This study was conducted to determine the effect of tamarind seed husk (TSH) as a source of tannin on various parameters of rumen fermentation in vitro. The TSH contained 14% tannin (DM basis). The biological interference of TSH tannin on rumen fermentation was assessed using polyethylene glycol (PEG) 6000 as an indicator. Three compound feed mixtures (CFM) were prepared either without TSH (CFM-I), with 2.5% TSH (CFM-II) and with 7.5% TSH (CFM-III). Parameters studied were in vitro gas production with PEG, rate of substrate degradation, and microbial protein synthesis. Addition of PEG to TSH resulted in an increase in gas production from 5.5 to 16.5 ml per 200 mg DM. Presence of TSH tannin depressed cumulative gas production by 16.8% in CFM-II, and by 29.2% in CFM-III during initial stages of fermentation (i.e. at 8 h). Rate of substrate disappearance (T1/2) was 14.4, 17.6 and 20.5 h in CFM-I, CFM-II and CFM-III, respectively. Irrespective of the carbohydrate source, presence of TSH tannin improved the efficiency of microbial protein synthesis in vitro. Thus, TSH is a natural source of tannin that can be used to beneficially manipulate rumen fermentation.  相似文献

This study investigated effects of dietary forage particle size (PS) and concentrate level (CL) on fermentation profiles of particle-associated rumen liquid (PARL) and free rumen liquid (FRL), in vitro degradation characteristics and concentration of bacterial mass attached to the solid or fluid rumen digesta phase in dairy cows. The experiment was a 4 × 4 Latin square design with four late-lactation dairy cows in four 23 day periods. Cows were restrictively fed (17 kg dry matter (DM)/d) one of four diets varying in the theoretical PS (6 and 30 mm) of grass hay and in the levels (approximately 200 and 550 g/kg, DM basis) of a cereal-based concentrate. Proportion of large particles (>6 mm) and the content of structural fibre in the diet increased by reducing dietary CL and, particularly, by increasing hay PS. This effect was not reflected by changes in mean total volatile fatty acid concentration or pH in the rumen. However, cows fed high concentrate diets had pH of 5.28 and 5.37 in PARL at 3 h after the last meal, when fine or long chopped hay was offered. The low pH may indicate a depression of the capacity of PARL to degrade fibre in vitro. Gas production in vitro of concentrate increased with the high concentrate diet at 12 h, suggesting that amylolytic capacity was affected only in early phases of fermentation. In addition, elevating dietary CL appeared to shift ruminal fermentation outputs from propionate to butyrate and valerate. Inclusion of coarsely chopped hay to a high concentrate diet does not appear to bring advantages due to increased structure in restrictively fed dairy cows. In addition, results suggest that the response of pH in PARL is more sensitive to dietary changes (i.e., forage PS and CL) than the response in FRL, and so PARL might be better to evaluate the risk of ruminal disfunction in dairy cows.  相似文献

An in vitro study was conducted to examine the effect of adding monensin, fish oil, or their combination on rumen fermentation and conjugated linoleic acid (CLA) production by mixed ruminal bacteria when incubated with safflower oil. Concentrate (1 g/100 ml) with safflower oil (0.2 g/100 ml) was added to a mixed solution (600 ml) of strained rumen fluid and buffer (control). Monensin (10 ppm), fish oil (0.02 g/100 ml), or monensin plus fish oil was also added into control mixture. All the culture solutions prepared were incubated anaerobically at 39 °C for 12 h. A higher pH and ammonia concentration were observed from the culture solution containing monensin at 12 h of incubation than those from the control or the culture containing fish oil. Monensin increased (P < 0.007) the C3 content over all the collection times of culture solution while reducing the C4 content at 6 h (P < 0.018) and 12 h (P < 0.001) of incubations. Supplementation of monensin, fish oil or their combination changed the content of C18-fatty acids of ruminal culture. Monensin alone reduced (P < 0.021) the content of cis-9, trans-11 CLA compared to fish oil at all sampling times, but increased (P < 0.041) the trans-10, cis-12 CLA production compared to fish oil addition and the control which were similar at incubation for 12 h. The combination of monensin and fish oil increased the content of cis-9, trans-11 CLA (P < 0.023) and transvaccenic acid (TVA, P < 0.018) significantly compared to the control or monensin alone at incubation for 12 h.  相似文献

