的单株及从R-A含量为9.10%的良种单株无性系中选出R-A含量为10.15~12.15%的单株,它们的R-A含量均大幅度提高。  相似文献

米根霉乙醇脱氢酶(ADH)突变菌株的诱变选育   总被引:4,自引:0,他引:4  
米根霉发酵生产L-乳酸过程中,由于丙酮酸在丙酮酸脱羧酶、乙醇脱氢酶(ADH)催化下生成乙醇,使得丙酮酸向乳酸转化的流量减少。采用亚硝基胍(NTG)诱变米根霉AS3.3462孢子液,诱变剂量为0.15 mg/ mL时,致死率为70%~80%。在含丙烯醇的YPD筛选培养基上筛选获得两株ADH活力降低的突变株mut-1和mut-2,检测突变株mut-1和mut-2的最大ADH活力分别为35.67和43.09U/mL,是原始菌株的41.63%和50.29%。发酵72h后,原始菌株的乙醇与乳酸浓度分别为28.9g/L和40.31g/L,而mut-1和mut-2突变株的乙醇产量分别为4.87g/L和6.56g/L,乳酸产量为54.45g/L和44.07g/L。在相同的发酵条件下,米根霉ADH突变株mut-1和mut-2对还原糖的利用速率高于出发菌株,其生物量积累亦高于出发菌株。  相似文献

对兔眼蓝浆果(Vaccinium ashei Reade)、北方高丛蓝浆果(V.corymbosum L.)和南方高丛蓝浆果(V.corymbosum hybrids)3个品种群及南方高丛蓝浆果实生系的花冠长度、花冠宽度、花冠长宽比和花冠口径大小等特征进行了测定和比较,并研究了花冠形态特征参数与果实横径的相关性。研究结果表明,蓝浆果不同品种群间和同一品种群内不同品种间的花冠形态特征参数差异较大;依据花冠形态特征参数,可将供试的兔眼蓝浆果和南方高丛蓝浆果各品种及南方高丛蓝浆果各实生系初步划分为4种类型,将供试的北方高丛蓝浆果各品种划分为3种类型。此外,只有南方高丛蓝浆果各品种及其实生系的果实横径与花冠宽度相关性显著(P〈0.05),相关系数分别为0.639和0.324。  相似文献

Antibiotic therapy is often used with mechanical therapy to treat periodontal disease. However, complications associated with antibiotic use can occur. A ‘bacteria-specific’ targeted approach would eliminate some of these complications and kill specific periodontopathogens without harming the commensal bacteria. One such approach is to couple antimicrobial peptides to a ligand, pheromone, or antibody specific for the periodontopathogen, Porphyromonas gingivalis. To assess the feasibility of this approach, we attached PQGPPQ, a peptide from proline-rich protein 1 to either the N-terminus of SMAP28 (peptide ZS37-37) or the C-terminus of SMAP28 (peptide ZS37-38) to see whether it has potential as a carrier ligand to deliver SMAP28 to the surface of P. gingivalis. For Escherichia coli and Aggregatibacter actinomycetemcomitans, the median minimal inhibitory concentration (MIC) of ZS37-37 was higher than the median of SMAP28 alone, although the median MIC of ZS37-38 was lower than that of SMAP28 alone. For P. gingivalis, there was no difference in the median MIC values. For S. aureus, the median MIC was higher for ZS37-37 and ZS37-38 compared to SMAP28 alone, particularly for ZS37-38. For Fusobacterium nucleatum, the median MIC values were equal for ZS37-37 and ZS37-38 and higher than the median MIC for SMAP28 alone. Attaching PQGPPQ to SMAP28 did not greatly increase the antimicrobial activity of ZS37-37 or ZS37-38 for P. gingivalis nor substantially decrease the antimicrobial activity of ZS37-37 or ZS37-38 for the four other microorganisms tested. This is an initial step to develop a selective antimicrobial agent that has ‘targeted’ antimicrobial activity without adverse reactions often associated with the use of broad-spectrum antibiotics.  相似文献

Summary The variability of 13C values was measured in leaf, stem and root tissues of several tree species growing in closed natural and plantation forests in the Luquillo mountains of Puerto Rico. Results confirm a significant decrease of 13C values from the tree canopy to the forest floor. The values measured in understory plants growing in gaps were not significantly different from the average for plants growing under the forest shade. Seedling leaf values tended to be more positive than those of saplings, probably reflecting the contribution of organic matter from the mother tree. Photosynthetic independence on the forest floor results in a reduction in °13C value. Stem and root tissue values of seedlings and saplings were less negative than those of the leaves of the same plants. It is suggested that this difference results from the slower change in isotopic composition experienced by the woody tissue, as the seedlings become photosynthetically independent in the forest floor.  相似文献

