Two hundred one American toads (Bufo americanus) from northern Michigan were examined for blood trypanosomes. Three species, Trypanosoma bufophlebotomi, T. schmidti-like sp. and T. pseudopodia, had prevalences of 27, 16 and 1%, respectively. Cross experimental inoculations showed that T. bufophlebotomi from toads is not the same as T. ranarum found in frogs of the family Ranidae of this region.  相似文献

Aspects of visual optics were investigated in the American toad (Bufo americanus). The development of the refractive state of the eye during metamorphosis was followed with IR photoretinoscopy. Frozen sections documented the changes in optical parameters before and after metamorphosis. There is a difference in light sensitivity between juvenile and adult toads. Binocular accommodation in adult toads was observed. 1. IR photoretinoscopic measurements showed that the refractive state of the eye changed very rapidly during metamorphosis, about 10 D/h while the animal entered the terrestrial habitat. 2. Frozen sections showed that the almost spherical lens in a tadpole eye had flattened in a just metamorphosed toad's eye while at the same time the distance of the lens to the retina had decreased. However, the morphological measurements were not sufficiently sensitive to record the relatively small changes in ocular dimensions that were responsible for the rapid changes in refractive state during metamorphosis. 3. Schematic eyes, with homogeneous and non homogeneous lenses, were constructed for tadpoles, juvenile toads, and adult toads. 4. Nonparaxial raytracing studies in schematic eyes suggested that the lenses of animals of the three developmental stages tadpole, juvenile toad, and adult are not homogeneous but have a refractive index gradient. The raytracing studies indicated that the refractive index gradient is different for the different developmental stages, being highest in the tadpole lens. 5. The observations of toads during feeding behavior at different light levels showed an increased light sensitivity in the adult nocturnal toads in contrast to the juvenile animals, which are diurnal. The increased light sensitivity could partly be explained with an increase in aperture and an increase in red rod outer segments. To fully explain the higher light sensitivity in adult toads, changes in neuronal parameters had to be assumed. 6. Retinoscopic measurements of the resting refractive state in the adult toad showed a hyperopic defocus of about +8 D. By subtracting the measurement artefact for retinoscopy, the true resting focus was found to be nearly emmetropic. 7. The amount of natural accommodation in adult toads during normal feeding behavior was investigated with IR photoretinoscopy. Binocular accommodation of about 8 D was observed.  相似文献

Very little is known about the effects of manipulating toad sperm activity in vitro, and such information is important in the development of a genetic resource bank for bufonid species. The specific objectives of this study were to: 1). identify the optimal inactivation and reactivation solutions for toad spermatozoa collected in urine; 2). establish the length of time toad spermatozoa can be exposed to an inactivation buffer and still resume motility upon reactivation; 3). evaluate the consequence of inactivation on specific sperm characteristics; and 4). characterize the sperm mitochondria vesicle (MV) and its relationship to motility. Reactivated sperm motility was similar after inactivation in either Simplified Amphibian Ringers (SAR) solution or DeBoer's (DB) solution. Diluting the buffer by 80% with water provided the best method for reactivating sperm. Dilutions with NaCl solutions (10-50 mM) produced inferior results. SAR-inactivated spermatozoa could remain suspended up to 4 hr and still regain 25% of initial motility upon reactivation in water. Compared to the controls, sperm motility was greater (P<0.01) over time for samples treated with SAR, although forward progression was significantly lower. Furthermore, SAR treatment resulted in sperm samples with a greater number of viable, morphologically normal, and intact MVs over time. Electron microscopy and fluorescent staining confirmed that the toad sperm's MV contains a large number of active mitochondria with very few other cytoplasmic structures. Nearly all spermatozoa exhibiting motility had an intact MV, and dissociation of this structure was clearly related to motility loss. In conclusion, toad spermatozoa can be effectively inactivated and reactivated by varying the osmolality of the external solutions and, although sperm forward progression is reduced, all other characteristics are well maintained. Moreover, the increased number of spermatozoa with intact MV after inactivation suggests the process may help preserve this important structure.  相似文献