The influence of insulin (17.4 nmol l-1) on total gas and methane production, the concentration of total and individual fatty acids and dry matter degradability was investigated in the rumen ciliate culture of Entodinium caudatum. The experimental groups consisted of control group (without insulin) and two groups with insulin application--single shot and long-term application (over 30 days). Fermentation activity of each experimental group was observed on two subgroups: whole protozoan culture (protozoa plus bacteria) and bacterial fraction (bacteria without protozoa). Long-term application of insulin significantly increased methane production and DM degradability in the whole protozoan culture. Total VFA concentration was significantly increased by long-term as well as single-dose application of insulin (by 255% and 158%, respectively). The growth of the protozoa was not influenced by insulin treatments. It can be concluded that the fermentation activity of the community of the rumen ciliate Entodinium caudatum culture was marked stimulated by application of insulin.  相似文献

目的 研究三种模拟结肠发酵培养基和粪菌固定化对体外结肠发酵体系菌群构成的影响,为建立高度模拟结肠体外结肠发酵提供参考。方法 采集健康女性成人粪便,粪便制备悬液或用结冷胶‒黄原胶固定化粪菌接种于模拟结肠发酵反应器,分别用三种培养基按0.07 mL/h稀释率进行连续发酵10 d,用末端限制性片段长度多态性(T-RFLP)与高通量测序技术分析菌群多样性,并分析发酵液短链脂肪酸含量。结果 T-RFLP分析显示,培养基类型主要影响粪菌固定体系菌群α多样性达到稳定的时间和多样性。β多样性分析显示,第10天时培养基III悬液培养和培养基I固定化培养系统的菌群构成与粪便菌群最相近。而培养基III在第10天达到较高而稳定的丁酸浓度。  相似文献

The potential of natural surfactant tea saponin to enhance uptake of polychlorinated biphenyls (PCBs) and cadmium (Cd) by Zea Mays L. and Saccharum officinarum L. was investigated. With addition of tea saponin at 0.01% in solution culture, the concentrations of PCB 14, PCB 18, PCB 77 and PCB 156 in root of corn seedling were 2.72, 2.68, 1.94 and 2.40 times as those of treatments without adding any surfactant, respectively. Application of tea saponin to the soil significantly elevated PCB 5 accumulation in shoots and roots (p < 0.05) by sugarcanes. With addition of 0.3% tea saponin, Cd concentration was increased by 96.9% in roots, 156.8% in stems and 30.1% in leaves compared with the treatment without addition of surfactant in sugarcane grown in soil. Tea saponin had potential of assisting the uptake of PCBs and Cd by plants from water solution and soil.  相似文献

Batch cultures of mixed rumen micro-organisms were used to study the effects of three fibrolytic enzymes (xylanase from Trichoderma viride (XYL) and fibrolytic enzymes from Aspergillus niger (ASP) and Trichoderma longibrachiatum (TR)) on the fermentation of three substrates composed of grass hay:concentrate in the proportions (dry matter (DM) basis) of 0.7:0.3 (HF), 0.5:0.5 (MF) and 0.3:0.7 (LF). Enzymes were characterized for xylanase, endoglucanase, exoglucanase and amylase activities, and were supplied at rates of 40 and 80 enzymatic units/g substrate DM. In 8 h incubations, all enzymes increased (P=0.048 to P<0.001) the true degradability of substrate DM and the production of acetate, propionate, total volatile fatty acids (VFA) and gas. After 24 h incubation, some of the observed effects disappeared, but all enzymes still increased (P=0.028 to P<0.001) the degradability of substrate acid detergent fibre and the production of acetate, propionate and total VFA. For all enzymes, the effects on ruminal variables were less marked at 24 than at 8 h of incubation. Only few significant (P=0.044 to P=0.001) enzyme × substrate interactions were detected, although the magnitude of the response for each substrate varied with the enzyme. When considering the amount of organic matter apparently fermented (OMAF) and the methane:OMAF ratio as main variables, TR80 produced the greatest increase in OMAF (17.0%) for HF substrate, with ASP80 and TR40 having similar values (11.1 and 12.6%), and XYL and ASP40 showing no effects (P>0.05). A decrease (P<0.05) of methane:OMAF ratio was only found for TR80 at 8 h (17.4%). All enzymes, with the exception of ASP40, increased (P<0.05) OMAF at 8 h for MF substrate (11.3–25.4%), TR80 showing the greatest response. After 24 h of incubation, both doses of XYL and TR increased (P<0.05) OMAF (mean value 8.2%) and decreased methane:OMAF ratio (mean value 9.5%). All enzymes increased significantly OMAF with LF substrate at 8 h (7.5–19.9%), but after 24 h no effect (P>0.05) was detected on OMAF and methane:OMAF ratio. In general, few differences were detected between both doses of enzymes, which indicate than the used enzymes would be effective in enhancing ruminal degradation of substrates at a dose lower than 80 enzymatic units/g substrate DM.  相似文献