Zellweger syndrome (ZS) is a consequence of a peroxisome biogenesis disorder (PBD) caused by the presence of a pathogenic mutation in one of the 13 genes from the PEX family. ZS is a severe multisystem condition characterized by neonatal appearance of symptoms and a shorter life. Here, we report a case of ZS with a mild phenotype, due to a novel PEX6 gene mutation. The patient presented subtle craniofacial dysmorphic features and slightly slower psychomotor development. At the age of 2 years, he was diagnosed with adrenal insufficiency, hypoacusis, and general deterioration. Magnetic resonance imaging showed a symmetrical hyperintense signal in the frontal and parietal white matter. Biochemical tests showed elevated liver transaminases, elevated serum very long chain fatty acids, and phytanic acid. After the death of the child at the age of 6 years, molecular diagnostics were continued in order to provide genetic counseling for his parents. Next generation sequencing (NGS) analysis with the TruSight One? Sequencing Panel revealed a novel homozygous PEX6 p.Ala94Pro mutation. In silico prediction of variant severity suggested its possible benign effect. To conclude, in the milder phenotypes, adrenal insufficiency, hypoacusis, and leukodystrophy together seem to be pathognomonic for ZS.  相似文献

A novel bacterium, strain 1ZS3-15T, was isolated from rhizosphere of rice. Its taxonomic position was investigated using a polyphasic approach. The novel strain was observed to be Gram-stain positive, spore-forming, aerobic, motile and rod-shaped. Phylogenetic analysis based on 16S rRNA gene sequences showed that strain 1ZS3-15T was recovered within the genus Paenibacillus. It is closely related to Paenibacillus pectinilyticus KCTC 13222T (97.9 % similarity), Paenibacillus frigoriresistens CCTCC AB 2011150T (96.8 %), Paenibacillus alginolyticus JCM 9068T (96.4 %) and Paenibacillus chondroitinus DSM 5051T (95.5 %). The fatty acid profile of strain 1ZS3-15T, which showed a predominance of anteiso-C15:0 and iso-C16:0, supported the allocation of the strain into the genus Paenibacillus. The predominant menaquinone was found to be MK-7. The polar lipids profile of strain 1ZS3-15T was found to consist of diphosphatidylglycerol, phosphatidylglycerol, phosphatidylethanolamine, one unidentified lipid and two unidentified aminophospholipids. The cell wall peptidoglycan contains meso-diaminopimelic acid. Based on draft genome sequences, the DNA–DNA relatedness between strain 1ZS3-15T and the closely related species P. pectinilyticus KCTC 13222T are 24.2 ± 1.0 %, and the Average Nucleotide Identity values between the strains are 78.9 ± 0.1 %, which demonstrated that this isolate represents a new species in the genus Paenibacillus. The DNA G+C content was determined to be 45.3 mol%, which is within the range reported for Paenibacillus species. Characterisation by genotypic, chemotaxonomic and phenotypic analysis indicated that strain 1ZS3-15T represents a novel species of the genus Paenibacillus, for which the name Paenibacillus oryzisoli sp. nov. is proposed. For 4-month-old Douglas-fir [Pseudotsuga menziesii (Mirb.) Franco] seedlings under 17-h photoperiods in controlled environment conditions, flurprimidol (-(1-methlyethyl--[4-(trifluoromethoxy)phenyl]-5-pyrimidine-methanol) decreased incidence of second flushing from 81 to 54% and significantly reduced height growth in the second year following low-temperature treatment, in comparison to untreated controls. For seedlings under 15-h photoperiods, flurprimidol had only limited early effects, but after a period of bud dormancy, shoot growth was reduced to one-third that of the untreated controls. Under both photoperiods flurprimidol significantly depressed abscisic acid levels and gibberellin-like bioactivity. Summary We describe the cytogenetic findings in a recurrent neurofibrosarcoma in a patient with nonfamilial von Recklinghausen disease. The composite karyotype was: 40,Y,-X,+dic r(X;20)(:Xp22.2q26::20p13 q13:), -1, +der(1)t(1;3) (p21;p24),-3,-4,-5,+der(5) t(5;?)(q31;?),-9,-9,+der(9)t(3;9)(q21 or q13;p24 or p22), -11,+der(11)t(11;?)(q22.2;?), -17,+der(17)t(17; 22;?)(q21;q13.1;?), -20, -21, -22, -22, +der(22)t(17; 22;?)(q21;q13.1;?),t(2;10)(q37;q22). The derivative chromosomes were demonstrated at the 500 band level. Chromosomes 17 and 22 were shown to be involved in an unbalanced three-way translocation: t(17;22;?)(q21;q13.1;?). This event was confirmed by in situ hybridization, using two probes mapped to chromosome 17. Hill H is a probe derived from the novel oncogene TRE and is located at 17q12–22. The second probe, derived from the granulocyte colony-stimulating factor (G-CSF), is located at 17q11–q21. The rearrangement between chromosomes 17 and 22 showed breakpoints similar or close to the gene loci for neurofibromatosis 1 (NF-1) and NF-2. Based on our observations we recommend that genetic studies on NF-1 tumors include both gene sites (NF-1 and NF-2) rather than focus on one gene locus.  相似文献