Summary Single unit recordings in the posterior nerve branchlet from the saccule have shown that, in the American toad (Bufo americanus), approximately 30% of the fibers respond to airborne sounds in a way similar to fibers from the two known auditory organs, the amphibian and basilar papillae. In response to tones, saccule fibers have best excitatory frequencies which fall into two disjoint populations: units in the low-frequency-sensitive group (below 300 Hz) show tone-on-tone suppression while those in the high-frequency-sensitive group (700–1,200 Hz) show no evidence of peripheral inhibition. Saccule units have somewhat higher thresholds than those from the other auditory organs. It is suggested that the high-frequency-sensitive fibers might be useful for discriminating mating calls in an intense chorus while the low-frequency-sensitive units likely respond to other high intensity sounds in the environment.Research supported by the U.S. Public Health Service (NIH grant NS-09244).  相似文献

Mating behaviour of B. americanus was observed from 1985 to 1987. The population contained 38–45 males and 11–26 females, depending on the year. The breeding season of this ‘explosive breeder’ usually encompassed less than 48 h. Male reproductive success varied from zero to two matings per season and zero to an estimated 15 126 zygotes per season. All females mated once per season and variation in their zygote production was estimated to be 4017–11 624 zygotes per season. Body length explained 76% of the variation in zygote production of females. However, male body length was only weakly correlated with mating success in two of three seasons, and with zygote production in one of three seasons. Male arm length was predicted to correlate with male mating success because longer arms should facilitate remaining clasped to females when challenged by rival males. However, mating males did not differ from non-mating males in arm length, and the relationship between arm length and body length was the same for the sexes. Various male behaviours were measured using focal-animal sampling but only call rate correlated with male mating success. The pattern of size dimorphism (females larger than males) is consistent with the observed sex-specific relationships between reproductive success and body size.  相似文献

Temperature and humidity are dominant environmental variables affecting performance of nocturnal, terrestrial amphibians. Toads are frequently active at body temperatures (T(b)) and hydration states (HS) that yield suboptimal performance. We investigated the combined effects of T(b) and HS on feeding, locomotion, and metabolism of Bufo americanus. More toads responded to the presence of prey when fully hydrated than when dehydrated, and times to orient to prey, maneuver around a barrier, and reach prey were less in hydrated than in dehydrated animals. Time to capture prey decreased with increasing T(b) in fully hydrated, but not dehydrated, toads, and hydrated animals caught prey more rapidly than did dehydrated animals. Distance traveled in 5 min and aerobic scope were affected by T(b). Generally, individuals that performed well in the feeding experiments at a particular T(b) and HS also performed well at a different T(b) and HS. The same was true for distance traveled and aerobic scope. However, within combinations of T(b) and HS, correlations between performance variables were minimal. Specialization of a particular variable resulting in high performance at a certain T(b) and HS does not appear to exact a cost in terms of performance at a different T(b) and HS.  相似文献

We reconstruct phylogenetic relationships among a well-studied group of toads and find relationships that differ greatly from the current taxonomic understanding. We use mitochondrial sequences encoding ND1, tRNA(Leu(UUR)), and part of 16S to infer relationships among members of the Bufo americanus complex. Focusing on the four taxa that historically have been most problematic due to morphological similarity and hybridization in sympatry, we sample 150 individuals from multiple populations across each species' geographic range. Our evidence conflicts with previous taxonomic hypotheses that were based on ability to hybridize, geographic distribution, and call variation. First, sequences from B. fowleri do not comprise the sister clade to sequences of B. woodhousii; therefore the previous classifications of B. fowleri as sister species to, or eastern subspecies of, B. woodhousii are both called into question. Second, sequences from B. americanus are more closely related to those of B. woodhousii than to those of B. terrestris, indicating that similar advertisement call characteristics evolved independently. Third, sequences of B. fowleri are paraphyletic, with sequences of B. terrestris embedded within. Lastly, sequences from B. fowleri cluster into three distinct mitochondrial clades, with some divergences corresponding to greater than 2mya. These clades are somewhat geographically structured, suggesting divergence in allopatry during the Pleistocene. These mitochondrial divergences are not accompanied by known phenotypic differences, however, suggesting either evolutionary stasis in morphology and behavior, cryptic phenotypic evolution, or that hybridization in secondary contact has homogenized phenotypic differences that may have arisen in allopatry.  相似文献