Effects of proportions of neutral detergent fibre (aNDFom) and starch, as well as their degradation rates, on rumen fermentation were tested using an in vitro rumen simulation system (SIMCO). The in vitro system was designed to simulate selective particle retention and had an average fluid volume of 1150 ml with a liquid dilution rate of approximately 0.07 h−1. Two types of hay (aNDFom sources) and two types of starch were each included at two different levels in the diet and were examined in an experiment following a 2×2×2 factorial arrangement of treatments (eight diet combinations). The hay was either late-cut timothy (Phleum pretense L.) or early cut meadow grass (Poa pratensis L.), with ruminal in situ aNDFom digestion rates of 0.03–0.04 and 0.07–0.08 h−1, respectively. The two starch types were raw (R) and cooked (C) potato starch with previously determined in vitro ruminal digestion rates of 0.04 and 0.20 h−1, respectively. The starch levels were 300 and 600 g/kg diet dry matter (DM) with the remaining being hay (282–682 g/kg DM) and peptone (14–111 g/kg DM). The aNDFom level varied among the diets with different starch levels and hay types. The peptone acted as a source of peptides and, together with ammonia salts from buffer, was used to balance the N contents of the diets. The feeding level for each of the eight vessels was 28 g DM/d. Two 10-day simulations were made with the system. The average pH was higher (P<0.05) for all treatments with raw potato starch (6.19) versus cooked starch (6.07). Protozoa scores, on a qualitative scale, declined faster at the higher starch level. The aNDFom digestibility was, as expected, higher (P<0.001) for meadow hay (0.57) than timothy (0.32), and was also higher (P<0.001) at the lower starch level (0.54) versus the higher (0.35). Microbial protein production efficiency (mg microbial N/g organic matter truly digested) was higher for the faster degrading aNDFom (P<0.01) and starch (P<0.05) sources, but was not affected by starch level. Cooked starch resulted in a lower acetate proportion (449 mmol/mol versus 591 mmol/mol VFA; P<0.001) but higher proportions of propionate (297 mmol/mol versus 236 mmol/mol VFA; P<0.001), and butyrate (169 mmol/mol versus 127 mmol/mol VFA; P<0.01). Butyrate increased with starch level (127 mmol/mol versus 169 mmol/mol VFA; P<0.01), and was also higher for meadow hay versus timothy (168 mmol/mol versus 128 mmol/mol VFA; P<0.01). Interactions between the treatments demonstrate that the response in VFA pattern to starch level is dependent on starch and aNDFom sources. Substrates such as starch and aNDFom are fermented differently depending on their rates of ruminal degradation.  相似文献