A tigloyl-CoA: 13-hydroxylupanine O-tigloyl-transferase could be demonstrated in crude enzyme preparations from Lupinus albus seedlings. The enzyme activity increases concomitantly with for formation of 13-tigloyloxylupanine in developing lupin seedlings. The transferase catalyzes specifically the transfer of an acyl group to 13-hydroxylupanine. The apparent Km-values are 140 M for tigloyl-CoA and 18 M for 13-hydroxylupanine. Other hydroxylated compounds, e.g., lupinine, 4-hydroxylupanine, and cholesterol are not acylated. The transferase shows optimal activity at pH 7–8 and at 30°C. It is activated by dithioerythritol and inhibited by thiol reagents. Tigloyl-CoA can be replaced as acyl donor by benzoyl-CoA and to a lesser extent by valeroyl-CoA, 3-methylbutyryl-CoA, butyryl CoA and propionyl-CoA but not acetyl-CoA. Preliminary evidence indicates that the transfer of the tigloyl and benzoyl moieties is catalyzed by different enzymes. To investigate how seed reserves affect early seedling performance, we conducted a factorial greenhouse experiment using Lithocarpus densiflora (Tanoak). Seedlings were grown from large (5.8±0.7 g) and small (3.2±0.4 g) seeds and, following shoot emergence, seeds were either removed or left attached. Seedlings were harvested for quantification of biomass and 13C at seven time periods following seed removal (2, 4, 8, 16, 32, 64, 128 days) and seedling photosynthesis was measured three separate time periods (2–4, 49–82, 95–128 days after seed removal). Biomass increased for all seedlings, but the increase was significantly larger for seedlings with attached seeds than with removed seeds. Seed removal just after shoot emergence significantly decreased seedling biomass, but seed removal 64 days after shoot emergence had no effect on seedling biomass. Seedling photosynthesis per unit leaf area varied by time and seed presence, but not by seed size. At the first period, seedlings with attached seeds had significantly higher photosynthetic rates than seedlings with removed seeds, at the second period there was no effect of seed removal, and at the third time period seedlings with attached seeds had significantly lower photosynthetic rates than seedlings with removed seeds. Despite temporal variation in photosynthesis per unit leaf area, seedlings with attached seeds always had significantly greater leaf area than seedlings with removed seeds, resulting in significantly higher total plant photosynthesis at all three time periods. The 13C values of both the leaves and roots were more similar to that of the seed for seedlings with attached seeds than for seedlings with removed seeds, however, seed removal and seed size strongly affected root 13C. This study demonstrates that seed reserves have important effects on the early growth, physiology, and 13C of L. densiflora seedlings.  相似文献

A new psychrotolerant methanogenic archaeon strain ZS was isolated from anoxic lake sediments (Switzerland). The cells of the organism were non-motile cocci, 1.5-3.5 microm in diameter. The cells aggregated and formed pseudoparenchyma. The cell wall was Gram-positive. The organism utilized methanol, mono-, di-, trimethylamine and H2/CO2 with methane production. The temperature range for growth was 1-35 degrees C with an optimum at 25 degrees C. The DNA G+C content of the organism was 43.4. mol%. Analysis of the 16S rRNA gene sequence showed that strain ZS was phylogenetically closely related to members of the genus Methanosarcina, but clearly differed from all described species of this genus (95.6-97.6% of sequence similarity). The level of DNA-DNA hybridization of strain ZS with Methanosarcina barkeri and Methanosarcina mazei was 15 and 31%, respectively. Based on the results of physiological and phylogenetic studies strain ZS can be assigned to a new species of the genus Methanasarcina. The name Methanosarcina lacustris sp. nov. is proposed. The type strain is ZS (= DSM 13486T, VKM B-2268).  相似文献

The excessive expression of catalase protein and its activity in cultured skin fibroblast from Zellweger Syndrome (ZS), a disorder of peroxisomal biogenesis, was found to be regulated at the translational level (J. Neurochem. 67: 2373-2378, 1996). Overall there is a considerable increase in the association of catalase mRNA with polysomes in ZS cell lines as compared to control indicating translational upregulation. To investigate the possibility that RNA-protein interactions are involved in the mediation of this increase in translation, the interaction between 3 untranslated region of human catalase mRNA and human fibroblast cytoplasmic proteins were investigated by RNA gel shift assay technique. Competition experiments demonstrated that all the 600 bases of 3 UTR (of human catalase gene) was required for efficient binding. Catalase RNA- protein interaction was sensitive to the altered redox state in these in vitro assays and this RNA-protein interaction could be enhanced by the addition of -mercaptoethanol in cytoplasm from control fibroblast but not in cytoplasm from ZS fibroblast. UV cross linked RNA-protein complexes on SDS polyacrylamide gel electrophoresis revealed the presence of at least four protein bands with approximate molecular masses of 38 kDa, 50 kDa, 66 kDa and 80 kDa. The potential role of these mRNA binding proteins in the regulation of catalase gene expression is discussed.  相似文献