Growth and survival of tadpoles (Bufo americanus) were used to assess the food value of three types of amorphous detritus thought to be derived from dissolved organic matter (DOM) transported in river water or groundwater: 1. Flocculent organic aggregates suspended at the mouth of a humic stained river which drains into Lake Superior; 2. Precipitates formed directly from humic stained river water; 3. The < 35 m fraction of littoral lake sediment. Tadpoles were unable to grow when fed the amorphous detritus, but experienced lower mortality than unfed animals. Although the food quality of certain types of detritus derived from DOM may be inadequate to support the growth of aquatic fine particle feeders it may contribute to the nutrition of facultative detritivores.  相似文献

Larvae of certain species of blowflies (Calliphoridae) cause myiasis in amphibians which may result in significant mortality, yet there are few reports from North America. In this study, we observed primary myiasis in a population of juvenile eastern American toads (Bufo americanus americanus) collected during May-July 1998 from southeastern Wisconsin (USA). Nine (6%) of 140 toads were infected by the green blow fly (Bufolucilia silvarum) with a mean intensity of 10.5 +/- 7.2 (range = 1-24). Weekly parasite prevalence and mean intensity remained low, ranging from 0-20% and 2 +/- 1.4 to 14 +/- 6, respectively. We found: 1) flies lay eggs on healthy toads, 2) eggs hatch with first instar maggots penetrating under the skin, 3) maggots develop to mature third instars within 5-7 days, 4) maggots leave the host and form pupa within 8-11 days of hatching, and 5) maggots pupate within 7-9 days at room temperature. All infected toads died within 1-2 wk as a result of the infection. The low prevalence observed in this study and other reports of this species from mammalian and bird carcasses indicated that B. silvarum is probably a facultative parasite of toads and other amphibians in the United States. This is the first report of B. silvarum causing myiasis in Wisconsin amphibians and the first report in eastern American toads in the United States.  相似文献