The present study was to evaluate effect of herbal feed additives on methane and total gas production during the rumen fermentation for environment and animal health concern. Different parts of the five medicinal plants were selected such as leaf and small stems of Ocimum sanctum (Tulsi), roots of Curcuma longa (Haldi), fruits of Emblica officinalis (Amla), leaves of Azadirachta indica (Neem) and leaves and small stem of Clerodendrum phlomidis (Arni) for our study. Addition of different herbal additive combinations did not influence IVDMD and total gas production however methane production (mg/g of substrate DM) was significantly (P<0.05) reduced in Amla: Neem and Neem: Arni combinations. Total nitrogen significantly (P<0.01) increased in the combinations of Tulsi: Haldi and Amla: Neem. TCA–ppt-N is significantly (P<0.01) increased in Tulsi: Haldi, Haldi: Amla, Amla: Neem and Neem: Arni however NH3-N (mg/dl) significantly decreased in all treatments. We conclude that the screening of plant combinations, Amla: Neem and Neem: Arni have potential to decrease methane production and our herbal feed supplements have no side-effects on the ruminant in small amount.  相似文献

采用注射、熏蒸、点滴、饲喂、内吸等方法测定了茶皂素对菜粉蝶Pieris rapae L.5龄幼虫的拒食活性。结果表明,茶皂素对5龄菜青虫具有很强的饲喂拒食活性,24小时选择性拒食中浓度AFC_(50)为40.88mg/L,非选择性拒食中浓度为321.92mg/L;并具有一定的内吸拒食作用,尤其是根部施药法内吸拒食活性较明显。茶皂素对菜青虫无显著的注射、熏蒸、点滴拒食作用。  相似文献

Stirred, pH controlled batch cultures were carried out with faecal inocula and various chitosans to investigate the fermentation of chitosan derivatives by the human gut flora. Changes in bacterial levels and short chain fatty acids were measured over time. Low, medium and high molecular weight chitosan caused a decrease in bacteroides, bifidobacteria, clostridia and lactobacilli. A similar pattern was seen with chitosan oligosaccharide (COS). Butyrate levels also decreased. A three-stage fermentation model of the human colon was used for investigation of the metabolism of COS. In a region representing the proximal colon, clostridia decreased while lactobacilli increased. In the region representing the transverse colon, bacteroides and clostridia increased. Distally a small increase in bacteroides occurred. Butyrate levels increased. Under the highly competitive conditions of the human colon, many members of the microflora are unable to compete for chitosans of low, medium or high molecular weight. COS were more easily utilised and when added to an in vitro colonic model led to increased production of butyrate, but some populations of potentially detrimental bacteria also increased.  相似文献

Aims: To determine the in‐vitro effect and mode of action of tea saponin on the rumen microbial community and methane production. Methods and Results: Saponin extracted from tea seeds was added to (1) an in‐vitro fermentation inoculated with rumen fluid and (2) a pure culture of Methanobrevibacter ruminantium. Methane production and expression of the methyl coenzyme‐M reductase subunit A (mcrA) were monitored in both cultures. Abundance of methanogens, protozoa, rumen fungi and cellulolytic bacteria were quantified using real‐time PCR, and bacterial diversity was observed using denaturing gradient gel electrophoresis. Addition of tea saponin significantly reduced methane production and mcrA gene expression in the ruminal fermentation but not with the pure culture of M. ruminantium. The abundance of protozoa and fungi were significantly decreased 50% and 79% respectively but methanogen numbers were not affected, and Fibrobacter succinogenes increased by 41%. Bacterial diversity was similar in cultures with or without tea saponin. Conclusions: Tea saponin appeared to reduce methane production by inhibiting protozoa and presumably lowering methanogenic activity of protozoal‐associated methanogens. Significance and Impact of the Study: Tea saponin may be useful as a supplement to indirectly inhibit methane production in ruminants without a deleterious effect on rumen function.  相似文献

This experiment was designed to study effects of exogenous fibrolytic enzymes on digestibility and ruminal fermentation of total mixed rations (TMR) with different forage:concentrate (F:C) ratios. The experimental design was a 6×6 Latin square with six ruminally cannulated Ramboulliet lambs (58 ± 7.4 kg body weight) used in a 3×2 factorial arrangement of treatments using three forage:concentrate (F:C) ratios (400:600, 500:500, 600:400 kg/kg) and two (0 or 2 g) levels of fibrolytic enzymes/kg TMR dry matter (DM). Exogenous fibrolytic enzymes increased the soluble fraction of DM (P<0.01) and ruminal in situ disappearance rate of DM and neutral detergent fibre (aNDF; P<0.001). However, there were no differences among treatments in DM and aNDF in vivo digestibility due to enzyme addition to the diet. Feed intake and N balance were also not affected by enzymes, and ruminal fermentation patterns were unchanged. Exogenous fibrolytic enzymes improved ruminal disappearance rates of DM and aNDF, but had no impact on lamb performance or ruminal fermentation.  相似文献

This study was conducted to investigate the impact of dietary inclusion of Moringa oleifera leaf meal (MLM) as a substitution for soybean meal on nutrient digestibility, rumen fermentation, rumen enzyme activity, blood metabolites, growth-related hormones, and growth performance of buffalo calves. Thirty buffalo calves eight to nine months of age with an average body weight of approximately 153.7 ± 0.97 kg were randomly distributed through three dietary treatments (ten calves/treatment). MLM inclusion rates were 15% (M15) and 20% (M20), replacing soybean meal by 50 and 75% in the concentrate mixture, respectively. The results indicated that, digestibility of dry matter, organic matter (OM), and crude fiber (CF) increased significantly (p < 0.05) with MLM inclusion, while the digestibility of crude protein (CP) and ether extract (EE) reduced significantly (p < 0.05) with MLM addition. Dietary supplementation with MLM significantly affected (p < 0.001) rumen fermentation by reducing ruminal enzymes, ruminal ammonia-N, total protozoa, and acetate/propionate ratio and increasing acetic, propionic, and butyric acids and total volatile fatty acid concentrations (p < 0.001). Furthermore, dietary inclusion of 15% MLM significantly improved (p < 0.001) final body weight, dry matter intake of feed, daily weight gain, feed conversion efficiency, blood metabolites, and plasma insulin growth factor-I (IGF-I). It can be concluded that MLM is a multi-purpose protein supplement that provides some nutritional and therapeutic advantages when replacing 50% of soybean meal. Dietary supplementation of 15% MLM improved rumen fermentation, growth performance, blood metabolites, plasma IGF-I and mitigated ammonia and methane without any adverse effects in growing buffalo calves.  相似文献

对粗茶皂素的提取纯化工艺进行了研究。以质量分数为70%的粗茶皂素为原料,经2%NaOH溶解、盐酸酸析、95%乙醇溶解、丙酮沉析工艺得到精制茶皂素。检测结果表明,茶皂素质量分数大于95%,提取率大于80%,这种简便易行的工艺得到的茶皂素可作为开发新型植物灭螺剂的原料。  相似文献

油茶茶枯是南方地区一种常见的加工副产物,含有较高的茶皂素。茶皂素是工业生产中应用广泛的一种表面活性剂,性能优良,但目前茶皂素的提取工艺并不成熟。为了优化油茶茶枯中茶皂素提取的最佳工艺,提高油茶茶枯的利用率,该文以脱脂油茶茶枯粉末为原料,乙醇溶液作为浸提剂,以茶皂素的得率和含量作为提取效果的评价指标,通过单因子实验和正交试验探求油茶茶枯中茶皂素的最佳提取条件。结果表明:以提取时间、3%NaOH添加量、乙醇浓度、料液比、温度和提取次数6个因素作为单因子,对油茶茶枯中茶皂素提取得率和含量进行研究,在单因子实验的基础上进行正交试验得到茶皂素提取的最佳工艺条件为料液比1∶17,乙醇浓度75%,提取温度81℃,提取时间88min,3%NaOH添加量2.5mL(即每10g茶枯粉末中添加0.075g NaOH),提取次数2次。通过论证该结果表明,在最佳的提取条件下,提取得到茶皂素的最高含量为0.54mg·mL-1,得率为21.35%。该实验方法添加了NaOH后,在最佳条件下茶皂素的得率明显提高,对今后优化油茶茶枯中茶皂素的提取工艺具有很好的借鉴意义。  相似文献