A population of Bufo americanus from southwestern Ohio exhibited an extreme degree of transferrin and albumin polymorphism. One hundred and eighty-five individuals were collected from this population, and their transferrin and albumin phenotypes were determined by vertical acrylamide gel electrophoresis. Thirteen transferrin alleles were present in 36 phenotypes, and 11 albumin alleles were present in 29 phenotypes. A deficiency of heterozygotes occurred at both protein loci. As many as 20% of all assessed amphibian species are threatened with extinction, and captive breeding programs are becoming important components of conservation strategies for this taxon. For some species, exogenous hormone administration has been integrated into breeding protocols to improve propagation. However, most treatments are administered by an intraperitoneal injection that can be associated with some risks. The general goal of this study was to identify a non‐invasive method of applying luteinizing hormone‐releasing hormone (LHRH), which reliably induces sperm release in toads. Specific objectives were to 1) test the spermiation response after topical application of different LHRH doses to the abdominal seat region, 2) evaluate the effects of adding the absorption enhancers dimethyl sulfoxide (DMSO), acetone, and glyceryl monocaprylate (GMC) to the LHRH, 3) assess the spermiation response after oral delivery of LHRH in a mealworm vehicle, and 4) compare sperm characteristics and spermiation responses to treatments in two different toad species. Male American (n = 9) and Gulf Coast (n = 7) toads were rotated systematically through a series of treatments. Urine was collected and evaluated for the presence of sperm at 0, 3, 7, 12, and 24 hours post‐treatment. There were no statistical differences in spermiation induction or sperm characteristics between American and Gulf Coast toads after the treatments. Oral administration of 100 μg LHRH was occasionally successful in inducing spermiation, but results appeared largely unreliable. Ventral dermal application of 100 or 10 μg LHRH in 40% DMSO were more effective (P < 0.05) at inducing spermiation compared with the other treatments tested, eliciting sperm release in more than 70% of toads tested. In breeding programs for rare and/or fragile anurans, these non‐invasive methods of exogenous hormone administration might be preferred over intraperitoneal injections. One-hundred twelve amphibians, including 51 blue-spotted salamanders, Ambystoma laterale, 30 eastern American toads, Bufo americanus americanus, and 31 northern leopard frogs, Rana pipiens, were collected during April-October 1996 from Waukesha County, Wisconsin and examined for helminth parasites. The helminth compound community of this amphibian assemblage consisted of at least 10 species: 9 in American toads, 8 in leopard frogs, and 3 in blue-spotted salamanders. American toads shared 7 species with leopard frogs, and 2 species occurred in all 3 host species. Although there was a high degree of helminth species overlap among these sympatric amphibians, statistically significant differences were found among host species and percent of indirect or direct-life cycle parasites of amphibian species individual component communities (chi2 = 1,015, P < 0.001). American toads had a higher relative abundance of nematodes, 59%, than larval cestodes, 31%, and larval and adult trematodes, 10%, whereas leopard frogs had a higher relative abundance of larval cestodes, 71.3%, and larval and adult trematodes, 25.3%, than nematodes 3.4%. This is related to ecological differences in habitat and dietary preferences between these 2 anuran species. Helminth communities of blue-spotted salamanders were depauperate and were dominated by larval trematodes, 94%, and few nematodes, 6%. Low helminth species richness in this host species is related to this salamander's relatively small host body size, smaller gape size, lower vagility, and more fossorial habitat preference than the other 2 anuran species. Adult leopard frogs and toads had significantly higher mean helminth species richness than metamorphs, but there was no significant difference in mean helminth species richness among adult and metamorph blue-spotted salamanders. Considering adult helminths, the low species richness and low vagility of caudatans as compared with anurans suggest that local factors may be more important in structuring caudatan helminth communities of salamanders than of anuran hosts. Helminth species infecting salamanders may be more clumped in their geographic distribution as compared with anurans, and the role of other hosts and their parasites at the compound community level may be important in structuring helminth communities of salamanders.  相似文献

Acidification of breeding ponds has been identified as a potential threat to the survival and health of North American amphibian populations. The effects of acid exposure on ion and acid-balance are well known, but there is little information on how environmental water pH influences nitrogen balance in amphibians. The aim of this study was to determine the effects of moderately acidic water (pH 6.0) on nitrogen excretion in early life stages of the toad, Bufo americanus. Acid exposure (pH 6.0, 54 h) resulted in a 20–80% increase in ammonia-N excretion rates in embryos and early, middle and late tadpoles stages, whereas there was no significant effect on urea-N excretion. Tissue ammonia concentrations were significantly higher (+ 33%) in the embryos and 35–65% lower in the three groups of tadpoles exposed to water of pH 6.0 compared to control animals (pH 8.5). In embryos, ammonia excretion accounted for greater than 90% of total nitrogen excretion (ammonia-N + urea-N), but by the late tadpole stage this value had decreased to approximately 65%. These findings indicate that exposure of embryonic and larval B. americanus to moderately acidic water disrupts nitrogen balance by increasing nitrogen loss as ammonia, with no compensatory decrease in urea excretion.  相似文献

Trypanosoma fallisi n. sp. is described from Bufo americanus in Ontario. The parasite was observed in 65 of 94 toads examined. The trypanosomes were pleomorphic with respect to the age of infections, being longer and broader in early infections (during spring and summer) and shorter and more slender during late summer and autumn. They ranged in size from 38–76 μm in body length and 3–8 μm in width, with a free flagellum 6–30 μm long. Epizootiological and experimental evidence suggests that this trypanosome is transmitted to the toads by the leech, Batracobdella picta. Trypanosoma fallisi is morphologically similar to T. bufophlebotomi described in Bufo boreas from California, but geographic isolation, host and vector differences as well as slight morphological differences indicate that speciation has occurred. Similar trypanosomes from Bufo americanus (which were identified as T. bufophlebotomi) in Michigan, are probably T. fallisi. This species shares many ultrastructual features with trypanosomes of other lower vertebrates and also of mammals.  相似文献
